Description
[1]CD4+T細胞において、Dedicator of cytokinesis 8(DOCK8)タンパク質、Mammalian STE20−like kinase 1(MST1)タンパク質、及びEndothelial PAS domain protein 1(EPAS1)タンパク質を含む複合体が形成不能となる遺伝子変異を有する、アトピー性皮膚炎モデル非ヒト動物。
[2]前記遺伝子変異が、DOCK8遺伝子又はMST1遺伝子のノックアウト又はノックダウンである、[1]に記載のアトピー性皮膚炎モデル非ヒト動物。
[3]再編成したT細胞受容体(TCR)が遺伝子導入されている、[1]又は[2]に記載のアトピー性皮膚炎モデル非ヒト動物。
[4]DOCK8−/−TCR Tg又はMST1−/−TCR Tgの遺伝子型を有する、[1]〜[3]のいずれかに記載のアトピー性皮膚炎モデル非ヒト動物(ここで、TCR Tgは再編成したTCRが遺伝子導入されていることを表す。)。
[5]前記TCRがANDである、[3]又は[4]に記載のアトピー性皮膚炎モデル非ヒト動物。

1実施形態において、本発明は、アトピー性皮膚炎の治療剤の創薬標的としてのEPAS1を提供する。EPAS1は、EPAS1タンパク質をコードする遺伝子であってもよいし、EPAS1タンパク質であってもよい。
1実施形態において、本発明は、EPAS1阻害剤を有効成分として含有するアトピー性皮膚炎の治療剤を提供する。
発現阻害物質としては、例えば、siRNA、shRNA、リボザイム、アンチセンス核酸、低分子化合物等が挙げられる。これらの発現阻害物質を生体に投与することにより、EPAS1タンパク質の発現を阻害することができる。その結果、IL−31の発現を抑制し、アトピー性皮膚炎を治療することができる。上記の低分子化合物としては、例えば実施例において後述するFM19G11(3−[(2,4−ジニトロベンゾイル)アミノ]−安息香酸2−(4−メチルフェニル)−2−オキソエチルエステル,[2−オキソ−2−(p−トリル)エチル]3−[(2,4−ジニトロベンゾイル)アミノ]安息香酸、例えば、Moreno-Manzano V., et al., FM19G11, a new hypoxia-inducible factor (HIF) modulator, affects stem cell differentiation status., J. Biol. Chem., 285 (2), 1333-1342, 2010. を参照。)等が挙げられる。
EPAS1タンパク質に対する特異的結合物質は、EPAS1タンパク質に結合し、例えばSP1タンパク質との結合を抑制することができる。その結果、IL−31の発現を抑制し、アトピー性皮膚炎を治療することができる。同様に、SP1タンパク質に対する特異的結合物質は、SP1タンパク質に結合し、EPAS1タンパク質との結合を抑制することができる。その結果、IL−31の発現を抑制し、アトピー性皮膚炎を治療することができる。
EPAS1タンパク質によるIL−31遺伝子の発現を低下させる物質は、アトピー性皮膚炎の治療剤であるといえる。このような物質は、例えば、上述したいずれかのスクリーニング方法により、化合物ライブラリー等からスクリーニングすることができる。
上述したアトピー性皮膚炎の治療剤は、それ自体を生体に投与してもよいし、薬学的に許容される担体と混合した医薬組成物として製剤化したものを生体に投与してもよい。
1実施形態において、本発明は、EPAS1阻害剤の有効量を、治療を必要とする患者又は患畜に投与する工程を備える、アトピー性皮膚炎の治療方法を提供する。
(マウス)
DOCK8−/−マウスは以前に作製した(非特許文献2を参照。)。ヘテロマウス(DOCK8+/−)はC57BL/6バックグラウンドに9世代以上戻し交配した。DOCK8+/−マウスはAND TCR Tgマウスと交配し、DOCK8+/−AND Tgマウス又はDOCK8−/−AND Tgマウスを得た。EPAS1lox/loxマウスはジャクソンラボラトリー社から購入し、CD4−Cre Tgマウスと交配してCD4−Cre+EPAS1lox/loxDOCK8−/−AND Tgマウスを得た。マウスは特定病原菌フリー(SPF)環境下において九州大学動物施設で飼育した。週齢及び性別が一致した同腹仔を対照に用いた。すべての実験は九州大学動物実験倫理委員会のガイドラインにしたがって行った。マウスはランダムに選択し、遺伝子型にしたがって各実験群に割り当てた。実験者はマウスの遺伝子型を知らされずに実験を行った。
マウスは11×14×20cmのアクリル製のかごに入れて少なくとも1時間順化させた後、その行動をビデオ撮影した。ビデオを再生し、2時間の期間における掻痒行動の合計を決定した。
皮膚組織は4%(w/v)パラホルムアルデヒドで固定し、パラフィンブロックに包埋した。厚さ3μmの切片をヘマトキシリン及びエオジンで染色し、明視野顕微鏡観察した。免疫蛍光解析においては、組織はOCTコンパウンド(サクラファインテック社)に包埋し、液体窒素で凍結した。厚さ10μmの凍結切片を4%(w/v)パラホルムアルデヒドで30分間固定し、10%ヤギ血清で1時間ブロッキングした。続いて、切片を、フィコエリスリン(PE)標識抗マウスCD3抗体(型式「17A2」、BioLegend社)、フルオレセインイソチオシアネート(FITC)標識抗マウスCD45R/B220抗体(型式「RA3−6B2」、BDバイオサイエンス社)、FITC標識抗マウスCD8a抗体(型式「53−6.7」、BDバイオサイエンス社)又はビオチン標識抗マウスCD4抗体(型式「H129.19」、BDバイオサイエンス社)及び後に続くアレクサフルオロ488標識ストレプトアビジン(サーモフィッシャーサイエンティフィック社)で染色した。EPAS1染色は、マウス胚性線維芽細胞(MEF)3×105個/mLをポリ−L−リジンコートされたガラスボトムディッシュ(マツナミ社)上で1%O2環境下で30時間培養し、4%(w/v)パラホルムアルデヒドで30分間固定し、0.2%Triton X−100で30分間処理することにより透過処理した。続いて、10%ヤギ血清で1時間室温で処理してブロッキングし、4’,6−ジアミジノ−2−フェニルインドール(DAPI、同仁化学研究所社)及びウサギ抗EPAS1抗体(NOVUS Biologicals社)で染色し、続いてアレクサフルオロ488標識ロバ抗ウサギIgG抗体(Fabフラグメント、ジャクソンイムノリサーチ社)で染色した。全ての画像は共焦点レーザー走査型顕微鏡(カールツァイス社)で撮影した。
血清サンプル中及び細胞培養上清中のIL−31の濃度は、ELISAキット(ヒトサンプルはR&Dシステムズ社、マウスサンプルはUSCN社)を用いて説明書にしたがって測定した。血清中のIgE及びIgG2bの濃度の測定は次のようにして測定した。まず、血清サンプルを段階希釈し、ヤギ抗マウスIgE抗体又はヤギ抗マウスIg(IgM+IgG+IgA,H+L)抗体(Southern Biotech社)でコートした96ウェルプレートに入れた。2時間インキュベート後、ウェルをリン酸緩衝液(PBS)で洗浄し、アルカリフォスファターゼ標識ラット抗マウスIgE抗体又はヤギ抗マウスIgG2b抗体(Southern Biotech社)と共にインキュベートした。
以下の抗体及び試薬を使用した。FITC標識抗マウスCD45R/B220抗体(型式「RA3−6B2」)、FITC標識講マウスCD4抗体(型式「RM4−5」)、ビオチン標識抗マウスCD4抗体(型式「RM4−5」)、PE標識抗マウスCD8a抗体(型式「53−6.7」)、PE標識抗マウスCD44抗体(型式「IM7」)、FITC標識抗マウスCD62L抗体(型式「MEL−14」)、FITC標識抗マウスVα11抗体(型式「RR8−1」)、FITC標識抗マウスVα2抗体(型式「B20.1」)、ビオチン標識抗マウスVβ3(型式「KJ25」)、ビオチン標識抗マウスVβ5(型式「MR9−4」)、アロフィコシアニン(APC)又はPerCP−5.5シアニン標識ストレプトアビジン(以上いずれもBDバイオサイエンス社)、ビオチン標識抗マウスCD90.2/Thy1.2(型式「30−H12」、eBioscience社)。抗体で染色する前に、細胞を抗FcγIII/IIレセプター抗体(型式「2.4G2」、BDバイオサイエンス社)と共に氷上で10分間インキュベートし、Fc受容体をブロックした。フローサイトメトリー解析はFACS Calibur(商品名、BDバイオサイエンス社)を用いて行った。
マウスCD4+T細胞は、脾臓及び抹消リンパ節からダイナビーズマウスCD4を用いた磁気ソーティングにより分離し、DETACHaBEADマウスCD4(ライフテクノロジーズ社)で処理し、10%加熱不活性化処理した仔ウシ胎児血清(FCS)、50μM 2−メルカプトエタノール(ナカライテスク社)、2mM L−グルタミン(ライフテクノロジーズ社)、100U/mLペニシリン、100μg/mLストレプトマイシン、1mMピルビン酸ナトリウム(ライフテクノロジーズ社)及びMEM非必須アミノ酸(ライフテクノロジーズ社)を含有するRPMI1640培地(和光純薬)に懸濁した。抗原刺激は、CD4+T細胞(3×105個/ウェル)を、24ウェルプレート中で、T細胞を除去し放射線照射した脾臓細胞(5×106個/ウェル)と共に、MCC88−103ペプチド(配列番号1、3μg/mL)又はOVA323−339ペプチド(配列番号4、1μg/mL)の存在下で培養することにより行った。
ISOGEN(商品名、ニッポンジーン社)を使用して全RNAを抽出した。RNaseフリーDNaseI(ライフテクノロジーズ社)で処理した後、RNAサンプルをオリゴ(dT)プライマー(ライフテクノロジーズ社)及びSuperScript III逆転写酵素(ライフテクノロジーズ社)を用いて逆転写し、PCR増幅した。
ISOGEN(商品名、ニッポンジーン社)を使用して全RNAを抽出した。cRNAの増幅及び標識を、市販のキット(型式「Low Input Quick Amp Labeling Kit」、アジレントテクノロジー社)を用いて行った。
DOCK8−/−又はDOCK8+/−AND Tg CD4+T細胞におけるEPAS1遺伝子又はMST1遺伝子の発現をノックダウンするために、市販のキット(商品名「Accell siRNA SMART pool」、型式「E−040635−00−0005」(EPAS1用)又は「E−059385−00−0005」(MST1用)、Dharmacon社)を使用した。
マウスIL−31レポータープラスミド(pGL4.10−IL−31)を作成するために、マウスIL−31遺伝子のプロモーターの−1367〜−1の位置をPCRにより増幅し、pGL4.10[luc2]ベクター(プロメガ社)にサブクローニングした。IL−31プロモーターの欠失及び変異はPCRにより行った。これらのプラスミドは、Lipofectamine2000試薬(ライフテクノロジー社)を用いてMEFに導入し、ルシフェラーゼレポーターアッセイを行った。
MEFに、pRL−SV40−Renilla luciferaseプラスミド(0.1μg、プロメガ社)、pGL4.10−IL−31(2μg)、及びpcDNA−EPAS1又はその変異体(2μg)を共導入した。
細胞を1%Triton X−100、150mM NaCl、1mM EDTA、1mM EGTA、2.5mMピロリン酸ナトリウム、1mM β−グリセロリン酸、1mM Na3VO4及びcomplete protease inhibitors(ロシュ社)を含有する20mM Tris−HClバッファー(pH7.5)で溶解した。
定法により、MEFから核抽出物を調製した。簡単には、細胞をPBSで洗浄し、バッファーA(10mM HEPES−K+、pH7.9、10mM KCl、0.1mM EDTA、0.1mM EGTA、0.5mM DTT、1mM PMSF)に懸濁し、氷上で15分間インキュベートした。続いて、NP−40を終濃度0.67%添加することにより細胞を溶解させ、直ちに10秒間ボルテックスした。
野生型(WT)及びEPAS1−/−のMEFを、2枚の100mm培養ディッシュ中で、10%FCSを含有する10mLのDMEM培地で12時間培養し、続いて1%O2環境下で24時間培養した。続いて、細胞を1%ホルムアルデヒドで10分間、室温でクロスリンクした。続いてグリシンを添加して5分間、室温で反応させ中和した。続いて、細胞を、プロテアーゼ阻害剤を含有するPBS中で掻き取って洗浄し、続いて市販のキット(型式「Magna ChIP(商標)HiSens Kit」、ミリポア社)を用いて説明書にしたがって核を抽出した。
群間の差は、対応のない片側スチューデントt検定(2群の場合)又は一元配置分散分析(多重群の場合)により比較し、続いて事後ボンフェローニテストを行った。また、0.05未満のP値を有意であると見なした。
[実験例1]
ANDは、MHCクラスII I−Eκ分子と複合体を形成した、moth cytochrome c(MCC)の第88番目〜103番目のアミノ酸からなるペプチド(配列番号1)を認識するTCRである。
DOCK8−/−AND Tgマウスの掻痒行動を解析した。図2は、6週齢(n=8)、12週齢(n=6)及び18週齢(n=8)のDOCK8+/−AND Tgマウス及びDOCK8−/−AND Tgマウスの2時間あたりの掻痒行動を定量した結果を示すグラフである。グラフの値は平均値±標準偏差を示す。図中の「*」は危険率5%未満で有意差があることを示す。図2に示すように、DOCK8−/−AND Tgマウスは、12及び18週齢において、激しい掻痒行動を起こした。この結果は、この皮膚炎が非常に痒いことを示す。
DOCK8−/−AND Tgマウスの皮膚の組織学的及び免疫組織学的解析を行った。図3(a)及び(b)は、18週齢のDOCK8+/−AND Tgマウス(図3(a))及び同腹のDOCK8−/−AND Tgマウス(図3(b))の皮膚をヘマトキシリン及びエオジン染色した結果を示す写真である。スケールバーは50μmを示す。
DOCK8+/−AND Tgマウス及びDOCK8−/−AND Tgマウスの血清中のIgE及びIgG2bの濃度を測定した。図5(a)及び(b)は、6週齢(n=13)、12週齢(n=8〜9)及び18週齢(n=4)のDOCK8+/−AND Tgマウス及びDOCK8−/−AND Tgマウスの血清中のIgE(図5(a))及びIgG2b(図5(b))の濃度を示すグラフである。グラフ中の線は平均値を示し、「*」は危険率5%未満で有意差があることを示す。
DOCK8+/−AND Tgマウス及びDOCK8−/−AND Tgマウスの血清中のIL−31の濃度を測定した。図6は、6週齢(n=6)及び18週齢(n=6)のDOCK8+/−AND Tgマウス及びDOCK8−/−AND Tgマウスの血清中のIL−31の濃度を示すグラフである。グラフ中の線は平均値を示し、「**」は危険率1%未満で有意差があることを示す。
[実験例6]
IL−31は、主に活性化されたCD4+T細胞から産生される。そこで、発明者らは、DOCK8+/−AND Tgマウス及びDOCK8−/−AND TgマウスのCD4+T細胞の免疫学的な機能を比較した。
DOCK8+/−AND Tgマウス由来のCD4+T細胞及びDOCK8−/−AND Tgマウス由来のCD4+T細胞の抗原特異的な増殖を検討した。図9は、I−Eκを発現するB10.BRマウスの脾臓細胞の存在下において、MCCペプチド(配列番号1)で刺激した、DOCK8+/−AND Tgマウス及びDOCK8−/−AND Tgマウス由来のCD4+T細胞の増殖を測定した結果を示すグラフである。
DOCK8+/−AND Tgマウス及びDOCK8−/−AND Tgマウス由来のCD4+T細胞をMCCペプチドで1次刺激し、サイトカイン遺伝子の発現を検討した。
DOCK8+/−AND Tgマウス由来のCD4+T細胞及びDOCK8−/−AND Tgマウス由来のCD4+T細胞を2次刺激した後のIL−31タンパク質の発現量をELISA法により測定した。
図10(b)に示したように、活性化したDOCK8−/−AND Tgマウス由来のCD4+T細胞は、IL−4遺伝子の発現を上昇させた。
上述した知見が、異なる抗原に対するTCRにまで拡張することが可能か否かを検討するために、発明者らはI−Ab分子によって提示されるオブアルブミン(OVA)ペプチド(配列番号4)を認識する、OTII TCRを発現するDOCK8−/−マウスを作製して解析した。
続いて、DOCK8+/−OTII Tgマウス由来のCD4+T細胞及びDOCK8−/−OTII Tgマウス由来のCD4+T細胞の抗原特異的な増殖を検討した。図15は、I−Abを発現するC57BL/6マウスの脾臓細胞の存在下において、OVAペプチド(配列番号4)で刺激した、DOCK8+/−OTII Tgマウス及びDOCK8−/−OTII Tgマウス由来のCD4+T細胞の増殖を測定した結果を示すグラフである。
続いて、DOCK8+/−OTII Tgマウス由来のCD4+T細胞及びDOCK8−/−OTII Tgマウス由来のCD4+T細胞を2次刺激し、IL−31遺伝子の発現量を測定した。図16は、IL−31遺伝子の発現を測定した結果を示すグラフである。IL−31遺伝子の発現量は、DOCK8+/−OTII Tgマウス由来のCD4+T細胞の結果を1とした相対値で示した。グラフは、3つの独立した実験の代表的な結果における、平均値±標準偏差を示す。図中の「*」は危険率5%未満で有意差があることを示す。
DOCK8+/−OTII Tgマウス由来のCD4+T細胞及びDOCK8−/−OTII Tgマウス由来のCD4+T細胞を2次刺激した後のIL−31タンパク質の発現量をELISA法により測定した。
続いて、18週齢のDOCK8+/−OTII Tgマウス(n=7)及びDOCK8−/−OTII Tgマウス(n=7)の血清中のIL−31の濃度を比較した。
続いて、18週齢のDOCK8+/−OTII Tgマウス(n=4)及び同腹のDOCK8−/−OTII Tgマウス(n=4)の皮膚の組織学的解析を行った。
[実験例17]
DOCK8−/−AND TgマウスのCD4+T細胞によるIL−31の過剰発現の機構を明らかにするために、発明者らはマイクロアレイ解析を行った。その結果、抗原刺激後において、対照であるDOCK8+/−AND TgマウスのCD4+T細胞と比較して、856個の遺伝子が、DOCK8−/−AND TgマウスのCD4+T細胞で高発現していることが明らかとなった。
DOCK8−/−AND TgマウスのCD4+T細胞におけるEPAS1遺伝子をノックダウンし、IL−31遺伝子の発現に及ぼす影響を検討した。
続いて、DOCK8−/−AND TgマウスのCD4+T細胞におけるEPAS1遺伝子の欠失が、IL−31遺伝子の発現に及ぼす影響を検討した。
CD4−Cre+EPAS1lox/loxDOCK8−/−AND Tgマウスにおける、アトピー性皮膚炎の発症、掻痒行動及び血清中のIL−31の濃度を検討した。
続いて、EPAS1がどのようにしてIL−31のプロモーターを活性化するのかについて検討した。
続いて、MEFに、実験例21のレポーターコンストラクト、野生型のEPAS1の発現ベクター、及びAryl hydrocarbon receptor nuclear translocator(ARNT)遺伝子に対するsiRNA又はSpecificity protein 1(SP1)遺伝子に対するsiRNAを導入し、IL−31プロモーターの活性を測定した。
続いて、発明者らは、実験例21のレポーターコンストラクトにおけるIL−31プロモーターを少しずつ短くしたレポーターコンストラクトを作製し、野生型のEPAS1の発現ベクターと共にMEFに導入し、IL−31プロモーターの活性を測定した。
続いて、発明者らは、IL−31プロモーターに部位特異的変異を導入したレポーターコンストラクトを作製し、野生型のEPAS1の発現ベクターと共にMEFに導入し、IL−31プロモーターの活性を測定した。
続いて、Electrophoretic mobility shift assay(EMSA)によりIL−31プロモーターにSP1が結合するか否かについて検討した。
続いて、クロマチン免疫沈降(ChIP)により、IL−31プロモーターへのSP1の結合に与えるEPAS1の影響を検討した。
[実験例27]
EPAS1は、細胞質から核に移動し、核内でIL−31プロモーターの活性化を媒介する。そこで、発明者らは、DOCK8がIL−31の誘導を負に制御する機構を解明するために、野生型のMEF及びDOCK8−/−のMEFの間でEPAS1の細胞内局在を検討した。
野生型のDOCK8(WT)又はDOCK8の変異体(ΔN及びΔDHR2)をDOCK8−/−MEFで発現させ、EPAS1の細胞内局在を検討した。
ところで、Mammalian STE20−like kinase 1(MST1)は、T細胞の接着、遊走、増殖及びアポトーシスに関与するセリン/スレオニンキナーゼである。DOCK8に結合するタンパク質を探索した結果、発明者らは、DOCK8がそのN末端領域を介してMST1に結合することを明らかにした。
続いて、野生型のMEFにおいて、MST1のノックダウンがEPAS1の核への局在に及ぼす影響を検討した。
DOCK8+/−AND TgマウスのCD4+T細胞のMST1遺伝子をノックダウンした場合のIL−31遺伝子の発現を検討した。
[実験例32]
EPAS1がマウスのCD4+T細胞におけるIL−31の誘導に必須であることが明らかになったため、発明者らは、ヒトのCD4+T細胞におけるEPAS1の役割を検討した。
続いて、アトピー性皮膚炎患者及び健常人(対照)の血清中のIL−31の濃度を測定して比較した。図41は血清中のIL−31の濃度を測定した結果を示すグラフである(n=6)。グラフ中の線は平均値を示す。図中の「**」は危険率1%未満で有意差があることを示す。
続いて、TCR刺激により誘導される、CD4+T細胞によるサイトカイン遺伝子の発現を検討した。
続いて、発明者らは、アトピー性皮膚炎患者由来のCD4+T細胞に対するEPAS1阻害剤の影響を検討した。EPAS1阻害剤としては、FM19G11及びHIFVIIを使用した。正確な機構は解明されていないが、FM19G11はEPAS1の発現を阻害することが知られている。
続いて、EPAS1阻害剤であるFM19G11(n=6)及びHIFVII(n=3)が、アトピー性皮膚炎患者由来のCD4+T細胞のTCR刺激により誘導されるサイトカイン遺伝子発現に及ぼす影響を検討した。
ところで、HIFVIIはEPAS1のPAS−Bドメインに結合し、EPAS1とARNTとの結合を阻害することが知られている。このことを確認するために、ARNT及びEPAS1をHEK−293T細胞で共発現させ、HIFVIIの存在下又は非存在下で免疫沈降を行った。図46(a)及び(b)は、免疫沈降の結果を示す写真である。その結果、HIFVII処理はARNT及びEPAS1の結合を阻害することが確認された。
[実験例38]
AND TCRを有しCD4+T細胞特異的にDOCK8を欠失したコンディショナルノックアウトマウスが、アトピー性皮膚炎を発症するか否かを検討した。
[実験例39]
実験例16において、DOCK8−/−OTII Tgマウスはアトピー性皮膚炎を発症しないことが示された。そこで、DOCK8−/−OTII Tgマウス由来のCD4+T細胞を、OVA抗原を全身に発現するCAG−OVAマウスに移入し、痒みを惹起するか否かを検討した。
Claims (21)
- アトピー性皮膚炎モデル非ヒト動物の製造方法であって、非ヒト動物に、CD4+T細胞において、Dedicator of cytokinesis 8(DOCK8)タンパク質、Mammalian STE20−like kinase 1(MST1)タンパク質及びEndothelial PAS domain protein 1(EPAS1)タンパク質を含む複合体が形成不能となる遺伝子変異、及び、AND T細胞受容体(TCR)遺伝子を導入する工程を有し、
前記遺伝子変異及びAND TCRを有する非ヒト動物がアトピー性皮膚炎モデル非ヒト動物である、製造方法。 - 前記遺伝子変異が、DOCK8遺伝子又はMST1遺伝子のノックアウト又はノックダウンである、請求項1に記載の製造方法。
- 前記アトピー性皮膚炎モデル非ヒト動物が、DOCK8−/− AND Tg又はMST1−/− AND Tgの遺伝子型を有する、請求項1又は2に記載の製造方法(ここで、AND TgはAND TCRが遺伝子導入されていることを表す。)。
- アトピー性皮膚炎モデル細胞の製造方法であって、細胞に、DOCK8タンパク質、MST1タンパク質及びEPAS1タンパク質を含む複合体が形成不能となる遺伝子変異を導入する工程を有し、前記遺伝子変異を有する細胞がアトピー性皮膚炎モデル細胞である、製造方法。
- 前記遺伝子変異が、DOCK8遺伝子又はMST1遺伝子のノックアウト又はノックダウンである、請求項4に記載の製造方法。
- 被験物質の投与下で、請求項1〜3のいずれか一項に記載の製造方法により製造された非ヒト動物の掻痒行動の量を定量する工程と、
定量された前記掻痒行動の量が、前記被験物質の非投与下における前記非ヒト動物の掻痒行動の量と比較して低下していた場合に、前記被験物質はアトピー性皮膚炎の治療剤であると判断する工程と、を備える、アトピー性皮膚炎の治療剤のスクリーニング方法。 - 被験物質の存在下で、アトピー性皮膚炎の患者由来のCD4+T細胞、DOCK8−/−CD4+T細胞又はMST1−/−CD4+T細胞のTCRを刺激し、前記CD4+T細胞によるインターロイキン(IL)−31の発現量を定量する工程と、
前記IL−31の発現量が、前記被験物質の非存在下で前記CD4+T細胞のTCRを刺激した場合のIL−31の発現量と比較して低下していた場合に、前記被験物質はアトピー性皮膚炎の治療剤であると判断する工程と、を備える、アトピー性皮膚炎の治療剤のスクリーニング方法。 - 被験物質の存在下で、T細胞にEPAS1遺伝子を発現させ、IL−31の発現量を定量する工程と、
