Description
〔産業上の利用分野〕
本発明はアルギン酸のアルカリ土類金属塩をア
ルギン酸リアーゼの作用により分解する方法、特
に上記方法においてアルカリ土類金属キレート能
物質および/または界面活性剤の存在下に行なう
アルギン酸のアルカリ土類金属塩の分解の改良方
法に関する。
〔従来の技術〕
アルギン酸リアーゼは、アルギン酸およびその
塩に作用してそれらを分解し、低分子化させる酵
素である。
アルギン酸およびその塩は多くの用途に使用さ
れている。例えば経糸剤、食品加工、塗料、歯科
印象材等にその用途が開かれている。そしてこれ
らの用途においてはアルギン酸および/またはそ
の塩は使用後これを除去する必要があることがあ
る。
例えば歯科印象材のアルギン酸カルシウムゲル
は使用後、トレイ等からこれらを除去しなければ
ならない。かかる場合従来はエチレンジアミン四
酢酸(EDTA)などのキレート能物質を主成分
とする溶解液を用い、化学的にアルギン酸およ
び/またはその塩を可溶化させている。しかしな
がらEDTAは金属を腐蝕する作用があり、基材
を損傷することがあるので好ましくない。またク
エン酸またはリン酸の如き酸あるいはそれらの塩
を用いてアルギン酸および/またはその塩のゲル
除去をする方法もあるが、これらはその溶解作用
が弱いため効率的でない。
上述した化学的方法に代るものとしてアルギン
酸リアーゼを使用した例としてはアルギン酸を分
解するアルギン酸リアーゼの力価測定方法が知ら
れている(例えばメソツズ・イン・エンジモロジ
ー第8巻第641頁〜第644頁、1966年、参照)。
アルギン酸リアーゼは上述した化学的方法と異
なり、アルギン酸および/またはその塩にのみ作
用して、これを分解し、低分子化して除去を容易
にすると共に、アルギン酸および/またはその塩
が適用された他の材料を損うことがないので非常
に有利である。
かかるアルギン酸リアーゼを生産する微生物と
してはシユウドモナス属(ジヤーナル・オブ・バ
イオケミストリー、第66巻第503頁〜第512頁、
1969年)、エーロモナス属(科学と工業、第43巻

〔発明が解決しようとする問題点〕
アルギン酸リアーゼは上述した化学的方法に比
してアルギン酸および/またはその塩を分解する
のに有利なのであるが、その分解能力、特に分解
速度において未だ充分とはいえず、特にアルギン
酸のアルカリ土類金属塩を分解する場合には分解
速度の向上が望まれているのが現状である。
従つて本発明の目的はアルギン酸リアーゼによ
るアルギン酸のアルカリ土類金属塩の分解速度を
向上させる改良法を提供することにある。
〔問題点を解決するための手段〕
本発明はアルギン酸のアルカリ土類金属塩をア
ルギン酸リアーゼの作用により分解するに当り、
アルカリ土類金属キレート能物質および/または
界面活性剤の存在下に行なうアルギン酸のアルカ
リ土類金属塩を分解する方法にある。
本発明方法で使用するアルギン酸リアーゼはそ
の起源は特に限定されるものではなく、前述した
公知の例えばシユードモナス属、エーロモナス
属、クレブシエラ属およびバシラス属等の微生物
を公知の方法で培養することによつて得られるア
ルギン酸リアーゼを使用できる。
また本発明者によつて新たに見出されたキサン
トモナス属(Xanthomonas)に属するアルギン
酸リアーゼ生産菌を培養し、この培養物より採取
したアルギン酸リアーゼも使用でき、これはその
生産量が大ですぐれているので好ましい。このキ
サントモナス属の微生物からのアルギン酸リアー
ゼを製造する方法は本願と同日付にて出願した特
許出願(特開昭63−39581号)に記載されている。
よつて同特許出願明細書の記載はここに引用して
組入れるものとする。
上記キサントモナス属に属する微生物はアルギ
ン酸リアーゼを培養物中に著量生産することがで
きるので有利である。これについて以下に更に説
明する。
かかる菌株としては、キサントモナス属に属す
るアンギン酸リアーゼ生産能を有する菌株であれ
ばいかなる菌株でもよく、またそれらの変異株で
もよい。そしてキサントモナス属に属し、アルギ
ン酸リアーゼ生産能を有する菌株の具体例として
は、例えばキサントモナス・エスピーN−26
(Xanthomonas spN−26)を挙げることができ
る。本菌株は京都府福知山市内の畑地の土壌より
分離された。本菌株の菌学的性質は次のとおりで
ある。
a 形態的性質
顕微鏡的観察(肉汁液体培地で30℃で培養)
(1) 細胞の形および大きさ:通常細胞の大きさは
4〜5μm×0.6〜0.7μm。
(2) 細胞の多形性の有無:なし、単独である。
(3) 運動性の有無:有、極鞭毛を有する。
(4) 胞子の有無:なし。
(5) グラム染色性:陰性。
b 各培地における生育状態
(1) 肉汁寒天平板培養:
30℃48時間の培養で、直径2〜5mmの凸円形コ
ロニーを形成する。表面は滑らかでやや黄色。
(2) 肉汁寒天斜面培養:
30℃の培養で糸状、周縁は滑らかで光沢があ
り、生育は普通。
(3) 肉汁液体培養:
30℃の培養で24時間で生育し、生育は普通。
(4) 肉汁ゼラチン穿刺培養:
30℃の培養で24時間で生育し、生育は普通。液
化は漏斗状。
(5) リトマスミルク培養:
30℃の培養で凝固せず、色調は青紫色で変化な
し。
c 生理学的性質
(1) 硝酸塩の還元:陰性
(2) 脱窒反応:陰性
(3) MRテスト:陽性
(4) VPテスト:陰性

ス、アントール、ガラクトース、ラムノース、
ラフイノース、イノシトール
以上の諸性質を「バージーズ・マニユアル・オ
ブ・デタミネイテイブ・バクテリオロジイ」第8
版(1974年)より検索するとキサントモナス・キ
ヤンベストリスと類似していることが分かつた
が、糖類からの酸の生成などにおいて異なつてい
るのでキサントモナス属の新種と判断しキサント
モナス・エスビーN−26と命名した。なお本菌株
は工業技術院微生物工業技術研究所に微生物受託
番号微工研菌寄第8800号として寄託されている。
本菌株は微生物の培養に通常用いられる栄養物
たとえば肉エキス、ペプトン、酵母エキス、麦芽
エキス、コーンスチープリカー、カザミノ酸など
にアルギン酸およびアルギン酸の塩を0.01〜2
%、好ましくは0.1%〜1%を加えた培地で20℃
〜40℃、好ましくは27℃〜36℃の好気的条件で良
好に増殖し、培養5〜40時間でアルギン酸リアー
ゼを生産する。
本発明の菌によつて生産されたアルギン酸リア
ーゼは菌体内に存在するので菌体からのアルギン
酸リアーゼの抽出はビーズや超音波を用いた細胞
破砕、有機溶媒抽出などが使用できるが好ましく
は以下の方法で行なう。アルギン酸リアーゼを生
産し菌体内に蓄積した菌体を遠心等によつて集め
緩衝液に分散させた液、あるいは菌体を含んだ培
養液に、界面活性剤たとえばトリトンX−100を
0.01%〜5%、好ましくは0.05%〜2%を加え
る。この時リゾチームを0.001〜10mg/ml、好ま
しくは0.01〜1mg/ml加えると抽出効果が良くな
る。処理後、添加量や菌体濃度によつても異なる
が、1分〜5時間放置したのち凝集剤による凝
集、遠心などにより菌体残さを除去すると粗酵素
液を得ることができる。本発明で使用される上記
アルギン酸リアーゼは上記の粗酵素液に限定する
ものではなく粗酵素液の処理物たとえば有機溶媒
沈殿、塩析、クロマトグラフイーなどの公知の方
法で精製した酵素液および、それらの乾燥物、固
定化物さらには細胞懸濁液でもよい。
次に上記のキサントモナス・エスビーN−26よ
り得られるアルギン酸リアーゼの酵素化学的およ
び理化学的性質は次のとおりである。
(1) 作用:アルギン酸を基質として反応させた
時、アルギン酸リアーゼの反応生成物である二
重結合に由来する235nmの特異吸収の増加が確
認された。
(2) 基質特異性:

In such cases, alginic acid and/or its salts have conventionally been chemically solubilized using a solution containing a chelating substance such as ethylenediaminetetraacetic acid (EDTA) as a main component. However, EDTA is not preferred because it has the effect of corroding metals and may damage the base material. There is also a method of gel removal of alginic acid and/or its salt using an acid such as citric acid or phosphoric acid or a salt thereof, but these methods are not efficient because their dissolving action is weak. As an example of using alginate lyase as an alternative to the chemical method described above, a method for measuring the titer of alginate lyase that decomposes alginic acid is known (for example, Methods in Engineering Vol. 8, pp. 641-644). p. 1966). Unlike the chemical methods mentioned above, alginate lyase acts only on alginic acid and/or its salts to decompose it and reduce its molecular weight to make it easier to remove. This is very advantageous because it does not damage the material. Microorganisms that produce such alginate lyase include the genus Pseudomonas (Journal of Biochemistry, Vol. 66, pp. 503-512).

Microorganisms such as the genus Bacillus (Applied Endo-Environmental Microbiology, Vol. 47, pp. 704-709) are known. [Problems to be Solved by the Invention] Although alginate lyase is more advantageous in decomposing alginic acid and/or its salts than the above-mentioned chemical methods, its decomposition ability, especially its decomposition rate, is still insufficient. However, the current situation is that it is desired to improve the decomposition rate, especially when decomposing alginic acid alkaline earth metal salts. Accordingly, it is an object of the present invention to provide an improved method for increasing the rate of decomposition of alginic acid alkaline earth metal salts by alginate lyase. [Means for Solving the Problems] The present invention provides the following steps when decomposing alginic acid alkaline earth metal salts by the action of alginate lyase.
A method for decomposing an alkaline earth metal salt of alginic acid in the presence of an alkaline earth metal chelating substance and/or a surfactant. The origin of the alginate lyase used in the method of the present invention is not particularly limited, and can be obtained by culturing the aforementioned microorganisms such as Pseudomonas, Aeromonas, Klebsiella, and Bacillus by a known method. The resulting alginate lyase can be used. In addition, alginate lyase produced by culturing alginate lyase-producing bacteria belonging to the genus Xanthomonas, which was newly discovered by the present inventor, can also be used. It is preferable because there is. This method for producing alginate lyase from microorganisms of the genus Xanthomonas is described in a patent application (Japanese Patent Application Laid-open No. 39581/1983) filed on the same date as the present application.
Therefore, the description of the patent application specification is hereby incorporated by reference. The microorganisms belonging to the genus Xanthomonas are advantageous because they can produce alginate lyase in significant amounts in culture. This will be further explained below. Such strains may be any strains belonging to the genus Xanthomonas that have the ability to produce anginate lyase, or may be mutants thereof. Specific examples of strains that belong to the genus Xanthomonas and have the ability to produce alginate lyase include, for example, Xanthomonas sp.
(Xanthomonas spN-26). This bacterial strain was isolated from the soil of a field in Fukuchiyama City, Kyoto Prefecture. The mycological properties of this strain are as follows. a. Morphological properties Microscopic observation (cultured at 30°C in broth liquid medium) (1) Cell shape and size: Normal cell size is 4-5 μm x 0.6-0.7 μm. (2) Presence or absence of cell pleomorphism: None, isolated. (3) Motility: Yes, with polar flagella. (4) Presence or absence of spores: None. (5) Gram staining: Negative. b Growth status in each medium (1) Broth agar plate culture: Form convex circular colonies with a diameter of 2 to 5 mm by culturing at 30°C for 48 hours. The surface is smooth and slightly yellow. (2) Juicy agar slant culture: When cultured at 30°C, the growth is normal, with thread-like appearance and smooth and glossy edges. (3) Meat juice liquid culture: Grows in 24 hours when cultured at 30℃, and growth is normal. (4) Meat juice gelatin puncture culture: Grows in 24 hours when cultured at 30°C, and growth is normal. Liquefaction is funnel-shaped. 本発明により、理論的には不明であるがアルギ
ン酸リアーゼと共に、アルカリ土類金属キレート
能物質および/または界面活性剤を使用すると、
従来のアルギン酸リアーゼ単独使用の場合に比し
て、アルギン酸のアルカリ土類金属塩の分解反応
速度が大となり、更に他の共存する材料を腐蝕等
により損うことがない。
〔実施例〕
以下に実施例を挙げて本発明を説明する。
実施例 1
歯科用印象剤ブロツク(2.5×2.5×2.5cm19.5
g)を、PH6.3に調整したアルカリ土類金属キレ
ート能物質および/または界面活性剤を含むアル
ギン酸リアーゼ水溶液にて30℃、20時間、静置反
応させた。溶け残つたブロツクの重さを天秤にて
測定し、下記式により溶解した印象剤ブロツクの
割合(溶解率(%))にてその効果を判定した。
ただし、PH調整はNaH2PO4水溶液にて実施し
た。その結果を表2に示す。
