Description
【発明の詳細な説明】
本発明はクロルヘキシジン類等のビグアニド系
化合物で代表されるカチオン性殺菌剤を配合した
口腔用組成物に関し、更に詳述すればクロルヘキ
シジン類等のカチオン性殺菌剤の非イオン性界面
活性剤による失活を防止した口腔用組成物に関す
る。
従来、歯磨等の口腔用組成物に有効成分として
クロルヘキシジン類等のビグアニド系化合物で代
表されるカチオン性殺菌剤を配合し、これらの強
力な殺菌作用により歯周疾患の予防,治療を行な
うことが知られており、またクロルヘキシジン類
等をシヨ糖脂肪酸エステル等の非イオン性界面活
性剤を含む口腔用組成物に配合することも知られ
ている。
しかしながら、本発明者らの検討の結果では、
クロルヘキシジン類等のカチオン性殺菌剤を非イ
オン性界面活性剤と併用した場合、カチオン性殺
菌剤が著しく失活し、その効果が十分に発揮され
ないことを知見した。
このため、本発明者らは、クロルヘキシジン類
等のカチオン性殺菌剤を非イオン性界面活性剤を
含む口腔用組成物に安定して配合し、その強力な
殺菌効果を有効に発揮させることについて鋭意検
討を行なつた結果、クロルヘキシジン類等のカチ
オン性殺菌剤と非イオン性界面活性剤とを併用し
た口腔用組成物に対し、高級アルコールを配合す
ると、クロルヘキシジン等のカチオン性殺菌剤の
失活が効果的に防止され、上記の目的が達成され
ることを知見し、本発明をなすに至つたものであ
る。
以下、本発明につき詳しく説明する。
本発明に係る口腔用組成物は、カチオン性殺菌
剤と非イオン性界面活性剤と高級アルコールとを
含有してなるもので、練歯磨、潤製歯磨等の歯磨
類、マウスウオツシユ、歯肉ヤツサージクリー

また、非イオン性界面活性剤としては、シヨ
糖、マルトース、マルチトール、ラクチトールか
ら選ばれる糖又は糖アルコールの高級脂肪酸エス
テル、特にそれぞれ脂肪酸残基の炭素数が12〜18
で平均エステル化度が1.1〜2.5、より好ましくは
1.2〜1.9のシヨ糖脂肪酸エステル、ラクトース脂
肪酸エステル、ラクチトール脂肪酸エステル、マ
ルトース脂肪酸エステル、マルチトール脂肪酸エ
ステルが好適に用いられ、更にマルトトリイトー
ル脂肪酸エステル、マルトテトライトール脂肪酸
エステル、マルトペンタイトール脂肪酸エステ
ル、マルトヘキサイトール脂肪酸エステル、マル
トヘプタイトール脂肪酸エステルなども好適に使
用される。なお、カチオン性殺菌剤の配合量は組
成物全体の0.001〜0.5%(重量%、以下同じ)、
非イオン性界面活性剤の配合量は組成物全体の
0.01〜5%、特に0.05〜3%とすることが好まし
い。
本発明は上述したカチオン性殺菌剤と非イオン
性界面活性剤とを含む系に高級アルコールを配合
するもので、これによりカチオン性殺菌剤の非イ
オン性界面活性剤による失活を可及的に防止し得
たものである。
この場合、高級アルコールとしては、炭素数10
〜20の脂肪酸アルコール(飽和でも不飽和でもよ
く、直鎖でも分枝鎖でもよく、更にアルコールは
一価でも多価でもよい)、特にラウリルアルコー
ル、ミリスチルアルコール、セチルアルコール、
ステアリルアルコールが好適に使用される。これ
ら高級アルコールの配合量は、組成物全体の0.1
〜10%とすることが好ましい。
クロライドの口腔用組成物には、上述した成分
に加えて更にその目的、組成物の種類等に応じた
適宜な成分を配合することができる。
例えば、歯磨類の場合には、第2リン酸カルシ
ウム・2水和物及び無水物、第1リン酸カルシウ
ム、第3リン酸カルシウム、炭酸カルシウム、ピ
ロリン酸カルシウム、水酸化アルミニウム、アル
ミナ、無水ケイ酸、シリカゲル、ケイ酸アルミニ
ウム、不溶性メタリン酸ナトリウム、第3リン酸
マグネシウム、炭酸マグネシウム、硫酸カルシウ
ム、ポリメタクリル酸メチル、ベントナイト、ケ
イ酸ジルコニウム、合成樹脂等の1種又は2種以
上を配合し得る(配合量通常20%〜90%、練歯磨
の場合には20%〜60%)。
また、練歯磨等のペースト状組成物の場合には
粘結剤としてカラゲナン、カルボキシメチルセル
ロースナトリウム、メチルセルロース、ヒドロキ
シエチルセルロース、カルボキシメチルヒドロキ
シエチルセルロースナトリウムなどのセルロース
誘導体、アルギン酸ナトリウムなどのアルカリ金
属アルギネート、アルギン酸プロピレングリコー
ルエステル、キサンタンガム、トラガカントガ
ム、カラヤガム、アラビヤガムなどのガム類、ポ
リビニルアルコール、ポリアクリル酸ナトリウ
ム、カルボキシビニルポリマー、ポリビニルピロ
リドンなどの合成粘結剤、シリカゲル、アルミニ
ウムシリカゲル、ビーガム、ラポナイトなどの無
機粘結剤等の一種又は2種以上が配合され得る
(配合量通常0.3〜5%)。
更に、歯磨類等のペースト状や液状口腔用組成
物の製造において、粘稠剤としてソルビツト、グ
リセリン、エチレングリコール、プロピレングリ
コール、1,3−ブチレングリコール、ポリエチ
レングリコール、ポリプロピレングリコール、キ
シリツト、マルチツト、ラクチツト等の1種又は
2種以上を配合し得る(配合量通常10〜70%)。
本発明の口腔用組成物には、更にメントール、
カルボン、アネトール、オイゲノール、サリチル
酸メチル、リモネン、オシメン、シトロネロー
ル、α−テルピネオール、メチルアセテート、シ
トロネリルアセテート、メチルオイゲノール、シ
ネオール、リナロール、エチルリナロール、ワニ
リン、チモール、スペアミント油、ペパーミント
油、レモン油、オレンジ油、セージ油、ローズマ
リー油、桂皮油、ピメント油、桂葉油、シソ油、
冬緑油、丁子油、ユーカリ油特の香料を単独で又
は組合せて全体の0〜10%、好ましくは0.5〜5
%程度配合し得るほか、サツカリンナトリウム、
ステピオサイド、ネオヘスペリジルジヒドロカル
コン、グリチルリチン、ペリラルチン、タウマチ
ン、アスパラルフエニルアラニンメチルエステ
ル、p−メトキシシンナミツクアルデヒドなどの
甘味剤(0〜1%、好ましくは0.01〜0.5%)等
を配合し得る。
なお、本発明においては、有効成分として、前
記カチオン性殺菌剤加えて、デキストラナーゼ、
アミラーゼ、プロテアーゼ、ムタナーゼ、リゾチ
ーム、溶菌酵素(リデツクエンザイム)等の酵
素、モノフルオロリン酸ナトリウム、モノフルオ
ロリン酸カリウムなどのアルカリ金属モノフルオ
ロホスフエート、フツ化ナトリウム、フツ化第1
錫等のフツ化物、トラネキサム酸やイプシロンア
ミノカプロン酸、アルミニウムクロルドヒドロキ
シアラントイン、ジヒドロコレステロール、グリ
チルレチン塩類、グリチルレチン酸、グリセロホ
スフエート、クロロフイル、塩化ナトリウム、カ
ロペプタイド、水溶性無機リン酸化合物等の有効
成分を1種又は2種以上配合し得る。
次に、実験例を示して本発明の効果を具体的に
説明する。
〔実験例〕
塩酸クロルヘキシジン0.1%、界面活性剤1.0%
及び添加剤の所定量を溶解した水溶液をPH7.0の
リン酸緩衝液で2倍(第1図の場合)又は5倍
(第2図の場合)に希釈して試料液を調製し、こ
の試料液に大腸菌浮遊液の一定量を加え、菌を加
えてから35℃の温浴中で1分間殺菌反応を行なわ
せた。その後、ポリオキシエチレンソルビタンモ
ノオレエート1%とレシチン0.2%とを含む希釈
液(生理食塩水)で希釈し、その一定量をポリオ
キシエチレンソルビタンモノオレエートとレシチ
ンとを含むSCDLP(大五栄養化学(株)寒天平板上で
37℃で一晩培養し、生存菌数を求めた。
結果を第1図及び第2図に示す。これらの結果
において、塩酸クロルヘキシジン(図中CHXで
示す)、界面活性剤及び添加剤の濃度はいずれも
試料液中の濃度として示してある。また、塩酸ク
ロルヘキシジン0〜0.05%を単独で大腸菌浮遊液
に作用させた場合の生存菌数をCHX活性として
菌数との対比において示した。従つて、各試料液
の結果より各試料液中の塩酸クロルヘキシジンの
残存活性が明らかになる。
なお、界面活性剤としては、構成脂肪酸がパル
ミチン酸、ステアリン酸約70:30であり、モノエ
ステル75%、ジエステル以上25%のシヨ糖脂肪酸
エステル(菱糖株式会社製P−1670)及びラウリ
ル硫酸ナトリウム(SDS)をそれぞれ使用した。
