〔産業上の利用分野〕
本発明は、イブプロフエン経口製剤に関し、さ
らに詳しくはステアリン酸マグネシウムを添加す
ることを特徴とする経口製剤に関する。
〔従来の技術〕
イブプロフエン〔2−(4−イソブチル)−プロ
ピオン酸、C13H18O2(分子量、206.28)〕は非ステ
ロイド系抗炎症剤で、鎮痛、解熱薬として広く用
いられている。
何ら処置をしない場合、イブプロフエン製剤か
らのイブプロフエンの最高血中濃度到達時間
(Tmax)は1.5〜3時間であり、〔塩川ら(診療
と新薬、第18巻、第2139頁、1981年)〕速効性を
必要とする解熱鎮痛剤としては不十分である。
このため、2対3のモル比でイブプロフエンを
β−シクロデキストリンで包接することにより、
Tmaxを約2.5倍早める試みがなされた。〔チヨウ

〔発明が解決しようとする問題点〕
しかしながら、この方法は分子量1000を越える
β−シクロデキストリンをイブプロフエンに対
し、1.5倍も加えてしまうと、製剤的に嵩高くな
るという欠点がある。
本発明の目的は最高血中濃度到達時間
(Tmax)が短いイブプロフエン経口製剤を提供
することにある。
〔問題を解決するための手段〕
本発明者らは、上述の問題に鑑み、鋭意検討を
重ねた結果、イブプロフエンに対し5重量%以上
好ましくは10〜50重量%のステアリン酸マグネシ
ウムを添加剤として加えることにより、最高血中
濃度到達時間(Tmax)を早め、バイオアベイラ
ビリテイーを高めることができた。すなわち、イ
ブプロフエンに対し、ステアリン酸マグネシウム
を1重量%添加したものではTmaxおよび血中濃
度曲線下面積(AUC)とも対照品と同等である
のに対し、5%添加するとTmaxが速くなり、吸
収が良くなる。さらに、30重量%添加することに
より、Tmaxは対照品の約1/2時間となり、AUC
は約2倍の値を示した。ここでイブプロフエンに
対し、5%以上の割合でステアリン酸マグネシウ
ムを添加することによつて、速効性で、かつ、バ
イオアベイラビリテイーを高めることができた。
一方、配合割合の上限に関しては、特に制限す
るものではないが、イブプロフエンと同量を越え
て配合すると、製剤上嵩高くなつてしまい、実用
上、1回の投与量が増すなどの問題を生じる。好
ましくは、イブプロフエンに対し、50重量%以下
であり、この範囲内の添加量であれば、イブプロ
フエン製剤の吸収性を改良するだけでなく、製剤
設計上問題が無い。
本経口製剤を製造するに当り、剤形を特に規定
するものではない。例えば、錠剤、丸剤、顆粒
剤、細粒剤、散剤、トローチ剤、カプセル剤など
が挙げられる。
ここでいうステアリン酸マグネシウムは、日本
薬局方に基づいたもの(成分的には、ステアリン
酸とパルミチン酸のマグネシウム塩の混合で、本
品を乾燥したものは、3.9〜5.1重量%のマグネシ
ウムを含有する。)でもよい。
〔発明の効果〕
本発明に係るステアリン酸マグネシウム配合イ
ブプロフエン製剤は、従来のものに比べ、速く薬
効を発揮することができる。また従来のものと同
等の薬効を発揮させるためのイブプロフエン配合
量を少なくすることができることから、潰瘍形成
等の副作用を低減させるだけでなく、製剤の小型
化およびコストの低減をもたらす。
(実施例)
以下、実施例及び試験例をもつて本発明を具体
的に説明する。
実施例 1
イブプロフエン 150g、低置換度ヒドロキシ
プロピルセルロース 70g及び乳糖70gから成る
均質な粉末混合物を調製し、常法により顆粒化し
た。別にステアリン酸マグネシウム7.5g、軽質
無水ケイ酸4.5gを均質化した。2つの粉末混合
物を合わせ、さらにタルク12.0gを加えて、混合
均質化したのち、常法により3号カプセル2000個
に充填し、1カプセル当り以下に示す処方製剤を
得た。

実施例 3
ステアリン酸マグネシウム 100g、軽質無水
ケイ酸、 60gを混合し、均質化した。その後乳
糖 740g、イブプロフエン 300g、デンプン
750gを加えて混合した。さらにヒドロキシプロ
ピルセルロース 50gに精製水500ml加えて溶解
したものを結合剤として、混練したのち、常法に
従つて、1包当り、以下の処方に示す顆粒剤2000
包を製造した。

実施例 4
ステアリン酸マグネシウム 300g、軽質無水
ケイ酸 100g、乳糖 1250g イブプロフエン
300gを加えて混合した。
さらにヒドロキシプロピルセルロース 50gに
精製水 500ml加えて溶解したものを結合剤とし
て混練し、乾燥した。その後、常法に従つて粉砕
し、1包当り、以下の処方に示す散剤2000包を製
造した。

プロピルセルロース 75g及び乳糖90gから成る
均質な粉末混合物を調製し、常法により顆粒化し
た。別にステアリン酸マグネシウム 20g、軽質
無水ケイ酸 10gを均質化した。2つの粉末混合
物を合わせ、さらに、タルク 15gを加えて混合
均質化した後、常法により、直径8mm、重量180
のmg以下の処方に示す錠剤2000錠を打錠した。

これら4つの製剤について雌性ビーグル犬各4
頭を用いて、バイオアベイラビリテイー評価を実
施した。
(方法)
体重約10Kgの雌性ビーグル犬を投与前日より、
18時間絶食させ、その後、試験製剤を水約100ml
とともに経口投与した。投与後の犬の状態は良好
で吐くことはなかつた。
採血は投与前、及び投与後、0.5,1,2,3,
4,6,8時間毎に前肢静脈より、ヘパリン処理
注射筒にて行なつた。採血した血液は、3000rpm
で10分間遠心分離し、その血漿中のイブプロフエ
ン量を高速液体クロマトグラフイー法により定量
した。その結果を表1および図1に示した。
