本発明が、その好ましい実施形態のうちのいくつかについて詳細に説明される前に、以下の一般的定義を提供する。
TCRは、2つの異なる、別々のタンパク質鎖、すなわちTCRアルファ(α)鎖およびTCRベータ(β)鎖から構成される。TCRα鎖は、可変(V)、連結(J)および定常(C)領域を含む。TCRβ鎖は、可変(V)、多様性(D)、連結(J)および定常(C)領域を含む。TCRα鎖とTCRβ鎖の両方の再配列V(D)J領域は、超可変領域(CDR、相補性決定領域)を含有し、これらの中で、CDR3領域は、特異的エピトープ認識を決定する。C末端領域において、TCRα鎖とTCRβ鎖の両方は、疎水性膜貫通ドメインを含有し、短い細胞質側末端で終わる。
本明細書に記載のTCRに対する標的は、MAGEA1(NCBI参照配列:NM_004988.4)由来のペプチドKVL(配列番号1)である。
一部の実施形態は、TCRα鎖およびTCRβ鎖を含む単離されたTCRに関し、ここで、
- TCRα鎖は、配列番号4の配列を有する相補性決定領域3(CDR3)を含み、
- TCRβ鎖は、配列番号7のアミノ酸配列を有するCDR3を含む。

本発明の別の態様は、本明細書に記載されているTCRの機能性部分を含むポリペプチドであって、機能性部分が、配列番号4および7のアミノ酸配列のうちの少なくとも1つを含む、ポリペプチドを指す。
一部の実施形態は、本明細書に記載されている単離されたTCR、本明細書に記載されているポリペプチド、本明細書に記載されている多価TCR複合体を指し、ここで、IFN-γの分泌は、エフェクター細胞上に発現された本発明のTCRの配列番号1からなる群から選択されるアミノ酸配列のHLA-A*02結合形態への結合によって誘導される。
本発明の別の態様は、本明細書に記載されているTCRをコードする核酸または本明細書に記載されているTCRをコードするポリヌクレオチドをコードする核酸に関する。
本発明の別の態様は、本明細書に記載されているTCRを発現する細胞を指す。

本発明の別の態様は、MAGEA1に対する特異性を媒介する、本明細書に記載されているTCRの一部に特異的に結合する抗体またはその抗原結合断片を指す。一実施形態では、MAGEA1に対する特異性を媒介するTCRの一部は、配列番号4、および/または配列番号7のベータ鎖のCDR3を含む。
本発明の別の態様は、本明細書に記載されているTCRを含む医薬組成物、前記TCRの機能性部分を含むポリペプチド、本明細書に記載されている多価TCR複合体、TCRをコードする核酸、前記核酸を含むベクター、前記TCRを含む細胞、または本明細書に記載されているTCRの一部に特異的に結合する抗体を指す。
1.実験:MAGEA1特異的TCRの単離
MAGEA1由来のエピトープに対する特異性および目的のいずれかのMHC分子に対する拘束性を有するT細胞を単離するために、in vitroプライミングアプローチを使用した。プライミング系は、抗原提示細胞としてHLA-A*02:01陰性ドナーの成熟樹状細胞(mDC)を使用し、抗原に特異的なT細胞応答および自己のCD8+T細胞の拡大を開始する。ミニ遺伝子(配列番号27)をコードし、MAGEA1由来のペプチドKVLEYVIKV(配列番号1において参照される)を含有するアミノ酸配列へと翻訳されるin vitroで転写されたRNA(ivtRNA)は、特異的抗原の供給源としての役割を果たす。同時に、ヒトのHLA-A*02:01をコードするivtRNAを、mDCにトランスフェクトされる拘束エレメントの供給源として使用し、この専用のHLAアレルについてアロジェニックプライミングを設定した(国際公開第2007/017201号パンフレットに記載されている)。mDCへの電気穿孔後に、ミニ遺伝子をコードするivtRNAは、タンパク質へと翻訳され、次に、プロセシングされ、同時にトランスフェクトされたmDCによって発現されるトランスジェニックHLA-A*02:01分子によってペプチドとして提示される。同じドナーに由来するT細胞のivtRNAがトランスフェクトされたmDCとのin vitro共培養によって、対応するTCRの供給源としての役割を果たす抗原特異的T細胞のde novo誘導がもたらされる。拡大後に、抗原特異的T細胞を濃縮し、希釈を制限するかまたはFACSに基づく単一細胞選別によって単一細胞をクローニングすることができる。
高親和性TCRの単離および同定のためのT細胞の樹状細胞によるプライミングは、以下のプロトコールに従ったアロジェニックHLA-A*02:01分子によるペプチド提示を使用して達成した:
記載したin vitroプライミングアプローチによって、候補TCR T15.8-4.3-83を発現するT細胞クローンが同定された。NGS分析を使用してTCR配列を特定し、再構築して、機能性および安全性についてのTCRの完全な特徴付けのために、好適なエフェクターT細胞において遺伝子導入により発現させた。
遺伝子導入により発現した、組換えTCRの機能性を確認するために、TCRを形質導入したエフェクターT細胞をペプチドを負荷したT2細胞(標的細胞)と共培養した。特異的ペプチド:MHC複合体のTCR媒介性認識により、TCRを発現するT細胞の活性化およびある種のサイトカイン、例えば、IFN-γの分泌がもたらされる。共培養上清において分泌されたIFN-γを分析するために、ELISAを実施した。
HLA-A*02を発現するT2細胞に飽和量のペプチド(10-5M)を負荷した。MAGEA1由来のKVLペプチド(配列番号1)を負荷したT2細胞は陽性標的としての役割を果たし、無関係のASTN1ペプチド(配列番号28)を負荷したT2細胞は陰性標的としての役割を果たした。エフェクター細胞として、2名の異なるドナーに由来するCD8濃縮T細胞集団に形質導入して、組換えTCR T15.8-4.3-83を安定して発現させた。エフェクターは、TCR T15.8-4.3-83の2つの異なるバージョンを発現するように調製し、一つのバージョンはマウスC領域(murC)を有し、一つのバージョンはヒトの最小限にマウス化されたC領域(mmC)を有した。対照のMAGEA1-TCR(ベンチマークTCR)も2名の異なるドナーに由来するT細胞において発現し、分析された。
TCRを発現するエフェクター細胞は、KVLペプチドを負荷したT2細胞と共に培養した際にのみIFN-γを分泌し、トランスジェニックTCR T15.8-4.3-83によって媒介される、提示されたペプチド:MHC複合体の特異的認識が示され、その機能性が証明された。陰性標的である、無関係のASTN1ペプチド(配列番号28、KLYGLDWAEL)を負荷したT2細胞は認識されなかった(図1)。トランスジェニックTCRは、TCRのC領域とは無関係に、特異的標的認識およびIFN-γの分泌を媒介する。
TCRは、ある種のMHC分子と組み合わせた場合にのみそれらの特異的エピトープを認識するため、抗原の発現レベルだけでなく、標的細胞のHLA型も(in vitroおよびin vivoで)標的を陽性または陰性の標的であると規定する。言い換えれば、HLA型に依存して、TCRベースのACTに関する処置レジメンに患者を含めることも含めないこともある。
53種のLCL細胞株に、飽和量のMAGEA1由来のKVLペプチド(10-5M;配列番号1)を約1.5時間負荷し、洗浄し、次に、異なるエフェクター調製物に由来するTCR T15.8-4.3-83を発現するエフェクターと共培養した。負荷されていないLCLを対照として使用した。共培養は、96ウェル丸底プレートにおいて1:1のE:Tで設定し、約20時間後に上清を回収し、ELISA(標準的なサンドイッチELISA、BDヒトIFN-γ ELISAセット)によってIFN-γのレベルを分析した。
TCR T15.8-4.3-83の拘束分析は、HLA-A*02:01に提示される場合だけでなく、様々な程度で、HLA-A*02:04、HLA-A*02:16、およびHLA-A*02:17に提示される、そのMAGEA1由来のエピトープをも認識するTCRの能力を示す。図2は、負荷されていないLCLに関して検出されたバックグラウンドレベルを超えるペプチド特異的認識をもたらすこれらのLCL(試験した53個のうち)に関するデータのみを示す。
T細胞受容体の有効性、特異性および臨床適用に対するその適合性を分析するために、腫瘍細胞株のセットを、TCRを形質導入したエフェクター細胞と共培養した際のIFN-γ分泌を測定することによって、TCR媒介性認識について試験した。
3名の異なるドナーに由来するエフェクター細胞にTCR T15.8-4.3-83およびベンチマークCtrl.MAGEA1-TCRを形質導入した。TCRを形質導入したかまたは形質導入されていないエフェクターを、様々な腫瘍細胞株と共培養した。細胞株U266、NCI-H1703、UACC-62、Saos-2およびMel624.38(HLA-A2+/MAGEA1+)およびHLA-A2をトランスフェクトした細胞株KYO-1およびOPM-2(HLA-A2+/MAGEA1+)は、陽性標的としての役割を果たした。細胞株NCI-H1755、647-VおよびCMK(HLA-A2+/MAGEA1-)、未改変のOPM-2およびKYO-1(HLA-A2-/MAGEA1+)は、陰性標的としての役割を果たした。細胞株のHLA-A2発現をFACSによって分析し(データは示さず)、qPCRを使用してMAGEA1発現データを作成し、当業者に公知の公共に利用可能なデータベースから抽出した。対照として、すべてのエフェクターも、飽和濃度(10-5M)のMAGEA1由来のKVLペプチド(配列番号1)または無関係のASTN1ペプチド(配列番号28)を負荷したT2細胞と共培養した。共培養のために、エフェクターと標的を2.5:1のE:Tで、96ウェル丸底プレートに播種し、約20時間の共培養後に上清を回収し、ELISAによってIFN-γのレベルを分析した。
腫瘍細胞を効果的に認識するTCRの能力を、T15.8-4.3-83を形質導入したエフェクターを様々な腫瘍細胞株のセットと共培養することによって評価した。図3に示されるように、T15.8-4.3-83とCtrl MAGA1-TCRの両方が、HLA-A2陽性およびMAGA1陽性の腫瘍細胞株U266、NCi-H1703、UACC-62、Saos-2およびMel624.38の特異的認識を媒介した。KYO-1およびOPM-2細胞は、HLA-A2をコードするivtRNAによるトランスフェクション後にのみ認識された。
20.000個のTCRトランスジェニックエフェクターT細胞を、96ウェル丸底プレートにおいて、20.000個の腫瘍細胞と共培養した。メラノーマ細胞株UACC-257、メラノーマ細胞株UACC-62または骨肉腫細胞株SAOS-2のいずれかとの20時間の共培養後に、標準的IFN-γ ELISAを実施する。4000pgを超える値は、三次多項式を使用して推定する。
MAGEA1およびHLA-A*02:01二重陽性腫瘍細胞株に応答してIFN-γを産生するTCRトランスジェニックエフェクターT細胞集団の能力を評価した。T15.8-4.3-83は、国際公開第2018/170338号パンフレットに開示されたTCR FH1、FH2、FH3およびFH4ならびに国際公開第2018/104438号パンフレットに開示されたR37P1C9と比較して、3種の細胞株すべて、UACC-257、UACC-62またはSAOS-2に対して増強された腫瘍細胞認識能を示す。UACC-62は、T15.8-4.3-83にのみ認識される。
腫瘍細胞を認識するだけでなく効率的に溶解し、これにより、腫瘍細胞を死滅させるTCRの能力は、最も重要であり、臨床的に適切である。TCR T15.8-4.3-83の殺滅能を分析するために、IncuCyte(商標) Zoomデバイスを使用して、6日にわたって長期死滅アッセイを実施した。IncuCyte(商標)デバイスは、細胞のライブイメージングを可能にする顕微鏡に基づくシステムである。腫瘍細胞株のセットを96ウェル平底プレートのウェル中に播種し、3名の異なるドナーに由来するTCRを発現するエフェクターと共培養した。腫瘍細胞株は、核に拘束される赤色蛍光タンパク質mKate2を安定して発現し、IncuCyte(商標) Zoomシステムが、各ウェルにおける赤色蛍光細胞の正確な数を決定することを可能にした。経時的にウェル当たりの増加した細胞数が腫瘍細胞の成長を示し、一方、ウェル当たりの細胞数の低下はTCR媒介性死滅を示すことになる。
腫瘍細胞株647-V(膀胱尿路上皮癌)、UACC-62(メラノーマ)、Saos-2(骨の骨肉腫)およびNCI-H1703(肺腺癌)に、製造業者の使用説明書に従って、IncuCyte(商標) NucLight(商標) Red Lentivirus Reagentを使用することによって、核に拘束される赤色蛍光タンパク質mKate2を安定して形質導入した。腫瘍細胞を96ウェル平底プレートに播種し、一晩、プラスチックに付着させた。陽性対照として、腫瘍細胞に、飽和濃度のMAGEA1由来のKVLペプチド(10-5M)を約1.5時間負荷させ、洗浄し、次いで、共培養に使用した。KVLを負荷したか、または未改変の腫瘍細胞を、単独で培養したか、または3名の異なるドナーに由来するTCR T15.8-4.3-83を形質導入したエフェクターと共培養した。形質導入されていないエフェクターとの共培養は、陰性対照としての役割を果たした。2時間ごとに、IncuCyte(商標) Zoomシステムを用いて写真を撮り、IncuCyte(商標) Zoomソフトウェア(バージョン2016B)を用いて分析した。
図4A~4Dは、それぞれ、赤色蛍光標的細胞株647-V(HLA-A2+/MAGEA1-)、UACC-62(HLA-A2+/MAGEA1+)、Saos-2(HLA-A2+/MAGEA1+)およびNCI-H1703(HLA-A2+/MAGEA1+)のTCR媒介性の、抗原特異的溶解を示す。形質導入されていないエフェクター細胞は、腫瘍細胞のいずれの成長(経時的な細胞数の増加)にも影響を及ぼさないが、TCR T15.8-4.3-83を形質導入したエフェクター細胞は、抗原MAGEA1およびHLA-A2を発現する腫瘍細胞NCI-H1703、UACC-62およびSaos-2を効率的に死滅させる(経時的な細胞数の減少)。MAGEA1陰性の647-V細胞は死滅しない。すべての腫瘍細胞は、対照としてのKVLペプチドを人工的に負荷した場合に死滅する。参照として、KVLペプチドを負荷していないかまたは負荷した、単独で培養した腫瘍細胞も分析し、各グラフに成長を示す。
20.000個のTCRトランスジェニックエフェクターT細胞を7.500個のIncuCyte(登録商標) NucLight Red Lentivirusを形質導入したSAOS-2細胞と共培養する。共培養開始の1日前に、腫瘍細胞を播種した。エフェクター細胞を添加した後、培養プレートをIncuCyte ZOOM(登録商標)デバイスに移し、37℃および6%のCO2で、72時間にわたり、赤色蛍光細胞の拡大をモニターし、4時間ごとに写真を撮る。各測定点におけるウェル当たりの赤色蛍光腫瘍細胞の細胞数(l/mm2)を、IncuCyte ZOOM(登録商標)ソフトウェアを使用して計算する。各測定点は、3つの技術的反復の平均を表す。
形質導入されていないT細胞の存在下で培養したSAOS-2細胞は、強力な増殖を示す。異なるTCRトランスジェニックT細胞の共培養は、SAOS-2細胞数の減少を誘導する。興味深いことに、死滅の速度および程度は、異なるTCRトランスジェニックエフェクター細胞間で変化する。TCR T15.8-4.3-83は、国際公開第2018/170338号パンフレットに開示されたTCR FH1、FH2、FH3およびFH4ならびに国際公開第2018/104438号パンフレットに開示されたR37P1C9と比較して、MAGEA1およびHLA-A*02:01二重陽性腫瘍細胞株を溶解する最も高い能力を示す。
TCRの特異的エピトープ認識モチーフの詳細な分析のために、トレオニン置換スキャンおよび/またはセリンスキャンを実施した。エピトープの元のアミノ酸をトレオニン(それぞれ、またはセリン)で置換することによって、TCR媒介性認識に必須であるエピトープ内の位置を特定することができる。この認識は、ペプチドのHLA分子への適切な結合およびペプチド:MHC複合体のTCR自体との相互作用に依存し、いずれも、1つの単一アミノ酸の置換によって影響を受け得る。
TCR T15.8-4.3-8のエピトープ認識モチーフを特異的に規定するために、TCR T15.8-4.3-83を発現する3名の異なるドナー由来のエフェクターおよびベンチマークMAGEA1-TCRを発現するエフェクターを調製した。T細胞を、TCRの特異的エピトープKVL(配列番号1)を負荷したか、またはトレオニンで連続的に置換された(トレオニン置換スキャン)それぞれ個々のアミノ酸残基を有するペプチドを負荷したT2細胞と共培養した。T2細胞に、飽和濃度(10-5M)のペプチドを別々に負荷し、洗浄し、1:1のE:Tでエフェクターと共培養した。約20時間の共培養後に、上清を回収し、ELISAによって分泌されたIFN-γを分析した。
TCR T15.8-4.3-83を形質導入したか(図6に示される)またはベンチマークCtrl.MAGEA1-TCRを形質導入した(図5に示される)エフェクターに関するトレオニン置換スキャンの結果は、それらのエピトープおよびエピトープ由来の誘導体に対する個々のTCRの別個の特異性を実証する。参照として、MAGEA1由来のKVLエピトープの認識を、各グラフの左側に示す。ベンチマークCtrl.MAGE A1-TCRは、位置4および7のトレオニン置換を有するペプチドを認識しない(EおよびI、図5)。
20.000個のTCRトランスジェニックT細胞を、20.000個の10-5Mのペプチドを負荷したT2細胞と共培養する。20時間の共培養後に、標準的IFN-γ ELISAを実施する(4000pgを超える値は、三次多項式を使用して推定する)。上記のトレオニン残基の代わりに、セリン残基を使用して、MAGEA1KVLペプチドにおける個々のアミノ酸を系統的に置き換える(セリンスキャン)。
T15.8-4.3-83を形質導入したT細胞は、FH1またはR37P1C9 TCRを形質導入したT細胞とは異なる固定位置を有する異なる認識モチーフを示す(セリンスキャンによる)。
MAGEファミリーの13個のメンバーは、MAGEA1由来のKVLペプチドと非常に類似する(2~4個のアミノ酸ミスマッチのみ)ペプチド配列を含有し、潜在的なTCR T15.8-4.3-83媒介性の交差認識をさらに調査するために、インデプス分析を行った。MAGEファミリーのメンバーの発現は、がんおよび精巣においてだけでなく、生命維持に必要な器官においても記載されており、タンパク質由来のペプチドの潜在的な交差認識が、臨床開発のためのTCRに対する除外基準であるとする。
MAGEファミリーのメンバーMAGEA8(NCBI Reference Sequence(受託):NM_001166400.1)、MAGEA9(NM_005365.4)、MAGEA11(NM_005366.4)、MAGEB1(NM_002363.4)、MAGEB2(NM_002363.4)、MAGEB3(NM_002365.4)、MAGEB5(NM_001271752.1)、MAGEB16(NM_001099921.1)、MAGEB17(NM_001277307.1)、MAGEB18(NM_173699.3)、MAGEC2(NM_016249.3)、MAGED2(NM_014599.5)およびMAGEE2(NM_138703.4)をコードするivtRNAを生成した。MAGEA1(NM_004988.4)およびASTN1ペプチドKLYをコードするivtRNAを陽性および陰性対照RNAとして生成した。腫瘍細胞株HEK293T(HLA-A2+/MAGE-)、LCL(HLA-A2+/MAGE-)およびHLA-A2を形質導入したK562(MAGE-)に、13個のMAGEファミリーのメンバー、MAGEA1またはASTN1ペプチドKLYのいずれかをコードする20μgのivtRNAを別々にトランスフェクトした。MAGEファミリーのメンバーのトランスジェニック発現をFACSによって分析した(データは示さず)。対照として、T2細胞および3種の細胞株に、MAGEA1由来のKVLペプチド(10-5M)またはASTN1由来のKVLペプチド(10-5M)のいずれかをさらに負荷した。ペプチドを負荷した対照細胞およびトランスフェクトした細胞株をトランスフェクションの3時間後に、96ウェル丸底プレートに播種し、2名のドナーに由来する形質導入されていないかまたはTCR T15.8-4.3-83を形質導入したエフェクターを1:1のE:T比で共培養した。約20時間の共培養後に上清を回収し、ELISAによって分泌されたIFN-γを分析した。
図7は、HEK 293T細胞(A)、K562細胞(B)およびLCL細胞(C)において発現された組換えMAGEファミリーのメンバーのTCR T15.8-4.3-83媒介性認識を示す。T2細胞ならびにKVLペプチドまたはASTN1ペプチドを外部から負荷したHEK 293T、K562またはLCL細胞の認識は、対照としての役割を果たした。形質導入されていないエフェクターは、いずれの特異的認識も示さなかったが、2名のドナーに由来するTCR T15.8-4.3-83を形質導入したエフェクターは、完全長MAGEA1をコードするivtRNAをトランスフェクトした3つの細胞株すべてを特異的に認識した。これは、MAGEA1をコードするRNAのMAGEA1タンパク質への効率的な翻訳およびタンパク質由来KVLペプチドのプロテオソームプロセシングとその後の細胞表面におけるHLA-A2分子によるエピトープの負荷および提示を示す。ASTN1ペプチドをトランスフェクトした細胞株のいずれについても認識されず、バックグラウンドレベルを超える組換え発現されたMAGEファミリーのメンバーのいずれも認識されなかった。標的細胞の人工的負荷(以前に記載されている)の際に認識され得る、MAGEファミリーのメンバーに由来するペプチドは、内因的に発現されたMAGEタンパク質からプロセシングされないか、MHC分子に負荷されないか、または細胞表面に提示されないかのいずれかである。したがって、これらのペプチドは、免疫原性T細胞エピトープとして適格ではなく、TCR T15.8-4.3-83媒介性交差認識に対する臨床的に適切な標的であるとも考えられない。
TCRの安全プロファイルを調査するために、異なる起源の健康な組織に由来する細胞のTCR媒介性認識が調査され、除外されなければならない。したがって、TCRを発現するエフェクターを、腎臓(PromoCellによって得られる腎皮質上皮細胞)、肺(Lonzaによって得られる肺線維芽細胞)、およびiPS由来の肝細胞、心筋細胞、および内皮細胞(Cellular Dynamicsによって得られる)に由来する正常細胞と共培養した。調査した細胞は、抗原MAGEA1に対して内因的に陰性であり、したがって、潜在的なMAGEA1に関連しないオフターゲット毒性の特定が可能になった。
正常細胞を解凍し、供給業者に指定されたように、共培養前の1週間培養した。細胞を96ウェル平底プレートに播種し、3名の異なるドナーに由来するTCR T15.8-4.3-83を形質導入したかまたは形質導入されていないエフェクター(データは示さず)と共培養した。標的細胞のHLA-A2発現を、抗体染色およびFACSによって確認し(データは示さず)、標的細胞を未改変または対照としてのMAGEA1由来KVL(配列番号1)ペプチドの過剰量(10-5M)を負荷したかのいずれかで試験した。対照として、エフェクター細胞および標的細胞を単独で播種し、T2細胞にTCRのエピトープKVL(10-5M;配列番号1)を負荷した。約20時間の共培養後に、上清を回収し、ELISAによって分析した。約48時間の共培養後に、IncuCyte(商標) Zoomデバイス(Essen BioScience Inc.)で位相差イメージを撮影し、接着標的細胞の溶解および剥離について潜在的な毒性効果を可視化した。
図8に示されるように、3名のドナーに由来するTCR T15.8-4.3-83を形質導入したエフェクターの共培養により、人工的にKVLペプチドを負荷した細胞のみの認識がもたらされ、MHC分子の文脈で、それらの細胞表面にTCRのエピトープを適切に提示する標的細胞の能力を表した。未改変正常細胞は、トランスジェニックTCR T15.8-4.3-83を発現するエフェクターによって認識されず、調査された受容体の好ましい安全プロファイルを示した。
様々なKVLペプチドに特異的なTCRの機能性アビディティを測定するために、段階的な量のKVLペプチドを負荷したT2細胞との共培養において、最大半量の相対的IFN-γ放出(EC50値)が決定される。
TCRトランスジェニックT細胞集団の機能性アビディティを、段階的な量のKVLペプチド(10-4M~10-12M)を負荷したT2細胞との共培養において、最大半量の相対的IFN-γ放出(EC50値)として決定する。20時間の共培養後に、標準的IFN-γ ELISAを実施する(4000pgを超える値は、三次多項式を使用して推定する)。
様々なKVL特異的TCRは、それらの機能性アビディティにおいて、KVLペプチドを負荷したT2細胞と異なる。TCR T15.8-4.3-83は、低いEC50値、すなわち、国際公開第2018/170338号パンフレットに開示されたTCR FH1、FH2、FH3およびFH4ならびに国際公開第2018/104438号パンフレットに開示されたR37P1C9と比較して、KVLペプチドを負荷した2種の細胞の高い機能性アビディティを示す。
異なるTCRを形質導入したT細胞のTH2特異的サイトカインIL-4、IL-5およびIL-13のTCR分泌パターンを比較する。Th2型の炎症は、腫瘍成長を容易にすることが提唱されている(Jager MJ, Desjardins L, Kivela T, Damato BE (eds): Current Concepts in Uveal Melanoma. Dev Ophthalmol. Basel, Karger, 2012, vol 49, pp 137-149、J Immunother. 2018 Oct;41(8):369-378)。したがって、低いまたは検出不能な分泌レベルのTh2は、腫瘍退縮に有利であり得る。
20.000個のTCRトランスジェニックエフェクターT細胞を、丸底の96ウェルプレートにおいて、20.000個の腫瘍細胞と共培養する。MAGPIX(登録商標)システムのプロトコールに従って、MILLIPPLEX(登録商標) MAP Kitを使用して、分泌されたサイトカインに関して、無細胞上清を分析した。
T1367 TCRを形質導入したT細胞は、いくつかのTH2サイトカイン、特にIL-4、IL-5およびIL-13を分泌する。15.8-4.3-83 TCRを形質導入したT細胞は、TH2サイトカインの同等の分泌を示さないが、国際公開第2014/118236号パンフレットに開示されたT1367は、これらのサイトカインのかなりの分泌を示す。

a)実施形態27で実現されている少なくとも1つの核酸を含む発現ベクター、または
b)実施形態1から21のいずれか1つで実現されているTCRのアルファ鎖をコードする核酸を含む第1の発現ベクター、および実施形態1から21のいずれか1つで実現されているTCRのベータ鎖をコードする核酸を含む第2の発現ベクター
を含む、実施形態32から34に記載の細胞。
Claims (12)
- MAGEA1に対して特異的な単離されたT細胞受容体(TCR)であって、
- 配列番号2のアミノ酸配列を有するCDR1、配列番号3のアミノ酸配列を有するCDR2および配列番号4のアミノ酸配列を有するCDR3を含むTCRα鎖、並びに、
- 配列番号5のアミノ酸配列を有するCDR1、配列番号6のアミノ酸配列を有するCDR2および配列番号7のアミノ酸配列を有するCDR3を含むTCRβ鎖
を含み、
配列番号1のアミノ酸配列のHLA-A2結合形態を特異的に認識する、前記TCR。 - 前記配列番号1のアミノ酸配列のHLA-A2結合形態が、HLA-A*02:01、HLA-A*02:04、HLA-A*02:16およびHLA-A*02:17からなる群から選択される遺伝子によってコードされた分子によって提示される、請求項1に記載の単離されたTCR。
- 配列番号8と少なくとも80%同一であるアミノ酸配列を有する可変TCRα領域および配列番号9と少なくとも80%同一であるアミノ酸配列を有する可変TCRβ領域を含む、請求項1または2に記載の単離されたTCR。
- 請求項1から3のいずれかに記載のTCRの機能性部分を含む単離されたポリペプチドであって、前記機能性部分が、配列番号2、3、4、5、6および7のアミノ酸配列を含み、前記機能性部分は、TCRの抗原への結合を媒介する、単離されたポリペプチド。
- 請求項1から3のいずれか一項に記載の少なくとも2つのTCRを含む多価TCR複合体。
- IFN-γ分泌が、
HLA-A*02:01、HLA-A*02:04、HLA-A*02:16、またはHLA-A*02:17の遺伝子によってコードされた分子によって提示される配列番号1のアミノ酸配列のHLA-A2結合形態への、
エフェクター細胞上に発現された、請求項1から3のいずれか一項に記載の単離されたTCR、請求項4に記載のポリペプチド、または請求項5に記載の多価TCR複合体の結合
によって誘導される、請求項1から3のいずれか一項に記載の単離されたTCR、請求項4に記載のポリペプチド、または請求項5に記載の多価TCR複合体。 - 請求項1から3のいずれか一項に記載のTCRをコードするかまたは請求項4に記載の単離されたポリペプチドをコードする核酸。
- 請求項7に記載の核酸を含むベクター。
- 請求項1から3のいずれか一項に記載のTCRを発現する細胞。
- MAGEA1に対する特異性を媒介する請求項1から3のいずれか一項に記載のTCRの部分に特異的に結合する抗体または抗原結合性断片。
- 医薬として使用するための、請求項1から3のいずれか一項に記載のTCR、請求項4に記載のポリペプチド、請求項5に記載の多価TCR複合体、請求項7に記載の核酸、請求項8に記載のベクター、または請求項9に記載の細胞。
- 前立腺がん、子宮がん、甲状腺がん、精巣がん、腎がん、膵臓がん、卵巣がん、食道がん、非小細胞肺がん、肺腺癌、扁平上皮癌、非ホジキンリンパ腫、多発性骨髄腫、メラノーマ、肝細胞癌、頭頚部がん、胃がん、子宮内膜がん、子宮頸がん、結腸直腸がん、胃腺癌、胆管細胞癌、乳がん、膀胱がん、骨髄性白血病および急性リンパ芽球性白血病、肉腫または骨肉腫からなる群から選択されるがんの処置において使用するための、請求項1から3のいずれか一項に記載のTCR、請求項4に記載のポリペプチド、請求項5に記載の多価TCR複合体、請求項7に記載の核酸、請求項8に記載のベクター、または請求項9に記載の細胞。
