本開示は、一般に、抗コネキシン(Cx)43抗体と、例えば骨細胞におけるCx43ヘミチャネルの開放に関連する疾患または症状の処置におけるその使用とに関する。
複数の予防的および治療的アプローチにもかかわらず、癌は、世界中で主な死因の1つである。2010~2020年に、米国における新たな癌症例の数は、男性では約24%増加して年間100万症例を超え、女性では約21%増加して年間90万症例を超えると予想される。最も増加すると予想される癌のタイプは、男性および女性の両方で黒色腫;男性では前立腺癌、腎臓癌、肝臓癌および膀胱癌;ならびに女性では肺癌、乳癌、子宮癌および甲状腺癌である。癌は依然として、米国における2番目に多い死因であり、死亡の4件ごとにほぼ1件を占めている。多くの癌は、現在のアプローチを用いて処置困難または不可能である。多くの癌は、現在の処置レジメンを回避するか、処置に対して耐性になるか、または処置後に再発する。
骨細胞におけるCx43ヘミチャネルの開放(例えば、開放不足または不全)に関連する疾患または症状、例えば癌、癌転移、骨肉腫、骨粗鬆症または骨減少症を処置するための組成物および方法が本明細書で提供される。
それぞれ配列番号1、2および3のアミノ酸配列を有する第1、第2および第3の重鎖相補性決定領域(CDR)配列と;

抗Cx43抗体は、当技術分野で一般に公知の様々な方法を使用して作製され得る。例えば、ファージディスプレイ技術を使用して、ヒト抗体ライブラリーをスクリーニングし、治療のための完全ヒトモノクローナル抗体を生産し得る。高親和性結合物は、中和研究の候補と考えられ得る。あるいは、従来のモノクローナルアプローチを使用し得、マウスまたはウサギをヒトタンパク質で免疫化し、候補結合物を同定および試験し、最後に、重鎖および軽鎖の結合部位をヒト抗体コード配列にグラフトすることにより、ヒト化抗体を生産し得る。
本開示は、Cx43分子の1つまたはそれを超えるドメイン、例えば細胞外ループ内に見られる1つまたはそれを超えるアミノ酸と相互作用する抗Cx43抗体を提供する。抗体が結合するエピトープは、1つまたはそれを超える細胞外ループ内に位置する2つまたはそれを超える(例えば、2つ、3つ、4つ、5つ、6つ、7つ、8つ、9つまたはそれを超える)アミノ酸の1つまたはそれを超える隣接配列を含み得る。あるいはまたは加えて、エピトープは、1つまたはそれを超える細胞外ループ(例えば、立体構造エピトープ)内に位置する1つまたはそれを超える非隣接アミノ酸(またはアミノ酸配列)を含み得る。

限定されないが、再構成製剤および液体製剤を含む本開示の製剤は、公知の方法、例えばボーラスとしての静脈内投与にしたがって、または、筋肉内、腹腔内、脳脊髄内、皮下、関節内、滑液嚢内、髄腔内、経口、局所もしくは吸入経路による一定期間にわたる持続注入により、抗Cx43抗体による処置を必要とする哺乳動物、好ましくはヒトに投与される。
以下のプロトコールにしたがって、Cx43への結合親和性について、最適化配列を試験した。
2.新たなCM5チップを挿入する
3.HBS-EP+バッファー(10mM Hepes、150mM NaCl、3mM EDTA、0.05%界面活性剤P20[tween20])で3回プライミングする。
4.前提条件:100ul/分で新たなセンサーグラムを開始する。高粘度溶液(エクストラクリーン)で再生コマンドを使用して、各10ulの2×100mM HCl、2×50mM NaOH、2×0.5%SDSを注入する。これは、カップリングのためにチップをクリーニングおよび調製する。タンパク質が付着した後ではなく、ブランクチップに対してのみこれを行う。
5.GEヒト抗体捕捉キットに付属の説明書にしたがって、使用する各表面(参照表面を含む)に抗ヒトIgG Fc(GE:BR-1008-39)を別個にアミンカップリングする。簡潔に言えば、Mabを10mM酢酸ナトリウムpH5で25ug/mlに希釈する。5ul/分で新たなセンサーグラムを開始する。立て続けに、NHS/EDCで7’を活性化し、Mab7’を注入し、エタノールアミン7’でブロッキングする。典型的には、10,000~12,000RU/表面が得られる。この後、3M MgCl2で20ul/分で30秒間10回再生する。(本発明者らは、GEマウス抗体捕捉キット(GE:BR-1008-38)を使用したことを除いて、マウスIgGを用いた実験を同様に行ったことに留意する。唯一の違いは、抗マウスIgG抗体を30ug/mlでカップリングし、10mMグリシン-HCl pH1.7で20ul/分で3分間再生することである)。
6.正規化の後、HBS-EP+バッファーで1回プライミングする。
7.次いで、アミンカップリングの直後に実験を実施した。実験の間にかなりの時間があった場合、チップを取り出して4℃で保存した。チップを機器に戻す場合には、この後、HBS-EP+で3回プライミングし、正規化し、HBS-EP+で1回プライミングした。

T200評価ソフトウェア2.0を使用して、データを分析した。動態に最適な条件下で平衡に達したので、3回反復の各セットからの(Fc2-Fc1)データを、1:1結合動態モデルまたは定常状態親和性モデルのいずれかにグローバルフィッティングした。両方法で得られた結果は類似していた。
1.製造業者の説明書にしたがって、GE Healthcareのビオチン捕捉キット(28920234)のCAPチップを調製した。簡潔に言えば、それを機器(T200)にドッキングし、ランニングバッファー(HBS-EP+)で3回プライミングし、スタンバイモードでランニングバッファーで一晩水和させた。次いで、再生溶液(6M GuHCl、250mM NaOH)を30ul/分で3×60秒間注入することにより、それをコンディショニングした。この後、1回の正規化および1回のプライミングを行った。次いで、それが実験に使用可能になった。以下のパラメータを使用して、プログラムを作成した。

2.濃度系列の各Mab(IまたはH):0、6.2nM、18.5nM、55.6nM、166.7nM、500nMをランした。

Fcエフェクター機能は、受容体へのFcの結合により媒介される。受容体としては、FCRI、FCRIIa、FCRIIb、FCRIIIa、FCRIIIb、C1qおよびFcRnが挙げられる。FcRnを除くほとんどのFc受容体への結合親和性を減少させて、抗体半減期を維持しながら、潜在的なインビボ毒性を最小化することが一般に望ましい。以下の表面プラズモン共鳴(SPR)および酵素結合免疫吸着測定法(ELISA)プロトコールを使用して、様々な抗体に対する様々なFc受容体結合を試験した。

注入の直前に400mM EDCおよび100mM NHSを混合することにより、アクチベーターを調製した。CM5センサーチップを混合物で420秒間活性化した。次いで、10mM NaAc(pH4.5)中30μg/mLのTHE(商標)Hisタグ抗体をチャネル1~8に30μL/分の流速で400秒間注入した。1Mエタノールアミン-HCl(GE)により、チップを不活性化した。
ランニングバッファー(1×HBS-EP+)中2μg/mL CD64をチャネル1~4のFc2に10μL/分の流速で30秒間注入した。6つの濃度(40、20、10、5、2.5および1.25nM)の分析物20170905-Ab#C-02、20170905-Ab#D-02、20170908-Ab#G-02、20170920-Ab#H-02およびランニングバッファーをチャネル1~4のFc1-Fc2に30μL/分の流速で180の会合段階、続いて400の解離で順番に注入した。分析物濃度にしたがって昇順で、リガンドの捕捉および分析物のランを6サイクル反復する。あらゆる解離段階の後に、再生バッファーとして10mMグリシンpH1.5を注入した。
チップを10mMグリシンpH1.5で再生した。
リガンドの捕捉なしの表面チャネルFc1を参照減算の対照表面として使用した。参照チャネルおよびバッファーチャネルデータから各相互作用の最終データを差し引いた。CD64への20170905-Ab#C-02、20170905-Ab#D-02、20170908-Ab#G-02、20170920-Ab#H-02結合の実験データを1:1結合モードでフィッティングした。分析物20170907-Ab#K-02、20170908-Ab#L-02、20170915-Ab#O-02、20170919-Ab#P-02、20170919-Ab#S-02および20170920-Ab#T-02の10240nM曲線を除去して、より良好なフィットを可能にした。相対的な実験データは定常状態親和性によりフィッティングし、以下の表2に示されている。

(1)バッファー交換
取扱説明書にしたがって脱塩カラムを使用して、ヒトFcRnのバッファーをランニングバッファー(50mM Na2HPO4、50mM NaH2PO4、150mM NaCl、0.05%Tween20、pH6.0)に交換した。Nanodropにより、濃度を決定した。
注入の直前に400mM EDCおよび100mM NHS(GE)を混合することにより、アクチベーターを調製した。CM5センサーチップを混合物で10μL/分の流速で420秒間活性化した。次いで、10mM NaAc(pH5.5)中5μg/mLの抗体をそれぞれチャネル1~8のFc2に10μL/分の流速で60秒間注入した。相対的Fc1をブロッキングした。1Mエタノールアミン-HCl(GE)により、チップを10μL/分の流速で420秒間不活性化した。
分析物FcRnをそれぞれチャネル1~8に注入した。8つの濃度のFcRn(0、93.75、187.5、375、750、1500、3000および6000nM)を30μL/分の流速で60秒間の会合段階、続いて90秒間の解離でモニタリングした。相互作用分析の各サイクルの後、センサーチップ表面を1×PBS(pH7.4)で10μL/分の流速で30秒間再生した。
チップを1×PBS(pH7.4)で再生した。
固定化抗体なしの表面チャネルFc1を参照減算の対照表面として使用した。参照チャネルおよびバッファーチャネルデータから各相互作用の最終データを差し引いた。実験データは定常状態親和性モードによりフィッティングし、以下の表5に示されている。
プレート(Nunc)を3μg/mLの抗体で4℃で一晩コーティングした。ブロッキングおよび洗浄の後、C1qをブロッキングバッファー(600、189.75、60.01、18.98、6.00、1.90、0.60、0.19、0.06および0.02μg/mL)で半対数滴定し、室温で2時間インキュベートした。次いで、プレートを洗浄し、続いて、二次抗体ヒツジ抗ヒトC1qAb-HRPと共に1時間インキュベートした。洗浄後、TMB基質を添加し、2M HClにより相互作用を停止した。マイクロプレートリーダー(Molecular Device)を使用して450nmの吸光度を読み取り、以下の表6に示した。
標的分子のエピトープを再構築するために、固相Fmoc合成を使用して、ペプチドベースのエピトープ模倣物のライブラリーを合成した。独自の親水性ポリマー製剤でグラフトし、続いて、N-ヒドロキシベンゾトリアゾール(HOBt)と共にジシクロヘキシルカルボジイミド(DCC)を使用してt-ブチルオキシカルボニル-ヘキサメチレンジアミン(BocHMDA)と反応させ、続いて、トリフルオロ酢酸(TFA)を使用してBoc基を切断することにより、アミノ官能化ポリプロピレン支持体を得た。標準的なFmoc-ペプチド合成を使用して、カスタム改変JANUS液体ハンドリングステーション(Perkin Elmer)により、アミノ官能化固体支持体上でペプチドを合成した。
抗体の安定性は、開発、有効性、生産コストなどに影響を与える重要な要素である。配列最適化後、重要な安定性パラメータを評価した。酸性および熱条件下における様々な抗体の種分布プロファイルを試験した。すべての抗体は、改善された安定性を示す。
製剤:PBS、pH6.5または7.2
濃度(mg/mL):5.28、5.13、5.00、5.17、5.01、5.26、4.92、5.04、4.99、5.12(すべて約5mg/mL)
条件:室温、酸性処理、次いで4℃で1週間または40℃で1週間保管
2μLのサンプルをACQUITY UPLC Protein BEH SEC 200、1.7μm、4.6×150mmカラムに0.3mL/分の流速で10分間注入した。50mMリン酸ナトリウム、500mM NaCl、pH6.2の移動相を使用した。すべての抗体は、様々なpH、熱および保管条件下で望ましい安定性を示す。
製剤:PBS、pH6.5、7.2、6.2
濃度(mg/mL):0.5
条件:室温、酸性処理、次いで4℃で1週間または40℃で1週間保管
LabChip GXIIシステム(PerkinElmer)に供する前に、還元標識バッファー中でサンプルを調製した。すべての抗体は、様々なpH、熱および保管条件下で望ましい安定性を示す。
A.インビトロアッセイ
本明細書に開示される抗体は、色素取り込みアッセイを使用して、ヘミチャネル開放に対するそれらの効果についてインビトロで試験され得る。色素は、蛍光トレーサー色素(例えば、エチジウムブロミドまたはルシファーイエロー)であり得る。
(a)コネキシン発現細胞または細胞株を単離、取得または生産すること。例えば、初代骨細胞は、頭蓋冠から単離され得る。他の細胞タイプは、当技術分野で公知の他の方法を使用して単離され得る。特定の態様では、頭蓋冠骨細胞は、動物(例えば、16日齢のニワトリ胚性頭蓋冠または新生児マウス)から単離される。動物を断頭し、頭蓋冠骨を解剖し、70%アルコールに迅速に浸漬する。次いで、頭蓋冠骨をαMEMに入れ、PBSで複数回洗浄する。クリーニングした骨を新鮮αMEMに入れる。骨を細断し、1.5mm領域サイズに切断する。骨片をコラゲナーゼで処理して軟組織および類骨を除去し、続いて、EDTAを使用して脱灰し得る。最後に、コラゲナーゼによる処理および激しい撹拌により、骨片から骨細胞を放出させる。
特定の態様では、骨細胞におけるCx43モジュレーションは、候補試薬を長骨に注入し、蛍光トレーサー色素(例えば、カルセインまたはエバンスブルー)を使用して、骨細胞におけるヘミチャネルの開放をインサイチューで検出することにより決定される。
A.インビトロアッセイ
癌細胞遊走のアッセイ。骨細胞におけるCx43ヘミチャネルは、ADまたはFFSSの投与により開放される。開放したヘミチャネルは、培地への様々な因子の放出を可能にし、馴化培地(CM)が得られる。ADまたはFFSS処理CM中の放出された因子は、軟寒天および創傷治癒アッセイにより決定した場合に、癌細胞遊走を減少させる。対照CMで処理された癌細胞は、正常な遊走を示す。軟寒天アッセイは、足場依存性成長とは対照的に、足場非依存性成長のアッセイである。癌細胞のみが軟寒天上で成長し得、このマトリックス上におけるそれらの成長は、癌細胞増殖の程度を示す。
インビボにおける骨転移に対する試験抗体の効果は、脛骨内注射骨転移モデルおよび/または心臓内注射癌転移アッセイを使用して決定される。

Claims (12)
- 抗Cx43抗体またはその抗原結合断片であって、
それぞれ配列番号1、2および3のアミノ酸配列を有する第1、第2および第3の重鎖相補性決定領域(CDR)配列と;
それぞれ配列番号4、5および6のアミノ酸配列を有する第1、第2および第3の軽鎖CDR配列とを含む、抗Cx43抗体またはその抗原結合断片。 - 配列番号7のアミノ酸配列を有する重鎖可変ドメインと、配列番号8のアミノ酸配列を有する軽鎖可変ドメインとを含む、請求項1に記載の抗体またはその抗原結合断片。
- 抗Cx43抗体またはその抗原結合断片であって、配列番号13、15、および17からなる群より選択されるアミノ酸配列を有する重鎖と、配列番号18のアミノ酸配列を有する軽鎖とを含む、抗Cx43抗体またはその抗原結合断片。
- Cx43に結合する場合、配列番号19のF1、S3、R4、P5、T6、E7、K8、T9およびI10からなる群より選択される1個またはそれを超えるアミノ酸を含むエピトープに結合する、請求項1~3のいずれか一項に記載の抗体またはその抗原結合断片。
- Cx43に結合する場合、配列番号19のF1、S3、R4、P5、T6、E7、K8、T9およびI10からなるエピトープに結合する、請求項1~3のいずれか一項に記載の抗体またはその抗原結合断片。

- 骨細胞におけるCx43ヘミチャネルの開放を促進するための方法における使用のための組成物であって、前記組成物は、請求項1~3のいずれか一項に記載の抗体またはその抗原結合断片を含み、前記方法は、前記骨細胞を前記抗体またはその抗原結合断片と接触させることを含むことを特徴とする、組成物。
