データを持ち、従来のADC薬物に存在する安定性問題および異質性問題が解決された。本
発明において、新規抗ヒトErbB2/Her2抗体-マイタンシン誘導体複合体、新規抗ヒトErbB2/Her2抗体-アウリスタチン誘導体複合体の作製およびErbB2/Her2陽性腫瘍標的治療に関する。
体の指向性および伝統細胞毒素を重ねた新世代抗腫瘍薬である。2011年にADC薬物Adcetris(brentuximab vedotin)がホジキンリンパ腫治療薬として、2013年にKadcyla(ado-trastuzumab emtansine)が進行性転移性乳がん治療薬として、米国のFDAにより承認された
。2015年4月まで、全世界において約50種類のADC候補薬のI、IIまたはIIIステージ臨床

、ADC薬物の開発段階では、リンカーとカップリングストラテジーの設計がADC薬物の安定性に極めて重要となり、複合体の生物活性、凝集状態、生物学的利用能、体内分布および代謝に影響を及ぼす。現在市販または臨床開発段階の多くのADC薬物に、リンカーの構造
とカップリングストラテジーが主にSeattle GeneticsとImmunogen社から提供されている
。この二つの会社におけるストラテジーが僅かな異なりがあるにもかかわらず、両社ともチオール(thiol)とマレイミド(maleimide)との反応によって形成されたチオスクシンイミド構造(thiosuccinimide linkage)を利用し、低分子薬物と抗体との連接を行った
(図1)。このカップリング法に反応性が高く、かつ定量可能および温和な反応条件をもつため、広く応用されている(Hermanson GT, Bioconjugate Techniques. 2nd edition, 2008)。しかし、チオスクシンイミド構造の安定性が低く、体内においてほかのチオールと可逆的に置換でき、マレイミド除去反応(maleimide elimination reaction)が起こる。体内にあるシステイン(cysteine)、グルタチオン(glutathione)およびアルブミン
(albumin)の構造に大量なチオールが存在する。これらのチオールがチオスクシンイミ
ド構造にあるスクシンイミドリング(succinimide ring)を認識し、ADC薬物にあるチオ
ールと置換する。この反応の発生は、ADC薬物に抗体と細胞毒素の分解の原因となる(Alley SC etal., Bioconjug. Chem. 2008; Shen BQ etal., Nat. Biotechnol. 30, 184-189,2012; Chudasama VL, etal., Clin. Pharmacol. Ther. 92, 520-527, 2012)。

ーを用い、チオスクシンイミド構造の加水分解を促進することにより、ADC薬物の安定性
を向上させるが、問題点がある。加水分解が抗体と細胞毒素を連接してから行われるため、ADC薬物の作製に複雑な開環操作が導入され、抗体失活のリスクを増大する可能性が考
えられる一方、チオスクシンイミド構造の加水分解は精確に制御できなくなり、品質管理標準の制定が困難となる。

異質性の問題を解決する。本発明に提供した解決案は核酸ターゲット薬物の作製および診断用トレーサーの作製に応用できる。本発明にはリンカーとその作製方法、リンカー―細胞毒素中間物(Linker-Payload intermediate)とその作製方法、開環安定構造リンカー―細胞毒素中間物とその作製方法が含まれる。本発明には遺伝子工学的手段で改良したHerceptinまたはほかの抗ヒトErbB2 /Her2抗体(Ab)、リンカー(Linker)および修飾さ
れたメイタンシンまたはその誘導体、オーリスタチン(Auristatins)またはその誘導体MMAEをLDC技術プラットフォームでカップリングすることによって形成した新世代ADC薬物
およびその作製方法と応用が含まれる。

(1)樹脂の選択
リンカーのC末端アミノ酸残基はアミド基の配列合成にはRink amide-MBHA Resinを用い、C末端アミノ酸残基はカルボキシル基の配列合成にはワング樹脂(Wang resin)を用いる。
(2)樹脂の膨潤
反応用の樹脂の使用量は、合成の目標モル数に基づいて算出されるものである。この樹脂をDCMで浸漬済みのカラムに移し、DCMで二回洗浄した後に、DCM溶液に十分膨潤になるま
で30分間浸漬する。
(3)Fmocの脱保護
20%ピペリジンが含まれるDMF溶液を用い、窒素ガスの保護で10分間反応させる。濾過により除去した後に、再び上記の溶液を加え、5分間反応させる。DCMで二回洗浄し、さらにDMFで三回洗浄した後に、ニンヒドリン法で検出された樹脂の色は、赤褐色あるいは濃い青色になるはずである。
(4)アミノ酸の結合
目標モル数2-4倍量の縮合反応をさせるアミノ酸をDMFに溶解する。適量の縮合剤DICあるいはHBTUを加え、5分間活性化させる。得られる溶液をカラムに移し、窒素ガスの保護で2時間反応させる。ニンヒドリン法で樹脂の色を検出し、無色に近づくまで上記の反応を繰り返す。その後、DCMで二回洗浄し、さらにDMFで三回洗浄をする。
(5)すべてのアミノ酸を連接したままでステップ(3)、(4)の操作を繰り返す。最後に連接されたアミノ酸にBoc保護を行う。
(6)分岐鎖SMCCまたはほかの二官能性分子との結合
Lys分岐鎖保護基の違いによって、脱保護の方法は異なる。例えば、触媒水素化処理法でZを脱離し、ヒドラジン水和物を用いivDdeを脱離する。その後、活性化されたSMCCまたは
ほかの二官能性分子を直接に結合させる。さらなる分岐鎖修飾はない場合には、反応を終了させる。
(7)樹脂の処理と切断
反応が終了した後に、窒素ガスで樹脂複合物を乾燥させ、10ml/g樹脂の量に基づいて、TFA/phenol/H20/EDT/TIS (85/5/5/3/2)の割合で切断液を加え、窒素ガスの保護で攪拌しな
がら0-5℃で2時間反応をさせる。濾過を行い、濾液に体積30倍の氷冷したエチルエー
テルに入れ、冷蔵庫に2時間放置した。遠心分離によって沈澱物を回収し、凍結乾燥で粗ペプチドを得る。
(8)純化と質量分析
粗ペプチドを適切な割合のアセトニトリル水溶液に溶解し、逆相クロマトグラフィーで必要な純度まで精製した後、MSによりその分子量が理論値と一致か否かを確認する。
本発明におけるペイロード(payload)は低分子化合物、核酸、核酸類似物、トレーサー
(caged放射性核種および蛍光分子など)などが含まれ、低分子化合物には、好ましく細
胞毒素がよく使われている。

上記のような、CCA1が含まれるリンカーには、マレイミドリング構造が含まれ、メルカプト基が含まれる分子とスルホスクシンイミジルアミン構造を形成する。メルカプト基を含む分子に低分子化合物、短ペプチド、ポリペプチド、模倣ペプチド、タンパク質、核酸および核酸類似物などが挙げられる。上記カップリング中間物をスクシンイミドリングの開環条件に従って処理し、安定性の高い開環構造を有する中間物を形成する。その構造式は下記に示す。

重要なことに、マレイミドリングの開環反応の効率に関わらず、半調製用/調製用HPLCまたはほかの分離手段で開環中間物の精製ができ、抗体との連接への応用を保証する(実施例に参照)。

重鎖および軽鎖の組み合わせから3種類の抗体が合成でき、それぞれ種類の組み合わせか
ら8種類の抗体が合成できる。アミノ酸残基の配列はアミノ酸配列表に参照する。
従来の化学方法で作製するADC薬物に連接部位の確定は難しく、産物のDAR(Drug Antibody Ratio)の分布範囲が広く、異質性があり、更なる純化は不可能などの欠点が存在する
。本発明に作製するADC薬物は部位特定連接との特徴を持つ。
1)作製方法
ステップ1:前記に記載した方法で式(I)、式(II)、式(III)および式(IV)に示す
リンカー―細胞毒素開環中間物を作製する。マレイミドリングの開環反応の効率に関わらず、半調製用/調製用HPLCまたはほかの分離手段で開環中間物の分離および精製をすることができる。
ステープ2:エンジニアリング技術で改変された、リガーゼ認識配列が含まれる組み換え
抗体をCHO細胞またはほかの哺乳類細胞培養システムを用いて発現させる。
ステープ3:組み換え抗体(または抗体類似物)とリンカー―細胞毒素開環中間物を連接
する。軽鎖および/または重鎖のC末端にリガーゼ特異認識配列を含む組み換え抗体とリ
ンカー―細胞毒素開環中間物をリガーゼ(またはリガーゼグループ)の触媒反応を介して連接する。
抗体とリンカー―細胞毒素開環中間物をリガーゼの触媒反応を介して連接し、数種類のADC薬物を得ることができる(図27-32に示し、実施例を参照)。
2)ADC薬物の構造分析
本発明におけるADC薬物が多種の方法で分析される。反応効率がSDS-PAGEにより分析され
、分子の構造がエレクトロスプレーイオン化質量分析(ESI-MS)により分析され、DAR分
布が疎水高速液体クロマトグラフィー(HIC-HPLC)により測定され、抗体凝集程度が分子サイズスクリーニング高速液体クロマトグラフィー(SEC-PAGE)により分析される。
1)ADC薬物の腫瘍細胞表面特定抗原との結合及び親和力の測定
本発明にADC薬物が腫瘍細胞表面特定抗原に結合する親和力の測定方法を提供し、腫瘍細
胞表面特定抗原または受容体に効率的に結合するADC薬物をスクリーニングする。例えば
、ADC候補薬物であるGQ1001を乳がん細胞と混合培養し(10-60min)、FACSでADC候補薬
物であるGQ1001と乳がん細胞表面受容体ErbB2/Her2との結合親和性を測定する。その結果、本発明に用いられるリンカーおよび連接法で作製したADC薬物であるGQ1001が細胞表面
受容体ErbB2/Her2に特異的に認識し、その親和力がHerceptinと同程度であることが分か
り、本発明に用いられるリンカーおよび連接法は抗体の活性に対する影響が少ないことが提示されている。
2)ADC薬物の腫瘍細胞増殖抑制作用
候補ADC薬物の腫瘍細胞(ErbB2/Her2高発現腫瘍細胞)に対する増殖抑制作用を確認する
ため、本発明は下記の方法で候補ADC薬物の細胞毒性または細胞増殖抑制活性を測定する
ことになる。ターゲット抗原または受容体発現哺乳類細胞(例えばErbB2/Her2高発現または低発現腫瘍細胞)に候補ADC薬物を加え、12-120hr培養してから、CellTiter Glo法で
細胞生存率を測定する(viability)。
実施例の一つとして、ヒト乳がん細胞(BT474、HCC1954、SK-BR-3、MCF-7、MDA-MB-468)、ヒト卵巣がん細胞(SK-OV-3)、ヒト胃がん細胞(NCI-N87)にADC薬物GQ1001を加え、12-120hr培養してから、CellTiter Glo法で細胞内のATP量を測定した。ATP量の変化によ
って、細胞生存率に換算した。その結果、GQ1001はErbB2/Her2高発現腫瘍細胞の増殖を特異的に抑制することがわかった。
3)ラット体内での代謝および安定性
ラットに候補ADC薬物を5-50 mg/kg尾静脈投与を行い、各時間点で採血し、ELISA法で血清中の候補ADC薬物濃度を測定する。実施例の一つとして、SDラットにADC薬物GQ1001またはKadcylaの尾静脈投与を行い、投与1時間から28日までの各時間点で採血し、ELISA法で血
清中のGQ1001またはKadcyla濃度を測定した。その結果、各時間点で、GQ1001またはKadcylaのラット体内における濃度に変化は認められなかった。
4)ADC薬物の体内薬効
本発明において、腫瘍細胞を移植するヌードマウスモデルを作製し、候補ADC薬物の抗腫
瘍作用を検討する。実施例の一つとして、ヒト乳がん細胞を移植するヌードマウスモデルを作製し、0.5-50 mg/kg のADC薬物GQ1001の尾静脈を通しての単回投与を行い、腫瘍のサイズを測定した。その結果、0.5-50 mg/kg のADC薬物GQ1001の単回投与では、ErbB2/Her2高発現腫瘍細胞の増殖が顕著に抑制された。
5)げっ歯類動物に対する毒性
本発明はラットを用い、候補ADC薬物の急性毒性を評価した。雌性Sprague-Dawleyラット
に高用量(60mg/kgまたはその以上)の候補ADC薬物を投与し、ラットの体重、症状、血液学、臨床生化学と組織病理学などの指標を通して、候補ADC薬物の毒性を評価した。その
結果、ADC薬物GQ1001のラットに対する毒性は同濃度(60mg/kg)のKadcylaより低いこと
を発見した。
本発明は特殊なリンカーを提供する。これらのリンカーはタンパク質、特に抗体と低分子化合物、ペプチド、核酸、トレーサーなどとの連接に応用でき、研究、臨床診断および治療に用いられる複合体の作製に関わっている。

2)本発明が提供するリンカーにスクシンイミド開環構造を有し、閉環構造より、哺乳類
動物体内での安定性が高い。また、連接された細胞毒素は体内に存在するシステイン、グルタチオンまたはアルブミンとメルカプト基置換反応を起こしにくい。このリンカーを用いて作製するADC薬物は体内での安定性が高く、ADC薬物の低分子が脱離しやすい問題点が解決された。
3)本発明が提供するADC薬物は精確カップリング複合体であり、均質性が高い。伝統的な化学カップリング複合体と比べ、本発明が提供するADC薬物は品質管理と薬物安全性の面
で改善され、ADC薬物の異質性問題点が解決された。
1)抗体T-LCCTL-HCの作製
SEQ ID No.1 抗体T-LCCTL-HC配列が組み込まれたプラスミドをCHO細胞に導入し、T-LCCTL-HC発現量の高い細胞をスクリーニングした。Trastuzumabの培養方法に基づき、5-10Lスケールで培養を行い、上清を回収した。
2)抗体T-LCCTL-HCの精製
MabSelect親和性クロマトグラフィー法およびSepharoseS陽イオン交換クロマトグラフィ
ー法を用い、T-LCCTL-HCの精製を行った。Trastuzumabの緩衝液(5mM histidine-HCl、2%
Trehalose、0.009% Polysorbate 20、pH6.0)に溶解し、分注保存した。
3)T-LCCTL-HC抗体の品質管理
精製されたT-LCCTL-HC抗体の純度をSDS-PAGEによって確認した結果、98.5%になったことが観察された。SEC-HPLCによって、T-LCCTL-HC抗体が形成された高分子量ポリマーの量は0.4%未満であった。また、エンドトキシンの量は0.098EU/mg未満であった。
1)リンカー2-DM1中間物(n=3、閉環)の作製および質量管理
等モル数のリンカー2(n=3)とDM1 を計り、溶解混合後、0-40℃で0.5-20hr反応させ、リンカー2-DM1中間物(n=3、閉環、構造は図12に示す)を得た。超高速液体クロマ
トグラフ-タンデム型質量分析法(UPLC-MS)を用い、リンカー2-DM1中間物(n=3、閉環)の純度と分子量を測定した。その結果は予想通りに、純度は100%に示し(同量の異
性体が含まれ、図33に示す)、分子量は1274である。
2)リンカー2-DM1中間物(n=3、閉環)の開環反応および精製
リンカー2-DM1中間物(n=3、閉環)溶液を適量のTris base溶液またはほかの開環反応を促進する溶液と混合し、0-40℃で0.5-20hr反応させ、リンカー2-DM1中間物(n=3
、開環、構造は図16に示す)を得た。開環反応が20分、40分、60分、2時間または4時間進んだ時点において、UPLCで測定した結果が図35A-Eに示す。反応の進行に伴い、リンカー2-DM1中間物(n=3、開環)の割合は増加し(10%から73%まで増加)、マレイミドリングの開環反応の効率に関わらず、半調製用/調製用HPLCでリンカー2-DM1中間物(n=3
、開環)の精製ができ、抗体との連接への応用を保証する。
3)リンカー2-DM1中間物(n=3、開環)の質量管理
適量のリンカー2-DM1中間物(n=3、開環)を計り、溶解し、超高速液体クロマトグラ
フ-タンデム型質量分析法(UPLC-MS)を用い、その純度と分子量を測定した。結果は図36と図37に示す。予想通り、HPLCで精製されたリンカー2-DM1中間物(n=3、開環)の純度は100%に達し、分子量は1291.8であり、ADC薬物GQ-1001の作製に理想的なリンカーを
提供した。
本発明はペプチド転移酵素を用い、リンカー2-DM1中間物(n=3、開環)を抗体T-LCCTL-HCと部位特定的に連接し、図28に示すようなADC薬物を得た。その中で、nは3であり、
dは2であり、リガーゼの認識配列LPXTにおけるXはグルタミン酸(E)である。
1)抗体T-LCCTL-HCの処理
限外ろ過、透析または脱塩カラム法によって抗体T-LCCTL-HCのバッファーを1×リガーゼ緩衝液に置換した。1×リガーゼ緩衝液に50mM Tris-HCl(pHは5-8)、150mM NaCl、CaCl2(ありまたはなし)が含まれる。
2)ADC薬物GQ1001の固相精製
本発明はSortase酵素を基づいた改変型ペプチド転移酵素を用い、抗体T-LCCTL-HCをリン
カー2-DM1中間物(n=3、開環)と連接し、ADC薬物GQ1001を作製した。
1×リガーゼ緩衝液中で抗体T-LCCTL-HCとリンカー2-DM1中間物(n=3、開環)を適量
のモル比(1:1~1:100)で混合し、固相カップリングカラムに加えた。固相カップリングカラムに充填した基質は転移酵素を有し、抗体T-LCCTL-HCをリンカー2-DM1中間物(n=3、開環)と連接させる。4-40℃で0.5-20hr連接反応を行い、固相カップリングカラ
ムから産物を回収し、限外ろ過または透析法で反応できていない薬物中間物を分離した。精製されたADC薬物GQ1001を4℃または―80℃において、Kadcyla緩衝液(10 mM Sodium Succinate、pH5.0;100 mg/ml Trahelose; 0.1% (w/v) Polysorbate 20; Kadcyla製剤の調製を参照)に保存した。
連接反応が完成された後、SDS-PAGE法によってGQ1001の純度および連接効率を測定した。その結果は図38に示すように、連接反応には部位特定的に抗体T-LCCTL-HCの軽鎖に連接された。なお、細胞毒素が連接されていない抗体T-LCCTL-HCの軽鎖と比べ、細胞毒素DM1が
連接されたGQ1001の軽鎖に顕著な分子量遷移が見られた。連接産物に細胞毒素が連接されていない軽鎖が検出されておらず、連接効率は95%以上になり、連接産物の純度も予想に達した。
高精度質量分析(ESI-MS)を行い、ADC薬物GQ1001の軽鎖を解析した結果、分子量が25281に示し、理論分子量の25284とほぼ一致の結果が得られ、各軽鎖に一つの細胞毒素分子が
連接されていることを確認した。高精度質量分析(ESI-MS)の結果を図39Aと39Bに示す。
Butyl-HICカラムを利用し、ADC薬物GQ1001のDAR分布を測定した。その結果は図40に示す
ように、細胞毒素が連接されていない抗体T-LCCTL-HCの割合は5%未満であり、連接産物DAR2がADC薬物GQ1001の主体として、1.8に達したことがわかった。
SECカラムを利用し、ADC薬物GQ1001の高分子量凝集程度を測定した。その結果は図41に示すように、ADC薬物GQ1001に高分子量凝集が検出されていないことから、カップリング反
応は抗体に影響を及ぼさないことがわかった。
1)ヒト乳がん細胞BT-474またはSK-BR-3単細胞懸濁液を(0.5-5)×106/mlになるように
調整した。5×105 cells/testに6.25nMのHerceptin、T-LCCTL-HCまたはGQ1001を加え、4
℃で60分培養した。さらに1ml洗浄液(PBS+1% BSA)を加え、1000rpmで5分間遠心分離し
、上清を除去した。この洗浄操作を二回繰り返した。
2)Herceptin、T-LCCTL-HCまたはGQ1001を加えた細胞に100 μLのFITC-Goat anti-human IgG抗体希釈液を加え、4℃で30分遮光培養した。1ml洗浄液を加え、1000rpmで5分間遠心
分離し、上清を除去した。この洗浄操作を二回繰り返した後、500μLのPBSで懸濁した。300目ふるいを通して、氷上において放置し、BD C6で測定を行った。その結果は図42-43
に示すように、Herceptin、T-LCCTL-HCまたはGQ1001は乳がん細胞BT-474またはSK-BR-3表面に発現する受容体ErbB2/Her2に対する親和力に顕著な差は認められなかった。
1)ErbB2/Her2低発現するヒト乳がん細胞をMCF-7、MDA-MB-468に、高発現するヒト乳がん細胞をBT-474、SK-BR-3、HCC1954に、ヒト卵巣がん細胞をSK-OV-3に、ヒト胃がん細胞をNCI-N87に使用された。各ウェルに100μLの細胞(1000-10000細胞数)を96ウェル細胞培
養プレートに播種し、37℃、5%CO2、95%空気、100%湿度の条件でO/N培養を行った。
2)O/Nで培養した細胞に各濃度(30、10、3.333、1.111、0.370、0.123、0.041、0.014、0.005 nM)のGQ1001、Kadcyla、Herceptin、DM1を加え、コントロールとして、50 μM の
Puromycinを加え、されに37℃で48-96hr培養を行った。
3)細胞培養プレートを室温になるまで放置した。各ウェルに100μM 的CellTiter Glo試
薬を加え、2分間振動培養を行い、10分間遮光反応を行った。最後にBioTech Gen5マイク
ロプレートリーダーで発光値(RLU)を測定した。
4)各薬物の腫瘍細胞に対する増殖抑制作用の結果は表1および図44-50に示す。DM1はErbB2/Her2高発現および低発現細胞に顕著な増殖抑制作用を示した一方、Herceptin、GQ1001はErbB2/Her2高発現細胞のみに顕著な増殖抑制作用を示した。また、Kadcylaは高濃度において、ErbB2/Her2低発現細胞の増殖に抑制作用を示した。
1)体重160-180gのSPF雌性SDラットを用い、各群に4匹でランダムにGQ1001群、Kadcyla
群、ブランク群に分けられた。
2)10 mg/kg のGQ1001(ロット:20141128、純度>98%)またはKadcyla(ロット N1003)
を尾静脈により投与を行った。ブランク群では、同体積のPBS(pH=7.4)を尾静脈により投与した。
3)投与1hr、1日、2日、3日、4日、6日、8日、13日、17日、21日、28日後、眼底採血により100-200μL採血を行った(抗凝固剤使用せず)。血液を氷上において1-2hr静置し、4000rpm、20min、4℃で遠心分離した。上清の血清画分を新しいEPチューブに分注し、-80℃で保存した。
4)ELISAで血清中のおけるGQ1001、Kadcyla量を測定した。
5)その結果、投与1日後、血中GQ1001およびKadcyla濃度は大幅に減少し、ADC薬物が体内で速やかに吸収されたことが考えられる。投与1hr後と比べ、それぞれ44.7%と42.5%ま
で減少し、両群には顕著な差は見られなかった。投与6日後、血中GQ1001およびKadcyla濃度は投与1hr後と比べ、それぞれ20.9%と20.5%まで減少した。投与13日後、血中GQ1001
およびKadcyla濃度は投与1hr後と比べ、それぞれ9.9%と12.2%まで減少した。投与21日
後、血中GQ1001およびKadcyla濃度は投与1hr後と比べ、それぞれ5.5%と7.7%まで減少し
た。投与28日後、血中GQ1001およびKadcyla濃度は投与1hr後と比べ、それぞれ3.5%と5.2%まで減少した。以上の結果から、GQ1001およびKadcylaは雌性ラットでの薬物動態に明
らかな差は認められなかった(図51)。
1)対数成長期における乳がん細胞HCC1954を使用し、マトリゲルバッファー(PBS:BD Matrigel=1:1)で細胞密度を2.5×107/mlまで調製した。6-8週齢のSPF雌性BALB/cヌードマ
ウスを使用し、右の肩甲骨に調製したHCC1954細胞懸濁液を0.2ml皮下投与した。
2)接種7日後、ノギスで腫瘍の直径を測定し、公式V=0.5a × b2に従って腫瘍の体積を
計算した(aは腫瘍における最長直径、bは腫瘍における最短直径)。腫瘍サイズが130-140mm3までになるマウスを使用し、各群に10匹でランダムにブランク群、GQ1001 0.5mg/kg、5mg/kg群、Kadcyla 5 mg/kg群、Herceptin 5 mg/kg群に分けられた。投与方法に尾静脈投与を採用し、ブランク群では等量の溶媒を投与することになった。投与から31日までに週二回腫瘍サイズの測定を行い、31日後週一回腫瘍サイズの測定を行った。各時間点における腫瘍サイズを各群において比較した。また、T-CまたはT/C値を抗腫瘍活性の指標として、薬物の薬効を評価した。T-C値の計算では、Tは治療群において、腫瘍平均サイズが予定サイズ(500 mm3)に達するまでの平均日数であり、Cはブランク群において、腫瘍平均サイズが予定サイズ(500 mm3)に達するまでの平均日数である。T/C値(%)は腫瘍抑制効果を評価する指標として、その中で、Tはある時間点で治療群における腫瘍平均体積
であり、Cはある時間点でブランク群における腫瘍平均体積である。
3)GQ1001 5 mg/kgおよびKadcyla 5 mg/kg投与10日後、ブランク群と比べ、腫瘍体積が顕著に減少した。その中に、腫瘍が完全に消えた例も発見し、38日まで腫瘍の再生は見られなかった(表2、図52)。以上の結果から、5 mg/kgの GQ1001またはKadcylaがErbB2/Her2陽性乳がん細胞に対して、顕著な抑制作用を有することが示唆された。
1)成年健康雌性SDラットを使用し、各群に6匹でランダムにブランク群(0 mg/kg)、GQ1001 6mg/kg、60mg/kg群、Kadcyla 60 mg/kg群に分けられた。単回尾静脈投与により、10ml/kgを1 ml/minの速度で投与した。投与期間では、ラットに臨床観察を行い、体重、摂食量を記録し、血液細胞計数、血液生化指標を測定した。試験が終わった後、ラットに安楽死を行い、解剖により可視病変を観察し、臓器を摘出し、重量を計り、臓器係数を計算した。
2)その結果、投与期間に、各濃度のGQ1001群では明らかな臨床症状は観察されていない
。Kadcyla 60 mg/kg群では、投与5日後(D5)鼻分泌物(2/6)、耳フラッシング(耳部
)と腫脹(6/6)、毛乱れ(6/6)、体重減少(1/6)、背曲げ(1/6)、耳と四肢青い(1/6)などの症状が観察された。耳腫脹の症状はD6に消失し、鼻分泌物の症状はD7に消失し
た。4/6のラットはD8に正常に回復し、残りの2/6のラットはD15の安楽死まで毛乱れ以外
の症状が回復した。ブランク群と比べ、各GQ1001投与群では、明確な症状は観察されていないのに対して、Kadcyla 60 mg/kg投与群では、投与後(D2-D12)体重減少が観察できた(図53)。
血液学と臨床生化指標の結果から、各GQ1001投与群では、明らかな毒性は観察されていないのに対して、Kadcyla 60 mg/kg投与群では、赤血球指標(RBC、HGB、HCT、Retic)の減少、白血球数の増加、ALT、AST、TBIL、GGT指標の上昇、特にALTおよびASTの上昇が観察
され、Kadcyla 60 mg/kg単回投与下、肝毒性に関わる可能性が示唆された(図54)。
解剖所見から、各GQ1001投与群では、明確な変化は観察されていないのに対して、Kadcyla 60 mg/kg投与群では、脾臓膨大(6/6)、肝臓周辺鈍い(4/6)、胸腺縮小(1/6)など
の症状が観察された。
以上の結果から、同濃度(60 mg/kg)条件下において、GQ1001の毒性はKdcylaより低いことが示唆された。
1)リンカー5-Mc-Val-Cit-Pab-MMAE薬物中間物(n=3、閉環)の作製および質量管理
等モル数のリンカー5(n=3)とMc-Val-Cit-Pab-MMAEを計り、溶解混合後、0-40℃で0.5-20hr反応させ、リンカー5-Mc-Val-Cit-Pab-MMAE中間物(n=3、閉環)を得た。構造は図19に示す。
2)リンカー5-Mc-Val-Cit-Pab-MMAE薬物中間物(n=3、閉環)の開環反応および質量管理
リンカー5-Mc-Val-Cit-Pab-MMAE薬物中間物(n=3、閉環)溶液は適量のTris base溶液ま
たはほかの開環反応を促進する溶液と混合し、0-40℃で0.5-20hr反応させ、開環構造になった薬物中間物を得た。構造は図23に示す。HPLCで薬物中間物(n=3、開環)の純度お
よび分子量を測定した。結果は図55に示すように、異性体がHPLCでは分離できなかった。開環反応液をMALDI-TOF質量分析器で解析した結果は図56に示すように、その分子量は1702または1718であり、Na、Kイオン化による原因と考え、理論分子量1681と一致し、開環産物ができたことが示唆された。マレイミドリングの開環反応の効率に関わらず、半調製用/調製用HPLCで開環薬物中間物の精製ができ、抗体との連接への応用を保証する。
Claims (15)
- 式(I)、式(II)、式(III)または式(IV)で示される構造の化合物:
PCAは、Sortase酵素であるリガーゼが特異的に認識する基質にあるアミノ酸配列を有する部位であり、グリシンまたはアラニンの一つまたは多数からなる群から選択されるユニットを1-100まで直列接続して含み;
LA1、LA2は連接ドメインであり、
前記LA1はPCAとCCA'との二官能性架橋部位であり、前記LA1は、C7~C34アルキレンを含み、前記アルキレンの1つもしくはそれ以上の(-CH2-)構造は、場合により、-O-で置換されてもよく;および/または前記LA1は、1~10個のアミノ酸を含むアミノ酸断片を
含み、各アミノ酸は独立して天然型もしくは非天然型アミノ酸であり、前記LA1は、PCAとCCA'とを、アミド結合、ジスルフィド結合、チオエーテル結合、チオエステル結合、ペプチド結合、ヒドラゾン結合、エステル結合、エーテル結合、もしくはウレタン結合により結合する;ならびに

- 下記に示すステップを含むことを特徴とする、請求項3~5のいずれかに記載の化合物を作製する方法:
i) 請求項1または2に記載の式(I)、式(II)、式(III)または式(IV)に示す化合物を作製するステップ;
ii) 請求項3~5のいずれかに記載のA-(LA3)b-Fを作製するステップ;
iii)リガーゼを加え、ii)で得られたA-(LA3)b-Fをステップi)の式(I)、式(II)、式(III)または式(IV)に示す化合物と、前記リガーゼの作用に適切な条件下で
連接するステップ。 - 動物における細胞増殖を抑制するための、請求項3~5のいずれかに記載の化合物。
- 動物が哺乳類動物である、請求項7に記載の化合物。
- 動物がヒトである、請求項8に記載の化合物。
- ヒトの疾病予防と治療に用いる薬物を製造するための、請求項3~5のいずれかに記載の化合物の使用。
- 疾病が腫瘍または自己免疫疾病を含む、請求項10に記載の使用。
- 腫瘍細胞表面が、化合物が認識、結合する特異的抗原または受容体タンパク質を有する、請求項11に記載の使用。
- 腫瘍細胞表面が有する特異的抗原または受容体タンパク質はErbB2/Her2である、請求項12に記載の使用。
- 腫瘍が乳がん、胃がん、卵巣がん、肺がん、結腸がん、直腸がん、大腸がん、または食道がんである、請求項13に記載の使用。
