Description
上記の方法は、T細胞クローン(クローン型)のライブラリー又は個々の若しくは特定の腫瘍型に特異的なネオ抗原を生成するために使用できる。
本発明に従って使用する分子は、カチオン性抗菌ペプチド(CAP)として一般に知られるペプチド群を含む。これらは正電荷を有する両親媒性ペプチドであり、この種のペプチドは多くの種に見出され、かつ先天性免疫系の一部を形成する。CAPラクトフェリシン(LfcinB)は、ミトコンドリアに影響を与えることが示された25個のアミノ酸ペプチドである(Eliasenら、Int.J.cancer(2006)119,493-450)。9個のアミノ酸ペプチド(WO 2010/060497に記載の種類の)である、はるかに小さいペプチドLTX-315もまた、ミトコンドリアを標的とすることも見出された。
「カチオン性側鎖を有するアミノ酸」とは、腫瘍細胞の細胞内pH、例えば約pH7.4で正味の正電荷を有する側鎖を有するアミノ酸を意味する。遺伝子コードされるアミノ酸の中で、これはリジン及びアルギニンを含むが、その側鎖上にかかる正味の正電荷を保有する任意の遺伝子コードされないアミノ酸又は修飾アミノ酸が使用され、例えばグアニジノ基又はアミン基又は別のカチオン性部分、例えばリジンの誘導体、及びアルギニンを有する側鎖を保有するそれらのアミノ酸を使用でき、プロトン化水素以外の側鎖中の任意の水素は、ハロゲン原子、例えばフッ素、塩素又は臭素、又は、直鎖、分枝鎖、脂肪族、不飽和又は飽和のC1~C4アルキル又はアルコキシ基、例えばメチル、エチル、プロピル、イソプロピル、ブチル、イソブチル、sec-ブチル、tert-ブチル、エチレン、プロピレン、ブチレン、ヒドロキシ、メトキシ、エチルオキシ、プロピルオキシ、イソ-プロピルオキシ、ブチルオキシ基、イソブチルオキシ、sec-ブチルオキシ、tert-ブチルオキシ又はハロゲン置換されたそのバージョンで置換される。
特許公報WO2010/060497に記載されているように、溶解性ペプチド又はペプチド模倣物は、以下の特性を有してよい:
a)直鎖状配列の9個のアミノ酸からなる;
b)これらの9個のアミノ酸のうち、5個はカチオンであり、4個は親油性R基を有する;
c)上記9個のアミノ酸の少なくとも1個は、遺伝子コードされないアミノ酸又は遺伝子コードされるアミノ酸の修飾誘導体であり;必要に応じて、
d)親油性及びカチオン性残基は、互いに隣接する2以下のいずれかのタイプの残基が存在するように配置されており;さらに必要に応じて、
e)分子は、2対の隣接するカチオン性アミノ酸及び1対又は2対の隣接する親油性残基を含む。

したがって、本発明に従って提供されるのはまた、LTX-301、LTX-303~LTX-310、LTX-312~LTX-321、LTX-323~LTX-327、LTX-329、LTX-331~LTX-336、及びLTX-338、又はその塩、エステル若しくはアミドからなる群から選択される化合物である。したがって、本発明は、配列番号10及び12~42、又はその塩、エステル若しくはアミドからなる群から選択される式を有する化合物を提供する。特に好ましいのは、化合物LTX-315である。
特許公報WO2011/051692に記載されているように、ペプチド、ペプチド模倣物又はアミノ酸誘導体は、少なくとも+2の正味の正電荷を有していてよく、二置換βアミノ酸を組み込んでおり、βアミノ酸中の置換基の各々は、同一である又は異なっていてよく、少なくとも7個の非水素原子を含み、親油性であり、少なくとも1個の環状基を有し、置換基内の1以上の環状基は他の置換基内の1以上の環状基に結合及び融合されていてよく、その場合の環状基は、2置換基の非水素原子の合計数が少なくとも12であるように融合される。βアミノ酸上の2つの置換基は好ましくは同一である。
環状基又は複数の環状基は典型的には、1~4個の、好ましくは1~3個の原子の鎖だけペプチド主鎖から(すなわち、βアミノ酸のα又はβ炭素原子から)離間されており;これらの結合原子は、窒素及び/又は酸素を含んでよいが、典型的には炭素原子であり、好ましくは結合原子は置換されていない。これらのスペーサは、もちろん、本明細書で定義される置換基の一部である。
これらの分子は好ましくは、長さが1若しくは2~12個のアミノ酸又は同等のサブユニットのペプチド又はペプチド模倣物である。特に文脈から明らかでない限り、本明細書での「アミノ酸」への言及は、ペプチド模倣物中の同等のサブユニットを含む。好ましい分子は、1~3又は4個のアミノ酸を有するが、代わりに長さが3~12個、好ましくは5~12個のアミノ酸であり得る。本発明に従って使用する分子は、単一のアミノ酸を単に含んでよいが、これは、電荷の要件を満たすために「修飾された」アミノ酸である。
3以上のアミノ酸を有するペプチドは典型的には、1以上の追加の親油性アミノ酸、すなわち親油性R基を有するアミノ酸を有する。典型的には親油性R基は、縮合又は結合されていてよい少なくとも1個の、好ましくは2個の環状基を有する。親油性R基は、O、N又はSなどのヘテロ原子を含有できるが、典型的には1以下のヘテロ原子が存在し、好ましくはそれは窒素である。このR基は、好ましくは2以下の極性基を有し、より好ましくは1つも無いか又は1個有し、最も好ましくは1つも無い。

βアミノ酸の好ましい置換基を定義する前の節は、変更すべきところは変更して、R1~4の2個の置換基に適用される。式Iの基を組み込むペプチド及ペプチド模倣物は、本出願の前の方に記載される分子の好ましいサブセットであるので、分子の好ましい特徴、例えばそれらの長さ及びそれらが含有する他のアミノ酸を定義する全ての前の節も、式Iの基の組み込みによって定義されるこれらの分子に適用され、その逆もまたあり得る。特に好ましい分子は、1~7又は8個(例えば、1~5個)、より好ましくは1、2、3又は4個のアミノ酸長である。ペプチド模倣分子は同じ数のサブユニットを含むが、これらのサブユニットは典型的にはアミド結合模倣物によって結合され;好ましい結合は前述されており、エステル及びアミノメチル及びケトメチレンを含む。
好ましくは、分子はLTX-401である。
上述したように、上で定義した分子は、ペプチド模倣物の形態であり得る。ペプチド模倣物は典型的には、そのペプチド同等物の極性、三次元サイズ及び機能性(生物活性)を保持することによって特徴づけられるが、ここでペプチド結合は多くの場合、より安定した結合で置換されている。「安定な」とは、加水分解酵素による酵素分解に対してより耐性であることを意味する。一般的には、アミド結合と置き換わる結合(アミド結合代用物)は、アミド結合の多くの特性、例えば立体構造、立体容積、静電的特徴、水素結合の可能性などを保つ。「薬物の設計と開発(Drug Design and Development)」の第14章、Krogsgaard,Larsen,Liljefors and Madsen(編)1996,Horwood Acad.Pubは、ペプチド模倣物の設計及び合成のための技術の一般的な説明を提供している。適切なアミド結合代用物は、以下の基を含む:N-アルキル化(Schmidt,R.ら,Int.J.Peptide Protein Res.,1995,46,47)、レトロインバースアミド(Chorev,M及びGoodman,M.,Acc.Chem.Res,1993,26,266)、チオアミド(Sherman D.B.及びSpatola,A.F.J.Am.Chem.Soc.,1990,112,433)、チオエステル、ホスホネート、ケトメチレン(Hoffman,R.V.及びKim,H.O.J.Org.Chem.,1995,60,5107)、ヒドロキシメチレン、フルオロビニル(Allmendinger,T.ら,Tetrahydron Lett.,1990,31,7297)、ビニル、メチレンアミノ(Sasaki,Y及びAbe,J.Chem.Pharm.Bull.1997 45,13)、メチレンチオ(Spatola,A.F.,Methods Neurosci,1993,13,19)、アルカン(Lavielle,S.ら,Int.J.Peptide Protein Res.,1993,42,270)及びスルホンアミド(Luisi,G.ら、Tetrahedron Lett.1993,34,2391)。
好ましいものとして記載される全てのペプチドのペプチド模倣等価物もまた好ましい。
本明細書に記載の溶解分子は、任意の便利な方法で合成できる。一般的に、存在する反応基(例えば、アミノ、チオール及び/又はカルボキシル)は、全合成中に保護される。合成での最後の工程はしたがって、本発明の保護された誘導体の脱保護である。
ペプチド合成の方法は当技術分野で周知であるが、本発明については、固相支持体上で合成を行うことが特に好都合であり得、かかる支持体は当技術分野で周知である。
対象は、典型的にはヒト患者であるが、家庭用又は家畜動物などの非ヒト動物も処置されてよく、実験又は試験用の動物が処置されてよい。
活性分子を含有する投与単位は好ましくは、0.1~10mgの、例えば1.5mgの本発明の抗腫瘍分子を含有する。
本発明を、以下の実施例及び図面を参照しながらさらに説明する。
LTX-315は骨肉腫細胞の細胞質膜を崩壊する
1.材料及び方法
FORVEILLE S.ら,Cell Cycle 2015,14:3506-12に記載のように細胞膜を崩壊させた。
細胞培養用の培地及び栄養補助剤をGibco-Life Technologies(Carlsbad,CA,USA)から入手し、Lytix Biopharma(Tromso,Norway)によって提供されたLTX-315(K-K-W-W-K-K-W-Dip-K-NH2)を除く化学物質をSigma-Aldrich(St.Louis,MO,USA)から入手した。プラスチック製品はGreiner Bio-One(Monroe,CA,USA)からのものであった。
超微細構造研究のために、ヒト骨肉腫U2OS細胞を、1時間、1.6%のグルタルアルデヒド(0.1Mのリン酸緩衝液中v/v)中で固定し、掻き取りによって回収し、遠心分離し、ペレットを1%四酸化オスミウム(0.1Mのリン酸緩衝液中w/v)で後固定した。段階的エタノール系列を通して脱水した後、細胞をEponTM 812中に包埋し、超薄切片を標準的な酢酸ウラニル及びクエン酸鉛で染色した。画像を、Tecnai 12電子顕微鏡(FEI、Eindhoven,the Netherlands)を用いて取得した。
LTX-315の投与後、細胞の大部分は、細胞質膜が崩壊した壊死形態をとった(図2)。
LTX-315は内部移行し、ミトコンドリアと相互作用する
本研究では、ヒト黒色腫細胞に対するLTX-315の殺腫瘍効果を調べた。ペプチドは内部移行し、ミトコンドリアに付随して、最終的に溶解された細胞死に至ることが示された。LTX-315ペプチドは、2段階の作用モードを介して腫瘍内注射で固形腫瘍を処置する。作用モードの第1は腫瘍自体の崩壊であり、第2は、瀕死の腫瘍細胞から放出される損傷関連分子パターン分子(DAMP)であり、これは再発及び転移に対するその後の免疫防御を誘発できる。
本研究を、EIKE L-Vら,Oncotarget 2015,6:34910-23に記載の通りに行った。
LTX-315及びLTX-328(K-A-Q-Dip-Q-K-Q-A-W-NH2)は、要求に応じてそれぞれBachem AG(Bubendorf,Switzerland)及びInnovagen(Lund,Sweden)によって作製された。LTX-315パシフィックブルー及びLTX-328パシフィックブルーは、要求に応じてそれぞれInnovagen(Lund,Sweden)、Norud(Tromso,Norway)から購入した。
A375細胞株A375(ECACC、88113005)は、患者物質由来のヒト悪性黒色腫であり、Public Health England(PHE Culture Collections,Porton Down,Salisbury,UK)から購入した。細胞を、抗生物質を含まない、10%FBS及び1%L-グルタミンを補充した高グルコース4.5% DMEM(完全培地)中で単層培養として維持した。この細胞株を、37℃にて加湿した5%CO2雰囲気中で増殖させ、MycoAlert(Lonza)を用いてマイコプラズマの存在について定期的に試験した。
非標識細胞を用いた生細胞イメージング-A375細胞を、25μg/mlのヒトフィブロネクチン(Sigma)でプレコートしたNunc Lab-Tec 8-ウェルチャンバーカバーガラス(Sigma)中の完全培地中に10,000細胞/ウェルで播種し、一晩接着させた。細胞を、無血清RPMIで2回洗浄し、RPMI中に溶解したペプチドで処置し、63X/1.2W対物レンズを用いて、多光子励起Leica TCS SP5共焦点顕微鏡を使用して調べた。この顕微鏡は、CO2及び温度制御を有するインキュベーションチャンバーを備えていた。
固定細胞、蛍光標識ペプチド―サブコンフルエントなA375細胞を、上記のように、8,000細胞/ウェルで播種し、製造業者のプロトコルに従ってプラストランスフェクション試薬(Invitrogen)と共にリポフェクタミンLTXを用いて2日目にトランスフェクトした。ミトコンドリアを、pDsRed2-Mitoを用いて標識し、核を、GFP-ヒストン2Bプラスミド(Imaging Platform,University of Tromso)を用いて標識した。トランスフェクションの翌日、細胞を無血清RPMIで2回洗浄し、異なる濃度及び異なるインキュベーション期間で、LTX-315パシフィックブルー又はLTX-328パシフィックブルーで処置した。MTTアッセイによって決定した場合、LTX-315 PBは、非標識LTX-315と同様の細胞傷害プロファイルを示した。対照細胞を、非標識LTX-315及び再び無血清RPMI単独で処置した。インキュベーション後、細胞をPBS中の4%パラホルムアルデヒドで固定し、ウェルをProlong Gold antifade(Invitrogen)で覆った。細胞を、693、1.2W対物レンズを用いて、Leica TCS SP5共焦点顕微鏡を使用してさらに分析した。パシフィックブルー、GFP及びDs Redを、488及び561レーザーと共にUVを用いて励起し、蛍光チャネルを、次のバンドパス:UV:420~480nm(減衰で)、488:501~550nm及び561:576~676nmを用いて順次検出した。
A375細胞を、6ウェルプレート中にウェル当たり1×105細胞で播種し、培養物中で膜構造を最適化するために3日間増殖させ、培地を2日目に交換した。細胞を無血清RPMIで2回洗浄し、その後、5、10及び25μg/mlで無血清RPMI中に溶解したLTX-315で処置し、陰性対照として無血清RPMIを用いた。細胞を次いでPBSで2回洗浄し、その後、4℃で24時間、pH7.8のHepes緩衝液中の4%ホルムアルデヒド及び1%グルタルアルデヒドで固定した。脱水及び後固定プロトコルは、5%緩衝タンニン酸中でのインキュベーション及び1%オスミウム低減フェロシアン化物中でのインキュベーションを含んだ。超薄切片を作製し、酢酸ウラニル(5%)及びレイノルズのクエン酸鉛を染色及び対比のために使用した。試料を、Jeol JEM-1010透過型電子顕微鏡で検査し、画像を、Olympus MoradaサイドマウントTEM CCDカメラ(Olympus soft imaging solutions,GmbH,Germany)で取得した。
DCFDA細胞活性酸素種の検出アッセイキットを、abcam(登録商標)から購入し、DCFDAアッセイ前にA375細胞を96ウェルのCostar黒色透明底プレートにウェルあたり20,000細胞で播種し、37℃で16時間インキュベートした。細胞を、100μL/ウェルの予め温めたPBSで1回洗浄し、細胞培養インキュベーター中37℃で45分間キット付属の緩衝液中で20μΜのDCFDAとインキュベートし、次いで100μL/ウェルの緩衝液で再び洗浄した。細胞を次いで、17μΜの濃度で緩衝液中に溶解した100μL/ウェルのLTX-315ペプチドで30分間刺激し、処置しなかった細胞を陰性対照として使用した。蛍光強度を、FLUOstar Galaxyプレートリーダーで485nmの励起波長及び530nmの発光波長で測定した。
A375細胞を、6ウェルプレート中の完全培地に3×105個の細胞/ウェルで播種し、一晩接着させた。細胞を、35μΜにてLTX-315又はLTX-328で処置し、37℃および5%CO2で異なる時間(5、10、15、30、60分間)インキュベートし、陰性対照は無血清RPMI-1650であった。上清(S)を収集し、5分間1,400gで遠心分離し、細胞溶解物(L)をPBSで2回洗浄した後に収集し、その後、4×試料緩衝液(Invitrogen,number)、0.1M DTT(Sigma number)及び水を用いて溶解した。上清をAmicon Ultra 50K遠心分離フィルター(Millipore UFC505024)を用いて濃縮し、細胞溶解物を超音波処理した。上清と溶解物の両方を煮沸し、10%ドデシル硫酸ナトリウムポリアクリルアミドゲル電気泳動(SDS-PAGE)で分離し、次いでポリフッ化ビニリデン(PVDF)膜(Millipore)に電気移動させた。膜を5%ミルク中でブロッキングし、HMGB1抗体(ウサギ、ポリクローナル、abcam ab 18256)とインキュベートした。膜を次いでTBSTで数回すすぎ、西洋ワサビペルオキシダーゼ(HRP)結合二次抗体(abcam ab6721)とインキュベートし、TBSTで再度すすぎ、次いでWBルミノール試薬(Santa Cruz Biotechnology,Heidelberg,Germany)を使用して発色させた。
A375細胞を、HMGB1研究と同様に播種し、異なる時間(5、15、45)、35μΜで処置した。HMGB1研究と同様に上清を収集し濃縮し、上清からの試料を、製造業者の説明に従って4.5時間固体形態チトクロムC-Elisaキット(R&D Systems,USA,♯DCTC0)を用いて分析した。その後すぐに、50%希釈した試料を分析し、450nmに設定したマイクロプレートリーダーを用いて光学密度を決定し、この読み取りを次いで540nmでの読取りから差し引いた。標準曲線を、アッセイした試料の各セットについて作製した。試料を4つ平行して実行し、上清中に放出されたチトクロムcを、未処置細胞の上清中のチトクロムcレベルに対する倍数として表した。
LTX-315処置A375細胞の上清を、Enliten ATPルシフェラーゼアッセイキット(Promega,USA)を用いて分析した。細胞を次いで、ROSアッセイと同様に播種し、2つ平行して1~15分の異なるインキュベーション時間で、LTX-315で処置し、これを次いで3回行った。陰性対照は、無血清培地のみに暴露した未処置A375細胞であった。試料を1:50及び1:100で希釈し、製造業者のプロトコルに従ってLuminoscan RTルミノメーターで分析した。
全てのデータは、少なくとも2つ平行して行った少なくとも2つの独立した実験を表し、平均±SDとして表した。チトクロムC放出及びATP放出データを、一方向ANOVA及び多重比較検定を用いて比較し、発明者らは、P値<0.05が統計的有意性を示すと考えた。
2.1 LTX-315は内部移行し、ミトコンドリアを標的にする
細胞内でのペプチドの内部移行及び運命を調べるために、LTX-315をパシフィックブルーで標識し、それぞれ3μΜ及び1.5μΜの濃度で細胞とインキュベートした。標識LTX-315は形質膜を素早く貫通し、1.5μΜで、ペプチドは、インキュベーションの30分後にミトコンドリアの周りに蓄積を示したが、細胞核中には検出されなかった(図3)。標識した非溶解性モック配列ペプチドLTX-328は、試験したいかなる濃度又はインキュベーション時間でも内部移行を示さなかった(図4)。
発明者らはさらに、透過型電子顕微鏡法(TEM)を実行することによって処置細胞における超微細構造的変化を評価し、その際A375細胞を、培地中に直接溶解したペプチド又は培地単独のいずれかで処置した。低濃度(3.5μΜ)のLTX-315ペプチドで60分間処置された多数の細胞は、空胞化、及びミトコンドリアの形態のある程度の変化を示した(図5)。ミトコンドリアは、より低い電子密度であるようにみえ、ある程度の再編成も示し、クリステはさらに離れて存在するか、又は全く見えなかった。これらの試料中の壊死細胞の数は5%未満であった。これらの低濃度において、細胞質の空胞化が観察された。これらの試料における別の一般的な知見は、末梢に位置する小胞であり、それは均質材料を含有する単一膜層で裏打ちされていた(図5B)。細胞をより高い濃度(17μΜ)で60分間処置した場合、それらの約40%が、原形質膜完全性を喪失した壊死形態を示した(図5C&E)。依然として無傷であった細胞は、微絨毛を有する正常な外観~丸い外観の、かなりの異質性を示し、ミトコンドリアは明らかに影響を受けていた。この高い濃度では、調べた細胞のわずか4%が空胞化を示し、クロマチン凝縮は、試験したいかなるペプチド濃度でもこの物質中で見えなかった。これらの結果は、LTX-315が溶解作用モードで腫瘍細胞を死滅させる、より低い濃度は、細胞に、空胞形成及び変化したミトコンドリア形態などの、超微細構造の変化を受けさせることを実証する。さらに、アポトーシス細胞死を示唆する有意な形態学的変化は観察されなかった。
DAMPは、細胞の損傷時に細胞内供給源から放出される分子である。DAMPは、抗原提示細胞(APC)上のパターン認識受容体(PRR)への結合を介して免疫応答を開始及び永続化できる。一般的に知られているDAMPは、ATP、HMGB1、カルレティキュリン、チトクロムC、ミトコンドリアDNA及び活性酸素種(ROS)である。発明者らは次に、ATPがLTX-315処置細胞から上清中に放出されたかどうかを調べることを望んだ。したがって、処置細胞及び非処置細胞からの上清を、ルシフェラーゼ検出アッセイを用いて分析した。図7に示すように、ATPは、LTX-315での処置後5分ほどで上清中に検出され、放出は濃度依存的であった。
LTX-315処置細胞が培地中にチトクロムCを放出するかどうかを評価するために、A375細胞を異なる時間で(5、15、45分)、35μΜのLTX-315で処置した。上清を続いて、ELISAアッセイを用いて分析した。35μΜ値で処置した細胞は、未処置対照細胞と比較して、上清中に3倍超のチトクロムCを有した。チトクロムCの増加は、処置のわずか5分後に検出され、ペプチド処置の15及び45分後にもそれぞれ増加した(図8)。
HMGB1は、非ヒストン、クロマチン結合核タンパク質である。受動的に壊死細胞から放出されると、HMGB1は、いくつかの免疫賦活効果の中で樹状細胞の機能的成熟、サイトカイン刺激及び走化性を誘発できる。
LTX-315処置後のROS生成を、CH2DCFDA蛍光定量アッセイによって測定した。著しい量のROSが、LTX-315とのインキュベーションの15分後に発生し、ROSレベルは濃度依存的であった(図10)。
蛍光分子パシフィックブルーで標識したLTX-315は、A375黒色腫細胞とのインキュベーション後数分以内に内部移行し、細胞質中に分布した(図3)。低濃度では、ミトコンドリア周囲のペプチド蓄積が明らかであったが、より高い濃度では、ペプチドは細胞質内により多く広がり、細胞膜付近に円形構造で蓄積した。ペプチドがミトコンドリア膜を攻撃するならば、ミトコンドリア膜電位の減少又はさらには完全な崩壊が予想される。膜電位依存性ミトコンドリア染色Mitotracker CMXh2ROSでの細胞の共焦点画像は、ペプチド処置後短時間でミトコンドリアシグナルの喪失を示した(データ示さず)。シグナルの喪失は、ミトコンドリアとのペプチド相互作用が、ミトコンドリアの最も重要な細胞機能に不可欠である、ミトコンドリア膜電位の喪失を引き起こすことを示す。変化したミトコンドリア形態はまた、TEMを用いても実証された。LTX-315で60分間処置した細胞は、未処置細胞と比較して、クリステの組織が変化したより低い電子密度のミトコンドリア、及びミトコンドリア内の空胞化を有した(図6)。さらに、空胞化は、3.5μΜのLTX-315で処置した細胞の約20%において明らかであった。ミトコンドリアが機能不全である場合、遊離酸素ラジカル(ROS)が形成され、蛍光定量アッセイを使用することによって、発明者らはペプチド処置後数分以内のROS形成を実証した(図10)。
LTX-401は黒色腫細胞中でDAMP放出を誘発する
本研究において、発明者らは、癌細胞株中で細胞死を誘発するアミノ酸誘導体LTX-401の能力、及びマウス黒色腫モデルにおいて退縮を誘発する能力を調べた。作用様式のインビトロ研究は、処置細胞での大規模な細胞質空胞化及び損傷されたリソソームが先行する、溶解細胞死及び危険関連分子パターン分子の放出を明らかにした。マウス黒色腫モデルの使用は、LTX-401の腫瘍内注射で処置された動物の大部分が完全なかつ長期的な寛解を示したことを実証した。まとめると、これらの結果は、固形腫瘍の処置のための免疫療法剤としてのLTX-401の可能性を実証する。
1.1 試薬
LTX-401(Mwnet=367.53)は、要求に応じて、Synthetica AS(Oslo,Norway)によって合成された。化学構造を図11に示す。
ラット肝細胞癌JM1、いずれもヒト肝細胞癌であるHepG2及びBEL7402は、オスロ大学病院移植医学部のDr.Pal-Dag Lineディレクターの厚意により提供された。B16F1(ATCC、CRL-6323)、マウス皮膚悪性黒色腫、MDA-MB-435S(HTB-129)、乳癌転移に由来するヒト悪性黒色腫、Malme-3M(HTB-64)、肺転移由来のヒト悪性黒色腫、MRC-5(ATCC、CCL-171)、正常ヒト肺線維芽細胞、SK-N-AS(ATCC、CRL-2137)、骨髄転移由来のヒト神経芽細胞腫細胞株、HT-29(ATCC、HTB-38)、ヒト結腸直腸腺癌及びHUV-EC-C(ATCC、CRL-1730)、ヒト血管内皮細胞株は全て、アメリカン・タイプ・カルチャー・コレクション(American Type Culture Collection(ATCC-LGC Standards,Rockville,MD,USA)から購入した。ヒトのケラチノサイト細胞株HaCatは、Tromso大学の宿主微生物相互作用学部(Department of Host Microbe Interactions)のDr.Ingvild Pettersenの厚意により提供された。ヒト由来の悪性黒色腫細胞株A375は、Public Health England(PHE Culture Conditions,Porton,Down,Salisbury,UK)から購入した。JM1、A375、BEL7402、HepG2及びB16F1は全て、DMEM(高グルコース)からなる培地中で維持され、SK-N-AS、HT-29及びMDA-MB-435Sは、2mMのL-グルタミン及び重炭酸ナトリウムを含有するRPMI-1640培地中で培養した。Malme-3Mは、4mMのL-グルタミン及び20%FBS(ウシ胎児血清)を含有するRPMI-1640中で培養した。最後に、3つの非悪性細胞株HUV-EC-C、MRC-5及びHaCaTをそれぞれ、基礎培地及び増殖因子を含有するEGM-2 Bulletkit(Lonza,MA)、2mMのL-グルタミン及び1%非必須アミノ酸も含有するMEM(正常グルコース)並びに2mML-グルタミンを含有するDMEM(正常グルコース)を用いて培養した。溶液中2%FBSの最終濃度を得るために10mlのFBSのアリコートで送達されるEGM-2 Bulletkitを除いて、全ての培地に(特に断らない限り)10%FBSを補充した。
MTTアッセイを用いて、癌及び非悪性細胞株の両方の選択に対するLTX-401のインビトロ細胞傷害性を調べた。前培養した細胞を、1×104~1.5×104細胞/ウェルの密度で播種し、実験をCamilio K.A.ら、Cancer Immunol.Immunother,2014,63:601~13で以前に記載されたように行った。結果を、それぞれ3連のウェルで、3回の実験の平均値を用いて計算し、50%阻害濃度(IC50)として表した。
LTX-401の死滅動態を、それぞれ54μΜ及び108μΜに対応する2×IC50 4hと4×IC50 4hの両方の値を使用して、B16F1黒色腫細胞に対して研究した。細胞を、前にMTTアッセイについて記載しされるように播種し、5、15、30、60、90、120及び240分間LTX-401溶液とインキュベートした。細胞を、インキュベーション後に100μlの無血清RPMI-1640で1回洗浄し、さらに2時間、10%のMTT溶液(無血清RPMI-1640で希釈した)中でさらにインキュベートした。
B16F1細胞を、2mlの培地容量中1×104細胞の密度で35mm滅菌組織培養皿に播種し、37℃にて、95%超の湿度及び5%CO2条件で、細胞インキュベーター中で接着及び増殖させた。培養細胞が許容されるコンフルエンス(約80~90%)に達した時、それらを異なる時間(5、15、30及び60分間)、4×IC50 4h値のLTX-401(108μΜ)とインキュベートし、続いて0.05%のマラカイトグリーンシュウ酸塩(Sigma)、0.5%のグルタルアルデヒド(GA,Electron Microscopy Sciences)及び4%のホルムアルデヒド(FA、電子顕微鏡科学)を含有するPHEM緩衝(0.1M)溶液中で固定した。マラカイトグリーンは、試料処理と一般的に関連する脂質抽出を減少させるために、一次固定への添加剤としてより頻繁に使用されている。試料調製を数日から数時間に低減するので、従来のベンチ・プロトコルよりも有利であると考えられる新たに導入された方法であるPELCO Biowave Pro Laboratory Microwave(Ted Pella社)を用いて、試料をすぐに添加し、一括処理した。全てのマイクロ波手順については、試料を50℃カットアウト温度で、23℃にてマイクロ波照射した。マラカイトグリーン/GA/FAとオスミウム還元フェロシアン化物(0.8%のK3Fe(CN)6/1%のOsO4)の両方の固定工程は、真空で、2分オン、2分オフ(100W)の「電源オン/オフ」サイクルで実行した。試料を、各工程の間に0.1M PHEM緩衝液で4回すすぎ(250Wで40秒間、2回のベンチリンス及び2回の最終リンス)、その後、真空で、1分間オン、1分間オフ(150W)の「電源オン/オフ」サイクル下で、1%タンニン酸(Electron Microscopy Sciences,PA,USA)及び1%水性酢酸ウラニル(Electron Microscopy Sciences,PA,USA)で染色した。試料を、250W(真空オフ)で水中にて2回マイクロ波照射することに加えて、前に記載されるように各染色手順の間にすすいだ。試料の脱水は、段階的エタノール系列(25%、50%、70%、90%及び100%)を通して生じ、各グレードで40秒間250Wでマイクロ波照射し(真空オフ)、その後50%エポン樹脂(AGAR、DDSA、MNA及びDNP-30)で、並びに2つの最終浸透工程については100%に増加して、250Wでそれぞれ3分間(真空サイクル;20秒オン、20秒オフ)一括して浸漬した。試料を次いで60℃で一晩重合させた。超薄切片(70nm)を調製し、標準ホルムバール、炭素安定化銅グリッド上に置いた。次に、試料をJEOL JEM-1010透過型電子顕微鏡で検査し、Olympus MoradaサイドマウントTEM CCDカメラ(Olympus soft imaging solutions,GmbH,Germany)で画像を取得した。
B16F1細胞を、6ウェルプレート中の完全培地に2×105細胞/ウェルの密度で播種し、一晩接着させた。細胞を108μΜのLTX-401(4×IC50 4h)で処置し、異なる時間(10、30、60、90及び120分間)、37℃(95%超の湿度及び5%CO2)でインキュベートした。無血清RPMI-1640を陰性対照として使用し、上清(S)を収集し、5分間1,400gで遠心分離し、その後Ultracel-50膜(Milipore,Norway)と共にAmicon Ultra-0.5遠心フィルターユニット(Centrifugal Filter units)を用いて濃縮アップした。細胞溶解物(L)を2mLの無血清RPMI-1640で洗浄した後に収集し、2×NuPAGE LDS試料緩衝液(Invitrogen,Norway)、1×NuPAGE試料還元剤(Invitrogen,Norway)及び50%滅菌水を含有する溶解緩衝液(mastermix)に供した。上清と溶解物の両方を10分間70℃で煮沸し、NuPAGE Novex 4~12% Bis-Trisゲル(Millipore,Norway)上で分離し、その後ポリフッ化ビニリデン(PVDF)膜(Millipore)に電気移動させた。膜を、TBS-T中5%無脂肪乾燥ミルクを用いて1時間ブロッキングし、次にHMGB1抗体(HMGB1に対するウサギポリクローナル、ChIPグレード、Abcam,UK)と一晩4℃でハイブリダイズさせ、続いて西洋ワサビペルオキシダーゼ(HRP)結合二次抗体(ヤギポリクローナル抗ウサギIgG,Abcam,UK)と室温でさらに2時間ハイブリダイズさせた後に、WBルミノール試薬(Santa Cruz Biotechnology,Heidelberg,Germany)を使用して発色させ、ImageQuant LAS 4000及びImageQuantソフトウェア(GE Healthcare)を用いて画像化/スキャンした。
B16F1細胞を、MTTアッセイについて前に記載されるように、96ウェル培養プレート上にプレーティングした。細胞を、指定した時間(30、60、90、120及び240分間)108μΜ(4×IC50 4h)で処置し、対照細胞は、実験の終点まで無血清RPMI-1640単独中で保存した。上清を収集し、無血清RPMI-1640中で1/5希釈し、その後製造業者の指示に従って、ラット/マウスのチトクロムcのQuantikine ELISAキット(R&D Systems,USA)を使用して評価した。チトクロムcの濃度を、提供される標準曲線上の補間値により計算した。
B16F1細胞を、MTTアッセイについて前に記載されるように播種し、異なる時間(10、30、60、90及び120分間)2×IC50 4h値のLTX-401(54μΜ)で処置した。無血清RPMI-1640処置細胞は、それぞれ陰性対照及びブランク対照として機能した。ATPの細胞外レベルを、ルシフェリンベースのENLITEN ATPアッセイキット(Promega,Madison,WI,USA)を用いて実験の終了時に測定し、その際、ATP駆動型化学発光を、発光マイクロプレートリーダー(Labsystems Luminoskan(登録商標),Finland)で記録し、相対的光単位(RLU)として表した。
フローサイトメトリー:B16F1細胞を6ウェルプレートに播種し、一晩接着させた。細胞をその後60分間、1×IC50 4hのLTX-401(27μΜ)で処置し、40nMのリソトラッカーDND-26(Invitrogen)を最後の5分で添加した。未処置細胞を対照として使用した。細胞をトリプシン処理し、FACSキャリバーフローサイトメーター上で調査し、結果をFLowJoソフトソフトウェア(Tree Star社,Ashland,OR,USA)を用いて処理した。損傷した原形質膜を有する細胞をゲーティングから外すために、PIをいくつかの実験で利用した。
結果を、少なくとも2つの独立した実験の平均±平均の標準誤差(SEM)又は標準偏差(SD)として示す。MTTアッセイを3つ並行して2回行い、チトクロムcアッセイを2つ平行して2回行い、ATPアッセイを2つ平行して3回行った。チトクロムc放出データ及びATP放出データを、一方向ANOVA及び多重比較検定を用いて比較し、発明者らは0.05以下のp値が統計学的に有意であるとみなした。
2.1 LTX-401は細胞死を急速に誘発する
本研究では、LTX-401は、インビトロでいくつかの腫瘍細胞株の生存率を効果的に低下させた(データ示さず)。LTX-401は、ヒト悪性黒色腫細胞株MDA-MB-435S(13.5μΜ)に対して最高の細胞傷害活性を示し、ヒト肝細胞癌細胞株HepG2(35.4μΜ)に対して最低の活性であった。残りの細胞株について、LTX-401は、19~32μΜの範囲内でわずかに変化する類似のIC50値を示した。
LTX-401の細胞傷害活性の基礎をなす作用機序をさらに調べるために、B16F1細胞を4×IC50 4hのLTX-401(108μΜ)とそれぞれ5分間及び60分間インキュベートした。未処置細胞を対照とし、実験の終点(60分)まで無血清RPMI-1640中で培養した。インキュベーション後に、全ての細胞をTEM研究用に固定し調製した。未処置B16F1細胞のTEM画像は、原形質膜上の頻繁な微絨毛状突起により特徴付けられる粗い表面を明らかにした(図13a、b)。細胞質は、いくつかの電子密度の高いミトコンドリア、並びに視覚的に平滑なER及びゴルジ装置からなった(図13a、b)。LTX-401処置細胞の大部分は、5分後にそれらの表面形態を失い、細胞の大部分は、細胞質のわずかな空胞化を除いて顕著な変化はなかった(図13c)。インキュベーションの60分後に、処置した培養物は、高度に空胞化した細胞と大きな非空胞化細胞の両方を含む、細胞の不均一な集団を示した(図13e)。原形質膜の完全性の喪失及び細胞構成成分の漏れにより明らかにわかるように、細胞の大部分は壊死した(図13e、f)。クロマチン構造は、いくつかの細胞でわずかに凝縮されたが、程度の差はあるもののLTX-401処置によって影響されなかった。さらに、より高い倍率は、対照(図13g)と比較して、LTX-401処置細胞(図13h)の大部分のミトコンドリアでの顕著な超微細構造変化を示さなかった。60分間処置した細胞(図13h)は、ある程度のミトコンドリアの膨潤を有するものの、無傷の内側と外側のミトコンドリア膜を示した。全体的に、これらの観察は、LTX-401が、細胞腫脹、ミトコンドリアの膨張及び細胞質の空胞化を伴い、溶解性作用機序により死滅させることを実証する。
次に、発明者らは、DAMPの放出を誘発するLTX-401の能力を特徴づけることを望んだ。HMGB1は、非ヒストン核タンパク質であり、細胞外に放出されると、toll様受容体(TLR)又は終末糖化産物受容体(RAGE)に結合することによってDAMPとして作用できる。B16F1細胞から細胞培養上清へのHMGB1の放出を、ウェスタンブロット分析により評価した。HMGB1の移行が検出され、HMGB1の放出は処置の30分後に生じた。溶解物中のタンパク質の完全留置によりわかるように、非処置対照細胞は、上清中へのHMGB1の放出を示さなかった(図14a)。
次に、発明者らは、LTX-401がリソソーム完全性に何らかの変化を引き起こすかどうかを調べることを望んだ。フローサイトメトリー及び共焦点顕微鏡で示されるように、LTX-401での処置は、好酸性染料リソトラッカーDND-26からのシグナルの喪失をもたらした。この染料は、リソソーム及びメラノソームなどの酸性オルガネラ中に蓄積する。同様の結果がリンパ腫細胞でも得られたので、この効果は、細胞型特異的ではないことが示された(データ示さず)。フローサイトメトリーと共焦点顕微鏡の両方によって示されるように、27μΜのLTX-401は、インキュベーションの60分後に、B16F1黒色腫細胞でシグナルを減少させた。
免疫監視を回避する癌細胞の能力は、癌の新たな特質として最近認められている。免疫原性細胞死(ICD)は、危険関連分子パターン分子(DAMP)と呼ばれる免疫増強分子の放出として定義され、従って癌細胞に対する免疫応答を確立する上で重要である。ICDを誘導するいくつかの細胞増殖抑制化合物の能力に起因して、免疫系が従来の処置への応答として、癌撲滅に重要な役割を果たしていることも示されている。ICDはさらに、抗癌治療の長期的な成功のための決定要因として示唆されている。腫瘍溶解性療法は、癌細胞をその場で溶解しその後のDAMPの放出を伴う、固形腫瘍に対する新しい有望な治療的アプローチである。
LTX-315はマウス黒色腫モデルにおいてT細胞クローン性を増加させる
1.材料及び方法
本研究は、Adaptive Biotechnologies(商標)による「immunoSEQ(商標)」T細胞受容体(TCR)のレパートリー特性評価プラットフォームを用いて行った。このプラットフォームは、高度に最適化されたプライマーセットを有する新規なマルチプレックスPCR1と、TCR遺伝子のみを標的にするディープシーケンシング技術を兼ね備える。immunoSEQプラットフォームは、研究者が例外的な深度及び特異性で適応免疫系を分析するのを可能にする。
マウスB16黒色腫細胞を収集し、洗浄し、10匹のマウスに皮内注射した。腫瘍注射後8日目に、5匹のマウスに1回又は2回、LTX-315(1.0mgのLTX-315/50μl生理食塩水)の単回腫瘍内注射を用いて、ペプチド処置を開始した。生理食塩水のみのビヒクル対照(滅菌水中0.9%のNaCl)を、他の5匹のマウスに投与した。動物を次いで15日目に安楽死させた。
腫瘍組織(10mg)及び全血(150~200μl)を、TCRレパートリー特性分析のために各マウスから採取した。血液試料は末梢における免疫レパートリーに関する情報を提供し、腫瘍試料はレパートリーの重点的な見解を提供する。
クローン性は、クローン頻度分布の形状を測定することにより、モノクローナル又はオリゴクローナル増殖の程度を定量する。値は0~1の範囲であり、1に近づく値は、ほぼモノクローナルの集団を示す。図15は、0.05(A)及び0.32(B)のクローン性を有する集団の例示的な分布曲線を提供する。
結果の要約を以下の表2に示す。
LTX-315は防御免疫応答を誘発する
LTX-315処置によって治癒された動物は、皮内及び静脈内の両方でのB16腫瘍生細胞の再負荷に対して保護された。まとめると、データは、LTX-315での腫瘍内処置が、局所腫瘍制御に続く防御免疫応答を提供でき、新しい免疫療法剤としての可能性を有することを示す。
本研究は、CAMILIO KAら,cancer Immunol.Immunother.2014,63:601-13に記載の通りに行った。
LTX-315及びLTX-328(K-A-Q-Dip-Q-K-Q-A-W-NH2)を、要求に応じて、それぞれBachem AG(Bubendorf,Switzerland)及びInnovagen(Lund,Sweden)から購入した。ダカルバジン(D2390)、テモゾロミド(T2577)及びシスジアンミン白金(II)ジクロリド(P4394)は全て、Sigma-Aldrichから購入した。
C57BL/6マウス由来の皮膚悪性黒色腫B16F1(ATCC、CRL-6323)、ヒト胎児肺線維芽細胞株MRC-5(ATCC、CCL-171)及びヒト臍帯静脈内皮細胞株HUV-EC-C(ATCC、CRL-1730)は全て、アメリカン・タイプ・カルチャー・コレクション(ATCC-LGC Standards,Rockville,MD,USA)から購入した。A375(ECACC、88113005)は、患者材料由来のヒト悪性黒色腫であり、Public Health England(PHE Culture Collections,Porton Down,Salisbury,UK)から購入した。B16F1及びA375細胞をDMEM(高グルコース)中で培養し、MRC-5細胞を、2mMのl-グルタミン(全て)及び1%非必須アミノ酸(A375のみ)を含有するMEM(正常グルコース)中で培養した。初代表皮メラニン細胞(ATCC、PCS-200-013)を、アダルトメラノサイト増殖キット(ATCC、PCS-200-042)を補充したDermal Basal Medium(ATCC,PCS-200-030)中で培養した。HUV-EC-Cを、LonzaからのEGM-2 Bulletkitを用いて培養し、全ての増殖培地は、抗生物質を含まず、10%FBSを補充した(無血清初代メラノサイトを除いて)。細胞培養を、37℃にて5%CO2及び95%超湿度の加湿雰囲気中で維持し、マイコプラズマと他の病原体(Rapid-MAPTM-27,Taconic,Europa)の両方又はマイコプラズマ単独のいずれかについて試験した。
5~6週齢の雌C57BL/6野生型マウスを、英国のCharles Riverから入手した。全てのマウスを、地方及びヨーロッパの倫理委員会のガイドラインに従って病原体を含まない動物施設内のケージに収容した。
腫瘍細胞を収集し、RPMI-1640で洗浄し、C57BL/6マウスの腹部の右側に皮内(i.d.)注射した(マウスあたり5×104個のB16F1細胞/50μlのRPMI-1640)。触知可能な腫瘍(20~30mm2)を、生理食塩水中に溶解したLTX-315又はLTX-328(1.0mgのペプチド/50μlの生理食塩水)の単回用量で3日間連続して1日1回、i.t.注射し、ビヒクル対照は生理食塩水のみであった(滅菌水中0.9%のNaCl)。腫瘍の大きさを、電子キャリパーを用いて測定し、楕円の面積として表した[(最大寸法/2)×(最小寸法/2)×Π]。垂直腫瘍寸法の積が130mm2に達した時、又は腫瘍潰瘍が発症した時に、動物を次いで安楽死させた。
LTX-315処置後に腫瘍が完全退縮(CR)した動物に、それらがLTX-315で治癒された4~5週間後に、腹部の左手側(最初の腫瘍部位に対して反対側)に第2のi.d.腫瘍細胞負荷(5×104個のB16F1細胞)を与えた。このi.d.腫瘍再負荷後に生存している動物に、その後、尾静脈を介して第2の腫瘍再負荷を静脈内で(i.v.)(2×105細胞)与えた。肺をi.v.再負荷後19日目に摘出した。全てのマウスを、腫瘍の大きさ及び生存について監視した。
全てのデータは、少なくとも3回の独立した実験を表し、平均値±標準偏差(SD)又は平均値の標準誤差(SEM)として表される。動物の生存曲線(カプランマイヤープロット)を、ログ・ランク(マンテル・コックス)検定を用いて比較した。発明者らは、p値≦0.05が統計的有意性を示すとみなした。
LTX-315での処置が、適応免疫応答を誘発できるかどうかを調べるために、治癒された動物(n=25)に、非処置対照動物(n=6)と共に、CRが達成された4~5週間後に、最初の腫瘍部位に対して反対側の腹部に5×104個の生存腫瘍細胞をi.d.で再負荷した。このi.d.腫瘍再負荷後CRに達した動物に、後に2回目のi.v.再負荷を行った。LTX-315により治癒された動物の大半は、成長阻害を示し、25匹中15匹の動物で腫瘍増殖はなかったが、全ての対照動物は、i.d.再負荷後に腫瘍を発症した(図21c)。以前にLTX-315のi.t.注射によって治癒された動物(n=7)は、未処置対照動物(n=7)と比較して、i.v.再負荷後にB16黒色腫に対する全身免疫防御を示した(図21d、e)。LTX-315治癒動物(腫瘍病巣の平均数=15.86)は、未処置対照動物(腫瘍病巣の平均数=123.3)と比較して、顕著により低い数の肺腫瘍病巣を示した。さらに、組織学的検査は、以前にLTX-315によって治癒された動物の肺腫瘍病巣が、未処置対照動物のあまり浸潤していない肺腫瘍病巣と比較して、CD3+T細胞で顕著に浸潤されたことを明らかにした(図21f)。
インビボでのLTX-315の免疫調節特性が長期防御免疫応答をもたらすかどうかを調べるために、動物を生存腫瘍細胞で再負荷した。全ての以前に治癒された動物は、有意な腫瘍増殖阻害を示し、生存細胞をi.d.投与した動物の60%で腫瘍取り込みは全く観察されなかった(図21a~c)。癌に対する可能性のある長期的な全身保護及び実験的肺転移モデルでのその効果を調べるために、i.d.再負荷後に生存している動物にLTX-315処置後9ヶ月で、生存B16黒色腫細胞をi.v.再負荷した。i.v.再負荷された全ての動物は、抗CD3での免疫標識によって見られるように(図21f)、再負荷された肺腫瘍病巣へのCD3+T細胞浸潤の量の増加に加えて、未処置対照動物(図21d、e)と比較してB16黒色腫に対する全身免疫防御を示した。これは、LTX-315が、T細胞媒介性抗腫瘍免疫の持続性、ひいては腫瘍の再発及び肺腫瘍病巣に対する長期的な保護と共に、全身抗腫瘍免疫応答を誘発することを示している。LTX-315の機構は、膜誘導性溶解及びDAMPの細胞外放出(HMGB1)を介してB16黒色腫に対する長期的な特異的細胞性免疫を誘発することによるものであると考えられる。まとめると、インビトロ及びインビボでの発明者らの観察は、LTX-315のi.t.投与が、腫瘍細胞の細胞膜上のペプチドの直接的な破壊効果によって開始される広範な腫瘍壊死をもたらすことを示している。さらに、LTX-315の壊死効果は、免疫応答及び腫瘍実質へのTILの浸潤を刺激するDAMPの放出をもたらし、これは、持続性の腫瘍免疫防御を誘発することにおけるそれらの可能性のある役割のため、固体B16黒色腫の根絶に重要であり得る。
LTX-315の投与後に作製されるT細胞はT細胞退縮に重要な役割を果たす
これから、発明者らは、LTX-315が、多機能性Tヘルパータイプ1/タイプ1細胞傷害性T細胞の頻度を増加させることによって腫瘍の微小環境を急速に再プログラムすることを示す。この増加は、腫瘍退縮において重要な役割を果たしている。
1.1 化学物質及び細胞培養
細胞培養のための培地及び栄養補助剤をGibco-Life Technologies(Carlsbad,CA,USA)から入手し、Lytix Biopharma (Tromso,Norway)によって提供されたLTX-315を除く化学物質をSigma-Aldrich(St.Louis,MO,USA)から入手し、プラスチック製品は、Corning BV Life Sciences (Amsterdam,The Netherlands)からのものであった。MCA205を、10%ウシ胎児血清、並びに2mMのl-グルタミン、100 IU/mlのペニシリンGナトリウム塩、100μg/mlの硫酸ストレプトマイシン、1mMのピルビン酸ナトリウム及び1mMの非必須アミノ酸を補充したRPMI-1640培地中で培養した。細胞を、5% CO2雰囲気下にて加湿インキュベーター中で、37℃で増殖させた。
マウスを、12時間の明と12時間の暗のサイクルで温度制御環境にて特定の病原体を含まない条件下で維持し、食物及び水を自由に与えた。欧州連盟実験動物学協会(FELASA)ガイドラインに従う動物実験は、EU指令63/2010に準拠しており、Gustave Roussy Cancer Campus(Villejuif,France)の倫理委員会によって承認された。7週齢~14種齢のマウスを使用した。WT特異的病原体フリー(SPF)C57BL/6 Jを、Envigo(Gannat,France)から入手し、Gustave Roussy,Villejuif,フランスで、SPF条件下で維持した。
マウスに、1×106個のMCA205細胞を右脇腹に皮下注射した。腫瘍細胞株を、C57BL/6マウスに接種した。腫瘍表面(最長寸法×垂直寸法)を、日常的にキャリパーによって監視した。腫瘍が20~25mm2の大きさに達したときに(0日目)、マウスに、300μgのLTX-315を3日連続、注射で毎日腫瘍内投与した。T細胞枯渇実験(図24)において、抗CD4及び抗CD8モノクローナル抗体(mAB)(それぞれGK1.5及び53~6.72;マウスあたり200μg)又はそれらのアイソタイプ対照(それぞれLTF-2及び2A3)を、最初のLTX-315の注入の3及び4日前に腹腔内注射し、7日おきに継続した。インビボでの使用のための全てのmAbを、推奨されるアイソタイプ対照のmAbを使用して、BioXcell(West Lebanon,NH,USA)から入手した。
腫瘍及び脾臓を、LTX-315の最初の注射後7日目に摘出した。切除した腫瘍を小片に切断し、25μg/mLのリベラーゼ(Roche,Boulogne-Billancourt,France)及び150 UI/mlのDNアーゼ1(Roche)を含有するRPMI-1640培地中で、37℃で30分間消化した。混合物をその後、100μmのセルストレーナーを通して継代した。2×106個の脾細胞(赤血球溶解後)又は腫瘍細胞を、膜染色前に、精製した抗マウスCD16/CD32(93;eBioscience,San Diego,CA,USA)と4℃で15分間プレインキュベートした。細胞内染色のために、FoxP3染色キット(eBioscience)を使用した。死んだ細胞を、生/死修正可能黄色死細胞染色キット(Live/Dead Fixable Yellow dead cell stain kit(Life Technologies,Carlsbad,CA,USA))を使用して除外した。サイトカイン染色のために、細胞を、50ng/mlのホルボール12-ミリステート13-アセテート(PMA;Calbiochem,San Diego,CA,USA)、1μg/mlのイオノマイシン(Sigma,St.Louis,MO,USA)、及びBD Golgi STOP(BD Biosciences,San Jose,CA,USA)を用いて37℃で4時間刺激した。抗IFN-γ(XMG1.2)及び抗TNF-α(MP6-XT22)を、eBioscienceから購入した。抗CD4(GK1.5)、抗CD8β(YTS1567.7)を、Biolegend(San Diego,CA,USA)から購入した。8色のフローサイトメトリー分析を、フルオレセインイソチオシアネート、フィコエリトリン、フィコエリトリンシアニン7、ペリジニンクロロフィルタンパク質-シアニン5.5、アロフィコシアニン-シアニン7、パシフィックブルー又はアロフィコシアニンに結合された抗体を用いて行った。全ての細胞を、FlowJo(Tree Star,Ashland,OR)ソフトウェアを使用して、CyAn ADP(Beckman Coulter,Marseille,France)フローサイトメーター上で分析した。
データを、マイクロソフト社のエクセル(Microsoft Co.,Redmont,WA,USA)及びプリズム5(GraphPad,San Diego,CA,USA)を用いて分析した。データを平均±S.E.M.として表し、P値をスチューデントt検定又は一方向ANOVA、その後、該当する場合、テューキー(Tukey)検定によって計算した。カプランマイヤー生存曲線の比較を、ログランクマンテル・コックス検定を用いて行った。全ての報告した検定は、両側検定であり、P値<0.05で有意とみなした。
発明者らは、皮下肉腫モデルにおいてLTX-315投与後7日目に形作られた腫瘍微小環境を構成する主要エフェクターの動態を検討した。発明者らは、IFNγ+(Tヘルパー1型、Th1)、IL-17+(Th17)及び二重陽性IFNγ+ IL-17+(pTh17)CD4+TIL(図22)、並びに多機能性IFNγ+ TNFα+ CD8+ T細胞(図23)の蓄積を観察した。
この前臨床研究において、発明者らは、LTX-315が、典型的なアポトーシス又は調節された壊死性細胞死を誘発せずに、制御されていない免疫原性腫瘍細胞死を引き起こすことを見出した。この細胞死の少なくとも一部は、多機能性CD4+及びCD8+ TILの蓄積を促進することを含む。免疫原性作用機序は、Tリンパ球の非存在下で抗腫瘍効果が消失されたという事実によって、重要であることが示される。
LTX-315の投与後の腫瘍へのT細胞浸潤の臨床組織学的証拠
1 材料及び方法
患者からのホルマリン固定パラフィン包埋腫瘍組織切片を、キシレン及び段階的アルコールで脱パラフィンし、水和し、PBSで洗浄した。電子レンジでのクエン酸ナトリウム緩衝液(pH6)中の抗原回復後に、内因性ペルオキシダーゼを0.3%のH2O2によって15分間ブロッキングした。切片を、4℃で一晩、一次抗体;ウサギポリクローナル抗CD3(クローンA0452 Dako)又はマウスモノクローナル抗CD8(clone OX8,ab33786,Abcam)とインキュベートした。二次抗体として、抗ウサギ西洋ワサビペルオキシダーゼ(HRP)SuperPicTureポリマー検出キット(Invitrogen)又はエンビジョンシステムHRP抗マウス(Dako)を使用した。一致したアイソタイプ対照を、非特異的バックグラウンド染色の対照として使用した。
図25~29は、転移性黒色腫、悪性黒色腫、ミオ上皮腫、乳癌及びデスモイド腫瘍患者の腫瘍への細胞傷害性T細胞の浸潤を示す。
LTX-315処置は二次性及び原発性腫瘍への細胞傷害性T細胞浸潤をもたらす
1つの腫瘍病巣におけるLTX-315の腫瘍内注射が転移性疾患にも影響を与え得るかどうかを明らかにするために、ラット肉腫モデルにおいて腹腔内腫瘍及び2つの皮下腫瘍を確立した。その後、LTX-315を皮下病巣の1つに注入し、腫瘍増殖をライブイメージングによって評価した。結果は、LTX-315の処置が全3つの病巣を根絶し、動物が耐久性のある完全寛解状態に入ったことを示した。
1.1 動物実験
近交系Piebald Virol Glaxo(PVG.RT7a,略称PVG)系統のラットを、PVG.RT7B系統(本研究でPVGと略す)と互換的に使用した。これらの系統は、白血球共通抗原(LCA/CD45)ファミリの1つの無関係なエピトープを除いて同一である。PVGラットは、Harlan(the Netherlands)から購入し、PVG.7Bラットは、基礎医学科学研究所(Basic Medical Sciences(IMB,University of Oslo,Norway))での自家繁殖から入手した。実験の間、240~270gの重量の雄ラットを、12時間の明/暗サイクル及び周囲温度で、気候制御された条件下にてケージ当たり2~3匹の動物のグループで飼育した。ラットを、標準の齧歯類用固形飼料及び水を自由に利用できるようにして、強化された個別通気ケージ(IVC)システムに収容した。動物を、実験手順中に、十分な程度の鎮静及び鎮痛を提供する、2.5%のイソフルランガス(Baxter Medical AB)又はフェンタニル/フルアニゾン(Hypnorm;VetaPharma社)の皮下注射(0.4ml/kg)のいずれかで麻酔した。動物を毎日監視し、大きな腫瘍を有するラットをCO2で安楽死させた。実行した全ての手順は、FOTS番号1957及び5917の下で行われ、ノルウェー農業省の実験動物委員会によって承認され、実験及び他の科学的な目的のために使用される脊椎動物の保護のための欧州条約を遵守した。実験動物施設は、日常の健康監視プログラムの対象となっており、欧州実験動物学会連合の勧告の改定に従って一般的な病原体についてスクリーニングした。
前培養したラットの形質転換間葉系幹細胞由来の肉腫モデル細胞(rTMSC)を、無血清RPMI-1640中に収集し、200,000個のrTMSCを、半日目にPVGラットの右脇腹に皮下接種し、20,000個のrTMSCを0日目に反対の脇腹に皮下接種した。確立された腫瘍(平均の腫瘍の大きさ25mm2)に、LTX-315(0.9% NaClを有する滅菌水に溶解した)(n=8)又はビヒクル(0.9% NaClを有する滅菌水)(n=6)を病巣内注射した。20mg/mlにて50μl(1mg)のLTX-315処置用量を、1mlのシリンジ(Myjector U-100,Terumo)、0.5×16mm針(Fine-Ject;Henke-Sass,Wolf GmbH)を用いて与えた。注射を、3日連続して毎日提供した。腫瘍の大きさを、キャリパーを用いて週に3回測定し、楕円の面積[(最大寸法/2)×(最小寸法/2)]として表した。腫瘍が400mm2を超えた場合に、動物の命を終了させた。
単一細胞懸濁液を、LTX-315処置の翌週の間の異なる時点で、新鮮な腫瘍組織から調製した。腫瘍をかみそりの刃で穏やかにミンチし、小片(4mm2)に切断した。腫瘍組織を、37℃で60分間穏やかに撹拌しながら、10mlのMEM培地(Sigma-Aldrich)中0.18 Wunsch単位/mlの濃度でのリベラーゼTM(Thermolysin Medium;Roche Diagnostics)とインキュベートした。酵素消化を、4℃で2mlのFCS(Invitrogen,Thermo Fischer)の添加によって停止させた。細胞懸濁液を、70μΜメッシュ(Cell Strainer;BD)を通して濾過し、PBSで洗浄し、細胞を次いでフローサイトメトリー染色のために直接使用した。
CD4(OX38)及びCD8(OX38)に対するマウスモノクローナル抗体は、ハイブリドーマからの培養上清から単離され、英国オックスフォードのMRC Cellular Immunology Unitからの贈り物であった。それらを、標準的なプロトコルに従ってIMBで結合させた。CD3に対する蛍光色素結合mAb(G4.18)を、PerCPストレプトアビジンを含めて、BD Biosciences社から入手した。FITC/AI488、PE、PerCPストレプトアビジン及びAI647中の試薬からなる4色パネルを、CellQuest software(BD)を備えたFACSキャリバー(BD)上にアプライし分析した。ドットプロット及びヒストグラムゲートは、アイソタイプ対照抗体を使用して設定した。
ホルマリン固定パラフィン包埋組織切片を、キシレン及び段階的アルコールで脱パラフィンし、水和して、PBSで洗浄した。電子レンジでクエン酸ナトリウム緩衝液(pH6)中の抗原を回収した後、内因性ペルオキシダーゼを0.3%のH2O2によって15分間ブロッキングした。切片を、一次抗体;ウサギポリクローナル抗CD3(クローンA0452 Dako)又はマウスモノクローナル抗CD8(クローンOX8、ab33786、Abcam)と4℃で一晩インキュベートした。二次抗体として、抗ウサギ西洋ワサビペルオキシダーゼ(HRP)SuperPicTureポリマー検出キット(Invitrogen)又はエンビジョンシステムHRP抗マウス(Dako)を使用した。一致したアイソタイプ対照を、非特異的バックグラウンド染色のための対照として使用した。
データを、平均±SDとして表す。2つのグループ間の統計的優位性を、両側スチューデントt検定によって分析し、P<0.05を統計学的に有意であるとみなした。統計分析を、GraphPad Prismソフトウェア(バージョン6、GraphPad)を用いて行った。
LTX-315媒介退縮及び長期防御免疫応答の基礎となる細胞メカニズムを調べるために、発明者らは、処置した腫瘍の細胞組成をフローサイトメトリー及び免疫組織化学によって分析した。最後の処置の翌週の間、腫瘍を異なる時点で切除し、その後LTX-315処置動物及び未処置動物の両方で腫瘍浸潤白血球の表現型を決定した(図30a)。
Claims (10)
- 腫瘍浸潤T細胞集団を生成する方法であって、腫瘍細胞膜を溶解できる正電荷を有する両親媒性ペプチドで処置された対象から採取された細胞腫瘍試料からT細胞集団を単離することを含む方法であって、
前記ペプチドが、LTX-315(SEQ ID NO:23)、若しくはその塩、又はLTX-401、若しくはその塩である、方法。 - 前記T細胞集団を培養する工程をさらに含む、請求項1に記載の方法。
- 前記T細胞集団をエクソビボで増殖させる工程をさらに含む、請求項1又は請求項2に記載の方法。
- 前記T細胞集団からの1以上のクローン型の特定及び/又は単離をさらに含む、請求項1~3のいずれか一項に記載の方法。
- それらの対応する腫瘍特異的抗原又はネオ抗原を特定するために作製されたT細胞集団を分析する工程をさらに含む、請求項1~4のいずれか一項に記載の方法。
- 前記分析が、(a)前記T細胞集団がエクソビボで増殖された後に、又は(b)単離T細胞集団上で直接行われる、請求項5に記載の方法。
- 請求項1~4のいずれか一項に記載の方法により得られる、単離T細胞集団。
- 腫瘍細胞を処置する又は腫瘍の増殖、確立、拡散若しくは転移を予防する又は低減することにおける使用のための、請求項7に記載の単離T細胞集団。
- 腫瘍細胞を処置する又は対象において腫瘍の増殖、確立、拡散若しくは転移を予防する又は低減するための医薬の製造における請求項1~4のいずれか一項に記載の方法により得られる、単離T細胞集団の使用。
- 請求項1~4のいずれか一項に記載の方法により得られる、単離T細胞集団であって、腫瘍細胞を処置する又は腫瘍の増殖、確立、拡散若しくは転移を予防する又は低減することにおける使用のための、単離T細胞集団であって、チェックポイント阻害剤と共に投与される、単離T細胞集団。
