modRNAの同時トランスフェクションが、cmsnGFP modRNA単独でのトランスフェクションと比較して、CM増殖を増加させることを見出した(補足動画1)。さらに、cmsPkm2 modRNAは、MI後2日または7日目の心筋においてアポトーシスを有意に減少させ、毛細血管密度を増加させた(補足図7a~e)。MRIまたはエコーは、cmsPkm2が駆出率のパーセンテージを有意に増加させ(図3a~dおよび補足動画2および3)、MI後2日目(ベースライン)から28日目までに左室内径短縮率のパーセンテージデルタを増加させた(図3d)ことを示した。MI後28日目の対照と比較して、cmsPkm2マウスでは左心室内径の収縮末期が増加し、左心室の内径の拡張末期が有意に増加した(補足図7f~h)。MIの28日後、Pkm2またはcmsPkm2の発現は心瘢痕形成を有意に減少させた。さらに、cmsPkm2の注射後に心臓組織の異常(例えば血管腫、浮腫)は観察されず(図3eおよびf)、心臓重量対体重比は有意に増加した(図3g)が、CMサイズは有意に減少し、対照と比較して、Pkm2またはcmsPkm2 modRNAトランスフェクションにおいて、CMの増殖(図3hおよび補足図7i)、および毛細血管密度が有意に増加した(図3i)ことを示している。最後に、cmsPkm2は、対照と比較して単核画分を増加させながら、CMあたりの核数を上昇させることなく心臓においてCM数を有意に増加させた(図3j~m)。重要なことに、cmsLucまたはcmsPkm2 modRNAでMIの直後に処理したマウスの長期生存曲線は、MIおよびcmsPkm2トランスフェクション後のマウスの生存において有意な改善を示した(図3n)。cmsPkm2がMI後の心機能を改善するメカニズムを理解するために、cmsmodRNAとR26mTmGとを組み合わせて(図4a~j)、CMにおいてPkm2/Lucを排他的に発現する(cmsPkm2/cmsLuc+cmsCre modRNAを混合することによって;GFP標識CM、図4aおよびb)系統追跡モデルを使用し、cmsmodRNAがもはや発現されなくなった後、経時的にトランスフェクトされたCMの運命および特性を追跡する。cmsPkm2+Cre modRNAをトランスフェクトしたCMの数は、対照と比較して、MI後3日でより多く、MI後28日で有意に多かった(図4cおよびd)。cmsPkm2+Cre modRNAで処理したマウスでは、対照と比較して心臓重量対体重比は有意に増加し(図4e)、一方、GFP+CMサイズ(図4f)および核数/細胞(図4g)が有意に減少した。重要なことに、cmsPkm2+Cre modRNAでの処理の28日後、GFP+CMは、Pkm2が発現されなかった後長期に、pH3およびKi67などの増殖性マーカーの発現の上昇を示した(図4h~j)。MI状況におけるcmsPkm2またはcmsLucと併せたcmsihCD25 modRNAの送達後の遺伝子発現の変化を、注射の2日後に測定した。単離された細胞は、CMマーカーが豊富であり、トロポニンTの発現が有意に低かった(図4k)。Pkm2発現細胞は、その細胞質の下流のエフェクター(G6pd)および核(c-Myc、サイクリンD1、Bcl2、VEGF-AおよびPdk1)機能を有意に上方制御した(図4l)。増殖の増加に従って、Luc+ihCD25+CMと比較して、Pkm2+ihCD25+CMにおいて細胞周期促進遺伝子(Cdc20、Cdk1およびCcnd2、Ccnb1)の上方制御、ならびに細胞周期阻害因子(p21およびp27)の下方制御が観察された(図4m)。
mir-L7AEおよび5’UTRにkモチーフを担持する目的の遺伝子を同時にトランスフェクトする。これら2つのmodRNAにより、目的の遺伝子を特定の細胞型に特異的に送達することができる。
pTEMPLZは、目的のORFをUTR間に挿入できるクローニングベクターである。一実施形態では、開示の方法で使用するためのプラスミドは表1に示すものが挙げられる。
排他的にCMにおいて本発明者らの目的の遺伝子の一過性遺伝子発現を可能にする本発明者らの新規なCM特異的modRNAベースのデザインが、革新的な不活性(細胞外ドメインのみ)ヒトCD25(ihCD25)に基づく選別(磁気ビーズ)システムによるトランスフェクトCMのみの単離に使用できるかどうかを試験するために、MI後、心臓にnGFP kモチーフをihCD25 kモチーフmodRNAと共に注射し、細胞を単離し、CD25特異的磁気ビーズで選別した。nGFP陽性のCMおよび非CMの両方が観察された。nGFP kモチーフ、ihCD25 kモチーフとL7AE miR1-miR208が同時トランスフェクトされ、磁気分離されなかった場合、CM特異的nGFP発現が見られた。培養物はまた、nGFP陰性のCMおよび非CMを含む。磁気分離を適用した場合、純粋なnGFP陽性CMのみが観察された(図9)。
目的の遺伝子、例えば増殖誘導遺伝子の発現は、一対の核酸のうちの一方が、標的心筋細胞特異的miRに特異的に結合する抗マイクロRNA(抗miR)をコードする転写/翻訳調節システムの制御下に該遺伝子の転写/翻訳を置くことによって、心筋細胞特異的にすることができる。第1の核酸は翻訳抑制タンパク質であり、第2の核酸は翻訳抑制タンパク質と結合する抑制タンパク質相互作用モチーフおよび目的のタンパク質をコードする遺伝子を含む。
一実施形態では、本開示は、対象における心筋梗塞(MI)または心不全(HF)後の対象を治療する方法であって、少なくとも2つの合成modRNAを含む組成物の有効量をそれを必要とする対象に投与することを含む方法に関する。
伝統的なFACS細胞選別による心筋細胞の細胞選択は、細胞の大きさのために問題となり得る。細胞選択に対する別のアプローチが考案された。一実施形態では、本開示の転写調節システムを使用して、hCD25細胞外ドメイン(ECD)をコードする核酸が、目的の遺伝子をコードする核酸を含む構築物に含まれる。対照または候補遺伝子modRNAのいずれかを一過性に発現するCMを単離するために、hCD25 ECDおよび目的の遺伝子を同時発現する細胞を、アフィニティークロマトグラフィーカラムにおいて抗CD25 ECD抗体を使用して、またはパニング法を使用して選択する。これらの細胞を使用して、RNA-seq技術を用いた遺伝子発現プロファイルを作成し、差次的に発現された遺伝子を同定する。

以下の方法を、開示された方法と併せて使用する。特記しない限り、全ての手順は室温において、非無菌環境中で実施した。使用する全ての材料はヌクレアーゼフリーでなければならない。
ModRNAを、先に記載のように48-50アンチリバースキャップアナログ、3´-O-Me-m7G(5’)ppp(5’)G(6mM、TriLink Biotechnologies)、グアノシン三リン酸、(1.5mM、Life Technology)、アデノシン三リン酸(7.5mM、Life Technology)、シチジン三リン酸(7.5mM、Life Technology)およびN1-メチルプソイドウリジン-5’-三リン酸(7.5mM、TriLink Biotechnologies)のカスタムリボヌクレオチドブレンドを用いて、プラスミド鋳型からインビトロで転写した。mRNAを、Megaclearキット(Life Technology)を用いて精製し、南極ホスファターゼ(New England Biolabs)で処理し、続いてMegaclearキットを用いて再精製した。mRNAをNanodrop(Thermo Scientific)で定量し、エタノールと酢酸アンモニウムで沈殿させ、10mM TrisHCl、1mM EDTAに再懸濁した。詳しいプロトコルについては本発明者らの最近の出版物48を参照されたい。
pTEMPLZは、目的のORFをUTR間に挿入できるクローニングベクターである(図1)。5’-および3’-UTRは合成オリゴによって初めから合成する。合成したUTRを1つにアニールして、フォワードプライマーとリバースプライマーを用いて増幅する。ORFの挿入部位を提供するために、AleIおよびAfeI制限部位を5’と3’UTRの間に導入する(第1コドン(ATG)のアデニンヌクレオチド(A)はAleI部位によって提供されるので、フォワードプライマー配列から省略することができる)。PCR増幅断片およびアンピシリン耐性を有するpZErO-2ベクターをHindIIIおよびNotIで消化し、pTEMPLZを作製するために一緒に連結する(pTEMPLZプラスミドおよび誘導体は、One Shot(登録商標)ccdB Survival(商標)2 T1 Phage-Resistant (T1R)細胞などのccdB遺伝子産物に耐性の細菌株において増殖させるべきである)。

プラスミドの形質転換をコンピテント細胞を用いて行い、アンピシリン寒天プレート上で増殖させる。
1600μlのPCRマスター溶液を、以下の反応に従って調製した:

50μlのPCRマスター溶液を32本の別々のPCRチューブに等分した。PCRはサーモサイクラー(図2に記載されている設定)を使用して行う。伸長ステップの長さは、使用するDNAポリメラーゼおよびORFの長さに応じて異なる(例えば、2X KAPA HiFi HotStart ReadyMixを使用する場合、サーモサイクラーの伸長ステップはORFの長さ1Kbあたり30秒の比率で設定すべきである)。
品質管理分析のために、テールDNA鋳型産物の純度を、元のDNAプラスミドと共に1%アガロースゲルでチェックする(図3a)。
カスタムNTP混合物を表2に従って1本のEPPENDORF(商標)チューブ中で調製する。IVT反応用の試薬を、次の順序で1つのEPPENDORF(商標)チューブ中に混合する:
a.400μlの表2のカスタムNTP。
b.400μlのDNAテール鋳型(200ng/μl)。
c.T7 megascriptキットの10x反応緩衝液をボルテックス混合して、全ての沈殿物を溶解させ、100μlを加える。
d.100μlのT7酵素を加える。これにより1mlのIVT反応液が得られる
e.よく混合し、37℃のサーモミキサーにおいて4~6時間インキュベートする。
30μlのT7 Turbo DNaseを添加し、穏やかに混合し、次にサーモミキサー中で37℃において15~20分間インキュベート(ncubated)して反応を停止させた(注5参照)。
Ambion MEGAclear(商標)Transcription Clean-Upキットを用いて反応液を精製し、各チューブを50μlの95℃の溶出緩衝液で3回溶出し、各チューブ中に150μlのRNA産物を得る。各チューブのRNA混合液を1本のEPPENDORF(商標)チューブに組み合わせる。
ヌクレアーゼフリー水をRNAに添加して1.5mlの溶液を得る。150μlの南極ホスファターゼ緩衝液(×10)および150μlの南極ホスファターゼ酵素を添加し、十分に混合し、サーモミキサー中で37℃において1時間インキュベートする。
1800μlのRNA溶液を15mlのコニカルチューブに移す。180μlの5M酢酸アンモニウムを添加し完全に混合する。5200μlの冷たい(-20℃)100%エタノールを溶液に添加し、3~4個の2mlのEPPENDORF(商標)チューブに等分する。チューブを-20℃で一晩放置する。チューブを4℃において30分間10,000rpmで遠心分離する。上清を慎重に廃棄する。各ペレットを500μlの70%エタノールに溶解する。各チューブのmodRNAエタノール溶液を1本のEPPENDORF(商標)チューブにまとめる。チューブを4℃において30分間10,000rpmで遠心分離する。上清を静かに注ぎ出して廃棄し、キムワイプを使用して、チューブの内側を穏やかに清浄にする。ペレットを乱さないように注意を払う。チューブを逆さにしてペレットを風乾させるために2分以内放置する。ピペットを使用して、ペレットの周囲に残っているエタノールの小滴を全てそっと取り除く。ペレットを45~50μlの溶出緩衝液を用いて再懸濁する。modRNAを溶出緩衝液中に5分間放置し、次いでペレットが溶解するまで穏やかにピペッティングする。ここで、RNA溶液はインビボで使用することができ、-20℃で6ヶ月間、または-80℃で5年間保存可能である。
濃度はNanodrop機を用いて測定する(図3b)。A260/A280の比は1.8より大きくなければならず、2.0に近い値はより高い純度を示す。収量に応じて、濃度は20μg/μlに近いはずである。より良い品質管理分析のために、最終modRNA溶液からの1μlのサンプルを100μlのヌクレアーゼフリー水で希釈する。試料をバイオアナライザー機を用いて分析する(図3c)。
40μlのRNAiMaxを、5μlのOptiMEMとEPPENDORF(商標)チューブ内で混合し、ボルテックス混合する。混合物を室温で10分間静置する。別のEPPENDORF(商標)チューブ中で150~200ugのmodRNAを5μlのOptiMEMと混ぜ合わせる。チューブの側面の液体を排除するためにチューブを回転させる。RNAiMAXとOptiMEMの混合物を室温で10分間静置した後、modRNA混合物を含むチューブの液体をRNAiMAX混合物を含む試験管に加える。(いくつかの実施形態では、modRNA混合物をRNAiMAX混合物に加えることが重要であり、その逆ではない。)合わせた混合物を室温で15分間放置する。混合物を31ゲージのインスリン注射器に抜き取り、マウス心筋に注射する。(図4に示す結果の例)
動物の処理は全て、Icahn School of Medicine at Mount Sinai Institutional Care and Use Committeeによって承認されたプロトコルの下で実施した。CFW(Swiss Webster)マウスまたはRosa26mTmGマウスの雄および雌を使用した。ModRNAは、上記のようにインビトロ転写によって合成する。修飾ヌクレオチド(Trident)は、プソイドウリジン、5-メチル-シチジン、およびキャップ類似体である。RNAiMaxトランスフェクション試薬と複合体を形成した合計100~200μgの修飾RNAを、MIの誘発後の開胸手術において心筋の梗塞周囲領域に注射する。MRIは軽い麻酔下で行う(心拍数と鎮静レベルに対して滴定する)。LAD結紮および組織学的分析は、以前に記載されている通りに実施される(46)。各実験に3~8匹の動物を用いた。長期生存のために、CFW(8~10週齢)をMI誘導後にCM特異的LucまたはPkm2 modRNA(n=10)で処理し、動物施設で6ヶ月間回復させるために放置した。死亡を経時的にモニターし、記録した。
心臓を摘出し、Langendorff技術を用いて灌流し、CD25特異的磁気ビーズ(dynabeads CD25、Thermo Fisher Scientific)を用いて細胞をさらに処理し、RNeasyミニキット(Qiagen)を用いてRNAを細胞から単離する。RNAはさらにRNA-seqおよびRT-PCR分析に使用する。
図5に記載のMIモデルを使用して、CFWマウスまたはRosa26CD25マウスにおけるCM特異的Lin28、Pkm2を用いた処理後の治療効果を試験した。実験計画は、a)ビヒクルのみ、b)100μg/心臓の、KモチーフmodRNAを担持するLuc、c)100μg/心臓の、miR-1およびmiR-208認識エレメントの両方を担持するL7AE modRNA(L7AE miR1+miR208)およびd)KモチーフmodRNAおよびL7AE miR1+miR208を、各modRNAから100μg/心臓(合計200μg/心臓)担持するLucの混合物を含有するLuc CM特異的modRNAで処理した4つの対照群を含む。対照群は、細胞周期誘導CM特異的modRNAに直接関係しない、心臓におけるmiR-1およびmiR-208の減少の非特異的効果を評価するために機能する。本出願人らは、対照群と、KモチーフmodRNAを担持する100μg/心臓のa)Lin28およびb)Pkm2、ならびにKモチーフmodRNAを担持するd)Lin28およびe)Pkm2とL7AE miR1+miR208との混合物(細胞周期誘導CM特異的modRNA、各modRNAから100μg/心臓(合計でmodRNA200pg/心臓)を用いた4つの実験群とを比較した。MIの28日後にMRIを用いて心機能の改善を分析し、対照modRNAと比較して、高率の長期生存につながる瘢痕形成の減少および毛細血管密度の増加を分析した。さらに、MIモデルにおけるトランスフェクトCMの系統追跡モデルのためにRosa26Tdtomatoマウスを使用する。本発明者らの実験計画には、100pg/心臓のL7AE miR1+miR208と混合したCM特異的Luc(50pg/心臓)およびDD-Cre(50pg/心臓)で処理した1つの対照群が含まれる。対照群とCM特異的細胞周期誘導因子modRNAで処理した3つの実験群とを比較する。DD-Cre(50pg/心臓)およびKモチーフmodRNAを担持する細胞周期誘導遺伝子(Lin28およびPkm2 50pg/心臓)を100pg/心臓のL7AE miR-1+miR-208と一緒に含む混合物(合計200pg/心臓)を使用する。Rosa26Tdtomatoマウスの心臓に、合計100または200pgのmodRNAを筋肉内注射する。CM系統追跡モデルを用いて、免疫蛍光分析において抗小麦胚芽凝集素(WGA)抗体を用いたCM断面積評価を使用して、注射後28日目のトランスフェクトCMサイズを試験する。左心室あたりのトランスフェクトCMの数を計数し、各処理においてCMあたりの核の数を評価する。Rosa26Tdtomatoマウスを用いたこれらの試験は、CM特異的modRNAを用いた様々な処理後のCM機能の変化を経時的に評価可能にするものである。
Luc kモチーフ、Luc kモチーフ+miR1-208、miR1-208、Pkm2 kモチーフおよびPkm2 kモチーフ+miR1-208 modRNAで処理したCFWマウス(8週齢)を、LAD結紮後28日目にMRI評価に供した。11心臓および呼吸ゲーティングで7-T Bruker Pharmascan(SA Instruments、Inc、Stony Brook、New York)において遅延造影CINE画像を得た。マウスを1~2%イソフルラン/空気混合物で麻酔した。ECGプローブ、呼吸プローブ、および温度プローブをマウスに配置し、これによりスキャン中は温かく保った。0.3mmol/kgのガドリニウム-ジエチレントリアミン五酢酸のIV注射の10~20分後にイメージングを行った。心尖から心底までの心臓の8~10個の短軸スライスのスタックを、以下のパラメータを有するECG誘発および呼吸同期FLASHシーケンスを用いて取得した:エコー時間(TE)2.7ミリ秒、分解能200μm×200μm;スライス厚1mm;R-R間隔あたり16フレーム;フリップ角60°で4回の励起。駆出率は、拡張末期と収縮末期の体積の差を拡張末期の体積で割ったものとして計算した。MRIの取得および分析は処理群に対して盲検化して実施した。左心室収縮機能のエコー評価のために、40MHzマウス超音波プローブを備えたGE cares in site(V7R5049)を使用した。Mモード超音波から得られた拡張末期および収縮末期の寸法に基づいて、左室内径短縮率を計算した。心エコー図を6~8心臓/治療群について行った。
全RNAを、RNeasyミニキット(Qiagen)を用いて単離し、Superscript III逆転写酵素(Invitrogen)を用いて製造元の指示に従って逆転写した。リアルタイムqPCR分析は、SYBR Green(Quantitect(商標)SYBR Green PCRキット、Qiagen)を使用してMastercycler realplex 4 Sequence Detector(Eppendorf)で行った。必要に応じて、データを18s発現に対して正規化した(内因性対照)。遺伝子発現の倍率変化は∂∂CT法によって決定し、内部対照と比較して提示した。PCRプライマー配列を補足表3に示す。
Rosa26mTmGマウスはJackson Laboratoryから入手した。全ての実験は、年齢および性別が一致した、等しい比率の雄マウスと雌マウスで実施した。各実験について、増殖コロニーから健康なマウスを無作為に選択した。このマウス系統では、膜標的化tdTomatoはRosa26遺伝子座上の遍在性プロモーターの制御下で発現されるのに対して、膜標的化eGFPはfloxed tdTomatoのCre媒介性切断後に活性となる。CM特異的Cre modRNA(Cre Kモチーフ+miR1-miR208)を用いて、トランスフェクトCMにおいてCreを排他的に発現させた。これにより、modRNA発現が減少した後も長期に(>10日)、トランスフェクトCMおよびそれらの子孫の系統追跡が可能となった。Rosa26mTmGマウスを、Jackson Laboratory Genotyping Protocolsに記載されているように、テールDNAを用いたPCRによって遺伝子型決定した。プライマー配列は以下の通りである:Rosa26mT/mG、野生型フォワード、5’CTCTGCTGCCTCCTGGCTTCT-3’、野生型リバース、5’-CGAGGCGGATCACAAGCAATA-3’、および変異型リバース、5’-TCAATGGGCGGGGGTCGTT-3。このモデルにおいて、CM特異的LucまたはPkm2 modRNAのトランスフェクション後のCMにおいて、CM特異的Cre modRNAのトランスフェクションレベル、CMのサイズおよび数、ならびにCMの核数を測定した。
3~4日齢の新生仔ラットの心臓のCMを前述のように単離した1。新生仔ラットの心室CMは4日齢のSprague Dawleyラット(Jackson)から単離した。0.14 mg/mLのコラゲナーゼII(Invitrogen)による複数回の消化を使用した。各消化後、上清をウマ血清(Invitrogen)中に回収した。全細胞懸濁液を1500rpmで5分間遠心分離した。上清を廃棄し、細胞を0.1mMアスコルビン酸(Sigma)、0.5%インスリン-トランスフェリン-セレン(100×)、ペニシリン(100U/mL)およびストレプトマイシン(100μg/mL)を含むDMEM(GIBCO)培地に再懸濁した。ほとんどの非筋細胞が皿に付着し、筋細胞が懸濁状態になるまで、細胞をプラスチック培養皿に90分間プレーティングした。次いで筋細胞を24ウェルプレートに1×105細胞/ウェルで播種した。新生仔ラットCMを、5%ウマ血清+Ara cを含有するDMEM培地中で48時間インキュベートした。インキュベーション後、本文中に記載されているように、細胞に異なる用量の異なるmodRNAをトランスフェクトした。MIの28日後に成体CMをCFWマウスから単離し、前述のようにLangendorffの方法を用いてmodRNAを注射した2。CM数については、血球計を用いて3回の異なる計数/サンプルおよび3つの心臓/群を平均した。計数されたCMの総数は約150~200CM/アリコート(消化後に得られたCMの全体積からの広口径ピペットを用いた10μlのアリコートサンプル)であった。培養したCMを、核計数のためにα-アクチニン(CM、赤色)抗体(abcam)およびHoechst 33342で染色した。核計数については、1群あたり3~4個の独立した試料を用いて、1試料あたり約1×103個のCMを計数した。核計数を、計数したCMのパーセンテージとしてプロットした。トランスフェクト成体CMの単離とRNAの単離については、補足図8を参照されたい。
マウスを用いた全ての外科的および実験的手順は、Institutional Animal Care and Use Committees at Icahn School of Medicine at Mount Sinai Institutional Animal Care and Use Committee(IACUC)およびMSSM Center for Comparative Medicine and Surgery(CCMS)によって承認されたプロトコルに従って行った。CFW、R26mTmGマウス(6~8週齢)をイソフルランで麻酔した。前述のように、MIはLADの永久結紮によって誘発した3。簡潔には、左胸部を剃毛して滅菌した。挿管後、心臓を左開胸術により露出させた。縫合糸を配置してLADを結紮した。開胸および皮膚を層にして縫合して閉じた。過剰な空気を胸腔から除去し、通常の呼吸が確立されたときにマウスを換気から外した。MI後の心血管転帰に対するmodRNAの効果を決定するために、modRNA(100~150μg/心臓)を、LAD結紮直後に梗塞領域に注射した。心尖付近の梗塞周囲領域は、RNA単離およびその後のリアルタイムqPCR研究のために急速冷凍するか、または凍結切片化および免疫染色のために4%PFAで固定した。全ての実験において、外科医は治療群に対して盲検化された。心臓組織学の評価のために、各研究の終わりに心臓を収集した。心臓を摘出し、PBS中で簡単に洗浄し、灌流緩衝液で灌流し、秤量し、そして4%PFA中で4℃において一晩固定した。翌日、心臓をPBSで洗浄し、30%スクロース中で一晩インキュベートした。次に、心臓をOCTに入れ、凍結し、-80℃で保存した。クリオスタットを使用して心臓ブロックを8~9μmに横断切片化した。スライドをさらに免疫染色(下記参照)またはMassonのトリクローム染色キット(Sigma)を使用した組織学的瘢痕染色を用いて評価するために処理し、標準的な手順に従って行った。心臓重量対体重の比率の測定は秤を用いて行った。各実験の終点で比率を測定した。この比率は、マウスの全体重に対する心臓組織重量としてグラム(g)で算出した。
マウス心臓を摘出し、灌流緩衝液および4%パラホルムアルデヒド(PFA)を用いて灌流した。心臓を4%PFA/PBS中で一晩振盪機において固定し、次いでPBSで1時間洗浄し、30%スクロース/PBS中で4℃において一晩インキュベートした。翌日、心臓をOCTで固定し、-80℃で凍結した。横断心臓切片(8~10μM)をクリオスタットによって作製した。凍結切片をPBS中で5分間再水和した後、0.1%トリトンX100を含むPBS(PBST)で7分間透過処理した。次にスライドを3%H2O2で5分間処理した。PBSTで5分間ずつ3回洗浄した後、サンプルをPBS+5%ロバ正常血清+0.1%トリトン X100(PBSST)で室温において2時間ブロックし、PBSSTで希釈した一次抗体を加えた。次にスライドを4℃において一晩インキュベートした。スライドをPBSTで(4分間ずつ5回)洗浄し、続いてPBSTで希釈した二次抗体(Invitrogen、1:200)と共に室温において2時間インキュベートした。サンプルをさらにPBSTで洗浄し(5分間ずつ3回)、PBSTで希釈したHoechst 33342(1μg/ml)で7分間染色した。PBSTでそれぞれ4分間、および水道水で1回(4分間)5回洗浄した後、スライドをイメージング用の封入剤(VECTASHIELD)で封入した。染色スライドを4℃で保存した。全ての染色は、3~8心臓/群、2~3切片/心臓で行った。CMサイズの定量化のための小麦胚芽凝集素(WGA)による免疫染色の場合、20倍の倍率で画像を捕捉し、ImageJを使用して各細胞の面積を決定した。定量分析は、1群あたり3~6個の独立した心臓、および3切片/心臓(評価された1視野あたり約50細胞、サンプルあたり合計約250細胞)からの複数視野の計数を含んだ。BrdU免疫染色のために、心臓を摘出する前に、BrdU(1mg/ml、Sigma)を7~10日間、成体マウス(2~3ヶ月齢)の飲料水に添加した。定量分析は、1群あたり3つの独立したサンプル、および3切片/心臓からの複数視野におけるBrdU陽性CMを計数することを含んた。カウントされたCMの総数は、1切片あたり約1~2x103CMであった。TUNEL免疫染色は、製造業者の推奨に従って行った(インサイチュ細胞死検出キット、フルオレセイン、カタログ番号11684795910、Roche)。modRNA処理後の新生仔CMの免疫染色のために、modRNAをトランスフェクトした新生仔CMを3.7%PFAを有するカバーガラス上に室温で15分間固定した。PBS中0.5%トリトンXを用いて室温で10分間透過処理した後、細胞を5%正常ヤギ/ロバ血清+0.5%ツイーン20で30分間ブロックした。カバーガラスを一次抗体(補足表1参照)と共に湿潤チャンバー内で室温において1時間インキュベートし、続いてAlexa Fluor 488、Alexa Fluor 647およびAlexa Fluor 555にコンジュゲートした対応する二次抗体とインキュベートし、核を可視化するためHoechst 33342染色した。(全てInvitrogeneから)。蛍光画像は、10倍、20倍および40倍の倍率でZeiss蛍光顕微鏡で撮影した。
nGFP CM特異的modRNAのトランスフェクション後またはnGFPおよびCM特異的Pkm2 modRNAの同時トランスフェクト後の単離ラット新生仔心筋細胞の経時的画像を、10秒ごとに10倍対物レンズで共焦点回転ディスク顕微鏡(Zeiss)を用いて、その後24時間の低速度撮影で取得した。
統計的有意性は、MRI結果については対応のあるt検定、生存曲線についてはログランク(マンテル-コックス)検定、またはそれぞれの図の説明に詳述するように、他の実験についてはスチューデントt検定、一元配置分散分析、ボンフェローニ事後検定によって決定した、*p<0.05以下を有意とみなす。全てのグラフは平均値を表し、値は平均値±平均値の標準誤差として報告した。両側スチューデントのt検定は、仮定された正規分布に基づく。CD31内腔構造、WGA、CD45、CD3、TUNEL、BrdU+、ki67+、pH3+またはAurora B+CMの数の定量化については、結果は少なくとも3つの心臓切片から得た。
