Description
本発明は一般式(I)の化合物に関する。
式中、Rは、COOR1及びCH2OR2、並びに一般式(A)の基から選択され、
R1は、哺乳動物の組織又は器官において加水分解により除去されて(R)9‐シス‐13,14‐ジヒドロレチノエート及び生物学的に許容可能な耐容性の化合物を生じる基であり、及び/又は
R2はH又はアシル基C(O)R3であり、ここでR3は哺乳動物の組織又は器官において加水分解により除去されて(R)9‐シス‐13,14‐ジヒドロ‐レチノール及び生物学的に許容可能な耐容性の化合物を生じる基であり、及び/又は
Rは一般式Aの基であり、
式中、Q1は、置換若しくは非置換のC6‐10アルケニル若しくはシクロアルケニル、好ましくは置換若しくは非置換のトリメチル‐シクロアルケニル、又はより好ましくは置換若しくは非置換の2,6,6‐トリメチルシクロヘキセニル、さらにより好ましくは非置換の2,6,6‐トリメチルシクロヘキサ‐1‐エン‐1‐イル又は2,6,6‐トリメチルシクロヘキサ‐2‐エン‐1‐イルであり、トリメチルシクロヘキセニルの場合、例えば、2,6,6‐トリメチルシクロヘキセニル基は置換されており、それは好ましくはヒドロキシル置換又はオキソ置換、好ましくはオキソ置換である、前記化合物は、投与されると、哺乳動物の組織又は器官又は細胞において9‐シス‐13,14‐ジヒドロ‐レチノールに変換される。

酸型のエステルに関しては、WO95/04018A1は9‐シス‐レチノイン酸エステルの製造に関する。化合物は、炭素11と12の間、及び炭素原子13と14の間に二重結合を含み、シス又はトランス配置のいずれかを有することができる。本明細書において方法を利用することができるが、化合物は必ずしも13,14ジヒドロ誘導体であることはできない。WO95/32946A1はまた、9‐シス‐レチノイン酸エステルの調製に関する。化合物は、シス異性体とトランス異性体の両方であり得る、炭素11‐12と13‐14の間の二重結合を含まなければならない。
出発化合物は市販されているか、又は標準的な方法に従って合成することができる。
実施例に示される実施形態において、(R)‐9‐シス‐13,14‐ジヒドロレチノイン酸の(R)‐4エチルエステル[(R)‐1]の調製は、エナンチオ純粋なトリエニルヨージド3とボロン酸2の鈴木カップリングに基づいた(実施例1のスキーム1参照)。3の合成は(Z)‐スタンニルジエノール5から開始し、これは対応するチオールとの光延反応によりベンゾチアゾリルアリルスルフィド6に変換され、続いてH2O2及びペルオキシモリブデート(VI)試薬を用いてスルホン7に酸化された。Julia‐Kocienskiオレフィン化は、わずかに過剰の塩基及び過剰のエナンチオ純粋なアルデヒド(R)‐8(エチルエステルである)を用いて行われた。アリルスルホンとアルデヒドとの反応の立体化学的結果に関する以前の知見から予想されるように、トリエニルエステル(R)‐9の新たに形成されたオレフィンはZ‐幾何学的配置のものである。前駆体スタンナンをCH2Cl2中のヨウ素溶液で処理すると、Sn‐1交換及び9Z、11Zジエンの所望の9Z、11E幾何異性体へのヨウ素促進異性化を介してヨウ化物(R)‐3が生成した。幾何異性体は、NOE実験によって確認することができる。
9‐シス‐13,14‐ジヒドロレチノールは、エチル13,14‐ジヒドロレチノエートのDIBAL‐H(水素化ジイソブチルアルミニウム)還元により、本明細書に記載されるように調製されている。
好ましい9‐シス‐13,14‐ジヒドロレチノイン酸エステル
好ましい9‐シス‐13,14‐ジヒドロレチノイン酸エステルは、典型的には9‐シス‐13,14‐ジヒドロレチノイン酸のメチル、エチル又はプロピルエステルのような低級アルキルエステルである。長鎖脂肪族アルコールと9‐シス‐13,14‐ジヒドロレチノイン酸とのエステルは、9‐シス‐13,14‐ジヒドロレチノールの長鎖脂肪酸のエステルよりも好ましさが劣る。
さらに好ましい9‐シス‐13,14‐ジヒドロレチニルエステル化合物としては、ギ酸エステル(メタノエート)、エタノエートエステル及びプロピオン酸エステル、さらに好ましさは劣るが酪酸エステル及び吉草酸エステルが挙げられる。

303>207m/zで検出された相対MS‐MSシグナルが9CDHRAに対応するかどうかを確認するために、9‐シス‐13,14‐ジヒドロレチノイン酸の両方のエナンチオマーの立体選択的合成を、パラジウム触媒Csp2‐Csp2鈴木カップリング(Pazos Y et al.2001)に基づいて先に記載された戦略に従って実施した。9‐シス‐13,14‐ジヒドロレチノイン酸の(R)‐及び(S)‐エナンチオマーの立体制御合成、精製及び特徴付けの詳細を以下に提供する。
(a)PPh3,BTSH,DIAD,CH2C12,2h(98%)。
(b)(NH4)6Mo7O24・4H20,30% H2O2,EtOH,-10℃,17時間(66%)。
(c)NaHMDS,THF,-78℃,30分(93%)。
(d)I2,CH2Cl2,25℃,30分。
(e)Pd(PPh3)4,10%aq.TlOH,THF,25℃,3時間(42%)。
(f)2M KOH,MeOH,80℃,45分(84%)。
溶媒は公表されている方法に従って乾燥し、使用前に蒸留した。他の全ての試薬は入手可能な最高純度の市販化合物であった。特記しない限り、全ての反応はアルゴン雰囲気下で行われ、水性試薬を含まないものはオーブン乾燥ガラス器具中で行われた。分析用薄層クロマトグラフィー(TLC)は、Merck Kieselgel 60F254を用いてアルミニウムプレート上で行い、UV照射(254nm)によって、又はホスホモリブデン酸のエタノール溶液で染色することによって可視化した。フラッシュカラムクロマトグラフィーは、加圧下でMerck Kieselgel 60(230~400メッシュ)を使用して実施した。電子衝撃(EI)質量スペクトルは、70eVで作動するHewlett‐Packard HP59970機器で得た。あるいは、7T能動遮蔽磁石を備えたAPEX III FT‐ICR MS(Bruker Daltonics)を使用し、(イオン化効率を最適化するために)1800~2200Vの電圧を針に印加し、450Vの対向電圧をキャピラリーに印加して、アポロAPIエレクトロスプレーイオン化(ESI)源を使用してイオンを生成した。ESIスペクトルについては、試料を70:29.9:0.1(v/v/v)CH3OH/水/ギ酸のスプレー溶液を1~5%のv/v比で試料の溶液に添加することによって調製し、最良の信号対雑音比を得た。高分解能質量スペクトルはVG Autospec機器で取った。1H NMRスペクトルは、内部標準として残留プロトン性溶媒を用いて400MHzでBruker AMX‐400分光計でC6D6及びアセトン‐d6中で周囲温度で記録した。
(2Z,4E‐3‐メチル‐5‐(トリブチルスタンニル)ペンタ‐2,4‐ジエン‐1‐オール(1.0g、2.58mmol)、2‐メルカプトベンゾチアゾール(0.65g、3.87mmol)及びPPh3(1.10g、4.21mmol)のTHF(14mL)中の溶液を0℃で5分間撹拌した。THF(5mL)中のDIAD(0.77mL、3.87mmol)の溶液を滴下し、混合物を25℃で30分間撹拌した。溶媒を除去し、残渣をカラムクロマトグラフィー(C18‐シリカゲル、CH3CN)により精製して、(2Z,4E)‐1‐(ベンゾチアゾール‐2‐イル)スルファニル‐5‐(トリ‐n‐ブチルスタンニル)‐3‐メチルペンタ‐2,4‐ジエン6と同定された1.11g(78%)の無色の油状物を得た。
EtOH(9mL)中の(2Z、4E)‐1‐(ベンゾチアゾール‐2‐イル)スルファニル‐5‐(トリ‐n‐ブチルスタンニル)‐3‐メチルペンタ‐2,4‐ジエン6(0.48g、0.89mmol)の溶液へ,-10℃で、過酸化水素水溶液(35%、7.7mL)中の(NH4)6Mo7O24・4H2O(0.44g、0.36mmol)の溶液を加えた。-10℃で17時間撹拌した後、混合物をH2Oでクエンチし、Et2O(3×)で抽出した。合わせた有機層をブライン(3×)で洗浄し、乾燥させ(Na2SO4)、溶媒を除去した。残留物をクロマトグラフィー(C18‐シリカゲル、MeOH)により精製して、(2Z、4E)‐1‐(ベンゾチアゾール‐2‐イル)スルホニル‐5‐(トリ‐n‐ブチルスタンニル)‐3‐メチルペンタ‐2,4‐ジエン7として同定される0.33g(66%)の無色の油状物を得た。
THF(9mL)中の(2Z、4E)‐1‐(ベンゾチアゾール‐2‐イル)スルホニル‐5‐(トリ‐n‐ブチルスタンニル)‐3‐メチルペンタ‐2,4‐ジエン(0.115g、0.20mmol)の冷却(-78℃)溶液をNaHMDS(0.23mL、THF中1M、0.23mmol)で処理した。この温度で30分間撹拌した後、THF(4.5mL)中の(S)‐エチル3‐メチル‐4‐オキソブタノエート(0.044g、0.30mmol)の溶液を加え、得られた混合物を-78℃で1時間撹拌し、3時間温度を室温に到達させるために放置した。Et2O及び水を低温で加え、混合物を室温に温めた。それを次いでEt2Oで希釈し、層を分離した。水層をEt2O(3×)で抽出し、合わせた有機層を乾燥し(Na2SO4)、溶媒を除去した。残渣をカラムクロマトグラフィー(C‐18シリカゲル、MeOH)で精製して、(3S、4Z、6Z、8E)‐エチル3,7‐ジメチル‐9‐(トリ‐n‐ブチルスタンニル)ノナ‐4,6,8‐トリエノエート(S)‐9として同定される0.94g(93%)の淡黄色油状物を得た。
CH2Cl2(5.3mL)中の(3R,4Z,6Z,8E)‐エチル3,7‐ジメチル‐9‐(トリ‐n‐ブチルスタンニル)ノナ‐4,6,8‐トリエノエート(R)‐9(0.060g、0.121mmol)の溶液へ、CH2Cl2(2.8mL)中のヨウ素(0.046g、0.182mmol)を滴下して加え、得られた混合物を25℃で30分間撹拌した。飽和Na2S2O3溶液を添加し、反応混合物をEt2O(3×)で抽出し、合わせた有機層を乾燥させ(Na2SO4)、溶媒を除去した。残渣をカラムクロマトグラフィー(シリカゲル、97:3ヘキサン/Et3N)で精製して、(3R,4E,6Z,8E)‐エチル9‐ヨード‐3,7‐ジメチルノナ‐4,6,8‐トリエノエート(R)‐3として同定される0.037g(92%)の淡黄色油状物を得た。
(R)‐9‐シス‐13,14‐ジヒドロレチノールは、-78℃でTHF中の(9Z,13R)‐エチル13,14‐ジヒドロレチノエート((R)‐4)のDIBAL‐H還元によって収率95%で合成した(スキーム1)。
ジメチルアミノピリジン(DMAP)の存在下、無水酢酸及びピリジンを用いて(R)‐9‐シス‐13,14‐ジヒドロレチノールをアセチル化することにより、(R)‐9‐シス‐13,14‐ジヒドロレチニルアセテート]を86%の収率で調製した。
(3R,4E,6Z,8E)‐3,7‐ジメチル‐9‐(2,6,6‐トリメチルシクロヘキサ‐1‐エン‐1‐イル)ノナ‐4,6,8‐トリエン‐1‐オール((R)‐10)
エチル(3R,4E,6Z,8E)‐3,7‐ジメチル‐9‐(2,6,6‐トリメチルシクロヘキサ‐1‐エン‐1‐イル)ノナ‐4,6,8‐トリエノエート[エチル(R)‐9‐シス‐13,14‐ジヒドロレチノエート、(R)‐4](32.0mg、0.097mmol)のTHF(1.0mL)中の冷却(-78℃)溶液へ、DIBAL‐H(0.242mL、0.242mmol、ヘキサン中1.0M)を加え、得られた混合物を-78℃で30分間撹拌した。混合物を2.5時間で-20℃まで温めた。H2Oを加え、混合物をEt2O(3×)で抽出した。合わせた有機層を乾燥し(Na2SO4)、溶媒を除去した。残渣のフラッシュクロマトグラフィー(シリカゲル、最初に98:2のヘキサン/Et3Nで中和し、次いで95:5のヘキサン/EtOAcからEtOAcへの勾配)により、(3R,4E,6Z,8E‐3,7‐ジメチル‐9‐(2,6,6‐トリメチルシクロヘキサ‐1‐エン‐1‐イル)ノナ‐4,6,8‐トリエン‐1‐オール(26.5mg、95%)を無色の油状物として得た。

9‐シス‐エチルレチノエート(III)。THF(2.0mL)中のエチル(E)‐4‐(ジエトキシホスホリル)‐3‐メチルブタ‐2‐エノエート(II)(0.227g、1.10mmol)の冷(0℃)溶液に、nBuLi(0.63mL、1.00mmol、ヘキサン中1.6M)及びDMPU(0.15mL、1.24mmol)を加えた。1時間撹拌した後、反応物を-78℃に冷却し、THF(2.5mL)中の(2Z,4E)‐3‐メチル‐5‐(2,6,6‐トリメチルシクロヘキサ‐1‐エン‐1‐イル)ペンタ‐2,4‐ジエナール(I)(0.100g、0.46mmol)を加え、2時間撹拌した。水を添加し、混合物をEt2O(3×)で抽出した。合わせた有機層を乾燥し(Na2SO4)、溶媒を蒸発させた。粗生成物をカラムクロマトグラフィー(シリカゲル、95:5ヘキサン/EtOAc)で精製して、エチル9‐シス‐レチノエート(III)として同定される0.144g(96%)の黄色の固体を得た。

(Z)‐3‐ヨード‐3‐メチルプロパ‐2‐エン‐1‐オール(X)。CuI(0.34g、1.78mmol)をプロパルギルアルコール(IX)(1.00g、11.84mmol)のEt2O(13.2mL)中溶液に添加した。MeMgBr(17.8mL、53.51mmol、Et2O中3M)の溶液を-20℃で加えた。この温度で2時間撹拌した後、溶液を25℃に到達させ、さらに12時間撹拌した。Et2O(40.0mL)中のI2(9.10g、35.67mmol)の溶液を0℃で添加し、冷却浴を取り外した。25℃で24時間撹拌した後、得られた混合物を0℃に冷却し、氷水を加えた。有機層をNa2S2O4の飽和水溶液(3×)で洗浄し、次いでセライト(登録商標)のパッドを通して濾過した。残渣を蒸留(0.2mmHg、60℃)により精製して、(Z)‐3‐ヨード‐3‐メチルプロパ‐2‐エン‐1‐オール(X)として同定された黄色油2.10g(60%)を得た。
動物:
雄マウス(Charles River、フランス)を、個々に換気されたケージ(Techniplast、イタリア)内の午前7時~午後7時の明/暗サイクルで、ケージあたり3匹のマウスの群に収容した。食物(標準食餌、フランスのSAFEからのD04)及び水は自由に入手可能であった。すべての実験は、1986年11月24日の欧州共同体評議会指令(86/609/EEC)に従い、CNRS及びフランスの農林省の指針(法令87848)に従って行われた。
行動学的にナイーブなマウス群を、薬理試験を容易にするための修正を加えて(Wietrzych‐Schindler et al.、2011)、以前に記載されたプロトコール(Wietrzych et al.、2005)に従ってT‐迷路のDNMTPで試験した。具体的には、本プロトコルでは、動物を最初に約15秒の最小試行間間隔(ITI)で10連続日にわたって訓練した。これは4連続日(9‐10日目)の間に正しい選択の90%以上という基準を達成するために必要であった。この期間の後、5日間連続して(11~15日目に示す)6分のブロックずつITIを半ランダムに増加させて、各動物について「試験ITI」を決定し、これはマウスがチャンスレベルで行動する最短ITIとして定義した。16日目から、マウスを16日目のビヒクル注射後(ビヒクル単独の効果について試験するため)及び17日目の9CDHROL(40mg/kg)注射後に対応する「試験ITI」遅延を用いて試験した。保持期間中に腕を選択するための潜伏期間が3日連続で1日2回以上の試行で3分を超えたため(除外基準)、3匹のマウスを試験から除外した。18日目にマウスに再度9CDHROL(40mg/kg)を注射し、9時間後に化学分析のために殺した。3日連続してビヒクル注射を受けたマウスを9CDHROL代謝の分析のためのビヒクル処置対照として使用した。急性処置後の9CDHROL代謝について試験するために、3匹の未処置マウスのさらなる群にも9CDHROL(40mg/kg)を注射した。
行動分析のために、9CDHROL及び9CDHBCをエタノールに溶解し、次いでひまわり油と混合し、最終溶液は3%のエタノールを含んでいた。ビヒクル処置は、ヒマワリ油中の3%エタノール溶液からなった。午前7時から試験開始前の7時間の間に、3ml/kgの容量/重量比で腹腔内注射により処置を施した。ストレスプロトコールの1日目から始まるストレスセッション直後のストレスプロトコールの2日目毎に5~6pmの間に、各物質について10mg/kg、体重/重量比3ml/kgで経口投与した。
学習段階中の行動成績の比較は、従属パラメータとしての時間と独立パラメータとしての正しい選択の割合を用いて、反復測定における一元配置分散分析(ANOVA)を用いて行われた。薬理学的データは、反復測定で一元配置分散分析を用いて、ビヒクル処置後及び9CDHROL処置後の動物の能力を比較することによって分析した。事後統計分析を行って、一群スチューデントt検定を用いて動物の能力を50%のチャンスレベルと比較した。重要な違いは対応する図に示されている。
9CDHROLがインビボで9CDHRAを産生するための基質として作用し得る可能性を試験するために、我々は9CDHRAの活性を検出するのに敏感な行動パラダイムとして作業記憶を遅延非場所合わせ(DNMTP)で試験した(Ruhl et al、2015)。この目的のために、我々は4日間連続して最大性能(90%超)の基準を達成するためにDNMTP課題においてC57BL6J雄マウスのコホートを訓練した(図1a)。野生型C57BL6N雄マウスは、15秒間の試行間隔(ITI)で訓練を受けたときに訓練日の有意な効果によって示されるように作業記憶課題を獲得した(F[9,39]=8,28;p<0.001、反復測定で一元配置分散分析)。マウスを3分というより長いITIで試験したとき、記憶能力は低下した。しかしながら、ITIが平均13分(グラフ中にgrl3として示される全群の平均)に達したとき、それらの能力は50%のチャンスレベルと変わらなかったので、マウスはDNMTP課題において完全な記憶喪失を示した(p<0.05、1群t検定)。各動物が完全な忘却を示したITIは、各個々の動物について「試験ITI」として同定され、9CDHROLの記憶促進活性(pro-mnemonic activity)を試験するときに使用された(図1b)。そのような試験の間、9CDHROL処置は記憶性能を有意に向上させたが、正しい選択の割合は50%のチャンスレベル(p=0.2、ns;1群t検定)に匹敵したので、ビヒクル適用は記憶を改善しなかった(F[1,4]16;p<0.05、治療効果の反復測定に関する一元配置分散分析)。9CDHROL処置マウスは、50%のチャンスレベルよりも有意に良好に機能した(p<0.05;一群のt検定)。
組織試料のLC‐MS分析
A.レチノイド分析のみ:分析手順:高速液体クロマトグラフィー質量分析(Agilent 1260 Infinity LCシステム;Madrid、Spain)‐質量分析(SCIEX Triple Quad 3500 System;Sciex、Madrid、Spain)は、以前に検証されたプロトコルを使用して濃い黄色/琥珀色の光で行った(Ruhl、2006;Ruhl et al.、2015)。13,14‐ジヒドロレチノイン酸の検出についてはMS‐MS設定は303→207m/zであり、13,14‐ジヒドロレチノールの検出についてはMS‐MS設定はMS‐MS特有のレチノイン酸の設定に匹敵する同じ滞留時間及び衝突エネルギーを用いて290→69m/zであった。したがって、試料調製のために、100mgの材料(試料が100mg未満の場合、使用される標準重量:100mgまで水を添加した)又は100μlの血清を3倍容量のイソプロパノールで希釈し、組織をハサミで刻み、10秒間ボルテックスし、5分間超音波浴に入れ、6分間振盪し、+4℃でHeraeus BIOFUGE Fresco中で13000rpmで遠心分離した。遠心分離後、上清を30℃のエッペンドルフ濃縮器5301(エッペンドルフ、ドイツ)で乾燥した。乾燥抽出物を60μlのメタノールで再懸濁し、40μlの60mM酢酸アンモニウム水溶液で希釈し、オートサンプラーに移して、引き続いて分析した。
行動分析及びレチノイド/カロテノイド分析のための統計分析は、2群比較のそれぞれについてスチューデントt検定を用いて行った。重要な差は対応する図に示されている。
内因性9CDHROLの初期同定及び9CDHROLのマウスへの処置後の9CDHROLの同定
最初に、LC‐MS分析を使用して9‐シス‐13,14‐ジヒドロレチノール(9CDHROL)と全トランス‐ジヒドロレチノール(ATDHROL)の混合標準を分析し(図2の上図)、ビヒクル処置マウスからの肝臓試料を分析した(図2の下図)。標準9CDHROLとATDHROLの両方のピークは、LC‐MSシステムの同じMS‐MS分画チャンネルを使用して、肝臓試料で同定されたピークと共溶出している。共溶出に基づいて同じLC‐MSパラメータと同じ分画パターンチャンネル(290‐>69m/z)を使用して、9CDHROLとATDHROLを哺乳動物の内因性レチノイドと主張して同定する。さらに、これらの共溶出ピークはマウスの血清及び脳でも低レベルで観察することができ、9CDHROLは9CDHROL処置後に強く増加する(図3)。
マウス脳を上記の手順に従って調製した。簡単に言うと、夕方のマウスに40mg/kgの用量の9CDHROL(又は他の引用物質)を単回経口投与した後、11時間後に屠殺し、脳を含む組織試料を遮光条件下で解剖し、秤量し、液体窒素中で凍結し、分析まで-80℃で保存した。血清を調製し、さらなる分析まで、直ちに茶色のバイアル中に-80℃で保存した。
9CDHBCは、缶の中のモモ(砂糖煮)における共溶出及び匹敵するUV/VISスペクトルを用いて同定された。
マウスにおける9CDHROL処置後の9CDHRAの同定
最近我々は9CDHRAがマウス(Ruhl et al.、2015)及びヒト(まだ未発表のデータ)において内因性の誘導体であることを確認した。次のステップは、インビボ及びインビトロモデルで9CDHRAの前駆体を同定することである。
9CDHROLは9CDHROL処置マウス及び9CDHBCインビトロ乏突起膠細胞培養からの代謝産物に蓄積される:経口投与マウスにおける9CDHROLによる処置は、対照処理脳(図14Aの中央クロマトグラム)と比較してマウス脳における9CDHROLを増加させる(図14Aの下のクロマトグラム)。本明細書では、9CDHBC及び9CDHROLの投与はヒト乏突起膠細胞培養物中の9CDHROLレベルを増加させるが、ATBCは増加を示さなかったことに留意する。9CBC(及びATROL)は、乏突起膠細胞培養において、9CDHROLの増加を全く示さなかったか、又はわずかな非異性体選択的増加を示しただけである(図14D)。まとめると、9CDHBCは、インビトロでのヒト乏突起膠細胞培養における9CDHROLの優れた前駆体基質である。
細胞培養実験
正常な不死化乏突起膠細胞系158Nを以前に報告されたように培養した(Feutzら、2001)。細胞が70%の集密度に達したとき、それらを以下の化合物のうちの1つの10‐2Mエタノール溶液で処理した:9CDHROL、9CDHROL‐アセテート、9CDHRA‐エチルエステル。対照としての9CDHRA及びエタノール、ビヒクル処置。処理の18時間後、細胞をアンバーエッペンドルフチューブに集め、分析まで-80℃で保存した。レチノイドの光分解を避けるために、光制限条件下で処理及び細胞収集を行った。
同様の実験において、我々は以下のレチノール種を必要な変更を加えて同じ細胞培養に適用した:ATROL及び9CROL。標準として通常のROL標準が適用された(黒い線)。種は矢印でも示されている(図11)。
分析手順は上記の通りに実施した。
我々は、9CDHRAがマウスの血清、脳及び肝臓における9CDHROL補給の代謝産物であることを確認した。さらに、乏突起膠細胞株を10‐5Mの9CDHROL、9CDHRET‐アセテート又は9CDHRA‐エチルエステルで18時間処理した。我々は9CDHROL、9CDHRET‐アセテートの投与後に9CDHRAレベルの強い増加が見られたことを観察した(図6上)。加えて、我々は9CDHRA‐エチル‐エステル適用後に非常に強い増加が確認されたことを見出した(図6下)。
9CDHBC及び9CDHROLの投与は、ヒト乏突起膠細胞培養物中の9CDHROLレベルを増加させるが、ATBCは増加を示さなかった。9CBC(及びATROL)は、乏突起膠細胞培養において9CDHROLの増加を全く示さないか、又はほんのわずかな非異性体選択的増加を示す(図14D)。要約すれば。9CDHBCは、インビトロでのヒト乏突起膠細胞培養における9CDHROLの優れた前駆体基質である。
9CDHBC及び9CBC投与後、ただしインビトロでの乏突起膠細胞培養へのATBC投与後ではない9CDHBCの同定(図16A及び16B):9CBC投与時の乏突起膠細胞培養では、これらの処理した細胞における411nmの検出波長で、保持時間26.1分で9CBCを検出でき(図16A、411nmの検出)、一方、ATBC又は9CDHBC処置後のいずれかと同様に、9CBCは対照処置では検出できなかった。この存在は、ダイオードアレイ検出器によって取られたUV/Visスペクトルによっても確認された(データはここには示されていない)。
10‐5Mの9CDHROL、9CDHROL‐アセテート、9CDHRA又は9CDHRA‐エチル‐エステルで18時間処理した乏突起膠細胞株では、9CDHROLの後、9CDHROL‐アセテートは9CDHRAレベルの強い増加を見いだした(図17A)。さらに、9CDHRA‐エチル‐エステル(及び9CDHRA、データは示されていない)の適用後、非常に強い増加が確認された(図17Aの下のクロマトグラム)。9CDHROL及び9CDHROL‐エステルは9CDHRAの優れた前駆体であるが、我々はまた、ATROL、9CDHBC、9CBC及びATBCは、9CDHRAに変換されないことを確認した(データは示されていない)。要約すると、9CDHROL、9CDHROL‐エステル及び9CDHRA‐エステルは、インビトロヒト乏突起膠細胞培養における優れた選択的かつ異性体特異的なRXR‐リガンド前駆体である。
実験は実施例5に記載したものと類似していた。
インビトロ実験に加えて、本発明者らは、経口補給マウスに9CDHBC及び9CDHROLを用いてインビボ補給実験も行った(図17B及び17C)。対照と比較して、9CDHBC補給後に9CDHRAの中程度だが有意な増加が観察された(図17B)。低レベルの9CDHRA及びその異性体ATDHRAは、マウスの対照マウス(図17Cの上のクロマトグラム)の血清、肝臓及び脳でも観察され、特に9CDHRAは9CDHROL補給後に著しく増加する(図17Cの下のクロマトグラム)。
例8.1:方法
動物:
Rbpl‐/‐及びRxry‐/‐変異体並びにそれらの野生型(WT)対照マウスを、記載のようにヘテロ接合性交配から混合遺伝的背景(60%C57BL/6J及び40%129SvEms/j)で飼育し(Ghyselinck NB 1999);Krezelら、1996)、3~6ヶ月の年齢で試験した。全てのマウスを、個々に換気されたケージ(Techniplast、イタリア)中で午前7時~午後7時の明/暗サイクルでケージ当たり4~5匹のマウスの群に収容した。食料と水は自由に手に入るようにした。すべての実験は、1986年11月24日の欧州共同体評議会指令(86/609/EEC)に従い、CNRS及びフランスの農林省の指針(法令87848)に従って行われた。
強制水泳パラダイム(Dalvi and Lucki、1999)は、22~23℃の水で半分満たされた2リットルのガラス製ビーカー中で午後1時~午後4時の間に行われた(水深は17cm)。全てのマウスはこの課題において一度だけ試験された。このために、各マウスを穏やかに水中に下げ、不動時間を6分間の試験期間中に記録した。マウスは直立姿勢で浮遊し、頭を水面上に保つためにわずかな動きしかしなかった場合に動けないと判断された。6分後、マウスを水から取り出し、赤色灯の下で乾燥させ、そのホームケージに戻した。各動物の不動スコアを絶望行動の指標として用いた。
次に動物を強制水泳及びスクロース試験で試験した。
インビボでのRXR機能の調節における9CDHRAの関連性に取り組むために、本発明者らは、R‐9CDHRAがRbp1‐/‐マウスにおける行動障害を逆転させることができるかどうかを試験した。R‐9CDHRAによる急性処置は、強制水泳試験において用量依存的にノックアウトマウスの不動性を低下させ、すでに1mg/kgで最大効果を達成し、これは同用量又は5mg/kgでのATRAによる処置で合成RXRアゴニストUVI2108の抗致死効果に匹敵した(図9)。処置の効果はWTマウスでは明白ではなかったが、それはこの系統における低いベースラインの不動性及びその結果としての床効果に関連しているかもしれない。R‐9CDHRAでの2mg処置はRxrγ‐/‐マウスの能力を改善しなかったので、そのような活性はRXRγによって媒介され、ビヒクル処置Rxrγ‐/‐動物における119±19秒間と比較して120±25秒間不動のままであった。
本発明者らは、R‐9CDHRA補給が、鬱病の慢性ストレスモデルにおいて鬱病行動を治療することを見出した。ストレスは、うつ病の病因における重要な環境要因である。ストレスによって誘発される鬱病行動の治療のための9cDHRAの効率を試験するために、我々は社会的敗北ストレス動物モデルを使用した(Berton et al.、2006、Hollis and Kabbaj、2014)。支配的な優性CD1雄との10日間の短い毎日の物理的接触とそれに続くその後の感覚的接触は、強制水泳課題における絶望を効率的に誘発した(図10.a)。ストレスプロトコール中の1又は3mg/kgの9cDHRAによる処置は、コントロールのストレスを受けていないマウスに匹敵する不動時間を効率的に標準化した。より低い用量とは対照的に、3mg/kgの9cDHRAでの治療は、ストレスを与えられた非処置マウスで観察された無動時間よりも有意に低いので抗絶望効果を示し、したがって9cDHRAはこの行動パラメータの制御において用量効果を示す。9cDHRAの効果は合成panRXRアゴニストUVI2108の活性と同等であり、9cDHRAによる抗絶望活性の達成におけるRXR活性化の重要な役割を示唆している。絶望行動に加えて、慢性的なストレスも無作為選択を反映して50%と有意に異ならないレベルで消費された甘味飲料の嗜好の欠如によって反映される無快感症を誘発した(図10.b)。無快感症は、低用量の9cDHRA(1mg/kg)ですでに処置されたストレスマウスでは、チャンスレベルの50%を有意に超えるスクロースの嗜好性によって示されるようになくなったが、この嗜好性は3mg/kgの9cDHRAの用量を上げて処置すると、さらに顕著であったのである。anRXRアゴニストUVI2108は9cDHRAと同様の活性を示し、抗うつ活性における9cDHRAによるRXR活性化の関与を支持した。
ストレスは、うつ病の病因における重要な環境要因である。ストレスによって誘発される鬱病行動の治療のための9cDHROL及び9cDHBCの効率を試験するために、我々は社会的敗北ストレス動物モデルを使用した(Berton et al.、2006、Hollis and Kabbaj、2014)。支配的な優性CD1雄との10日間の短い毎日の物理的接触とそれに続くその後の感覚的接触は、強制的な水泳課題において効率的に絶望を誘発した(図18.8A)。ストレスプロトコール中の10mg/kgの9CDHROL又は10mg/kgの9CDHBCによる治療は、対照のストレスを受けていないマウスに匹敵する不動時間を正常に標準化した。絶望行動の慢性的なストレスは無作為選択を反映して50%と有意に異ならないレベルで消費された甘味飲料の嗜好の欠如によって反映される無快感症を誘発した(図18.B)。ストレスプロトコルの間に10mg/kgの9CDHROL又は10mg/kgの9CDHBCで処置されたストレスマウスにおいては、スクロース嗜好性が50%のチャンスレベルを有意に超えたことにより示されるように、無快感症はなくなった。
ビタミンAは体内で視覚色素と核ホルモン受容体のリガンドに変換されることができる誘導体の集まりである。ビタミンA1(レチノール)はRARリガンド全トランス‐レチノイン酸(ATRA)の前駆体であることがよく知られているが、最近発見されたレチノイドX受容体(RXR)の内因性のリガンドである9‐シス‐13,14ジヒドロレチノイン酸(9CDHRA)の前駆体の同一性は知られていない。本研究では、9cDHRAの栄養前駆物質は9‐シス‐13,14‐ジヒドロレチノール(9CDHROL)と9‐シス‐13,14‐ジヒドロ‐βカロテン(9CDHBC)であり、それぞれ新しいタイプのレチノイドとカロテノイドであって、これまで記載されてはいないが、食品マトリックス中及びヒトを含む哺乳動物生物中に内因的に高レベルで存在する。インビトロ及びインビボ実験を用いて、我々はそのような前駆体が直接又は間接的に9CDHRAに代謝され得ることを実証する。対照的に、全トランス‐レチノール、9‐シス‐レチノール及び全トランス‐β‐カロテンのような周知の内因性レチノイド/カロテノイドは、9CDHRAの弱い選択的な又は非選択的な前駆体にすぎない。我々はまた、既知の内因性カロテノイド9‐シス‐β‐カロテン(9CBC)が脱水素化によって9CDHBCに弱く変換されるだけであることを実証する。しかしながら、9CDHBCは容易に9cDHRAに変換され、それ自体RXRリガンド‐9cDHRAの間接的な前駆体として提案することができる。我々の代謝スクリーニングを通して、我々は初めて9CDHRAエステル、9CDHROL、9CDHROLエステル、9CDHBCが内因性RXRリガンド9CDHRAの栄養学的及び生理学的に関連のある選択的前駆体を元の形に戻すのに優れ、RXR媒介シグナル伝達を誘導することを証明した。我々はさらに、この新しいクラスの物質を、9cDHROLの場合はビタミンA5、9CDHBCの場合はプロビタミンA5と命名された新しい独立した選択的な新しいタイプのビタミンAとして説明した。これらの化合物の予防的/医薬的使用のための概念の証明は、うつ病の慢性ストレス動物モデルにおけるうつ病様行動の治療に示されている。これらの化合物の同様の使用は、RXR媒介シグナル伝達が影響を受けるか又は治療標的として提案されている様々な疾患に対して想定することができる。そのような疾患には、神経変性疾患及び代謝疾患、皮膚機能障害及び免疫機能障害(炎症を含む)、並びに心血管疾患が含まれ得るが、記憶亢進効果のような生活様式の用途にも関係し得る。

- 精神的健康又は性能を維持するため、又は請求項10~15のいずれかに定義された状態を予防するために健康を維持するための、請求項1~6のいずれかに記載の化合物を含む、医薬組成物又は栄養補助食品組成物。
- 非治療的応用における化合物9‐シス‐13,14‐ジヒドロ‐ベータ‐カロテン(9CDHBC)のレベルを増加させるための栄養補助食品としての、対応する自然環境又は食品マトリックス中に存在する量よりも多い量で9-シス-ベータ-カロテン(9CBC)を含む、栄養補助食品組成物。
- 請求項11~15のいずれかに定義された疾患の治療又は予防に使用するための、請求項19に記載の栄養補助食品組成物。
- VA5欠乏症の予防に使用するための、及び/又は一般的なVA5補給のための、又はRXR依存性機能障害を予防するための、請求項19に記載の栄養補助食品組成物。
- 記憶能力を増強するための、請求項19に記載の栄養補助食品組成物。
- R3は、C1‐6アルキル、C9‐23アルキル、C2‐6アルケニル、及びC13‐23アルケニルから選択される、請求項23に記載の化合物。
- 9‐シス‐13,14‐ジヒドロ‐ベータ,ベータ‐カロテン又は9‐シス‐13,14‐ジヒドロ‐ベータ,アルファ‐カロテンである、9‐シス‐カロテノイド化合物。
- 哺乳動物の被験体の治療に、RXR関連疾患又はビタミンA5欠乏症に使用するための請求項26に記載の9‐シス‐カロテノイド化合物であって、該9‐シス‐カロテノイド化合物は栄養補助食品組成物中に存在し、該栄養補助食品組成物は、天然の食品マトリックス中に存在するよりも多い量又は濃度で該化合物を含有する、及び/又は追加量の化合物を含む、上記9‐シス‐カロテノイド化合物。
