Description
されるアミノ酸配列の何れかの配列を含むことを特徴とする。
(1)ペプチドライブラリーを作製後、ベクターに挿入して組み換えファージを製造する段階と、
(2)前記組み換えファージとアセチルコリン受容体とを混合した後、バイオパニング(biopanning)を実施してアセチルコリン受容体と結合するファージを選別する段階と、
(3)前記段階(2)で選別されたファージに対して、アセチルコリン受容体と、対照群との酵素免疫分析(ELISA)を実施する段階と、
(4)前記酵素免疫分析の実施結果を分析し、対照群に対してアセチルコリン受容体と結合シグナルの強度が1.5倍以上であるファージを選別する段階と、を含むアセチルコリン受容体結合ペプチドのスクリーニング方法を提供することを本発明のまた別の側面とする。
ましくは90%以上の相同性を示す塩基配列を含むポリヌクレオチドになることができ、最も好ましくは95%以上の相同性を示す塩基配列を含むポリヌクレオチドになることができる。
これを粉砕し、適合な補助剤を添加した後、顆粒混合物で加工することによって、錠剤又は糖衣錠剤を収得することができる。適合な賦形剤の例としては、ラクトース、デキストロース、スクロース、ソルビトール、マンニトール、キシリトール、エリスリトール、及びマルチトール等を含む糖類と、トウモロコシ澱粉、ミール澱粉、米澱粉、及びジャガイモ澱粉等を含む澱粉類、セルロース、メチルセルロース、ナトリウムカルボキシメチルセルロース、及びヒドロキシプロピルメチル-セルロース等を含むセルロース類、ゼラチン、ポリビニルピロリドン等のような充填剤が含まれることができる。また、場合によって、架橋結合ポリビニルピロリドン、寒天、アルギン酸、又はナトリウムアルジネート等を崩壊剤として添加できる。
ベクター溶液4μLと混合した後、冷却して準備された0.2cmのキュベットに入れた後、1分の間に氷上に置いた。
めに、OD=0.7であるE.coilに混ぜてアンピシリンが含まれたagar培地に塗抹した。パニングを繰り返すために、outputファージ500μLを5mLのE.coilと混ぜて、37℃で200rpmの速度で30分間混合して培養した後、アンピシリン(50μg/mL)及びヘルパーファージ(1×1010pfu)添加し、同じ方法で30分間培養した。その後、アンピシリン(50μg/mL)とカナマイシン(10μg/mL)が含まれた50mLのSB培地に移して、同じ方法で一日間培養した。培養液を4℃で5,000rpmの速度で10分間遠心分離し、上層液にPEG/NaCl[20%PEG(w/v)及び15%NaCl(w/v)]を5:1の割合で添加した後、氷に1時間静置させた。4℃で13,000rpmの速度で20分間遠心分離後、上層液を完全に除去し、1mLのPBS溶液でファージペレットを懸濁した後、2次バイオパニングに使用した。各パニングの段階毎に前記と同じ方法で使用し、洗浄過程の回数を3回、5回、7回、及び10回として、6merのライブラリー(S6)をアセチルコリン受容体たんぱく質に対して5回にかけてバイオパニングを実施した条件とInputファージ、Outputファージの結果を下記表1に整理した。
