Description
脱分極に伴う活動電位の持続時間を延長する電位依存性Naイオンチャネルと、
静止膜電位を負方向に深くするKイオンチャネルと、
静止膜電位を負方向に深くすること及び/又は脱分極に伴う活動電位の持続時間を短縮することに寄与するイオンチャネルである標的イオンチャネルと、
を備える細胞を含み、
前記Naイオンチャネルと前記Kイオンチャネルと前記標的イオンチャネルとが、前記Kイオンチャネルに対する阻害剤の存在下で脱分極の惹起により細胞死が誘導される細胞死誘導系を構築している、スクリーニング材料。
(2) 前記標的イオンチャネルは、前記細胞死誘導系によって誘導される細胞死に抑制的に作用する、(1)に記載のスクリーニング材料。
(3) 前記Kイオンチャネルに対する選択的阻害剤を用いる、(1)又は(2)に記載のスクリーニング材料。
(4) 前記標的イオンチャネルは、前記Kイオンチャネルの阻害剤に対する感受性が、前記Kイオンチャネルよりも低い、(3)に記載のスクリーニング材料。
(5) 前記標的イオンチャネルは、静止膜電位を負方向に深くすることに寄与するイオンチャネルであって、前記Kイオンチャネルとは異なる他のKイオンチャネルである、(1)~(4)のいずれかに記載のスクリーニング材料。
(6) 前記他のKイオンチャネルは、K2Pチャネルである、(5)に記載のスクリーニング材料。
(7) 前記標的イオンチャネルは、脱分極に伴う活動電位の持続時間を短縮することに寄与するイオンチャネルである、(1)~(6)のいずれかに記載のスクリーニング材料。
(8) 標的イオンチャネルに対する作用剤又は阻害剤をスクリーニングする方法であって、
脱分極に伴う活動電位の持続時間を延長する電位依存性Naイオンチャネルと、
静止膜電位を負方向に深くするKイオンチャネルと、静止膜電位を負方向に深くすること及び/又は脱分極に伴う活動電位の持続時間を短縮することに寄与するイオンチャネルである標的イオンチャネルと、を備える細胞であって、前記Naイオンチャネルと前記Kイオンチャネルと前記標的イオンチャネルとが、前記Kイオンチャネルに対する阻害剤の存在下で脱分極の惹起により細胞死が誘導される細胞死誘導系を構築している細胞を用い、
前記細胞に対して、前記阻害剤を供給する工程と、
前記細胞に対して、前記標的イオンチャネルを阻害する又は活性化する可能性のある被験化合物を供給する工程と、
前記被験化合物の供給による前記細胞の細胞死への影響を評価する工程と、
を備える、方法。
(9) 標的イオンチャネルに対する作用剤又は阻害剤をスクリーニングするためのキットであって、
脱分極に伴う活動電位の持続時間を延長する電位依存性Naイオンチャネルと、静止膜電位を負方向に深くするKイオンチャネルと、静止膜電位を負方向に深くすること及び/又は脱分極に伴う活動電位の持続時間を短縮することに寄与するイオンチャネルである標的イオンチャネルと、を備える細胞であって、前記Naイオンチャネルと前記Kイオンチャネルと前記標的イオンチャネルとが、前記Kイオンチャネルに対する阻害剤の存在下で脱分極の惹起により細胞死が誘導される細胞死誘導系を構築している細胞と、
前記阻害剤と、
を備える、キット。
本スクリーニング材料は、脱分極に伴う活動電位の持続時間を延長する電位依存性Naイオンチャネルを備えている。また、本スクリーニング材料は、静止膜電位を負方向に深くするKイオンチャネルを備えている。さらに、本スクリーニングは、静止膜電位を負方向に深くすること及び/又は脱分極に伴う活動電位の持続時間を短縮することに寄与するイオンチャネルである標的イオンチャネルを備えている。
本スクリーニング材料が備えるNaイオンチャネルは、脱分極に伴う活動電位の持続時間を延長する電位依存性Naイオンチャネルである。かかるNaイオンチャネルは、換言すれば、不活性化を抑制されたNaイオンチャネルということもできる。
本スクリーニング材料が備えるKイオンチャネルは、静止膜電位が負の方向に深くなるように、換言すれば、より負の電位が大きくなるような機能(活性)を有するイオンチャネルである。すなわち、かかるKイオンチャネルによれば、その発現や活性によるKイオン透過性の亢進により深い静止膜電位を形成することができる。上記したNaイオンチャネルを備えると、細胞内外におけるNaイオンの濃度差により、Naイオンが細胞内に過剰に流入する状態となり、最終的には細胞内のNaイオン濃度が高まり細胞は死んでしまう。細胞をスクリーニング材料として用いるには、かかる細胞死誘導系が誘導されるまでは細胞が生存している必要がある。そこで、静止膜電位を深く(低く)するために、静止膜電位が負の方向に深くするKイオンチャネルを用いる。
本スクリーニング材料が備えることができる抑制的標的イオンチャネルは、静止膜電位を負方向に深くすること及び/又は脱分極に伴う活動電位の持続時間を短縮することに寄与するイオンチャネルである。抑制的標的イオンチャネルは、本スクリーニング材料が備える、Kイオンチャネルに対して阻害的に作用して、細胞死誘導系において脱分極を惹起して細胞死を誘導できる阻害剤(以下、Kイオンチャネル阻害剤ともいう。)による脱分極の惹起や細胞死誘導に抑制的に作用することができる。抑制的標的イオンチャネルは、当該阻害剤とは独立して当該阻害剤による作用、すなわち、脱分極の惹起やそれによる細胞死誘導を抑制することができる。こうしたイオンチャネルを選択することで、当該イオンチャネルに関する効率的な評価系を構築できる。
Kイオンチャネル阻害剤は、Kイオンチャネルの作用を阻害して、静止膜電位を負方向に深くすることを抑制する。こうした阻害剤は、例えば、Kイオンチャネルの種類によっても異なる。公知情報から、Kイオンチャネル阻害剤を取得できるほか、種々のKイオンチャネルを含む細胞死誘導系を備える細胞を準備して、この細胞に対して、可能性あるKイオンチャネル阻害剤を供給して脱分極の惹起や細胞死が誘導されるか否かを評価することで、取得することができる。
本明細書に開示されるスクリーニング方法(以下、本スクリーニング方法ともいう。)は、標的イオンチャネルに対する作用剤及び阻害剤をスクリーニングする方法である。本スクリーニング方法は、本スクリーニング材料に対して、Kイオンチャネルの作用を阻害するようにKイオンチャネルの阻害剤を供給する工程と、本スクリーニング材料に対して、抑制的標的イオンチャネルを阻害する又は活性化する可能性のある被験化合物を供給する工程と、被験化合物の供給による本スクリーニング材料の細胞死への影響を評価する工程と、を備えることができる。本スクリーニング方法によれば、Kイオンチャネルの阻害剤を供給してKイオンチャネルを阻害することで、抑制的標的イオンチャネルに対する被験化合物の作用を簡易に評価することができる。例えば、電気刺激を用いなくてもスクリーニング材料の脱分極を惹起することができる。この結果、効率的なスクリーニングが可能となっている。
本スクリーニング方法は、標的イオンチャネルに対する被験化合物の作用の評価方法としても実施できる。本評価方法によれば、簡易かつ迅速に、被験化合物の標的イオンチャネルへの作用(作用性や阻害性)を測定できる。したがって、被験化合物に一定以上の作用が求められる場合における迅速かつ簡易な評価方法として有用である。本評価方法には、すでに説明した本スクリーニング方法における各種態様をそのまま適用することができる。
本明細書に開示されるキットは、標的イオンチャネルに対する作用剤又は阻害剤をスクリーニングするためのキット(以下、本キットともいう。)である。本キットは、本スクリーニング材料と、Kイオンチャネルの作用を抑制する阻害剤と、を備えることができる。本キットによれば、標的イオンチャネルに作用する化合物のスクリーニングを容易にかつ効率的に行うことができる。本キットは、本スクリーニング材料と阻害剤のほか、細胞死の測定用の試薬を備えていてもよい。また、本スクリーニング材料に適した培地を備えていてもよい。また、標的イオンチャネルの発現や活性を制御する化合物や、必要に応じて行う電気刺激用の装置などの機材を備えていてもよい。なお、本キットは、被験化合物の標的イオンチャネルに対する作用を評価するためのキットとしても用いることができる。
本明細書は、Kイオンチャネルに対する阻害剤のスクリーニング方法(以下、阻害剤スクリーニング方法ともいう。)も開示する。阻害剤スクリーニング方法によれば、本スクリーニング材料におけるKイオンチャネルに対する好適な阻害剤、すなわち、用いるKイオンチャネルに対して特異性がより高く、感受性のより高い阻害剤をスクリーニングすることができる。
本明細書は、Naイオンチャネルに対する阻害剤及び活性化剤のスクリーニング方法も開示する。脱分極に伴う活動電位の持続時間を延長する電位依存性Naイオンチャネルの当該作用を阻害する阻害剤や当該作用を活性化する活性化剤は、いずれも、本明細書において開示される標的イオンチャネルに対する作用剤等のスクリーニング系に有用であるからである。
Nav1.5チャネルの不活性化を制御しているIII-IVリンカー領域中に存在する疎水性アミノ酸配列Ile-Phe-Met(IFMモチーフ)をすべてGlnに変異させることとした。変異後のアミノ酸配列(部分)を以下に示す。

ヒト胎児腎由来細胞(HEK293細胞)はヒューマンサイエンス研究資源バンク(HSRRB)から購入した。10% FBS(Fatal Bovine Serum; Gibco)を添加し、100 U/ml ペニシリン(和光純薬)、100μg/ml ストレプトマイシン(明治製菓)を加えたD-MEM(Dulbeco’s Modified Eagle Medium)培地(和光純薬)中で37℃、5% CO2条件下で培養した。ヒト由来のKir2.1(NM#000891)をpcDNA3.1(+)(Invitrogen)にサブクローニングしたpcDNA3.1/Kir2.1をLipofectamine 2000 (Invitrogen)を用いて遺伝子導入し、上記のD-MEM培地に0.2 mg/ml Zeocin(Invitrogen)を加えた培地で培養し、Zeocin耐性細胞をクローニングし、Kir2.1定常発現細胞(HEK#Kir)を作製した。また同様の方法でKir2.1定常発現細胞に不活性化変異体Nav1.5 IFM/QQQを遺伝子導入した(HEK#Kir#mutated Nav)。

Claims (9)
- 標的イオンチャネルに対する作用剤又は阻害剤をスクリーニングする方法であって、
電位依存性Naイオンチャネルの変異体であって脱分極に伴う活動電位の持続時間が1分以上であるNav1.5チャネルの変異体と、
静止膜電位-50mV以下であるKイオンチャネルと、静止膜電位を負方向に深くすること及び/又は脱分極に伴う活動電位の持続時間を短縮することに寄与するイオンチャネルである標的イオンチャネルと、を備える細胞を用い、
前記細胞に対して、前記Kイオンチャネルに対する選択的阻害剤を供給する工程と、
前記細胞に対して、前記標的イオンチャネルを阻害する又は活性化する可能性のある被験化合物を供給する工程と、
前記被験化合物の供給による前記細胞の細胞死への影響を評価する工程と、
を備え、
前記標的イオンチャネルは、前記選択的阻害剤によって惹起される前記細胞における脱分極又は該脱分極による前記細胞の細胞死を抑制するイオンチャネルである、方法。 - 前記標的イオンチャネルは、前記Kイオンチャネルの前記選択的阻害剤に対する感受性が、前記細胞の前記選択的阻害剤による前記細胞死を抑制できる程度に低いイオンチャネルである、請求項1に記載の方法。
- 前記Kイオンチャネルは、Kir2.1、Kir2.2、Kir3.X、Kir4.X、Kir4.X、Kir5.X及びKir6.X及びKir7.Xからなる内向き整流性Kイオンチャネル群、K2Pイオンチャネル群及びCaイオン活性化Kイオンチャネル群から選択される1種以上である、請求項1又は2に記載の方法。
- 前記Kイオンチャネルは、Kir2.1イオンチャネルである、請求項1~3のいずれかに記載の方法。
- 前記標的イオンチャネルは、電位依存性イオンチャネル、リガンド依存性イオンチャネル、機械刺激依存性イオンチャネル、温度依存性イオンチャネル、漏洩チャネルイオンチャネル及びリン酸化依存性イオンチャネルからなる群から選択される、請求項1~4のいずれかに記載の方法。
- イオンチャネルに対する作用剤又は阻害剤をスクリーニングするためのキットであって、
電位依存性Naイオンチャネルの変異体であって脱分極に伴う活動電位の持続時間が1分以上であるNav1.5チャネルの変異体と、静止膜電位が-50mV以下であるKイオンチャネルと、静止膜電位を負方向に深くすること及び/又は脱分極に伴う活動電位の持続時間を短縮することに寄与するイオンチャネルである標的イオンチャネルと、を備える細胞と、
前記Kイオンチャネルの選択的阻害剤と、
を備え、
前記標的イオンチャネルは、前記選択的阻害剤によって惹起される前記細胞における脱分極又は該脱分極による前記細胞の細胞死を抑制できるイオンチャネルである、キット。 - 前記標的イオンチャネルは、前記Kイオンチャネルの前記選択的阻害剤に対する感受性が、前記細胞の前記選択的阻害剤による前記細胞死を抑制できる程度に低いイオンチャネルである、請求項6に記載のキット。
- 前記標的イオンチャネルは、電位依存性イオンチャネル、リガンド依存性イオンチャネル、機械刺激依存性イオンチャネル、温度依存性イオンチャネル、漏洩チャネルイオンチャネル及びリン酸化依存性イオンチャネルからなる群から選択される、請求項6又は7に記載のキット。
- 前記Kイオンチャネルは、Kir2.1イオンチャネルであり、
前記選択的阻害剤は、Baイオン含有化合物である、請求項6~8のいずれかにに記載のキット。
