Claims (9)
- 1.生物学的試料から出発して、それが含有するmRNAの逆転写を行い、 転写生成物について(または直接試料から抽出されたDNAについて)、別個の 増幅を次いで各プライマー対V、C(Vは問題のレパートリーの可変セグメント に対応し、Cは研究中のレパートリーの一定セグメントでハイブリッド形成する )用のPCR型方法によって行い、プライマーとしてのJセグメントおよび鋳型 としての増幅生成物に特異的なオリゴヌクレオチドを使用して、これらの増幅生 成物の各々について、延長工程を標識レパートリーの各Jセグメントの場合に行 い、それによって得られるトリプレット(V、C)Jに対応する各延長生成物に 対して、異なる延長生成物の大きさを現わし、 レパートリーの記載はレパートリーの各エレメントに対してVCJトリプレット およびエレメントの大きさに対応する ことを特徴とする個体の免疫系の抗体(Ab)およびT細胞受容体(TcR)の レパートリーを記載するための方法。
- 2.延長生成物の大きさを延長混合物の電気泳動および標識の検出によって現わ すことを特徴とする請求項1に記載の方法。
- 3.標識が放射性、比色または蛍光であることを特徴とする、請求項1ないし2 のいずれか1項に記載の方法。
- 4.工程をMg2+イオン1〜3mMの存在下で行うことを特徴とする、請求項 1ないし3のいずれか1項に記載の方法。
- 5.分子大きさの各信号の強度を測定して対応配列の程度の定量的測定を得るこ とを特徴とする、請求項1ないし4のいずれか1項に記載の方法。
- 6.各信号の強度を内標準と比較して評価することを特徴とする、請求項5に記 載の方法。
- 7.同じ分子大きさの生成物を固定プローブに対する多少の親和力に従って分離 することを特徴とする、請求項1ないし6のいずれか1項に記載の方法。
- 8.温度または他の変性剤の勾配を有する電気泳動を使用して、この工程を行う ことを特徴とする、請求項7に記載の方法。
- 9.自動配列決定装置を使用して延長生成物の大きさを現わし且つ記載すること を特徴とする、請求項1ないし8のいずれか1項に記載の方法。
