Description
- R2は、水素原子又はメチル若しくはエチル基を表し;
- R3は、ヒドロキシル基で任意選択的に置換された直鎖C1~C12アルキル基(飽和);又はヒドロキシル基で任意選択的に置換された直鎖C2~C12アルケニル基(C=C不飽和)を表す)
の少なくとも1種の化合物及びまたその有機若しくは無機の酸若しくは塩基の塩又は水和物等のその溶媒和物の使用であって、特にBcc菌群の細菌によって汚染され得る製造物、例えば水、水性組成物、無水組成物、産業用又は家庭用であり得る、特に食品、化粧品又は医薬品用の設備、産業用又は家庭用調合物に使用される溶媒、好ましくは水性組成物又は水の、セパシア菌群の細菌に対する処理における使用に関する。
「製造物の処理又は複数の製造物の処理」という表現は、製造物をセパシア菌群の細菌に対して処理することを意味することを意図しており、前記製造物のセパシア菌群の細菌による汚染を防止すること又は前記処理される製造物からセパシア菌群の細菌の一部若しくは全部を除染することのいずれかを目的として、処理される製造物のサンプルに連続的又は回分式(回分方式)に物質を添加することを含む。

の化合物及びまたその有機若しくは無機の酸若しくは塩基の塩又は水和物等のその溶媒和物。
- R2は、水素原子及びCH3基から選択され;一層より良好には、R2は、水素原子を表し;及び/又は
- R3は、(i)C1~C10アルキル基;(ii)C2~C10アルケニル基、特に-CH=CH-R4基(ここで、R4は、直鎖C1~C6アルキル基を表す);又はさもなければ(iii)構造-CH2-CH(OH)-R5(ここで、R5は、直鎖C1~C10、好ましくはC4~C10アルキル基を表す)を有するヒドロキシアルキル基を表す。
- δdは、分子の影響中に誘起される双極子の生成に由来するロンドン分散力を特徴付け;
- δpは、永久双極子間のDebye相互作用による力を特徴付け;
- δhは、特殊な相互作用による力(水素結合、酸/塩基、供与体/受容体等)を特徴付ける)
で定義されている。ハンセン溶解度の三次元空間における溶解度の定義は、C.M.Hansenによる論文:“The three dimensional solubility parameters”J.Paint Technol.39,105(1967)に記載されている。Hansen:“The three dimensional solubility parameters”J.Paint Technol.39,105(1967)。
好ましくは、処理される水及び水性組成物を式(I)の化合物と接触させる前記ステップは、特に、前記化合物を液体形態で注入することによるか、前記化合物を含むフィルター若しくは濾過カートリッジを通過させることによるか、又は前記化合物を固体形態、特に顆粒形態、薬用ドロップ形態若しくはペレット形態で投与することによって実施することができる。
本発明による特定の一実施形態によれば、セパシア菌群の細菌によって汚染され得る製造物を処理、特に防腐するための方法は、上に定義した式(I)の1種又は複数の化合物、好ましくはEZを、前記製造物を製造する段階中に組み込むステップを含む。
本発明の式(I)の化合物、特にEZの最小阻害濃度(MIC)及び最小致死濃度(MBC)を、Rushton et al.Key role for efflux in the preservative susceptibility and adaptive resistance of Burkholderia cepacia complex bacteria.Antimicrobial Agents and Chemotherapy,2013.57(7):p.2972-2980 Key role for efflux in the preservative susceptibility and adaptive resistance of Burkholderia cepacia complex bacteria.Antimicrobial Agents and Chemotherapy,2013.に記載されている標準化された寒天希釈試験及びブロス希釈試験により、TSA/TSBを用いて決定した。産業製造物の製造及び保存を代表するように、培養を30℃で行った。DMSO(Sigma-Aldrich、United Kingdom)中で防腐剤原液(50%w/v)を調製し、試験濃度となる量で培地に添加した。細菌が存在するDMSOの最終濃度が無毒であることを確認し(4%v/v以下)、増殖への影響を排除するため、対照条件として試験に含めた。MICを、培養液OD(630nm)が80%低下するか又は寒天培地上で被験生物の増殖が目視で確認できなくなる防腐剤の最小濃度と定める。MBCを、死滅率が99%となり、且つ回復培地(TSA)上で増殖が停止する最小濃度として求めた。回復及び被験菌の生菌数の計数を行う前に、防腐剤を、Rushtonらによる科学論文(Rushtonらの同文献)に記載されている通りに、レシチン3%を含むTween 80を1.5%v/v含む中和液で希釈することにより不活性化した。(Rushton et al,ibid).実験を行う前に、中和液の有効性及び毒性を、J.C.Learらによる科学論文(J.C.,et al.,Chloroxylenol- and triclosan-tolerant bacteria from industrial sources - Susceptibility to antibiotics and other biocides.International Biodeterioration and Biodegradation,2006.57(1):p.51-56)International Biodeterioration and Biodegradation,2006.に記載されている通りに評価した。
細菌株及び培養条件
式(I)の化合物、特にEZに対する感受性のプロファイリングに使用する59種のバークホルデリア(Burkholderia)属のパネルを作成した。このコレクションは、セパシア菌群(Bcc)の現時点における20の菌種群、Bcc実験菌株のパネルの参照株(Mahenthiralingam,E.,et al.,Diagnostically and experimentally useful panel of strains from the Burkholderia cepacia complex.Journal of Clinical Microbiology,2000.38(2):p.910-913)Journal of Clinical Microbiology,2000.及びRushtonらによって防腐剤の感受性に関して既にプロファイリングされた39種の菌株(Rushton,L.,et al.,Key role for efflux in the preservative susceptibility and adaptive resistance of Burkholderia cepacia complex.bacteria.Antimicrobial Agents and Chemotherapy,2013.57(7):p.2972-2980)からなるものとした。Antimicrobial Agents and Chemotherapy,2013.57(7):p.2972-2980).バークホルデリア(Burkholderia)属菌株のパネルは、臨床(n=28)、環境(n=23)及び産業(n=8)由来の菌株から分離したものである。菌株をトリプトンソイ(TSB/TSA)(Oxoid Ltd、United Kingdom)上で30℃において1回培養した。菌株を、ジメチルスルホキシド(DMSO)(Sigma-Aldrich、UK)8%v/vを含むTSB中、-80℃で保存した。

- 箱の幅は、サンプルのサイズの平方根に比例する。
Claims (16)
- 式(I):

- R3は、ヒドロキシル基で任意選択的に置換された直鎖C1~C12アルキル基(飽和);又はヒドロキシル基で任意選択的に置換された直鎖C2~C12アルケニル基(C=C不飽和)を表す)
の少なくとも1種の化合物及びまたその有機若しくは無機の酸若しくは塩基の塩又は水和物等のその溶媒和物の、セパシア菌群(Bcc)の細菌に対する使用。 - 前記Bcc菌群の細菌によって汚染され得る製造物の汚染を防止するための、請求項1に記載の使用。
- 前記Bcc菌群の細菌によって汚染され得る製造物を処理するための、請求項1に記載の使用。
- 式(I)において、
- R2が、水素及びメチルから選択され、並びに/又は
- R3が、(i)C1~C10アルキル基;(ii)C2~C10アルケニル基;若しくはさもなければ(iii)構造-CH2-CH(OH)-R5(ここで、R5は、直鎖C1~C10 アルキル基を表す)を有するヒドロキシアルキル基を表す
、請求項1~3のいずれか一項に記載の使用。 - 前記式(I)の化合物が、以下の化合物:

- 前記式(I)の化合物が、単独で又は混合物として、処理される水又は水性組成物の総質量に対して少なくとも0.06質量%の比率で使用される、請求項1~6のいずれか一項に記載の使用。
- 製造物を処理するための方法であって、i)前記製造物の、セパシア菌群の細菌を含む1種若しくは複数の微生物による汚染を防止すること、又はii)前記Bcc菌群の細菌を含む1種若しくは複数の微生物によって汚染された、前記汚染された製造物を処理することのいずれかを可能にし、前記製造物のサンプルを、連続的又は回分式に、請求項1~6のいずれか一項に記載の式(I)の少なくとも1種の化合物と接触させる少なくとも1つのステップを含む、方法。
- 前記製造物が、請求項1~6のいずれか一項に記載の式(I)の化合物を有する、家庭用又は産業用製造物から選択される、請求項8に記載の方法。
- 前記式(I)の化合物が、単独で又は混合物として、処理される水又は水性組成物の総質量に対して少なくとも0.06質量%の比率で使用される、請求項8又は9に記載の方法。
- セパシア菌群の細菌によって汚染され得る製造物を処理するための方法であって、前記製造物に、その製造中又はその製造後に、請求項1~6のいずれか一項に記載の式(I)の1種又は複数の化合物を組み込むステップを含む、方法。
- 請求項1~6のいずれか一項に記載の式(I)の1種又は複数の化合物を、前記製造物を製造する段階中に組み込むステップを含む、請求項11に記載の方法。
- 前記組み込みが、調合槽に予め式(I)の1種又は複数の化合物を導入し、その後、前記槽に、処理及び防腐される前記製造物又は前記製造物を製造するために必要な成分を導入することによって実施される、請求項11に記載の方法。
- 前記製造物の前記製造に必要な各成分が、請求項1~6のいずれか一項に記載の式(I)の1種若しくは複数の化合物で予め処理されているか、又はさもなければ、各成分が、式(I)の1種若しくは複数の化合物との混合物として導入され;その後、前記製造物が、防腐され、且つ請求項1~6のいずれか一項に記載の式(I)の1種又は複数の化合物の存在下で使用者に提供される、請求項11又は13に記載の方法。
- 前記方法が、請求項1~6のいずれか一項に記載の式(I)の1種又は複数の化合物を、前記製造物を製造する前記段階後に組み込むステップを含み、前記製造物は、製造されると、式(I)の1種又は複数の化合物と接触され、その後、前記製造物は、防腐され、且つ式(I)の1種又は複数の化合物の存在下で使用者に提供される、請求項11又は12に記載の方法。
- セパシア菌群(Bcc)の細菌に対する使用のための、請求項1及び4~6のいずれか一項に記載の式(I)の1種又は複数の化合物を含む「無水」組成物、即ち5%未満の量の水を有する組成物であって、前記組成物が、i)アニオン性、非イオン性、カチオン性又は両イオン性界面活性剤、ii)脂肪物質、iii)アニオン性、非イオン性、カチオン性又は両イオン性の、会合性又は非会合性の増粘性又は非増粘性ポリマー、iv)染料、v)顔料、vi)充填剤、及びvii)これらの混合物から選択される1種又は複数の成分を含む、組成物。
