Description
腫瘍内皮細胞(TEC)は、正常内皮細胞とは異なり、血管新生促進表現型(pro-angiogenic phenotype)を呈する。例えば、TECは、in vitroにおいて血清とは無関係な高い運動および増殖、ならびにAktシグナル伝達を介した生存率の向上を実証している。表現型の上では、TECは、VEGFおよびEGF受容体を含む増殖因子受容体の発現を向上させることがあった。TECは、ある化学療法薬物にも耐性であり、VEGFを標的とする抗血管新生薬にさほど感受性ではない。さらに、TECは、遺伝子学的に正常内皮細胞と異なる。
細胞外小胞(EV)は、細胞間コミュニケーションの重要な機構と思われ、その活性カーゴは、レシピエント細胞の機能および表現型を改変することにより、それを再プログラムし得る。実際に、EVの活性は、タンパク質、RNA、DNAおよび脂質を含むいくつかの異なる因子の移動にリレーするとみられ、中でもマイクロRNAは、主要な役割を有すると思われる。幹細胞に由来するEV、詳細にはヒト骨髄由来間葉系間質細胞(BM-MSC)は、腫瘍型および発現段階に応じて、腫瘍形成促進活性(pro-tumorigenic activity)および抗腫瘍形成活性の両方を呈することが公知である。同様に、MSC-EVも、腫瘍の血管形成を陽性にも、または陰性にも調整し得る(Zhu W, Huang L, Li Y, Zhang X, Gu J, Yan Y, et al. Exosomes derived from human bone marrow mesenchymal stem cells promote tumor growth in vivo. Cancer letters. 2012;315(1):28-37)。MSC-EVは、腫瘍を抱えるマウスへの投与後に、in vivoで血管新生促進性になると報告された(Zhu W, Xu W, Jiang R, Qian H, Chen M, Hu J, et al. Mesenchymal stem cells derived from bone marrow favor tumor cell growth in vivo. Exp Mol Pathol. 2006;80(3):267-74)。他の研究では、MSC-EVの、腫瘍細胞によるVEGF分泌に対する間接的効果が観察された。
HLSCは、国際公開第2006/126236号に記載されている。
さらに、本発明者らの知る限りでは、腎癌に対して、特定のmiRNAの送達による、幹細胞に由来するEVの抗腫瘍効果は、これまで報告されていない。
薬学的効果は、薬学的キャリアに含有されるmiRNAに起因し得る。miRNAを用いた標的細胞の任意の効率的トランスフェクションは、腎癌の処置における有効な使用のために想定される。miRNAの効率的トランスフェクションは、好ましくはマイクロまたはナノ粒子の形態の適切な薬学的キャリアを必要とする。そのようなキャリアは市販されており、アルギネートベース(GEM、Global Cell Solutions)、デキストランベース(Cytodex、GE Healthcare)、コラーゲンベース(Cultispher、Percell)およびポリスチレンベース(SoloHill Engineering)微小キャリアを含む。
miRNAにさらに好ましい薬学的キャリアは、小胞、例えばリポソームまたは細胞外小胞(EV)である。細胞外小胞、例えば細胞由来微小胞またはエキソソームが、最も好ましい薬学的キャリアである。
したがって、本発明の別の好ましい実施形態によれば、薬学的に許容できるキャリアは、幹細胞に由来する、好ましくは成体幹細胞からの、より好ましくは間葉系幹細胞(MSC)、例えば骨髄間質幹細胞からの、もしくは脂肪由来幹細胞(ADS)からの、または非卵形ヒト肝臓前駆細胞(HLSC)からの細胞外小胞(EV)である。HLSCおよびそれを得る方法は、国際公開第2006126219号として公表されている国際特許出願で開示されている。
したがって、本発明に従って使用するための細胞外小胞(EV)は、単離した、天然に存在するEV、あるいは、miR-15a、miR-181b、miR-320c、miR-874およびそれらの任意の組合せからなる群から選択される1つまたは複数のマイクロRNAを含有するように操作されたEVである。
本発明に従って使用するための細胞外小胞(EV)は、天然に存在するEV、あるいは、天然に存在する細胞外小胞(EV)と比較して著しく多量の、miR-15a、miR-181b、miR-320c、miR-874およびそれらの任意の組合せからなる群から選択される1つまたは複数のマイクロRNAを含有するように操作されているEVである。操作されたEVは、miR-15a、miR-181b、miR-320c、miR-874およびそれらの任意の組合せからなる群から選択される1つまたは複数のマイクロRNAを、単離した細胞外小胞にex vivoで負荷することにより得ることができる。別法として、操作されたEVは、上で定義されている幹細胞にmiRNAをトランスフェクトすること、次いでトランスフェクトされた幹細胞の馴化培地からEVを単離し精製することにより得ることができる。
天然に存在する細胞外小胞(EV)と比較して著しく多量のmiRNAを見積もる好適な方法は、qPCRデータ分析のΔΔCT法である。
MSCおよびHLSCに由来するEVを用いて本発明者らにより実行された実験を開示する以下の実験の部は、例示の目的でのみ示されており、添付の特許請求の範囲により決定される本発明の範囲を限定することを意図していない。
材料および方法
細胞培養
TECは以前に、本発明者らの研究室において、腎臓癌患者の切除標本4例から単離され、培養されている。TECは、磁気細胞選別(MACSシステム、Miltenyi Biotech、Auburn、CA)により、抗CD105ポジティブ選択を使用して、消化された組織から単離し、記載されているように(Bussolati B, Deambrosis I, Russo S, Deregibus MC, Camussi G. Altered angiogenesis and survival in human tumor-derived endothelial cells. FASEB journal, official publication of the Federation of American Societies for Experimental Biology. 2003;17(9):1159-61)、EndoGro完全培地(Millipore)において成長させた。
MSCは、Lonzaから購入し、MSCBM完全培地(Lonza)中で培養した。
EV単離は、改変をわずかにして、以前に記載されている(Herrera MB, Fonsato V, Gatti S, Deregibus MC, Sordi A, Cantarella D, et al. Human liver stem cell-derived microvesicles accelerate hepatic regeneration in hepatectomized rats. Journal of cellular and molecular medicine. 2010;14(6B):1605-18)ように行った。簡潔には、HLSCまたはMSCの密集培地は、FBSを含まないRPMIに18時間変えた。翌日、この培地は、3000gで30分遠心分離して、細胞デブリおよびアポトーシス小体を除去した。その後、上澄みは、Beckman Coulter Optima L-100K Ultracetrifugeとロータータイプ45 Ti、45000RPMを使用して、100000gで2時間、超遠心分離にかけた。EVペレットは、10%のDMSOを補充したRPMIに再懸濁した。HLSC-EVの懸濁液は、次いで、使用するまで-80℃にて冷凍した。EVは、NTA分析および電子顕微鏡法を使用して分析した。EVの平均径は、90nm(±20)であった。いくつかのin vivo撮像実験のために、EVは、1μM Vybrant Cell Tracers DiD(Ex:640nm、Em:700nm)または血清なしのDil溶液(Ex:530nm、Em:580nm)(Molecular Probes、Oregon、USA)により標識し、次いで、PBS 1×40中での超遠心分離により2回洗浄した。
増殖のテストでは、TECは、2×103個/ウェルの密度で96ウェルプレートにシードした。翌日、細胞は、EndoGro完全培地(Lonza)中1×1010または5×1010または10×1010個のEV/TECの濃度で、HLSC-EVまたはMSC-EVを用いて処理した。増殖は、細胞増殖ELISA、BrdU(比色分析)キット(Roche、11647229001)を製造者の指示に従って使用したEV刺激後の24、48および72時間にて、BrdUの組み込みにより測定した。移行テストは、24ウェルプレートにシードしたTECで行い、密集するまで成長させた。EVを、スクラッチ直後に1×1015または5×1015または10×1015個のEV/ウェルの濃度で添加した。顕微鏡での優れた10×の撮像は、スクラッチ後0、3、7および24時間で行った。距離は、LASソフトウェア(Leica)により測定した。結果は、平均放射輝度±SDとして提示する。
TECを、Matrigelでコーティングした24ウェルプレートに、ウェル当たり25×103個の細胞の密度でシードし、1×1010、5×1010、10×1010または20×1010個のEV/TECの存在下で、EndoGro完全培地において培養した。EVなしのTECを対照として使用した。24時間のインキュベーション後、位相差像(倍率×10)を記録し、LASソフトウェア(Leica)を使用して、ネットワーク構造の全長を測定した。フィールド当たりの全長を5つのランダムなフィールドで計算し、それぞれの対照に対する比として表現した。データは、平均放射輝度±SDとして表現した。
動物研究は、国のガイドラインおよび規制に従って実施し、University of Torinoの治験審査委員会の承認を受けた(プロトコールナンバー:338/2016- PR)。Matrigel内にTECを注入することにより得られたin vivoにおける腫瘍の血管新生のモデルを使用して、記載されている(ref FASEB 2003)ように幹細胞に由来するEVの効果を見積もった。腫瘍の血管新生発現を防止するために、TECは、前処理してから注入した。この目的のために、SCIDマウス(6~8週齢)(Charles River Laboratories、Lyon、France)に、HLSC-EV/MSC-EVで前処理した、またはしていないMatrigel内の1×106個のTECを皮下注入した(細胞当たり10×103個のEV):(対照ではn=8、それぞれEV処理)。7日後、Matrigelプラグを切除し、血管密度をMassonの三色反応で分析した。EVの、確立された腫瘍血管に対する影響を評価するために、1×106個のTECをSCIDマウスのMatrigel内に皮下注入した。HLSC-EV(細胞当たり10×103個のEV)を、TEC注入から3日後および7日後に、Matrigelプラグに2回注入した。対照マウスは、ビヒクル(PBS)を注入した。実験の10日目に、マウスを屠殺し、組織化学的分析のためにMatrigelプラグを切除した(対照ではn=8、HLSC-EV処理)。
TECによるEV取込みをin vivo撮像するために、SCIDマウス25匹に、Matrigel内の1×106個のTECを皮下注入した。腫瘍血管が発達した(1週間)後、マウスを、ビヒクル(PBS)を受けた対照、DiL MSC-EV、DiD MSC-EV、DiL HLSC-EVおよびDiD HLSC-EVの5群に分けた(n=それぞれ5)。DiDまたはDiLで標識したEV(1.3×1010個のEV/マウス)を静脈内注入し、5時間後、マウスを屠殺した。DiL EVで処理したマウスから得た器官(Matrigelプラグ、皮膚、腎臓、脾臓、肝臓および肺)を、免疫蛍光のために回収した。各器官の冷凍切片を、核対比染色のためにDapiで染色し、共焦点顕微鏡法により分析して、DiL標識EVを検出した。
HLSC-EVまたはMSC-EVにおけるmiRNA発現レベルは、Applied Biosystems TaqMan(登録商標)アレイヒトマイクロRNA A/Bカード(Applied Biosystems、Foster City、CA)を使用して分析して、qRT-PCRにより754種の成熟miRNAの特性を明らかにした。キットは、マイクロRNA特異的ステムループ逆転写プライマーおよびTaqManプローブを使用して、2-ステップリアルタイム逆転写PCRアッセイで成熟miRNA転写産物を検出した。簡潔には、一本鎖cDNAは、全RNA試料(80ng)から、ループプライマー(マルチプレックスRTキット、Applied Biosystems)の混合物を製造者のプロトコールに従って使用した逆転写により生成した。RT反応を、次いで希釈し、Taqman universal master Mix(Applied)と1:1比で混合し、TaqManマイクロ流体カードに負荷し、qRT-PCR実験を行った。すべての反応は、384ウェル反応プレートを備えたApplied Biosystems 7900HTリアルタイムPCR機器を使用して行った。生Ct値は、SDSソフトウェアバージョン2.3を使用し、自動ベースラインおよび閾値を使用して計算した。筆者らは、3例のHLSC-EV試料でmiR発現を分析した。35サイクルのPCR後に増幅したマイクロRNAは、すべて非発現として分類した。検出された、または2例を超える試料で検出されていないマイクロRNAのみを考慮に入れた。
TECのトランスフェクションは、HiPerfect試薬(Qiagen)を使用して行った。理想的なトランスフェクション濃度を見出すために、TECに、模倣miR-FITCおよびHiPerfect試薬の推奨される濃度すべてをトランスフェクトした。HiPerfectの最大用量(50×103個のTEC当たり9μlのHiPerfect)の倍増を、同様に行った。FACS分析をトランスフェクションの翌日行い、この分析により、HiPerfectの最大用量の倍は、TECの60%超に生存能力および増殖にダメージを与えずにトランスフェクトできたことが明らかになった。この用量を、すべてのトランスフェクション実験に使用した。
HLSC-EVおよびMSC-EVのFACS分析は、CytoFLEXフローサイトメーター(Beckman Coulter)を使用して行った。使用される抗体は、FITC-結合体化抗体抗CD63(Abnova)、抗CD105(Dako Cytomation、Copenhagen、Denmark)、抗CD90(BD Pharmigen)、抗CD44(Miltenyi Biotech)、CD45(BD Pharmigen)、抗ICAMおよび抗VCAM(Serotec)、CD31(BioLegend)、インテグリンサブユニットα4、α5、α6(BD Pharmigenから);PE-結合体化抗体抗CD73(BD Pharmigen)、抗インテグリンサブユニットα4、α5(すべてBD Pharmigenから)およびVE-カドヘリン(BioLegend)であった。FITCまたはPEマウス非免疫アイソタイプIgG(Dako Cytomation)を対照として使用した。
タンパク質試料は、4%~15%勾配硫酸ドデシルナトリウム-ポリアクリルアミドゲル電気泳動(SDS PAGE)により分離し、PLAU(Abcam、ab131433)またはFGF1(Abcam ab9588)に対する抗体を用いて、免疫ブロッティングを施した。タンパク質バンドは、増強化学発光(ECL)検出キットおよびChemiDoc(商標)XRS+System(BioRad)で視覚化した。20μg/ウェルの細胞溶解物を負荷した。
データは、Kolmogorov-Smirnovテストを使用して、分布の正常性について見積もった。統計分析は、SigmaPlot 11.0ソフトウェアを使用して行った。処理群と対照群との間の差は、分布が正常である場合、Studentのt-検定を使用して分析した。データは、平均±SEMとして表現した。差は、p<0.05の場合、有意とみなした。
HLSC-EVは、in vitroでの腎臓TECの血管新生能力、および移行を阻害する
MSCEVおよびHLSC-EVのTECに対する影響は、増殖、アポトーシスおよび血管様構造形成のin vitro評価により見積もった。HLSC-EVの刺激により、用量依存手段で、in vitroでのTECの血管新生が有意に阻害され、TEC当たり10×103個のEVの添加により、血管様構造形成が37%減少し、TEC当たり20×103個のEVの用量では、血管様構造形成が44%減少した(図1、A、B、D)。MSCEVおよびHLSC-EVのいずれも、TECの生存能力を変化させなかった(示されていないデータ)。MSC-EVおよびHLSC-EVの、TECの運動に対する効果も、創傷治癒アッセイにより評価した。いずれのEVも、TECの移行を有意に阻害し、HLSC-EVは、TEC当たり1×103個の用量で早くも有効となった(図1、E)。
対照実験では、MSC-EVおよびHLSC-EVの、正常内皮細胞に対する効果を評価した。MSC-EVは、ヒト微小血管内皮細胞(HMEC)の血管新生特性を向上できるが、HLSC-EVは、効果を一切示さなかった(図1、F)。これにより、MSCおよびHLSCからのEVは、正常な血管新生および腫瘍の血管新生に対して、異なる作用を有することが示される。

MSC-EVおよびHLSC-EVの効果は、SCIDマウスに皮下移植された場合、Matrigel内のTEC組織により誘導されるヒト腫瘍の血管新生モデルを使用することによりin vivoで評価した。このモデルでは、TECは、マウスの血液循環に接続した構造において7日以内に組織化する。前処理設定では、TECをHLSC-EVまたはMSC-EVで24時間処理し、SCIDマウスに皮下移植した。埋め込んでから7日後、Matrigelプラグを切除し、血管密度を免疫組織化学的に分析した。対照プラグの分析から、予想通り、マウスの血管系に接続した血管の存在が示された(図2、A)。埋め込み前にHLSC-EVで24時間処理したTECのプラグは、赤血球を含有する血管を提示しなかった(図2B、D)が、MSC-EVで前処理したものは、効果を一切示さなかった(図2C、D)。
HLSC-EVは、腫瘍の血管新生を防止できるので、確立された腫瘍の血管に対するその効果も評価した。この目的のために、HLSC-EVを、TECを埋め込みから3日後および7日後にMatrigelプラグ中に注入し、プラグを10日後に回収した。HLSC-EVの処理は、血管密度をほぼ50%有意に低下させた(図2、E)。

in vivoにおいて、既に血管へと組織化したTECがEVを取り込む能力を評価するために、標識したMSC-EVおよびHLSC-EV(1.3×1010個のEV/マウス)を静脈内注入した。EVを、蛍光色素、光学撮像の場合は近赤色(DiD)、また、免疫蛍光の場合は赤色(DiL)で標識した(方法を参照されたい)。5時間後、マウスを屠殺し、器官を回収した。EVの用量および時間を、予備実験(示されていない)に基づき選択した。光学撮像で、筆者らは、in vivoでMatrigel内に注入されたTECが、図3AおよびCで示されているように、両方のタイプのDiD標識EV(MSC-EVおよびHLSC-EV)を取り込めることを実証した。対照的に、プラグの近くで単離した皮膚組織では、きわめて蛍光性が低いシグナルが生じた(図3BおよびC)。すべての器官内における生体内分布に関して、EVは、主に肝臓内に蓄積した。

これらの結果に基づき、HLSC-EV刺激後、in vitroにおける血管様構造の組織化中に、TECにおいて発生する変化の分子分析は、血管新生アレイを使用することにより行った。TECは、HLSC-EV(10×103個のEV/TEC)で処理し、血管新生アッセイからの伸展細胞(spreading cell)を採取した。テストした84種の遺伝子のうち、筆者らは、in vitroでの血管新生中に、HLSC-EVによりTECにおいて有意に下方調節された11種の血管新生促進因子を同定した(図5、A)。詳細には、TECは、Tie-1、ベータ3インテグリン(ITGB3)、エフリン受容体B4(EPHB4)およびエンドグリン(またはCD105))を含む血管新生促進表面受容体、ならびに、増殖因子、例えばFGF1、TGFファミリーメンバー、ウロキナーゼ-型プラスミノーゲンアクチベーター(PLAU)および組織因子(F3)を下方調節した。最終的に、TECの血管新生促進効果に関与することが公知のAkt1も下方調節された。

続いて、観察された遺伝子調節の考えられるエフェクターを詳細に分析するために、関与すると考えられるHLSCのマイクロRNA含有量を研究した。この目的のために、バイオインフォマティクス分析の方略、続いてin vitro機能確認を使用した。
これら別々のmiRNAの、TEC血管新生特性に対する特異的効果を実証するために、TECに、選択された模倣体をトランスフェクトした。トランスフェクションの2日後、血管様構造形成のin vitroアッセイを行った。4種のmiRNA、miR-15a、miR-181b、miR-320cおよびmiR-874は、in vitroにおいて血管様構造形成を有意に阻害した(図6、A)。模倣体の効果は、増殖またはアポトーシスで観察されなかった(図6、BおよびC)。

したがって、本発明者らは、4種の活性miRNA模倣体を用いたトランスフェクションが、予測された標的(EPHB4、ITGB3、FGF1およびPLAU)の発現をどのように変化させたかを調査した。TECトランスフェクションは、その標的を有意に下方調節した(表1、図7)。表1.選択されたmiRNAを用いてトランスフェクトしたTEC対対照TECにおける血管新生促進遺伝子の相対発現。
これらのmiRNAの発現から、HLSC-EVで処理したTECにおいて有意に向上したことが見出され(図8、A)、in silico データの正当性を指し示した。並行して、模倣体のHLSC-EVの4種の標的遺伝子に対する効果(EPHB4、ITGB3、FGF1およびPLAU)をRNAおよびタンパク質レベルの両方で評価した。
TECは、HiPerfect試薬(Qiagen)を使用し、以下の組合せのmiRNA:miR-874/miR-15a;miR-874/miR-181b;miR-847/miR-320c;およびmiR181c/miR-320を使用してトランスフェクトした。トランスフェクションの2日後、血管新生in vitroアッセイを行った。miRNAのすべての組合せが、TECの血管新生促進特性を有意に低下させた。さらに、miRNAの組合せは、単一のmiRNAより有効であった。
Claims (6)
- miR-15a、miR-181b、miR-320c、miR-874およびそれらの任意の組合せからなる群から選択されるマイクロRNAを含む薬学的に許容できる担体を含む、腎癌の抗血管新生治療における使用のための組成物。
- miR-15a、miR-181b、miR-320cおよびmiR-874からなる群のうち少なくとも2つの組合せを含む、請求項1に記載の組成物。
- 薬学的に許容できる担体が、マイクロまたはナノ粒子であり、1以上のマイクロRNAが、マイクロもしくはナノ粒子の内側に含有されているか、またはマイクロもしくはナノ粒子の表面に付着している、請求項1または2に記載の組成物。
- 薬学的に許容できる担体が、非卵形ヒト肝臓前駆細胞(HLSC)由来の細胞外小胞(EV)である、請求項1~3のいずれか1項に記載の組成物。
- 薬学的に許容できる担体が、miR-15a、miR-181b、miR-320c、miR-874およびそれらの任意の組合せの群から選択されるマイクロRNAを含有するように操作されている、成体幹細胞に由来する細胞外小胞(EV)である、請求項1~3のいずれか1項に記載の組成物。
- 細胞外小胞(EV)が、間葉系幹細胞(MSC)および脂肪由来幹細胞(ASC)からなる群から選択される成体幹細胞に由来する、請求項5に記載の組成物。
