Description

本発明は、微生物を提供し、加えて、バイオサーファクタントなどの、それらの増殖時の副生成物も提供する。本発明はまた、これらの微生物およびそれらの副生成物を用いる方法を提供し、加えて、それらを生産するための方法およびシステムをも提供する。有利なことに、本発明である微生物ベースの生成物および方法は、環境に優しく、非毒性であり、かつ費用対効果が高い。
本明細書において使用される場合、「微生物ベースの組成物」への言及は、微生物または他の細胞培養物の増殖の結果として産生された成分を含む、組成物を意味する。したがって、微生物ベースの組成物は、微生物それ自体、および/または微生物の増殖時の副生成物を含み得る。微生物は、生長能力のある(vegetative)状態、胞子の形、菌糸体の形、繁殖体の任意の他の形、またはこれらの混合物であり得る。微生物は、浮遊性であってよく、もしくはバイオフィルムの形であってもよい、または両方の混合物であってもよい。増殖時の副生成物は、たとえば、代謝物、細胞膜の成分、発現されたタンパク質、および/または細胞の他の成分であり得る。微生物は、インタクトなものであってよく、または溶解されてもよい。いくつかの態様において、微生物は、そこでそれらが増殖した培地とともに、微生物ベースの組成物中に存在する。細胞は、たとえば、1ミリリットルの組成物あたり1 x 104の、1 x 105の、1 x 106の、1 x 107の、1 x 108の、1 x 109の、1 x 1010の、1 x 1011の、1 x 1012の、もしくは1 x 1013の、またはより大きいCFUという濃度で、存在し得る。
1つの態様において、殺真菌性組成物が提供され、これは、1つもしくは複数の酵母株、および/またはそれらの増殖時副生成物を含む。好ましい態様において、1つまたは複数の酵母株は、ピキア属クレードの酵母から選択される。いっそうより好ましくは、酵母株は、ピキア・オキシデンタリス、およびピキア・アノマラ(ウィッカーハモマイセス・アノマルスとしても知られる)から選択される。
本発明は、微生物の培養のための、ならびに微生物代謝物および/または微生物の増殖時の他の副生成物の生産のための方法を利用する。本発明は望ましいスケールでの、微生物の培養、および微生物代謝物の生産に適した培養プロセスを、さらに利用する。これらの培養プロセスは、液内培養/液内発酵、固相発酵(SSF)、ならびにそれらの改変物、それらの混成物、および/またはそれらの組み合わせを含むが、それらに限定されるものではない。
本発明のシステムおよび方法によって増殖される微生物は、たとえば、細菌、酵母、および/または真菌類であり得る。これらの微生物は、天然の微生物であってよく、または遺伝的に改変された微生物であってもよい。たとえば、微生物は、特定の特徴を示す特定の遺伝子を用いて、形質転換されてよい。微生物はまた、望ましい株の変異体であってもよい。本明細書において使用される場合、「変異体」とは、基準となる微生物の、株、遺伝的バリアント、またはサブタイプを意味し、ここで変異体は、基準となる微生物と比べて、1つまたは複数の遺伝的な変化(たとえば、点変異、ミスセンス変異、ナンセンス変異、欠失、重複、フレームシフト変異、またはリピート伸長)を有する。変異体を作製するための手順は、微生物学の技術分野において周知である。たとえば、UV変異導入およびニトロソグアニジンが、この目的に向けて広く使用される。
本発明の微生物ベースの生成物の1つ(たとえば土壌処理組成物)は、単に、微生物、および/または微生物によって産生された微生物代謝物、および/または任意の残存栄養素を含む、発酵培地である。発酵の生成物は、抽出も精製も無しに、直接的に使用され得る。望ましい場合、抽出および精製は、標準的な抽出方法および/もしくは精製方法を用いて、または文献に記載される技術を用いて、容易に達成され得る。
1つの態様において、方法は、植物および/もしくは植物の周辺環境内で、または植物および/もしくは植物の周辺環境上で、フザリウム属菌を制御するために提供され、ここで1つもしくは複数の非病原性微生物、および/またはそれらの増殖時副生成物は、植物および/または周辺環境に接触する。ある態様において、フザリウム属感染は、植物内および/もしくは周辺環境内で、もしくは植物上および/もしくは周辺環境上で成立する、またはすでにそこに存在している。有利なことに、組成物は、植物毒性を引き起こすこと無しに、非休眠状態の植物組織に適用され得る。
本発明は、植物において病気を引き起こす真菌感染の防止および/または処置において、有用であり得る。好ましい態様において、真菌感染は、フザリウム属菌によって引き起こされる。
本発明のある態様において、微生物の増殖設備は、関心対象の、新鮮な、高密度の微生物、および/または微生物の増殖時副生成物を、所望のスケールで生産する。微生物の増殖設備は、適用の場所に、またはその近くに設置され得る。設備は、高密度の微生物ベースの組成物を、バッチ培養、準連続的な培養、または連続的な培養において生産する。
バイオマスの産生のために、コメをベースとする培地が使用される。およそ200グラムのコメが、600 mlのGUY培地(グルコース、尿素、および酵母抽出物、pH 5.71)または250 mlの濃縮GY培地(グルコースおよび酵母抽出物、pH 5.69)ならびに水と混合される。培地は、ステンレススチールの皿に、約1~2インチの厚さの層として広げられ、そして滅菌される。
発酵の温度およびpHは厳密ではないが、一般的に温度は、約25℃と約37℃未満との間、好ましくは約25~約30℃の間とすべきである。pHレベルは、約3.0~約5.0の間、好ましくは約3.5~約4.5の間の範囲とすべきである。発酵中のpHの安定化は必須ではないが、それが3.0を下回る場合には、pHを3.5~4.0に上げることが推奨される。
いくつかの処理が、ATCCから得られたフザリウム・オキシスポラムの増殖を阻害する能力について試験された。処理は、ピキア・アノマラ、ピキア・アノマラ変異体株(UV光のもとで30分、そしてさらに選択)、1% SLP処理、およびピキア・オキシデンタリスを含んでいた。
トリコデルマ属、P. アノマラ、SLP、変異体P. アノマラ、およびP. オキシデンタリスを含む、すべての処理は、F. オキシスポラムに対して阻害効果を有していた。トリコデルマ属は、F. オキシスポラムと、資源に関して競合し、そしてしたがって、その将来的な増殖を減速させたが、F. オキシスポラムそれ自体に対しては、直接的な効果は有していなかった。
10% SLPを用いて処理された培養物の色はほぼ白になり、そしてF. オキシスポラムの菌糸は、SLPによって破壊された。1% SLPを用いた3回の処理は、菌糸の破壊を引き起こした、図4A~4B、しかし、胞子に対する強力な効果は有さなかった。それは、P. アノマラよりもわずかに優れて作用した。
P. アノマラは、F. オキシスポラム菌糸の細胞壁を破壊する物質を分泌するように見受けられた。それは、菌糸に対して強い阻害効果を有していた。図5~7。変異体P. アノマラは、類似の効果を示したが、いくぶん、より急速かつより強力であった。図8A~8B。
P. オキシデンタリスは、フザリウム属に対して、菌糸および胞子の両方において、最も強い死滅効果を示した、それは、顕微鏡下、かつ寒天プレート上で、調べられた。それは、2つの異なるタイプの阻害分子を分泌しているように見受けられた:その1つは、フザリウム属の細胞壁を破壊し、そしてもう1つは、胞子からの、新しい菌糸の形成および発達を阻害した(図9~12)。
Claims (6)
- ピキア属種(Pichia sp.)の酵母および/またはその増殖時副生成物を含む殺真菌性組成物を植物が生育する土壌に適用する段階を含む、ムサ属種(Musa sp.)またはコフィア属種(Coffea sp.)植物においてフザリウム属種(Fusarium spp.)感染を制御および/または防止する方法であって、
該ピキア属種の酵母が、ピキア・オキシデンタリスおよびピキア・アノマラより選択され、
ピキア・オキシデンタリスとピキア・アノマラとの組み合わせが適用される、該方法。 - 前記殺真菌性組成物が、灌水システムを用いて前記土壌に適用される、請求項1に記載の方法。
- 前記植物が、フザリウムウィルト(Fusarium wilt)またはパナマ病によって影響を受ける、請求項1に記載の方法。
