3.1 細胞培養
Kasumi-3細胞を、5%CO2インキュベーター中で37℃で完全成長培地(20% FBSを含むRPMI-1640)と共に懸濁液中に維持した。細胞密度は、3×105~3×106細胞/mlの間で維持され、培地は、2~3日の間隔で変更した。
i 新たに分裂したKasumi-3細胞を、100μl培養培地中のウェル当たり20,000細胞の密度で培養培地中の96ウェル白色プレートに播種した。
ii.一次モノクローナル抗体(CD33、CD7およびCD13)を培養培地で希釈し、プレートに加えて所望の濃度範囲(0.0001~1000nM)を得た。
iii.次いで、細胞を、一次抗体の存在下で5~10分間インキュベートした。
iv.一方、二次抗体(αMFc-CL-MMAE)は、培養培地で希釈した。一次抗体と共にインキュベートした後、αMFc-CL-MMAEをウェルに加えて、ウェル当たり13.2nMを得た。
v.ブランク、対照、陰性対照および陽性対照などの条件も、各アッセイプレートに含めた。抗CD33、抗CD7、抗CD13、抗CD33+抗CD7、抗CD33+抗CD7併用および抗CD33+抗CD13併用によるKasumi-3のプレートレイアウトについては、それぞれ表1~5を参照する。
・ブランク=培養培地のみ
・対照=処理していない細胞
・陰性対照=13.2nMのαMFc-CL-MMAEで処理した細胞
・陽性対照=0.1nMの一次抗体で処理した細胞。
vi.プレートを72時間インキュベートした。
vii.72時間のインキュベーション後、100μlのCellTiter-Glo試薬を各ウェルに添加した。
viii.内容物を軌道シェーカー上で2分間混合して、細胞溶解を誘導し、室温で10分間インキュベートした。
ix.発光強度は、Promega GloMax(登録商標)Explorer機器を用いて測定した。
ブランクの平均読み取り値は、他のすべての読み取り値から差し引いた。ブランクは培養培地のみを含み、アッセイプレートの背景として表された。その後、データを統計的に分析し(一元配置分散分析、続いて事後ダネット多重比較検定)、GraphPad Prism 7.02ソフトウェアを用いてプロットした。ルミネセンス強度はy軸上の相対的ルミネセンス単位(RLU)として表され、種々の条件および一次抗体濃度は、x軸上に表された。
4.1 Kasumi-3細胞に対する抗CD33抗体の細胞毒性プロファイル
- 抗CD33処理のためのルミネセンス強度についての未加工のデータおよびブランク減算後のデータを、それぞれ表6および7に示す。表7のグラフ表示を、図1に示す。図1は、13.2nMのαMFc-CL-MMAE存在下で、0.01nMを超える抗CD33抗体の濃度で、一元配置分散分析、続いてダネット事後多重比較検定(*p<0.02;**p<0.005;***p<0.0003、<0.0001;±SEM)を用いて分析した場合に、Kasumi-3細胞生存率の統計的に有意な低下が観察されたことを示す。定量は、平均値(±標準誤差平均値[SEM])で表す。
- 抗CD7処置のためのルミネセンス強度についての未加工のデータおよびブランク減算後のデータを、それぞれ表9および10に示す。表12のグラフ表示を、図3に示す。図3は、13.2nMのαMFc-CL-MMAE存在下で、10、100および1000nMの抗CD7抗体で、一元配置分散分析、続いてダネット事後多重比較検定(***p<0.0006;±SEM)を用いて分析した場合に、Kasumi-3細胞生存率の統計的に有意な低下が観察されたことを示す。定量は、平均値(±標準誤差平均値[SEM])で表す。

- αMFc-CL-MMAE存在下で抗CD33抗体で処理した場合のKasumi-3細胞は、0.01、0.1、1、10、100および1000nM濃度で細胞生存率の統計的に有意な低下を示した(*<0.02;**p<0.005;***<0.001)。IC50=163.6nMを、%生存率曲線から計算した。
- αMFc-CL-MMAE存在下で抗CD33および抗CD13抗体の併用で処理した場合のKasumi-3細胞は、すべての濃度で細胞生存率の統計的に有意な低下を示した(***p<0.0001)。IC50=9.378nMを%、生存率曲線から計算した。
- 二次抗体-薬物結合体αMFc-CL-MMAE単独で処理した場合のKasumi-3細胞(陰性対照)は、最小の毒性を示した。

3.1 細胞培養
HEL 92.1.7細胞を、5%CO2インキュベーター中で37℃で完全成長培地(10%FBSを有するRPMI-1640)を有する懸濁液中で維持した。2~3日の間隔で培地を交換しながら、細胞密度を1×105~1×106細胞/mlに維持した。
i.新たに分割したHEL 92.1.7細胞を、96ウェル白色プレート中に培養培地中に100μl培養培地中ウェル当たり5000細胞の密度で播種した。
ii.一次モノクローナル抗体(CD33、CD56)を培養培地で希釈し、プレートに加えて所望の濃度範囲(0.0001~1000nM)を得た。
iii.次いで、細胞を、一次抗体の存在下で5~10分間インキュベートした。
iv.一方、二次抗体(αMFc-CL-MMAE)は、培養培地で希釈した。一次抗体と共にインキュベートした後、αMFc-CL-MMAEをウェルに加えて、ウェル当たり13.2nMを得た。
v.ブランク、対照、陰性対照および陽性対照などの条件も、各アッセイプレートに含めた。それぞれ抗CD33、抗CD56および抗CD33+抗CD56組み合わせによるHEL 92.1.7のプレートレイアウトについては、表21、22および23を参照のこと。

- αMFc-CL-MMAE存在下で抗CD33抗体で処理した場合のHEL 92.1.7細胞は、10、100および1000nM濃度で細胞生存率の統計的に有意な低下を示した(*p<0.05;**p<0.01;****p<0.0001)。IC50=915.7nMを、%生存率曲線から計算した。
- 二次抗体-薬物結合体αMFc-CL-MMAE単独(陰性対照)で処理した場合のHEL 92.1.7細胞は、最小毒性を示した。
Kasumi-3細胞株に対してBiFabを用いた二次細胞死滅アッセイを繰り返すための実験を行った。

Kasumi-3細胞を回収し、計数し、ウェル当たり100μlの成長培地中で2×104細胞で96ウェルプレートに播種した。WT Bifabの5ポイント用量応答を、最終アッセイ濃度の50倍の成長培地で調製した。2μlのWT Bifab滴定を、播種細胞上にピペットし、各濃度を3つのウェルにわたってアッセイした。細胞を室温で10分間インキュベートして、Bifabが結合できるようにした。一方、抗ヒトIgG Fab-MMAE抗体を成長培地で希釈して、製造業者のプロトコルに記載されているように、ウェル当たり6nMのMMAEの最終アッセイ濃度を得た。希釈したMMAE抗体2μlを、ウェルごとにピペットした。BifabまたはMMAE結合抗体を含まない対照ウェルを、含めた。プレートを、37℃、5%CO2で72時間インキュベートした。72時間後に、CellTiter 96 AQueous One Solutionを添加し、プレートを37℃、5%CO2でさらに3時間インキュベートした。吸光度は、492および690nmで読み取った。OD492nmからOD690nmを差し引き、データをGraphPad PRISMソフトウェアを用いてプロットし、図17に示した。
図17にプロットしたOD492~690nmは、細胞をbi-Fab/抗ヒトFab-MMAE複合体で96時間処理した後に残った生存Kasumi-3細胞の尺度である。bi-Fabの濃度の増加は、生存細胞の低下に反映される細胞死滅の増加をもたらした。CD7/CD33 bi-Fabの非存在下での抗ヒトFab-MMAEによるKasumi-3細胞の処理は、細胞死滅を引き起こさない。
CD7/CD33 bi-Fab/抗ヒトFab-MMAE複合体がCD7/CD33二重抗原陽性細胞に結合する結果、複合体の内部移行および生存細胞数の減少によって反映されるそれに続く細胞死滅がもたらされる。
直接結合BiFabを用いて細胞死滅アッセイを繰り返すように実験を行った。

図18にプロットしたOD492~690nmは、抗CD7/CD33 bi-Fab-MMAE結合体で細胞を96時間処理した後に残存する生存Kasumi-3細胞の尺度である。bi-Fab-MMAEの濃度の増加は、生存細胞の低下に反映される細胞死滅の増加をもたらした。
CD7/CD33 bi-Fab-MMAEがCD7/CD33二重抗原陽性細胞に結合する結果、複合体の内部移行およびその後の生存細胞数の減少によって反映される細胞死滅がもたらされる。

Kasumi-3細胞(図19)およびSET-2細胞(図20)を回収し、計数し、それぞれの細胞株について80μlの成長培地当たり2×104細胞で再懸濁した。CD7およびCD33抗体の各々の5ポイント用量応答を、最終アッセイ濃度の10倍の成長培地で調製した。10μlの抗体滴定および10μlの成長培地を96ウェルプレートにピペットし、CD7またはCD33抗体の各濃度を3つのウェルにわたって試験した。CD7抗体およびCD33抗体の両方の10μlの抗体組み合わせについて、同じ濃度で、ウェルに加えた。細胞80μlをウェルにピペットし、プレートおよび細胞を室温で10分間インキュベートして、抗体が結合できるようにした。抗マウスFc-MMAE抗体を成長培地で希釈して、製造業者のプロトコルに記載されるように、ウェル当たり13.2nMのMMAEの最終アッセイ濃度を得た。希釈したMMAE抗体2μlを、ウェルごとにピペットした。抗体またはMMAE結合抗体を持たない対照ウェルを含めた。プレートを、37℃、5%CO2で96時間インキュベートした。96時間後、CellTiter 96 AQueous One Solutionを、およびプレートを、37℃、5%CO2でさらに3時間インキュベートした。吸光度は、492および690nmで読み取った。OD492nmからOD690nmを差し引き、データをGraphPad PRISMソフトウェアを用いてプロットし、図19および20に示した。
図19および20にプロットしたOD 492~690nmは、抗マウスFc-MMAEに結合した抗CD7単独、抗CD33単独、または抗CD7+抗CD33抗体を用いて細胞を96時間処理した後に残った、生存可能なKasumi-3細胞(図19)およびSET-2(図20)の尺度である。細胞死滅の増加は、抗CD7および抗CD33/抗マウスFc-MMAE複合体の濃度の増加に伴って観察された。抗マウスFc-MMAE単独では、いかなる細胞死滅も引き起こさなかった。
抗マウスFc-MMAEと複合した抗CD7抗体および抗CD33抗体が二重抗原陽性細胞に結合する結果、抗原内部移行を介して細胞死滅が起こる。Kasumi-3およびSET-2細胞において、抗CD7および抗CD33抗体の両方の添加は、抗CD7抗体単独の添加よりも強力な細胞死滅をもたらした。
結合アッセイにおいて標的CD7とCD33との間に相乗作用が存在するかどうかを確認するために実験を行った。

Kasumi-3、SET-2、MOLM-16、ALL-SIL、Jurkat、HEL-92、SHI-1、MV4-11、THP-1、DND-39およびRamos細胞を回収し、計数した。各細胞株の3×106個の細胞を、5分間1000rpmで遠心分離することによってペレット化した。上清を流し落とし、細胞を10mlのPBSに再懸濁した。細胞懸濁液の1mlアリコートを5mlのFACSチューブに移した。チューブに蓋をし、細胞を1000rpmで5分間、4℃で遠心分離することによって再ペレット化した。上清を注ぎ出し、細胞を100μlの1nMのWT CD7/CD33 Bifab、CD7 monofabもしくはCD33 monofab、または1nMの各CD7およびCD33 monofabの組み合わせに再懸濁し、氷冷PBS/1%BSA中で調製した。各抗体条件を、2つ1組で試験した。細胞を二次抗体のみと共にインキュベートした対照試料も、含めた。細胞を氷上で1時間インキュベートして、Fabを結合させた。インキュベーション後、各FACSチューブに氷冷PBS4mlを添加することによって、結合していないFabを除去した。細胞を、1000rpmで5分間、4℃で遠心分離することによってペレット化した。上清を流し出し、チューブを組織に反転させて可能な限り多くのPBSを除去した。一方、二次マウス抗Fab PE抗体をPBS/1%BSA中で調製し、製造プロトコルに記載されているように最終濃度を6.6μgとした。細胞を、さらに45分間氷上でインキュベートした。細胞をPBSで洗浄し、前述のように遠心分離することにより、余分な二次抗体を除去した。上清を注ぎ出し、細胞ペレットを300μlのPBSに再懸濁した。細胞上のPE標識は、FACS Calibur,BD Biosciencesを用いてFL2で検出した。各fab条件のGeomeanをプロットし、図21および22に示した。
二次抗ヒトFab-PE抗体FACS分析を用いて、白血病細胞株に対するCD7/CD33 bi-Fab、CD7 Fab、CD33 FabまたはCD7+CD33 Fabの結合を検出した。図21および22のグラフは、細胞に結合した各Fabアームの1nMについて測定した蛍光のGeomeanを示す。
FACS解析は、二重抗原陽性細胞株Kasumi-3およびSET-2について、単独または組み合わせでインキュベートした場合、bi-Fabの多くが個々のFabアームよりも細胞に結合することを図21に示す。対照的に、図22にプロットしたFACSデータは、細胞が1つの抗原のみを発現する場合を示し、bi-Fabは、CD7+ve/CD33-ve細胞中のCD7 FabおよびCD7-ve/CD33+ve細胞株中のCD33 Fabと同程度に結合する。例外は、MOLM-16およびHEL-92細胞株であり、その後、CD33のみおよびCD7/CD33二重陽性細胞の亜集団を含むことが示された。
CD7、CD33、CD7+CD33およびBifabで処理した二重抗原陽性細胞株を評価するために、細胞死滅実験を行った。

Kasumi-3細胞およびSET-2細胞を回収し、計数し、各細胞株の90μlの成長培地当たり2×104細胞で再懸濁した。Bifab-MMAE、CD7-MMAE、CD33-MMAE、CD7-MMAE+CD33-MMAEまたはゲムツズマブ-MMAEの7ポイント用量応答を、それぞれ、最終アッセイ濃度の10倍で成長培地中に調製した。10μlの各滴定を、96ウェルプレートに別々にピペットし、各濃度を2つ1組のウェルにわたって試験した。90μlの細胞をウェル中にピペットし、プレートを37℃、5%CO2で96時間インキュベートした。インキュベーション後、CellTiter 96 AQueous One Solutionを加え、プレートを37℃、5%CO2でさらに3時間インキュベートした。吸光度は、492および690nmで読み取った。OD492nmからOD690nmを差し引き、データをGraphPad PRISMソフトウェアを用いてプロットし、図23~25に示した。
図23~25にプロットしたOD492~690nmは、指示されたbi-Fab、Fabまたは抗体MMAE結合体による細胞の96時間処理後に残存する生存Kasumi-3(図23Aおよび24)ならびにSET-2(図23Bおよび25)細胞の尺度である。細胞生存率の低下は、MMAE結合体による内部移行および細胞死滅を反映する。Kasumi-3細胞についての結果は、CD7/CD33 biFab-MMAEおよびゲムツズマブ-MMAEは強力な細胞死滅をもたらすが、試験された濃度では、個々のFab-MMAE結合体では細胞死滅は見られなかったことを示す。SET-2細胞についての結果は、bi-Fab-MMAE結合体での処理のみが細胞死滅をもたらしたことを示す。
以前のデータは、個々のCD7およびCD33 FabsがKasumi-3およびSET-2細胞に結合できることを示したが、これらのFabsのMMAE結合体では細胞死滅は観察されなかった。対照的に、CD7/CD33 bi-Fabは、Kasumi-3およびSET-2細胞の両方において強力な細胞死滅を引き起こす。
CD7/CD33二重陽性サブ集団を評価するために、細胞死滅実験を行った。

HEL-92およびMOLM-16細胞を回収し、1.5mlのRPMI成長培地中で1ウェル当たり300,000細胞で24ウェルプレートに播種した。WT bi-Fab滴定は、最高最終濃度0.3nMで、最終アッセイ濃度の10倍の成長培地中で調製した。bi-Fab滴定の150μlをウェルごとにピペットし、その結果、双方の細胞株を各濃度のbi-Fabで試験した。細胞を、37℃、5%CO2で72時間インキュベートした。インキュベーション後、各試験ウェルからの細胞を回収し、4℃で5分間1000rpmでペレット化した。細胞を、300μlの氷冷PBS/1%BSAに再懸濁した。
HEL-92およびMOLM-16細胞株に対する抗CD7-PEおよび抗CD33-FITC抗体の結合のFACS分析は、CD7およびCD33またはCD33のみを発現するこれらの細胞株内にサブ集団が存在することを示した。bi-Fab-MMAEの増加する濃度を処理した細胞株の試料を、その後、抗CD7-PEおよび抗CD33 FITCで処理した。CD33のみおよびCD7/CD33細胞サブ集団をゲートし、各試料について各ゲートした領域内の細胞の数を、FACSによって記録した。bi-Fab-MMAEの濃度の増加に伴う細胞の消失百分率を計算し、データを図26に示すようにプロットした。
細胞の試料をCD7/CD33-biFab-MMAEで処理した後のこれらのサブ集団のFACS分析は、bi-Fab-MMAEがCD7/CD33サブ集団のみを特異的に死滅させることができることを示した。
さらなるCD7/CD33二重陽性AML系にわたるWT bi-Fab-MMAEの細胞死滅を分析するために、実験を行った。

UOC-M1およびHNT-34細胞を回収し、計数し、90μlの成長培地当たり2×104細胞で再懸濁した。Bifab-MMAEの8ポイント用量応答を、最終アッセイ濃縮液の10倍で成長培地中で調製した。WT Bifab-MMAE滴定の10μlを96ウェルプレートにピペットして、各濃度が2つ1組のウェルにわたって試験されるようにした。90μlの細胞をウェル中にピペットし、プレートを37℃、5%CO2で96時間インキュベートした。
図18にプロットしたOD 492~690nmは、抗CD7/CD33 bi-Fab-MMAE結合体で細胞を96時間処理した後に残存する生存HNT-34(図27)およびUOC-M1(図28)細胞の尺度である。bi-Fab-MMAEの濃度の増加は、生存細胞の低下に反映される細胞死滅の増加をもたらした。
CD7/CD33 bi-Fab-MMAEがCD7/CD33二重抗原陽性細胞に結合する結果、複合体の内部移行および生存細胞の数の減少によって反映されるその後の細胞死滅がもたらされる。
Claims (13)
- CD33およびCD7に二重特異的に結合して、CD7+CD33+血液悪性腫瘍の処置に使用するための細胞阻害剤であって、CD33およびCD7に結合する二重特異性抗体またはその抗原結合部分を含む、前記細胞阻害剤。
- 前記細胞阻害剤が、CD33+およびCD7+細胞への前記細胞阻害剤のCD33およびCD7受容体媒介内部移行を誘導することができる、請求項1に記載の使用のためのCD33およびCD7に二重特異的に結合する細胞阻害剤。
- 前記CD33+およびCD7+細胞がAML細胞である、請求項2に記載の使用のためのCD33およびCD7に二重特異的に結合する細胞阻害剤。
- 前記二重特異性抗体またはその抗原結合部分が抗体依存性細胞傷害性を媒介することができる、請求項1~3のいずれか一項に記載の使用のためのCD33およびCD7に二重特異的に結合する細胞阻害剤。
- 前記細胞阻害剤が免疫エフェクター細胞を含む、請求項1~3のいずれか一項に記載の使用のためのCD33およびCD7に二重特異的に結合する細胞阻害剤。
- 前記細胞阻害剤がT細胞および/またはNK細胞を含む、請求項5に記載の使用のためのCD33およびCD7に二重特異的に結合する細胞阻害剤。
- 細胞阻害細胞がT細胞である、請求項6に記載の使用のためのCD33およびCD7に二重特異的に結合する細胞阻害剤。
- 前記免疫エフェクター細胞が二重特異性抗CD33抗CD7 CAR-Tである、請求項5に記載の使用のためのCD33およびCD7に二重特異的に結合する細胞阻害剤。
- 前記T細胞がCD33+T細胞、CD7+T細胞、またはそれらの組み合わせを含む、請求項7に記載の使用のためのCD33およびCD7に二重特異的に結合する細胞阻害剤。
- 前記細胞阻害剤が、細胞死滅部分を更に含む、請求項1~3のいずれか一項に記載の使用のためのCD33およびCD7に二重特異的に結合する細胞阻害剤。
- 前記CD33結合部分が抗体の抗原結合断片を含む、請求項10に記載の使用のためのCD33およびCD7に二重特異的に結合する細胞阻害剤。
- 前記CD7結合部分が抗体の抗原結合断片を含み、前記細胞死滅部分が細胞毒素である、請求項10または請求項11に記載の使用のためのCD33およびCD7に二重特異的に結合する細胞阻害剤。
- 前記細胞阻害剤が、連結部分をさらに含む、請求項10~12のいずれか一項に記載の使用のためのCD33およびCD7に二重特異的に結合する細胞阻害剤。
