<発明を実施するための形態>
Claims (7)
- SEQ ID NO:1のアミノ酸配列を含むα-L-フコシダーゼ(α-L-fucosidase);及び
ラクトース及び重合度(degrees of polymerizations)2~4のマルトオリゴ糖からなる群より選択された一つ以上の基質を含むことを特徴とする、単糖類又はマルトオリゴ糖生産用組成物であって、
ラクトースはα-L-フコシダーゼによって単糖類であるグルコース及びガラクトースに分解され、
重合度2~4のマルトオリゴ糖はα-L-フコシダーゼによって分解され、基質で用いられるマルトオリゴ糖より高い重合度のマルトオリゴ糖が生成する、
組成物。 - α-L-フコシダーゼは、α-1,4グルコシダーゼ及びβ-1,4ガラクトシダーゼ活性とトランスグリコシル化活性を有することを特徴とする、請求項1に記載の単糖類又はマルトオリゴ糖生産用組成物。
- α-L-フコシダーゼは、ビブリオ属(Vibrio sp.)菌株EJY3から分離したものであることを特徴とする、請求項1に記載の単糖類又はマルトオリゴ糖生産用組成物。
- α-L-フコシダーゼは、前記α-L-フコシダーゼをコーディングする遺伝子を含む組換えベクターに形質転換された宿主細胞又はその培養物から得たものであることを特徴とする、請求項1に記載の単糖類又はマルトオリゴ糖生産用組成物。
- 遺伝子は、SEQ ID NO:2の塩基配列を含むことを特徴とする、請求項4に記載の単糖類又はマルトオリゴ糖生産用組成物。
- ラクトース及び重合度(degrees of polymerizations)2~4のマルトオリゴ糖からなる群より選択された一つ以上の基質とSEQ ID NO:1のアミノ酸配列を含むα-L-フコシダーゼ(α-L-fucosidase)を反応させる段階;及び
前記反応の生成物から単糖類及び基質で用いられるマルトオリゴ糖より高い重合度のマルトオリゴ糖を回収する段階を含むことを特徴とする、単糖類又はマルトオリゴ糖の生産方法であって、
ラクトースはα-L-フコシダーゼによって単糖類であるグルコース及びガラクトースに分解され、
重合度2~4のマルトオリゴ糖はα-L-フコシダーゼによって分解され、基質で用いられるマルトオリゴ糖より高い重合度のマルトオリゴ糖が生成する、
生産方法。 - 反応は、30~40℃、pH6~8、30分~48時間の条件で行うことを特徴とする、請求項6に記載の単糖類又はマルトオリゴ糖の生産方法。
