【発明の詳細な説明】
発明の背景
を椎動物は2つの基本的免疫応答、即ち体液性および細胞性免疫応答を有する。
体液性免疫はBりンパ細胞によって産生される特別な組の細胞によってもたらさ
れる。これらの細胞は抗体を生産し、該抗体は血液およびリンパ液中を循環する
。他方、細胞媒介免疫はリンパ系のT細胞によってもたらされる。
この細胞性免疫応答は真菌、寄生虫、細胞内ウィルス感染、ガン細胞および異物
質に対して特に有効であり、一方で体液性応答は細菌並びにウィルス感染の細胞
外段階に対する防禦を行う。
侵入位置における抗原の拘束は局所的な炎症による領域の隔絶によって達成され
る。急性の炎症は血漿タンパクおよび多形核白血球の流入によって特徴付けられ
る。慢性炎症はT−リンパ細胞およびマクロファージの浸潤により特徴付けられ
る。急性(抗体誘起)および慢性(T−細胞誘起)の炎症が皮膚内で生ずる場合
、これらは夫々即時型過敏症反応(ITH)および遅延型過敏症反72時間でピ
ークに達する。DTH反応に関与するT細胞のサブセットはここではDTH−エ
フェクタ細胞と呼ぶ。
エビグリカニン(epi) (epiglycanin)は哺乳頻脈ガン移植可

入した。我々はガン−関連の十分に特徴付けされた炭水化物抗原を用いて、エフ
ェクタ細胞を感作し、これらの治療上の使用を教示した。
腫瘍はIL−2およびIL−2−活性化キラー細胞の組合せによって治療できる
ことが知られている。スチープンローゼンバーグ(Steven Rosenb
erg)の研究に関する報告Fortune、16−21(1985年11月2
5日)参照、このローゼンバーグの研究に係る問題はこの治療がLAK (リン
ホカイン活性化キラー)細胞として知られている細胞の集団を誘起することであ
る。LAK細胞は抗原−特異的ではないので、正常な組織が攻撃される可能性が
ある。また、LAK細胞は非特異性であるから、所定の抗−腫瘍活性を誘起する
ためには比較的大量のIL−2が必要である。
最後に、LAK細胞は免疫系に記憶を残すとは考えられない、逆に、DTHエフ
ェクタ細胞は抗原特異的であるから、感作後はより少量のリンホカインが増殖を
誘起するのに必要とされる。また、その特異性のために、DTHエフェクタ細胞
は自己を攻撃する恐れが少ない、更に、DTHエフェクタ細胞は記憶をもつと思
われる。
発明の概要
我々は、ガン細胞に対する細胞−媒介免疫応答が天然もしくは合成ガン−関連炭
水化物抗原の非経口投与により、あるいはこのような抗原により感作されたDT
)Iエフェクタ細胞の非経口投与によって増強されることを見出した。この増強
は、ある攻撃に対する身体の一次防禦が細胞免疫応答、例えば腫瘍に対する応答
である場合に治療並びに予防両面に対して有利である。多糖、糖脂質および糖タ
ンパクを含む他の特異的炭水化物抗原の使用は、他のT細胞によって制御される
脅威に対し対象を保護において有用であり得る。寄生虫、ウィルス、細菌および
真菌は表面炭水化物決定基をもつことが知られている。
リンホカインは特異的抗原または本発明の感作DTH−エフェクタ細胞と共に用
いて、特異的CMI応答を増強できる。

発明の詳細な説明
爽施■上 腫瘍に対する細胞免疫応答を増強するための5−TAGSの使用
合成腫瘍−関連塘タンパク(S−TAGS)および他の炭水化物抗原が当分野に
おいて知られており、任意の有利な方法で調製できる。

可溶性S−Tagsまたはその会合体を皮肉、筋肉内または腹腔内投与用のアジ
ュバントと混合する。静脈内投与も同様に有効であると考えられる。
ある時点で5匹のマウスからなる群をTα(TF) 、TF(アシアロ−GMI
ジサンカライド)およびTn S−Tagsの特定量で感作した。7〜10日後
、これらを3X10”個の生きたTA3Ha細胞で攻撃した。著しく延長された
生存率が約lμgの合成抗原を与えたマウスについて記録された。かくして、T
Fについては、以下の結果が見られた(第1表)。

であることに注目すべきである。マウスの正常(Norm)な後−移植平均余命
はわずかに15〜20日である。
! 腫瘍阻害剤としての培養DTHエフェクタ細胞の使以下の手順により、我々
はマウスの感作DTHエフェクタ細胞を得た。我kO)動物部門から(7) C
AFI/J (Balb x A/J) マウスを、50%完全フロインドアジ
ュバント中の30μgのEpi (または2μgの任意の合成抗原)で腹腔内経
路で免疫し、1週間後に同れら動物を殺し、その肺臓を取出し、単一細胞懸濁物
とするためにナイロンメツシュに通した。この細胞を洗浄し、1.5X10’細
胞/8mlにてコスターウェル(costar well)内で培養した((R
PMI十P、S、 + l Q%FC5) /ウェ/lz)、各ウェルには10
μgのepiを入れた(または任意の合成抗原1〜2μgを入れた)。
これら細胞を37℃、5%CO8にて6日間培養し、次いでゴムポリスマンでプ
ラスチックから細胞を穏かに分離することにより収穫した0次に、この感作細胞
を多数の抗原とのDTH反応性およびインビボでのTA3Haセルラインの生長
阻害能につきテストした。
ヒト感作DTHエフェクタ細胞を得るために、我々はローゼンバーグの方法を改
良して用いた。血漿分離交換法により、リンパ球を生きた対照の血液から取出し
た。このリンパ球を特異的抗原、例えばガンー関連娠タンパク(エビグリカニン
またはTもしくはTn S−丁AG)あるいはIL−2単独ではなく、むしろ特
異的抗原とIL−2との組合せなどと共に培養する。ヒトリンパ細胞を、好まし
い1度、即ち約10′細胞/mlにて適当な培地中で6日間培養する。この細胞
を洗浄して、抗原を除く0次いで、この細胞を患者に、静脈内経路で、治療また
は予防上の目的で戻すことができる。
我々は哺乳動物に、エピグリカニンー感作DTI(エフェクタ細胞と抗原(照射
腫瘍細胞)とを同時に注入することにより、DTH反応が誘起されることを立証
した(第1図)、30μgおよび20μgのTA3Ha腹水から抽出かつ精製し
たepiおよびCFA(予め1および2週間前に腹腔内投与された)により感作
されかつ二次免疫されたマウスからの肺細胞を、(81108g epi/ルミ
/ウェルおよび(′b)抗原なしの状態で、1.5X10’細胞/ウエルにて培
養した。更に、未感作牌細胞を、(C110μg epi/ルミ/ウェルおよび
ldl抗原のない状態で培養した。培養物を68目に収穫し、かつ10?個の細
胞を、106個の照射TA3Ha細胞と共に免疫されていないマウスの足の肉踵
に皮下投与した。コントロールtelとして、106照射TA3Ha細胞を単独
で注射した。すべてのDTH価を正味のDTH腫張、即ち〔感作細胞の腫張+A
g、 ) −〔感作細胞の腫張のみ〕として表す。大きな応答差は、ガンムチン
にする免疫が、細胞表面にムチンを有するガン細胞にDTH反応を誘起できるこ
とを立証した。
次に、この反応が腫瘍特異性であることを我々は立証した(第2図)。DTHエ
フェクタ細胞集団を実施例1のように発生させ、a) TA3Ha、 b) E
pi−M (セファ0−ス微小球に結合したepi)、C)EKSAにより、お
よびd)抗原なしの状態でテストした。すべてのDTH価を正味のDTH腫張、
即ち〔感作細胞の腫張十Ag、 ]−〔感作細胞の腫張のみ〕によって示す。結
果は、epiルミムチン免疫がムチンおよびムチンをもつ細胞(TA3Ha)に
対し特異的DTH応答を誘起したが、ムチンをもたないもの(mKsA)に対し
ては誘起しなかったことである。
我々は、またDTH反応の抗原特異性をより一層詳細に調べた(第3図)。10
7個のT細胞をepiで感作し、一連の抗原(TA3Ha、 Tn、 Ta、T
a、Epi 、フコース(“Fuc”: Fucose)、H3A、BSA、セ
ファロース微小球および抗原なし)と共に同時注入した。すべての可溶性抗原は
セファロース微小球に結合され、かツepiを除き(これは50μg/足肉!t
h)、12μg/足肉跡の量で用いた。すべてのDTHの結果は正味のDTH測
定による。この実験は、epiの注入により測定されたDTHがepi決定基と
しても知られるS−Tags上の炭水化物決定基と反応するがEpi上に表現さ
れていない炭水化物決定基(例えばフコース)と反応しないことを示す。
DTH応答の特異性の概要

Fuc BSA我々が目撃した反応が実際にDTH反応であったことを確認す
るために、我々はDTHエフェクタ細胞の細胞表面表現型をチェックするための
実験を行った(第4図)、エフェクタ細胞集団を前と同様に調製し、a)処理せ
ず、b)抗−Thyl、2抗体+補体、C)抗−Lytl、2+補体、d)抗−
t、ytz、z+補体およびe)補体のみ、で処理した。この抗原のみをコント
ロールとして用いた。
すべての値は正味のDTHで表されている。DTHエフェクタ細胞を抗−rhy
または補体を含む抗−Lyt −1抗体で処理すると、DTHエフェクタ細胞活
性がなくなるが、抗−Lyt −2+補体による処理はDTHエフェクタ細胞活
性を失わない、かくして、特異的抗−炭水化物DTHエフェクタ細胞の表現型は
rhy−1+、t、yt −1+、tyt−2−・であり、他のDTHエフェク
タ細胞により示される表現型パターンはタンパクと反応する。フジワラ(Fuj
iwara)、 J、 1μmuna1..135 : 2187 (1985
,9月)参照。
この比較のプロトコールは以下の通りであった。感作細胞集団を得、洗浄し、計
数し、処理用の別々の管に分割した。この細胞を穏かにベレント化し、ルイボヴ
イフッ(Leibowi tz)培地〔1)抗−Thyl、2抗体(1/100
0希釈率; NENから得た)、2)抗−Lytl、2抗体(1/1000希釈
率、NENから得た)または3)抗−tyt−2,2抗体(1/1000希釈率
、NENから得た)を含む〕に再懸濁し、4℃で45分間インキュベートした0
次に、この細胞を遠心沈降させ、ルイボヴインツ培地中のモルモット補体(1/
10希釈率)中に2X10’細胞/whlで再懸濁し、37℃水浴中で30分間
インキュベートした。インキュベーシッン後、細胞をルイボヴイッツ培地+10
%FC3で洗浄し、計数した。
比較としての研究を行って、この反応が遅延型であるか否かを決定した。DTH
エフェクタ細胞を丁^311aと共に注入し、この反応をDTHエフェクタ細胞
のみまたはTA3Haのみを注射した他のマウス内での反応と比較した。この反
応が24時間でピークに達し、96時間で消失することがわかった。2種のコン
トロールについてはDTH(遅延増強)は見られなかった。
観測された反応がDTHであることを更に確認したところ、単核浸潤物が腫張サ
イトに見られた。
上記実験はすべて局所的なりTHの伝達に関連した。我々は、またDTHエフェ
クタ細胞が全身的作用をもつ可能性をも調べた。
DTHエフェクタ細胞は宿主動物に静脈内投与された。4時間後照射TA3Ra
細胞を足内証に注射した。6.24.48および72時間後に、このDTH反応
を測定しく白抜きの四角)、負のコント白−ル(+)と比較した(第5図)。
上記実験で用いた照射腫瘍細胞は増殖しない、しかし、次の実験では未処理腫瘍
細胞を用いた。
DTH感作細胞を上記のように誘発した*2.5xlG’生腫瘍細胞十■を注入
した5X10’感作細胞を足肉証に皮下注射した。
コントロールマウスに、正常肺細胞+腫瘍細胞を注入した。該肉跋の腫張を典型
的には12.24.48および72時間並びにその後2日毎に測定した。DTH
と腫瘍生長の組合せに反して、DTH反応による腫張の成分を決定するために、
マウスに感作または正常細胞+照射腫瘍細胞を注入した。腫張はDTH反応のみ
に起因するものであった。腫瘍生長はピークDTHIl!張の48時間後を始点
として2日毎に測定した。DTHおよび腫瘍サイズをカリパスを用いて測定して
肉鉦厚さの増加を見積った。
第6図はこの感作細胞がTA3Ha腫瘍の成長を阻害したことを示している。(
白抜きの四角、=感作DTHエフェクタ細胞÷丁A3Ha ;+=正常細胞十T
^3Ha) 。
他の実験は、この反応の抗原に対する性質がDTHエフェクタ細胞に投与したプ
ライマーの投与量に依存することを示した。低投与量では、細胞媒介免疫が支配
的であるが、高投与量では逆が正しかった。
肺細胞が培養中にTnで感作された場合にも、投与量依存性が見られた(第7図
)、(ここで、立上り斜綿棒は正味のDTHを示し、下降斜線棒を標準偏差を示
す)。
我々は、更にepiで刺激されたDTHエフェクタ細胞が腫瘍上に“進み(ho
me in) ”かつ腫瘍生長を阻害することを明らかにした。上記の如く、D
THエフェクタ細胞をepiで感作した。epi−感作DTHエフェクタ細胞を
静脈内注射する1日前に、TA3Ha腫瘍をマウスの足の内証に移植した。同時
に腫瘍を移植したマウスをコントロールとして用い、腫瘍生長はカリパスで該内
証の腫張を測定することにより決定した。

うに、■−投与、epi−感作DTHエフェクタ細胞は腫瘍移植した内証サイト
において腫瘍阻害作用を及ぼした。
ffi 腫瘍阻害剤としてのエピグリカニンの使用エビグリカニン(epi)は
TA3Ra腫瘍にかかったマウスの腹水か゛ら抽出した。
異系交配TA3Ha ip 11瘍からの腹水を集め、細胞を遠心により取出し
た。サンプルを凍結保存した。抽出前に、この腹水を解凍し、37℃で2時間イ
ンキュベートし、凝集物をすべて遠心により除いた。凍結、解凍および凝固操作
をPNA抽出前に2度行った。
25〜40−1の床体積をもつPNAアガロース(E、Y、ラボラトリーズ(L
aboratories) )をPBSで洗い、腹水と混合し、4℃にて24〜
48時間穏かに混転した。このスラリーをカラムに注ぎ、室温にてPBSで徹底
的に洗浄した。ガラクトースを用いてEpiをPNAアガロースから溶出した。
次に、溶出したepi−ガラクトース混合物を水に対して透析してガラクトース
を除いた。このサンプルを凍結乾燥し、デシケータ中で一20℃にて保存した。
同時に、5匹のマウスからなる群を特定量のepiで感作した。
2〜3日後、これらを3X10’生TA3Ha細胞により攻撃した。
以下の第2表は感作投与量と生存率との関係を示す。

たデータは、これらマウスが依然として生存していることを示す。これらマウス
を殺しく88日目)で、実験を続けた。
かくして、好ましい感作投与量は1μgであり、かつこの投与量はTA3Ha細
胞に対し防禦免疫性を付与すると思われた。
もう一つの実験において、この投与量における液性応答は無視でき(第8図)、
このことはこの腫瘍が抗体以外の機序、恐らくCMIによって阻害されることを
示唆するものと理解された。マウス(CAFI/J)に、i、p、経路で、50
%CFA中の指定したepiO量を注射した。10日後に、これらを照射TA3
Ha細胞で攻撃して、DTH反応性を調べた(黒塗りの四角)、epiに対する
抗体(白抜きの丸)をELISAで決定した。
腫瘍阻害におけるEpiおよびTαによる我々の経験をまとめて以下の第3表に
与える。

Claims (30)
- 1.特異的抗原担持炭水化物決定基を感作量で含む媒質中でDTHエフェクタ細 胞を培養する工程を含む、該DTHエフェクタ細胞の免疫活性を増強するための 該細胞の感作法。
- 2.該決定基が腫瘍関連決定基である請求の範囲第1項記載の方法。
- 3.該抗原がムチンである請求の範囲第1項記載の方法。
- 4.該抗原がエピグリカニンである請求の範囲第3項記載の方法。
- 5.該抗原が糖タンパクである請求の範囲第1項記載の方法。
- 6.該抗原が糖脂質である請求の範囲第1項記載の方法。
- 7.該抗原がTα決定基を担持する請求の範囲第1項記載の方法。
- 8.該抗原がTβ決定基を担持する請求の範囲第1項記載の方法。
- 9.該抗原がTn決定基を担持する請求の範囲第1項記載の方法。
- 10.該抗原が免疫担体と複合化された合成炭水化物ハプテンを含む請求の範囲 第1項記載の方法。

- 21.請求の範囲第19または20項に記載の治療組成物を投与することを特徴 とする哺乳動物中の細胞免疫を増強する方法。
- 22.製薬上許容される担体中に、有効量で特異的抗原担持炭水化物決定基を含 むことを特徴とする哺乳動物の細胞免疫を増強するための組成物。
- 23.該決定基が腫瘍関連決定基である請求の範囲第22項記載の組成物。
- 24.該抗原がムチンである請求の範囲第22項記載の組成物。
- 25.該ムチンがエピグリカリンである請求の範囲第24項記載の組成物。
