Description
産業上の利用分野
本発明は抗真菌外用ゲル剤に関するものであ
る。
従来技術
今日、トルナフテート及びウンデシレン酸が外
皮用抗真菌剤として繁用されている。
トルナフテート、すなわちN−メチル−N−
(3−トリル)チオカルバミン酸−2−ナフチル
エステルは抗真菌作用とくに白癬菌に対しすぐれ
た作用を示す。
また、ウンデシレン酸は人間の汗の中に含ま
れ、生理的に生じる脂肪酸であり、人体に対する
作用は温和であるが、白癬菌に対しては、10-4μ
g/ml、カンジダ症には10-3μg/ml程度の濃度
で抑制作用を示し、いずれも水虫、いんきんたむ
し、ぜにたむしの治療薬として用いられている。
一般に、ゲル剤は透明でゼリー状の外用剤であ
り、主薬の皮膚への浸透性が大であり、塗布した
場合べとつきが少ないという特徴を有している。
しかし、主薬を溶解させなければならないため、
その溶媒の選択がむづかしくなる。
トルナフテートは各種極性溶媒に対して難溶性
であり、溶解可能な溶媒は限定され、その含有製
剤はマクロゴール軟膏、乳剤性軟膏、液剤が主で
あり、一部に外用ゲル剤がある。
一方、ウンデシレン酸は溶媒に対する溶解性に
は問題がないが、これをゲル基剤に配合するとゲ
ルを破壊し粘度が低下する。したがつて現在ウン
デシレン酸含有の抗真菌外用ゲル剤は市販されて
いない。
トルナフテートを溶解させる溶媒としてはメチ
ルエチルケトンがあり、またマクロゴール軟膏に
用いられているポリエチレングリコール類が公知
である。
たとえば、特開昭55−45662号公報にはトルナ
フテート溶解型外用ゲル剤が開示されている。す
なわち該公報に於てはメチルエチルケトンとポリ
エチレングリコールにトルナフテートを溶解させ
ゲル化剤を加え、さらにトルナフテートが析出し
ない程度に水、アルコールを加えている。しか
し、ポリエチレングリコールを配合しているた
め、塗布面にべたつきを生じて使用感がわるく、
さらにトルナフテートを皮膚に浸透させる溶媒と
してポリエチレングリコールはすぐれたものとは
いえず、ゲル基剤としての特徴を充分に生かした
ものではない。
発明の構成
本発明者らは、トルナフテート、ウンデシレン
酸の皮膚浸透性を重視し、外用ゲル剤について鋭
意研究を行ない、数多くの知見を得て本発明によ
る抗真菌外用ゲル剤を完成させたものである。
本発明に係わる抗真菌外用ゲル剤はトルナフテ
ート及び/又はウンデシレン酸と、プロピレング
リコール及び又は1,3−ブチレングリコールを
含有する外用ゲル剤であつて、低級一価アルコー
ルを含有しないことを特徴とするものであり、好
ましくは該剤に対して1〜4重量%のトルナフテ
ート及び/又は3〜10重量%のウンデシレン酸が
含有されたものである。
本発明による外用ゲル剤に配合するグリコール
類としては、プロピレングリコール、1,3−ブ
チレングリコールあるいはそれらの混合物であ
り、その配合量は該剤に対して3〜60重量%、好
ましくは10〜45重量%である。
本発明による外用ゲル剤はケトン類、水、カル
ボキシビニルポリマー及び中和剤を配合すること
もできる。ケトン類としては、好ましくはメチル
エチルケトン、アセトンあるいはそれらの混合物
が好ましく、その配合量は該剤に対して10〜60重
量%、好ましくは15〜55重量%である。
本発明に於て水の含有量は1〜35重量%、好ま
しくは5〜30重量%である。
また、本発明による抗真菌外用ゲル剤に配合す
るカルボキシビニルポリマーは酸性高分子化合物
で、主としてアクリル酸重合体であり、市販品と
してはカーボポール(B.F.Goodrich Chemi cal
Co.製)あるいはハイビスワコー(和光純薬工業
株式会社製)等が使用される。カルボキシビニル
ポリマーの配合量は該剤に対して0.3〜5重量%、
好ましくは0.5〜2.5重量%である。
通常、カルボキシビニルポリマーは、必要に応
じて塩基性物質で中和して適当なゲルを得る。こ
の塩基性物質としてはアミン類、たとえばモノエ
チルアミン、ジエチルアミン、トリエチルアミ
ン、モノエタノールアミン、ジエタノールアミ
ン、トリエタノールアミン、モノイソプロパノー
ルアミン、ジイソプロパノールアミン、トリイソ
プロパノールアミン、トリブチルアミン、トリオ
クチルアミン等;アンモニア;アルカリ金属塩、
たとえば水酸化ナトリウム、水酸化カリウム、ホ
ウ酸ナトリウム、炭酸カリウム等が挙げられ、こ
れらのうちアミン類がとくに好ましく使用され
る。中和剤は外用ゲル剤のPHが4〜10になるよう
に使用される。
実施例
以下に実施例により本発明を具体的に詳述す
る。
実施例 1
トルナフテートを使用した第1表に示す如き抗
真菌外用ゲル剤をヘアレスマウスの背部皮膚に塗
布しトルナフテート透過量測定を行つた。
結果は第1表に示す通りであつた。

実施例 2

真菌外用ゲル剤のウンデシレン酸透過量を実施例
1と同様にして測定した。

ート及びウンデシレン酸の透過量を実施例1と同
様にして測定した。結果は第3表に示す通りであ

を40.0g配合したもののトルナフテート透過量が
それぞれ18.8、17.9、ng/πcm2/24hrであるの
に対し、第1表に示す如くトルナフテート単独使
用の場合はトルナフテー2.0gにプロピレングリ
コールあるいは1,3−ブチレングリコールを
45.0g配合したもののトルナフテートの透過量が
それぞれ16.8、17.3ng/πcm2/24hrであり、ト
ルナフテート及びグリコール類の配合量がいずれ
も少ないにもかかわらず、併用の場合の方が単独
使用よりその透過量に於てすぐれ、この傾向はウ
ンデシレン酸に於ても全く同様であつた。
併用配合の場合もプロピレングリコール、1,
3−ブチレングリコールを使用するととくにその
透過量に於てすぐれていた。
実施例 4
トルナフテート及びウンデシレン酸を併用する
本発明による下記配合の抗真菌外用ゲル剤のトル
ナフテート及びウンデシレン酸の透過量を実施例
1と同様にして測定した。
結果は第4表に示す通りであつた。

トルナフテート及びウンデシレン酸を併用する
本発明による下記配合の外用ゲル剤のトルナフテ

様にして測定した。結果は第4表に示す通りであ

同様にして測定した。結果は第4表に示す通りで
あつた。

ウンデシレン酸単独配合の本発明による下記外
用ゲル剤のウンデシレン酸の透過量を実施例1と
同様にして測定した。結果は第4表に示す通りで
あつた。

第4表の結果より本発明によるプロピレングリ
コールあるいは1,3−ブチレングリコール配合
の抗真菌外用ゲル剤のトルナフテート及びウンデ
シレン酸はポリエチレングリコールを配合した市
販品すなわち比較例A及びBに比し、その透過性
に於てすぐれていることが明らかであつた。
本発明による抗真菌外用ゲル剤には、実施例に
示す如く必要に応じてジフエンヒドラミン等の鎮
痒剤、リドカイン等の鎮痛剤、尿素等の角質溶解
剤、グリチルレチン酸等の抗炎症剤、エデト酸ナ
トリウム、抗酸化剤等の安定化剤、ジエチルセバ
ケート、アジピン酸ジイソプロピル等の浸透促進
剤、メチルセルロース等の増粘剤及び香料等を適
宜配合することができる。
発明の効果
本発明による抗真菌外用ゲル剤は、トルナフテ
ート、ウンデシレン酸の溶解性にすぐれ、低温下
長期保存に於ても主薬の析出がなく、皮膚中への
浸透性にとくにすぐれ、塗布した場合にべとつく
ことがなく、高い治療効果を発揮するものであ

て、プロピレングリコール及び/又は1,3−ブ
チレングリコールの含有量が外用ゲル剤に対し3
