Description

請求の範囲 １ カプセル状の容器に入つた複数のマイクロペ
レツトからなり、該マイクロペレツトの合計がテ
オフイリンの投与単位を形成し、該マイクロペレ
ツトの各々が、その上をテオフイリンとポリビニ
ルピロリドンの混合物で被覆されシードを基礎と
し、さらに、かかる被覆シードがその上をヒドロ
キシプロピルセルロースとエチルセルロースの混
合物で被覆されていることを特徴とする食物に散
布して摂取させるテオフイリンの徐放性投与処
方。
２ 該ポリビニルピロリドンが約30000〜50000の
分子量を有する前記第１項の徐放性投与処方。
３ 該ポリビニルピロリドンが約40000の分子量
を有する前記第２項の徐放性投与処方。
４ 該シードが60〜80メツシユ（0.18〜0.25mm）
の糖である前記第１項の徐放性投与処方。
５ 該マイクロペレツトが約0.6mmの平均直径を
有する前記第１項の徐放性投与処方。
明細書 ゼロ次放出テオフイリン製品、THEO−DUR
は、気管支拡張剤として市場および医者の間で、
広範に受け入れられている。子供および年長者の
投与により適した形での徐放性気管支拡張剤が提
供されることが望まれている。
本発明に従えば、比較的大きな錠剤を飲みこむ
ことが困難である子供または年長者に投与するた
めに設計された徐放性テオフイリンのマイクロペ
レツトが提供される。マイクロペレツトは、投与
単位形態としてカプセルで提供され、各々のカプ
セルは所望の総投与量をもたらすために、約1000
個またはそれ以下の微粒子を含有する。例えば、
一つの態様において、100mg用量のテオフイリン
は845個の微粒子で提供される。該微粒子は分子
量約30000〜約50000を有するポリビニルピロリド
ン、好ましい態様では約40000のもの
（Kollidon30）と共に60〜80メツシユ（0.18〜
0.25mm）の糖シード（Seed）上に微細化した形の
テオフイリンを被覆することにより与えられる。
徐放性被膜はセルロース誘導体で与えられる。セ
ルロース誘導体としては、エチルセルロース
（Dow，Etocel Ｎ−10）およびヒドロキシプロ
ピルセルロース（Hercules，Klueck LF））の混
合物が使用される。各々の粒子の平均直径は、お
およそ約0.6mmである。
次の実施例により、本発明を説明する。
実施例 ポリビニルピロリドン（分子量40000、
Kollidon30）3.2Kgをイソプロパノール32に溶
解させる。その中に、微細化したテオフイリン
12.8Kgを分散させる。グラツト（Glatt）カラム
（６インチ、Glatt CPCG）の中に、60〜80メツ
シユの砂糖４Kgを充填する。該砂糖で空気懸濁系
を活動させ、温度60℃の流入空気を用い、スプレ
ー圧４バールおよびスプレー速度100ml／分で、
分散させたテオフイリンを該グラツトカラムに充
填する。
上記操作の完了後、グラツトカラムを停止し、
この製品を「テオフイリンペレツト、活性」と
して、取つておく。
ポリビニルピロリドン（分子量40000、
Kollidon 30）3.2Kgの第２番目のバツチをイソプ
ロパノール32.0中に溶解させ、得られた混合物
中に微細化したテオフイリン12.8Kgを分散させ
る。ついで、テオフイリンペレツト、活性４Kg
を温度、圧力および速度の同一条件下で、同一の
グラツトカラムの中に充填する。ついで、その中
にテオフイリンを分散させた該第２番目のバツチ
をグラツトカラムに充填してさらに被覆する。
グラツトカラムをからにし、その製品に「テオ
フイリンペレツト、活性」のラベルを付す。
クロロホルム13.2およびメタノール3.3の
混合物を調製し、その中にエチルセルロース
（Dow，Ethocel Ｎ−10）992.0ｇおよびヒドロキ
シプロピルセルロース（Hercules，Klucel LF）
329.0ｇを分散させる。グラツトカラムの中に、
テオフイリンペレツト、活性19.0Kgを充填し、
ついで、それらを、30℃、スプレー圧３バールお
よびスプレー速度100ml／分の条件下、該被膜混
合物で被覆する。得られた被覆ペレツトは小さな
白色マイクロペレツトである。845個の該マイク
ロペレツトを用いて100mgの投与単位をつくり、
便宜のためにカプセルに入れる。子供または年長
の患者については、典型的には、該微粒子を食物
の中に散布し、ついで食事の一部として摂食さ
せ、持続性のテオフイリン効果を与える。
