Description

CARD14のアミノ酸配列としては、
(a)配列番号2または4に記載のアミノ酸配列またはそのフラグメントからなる、ポリペプチド;
(b)配列番号2または4に記載のアミノ酸配列において、1以上のアミノ酸が置換、付加および欠失からなる群より選択される1つの変異を有し、かつ、生物学的活性を有する、ポリペプチド;
(c)配列番号1または3に記載の塩基配列のスプライス変異体または対立遺伝子変異体によってコードされる、ポリペプチド;
(d)配列番号2または4に記載のアミノ酸配列の種相同体である、ポリペプチド;または
(e)(a)〜(d)のいずれか1つのポリペプチドに対する同一性が少なくとも70%、少なくとも80%、少なくとも90%、少なくとも95%、少なくとも96%、少なくとも97%、少なくとも98%、または少なくとも99%であるアミノ酸配列を有し、かつ、生物学的活性を有する、ポリペプチド、であり得る。ここで、生物学的活性とは、代表的に、CARD14の有する活性またはマーカーとして同じ生物内に存在する他のタンパク質から識別し得ること(例えば、抗原として用いられる場合特異的エピトープとして機能し得る領域を含むこと)をいう。
等を挙げることができるがこれらに限定されない。
本発明で使用され得るCARD14阻害剤は、CARD14の作用を阻害する任意の物質をいい、その作用機序はCARD14の発現を阻害すること、機能を阻害することをなどを含む。したがって、「CARD14阻害剤」は、CARD14の発現を阻害する物質およびCARD14の機能を阻害する物質、このほか作用機序が不明であっても最終的にCARD14の作用が阻害される任意の物質を含み、抗体、抗体フラグメント、その誘導体、核酸、低分子、その他のアンタゴニスト等であり得るがこれらに限定されない。このような種々の抗体等については、本願明細書の他の箇所において説明される。好ましくは、このようなCARD14阻害剤は、CARD14を特異的に阻害するものであることが有利である。CARD14以外の因子への影響による副作用を低減することができるからである。
(1)CARD14遺伝子のmRNAに対するアンチセンス核酸、
(2)CARD14遺伝子のmRNAに対するリボザイム核酸、
(3)CARD14遺伝子のmRNAに対してRNAi活性を有する核酸もしくはその前駆体。
本発明における「CARD14遺伝子のmRNAに対するアンチセンス核酸」とは、該mRNAの塩基配列と相補的もしくは実質的に相補的な塩基配列またはその一部を含む核酸であって、標的mRNAと特異的かつ安定した二重鎖を形成して結合することにより、タンパク質合成を抑制する機能を有するものである。
CARD14遺伝子のmRNAの塩基配列と相補的もしくは実質的に相補的な塩基配列またはその一部を含む核酸の他の好ましい例としては、該mRNAをコード領域の内部で特異的に切断し得るリボザイム核酸が挙げられる。「リボザイム」とは、狭義には、核酸を切断する酵素活性を有するRNAをいうが、本明細書では配列特異的な核酸切断活性を有する限りDNAをも包含する概念として用いるものとする。リボザイム核酸として最も汎用性の高いものとしては、ウイロイドやウイルソイド等の感染性RNAに見られるセルフスプライシングRNAがあり、ハンマーヘッド型やヘアピン型等が知られている。ハンマーヘッド型は約40塩基程度で酵素活性を発揮し、ハンマーヘッド構造をとる部分に隣接する両端の数塩基ずつ(合わせて約10塩基程度)をmRNAの所望の切断部位と相補的な配列にすることにより、標的mRNAのみを特異的に切断することが可能である。このタイプのリボザイム核酸は、RNAのみを基質とするので、ゲノムDNAを攻撃することがないというさらなる利点を有する。CARD14遺伝子のmRNAが自身で二本鎖構造をとる場合には、RNAヘリカーゼと特異的に結合し得るウイルス核酸由来のRNAモチーフを連結したハイブリッドリボザイムを用いることにより、標的配列を一本鎖にすることができる[Proc. Natl. Acad. Sci. USA, 98(10): 5572-5577 (2001)]。さらに、リボザイムを、それをコードするDNAを含む発現ベクターの形態で使用する場合には、転写産物の細胞質への移行を促進するために、tRNAを改変した配列をさらに連結したハイブリッドリボザイムとすることもできる[Nucleic Acids Res., 29(13): 2780-2788 (2001)]。
本明細書においては、CARD14遺伝子のmRNAに相補的なオリゴRNAとその相補鎖とからなる二本鎖RNA、いわゆるsiRNAもまた、CARD14遺伝子のmRNAの塩基配列と相補的もしくは実質的に相補的な塩基配列またはその一部を含む核酸に包含されるものとして定義される。短い二本鎖RNAを細胞内に導入するとそのRNAに相補的なmRNAが分解される、いわゆるRNA干渉(RNAi)と呼ばれる現象は、以前から線虫、昆虫、植物等で知られていたが、この現象が動物細胞でも広く起こることが確認されて以来[Nature, 411(6836): 494-498 (2001)]、上記のアンチセンス核酸やリボザイムの代替技術として汎用されている。
がこれらに限定されない。
以下に本発明の好ましい実施形態を説明する。以下に提供される実施形態は、本発明のよりよい理解のために提供されるものであり、本発明の範囲は以下の記載に限定されるべきでないことが理解される。従って、当業者は、本明細書中の記載を参酌して、本発明の範囲内で適宜改変を行うことができることは明らかである。また、本発明の以下の実施形態は単独でも使用されあるいはそれらを組み合わせて使用することができることが理解される。

1つの局面では、本発明は、造血細胞におけるCARD14の発現に基づく多発性硬化症の治療剤または予防剤のスクリーニング方法を提供する。CARD14は従来知られていた分子ではあるが、その造血細胞、特にγδT細胞、中でも、表皮γδT細胞における発現をみることで、γδT細胞媒介性疾患および/または免疫介入の合併症の治療剤または予防剤をスクリーニングすることができることは見出されておらず、本発明において初めて提供されたものである。理論に束縛されることを望まないが、特に、造血細胞、特にγδT細胞、中でも、好ましくは表皮γδT細胞におけるCARD14の発現を抑制ないし消失させる剤等のCARD阻害剤は、γδT細胞媒介性疾患および/または免疫介入の合併症の治療剤または予防剤として使用することができることが理解される。したがって、本発明の方法は、造血細胞に候補物質を接触させる工程、および造血細胞におけるCARD14の発現を決定する工程であって、ここで該候補物質がCARD14の発現を抑制ないし消失させる場合該候補物質が乾癬の治療剤または予防剤として使用することができると決定される、工程を包含する。
CARD14の発現または機能を阻害する化合物をスクリーニングする場合、該スクリーニング方法は、CARD14を産生する能力を有する細胞を、被検物質の存在下および非存在下に培養し、両条件下におけるCARD14の発現量または機能の程度を比較することを含む。また、CARD14の機能を阻害する化合物は、精製したCARD14タンパク質への結合能、CARD14タンパク質とその結合タンパク質(例えば、CARD14受容体)との結合阻害活性を試験することによっても、スクリーニングすることができる。特に、このような細胞としては、造血細胞、好ましくはγδT細胞が有利であり、さらに有利には表皮のγδT細胞が好ましい。あるいは遺伝子組換え細胞を用いてもよい。このような細胞は、具体的には、CARD14をコードするDNA(即ち、配列番号1または3、好ましくは配列番号1で表される塩基配列または該塩基配列に対し相補性を有する塩基配列とストリンジェントな条件下でハイブリダイズし、且つ配列番号2または4、好ましくは配列番号2で表されるアミノ酸配列からなるタンパク質と同質の機能を有するポリペプチドをコードする塩基配列を含むDNA)を、適当な発現ベクター中のプロモーターの下流に連結して宿主動物細胞に導入することにより調製することができる。
(1)疾患(例えば、乾癬、多発性硬化症、TLRベース/IMQ/免疫刺激アジュバントによる副作用を減少させるための、またはγδT細胞媒介性疾患および/または免疫介入の合併症等)の標的細胞を、CARD14の存在下および非存在下で被検物質に接触させる工程
(2)各条件下での前記細胞における疾患(例えば、乾癬、多発性硬化症、TLRベース/IMQ/免疫刺激アジュバントによる副作用を減少させるための、またはγδT細胞媒介性疾患および/または免疫介入の合併症)の反応(例えば、乾癬特有の反応、多発性硬化症特有の反応、TLR関連の反応、合併症特有の症状)の程度を測定する工程
(3)被検物質の非存在下において測定した場合と比較して、CARD14の存在下で当該反応を抑制し、CARD14の非存在下で当該反応を抑制しなかった被検物質を、CARD14の機能を阻害して疾患(例えば、乾癬、多発性硬化症、TLRベース/IMQ/免疫刺激アジュバントによる副作用を減少させるための、またはγδT細胞媒介性疾患および/または免疫介入の合併症等)の治療または予防作用を示す物質の候補として選択する工程を含む。
本発明の医薬は、それ自体を投与してもよいし、または適当な医薬組成物として投与してもよい。投与に用いられる医薬組成物としては、本発明の医薬と薬理学的に許容され得る担体、希釈剤もしくは賦形剤とを含むものであってよい。このような医薬組成物は、経口または非経口投与に適する剤形として提供される。
本明細書において用いられる分子生物学的手法、生化学的手法、微生物学的手法は、当該分野において周知であり慣用されるものであり、すでに引用されたものも含め、例えば、Sambrook J. et al.(1989). 本実施例では、CARD14欠損マウスを作製し、2種のマウス乾癬様モデル(1つは、IMQクリームにより誘導されるモデルであり、もう1つは、組み換えIL−23タンパク質のマウスの耳への注射により誘導されるモデルである)を使用して、それらの免疫学的機能を評価した。
(マウス株)
Card14−/−(詳細は下記および図2に説明する)、Tlr7−/−、Tlr9−/−およびTlr7−/−Tlr9−/−二重欠損マウス(Onishi et al., 2015)を本発明者らの動物施設で飼育し、特定病原体除去条件下で維持した。Rag2−/−Il2rg−/−マウスをTaconic Biosciencesから入手した。Tcrd−/−マウスをJackson Laboratoriesから入手した。C57BL/6(WT)マウスをCLEA Japan, Inc.(東京、日本)から購入した。すべての実験を、医薬基盤・健康・栄養研究所の実験動物の管理および使用に関する施設倫理ガイドラインに従って行った。
Card14−/−マウスは、相同組換えによってエキソン2およびエキソン3の一部を取り除くように設計された標的化ベクターを使用して、Lexicon Pharmaceuticals(The Woodlands,TX,USA)により作製された。PCRを慣例的なマウス遺伝子型判定のために使用した。プライマーは以下を使用した。

Card14−/−標的化マウスを、C57BL/6バックグラウンドに対して少なくとも8回戻し交配し、マーカー支援スピードコンジェニック法を使用して決定した。結果として、N8世代でB6バックグラウンドゲノムの99%を超える置換が生じた(Markel et al.,1997,Nature Genetics. 17:280-284)。
以前記載されたように、乾癬状皮膚炎の誘導のために、6日間連続で、62.5mgの5%IMQを含有するBeselna(日本以外ではAldaraTMとして販売されている;3M Pharmaceticals, St Paul, Minn)クリーム(Mochida Pharmaceutical Co.,Ltdより購入)の局所投与によりマウスの両耳を処置した(非特許文献3(van der Fits et al., 2009))。
BM細胞(5×106)をドナーWTマウスおよびドナーCard14−/−マウスの大腿骨および脛骨をフラッシングすることにより得た。マウス(WTおよびCard14−/−)は致死線量のX線(9Gy)を受け、その後、眼窩静脈叢を介したBM細胞の静脈内注射によって再構成した。IMQクリームによる処置を再構成してから6週間後に行った。
実験の最終日にマウスを屠殺し、耳を回収して、その後のパラフィンにおける包埋のために10Nのホルマリンで固定した。パラフィン組織切片(直径4μm)を脱パラフィンし、CARD14に対するウサギ抗マウスポリクローナル抗体(1:100; Proteintech, Chicago, IL, USA)、ケラチン5に対するウサギ抗マウスポリクローナル抗体(1:400;Covance, Berkeley, CA, USA)およびKi67に対するウサギ抗マウスモノクローナル抗体(Thermo Fisher Scientific)で染色した。一次抗体を添加する前に切片を0.01Mクエン酸(pH6.0に調節)中で10分間80℃で処理した。二次抗体は、ヤギ抗ウサギ免疫グロブリンにコンジュゲートしたペルオキシダーゼ標識ポリマー(Dako)を使用した。3,3’−ジアミノベンジジン(Dako)を色素源として使用すると、褐色の染色が生じた(Uchio et al., 2002, Lab Invest. 82:619-628)。Aperio ScanScope (Aperio Technologies)を使用して、すべてのスライドを絶対倍率20倍でスキャンした。
マウスを屠殺し、それらの耳を回収した。この耳の皮膚組織を封入剤(Tissue-Tek OCT compound, Sakura Finetechnical. Co., Tokyo, Japan)中で−80℃で瞬間冷凍した。CARD14およびγδTCRについての染色を、クライオスタット(Leica)を使用して全マウスの耳6μmの切片に対して行った。切片を典型的には、15分間室温(RT)で4%パラホルムアルデヒドで固定し、1%BSAおよび0.2%Triton X−100を含有するPBS中の2%ヤギ血清で1時間室温でブロックし、抗マウスγδTCR(1:50;BD PharMingen, GL3, PE)およびCARD14に対するウサギ抗マウスポリクローナル抗体(1:150)と共に一晩4℃でインキュベートした。PBS中で洗浄した後、上記切片をAlexaFluor 488コンジュゲート化ロバ抗ウサギIgG(H+L)(1:250; Invitrogen)と共に室温で1時間インキュベートし、次いでPBS中で洗浄し、DAPIを使用して共染色した。LeicaTCS SP2共焦点顕微鏡システムを使用して画像を取得し、Volocity (version 6.2.1)を使用して分析した。
表皮細胞懸濁液を、表皮をトリプシン−EDTAと共に45分間37℃でインキュベートすることにより調製した。完全RPMI培地(Nacalai Tesque)中の1.6mg/mLのコラゲナーゼIV(Worthington)および0.1mg/mL Dnase-I(Sigma)含有する緩衝液を使用して、真皮懸濁液を得た。その後、細胞を70μmのポアサイズのセルストレーナー(BDFalcon)を通してろ過し、単一細胞懸濁液を得た(Nakashima et al. The Journal of Allergy and Clinical Immunology. 134:100-107, 2014; Tokura et al., 1994, The Journal of Investigative Dermatology. 102:31-38)。
保存した細胞を400×gで5分間遠心分離することによって回収した。RNAをQiagen RNeasy kit (Qiagen)を使用して単離した。その後、RNAを逆転写酵素(ReverTra Ace, Toyobo, Osaka, Japan)とオリゴdTプライマーとを使用してcDNAに逆転写した。リアルタイムPCR混合物は、総量20μLで、10μLのSYBR Green Master mix (Roche)、フォワードプライマーおよびリバースプライマー(CARD14およびグリセルアルデヒド−3−リン酸脱水素酵素;GAPDHに対しては200nM)ならびに2μLのcDNAサンプルからなる。増幅用のプライマーは以下の通りである。

PCR反応を、LightCycler480 System II (Roche Life Science)のMicroAmp optical 384ウェル反応プレートにおいて40サイクル(95℃で15分間、60℃で1分間)行った。閾値サイクルの間に検出される蛍光シグナル(ΔCt)を、上記装置にインストールされたソフトウェアにより記録した。RNAおよびcDNA品質におけるばらつきに関して標的遺伝子レベルを標準化するために、GAPDHを内部コントロールと同じ条件下で増幅した。
以下の抗体をBDPharMingenから購入した:抗CD8a(Ly-2,APC);抗CD45(30-F11,PerCP/Cy5.5);抗マウスγδTCR(GL3,PE);抗IL-17A(TC11-18H10,PE);および抗NK1.1(PK136,PE)。BioLegendからの以下の抗体を使用した:抗CD3e(134-2C11,PerCP/Cy5.5);抗CD4(RM4-5,PE/Cy7);抗CD11b(M1/70,FITC);抗CD45(30-F11,PE/Cy7);および抗TCRβ(H57-597,PerCP/Cy5.5)。以下の抗体をeBioscienceから購入した:抗CD19(1D3,PacificBlue);抗IL-22(IL22JOP,APC);および抗TCRγδ(GL3,APC)。FACS分析のために、耳の固定された領域をTrephine(8-mm,Kai Industries Co., Ltd.)を使用してくりぬいた。細胞をFc-block (BioLegend)と共に15分間予めインキュベートし、その後、30分間4℃で適切な細胞表面抗体により標識した。細胞内サイトカイン染色のために、細胞を50ng/ml PMA(Sigma)および500ng/mlイオノマイシン(Sigma)で刺激し、GolgiStop(BD Bioscience)を用いて5分間37℃で処理した。細胞内染色のために、細胞を、BD Cytofix/Cytoperm 固定/透過処理キット(BDBiosciences)で固定および透過処理し、その後30分間4℃で抗体により染色した。BD LSR IIフローサイトメーターをFACS分析のために使用した。FACSAriaフローサイトメーターをセルソーティングのために使用した。その後、試料を、FlowJoソフトウェア(Treestar)を使用して分析した。
単独または100μgのLPSを含まないOVA(Seikagaku)を含有するかのいずれかで、20μl PBSの皮内注射を0日目および7日目に行った。採血後、抗体力価を14日目に測定した。マウスの耳を、以前記載されたように、IMQを使用して6日間連続で処理して、乾癬状皮膚炎を誘導した。OVA特異的抗体を、ELISAを使用して定量化した。簡単に説明すると、平底96ウェルMaxiSorpプレート(Nunc)を、0.5M NaHCO3中の50μg/mLのOVAでコーティングした。プレートを4℃で一晩インキュベートし、0.05%Tween20を含むPBSで洗浄し、1時間室温でブロックした。洗浄後、50μLの希釈された血清を2時間室温でインキュベートした。その後、プレートを洗浄し、アルカリホスフェートコンジュゲート化ヤギ抗マウスIgG1抗体およびIgG2C抗体(1:4000,Southern Biotech)と共に2時間室温でインキュベートした。プレートを洗浄し、その後、TMB基質溶液(Kirkegaard & PerryLaboratories)を使用して、結合抗体を可視化した。反応を2N H2SO4を添加することにより停止し、Gen5ELISA (Biotek(登録商標))プログラムを使用して、450nmの吸光度をBioStackTM (BioTek Instruments Inc.)マイクロプレートリーダーで読み取った。
Mann−Whitney U検定またはScheffe検定を使用して、群間の差の統計学的有意性ついて試験した。統計学的有意性をP<0.05として認めた。
(TLR7およびTLR9の両方がIMQ誘導性乾癬状皮膚炎に必要だが、IL−23誘導性乾癬状皮膚炎には必要でない)
インビボの乾癬病態形成における自然免疫系でのCARD14の役割を評価するために、本発明者らは2種の確立したマウス乾癬様モデルを利用した。一方のモデルは、IMQを含有するAldaraTMクリームを6日間両耳に局所適用することにより誘導される(非特許文献3(van der Fits et al., 2009))。IMQはTLR7アゴニストであるので、本発明者らはTLR欠損マウスを使用して、TLR7が本発明者らの実験乾癬モデルにおいて必須であるのか、または不必要であるのかを評価した。以前の報告(非特許文献4(Walter et al., 2013))と一貫して、TLR7単一欠損(Tlr7−/−)マウスは、WTマウスと同様の乾癬状皮膚炎を発症した(データは示さず)。同様に、TLR9単一欠損(Tlr9−/−)マウスは、WTマウスと同一の表現型を示した(データは示さず)。著しく対照的には、同じ実験設定において、TLR7およびTLR9の二重欠損(Tlr7−/−Tlr9−/−)マウスを評価した際に、Tlr7−/−Tlr9−/−マウスは、WTマウスと比較して、適用の最後の3日間、耳の厚み、表皮過角化および真皮細胞浸潤の顕著な減少を示した(図1Aおよび1B)。多くのグループによって以前に推測されてきたが(Pantelyushin et al., 2012, The Journal of Clinical Investigation.122:2252-2256; 非特許文献4(Walter et al., 2013))、本発明者らの結果は、初めてIMQ誘導性乾癬状皮膚炎がTLR7およびTLR9の両方を必要とするという証拠を示した。
よりCARD14のインビボにおける役割を特徴づけるために、本発明者らは、CARD14欠損(Card14−/−)マウスを作製し(図2)、IMQ誘導性乾癬様モデルおよびIL−23誘導性乾癬様モデル(非特許文献3(van der Fits et al., 2009);非特許文献8(Zheng et al., 2007))の両方におけるCARD14の役割を評価した。本発明者らは、IMQクリームを6日間連続で適用するか、または1日おきに5日間IL−23を注射した。WTマウスは、IMQの適用の後、悪化した皮膚の状態を有し、皮膚の厚みが増加したが、Card14−/−マウスを皮膚炎症から有意に保護した(図3A)。組織化学的実験は、Card14−/−マウスの耳の皮膚におけるケラチン5およびKi67を染色した過剰増殖細胞が、コントロールマウスのものと同様であり、他方で、IMQの適用を受けたWTマウスにおける過剰増殖細胞は激しく増加したことを明らかにした(図3B)。さらに、IL−23誘導性の耳の膨化は、Card14−/−マウスにおいて顕著に抑制された(図3C)。表皮過角化(ケラチン5およびKi67が染色された過剰増殖細胞として見られる)および真皮炎症(H&E染色における浸潤細胞として見られる)は、IL−23注射されたCard14−/−マウスにおいて両方とも減少した(図3D)。これらの結果は、CARD14がIMQクリームの適用およびIL−23の注射によって誘導される乾癬様皮膚炎の形成に必須であることを示している。したがって、本発明者らは、CARD14が、TLR7およびTLR9の下流のシグナル伝達に必要であり、さらにTLR7およびTLR9を必要としないIL−23の下流のシグナル伝達にも必要であるので、自然免疫細胞よりもむしろ、適応免疫細胞におけるシグナル伝達経路に関連すると結論付けた。
CARD14は、ケラチノサイトノの自然免疫に重要であることが示されたが(非特許文献12(Fuchs-Telem et al., 2012); 非特許文献9(Jordan et al., 2012))、本発明者らの上記結果は、他の細胞型におけるCARD14についての潜在的な役割を示唆しており、CARD14はIL−23シグナル伝達の下流で機能することを示唆している。この問題をさらに明確にするために、本発明者らは相反性骨髄(BM)キメラを作製し、IMQ誘導性乾癬様モデルを使用してCARD14の役割をさらに評価した。WTマウスおよびCard14−/−マウス由来のBM細胞は、X線照射されたCard14−/−マウスおよびWTマウスにそれぞれ移植した。6週間後、マウスを6日間連続でIMQクリームで両耳に対して処置した。乾癬様炎症が、WTマウス由来のBMが移植されたCard14−/−マウスおよびWTマウスにおいて誘導されたが、Card14−/− BMが移植された照射されたWTマウスまたはCard14−/−マウスでは誘導されなかった(図4A)。これは、造血BM由来細胞におけるCARD14発現は、IMQ誘導性モデルにおける乾癬の病態形成に必須であるが、放射線抵抗性皮膚常在細胞におけるCARD14発現は必須ではなかったことを示している。本発明者らは、RAG遺伝子に加えて、ナチュラルキラー(NK)細胞、ナチュラルキラーT(NKT)細胞、自然リンパ球(ILC)およびγδT細胞によって制御されるT細胞およびB細胞の潜在的な寄与について明らかにした。本発明者らは、γδT細胞を欠いているT細胞レセプターδ欠損(Tcrd−/−)マウスと、T細胞、NK細胞、NKT細胞およびILCを欠いているRag2−/−Il2rg−/−マウスにおける乾癬様モデルの重症度を比較した。以前の報告(Pantelyushin et al., 2012, The Journal of Clinical Investigation.122:2252-2256)と一貫して、Tcrd−/−マウスおよびRag2−/−Il2rg−/−マウスが、IMQクリームの適用後、WTマウスと比較した皮膚の膨化の顕著な減少を示した(図4B)。ケラチノサイトを含む非造血細胞におけるCARD14が、乾癬様炎症における役割を担うという可能性を除外することはできないが、上記結果は、BM由来細胞、潜在的にはγδT細胞におけるCARD14は、IMQ誘導性乾癬様皮膚炎の発症に必要であることを強く示唆している。
いくつかの異なる細胞型におけるCARD14発現を試験するために、耳の皮膚由来の細胞を単離し、リアルタイム定量ポリメラーゼ連鎖反応(qPCR)を使用してmRNAを測定した。以前の報告(非特許文献9(Jordan et al., 2012))と一貫して、CARD14 mRNAは、ケラチノサイトにおいて高レベルで発現された(図5A)。免疫細胞の中で、耳の皮膚のγδT細胞におけるCARD14 mRNAは、ケラチノサイトと同程度に高く、他の免疫細胞型よりも高かった(図5Aおよび5B)。CARD14を発現する細胞型をさらに明らかにするために、本発明者らは、IL−23の投与の5日後に、CARD14およびT細胞レセプターγδ(TCRγδ)に対する抗体を使用して耳の皮膚の共焦点顕微鏡分析を行った。IL−23を受けたWTマウスの表皮領域において、CARD14陽性シグナルを検出し、他方で、IL−23を受けたCard14−/−マウスにおいて、CARD14陽性シグナルを検出しなかった。また、WTマウスおよびCard14−/−マウスの両方において、TCRγδ陽性シグナルを表皮と真皮との間の周辺領域において検出し、特定のTCRγδ陽性シグナルと、細胞質で発現されたCARD14陽性シグナルとが重なり、他方で、Card14−/−マウスにおいて重なった領域は検出されなかった(図5C)。これらのデータは、CARD14陽性γδT細部が乾癬様皮膚に存在することを示している。
IMQは、多くの他の適用において一般的に使用されるワクチンアジュバントであるTLR7のアゴニストであるので、本発明者らは、CARD14が、共投与される抗原に対するIMQのアジュバント効果に影響を与えるかどうかを対象とした。この疑問を解決するために、本発明者らは、IMQクリームを使用する乾癬様皮膚炎モデルの実験プロトコールを改変し、WTマウス、Tlr7−/−Tlr9−/−マウスおよびCard14−/−マウスを、両耳における皮内注射を介してオボアルブミン(OVA)で免疫化し、他方で、IMQクリームを同じ耳に局所的に適用した。耳の厚みの増加をWTマウスにおいて観察し、他方で、マウスにOVAタンパク質を注射しても、Tlr7−/−Tlr9−/−マウスおよびCard14−/−マウスは、7日目で一貫して耳の厚みが顕著に薄いことを示した(図6A)。WTマウスをOVA免疫化と共にIMQクリームで処置し、予想されるように、処置されていないOVA免疫化群と比較して、顕著に高いOVA特異的免疫グロブリン(IgG)1およびIgG2c抗体応答を示した(図6B)。OVA特異的IgG2c抗体力価は、Tlr7−/−Tlr9−/−マウスにおいて顕著に減少したが、Card14−/−マウスは、WTマウスに匹敵するレベルの血清抗OVA IgG2cを示した(図6B)。これらの結果は、IMQクリームが、共投与されたタンパク質抗原に対してアジュバント効果を有し、CARD14がアジュバント効果に影響を与えないことを示している。IMQ誘導性乾癬がIMQクリームの合併症であるので、CARD14阻害剤を開発すると、IMQ誘導性自然免疫機能およびそのアジュバント効果に影響を与えることなく乾癬状皮膚炎の病態形成を抑制する可能性がある。
次に、本実施例では、CARD14欠損により、多発性硬化症の実験的脱髄性疾患モデルの症状軽減効果がみられるかどうかを調べた。本実施例では、基本的にRuiらの文献(Yuxiang Rui, Tasuku Honjo, and Shunsuke Chikuma. Programmed cell death 1 inhibits inflammatory helper T-cell development through controlling the innate immune response. PNAS 2013 110 (40) 16073-16078)の記載に基づいて行った。
CARD14−/−マウスは、Ruiらの文献に準じて遺伝子をノックアウトすることにより作製した。
Ruiらの文献に準じてマウスを200μgのMOG35−55ペプチド(MOG35−55残基に相当するペプチド)、完全フロイントアジュバント(CFA)、および250μgの熱殺傷マイコバクテリア(Heat−killed mycobacteria;HKMTB)を含む0.2mLのエマルジョンで皮下(sc)免役した。加えて、マウスに、200ngの百日咳毒素(PTX)を免疫時および48時間後に2回の腹腔内(ip)注射を行った。いくつかの実験において、HKMTB、PTXまたはその両方を除いた最適未満(suboptimal)の免疫条件も用いた。マウスは毎日観察し、疾患の臨床的兆候を観察し、EAEスコアを以下のように評価した。
0, 無症状(no disease);
1, 尾の下垂(limp tail);
2, 後肢運動低下(hind limb weakness);
3, 一方の後肢の麻痺(paralysis of one hind limb);
4, 両方の後肢の麻痺(paralysis of both hind limbs);
5, 運動障害(limited movement);
6, 瀕死または死亡(moribund or death)。
本実施例では、乾癬の治療または予防剤、多発性硬化症の治療または予防剤、TLRベース/IMQ/免疫刺激アジュバントによる副作用を減少させるための治療剤または予防剤、γδT細胞媒介性疾患および/または免疫介入の合併症の治療または予防剤のスクリーニングを行う。

Claims (16)
- 造血細胞に候補物質を接触させる工程、および造血細胞におけるCARD14の発現を決定する工程であって、ここで該候補物質がCARD14の発現を抑制ないし消失させる場合該候補物質が乾癬の治療剤または予防剤として使用することができると決定される、工程を包含する、造血細胞におけるCARD14の発現に基づく乾癬の治療剤または予防剤のスクリーニング方法であって、該造血細胞はγδT細胞である、方法。
- 造血細胞に候補物質を接触させる工程、および造血細胞におけるCARD14の発現を決定する工程であって、ここで該候補物質がCARD14の発現を抑制ないし消失させる場合該候補物質が多発性硬化症の治療剤または予防剤として使用することができると判定される工程を包含する、造血細胞におけるCARD14の発現に基づく多発性硬化症の治療剤または予防剤のスクリーニング方法であって、該造血細胞はγδT細胞である、方法。
- 造血細胞に候補物質を接触させる工程、および造血細胞におけるCARD14の発現を決定する工程であって、ここで該候補物質がCARD14の発現を抑制ないし消失させる場合該候補物質が、TLRベース/IMQ(イミキモド)/免疫刺激アジュバントによる乾癬の副作用を減少させるための治療剤または予防剤として使用することができると判定される工程を包含する、造血細胞におけるCARD14の発現に基づくTLRベース/IMQ/免疫刺激アジュバントによる乾癬の副作用を減少させるための治療剤または予防剤のスクリーニング方法であって、該造血細胞はγδT細胞であり、該乾癬はTLR7およびTLR9の両方に媒介性の乾癬またはIL−23誘導性乾癬を含む、方法。
- 造血細胞に候補物質を接触させる工程、および造血細胞におけるCARD14の発現を決定する工程であって、ここで該候補物質がCARD14の発現を抑制ないし消失させる場合該候補物質が、γδT細胞媒介性疾患および/または免疫介入の合併症の治療剤または予防剤として使用することができると判定される工程を包含する、造血細胞におけるCARD14の発現に基づくγδT細胞媒介性疾患および/または免疫介入の合併症の治療剤または予防剤のスクリーニング方法であって、該造血細胞はγδT細胞であり、該γδT細胞媒介性疾患および/または免疫介入の合併症は乾癬または多発性硬化症を含む、方法。
- 前記造血細胞は表皮γδT細胞である、請求項1〜4のいずれか1項に記載の方法。
- 前記γδT細胞はIL−17およびIL−22を産生する細胞である、請求項1〜5のいずれか1項に記載の方法。
- 非造血細胞においてCARD14が発現していないことを決定する工程をさらに包含する、請求項1〜6のいずれか1項に記載の方法。
- 非造血細胞に前記候補物質を接触させ、CARD14が発現に実質的な影響がない場合、該候補物質を前記治療剤または予防剤として選択する工程をさらに包含する、請求項1〜7のいずれか1項に記載の方法。
- 前記非造血細胞は皮膚常在細胞を含む、請求項7または8に記載の方法。
- 前記乾癬はTLR7およびTLR9の両方に媒介性の乾癬およびIL−23誘導性乾癬を包含する、請求項1または3〜9のいずれか1項に記載の方法。
- 造血細胞特異的CARD14阻害剤を含む乾癬の治療剤または予防剤であって、該造血細胞はγδT細胞であり、該乾癬はTLR7およびTLR9の両方に媒介性の乾癬またはIL−23誘導性乾癬を含み、該造血細胞特異的CARD14阻害剤は、CARD14に対する抗体もしくは抗体フラグメント、CARD14遺伝子のmRNAに対するアンチセンス核酸、CARD14遺伝子のmRNAに対するリボザイム核酸、またはCARD14遺伝子のmRNAに対してRNAi活性を有する核酸から選択される、治療剤または予防剤。
- 造血細胞特異的CARD14阻害剤を含む多発性硬化症の治療剤または予防剤であって、該造血細胞はγδT細胞であり、該造血細胞特異的CARD14阻害剤は、CARD14に対する抗体もしくは抗体フラグメント、CARD14遺伝子のmRNAに対するアンチセンス核酸、CARD14遺伝子のmRNAに対するリボザイム核酸、またはCARD14遺伝子のmRNAに対してRNAi活性を有する核酸から選択される、治療剤または予防剤。
- 造血細胞特異的CARD14阻害剤を含むTLRベース/IMQ/免疫刺激アジュバントによる乾癬の副作用を減少させるための治療剤または予防剤であって、該造血細胞はγδT細胞であり、該乾癬はTLR7およびTLR9の両方に媒介性の乾癬またはIL−23誘導性乾癬を含み、該造血細胞特異的CARD14阻害剤は、CARD14に対する抗体もしくは抗体フラグメント、CARD14遺伝子のmRNAに対するアンチセンス核酸、CARD14遺伝子のmRNAに対するリボザイム核酸、またはCARD14遺伝子のmRNAに対してRNAi活性を有する核酸から選択される、治療剤または予防剤。
- 造血細胞特異的CARD14阻害剤を含むγδT細胞媒介性疾患および/または免疫介入の合併症の治療剤または予防剤であって、該γδT細胞媒介性疾患および/または免疫介入の合併症は乾癬または多発性硬化症を含み、該造血細胞はγδT細胞であり、該乾癬はTLR7およびTLR9の両方に媒介性の乾癬またはIL−23誘導性乾癬を含み、該造血細胞特異的CARD14阻害剤は、CARD14に対する抗体もしくは抗体フラグメント、CARD14遺伝子のmRNAに対するアンチセンス核酸、CARD14遺伝子のmRNAに対するリボザイム核酸、またはCARD14遺伝子のmRNAに対してRNAi活性を有する核酸から選択される、治療剤または予防剤。
- 前記造血細胞は表皮γδT細胞である、請求項11〜14のいずれか1項に記載の治療剤または予防剤。
- 前記γδT細胞はIL−17およびIL−22を産生する細胞である、請求項11〜15のいずれか1項に記載の治療剤または予防剤。
