Description
本発明は、CYP26として知られる、ある種のシトクロムP450酵素(P450RAI)を阻害する化合物に関連する。本発明はまた、レチノイド様活性を有する化合物にも関連する。

このレチノイド活性アッセイは、一過性にトランスフェクトされた、RAに感受性のレポーター構築物の発現を、試験化合物が誘導する能力を測定する。このアッセイにおいて、MCF-7細胞には、ホタルルシフェラーゼの発現を引き起こす2つのRA応答エレメントを含むCYP26A1遺伝子の上流プロモーターを含む構築物(pCYP26A1-luc)が、トランスフェクトされる。CYP26A1は、これらの細胞において、RAによって高度に誘導性であるので、この構築物は、レチノイド様転写活性に感受性であるレポーターとして利用される。

この実験は、造血幹・前駆細胞、および白血病幹細胞の、長期的な増殖能力を試験するために使用される。長期培養におけるコロニー形成能力は、幹細胞/前駆細胞の活性を示す。したがって、白血病幹細胞への影響の測定として、骨髄間質の存在下で、RAおよび/またはCYP26阻害剤で処理された白血病細胞は、コロニー形成能力について試験された。これらの実験のため、骨髄間質細胞および白血病細胞の共培養が、上述のように調製され、そしてRAおよび/またはCYP26阻害剤化合物で72時間処理される。処理期間後、白血病細胞は、収集され、そして、コロニー形成能力を評価するために、半固体のメチルセルロース培地に再プレーティングされる。35 mmプレートに、培養皿1枚につき、1.1 mlのMethoCult(STEMCELL Technologies, Vancouver, Canadaより入手可能)中の500個の白血病細胞がプレーティングされる。1回の処理につき、3連のプレートが調製され、そして培養物は、14~16日間にわたり、CO2インキュベーター内で増殖させる。培養期間後、コロニーは、解剖顕微鏡のもとで計数される。
RAは、骨細胞(軟骨細胞/骨芽細胞)の分化の種々の段階を妨害することによって、骨形成に影響をおよぼすことが示されている。これらの実験は、細胞培養モデルを用いて、インビトロでの骨芽細胞分化を減少させるまたは妨害する、CYP26阻害剤の能力を試験するために、使用される。MC3T3-E1マウス頭蓋冠前骨芽細胞(American Type Culture Collectionより入手可能)は、アスコルビン酸無しで、MEM-α中で培養された。骨芽細胞の分化、およびマトリックスのミネラリゼーションを誘導するため、細胞は、アスコルビン酸(50 μg/ml)およびリン酸(10 mM β-グリセロリン酸)を含む培地へと交換され、そして2~3日ごとに培地を代えながら、21日間まで培養される。細胞はまた、骨芽細胞分化への影響を評価するため、変化させた濃度のRAまたはCYP26阻害剤化合物で処理される。処理期間の最後で、24ウェルプレート中の細胞は、70%エタノール中で固定され、そして、骨芽細胞分化を示す、石灰化した細胞外マトリックスを可視化するため、アリザリンレッド(40 mM、pH 4.2)で染色される。染色されたウェルは、Leica MZ95解剖顕微鏡のもとで写真撮影された。6ウェルプレート中で増殖させた他の細胞は、RNA抽出、ならびに骨分化マーカー、およびRAの標的遺伝子のqPCR分析のために、使用される。
これらの実験は、初代細胞または細胞株の、RAを代謝する能力を評価するためのものであり、かつこの能力に対する、CYP26A1またはCYP26B1の阻害の影響を決定するためのものである。これらの実験のため、細胞は、48ウェルプレートへとプレーティングされ、そして放射能標識(3H)されたRAで処理される。この標識されたRAの、水溶性代謝物への変換の度合いが、上述のように、細胞ベースの阻害剤アッセイにおいて決定される。初代ヒト骨髄間質細胞(Lonzaより入手可能)は、MyeloCult(Stem Cell Technoloigies, Vancouver, Canadaより入手可能)中で培養された。マウスOP9骨髄間質細胞およびMC3T3前骨芽細胞は、上述のように培養された。細胞は適した培養培地にプレーティングされ、そして単層を形成させる。細胞はその後、CYP26発現を誘導するために、DMSO(ビヒクル対照)、またはRA(1 μM)のいずれかで、24時間にわたり前処理される。ビヒクルの、およびRAに誘導される細胞は、その後、RA代謝に対する影響を調べるため、試験CYP26阻害剤の存在下または非存在下で、3H-RAに曝露される。細胞は3時間にわたり標識されたRAとインキュベートされ、そして代謝物は上述のように抽出されそして分析される。
ヒトおよびマウス骨髄間質細胞はCYP26酵素を発現しており、かつCyp26B1は、これらの細胞において、RAによって高度に誘導性である。この実験は、CYP26阻害剤への骨髄の曝露が、骨髄におけるRAレベルを上昇させるのに、およびRAの標的遺伝子を誘導するのに、十分であるかどうかを試験する。マウス(8~12週齢)は、連続した2日間(1日につき1回)に、0.9%塩化ナトリウム/0.1% DMSOに溶解されたCYP26阻害剤化合物の静脈内注射(尾静脈)を受けた。動物には、40 μg/kgの用量の阻害剤が、または同量のビヒクルのみが投与された。2回目の注射後の朝、動物は犠牲にされ、そして骨髄単核細胞が、Lympholyte-mammal試薬(Cedarlane, Burlington, Canadaより入手可能)を用いて大腿骨から採取された。RNAはその後、単離された細胞から抽出され、そしてRAの標的遺伝子の発現は、qPCRによって評価された。
レチノイドは、ざ瘡を含む、ヒトにおける多様な皮膚疾患の処置において有用であることが示されている。これらの実験は、ざ瘡のマウスモデルにおける、潜在的な抗ざ瘡の効力についての、CYP26阻害剤化合物、および他のレチノイドの能力を試験する。ライノマウスモデルは、ざ瘡に類似する皮膚病変を発症する、無毛の変異体であり、かつ、ざ瘡の新規な治療法を評価するための、そしてレチノイドのインビボ活性を評価するためのモデルとして、広く使用されている。試験化合物またはRAは、アセトン/10% DMSO中に溶解され、そして0.4 mlが、動物(2匹の雄 + 2匹の雌)の背部の皮膚に、1日に1回、1週間につき5日、連続した2週間にわたり、局所適用された。最後の処置の翌日に、動物は犠牲にされ、そして皮膚は、処置動物から収集され、そして組織学上の処理がなされ、そして表皮シートは、嚢の可視化のため調製された。組織切片は、ホルマリン固定された試料をパラフィン包埋することによって調製され、そしてヘマトキシリンおよびエオシンで染色された。写真は、Nikon Eclipse E600顕微鏡のもとで、Q Imagingソフトウェアを用いて、20Xの拡大率で撮影された。表皮シートは、皮膚試料を5日間、0.5%酢酸中で4度で5日間処理し、その後で真皮層と表皮層とが慎重に分離され、そして表皮がその後、一連のエタノール処理(70%、80%、90%、および100%)を用いて脱水されたことによって、調製された。100%エタノール中での一晩の脱水後、表皮層は、CitriSolv透徹剤(Fisher Scientificより入手可能)を用いて透徹され、そしてパーマウント(Fisher Scientificより入手可能)中で、ガラススライドにマウントされた。表皮層はその後、上述のように写真撮影された。
ヒト組織試料は、ヒトCYP26B1に対する抗体(Sigma-Aldrich, Canadaより入手可能)を用いて染色された。染色は、Queen’s Laboratory for Molecular Pathologyにおいて、ベンタナシステムを用いて実施された。

Claims (23)
- 請求項1~4のいずれか一項に記載の化合物を含む、哺乳類における皮膚の状態を処置するための薬学的組成物。
- 皮膚の状態が、皮膚の疾患である、請求項5に記載の薬学的組成物。
- 皮膚の状態が、光線性角化症、ヒ素性角化症、炎症性のおよび非炎症性のざ瘡、乾癬、魚鱗癬、および他の角化、皮膚の過増殖性の障害、湿疹、アトピー性皮膚炎、ダリエー病、扁平苔癬、グルココルチコイドによる損傷、またはステロイド性萎縮である、請求項5に記載の薬学的組成物。
- 化合物が、局所抗微生物薬として、皮膚の抗色素沈着薬として、加齢および光による皮膚の損傷の影響を処置および回復させるために、適用される、請求項5~7のいずれか一項に記載の薬学的組成物。
- 請求項1~4のいずれか一項に記載の化合物を含む、哺乳類におけるがん性の状態または前がん性の状態を処置または予防するための薬学的組成物。
- 状態が、前悪性のもしくは悪性の過増殖性の疾患、乳房の、皮膚の、前立腺の、子宮頸部の、子宮の、結腸の、膀胱の、食道の、胃の、肺の、喉頭の、口腔の、血液の、リンパ系のがん、血液障害、化生、異形成、新形成、白板症、粘膜の乳頭腫、またはカポジ肉腫である、請求項9に記載の薬学的組成物。
- 請求項1~4のいずれか一項に記載の化合物を含む、哺乳類における増殖性硝子体網膜症(PVR)、網膜剥離、ドライアイ、またはコルネオパシー(corneopathy)を含む、目の疾患を処置するための薬学的組成物。
- 請求項1~4のいずれか一項に記載の化合物を含む、哺乳類における血管疾患を処置するための薬学的組成物。
- 血管疾患が、脂質代謝に関連する疾患、脂質異常症、または血管形成術後の再狭窄の予防を含む、請求項12に記載の薬学的組成物。
- 請求項1~4のいずれか一項に記載の化合物を含む薬学的組成物であって、哺乳類における、
循環中の組織プラスミノーゲンアクチベーター(TPA)のレベルを増加させるための、または
ヒトパピローマウイルス(HPV)に関連する状態および疾患、炎症性疾患、神経変性疾患、脳卒中、成長ホルモンの異常な産生を含む異常な下垂体機能、免疫系に関連する疾患を処置するもしくは予防するための、
アポトーシスの誘導およびT細胞により活性化されたアポトーシスの阻害の両方を含む、アポトーシスを調節するための、

- 請求項1~4のいずれか一項に記載の化合物を含む、哺乳類における、異所性骨形成の障害、または筋組織石灰化の処置のための薬学的組成物。
- 粉末、スプレー、丸剤、錠剤、シロップ、エリキシル、注射によって投与され得る溶液もしくは懸濁液、坐剤、皮膚下に留置するための徐放製剤、または筋肉内注射として投与される、請求項5~16のいずれか一項に記載の薬学的組成物。
- 化合物が、1 mLにつき0.01ミリグラム~1 mgの該化合物を含む製剤の状態で局所適用のための医薬中に含まれる、請求項5~8のいずれか一項に記載の薬学的組成物。
- 化合物が、1日につき体重1 kgあたり0.01~5 mgを含む製剤の状態で全身投与のための医薬中に含まれる、請求項5~16のいずれか一項に記載の薬学的組成物。
- 化合物が、レチノール、レチンアルデヒド、RA、もしくは他の天然のもしくは合成レチノイドから選択される、レチノイドまたはレチノイド前駆体との組み合わせとして提供される、請求項5~19のいずれか一項に記載の薬学的組成物。
- 組み合わせが、錠剤、カプセル、注射剤、または局所製剤として提供される、請求項20に記載の薬学的組成物。
