Description
開示される方法及び組成物は、方法、プロトコール、及び試薬はさまざまに異なりうるため、特定の方法、プロトコール、及び試薬に限定されない点を理解されたい。本明細書で使用される用語は、あくまで特定の実施形態を説明する目的のものであり、本発明の範囲を限定しようとするものではない点も理解されるべきであり、本発明の範囲は付属の特許請求の範囲によってのみ限定されるものである。
開示される組成物を調製するために使用される各成分、及び本明細書に開示される方法の中で用いられる組成物自体を開示する。これらの材料及び他の材料を本明細書に開示するが、これらの材料の組み合わせ、サブセット、相互作用、グループなどが開示される場合、これらの化合物のそれぞれ異なる個々の、及び集合的な組み合わせ及び順列の具体的な言及が明示的に開示されていない場合もあるが、それぞれが具体的に想到され、本明細書に記載されているものと理解される。1種以上のMGSペプチドと結合された抗体を含む組成物が開示される。いくつかの態様では、抗体は細胞内の標的をターゲティングする。

細胞内標的をターゲティングする方法であって、治療有効量の、開示される組成物のうちの1種以上のものを投与することを含む方法が開示される。
開示される組成物のうちの1種以上のものをコードした核酸配列を含むベクターが開示される。いくつかの態様では、ベクターは、開示されるMGSペプチドのうちの1種以上のものをコードすることが可能な核酸配列のみを含む。
上記に記載した材料及び他の材料は、開示される方法を実施する、またはその実施を助けるうえで有用なキットとして任意の適当な組み合わせで一緒にパッケージングすることができる。特定のキットの中のキット要素は、開示される方法で一緒に使用されるように設計及び適合されていれば有用である。例えば、開示される組成物のうちの1種以上のものを含むキットが開示される。
細胞内/細胞下区画を標的とするシステマチックな手法は最近まで報告されていなかった。特許技術であるFOX-Three技術は、MAbを細胞及び細胞下標的に送達することが可能なMGSをシステマチックに発見するためのプラットフォームである。FOX-Threeプラットフォームは、所望の細胞タイプに誘導する能力、細胞の取り込みを誘発する能力、及び標的細胞の個別の場所にペイロードを送達する能力の3つの主要な特性によりMGSを同定する。MGSの働き方を示す概観を図1に示す。
MGSは多くの疾患の治療に影響し、新たな治療選択肢を開きうるものである。タンパク質の相互作用をターゲティングすることで、「薬剤治療不能」(undruggable)な標的を治療できる可能性を有する、新たなクラスの細胞内で作用する薬剤が創出される。治療適応症としては、がん、新たに出現する感染症及びバイオテロリズム、心臓疾患、糖尿病、線維症、神経及び神経変性疾患が挙げられる。MGSは広範な利用可能性を有し、小分子薬剤、造影剤、核酸及びタンパク質の送達、ナノ粒子、及び細胞療法を送達するために用いることができる。
本発明のまた別の態様は、以下のとおりであってもよい。
〔1〕1種以上の分子誘導システム(MGS)ペプチドと結合された抗体を含む、組成物。
〔2〕前記抗体が、細胞内標的をターゲティングする、前記〔1〕に記載の組成物。
〔3〕前記抗体が、モノクローナル抗体である、前記〔1〕または〔2〕に記載の組成物。
〔4〕前記モノクローナル抗体、が抗Rasモノクローナル抗体である、前記〔3〕に記載の組成物。
〔5〕前記1種以上のMGSペプチドが、EHPWFNMWSWATQVQE(配列番号38)、YPGSPTQYPSSMHEYHSSSE(配列番号39)、AHTIDDEWASYHMQQWNSPP(配列番号40)、FEEFYSRQSNTIPYPQQYKG(配列番号41)、THGNKHQSWTYPSEINHKNY(配列番号19)、NLADTWTQTQQHDFHVLRGTR(配列番号20)、GYSWWQPNWPSSTWDT(配列番号21)、またはこれらの組み合わせの配列を含む、前記〔1〕~〔4〕のいずれか1項に記載の組成物。
〔6〕前記1種以上のMGSペプチドが、配列番号1、2、3、34、35、36、37、38、39、40、41、41、42、43、44、45、46、47、48、49、50、51、52、5、54、55、56、57、58、59、60、61、62、63、64、65、66、67、68、69、70、71、72、73、74、75、76、もしくは77、またはこれらの組み合わせの配列を含む、前記〔1〕~〔4〕のいずれか1項に記載の組成物。
〔7〕前記1種以上のMGSペプチドが、EHPWFNMWSWATQVQE(配列番号38)の配列を含む、前記〔5〕に記載の組成物。
〔8〕前記1種以上のMGSペプチドが、1種以上の細胞内標的に局在化する、前記〔1〕~〔6〕のいずれか1項に記載の組成物。
〔9〕前記細胞内標的が、リソソーム、ゴルジ装置、小胞体、細胞質、または細胞核である、前記〔8〕に記載の組成物。
〔10〕前記抗体が、リンカーを介して前記1種以上のMGSペプチドと結合されている、前記〔1〕~〔9〕のいずれか1項に記載の組成物。
〔11〕前記1種以上のMGSペプチドと結合された前記抗体が、融合タンパク質である、前記〔1〕~〔9〕のいずれか1項に記載の組成物。
〔12〕細胞内標的をターゲティングする方法であって、1種以上のMGSペプチドと結合された抗体を投与することを含み、前記抗体が細胞内標的をターゲティングする、前記方法。
〔13〕前記細胞内標的が、リソソーム、ゴルジ装置、小胞体、細胞質、または細胞核である、前記〔12〕に記載の方法。
〔14〕治療を要する対象を治療するための方法であって、前記治療を要する対象に、有効量の、1種以上のMGSペプチドと結合された抗体を投与することを含み、前記抗体が、疾患プロセスに関与する細胞内標的をターゲティングする、前記方法。
〔15〕前記治療を要する対象が、感染症、がん、糖尿病、神経もしくは神経変性疾患、遺伝性疾患を有するか、またはバイオテロリズム剤に曝露されている、前記〔14〕に記載の方法。
〔16〕CH 3 CO-YAAWPASGAWT-PEG 11 -C-NH 2 の配列を含む、改変ペプチド。
〔17〕CH 3 CO-LQWRRNFGVWARYRL-PEG 11 -C-NH 2 の配列を含む、改変ペプチド。
〔18〕CH 3 CO-d(Leu)-RGDLATLRQL-PEG 11 -YC-NH 2 の配列を含む、改変ペプチド。

Claims (11)
- 1種以上の分子誘導システム(MGS)ペプチドと結合された抗Rasモノクローナル抗体を含む組成物であって、前記1種以上のMGSペプチドが、EHPWFNMWSWATQVQE(配列番号38)、YPGSPTQYPSSMHEYHSSSE(配列番号39)、AHTIDDEWASYHMQQWNSPP(配列番号40)、またはFEEFYSRQSNTIPYPQQYKG(配列番号41)である、組成物。
- 前記抗Rasモノクローナル抗体が、細胞内標的をターゲティングする、請求項1に記載の組成物。
- 前記1種以上のMGSペプチドが、EHPWFNMWSWATQVQE(配列番号38)の配列を含む、請求項1に記載の組成物。
- 前記1種以上のMGSペプチドが、1種以上の細胞内標的に局在化する、請求項1または2に記載の組成物。
- 前記細胞内標的が、リソソーム、ゴルジ装置、小胞体、細胞質、または細胞核である、請求項4に記載の組成物。
- 前記抗Rasモノクローナル抗体が、リンカーを介して前記1種以上のMGSペプチドと結合されている、請求項1~5のいずれか1項に記載の組成物。
- 前記1種以上のMGSペプチドと結合された前記抗Rasモノクローナル抗体が、融合タンパク質である、請求項1~5のいずれか1項に記載の組成物。
- 細胞内標的をターゲティングするための医薬の製造のための1種以上のMGSペプチドと結合された抗Rasモノクローナル抗体の使用であって、前記1種以上のMGSペプチドが、EHPWFNMWSWATQVQE(配列番号38)、YPGSPTQYPSSMHEYHSSSE(配列番号39)、AHTIDDEWASYHMQQWNSPP(配列番号40)、またはFEEFYSRQSNTIPYPQQYKG(配列番号41)であり、前記抗Rasモノクローナル抗体が細胞内標的をターゲティングするものである、前記使用。
- 前記細胞内標的が、リソソーム、ゴルジ装置、小胞体、細胞質、または細胞核である、請求項8に記載の使用。
- 感染症、がん、糖尿病、神経もしくは神経変性疾患、遺伝性疾患、またはバイオテロリズム剤への曝露の治療を要する対象における感染症、がん、糖尿病、神経もしくは神経変性疾患、遺伝性疾患、またはバイオテロリズム剤への曝露を治療するための方法において用いるための医薬の製造のための1種以上のMGSペプチドと結合された抗Rasモノクローナル抗体の使用であって、前記方法が前記治療を要する対象に、有効量の、前記1種以上のMGSペプチドと結合された抗Rasモノクローナル抗体を投与することを含み、前記1種以上のMGSペプチドが、EHPWFNMWSWATQVQE(配列番号38)、YPGSPTQYPSSMHEYHSSSE(配列番号39)、AHTIDDEWASYHMQQWNSPP(配列番号40)、またはFEEFYSRQSNTIPYPQQYKG(配列番号41)であり、前記抗Rasモノクローナル抗体が、疾患プロセスに関与する細胞内標的をターゲティングするものである、前記使用。
- 1種以上のMGSペプチドと結合された抗Rasモノクローナル抗体を含む、感染症、がん、糖尿病、神経もしくは神経変性疾患、遺伝性疾患、またはバイオテロリズム剤への曝露の治療をするための医薬組成物であって、前記1種以上のMGSペプチドが、EHPWFNMWSWATQVQE(配列番号38)、YPGSPTQYPSSMHEYHSSSE(配列番号39)、AHTIDDEWASYHMQQWNSPP(配列番号40)、またはFEEFYSRQSNTIPYPQQYKG(配列番号41)であり、前記抗Rasモノクローナル抗体が、疾患プロセスに関与する細胞内標的をターゲティングするものである、前記医薬組成物。
