本明細書で使用されるとき、「ナノ粒子」はナノスケールを有する粒子を意味し、いずれかの特定な形状に限定することは意図されていない。特に、「ナノ粒子」はナノスフェア、ナノチューブ、ナノボックス、ナノクラスター、ナノロッドなどを包含する。特定の実施形態では、本明細書で検討されるナノ粒子および/またはナノ粒子コアは、一般的な多面体または球体形状を有する。ナノ粒子またはナノ粒子コアの「直径」と言った場合、それぞれナノ粒子またはナノ粒子コアの最も長い寸法を意味するように一般的に考えられる。ほぼ多面系または球体形状を有するナノ粒子では、粒子を横切る最短の寸法は、粒子を横切る最も長い寸法の一般的には50%以内であり、例えば25%または10%以内であり得る。

本発明のナノ粒子および組成物は、腸内または非経口経路を含むいくつかの異なる経路によって患者に投与され得る。非経口投与は、静脈内、皮膚または皮下、鼻腔、筋肉内、眼内、経上皮、腹腔内、および局所(皮膚、眼、結腸、鼻腔、吸入、およびエアゾールを含む)の経路、ならびに結腸全身経路による投与を含む。
例示的なリガンドおよびリンカーの合成
ラクトース長鎖リンカーリガンド
Lac−O−EG6−C11−SHおよびLacLLとも呼ばれ、このリガンド+リンカー部分のIUPAC名は(ω−11−チオウンデシル)−(ヘキサエチレングリセリルβ−D−ラクトースである。これはA.G.Barrientos,J.M.de la Fuente,T.C.Rojas,A.Fernandez,S.Penades,Chem.Eur.J.2003,9,1909−1921に従って合成することができる。
金表面に結合された肝臓標的化分子、結合リンカー、および炭水化物希釈剤を有する金ナノ粒子の調製および特徴付けが下記で説明される。
LAcLL、AL、およびGlcC2を参考に従って合成した。HAuCl4、NaBH4、KCN、KOH、およびメタノールをシグマアルドリッチから入手した。すべての試薬はさらに精製することなく使用した。ミリQ水(18.2mΩ)をSimplicity水精製システム(メルクミリポア)から得た。ナノ粒子をKCN/KOHエッチング後の1H NMR、DLS、TEM、およびゼータ電位によって特徴付けした。ナノ粒子溶液の金濃度をICP−MSまたはMPAESによって測定した。
LacLL(0.0410mmol、32.5mg、1.41mL)、GlcC2(0.185mmol、44.5mg、1.58mL)、およびAL(0.228mmol、67.7mg、2.40mL)のメタノール溶液を100mL丸底フラスコに加え、メタノール(32.4mL)で希釈して、0.012M濃度のリガンド溶液を得た。ついでHAuCl4(60.0mg、0.152mmol、1当量)の水溶液(6.09mL)を加えた。反応混合物をNaBH4(126mg、3.34mmol、22当量)の水溶液(3.33mL)によってボルテックス撹拌下で還元した。得られる黒色ナノ粒子溶液を楕円式振とう器によって室温で35分間振とうさせた。時間が経過とともにナノ粒子が沈殿した。反応終了後、溶液中に残存するナノ粒子を遠心分離(4500rpmで1分間)によってスピンダウンさせ、沈殿を4mLのミリQ水で再溶解した。水性NP懸濁液を洗浄済みのアミコンフィルター(4mL、10kDa)に移した。濃縮後、ナノ粒子を遠心ろ過によってミリQ水(3〜4mL)を用いて4回洗浄した。最後に、ナノ粒子をミリQ水の最終容量6mLに収集した。ナノ粒子をKCN/KOHエッチング後の1H NMR、DLS、TEM、およびゼータ電位によって特徴付けした。ナノ粒子溶液の金濃度をICP−MSによって測定した。TEM:平均直径2.06nm。
LacLL(0.0870mmol、69.8mg、3.55mL)、GlcC2(0.0710mmol、17.0mg、1.67mL)、およびAL(0.159mmol、47.2mg、1.96mL)のメタノール溶液を100mL丸底フラスコに加え、メタノール(19.3mL)で希釈して、0.012M濃度のリガンド溶液を得た。ついでHAuCl4(40.0mg、0.102mmol、1当量)の水溶液(4.24mL)を加えた。反応混合物をNaBH4(84.8mg、2.24mmol、22当量)水溶液(2.33mL)によってボルテックス撹拌下で還元した。得られる黒色ナノ粒子溶液を楕円式振とう器によって室温で35分間振とうさせた。時間経過でナノ粒子が沈殿した。反応終了後、溶液中に残存するナノ粒子を遠心分離(4500rpmで1分間)によってスピンダウンさせ、沈殿を4mLのミリQ水で再溶解した。水性NP懸濁液を洗浄済みのアミコンフィルター(4mL、10kDa)に移した。濃縮後、ナノ粒子を遠心ろ過によってミリQ水(3〜4mL)を用いて4回洗浄した。最後に、ナノ粒子をミリQ水の最終容量4mLに収集した。ナノ粒子をKCN/KOHエッチング後の1H NMR、DLS、TEM、およびゼータ電位によって特徴付けした。ナノ粒子溶液の金濃度をICP−MSによって測定した。TEM:平均直径1.98nm。
結合リンカーの官能化ならびに化学療法剤および蛍光色素を備えた肝臓標的化金ナノ粒子の特徴付けを下記で説明する。
スルホローダミンB酸クロリド、EDC・HCl、NHS、およびDMSOはシグマアルドリッチから入手した。Pt(IV)−コハク酸はCharnwood Molecularから入手した。ドキソルビシンはLC Labsから入手した。すべての試薬はさらに精製することなく使用した。ミリQ水(18.2mΩ)はSimplicity水精製システム(メルクミリポア)から得た。ナノ粒子をKCN/KOHエッチング後の1H NMR、DLS、TEM、およびゼータ電位によって特徴付けした。ナノ粒子溶液の金濃度をICP−MSまたはMPAESによって測定した。
5mLのLacLL−NP1ナノ粒子水溶液(低濃度の標的化リガンド)(21.4μmol反応性AL)をアミコンフィルター(4mL、10kDa)中で遠心分離(4500rpmで15分×2)によって濃縮し、DMSOで2.5mLの容量に希釈した。Pt(IV)−コハク酸(22.9mg、52.9μmol)のDMSO溶液(528μL、0.1M)に添加する前に、EDC・HCl(12.2mg、63.4μmol)のDMSO溶液(127μL、0.5M)およびNHS(7.29mg、63.4μmol)のDMSO溶液(63.4μL、1.0M)を混合して室温で15分間インキュベーションした。前活性化の30分後、反応混合物をナノ粒子溶液に加えて混合物を楕円式振とう器を用いて室温で一昼夜振とうさせた。反応溶液を25mLのミリQ水で希釈し、濃縮して繰り返しミリQ水で洗浄した。黒色残渣を5.00mLのミリQ水に採取した。ナノ粒子をKCN/KOHエッチング後の1H NMR、DLS、TEM、およびゼータ電位によって特徴付けした。ナノ粒子溶液の金およびプラチナの濃度をICP−MSによって測定した。[Au],MPAES:2.84mg/mL、[Pt],MPAES:0.19mg/mL
LacLL−NPにおけるアミノ官能基のコハク酸化:
8.0mLのLacLL−NP1ナノ粒子水溶液(低濃度の標的化リガンド)(33.8μmol反応性AL)をアミコンフィルター(15mL、10kDa)で遠心分離(4500rpmで15分)によって濃縮し、DMSOで8.0mLの容量に希釈した。無水コハク酸(16.9mg、169μmol)をDMSO(564μL)で希釈して0.5M溶液を得、ナノ粒子溶液に加えた。反応混合物を楕円式振とう器を用いて室温で一昼夜振とうさせた。反応溶液を25mLのミリQ水で希釈して濃縮し、繰り返しミリQ水で洗浄した。黒色残渣を5.00mLのミリQ水に採取した。ナノ粒子をKCN/KOHエッチング後の1H NMR、DLS、TEM、およびゼータ電位によって特徴付けした。ナノ粒子溶液の金およびプラチナの濃度をICP−MSによって測定した。
3.0mLのLacLL−NPナノ粒子水溶液(10.0μmol反応性コハク酸化AL)をアミコンフィルター(4mL、10kDa)中で遠心分離(4500rpmで15分)によって濃縮し、DMSOで3.0mLの容量に希釈した。15分間インキュベーションしたEDC・HCl(4.82mg、25.1μmol)およびNHS(5.75mg、50.0μmol)のDMSO溶液(416μL)をナノ粒子溶液に加え、混合物を楕円式振とう器を用いて室温で2時間振とうさせた。水(80mL)で希釈したナノ粒子溶液を遠心分離によってろ過し、HEPES緩衝液(pH7.8、25.0mL)で希釈し、ドキソルビシンの溶液(20mMヘペスで2.00mg/mL、2.50mL)を直ちに加えた。結合反応物を室温で1時間インキュベーションした。ドキソルビシンと結合されたナノ粒子を遠心分離ろ過(アミコン、15mL、10kDa)によってミリQ水で精製した。残存溶液をMES緩衝液(3.0mL)で収集した。ナノ粒子をKCN/KOHエッチング後の1H NMR、DLS、TEM、およびゼータ電位によって特徴付けした。ナノ粒子溶液の金濃度をICP−MSによって測定した。[Au],比色分析:1.05mg/mL、[Doxo],比色分析:0.66mg/mL
3mLのLacLL−NP1ナノ粒子水溶液(0.609μmolナノ粒子)(低濃度の標的化リガンド)を遠心分離ろ過(アミコン、4mL、10kDa)によって濃縮し、Na2CO3/NaHCO3緩衝液(0.1M、pH9.3)で1回洗浄した。残存溶液を1.5mLの緩衝液に溶解した。NP溶液に、DMF中のスルホローダミンB酸クロリド溶液(133μL、9.14μmol、5.27mg)を加え、日光を遮断して楕円式振とう器によって室温で一昼夜振とうさせた。反応混合物を事前に洗浄したアミコンフィルター(4mL、10kDa)に移した。ナノ粒子溶液を遠心分離して、Na2CO3/NaHCO3緩衝液(0.1M、pH9.3)で3回洗浄を繰り返し、ミリQ水でろ液が透明になるまで繰り返した。最後に、ナノ粒子を3mLのミリQ水に収集した。ナノ粒子をKCN/KOHエッチング後の1H NMR、DLS、TEM、およびゼータ電位によって特徴付けした。ナノ粒子溶液の金濃度をICP−MSによって測定した。
ラクトース長鎖リンカー金ナノ粒子の肝臓標的化特性は、インビボ体内分布試験における異なるナノ粒子構築体の比較によって示すことができた。高含量および低含量の肝臓標的化分子を有する2種のラクトース長鎖リンカーナノ粒子(LacLL−NP1(低)+LacLL−NP2(高))、2種の同様なラクトース短鎖リンカーナノ粒子(LacSL−NP1(低)+LacSL−NP2(高))(ラクトースはC2リンカーに結合される)、および非標的化ナノ粒子(Glc−NP)をそれぞれマウスに静脈内注入した。
細胞株およびトランスフェクション
肝癌細胞株であるHepG2細胞を、10%FCS(ギブコ)を添加したDMEM(シグマアルドリッチ)で、10cmペトリ皿(BD)において37℃、95%空気および5%CO2で増殖させ、PBS 1×(シグマアルドリッチ)で洗浄し、0.05%トリプシン・EDTA(ギブコ)による処理で継代した。生きた細胞をトリパンブルー排除アッセイにおいて血球計数器で計数した。HepG2細胞は、マイコプラズマ属のすべてのメンバーに共通であるプライマーセットを用いてマイコプラズマについて定期的に試験した(Choppaら,1998)。細胞を6ウェルプレートに2〜3×104個/cm2の密度で接種し、続いてpEGFP−Lucベクター(Clontech)とPEI25(シグマアルドリッチ)の1:3および1:5のモル比でトランスフェクションした。トランスフェクションでは、800μg/μgのG418を培養培地に加えることによって細胞を48時間で選択した。続いて、細胞を新鮮な培地に維持してコンフルエントまで増殖させた。インビトロ生物発光シグナルを評価するため、Mithrasマルチモードプレートリーダーを補助として用いたシンプルルシフェラーゼアッセイを、CCLR緩衝液中で細胞溶解物にD−ルシフェラーゼを加えることによって実施した(25mMトリスHCl pH7.8、2mMのDTT、2mMのEGTA、10%グリセロール、1%トライトンX−100)。
70匹の雌BALB/cヌードマウス(6週齢)を認定供給業者(Janvier Labs)から入手した。すべてのマウスは特定病原体フリー条件下の層流キャビネットにおいて室温、12時間明/暗サイクルで飼育し、食餌はペレットおよび水が自由に与えられた。USCの実験動物委員会が動物試験を承認し、結果として、すべての動物実験が動物愛護ガイドラインに適合した。
対数増殖期のHepG2細胞(0.1mLのPBSに細胞105個)を無胸腺ヌードマウス(各実験群n=6)の左脇腹に皮下注射して腫瘍保持マウスモデルを確立した。腫瘍移植を目視検査で定期的に確認し、最後にIVISスペクトル(Caliper LifeScience)における生物発光シグナルの表示によって確証した。D−ルシフェリンを投与(150mg/kg)して、生物発光を腫瘍細胞に蛍光NPとともに共局在化させた。腫瘍保持マウスをそれぞれ6匹の5群(コントロール群を含む)に分けた。腫瘍容積を[(長さ2×幅)/2]の式によって計算した(Soengasら,1999)。腫瘍が400mm3の容積に達したとき、ナノ粒子を静脈内に投与した。
化学療法剤Pt(IV)−コハク酸でペイロードされた肝臓標的化ラクトース長鎖リンカー金ナノ粒子(Pt−LacLL−NP+プラチナ)およびドキソルビシンでペイロードされた肝臓標的化ラクトース長鎖リンカー金ナノ粒子(Doxo−LacLL−NP+ドキソルビシン)の細胞毒性を、MTT(3−(4,5−ジメチルチアゾール−2−イル)−2,5−ジフェニルテトラゾリウムブロマイド)細胞生存アッセイにおいて遊離薬剤と比較して試験した(図7)。ヒト肝細胞癌細胞株HepG2をアッセイに使用した。
96ウェルマイクロタイタープレートにおける細胞接種
HepG2細胞をT−75フラスコ中で増殖させた。細胞を移動のため、培地をフラスコから取り出して、細胞を5分間トリプシン処理した。細胞をファルコンチューブに収集して完全細胞培地で希釈した。細胞計数をノイバウエルチャンバーで実施した。細胞溶液を調製してウェルあたり4000個(200μL/ウェル)を接種した。マイクロタイタープレートを37℃で24時間インキュベーションした。
各NPおよび薬剤について、細胞培地における組成物を調製した。化合物を異なる濃度で試験した(薬剤量に基づいて0.01、0.05、0.2、1、5、25、および125μM)。処理のため、細胞培地をすべてのウェルから取り除き、薬剤組成物用に交換した。200μL処理液を三つ組で各ウェルに加えた。マイクロタイタープレートを37℃で24、48、および72時間インキュベーションした。
1.5mLのMTT溶液(1.6mLのDMSO中に8.0mg)を完全細胞培地(フェノールレッド無含有)で希釈した。処理細胞培地をマイクロタイタープレートから除いて、ウェルを100μLのPBSで洗浄し、100μLのMTT反応性溶液を各ウェル加えた。37℃、1時間のインキュベーション後、MTT溶液を取り除いて、100μLのDMSOを加えてホルマザン色素を溶解した。吸光度を570nmで測定した。
データをOriginPro8を用いて分析した。基準化したデータをプロットして、曲線を非直線回帰曲線フィット(可変傾きを有するシグモイド用量−反応曲線)を用いてフィッティングさせた。IC50絶対値を補間によって得た。
いくつかのベース/ペプチド標的化GNPをインビトロでスクリーニングした。ガラクトース−C2−SH(Gal−C2)リガンドおよびHSPEG8COOHを含む混合コロナを有するGNPを、例1および2における前述と類似した方法論によって合成したが、グルコース−C2−SHをガラクトース−C2−SHで置き換え、アミノリンカーNH−2−EG6−SHをHSPEG8COOH(SH−EG8−COOHまたはSH−(OCH2CH2)8−COOHとも略す)で置き換えた。
次のGNP構築体を合成して、HepG2(肝臓肝細胞癌)細胞に対するこれらの腫瘍細胞致死能力を評価した。
Claims (11)
- 複数のナノ粒子と、少なくとも1種類の医薬品として許容可能な担体または希釈剤とを含む、哺乳動物対象における肝臓障害の治療用の医薬組成物であって、
前記ナノ粒子が、
金属を含むコアであって、前記コアの直径が1nm〜5nmの範囲である、コアと、
前記コアと共有結合された複数のリガンドと
を含み、
前記リガンドが
(i)ガラクトース、ラクトース、N−アセチルガラクトサミンから選択される少なくとも1種類の肝臓標的化リガンドと、
(ii)抗増殖剤である生物活性剤を含む少なくとも1種類のペイロードリガンドと、
(iii)モノサッカライドである炭水化物を含む少なくとも1種類の希釈リガンドであって、少なくとも1種類の希釈リガンドが、2〜10原子の鎖長を有する第三リンカーを介して前記コアと共有結合されている、少なくとも1種類の希釈リガンドと、
を含み、
前記コアと共有結合された複数のリガンドの総数は、20個以上である、
医薬組成物。 - 前記肝臓標的化リガンドが、第一リンカーを介して前記コアと共有結合され、前記第一リンカーが2〜50原子の鎖長を有する、請求項1に記載の医薬組成物。
- 前記第一リンカーが末端イオウ原子を介して前記コアと結合される、請求項1又は2に記載の医薬組成物。
- 前記ペイロードリガンドが、メイタンシノイドDM4、ドキソルビシン、イリノテカン、プラチナ(II)、プラチナ(IV)、テモゾロミド、カルムスチン、カンプトテシン、ドセタキセル、ソラフェニブ、メイタンシン、メイタンシノイドDM1、モノメチルアウリスタチンE(MMAE)、およびパノビノスタットからなる群から選択される化合物を含む、請求項1〜3のいずれか一項に記載の医薬組成物。
- 前記ナノ粒子が、検出可能な標識を含む少なくとも1種類のさらなるペイロードリガンドをさらに含む、請求項1〜4のいずれか一項に記載の医薬組成物。
- 前記少なくとも1種類の希釈リガンドがグルコースまたはガラクトースを含む、請求項1〜5のいずれか一項に記載の医薬組成物。
- 前記コアが、Au、Ag、Cu、Pt、Pd、Fe、Co、Gd、Zn、またはこれらのいずれかの組み合わせからなる群から選択される金属を含む、請求項1〜6のいずれか一項に記載の医薬組成物。
- 前記組成物が注入可能な剤形である、請求項1〜7のいずれか一項に記載の医薬組成物。
