【発明の詳細な説明】
〔技術の分野〕
本発明は、カルシウム依存性酸素添加酵素の調
製法に関する。

リンの蛋白部分を還元剤及び金属イオン処理する
ことにより、基質に酵素を連続的に添加し、連続
的発光能力を有する酵素を調製する方法に関す
る。

天然に存在するカルシウム受容発光蛋白エクオ
リンは、微量のカルシウムイオンの存在により、
数秒内にその蛋白内に存在するエミツターである
セレンテラジンが酸化して発光する。この一連の
反応は不可逆であり、連続的に長時間発光を持続
することは困難である。

本発明者は、前述の問題を解決すべく遺伝子工
学及び蛋白質工学の手法を用いて研究を行つた。

酸素反応を行なわせることができれば、瞬時に発
光するエクオリンより検出方法としての汎用性が
増大し、連続の発光をX−線フイルム等で検出す
ることが可能となる。このことはエクオリンの放
射性同位元素の代用としての利用も可能になり、
利用価値がある。

カルシウム添加による分解・発光する機能を利用
することにある。

天然あるいは組換えDNAで製造したエクオリ
ンは、アポエクオリン−酸素−セレンテラジンが
コンプレツクス状に存在し、Ca2+イオンで瞬時
に発光する。この反応は不可逆であり、基質添
加、還元剤の添加で、非常に微弱の発光しか検出
できず、実用性に乏しい。

そこで本発明は、アポエクオリンの高次構造の
変化を考慮して還元剤の処理方法、Ca2+イオン
等の添加順序等の検討を行うことならびに十分な
実用性を有するアポエクオリン、すなわち連続的
発光を触媒するカルシウム依存性酸素添加酵素の
調製法を確立することを目的とする。

本発明者は、以上の目的で研究中のところ、ア
ポエクオリンを適当な濃度の還元剤で処理して還
元型アポエクオリンとし、金属イオンを反応させ
ることにより得られたカルシウム依存性酸素添加
酵素が上述の問題点(註.連続的長時間発光の持
続の不能)を解決し得ることを知つて本発明を完
成した。

本発明は下記(1)の主要構成と(2)ないし(3)の実施
態様的構成を有する。

本発明の構成と効果につき以下に詳述する。
A 本発明に使用するアポエクオリン(E1):
天然産の発光蛋白(エクオリン)を常法により
分離精製して得られたアポ蛋白である。

量のカルシウム源(例えばCaCl2)を添加して発
光させ、該発光後の反応物にカルシウム除去剤例
えばEDTA(エチレンジアミンテトラ酢酸)の適
量を加えてエクオリンと結合しているCa2+を除
去し、被除去物を分離処理例えば、カラムクロマ
トグラフイーによつて分離することによりアポエ
クオリンを取得する。

したアポエクオリン(註.先行文献については後
述実施例参照)または、このものを例えば陰イオ
ン交換クロマトグラフイーによりさらに精製した
ものであつてもよい。

B アポエクオリンの還元:
アポエクオリンの還元とは、該蛋白のジスフイ
ド結合(−S−S−)の開裂を意味する。かゝる
還元に使用可能な還元剤は限定されないが例え
ば、メルカプトエタノール若しくはジチオスレイ
トールを挙げることができる。還元条件は限定さ
れないが、例えば0℃〜40℃好ましくは0℃〜10
℃で、10分〜10時間好ましくは30分〜3時間アポ
エクオリン溶液と還元剤の混合液を静置する。

トしたのち、適量の例えば30mM CaCl2溶液を注
入する。基質セレンテラジンと30mM CaCl2溶液
の混合順序は逆にしてもよい。このようにセツト
されたカルシウム依存性酸素添加酵素の連続的な
発光を相対発光値として記録する(註.後述実施
例及び図1,2参照)。

例及び図1,2参照)。

本発明によれば、天然のエクオリンより分離し
たアポエクオリンまたは、組換えDNA法で製造
したアポエクオリンを適当濃度の還元剤で処理し
て還元型アポエクオリンを取得したのち、これに
カルシウムイオン及び基質セレンテラジンを添加
することにより連続的に発光させることが可能と
なる。また、上述のような連続発光が可能となる
ことにより、この発光をX線フイルム等で撮影
し、最終的にカルシウムイオンの検出が可能にな
る。

〔実施例〕
アポエクオリンの調製法
(1) 天然エクオリンからの調製法
天然のエクオリン(100μg)に30mM CaCl2
(500μ)を添加し、発光させる。発光後100mM
EDTA(エチレンジアミンテトラアセト酢酸500μ
)を加えCa2+を除去した後、セフアテツクス
G25カラムクロマトグラフイー(100×10φmm)に
よりアポエクオリンを得る。

ラジン2μを注入し、連続的な発光をレコダー
により相対発光値(relative light units, rlu)
で記録する。

そこで、最初に還元剤で処理(12時間処理)し
たアポエクオリンについて示したのが図2であ
る。図2において、 aは還元剤処理をしたアポエクオリン(A′)
と基質セレンテラジン(B)を混合し、2分間後
にカルシウム(D)に注入し、その連続発光を示
したものであり、 bは還元剤処理したアポエクオリン(A′)と
カルシウム溶液(D)を混合し、2分間後に基質
セレンテラジン(B)を注入し、その連続発光を
示したものである。図2a,bの場合明らかに図
1の未処理のアポエクオリンに比べ、約5倍程度
発光活性が高く、しかも同程度に連続的に発光す
る。すなわち、連続的に機能するカルシウム依存
性酸素添加酵素が調製できたわけである。

性酸素添加酵素が調製できたわけである。
これらの一連の反応を、エクオリンを含めて示
したのが、図3であり、還元剤処理により連続的

Claims (1)
- 【特許請求の範囲】 1 アポエクオリン(E1)を還元剤で処理して
得られた還元剤処理アポエクオリン(E2)に金
属イオンを反応せしめることを特徴とするカルシ
ウム依存性酸素添加酵素の調製法。 2 アポエクオリンとして、天然産の発光蛋白よ
り得られたまたは組換えDNAの手法により得ら
れたアポ蛋白を用いる特許請求の範囲第1項に記
載の方法。 3 金属イオンとしてカルシウムイオン若しくは
ストロンチウムイオンを用いる特許請求の範囲第
1項に記載の方法。
