Description
本発明は、成人皮膚試験の分析による皮膚保護のレベルを評価すると同時に、特に乳児又は幼児用の皮膚バリアシステムの開発に関する。本発明により、幼児又は乳児に対する試験の必要性を回避しながら、客観的データにより皮膚バリアシステムの保護レベルを評価することが可能となる。
A.直接適用又はパッチ若しくは他の送達システムへの適用などによって、成人皮膚に局所物質を適用すること。
B.マーカーを成人皮膚に局所的に適用し、物質で処理された成人皮膚に対する当該マーカーの浸透データを収集する。この工程は、マーカーを適用すること、次いで、例えば共焦点ラマンマイクロ分光法(CRM)を使用して、皮膚を通過するその濃度プロファイルに従うことを含む。
C.情報(浸透データ)を成人皮膚の計算モデルに伝送し、このモデルから浸透パラメータを抽出すること。
この工程はまた、モデルの浸透プロファイルが実験データと一致するように浸透パラメータ(例えば、局所表面濃度及び透過係数)を最適化することにより、計算による成人皮膚の浸透モデルを使用してマーカーの浸透を視覚化するものとして説明することもできる。
D.(適切な変換後に)浸透パラメータを乳児皮膚の計算モデルに伝送し、乳児皮膚モデル内のマーカーの浸透を視覚化すること。
E.乳幼児皮膚モデルに浸透したマーカーの量に基づいて、乳児皮膚に対する局所製品のマイルドさ(影響)についての結論を導き出すこと。
本発明は、評価される1つ以上の局所物質を含み、局所物質は最終的な界面活性剤系で使用されることが望ましい。局所物質は、角質層の透過性に影響を及ぼす、皮膚に適用される任意の種類の物質である。局所物質は、皮膚を通過するマーカーの透過を変化させる。マーカーの透過を測定することにより、局所物質の効果を評価することができる。典型的には、上記で概説した試験では、局所物質は、マーカーが適用される前に30分間皮膚と接触して保持されるパッチに含浸される。パッチは、適用試験のための1つ以上の局所物質を含んでもよい。
本発明は、評価方法において1つ以上のマーカー又はバイオマーカーを使用する。マーカーを追跡し、濃度プロファイル(例えば、浸透データ)を作成する方法がある限り、あらゆる種類のマーカーが好適である。共焦点ラマンマイクロ分光法を使用する例では、マーカーは、ラマンスペクトルにおいて追跡可能なシグナルを有する必要がある。別の例では、蛍光マーカーは、共焦点蛍光顕微鏡法を用いて追跡することができる。マーカーの浸透速度論は、理想的には、濃度プロファイルの定常状態が妥当な時間内(例えば、最大1時間)に達するようなものでなければならない。
-安全性;毒性の理由により、過去に使用されたいくつかのマーカーは許容されない(塩化ダンシル(Paye et al.「Dansyl chloride labelling of stratum corneum:its rapid extraction from skin can predict skin irritation due to surfactants and cleansing products」Contact Dermatitis 30(2),91-96,1994で提案される)は、感作及び皮膚接触時の皮膚腐食のリスクのため、中止された)。
-浸透速度論;皮膚に浸透する分子は、例えば、十分な速さではあるが速すぎない分子である。例えば、カフェインに近い透過係数:Dias M et al.「Topical delivery of caffeine from some commercial formulations」Int J Pharm 1999182(1):41-7に報告される、kp=1.16×10-4cm/時間、を有する分子を用いることができる
-共焦点分析は非侵襲性であり、マーカー浸透の深さについてのデータを提供する。対照的に、例えば、テープ剥離は侵襲性であり、バリアを破壊する。これは、本発明では許容できない。
本発明は、浸透データを分析する。浸透データは、濃度プロファイルであり、これは、通過する皮膚の深さの関数としてのマーカーの濃度を意味する。本発明は、皮膚内、より正確には表皮及び特に角質層における深さの関数としてマーカーの濃度プロファイルを測定するのに好適な任意の所望の分析方法を使用することができる。任意の所望の方法を使用することができるが、非侵襲的方法が好ましい。共焦点技術は非侵襲性であり、適度な解像度、例えば皮膚表面に対して垂直方向に3~5μmの解像度、最大200μmの深度を提供するため好ましい。そのような方法の1つは、共焦点ラマンマイクロ分光法を含むが、共焦点蛍光顕微鏡法を含む他の方法が使用されてもよい。
本発明は、試験した界面活性剤系の成分を評価するための計算モデルを使用する。皮膚を通過するマーカーの濃度プロファイルをもたらすことができる任意のモデルを用いることができる。ユーザーは、浸透パラメータを考慮すると、皮膚を通過するマーカーの濃度プロファイルをもたらすことができる、任意のタイプの計算による皮膚浸透モデルを選択することができる。成人皮膚及び皮膚モデルの両方の使用は、モデルが皮膚アーキテクチャの構造を考慮し、2つの間に差異が存在することを必要とする。
浸透パラメータは、浸透速度論、つまり物質が表面を横切って皮膚の奥深くまで浸透することがどれほど容易かを特徴付けている。浸透パラメータは、例えば、分配係数、拡散係数、及び/又は透過係数であってもよい。
定常状態では、成人皮膚モデルにおけるマーカーの濃度プロファイルは、実験濃度プロファイルと比較される。プロファイルが一致しない場合、浸透パラメータ(C表面、PSC、及びPVE)が調整され、シミュレーションが繰り返される。2つのプロファイルが一致すると、パラメータは、乳児皮膚モデル内のマーカー浸透に対応するパラメータを計算するために使用される。乳幼児皮膚の親水性は高いため、C表面パラメータは高くなる(通常、成人皮膚の2倍*)のに対し、他の浸透パラメータは2つのモデル間で同じままである。
目標:
●2つの極端な局所溶液、すなわち1つの刺激性(0.1% SLSを含有する)局所溶液、及び1つのマイルドな(水)局所溶液の乳児皮膚に対する予測効果を示す。
●成人モデルが実験データと適合することを示す。
●局所物質が成人及び乳児皮膚に対して同じ効果を有さないことを示し、乳児皮膚はマーカーに対してより透過性であることを示す。
目標:
●乳児皮膚に対するそれらのマイルドさに基づいて、予測される界面活性剤製剤の分類を作成する。
驚いたことに、このグラフから、局所薬剤は、常に、成人皮膚で得られた実験値によって反映される乳児皮膚に関するマイルドさ指数値を持たないと予測できる。
●製剤3は、製剤5よりもマイルドである。
●製剤4は、製剤2よりもマイルドである。
上記の工程I及びIIは、同じままである。マーカーの乳児皮膚浸透に対する予測データを生成する。
実施例3
材料及び方法
研究前及び研究中に少なくとも24時間、前腕に他のいかなるスキンケア処理も行わないことに同意した、正常な皮膚の健康な被検者で成人皮膚のデータを収集した。
in vivo共焦点ラマンマイクロ分光計(Skin Composition Analyzer Model 3510,River Diagnostics,Rotterdam,The Netherlands)
カフェインパッチ:脱イオン水10mL中カフェイン180mg、1.8%
実験データを、20~35歳の5名の女性の被検者で収集した。
バリアクリーム:Dsitin(登録商標)クリーム(おむつかぶれクリーム)
US INICIリスト:酸化亜鉛10%、不活性成分(アロエ・バルバデンシス葉汁)、シクロメチコン、ジメチコン、香料、メチルパラベン、微結晶ワックス、鉱油、プロピルパラベン、精製水、ホウ酸ナトリウム、セスキオレイン酸ソルビタン、ビタミンE、白色ワセリン、白ろう。
1-温度及び湿度が管理された室内で5分間の順化
2-前腕への局所物質の適用
3-温度及び加湿が管理された室内で30分間の順化
4-前腕(同じ場所)での30分間のカフェインパッチの適用
5-ラマン指紋領域における測定
1-成人皮膚に対する実験データ。図8参照。
Desitin処理した皮(四角)からのデータを、参照(円)未処理皮膚からのデータ(すなわち、プロトコルの工程2で適用された局所物質なし)と比較する。
次の工程は、成人皮膚の計算モデルに対して皮膚透過パラメータを定義して、上に提示された実験データを正確にシミュレートすることができるようすることである。
最後の工程は、適切な変換を用いてカフェイン浸透パラメータを乳児皮膚の計算モデルに伝送し、乳児皮膚の推定カフェイン浸透をシミュレートすることを含む。
%保護=100×(AUC(未処理)-AUC(製品))/AUC(未処理)=89.18%
実験データを、18~40歳の6名の女性ボランティアで収集した。
-保湿剤A:グリセリン(12%)、ワセリン(4%)、ジステアリルジモニウムクロリド、水を含むエマルジョン
-保湿剤B:ワセリン(40%)、グリセリン(12%)、ジステアリルジモニウムクロライド、水を含む構造化エマルジョン
1-温度/湿度が管理された室内で5分間の順化
2-前腕への局所物質の30分間の適用
3-前腕(同じ場所)での30分間のカフェインパッチの適用
4-指紋領域における測定

保湿剤Aで処理した皮(四角)からのデータを、保湿剤Bで処理した皮膚(三角)及び参照未処理皮膚(〇)からのデータ(すなわち、プロトコルの工程2で適用された局所物質なし)と比較する。
次の工程は、成人皮膚の計算モデルに対して皮膚透過パラメータを定義して、上に提示された実験データを正確にシミュレートすることができるようすることである。
最後の工程は、適切な変換を用いてカフェイン浸透パラメータを乳児皮膚の計算モデルに伝送し、乳児皮膚の推定カフェイン浸透をシミュレートすることを含む。
保護%:該当なし
保湿剤Aの曲線下面積は、未処理参照の曲線下面積よりも優れている。シミュレーションは、乳児皮膚に対する保護効果を予測しない。
保護%=100×(AUC(未処理)-AUC(製品))/AUC(未処理)=17.72%。

d)浸透パラメータを最適化することによって、成人皮膚浸透の計算モデルを使用して前記マーカーの浸透を可視化して、前記成人皮膚浸透のモデルのプロファイルが実験データと一致するようにすることと、
e)最適化された前記浸透パラメータを乳児皮膚の計算モデルに伝送することと、
f)前記乳児皮膚の計算モデルにおける前記マーカーの浸透を決定することと、を含む、方法。
(2) 前記マーカーがカフェインである、実施態様1に記載の方法。
(3) 前記成人皮膚浸透の計算モデルとしてEPISIMを用いる、実施態様1に記載の方法。
(4) 実施態様1に記載の方法により選択される、バリアシステム。
(5) 前記成人皮膚浸透の計算モデルが、薬剤ベースのモデルである、実施態様1に記載の方法。
Claims (11)
- 乳児皮膚に対するバリアシステムの潜在的影響を評価する方法であって、
a)前記バリアシステムを成人皮膚に局所的に適用することと、
b)前記バリアシステムで処理された前記成人皮膚にマーカーを局所的に適用することと、
c)前記バリアシステムで処理された前記成人皮膚への前記マーカーの浸透を測定することと、
d)浸透パラメータを最適化することによって、成人皮膚浸透の計算モデルを使用して前記マーカーの前記浸透を可視化して、前記成人皮膚浸透のモデルのプロファイルが実験データと一致するようにすることと、
e)最適化された前記浸透パラメータを前記乳児皮膚の計算モデルに伝送することと、
f)前記乳児皮膚の前記計算モデルにおける前記マーカーの浸透を決定することと、を含む、方法。 - 前記マーカーがカフェインである、請求項1に記載の方法。
- 前記成人皮膚浸透の前記計算モデルとしてEPISIMを用いる、請求項1に記載の方法。
- 前記工程a)は、前記バリアシステムを直接適用またはパッチへの適用により局所的に適用することを含む、請求項1に記載の方法。
- 前記工程a)は、前記バリアシステムを前記パッチへの適用により局所的に適用することを含む、請求項4に記載の方法。
- 前記工程c)は、前記成人皮膚を通過する前記マーカーの濃度プロファイルに従うことを含む、請求項1に記載の方法。
- 前記濃度プロファイルは、共焦点ラマンマイクロ分光法(CRM)又は共焦点蛍光顕微鏡を用いて測定される、請求項6に記載の方法。
- 前記浸透パラメータは、肌表面濃度(C 表面 )パラメータ、角質層に対する透過係数(P SC )、及び生存表皮に対する透過係数(P VE )である、請求項1に記載の方法。
- 前記肌表面濃度(C 表面 )パラメータが前記乳児皮膚の前記計算モデルにおいて前記成人皮膚浸透の前記計算モデルおけるよりも高くなるように、かつ、前記角質層に対する透過係数(P SC )および前記生存表皮に対する透過係数(P VE )が前記2計算モデル間で同じになるように、前記浸透パラメータは前記工程e)において伝送される、請求項8に記載の方法。
- 前記乳児皮膚の前記計算モデルにおける前記肌表面濃度(C 表面 )パラメータは、前記成人皮膚浸透の前記計算モデルにおける前記肌表面濃度(C 表面 )パラメータの2倍である、請求項9に記載の方法。
- 請求項1に記載の方法を用いて前記バリアシステムを選択することを含む、バリアシステムの調製方法。
