Abstract
Description
Claims (10)
- 1.サンプル溶液から所望の成分を回収する方法であって、 (i)回収する所望の成分を含むサンプル溶液を、該成分に親和性を有するビー ズを含むカラムに、差流機を備えた下部出口から、安定化流動床の条件で供給し 、それによってサンプル溶液の未結合成分を該カラムの上部出口から排出させ、 (ii)膨張床を維持する流れ条件で、該下部出口から導入した溶液でカラムを 洗浄し、それによってサンプル溶液中の残存固体成分および他の不純物をカラム の上部出口から排出し、 (iii)該ビーズに結合したサンプル成分を遊離させる溶出溶液を導入し、該 成分を採取する ことから成る当該回収方法。
- 2.工程(iii)が、該ビーズを落ち着かせ、場合によっては上部アダプター を該床表面に下げ、さらに充填床が形成されるように該カラムの上部出口から溶 液を導入し、その後該成分をビーズから遊離させる溶出溶液を上部出口から導入 して該カラムから溶出させることによって行われる、請求項1に記載の方法。
- 3.流動床を形成するビーズが、請求項5〜10のいずれかに記載のクロマトグ ラフィー用支持体ビーズであることを特徴とする、請求項1または2に記載の方 法。
- 4.該サンプル溶液が固形細胞粒子を含む細胞培養液であることを特徴とする、 請求項1〜3のいずれかに記載の方法。
- 5.重合体マトリックスから成り、その中にガラスまたはシリカ粒子を含有する ことを特徴とする、ダウンストリーム処理で使用する支持ビーズ。
- 6.該重合体が、場合によって架橋された、多糖類であることを特徴とする、請 求項5に記載の支持ビーズ。
- 7.該重合体がアガロースであることを特徴とする、請求項6記載の支持ビーズ 。
- 8.100〜1000μmの直径を有し、シリカまたはガラスの含有量が5〜5 0%(湿潤重量)であることを特徴とする、請求項5〜7のいずれかに記載の支 持ビーズ。
- 9.該ガラスまたはシリカ粒子が1〜100μmの範囲の粒度分布を有すること を特徴とする、請求項5〜8のいずれかに記載の支持ビーズ。
- 10.用いるシリカ粒子が石英粒子であることを特徴とする、請求項5〜9のい ずれかに記載の支持ビーズ。
