Description
本出願は、2014年9月22日に出願の米国特許仮出願第62/053,602号の利益を主張する。上記出願の全教示は参照により本明細書に組み込まれる。
本発明は、米国国立衛生研究所により授与されたNIH認可番号第R01 GM095654号に基づく政府の支援によりなされた。政府は本発明に対し一定の権利を有する。
本発明は、高周波または磁界を使用して、特定の細胞型により産生される内在性常磁性ナノ粒子を励起することにより、細胞機能の遠隔制御を行うための方法および組成物を提供する。細胞は、温度感受性および/または機械感受性チャネルの発現を指示する一連のDNAコンストラクトを発現する。このチャネルでは、常磁性金属ナノ粒子が励起されて物理的変化が生じ、これが、Ca2+もしくはNa+などの陽イオンまたはClなどの陰イオンを含むイオンの流入を誘導することにより細胞応答に変換される。内在性ナノ粒子と、他のチャネルとの共発現は、他のシグナル伝達経路の調節も同様に可能とすると思われる。このような誘導可能な細胞応答には、例えば、1種または複数の生理学的に活性なタンパク質の産生をもたらす遺伝子発現の増加を含んでよい。このようなタンパク質の発現を使って、ヒトまたは動物対象の種々の異なる遺伝性または後天性の疾患または障害を処置することができる。また、該方法を使って、イオンの流れにより活性を調節することが可能な内在性細胞を活性化または抑制することができる。したがって、該システムを使って、ニューロン、内分泌細胞、分泌細胞、収縮細胞およびイオン流の変化が細胞の活性を変える任意の他の細胞型の活性を調節することも可能である。
実施例1 遺伝的にコードされたナノ粒子を使ったマウスのグルコース恒常性の遠隔調節
方法
高周波電磁界
信号発生器により465Hz正弦波信号を供給し、増幅器(信号発生器、増幅器の両方とも、Ultraflex,Ronkonkoma,NY製)を介して、半径2.5cmの2ターンソレノイドに印加し、5mT以上の磁界強度を生成した。試料をソレノイド内に配置した。
24G銅線を1/4”パーマロイ−80ロッドの周りに1200回巻き付け、先端を直径100umの半球に旋盤加工することにより、ソレノイド磁気マイクロニードルを作製した。コイルを通る電流は、Beckman3A/30V調節可能電源により制御した。ニードル先端を引き寄せられる細胞から約30umの位置に配置し、磁力は1.8Aの電流により生成した。
イオンチャネルを機械的に開かせる力は、約0.2〜0.4pN31であると推定された。したがって、我々は、Kimらにより記載の方法を使って、磁気処理に由来する細胞に与える力を測定した。簡単に説明すると、鉄をロードされ、フェリチンを発現している細胞をパラホルムアルデヒドで固定後、緩衝水溶液に加え、静磁界に供した。細胞は磁石に向けて加速され、その速度は磁力の大きさに比例する。これは、球体の周りのクリープ波としてモデル化され、粒子に加わる流体の抗力は、細胞に加わる磁力に等しい32。Uを細胞速度、μを緩衝液の動粘度、D細胞を細胞の直径として、この直線関係は、次のようになる。

Wolfgang Liedkte(Duke University,NC)から送呈されたTRPV1(pcDNA3.1中の)を、pEGFPN1(Clontech,Mountainview,CA)に挿入した。GFP結合ナノボディ配列をIntegrated DNA Technologies(Coralville,IA)で合成して、TRPV1のN末端に融合し、αGFP−TRPV1を生成した。ミリストイル化シグナルをフェリチン軽鎖のN末端に挿入することにより、EF1alpha−フェリチンキメラ含有pCR2.1を改変してミリストイル化フェリチンを生成、またはPegfp−n1由来のGFP配列をフェリチン軽鎖のN末端に付加することによりGFP−フェリチンを生成した。phoenixパッケージング細胞を使ったレトロウイルス産生のために、TRPV1の後に2A配列およびフェリチンもしくはミリストイル化フェリチンまたはαGFP−TRPV1の後に2A配列およびGFP−フェリチンをMSCV−hygroプラスミドに挿入し、カルシウム応答性フューリンインスリンをMSCV−puroプラスミド(Clontech)に挿入した。複製欠損アデノウイルスの生成のために、これらの配列をまた、pVQ Ad CMV KNpAに挿入した。PCR産物の正確さおよびクローニングは、DNA塩基配列決定法により確認した。
ヒト胎児腎臓細胞(HEK293T)を10%のウシ胎仔血清を含むダルベッコ改変イーグル培地(Gibco,Carlsbad,CA)を用い、5%CO2下、37℃で培養した。Phoenixエコトロピックパッケージング細胞(Stanford University)を10%のウシ胎仔血清を含むダルベッコ改変イーグル培地(Gibco)を用い、5%CO2下、37℃で増殖させた。マウス間葉系幹細胞(Gibco)を10%のウシ胎仔血清を含むDMEM/F12培地を用い、5%CO2下、37℃で増殖させた。Phoenixシステムを使って、MSCのレトロウイルス感染により安定な細胞株を産生した。簡単に説明すると、Phoenix eco細胞(6cmディッシュ当たり2x106細胞)に、MSCV−puroまたはhygroプラスミドを遺伝子導入した。24時間後、培地を置換し、細胞を32℃に置いた。さらに24時間後、培地を吸引し、遠心分離して細胞残屑を除去した。ポリブレン(4μg/ml、Sigma−Aldrich、St.Louis,MO)含有RPMI培地/10%FBS中の1:2希釈を使って、phoenix細胞上清をMSCに加えた(6cmディッシュ当たり1x106細胞で播種)。細胞を32℃でさらに24時間インキュベートした後、培地をDMEM/F12培地/10%FBSで置換した。感染の48時間後に選択培地を加えた。安定に遺伝子導入されたMSCを、PBS中でプレインキュベーションした5x5x5mmゼラチンスポンジ足場(Gelfoam)上に、60μlの培地に再懸濁した2x106細胞を足場に直接添加することにより播種した。
遺伝子導入細胞をPBSで3回洗浄し、その後、スルフィンピラゾン500μM(Sigma)の存在下で、Fluo−4 3μM(Invitrogen)を室温で60分間ロードした。細胞をPBSで再度洗浄後、スルフィンピラゾンを含むPBS中で、30分間インキュベートした。細胞を洗浄後、カルシウムイメージングバッファー中で画像形成した。イメージングは、特注のセラミックレンズを備えたDeltavision personal DVイメージングシステム(Applied Precision,Issawaq,WA)を使って行った。RF処理の前および処理中、磁石処理の前もしくは処理中にまたは200μMの2−アミノエトキシジフェニルボレート(2−APB)による処理の前および処理の後で、細胞の画像形成を行った。
免疫細胞化学(ICC)および免疫組織化学(IHC)を使って、TRPV1、GFPおよびHAタグフェリチンの発現を検出し、細胞および組織中のアポトーシス細胞を定量した。細胞をPBSで洗浄後、2%パラホルムアルデヒド(Electron Microscopy Services,Hatfield,PA)で15分間固定した。組織を10%のホルマリン(Sigma)中、4℃で一晩固定し、PBS中30%のスクロース中に、4℃で、さらに24時間置いた。組織をOCTコンパウンドに包埋し、凍結後、20μmの凍結切片を切りだし、スライドガラス上に直接配置する。スライドを55℃で1時間置いた後、染色する前に、−80℃で保存する。細胞または組織切片を洗浄し、上記のように固定後、ブロッキング緩衝液(0.1%のトリトンX(Sigma)を含むPBS中の3%BSA(Sigma)および2%ヤギ血清(Sigma))中で1時間インキュベートした。その後、細胞をブロッキング緩衝液で希釈した一次抗体(ウサギ抗TRPV1 1:500、マウス抗Ha 1:1000(Cell signaling)、ニワトリ抗GFP 1:1000(Abcam)、ウサギ抗活性化カスパーゼ3 1:250(Promega))中、4℃で一晩インキュベートした。細胞または組織をPBS中で3回洗浄後、ブロッキング緩衝液中で2時間希釈した二次抗体(ヤギ抗ウサギ594またはヤギ抗ウサギ488、ヤギ抗ニワトリ488、ヤギ抗マウス360、全て1:1000)中でインキュベートした。細胞または組織をPBS中でさらに3回洗浄した後、Fluoromount(Southern Biotech,Birmingham,AL)を使ってマウントした。
雄無胸腺NCr−nu/nuマウス(NCI−Frederick、6〜8週齡)の非近交種、または雄C57Bl6マウスを使用し、制御された照明条件下(12時間の明/12時間の暗)、および温度(22℃)で、単一ケージに収容し、標準的マウス固形飼料を自由に摂取させた。動物飼育および実験手順は、既定ガイドラインに基づいて、Rockefeller Universityの実験動物委員会の承認を得て実施した(プロトコル11421)。
製造業者のプロトコルに従って、細胞上清中のプロインスリンをELISA(Alpco,Salem,NH)により測定した。Breeze2血糖計(Bayer;Leverkusen,Germany)を使って、血糖を測定した。血液を10分間遠心分離し、血漿を収集した。ヒト特異的ELISA(Alpco)により、マウス血漿中のヒトインスリンの血漿中濃度を測定した。
組織をトリゾール試薬(Invitrogen)中で、または細胞を緩衝液RLT中でホモジナイジングし、Qiagen RNAプレップキットを使ってRNAを精製することにより全RNAを単離した。Qiagen omniscript RTキットを使って、相補DNAを合成した。製造業者のプロトコルに従い、TaqManシステム(Applied Biosystems;Foster City,CA)を使って、リアルタイムPCRを行った。
全データに対し、スチューデントt検定を使って、統計的有意性を分析した。P値は、示した通りである。
我々は、最初に、フェリチンナノ粒子のTRPV1チャネルへの近接度に関して異なる3つの別々のコンストラクトを試験することにより、RFを使って遺伝子発現を調節するための遺伝的にコードされた発現系を最適化することに着手した。第1のコンストラクトでは、野生型温度感受性一過性受容器電位バニロイド1(TRPV1)陽イオンチャネルを、細胞質中で発現する、フェリチン軽鎖、可動性リンカー領域およびフェリチン重鎖12からなるフェリチンキメラタンパク質(TRPV1/フェリチン)と共発現させた(図1A左パネル)。第2のコンストラクトでは、野生型TRPV1チャネルを、細胞膜の方向にフェリチンを向けるミリストイル化シグナルとのキメラフェリチン融合タンパク質と共発現させた(TRPV1/ミリストイル化フェリチン)(図1A、中央パネル)。第3のコンストラクトでは、単一ドメイン抗GFPラクダ抗体13へのN末端融合により改変されたTRPV1チャネルを、GFPへのN末端融合を有するキメラフェリチンタンパク質と共発現させた。これにより、GFPタグフェリチンキメラの修飾TRPV1への係留が生じ、それにより、その成分が細胞膜に並置される(αGFP−TRPV1/GFP−フェリチン)(図1A、右パネル)。遺伝子導入されたHEK細胞のTRPV1、GFPおよびHAタグ(フェリチンキメラの可動性リンカー領域の)に対する免疫組織化学により、発現成分の予測された位置が確認された(図1B)。αGFP−TRPV1を遺伝子導入したHEK細胞は、2APBによる細胞内カルシウムの有意な増加を示した(Fluo−4蛍光の2.0倍の変化に対する0.85倍の変化(図1C))ので、TRPV1のN末端修飾は、TRPアゴニスト2APBに対して応答する能力を妨害することはなかった。さらに、αGFP−TRPV1/GFP−フェリチンを発現しているHEK細胞のRF(465kHz)による処理は、非遺伝子導入対照に比較して、細胞内カルシウムの有意な増加を生じた(Fluo−4蛍光の2.9倍の変化に対する0.8倍の変化、図1D)。
ここで、我々は、電波または磁界を使って、インビボで遺伝子発現を調節する遺伝的にコードされた系の開発につて報告する。以前の調査で、我々は、外部から送達された機能化ナノ粒子がエピトープタグTRPV1チャネルに結合し、遠隔操作によるRFシグナルをインビボでカルシウム流入および遺伝子発現に変換することを示した。しかし、この系を使って調節された遺伝子発現は、局所的細胞集団において達成できるのみであり、粒子の内部移行の結果として連続的応答を誘発するためには、反復ナノ粒子投与が必要である。遺伝的にコードされたフェリチン係留TRPV1系の使用は、それが、モノマー結合タンパク質を介して酸化鉄ナノ粒子封入GFPタグフェリチンに結合したTRPV1を活性化するために、非侵襲的低周波数RF電磁界または間欠的磁界を使って遠隔操作によりインビボで確実に反復される遺伝子発現の時間的制御を可能とする限り優れている。我々は、インビボでのインスリン遺伝子発現および放出を制御することにより、この遺伝的にコードされた系が、効果的に、繰り返して血糖を調節することができることを示すことにより、この系を検証する。我々はまた、この方法を移植幹細胞に使って、潜在的に、遺伝子操作された幹細胞中の重要なタンパク質発現の調節を可能とすることを示す。
方法
高周波電磁界および静磁界
信号発生器により465kHz正弦波信号を供給し、増幅器(信号発生器、増幅器の両方とも、Ultraflex,Ronkonkoma,NY製)を介して、半径2.5cmの2ターンソレノイドに印加し、電磁界を生成した。試験した電磁界強度は、31mT、27mTおよび23mTであった。試料をソレノイド内に配置した。
pEGFPN1およびMSCV−hygro中の抗GFPナノボディ−TRPV1−2A−GFPフェリチンを以前に記載30のように生成した。QuikChange XL部位特異的変異誘発キット(Agilent,Santa Clara,CA)を使って、部位特異的変異誘発により、ラットTRPV1中の残基I679のKへの変異を導入した。複製欠損アデノウイルスの生成のために、これらの配列を、pVQ Ad CMV KNpAに挿入した。Cre活性化組換え型アデノウイルスベクターを構築するために、2対の互換性のないlox部位、loxNとlox2722、を有するDNAコンストラクトを合成し、抗GFPナノボディ−TRPV1−2A−GFPフェリチンを2つの対の間に、アンチセンスの向きに挿入した。その後、複製欠損アデノウイルスの生成のために、flox化した逆転抗GFPナノボディ−TRPV1−2A−GFPフェリチンカセットをpVQ Ad CMV KNpAに挿入した。PCR産物の正確さおよびクローニングは、DNA塩基配列決定法により確認された。
組換え型アデノウイルス(Ad−CMV−GFP、Ad−CMV−αGFP−TRPV1/GFP−フェリチン、Ad−FLEX−αGFP−TRPV1/GFP−フェリチンおよびAd−FLEX−αGFP−TRPV1変異体/GFP−フェリチン)をViraquest(Iowa)でパッケージングした。最終の力価は、4x1010プラーク形成単位(pfu)/mlであった。AAV−EF1a−DIO−hChR2(H134R)−EYFPをUNC Viral Coreから購入した。
ヒト胎児腎臓細胞(HEK293T、(ATCC(登録商標)CRL−3216(商標)、マイコプラズマ試験およびSTRプロファイリングはATCCで実施)を10%のウシ胎仔血清(Gibco,Carlsbad,CA)を加えたダルベッコ改変イーグル培地(Gibco,Carlsbad,CA)を用い、5%CO2下、37℃で培養した。Phoenixエコトロピックパッケージング細胞(Stanford University)を10%のウシ胎仔血清を加えたダルベッコ改変イーグル培地(Gibco)を用い、5%CO2下、37℃で増殖させた。胎仔マウス視床下部N38細胞(Cellutions Biosystems Inc)を10%のウシ胎仔血清を加えたダルベッコ改変イーグル培地を用い、5%CO2下、37℃で増殖させた。
TRPV1は、非選択的陽イオンチャネルであり、2価陽イオン、特にカルシウムに対する比較的高い透過性を有する(Ca2+>Mg2+>Na+ K+ Cs+)55。ルテニウムレッドのある場合とない場合について、RF(31mT)または磁石(280mT)の効果を調査するために、安定に遺伝子導入した細胞をPBSで3回洗浄した後、スルフィンピラゾン500μMの存在下、室温で、45〜60分間、Fluo−4 3μM(Invitrogen)をロードした。細胞をPBSで再度洗浄後、スルフィンピラゾンを含むPBS中で、15〜30分間インキュベートした。細胞を洗浄後、カルシウムイメージングバッファー中で画像形成した。イメージングは、特注のセラミックレンズを備えたDeltavision personal DVイメージングシステム(Applied Precision,Issawaq,WA)を使って行った。3秒毎に3分間、画像を取得した。処理なし(8つのケース)、RF処理の前および処理中(9つのケース)、ネオジウム磁石の適用の前および適用中(45秒間、3つのケース)または200μMの2−アミノエトキシジフェニルボレートによる処理の前および処理の後(2−APB、2つのケース)で、細胞の画像形成を行った。ルテニウムレッド(100μM)の存在下でイメージングを繰り返した(条件毎に2つのケース)。画像をImage Jソフトウェアを使って解析した。
安定に遺伝子導入した細胞をKrebs−HEPES緩衝液で3回洗浄後、MQAE(N−(エトキシカルボニルメチル)−6−メトキシキノリニウムブロミド、5mM、Invitrogen)を室温で60分間ロードした。細胞をKrebs−HEPES緩衝液で洗浄した後、イメージング前に緩衝液中で15分間インキュベートした。イメージングを、750nmでの2光子励起による、40x対物レンズを用いたLSM510NLO多光子共焦点倒立システム(Zeiss)を使って行った。処理なし(4つのケース)、ネオジウム磁石の20秒間の適用の前および適用中(280mT)(6つのケース)、200μMの2−アミノエトキシジフェニルボレートによる処理の前および処理の後(2−APB、2つのケース)で、細胞の画像形成を行った。ルテニウムレッド(100μM)の存在下でイメージングを繰り返した(条件毎に2つのケース)。画像をImage Jソフトウェアを使って解析した。
免疫細胞化学(ICC)および免疫組織化学(IHC)を使って、TRPV1、GFPおよびFLAGタグフェリチンの発現を検出し、c−fos発現場所を特定し、細胞および組織中のアポトーシスを定量化した。細胞をPBSで2回洗浄後、2%パラホルムアルデヒド(Electron Microscopy Services,Hatfield,PA)で15分間固定した。組織を10%のホルマリン(Sigma)中、4℃で一晩固定し、振動ミクロトームで40μm切片を切り出した。固定細胞または組織切片を洗浄後、ブロッキング緩衝液(0.1%のトリトンX(Sigma)を含むPBS中の3%BSA(Sigma)および2%ヤギ血清(Sigma))中で1時間インキュベートした。その後、細胞および組織を、ブロッキング緩衝液で希釈した一次抗体(ウサギ抗TRPV1 1:500(AB95541、Chemicon)、マウス抗FLAG 1:1000(FLAGタグマウスmAb #8146、Cell signaling)、ニワトリ抗GFP 1:1000(b139703、Abcam)、ウサギ抗活性化カスパーゼ3 1:250(G7481、Promega4))またはウサギ抗cFos1:(PC38、Calbiochem)中、4℃で一晩インキュベートした。細胞または組織をPBS/0.1%トリトンX中で3回洗浄後、ブロッキング緩衝液中で2時間希釈した二次抗体(ヤギ抗ウサギ594(A1012)またはヤギ抗ウサギ488(A11008)、ヤギ抗ニワトリ488(A11039)、ヤギ抗マウス350(A11045)、全て1:1000)中でインキュベートした。細胞または組織をPBS/0.1%トリトンX中でさらに3回洗浄した後、Fluoromount(Southern Biotech,Birmingham,AL)を使ってマウントした。
マウス脳を4%PFAで潅流し、ビブラトーム(Leica VT 100S)で50μmの切片を作成した。切片を、20mMトリス緩衝液中の4%BSAおよび0.15%サポニン(pH7.4)により室温で、2時間、ブロッキングした。その後、抗GFP(1:1000)(Aves Lab Inc.)と共に一晩4℃でインキュベートし、続けて、ビオチン化抗ニワトリ(1:1000、Vector Laboratories,Inc.)を、GoldEnhance(#2114、Nanoprobes)で処理したNanogoldストレプトアビジン(1:100、Nanoprobes,Yaphank,NY)と共にインキュベートした。一次抗体インキュベーションを除いた以外は、陰性対照を同一手順を用いて行った。組織切片をカコジル酸ナトリウム緩衝液中の2%グルタルアルデヒドでの固定、15分間の軽いオスミウム酸染色(0.5%四酸化オスミウム)および1%酢酸ウラニウムによる30分のブロック染色を行った。その後、組織をエタノール系を通して脱水し、続けて、Eponate12(Ted Pella Inc.)と共にインキュベートした。試料をレジンに埋め込み、60℃で48時間重合させた。極薄(70nm)の切片を切り出し、Rockefeller University電子顕微鏡センターのJEOL JEX100CX透過型電子顕微鏡下で検査した。
細胞培養
室温下、−60mVで、αGFP−TRPV1/GFP−フェリチンまたはαGFP−TRPV1変異体/GFP−フェリチンコンストラクトを発現しているHEK細胞およびN38細胞からホールセル電圧クランプ記録を作成した。落射蛍光を使って、Hamamtsu CCDカメラを備えた直立型Zeiss Axioskop 2FS Plus顕微鏡でGFPを発現しているニューロンを可視化した。外液には、次を含めた(mM単位):140NaCl、2.8KCl、2CaCl2、1MgCl2、1HEPES、10グルコース、pH7.4。ボロシリケートガラスから引いたパッチピペット(World Precision Instruments)は、チップ抵抗5〜10オームで、135グルコン酸カリウム、4KCl、0.05EGTA、10Hepes、4MgATP、10ホスホクレアチンナトリウム(mM単位)(pHはKOHで7.3に調節、特に指示がない限り、浸透圧は、290OSMである)を含むグルコン酸カリウム内部溶液を充填した。浸透圧が290OSM以外の場合は、次のCsCl内部溶液を使用した(mM単位):125CsCl、10HEPES、4MgATP、0.5CaCl2。2APB(200μM)を10mMのDMSOストックから調製し、記載がある場合は、浴を通して潅流した。200μMの2APBの存在下、5mVステップによる電圧増加に対する電流応答を測定することにより、I−V関係を得た。細胞を−60mVに保持した。ミクロマニピュレーターを使って、記録対象細胞の500ミクロン以内に永久磁石を5秒間配置することにより磁気活性化を適用した。記録をAxopatch 200B増幅器で取得し、2kHzに濾波し、10kHzでデジタル化する(pClamp software,Molecular Devices)。IGOR Pro(Wavemetrics)およびNeuroMatic.(neuromatic.thinkrandom.com)を使って、データを解析した。直列抵抗をモニターしたが、補償はしなかった。直列抵抗に20%を超える変化があった場合は、記録から除外した。
VMHにAd−αGFP−TRPV1/GFP−フェリチンまたはAd−αGFP7 TRPV1変異体/GFP−フェリチンを注入したグルコキナーゼ−cre Rosa−TdTomatoマウスに、イソフルランで深く麻酔をかけた後に、断頭し、全体脳を取り出して、85NaCl、2.5KCl、0.5CaCl2、4MgCl2、25NaHCO3、1.25NaH2PO4、64スクロース、25グルコースおよび0.02D−2−アミノ−5−ホスホノペンタン酸(D−AP5,Tocris Bioscience)(mM単位)を含む氷冷「切断」溶液中に直ちに浸漬した。これを95%O2、5%CO2でバブリングした(pH7.4)。ムービングブレードミクロトーム(VT1000S,Leica)を使って、冠状視床下部切片(200μm)を作製した。125NaCl、2.5KCl、1.25NaH2PO4、26NaHCO3、10グルコース、2CaCl2および1MgCl2(pH7.4)(mM単位)を含む記録溶液中で、95%O2、5%CO2でバブリングし、切片を32℃で40分保持した。αGFP−TRPV1/GFP−フェリチンまたはαGFP−TRPV1変異体/GFP−フェリチンコンストラクトの発現を示すtd−tomatoおよびGFPの両方を発現しているVMH中のニューロンからのホールセル電流クランプモードのパッチクランプ記録を室温で行った。ニューロンを可視化し、上述のように記録した。ニューロンの活性化を観察するために、ニューロンを閾値未満に過分極させた。
C57Bl6マウス(8〜9週齡、Jackson laboratories,Bar Harbor,MA)、Nestin cre(8〜9週齡、Jackson labs)およびグルコキナーゼcre(8〜16週齡)を使用し、制御された照明条件下(12時間の明/12時間の暗)、および温度(22℃)で、単一ケージに収容し、標準的マウス固形飼料を自由に摂取させた。動物飼育および実験手順は、既定ガイドラインに基づいて、Rockefeller Universityの実験動物委員会の承認を得て実施した(プロトコル12561および14712)。いずれの場合も、体重に従ってマウスを無作為化した。処理群に対する評価者盲検方式とはしなかった。
Breeze2血糖計(Bayer;Leverkusen,Germany)を使って、血糖を測定した。血液を10分間遠心分離し、血漿を収集した。インスリンの血漿中濃度(Mercodia,Winston Salem,NC)およびグルカゴン濃度(Mercodia)をELISAにより測定した。
RIPA中に溶解することによりタンパク質を単離し、4℃で16000rpm、5分間遠心分離した後、4xLaemeli緩衝液を添加した。試料を95℃で5分間変性し、アッセイまで−20℃で凍結した。試料(15ul)を4〜15%ゲルで泳動させた後、PVdF膜に転写した。膜を室温で1時間ブロッキングし(TBST緩衝液中の3%ドライミルク)、その後、TBST中の一次抗体(ホスホCREB(Ser133)(87G3)ウサギmAb(1:1000)またはベータアクチンウサギAb(1:1000)、Cell Signaling)中で一晩4℃でインキュベートした。膜をTBST中で3回洗浄後、ブロック中の二次抗体(ヤギ抗ウサギIgG−HRP、1:5000、Santa Cruz)中で、室温で2時間インキュベートした。膜をさらに5回洗浄した後、基質(Supersignal West Femto最大感度基質、Life Technologies)で5分間現像し、イメージングした(C−DiGit blot scanner,Licor)。pCREB密度シグナルを、ハウスキーピング遺伝子の密度シグナルで除算することにより、生じ得るタンパク質添加量の変動に関し補正した。

調査の前に決めたように、2SDを超えて平均の外側にあるデータは、さらなる分析から除外した。全データをガウス分布および分散に対し試験した。別段の指示がない限り、対応のない両側スチューデントt検定を使って、正規分布および類似の分散のデータを統計的有意性について分析した。正規変動および不等分散を有するデータは両側ウェルチのt検定により解析した。対応のあるデータは対応のあるt検定により解析した。3つ以上の群のあるデータは、パラメトリックデータに対して、1元配置分散分析とテューキーの事後解析により解析した。正規分布ではないデータは、事後的ダンの調整を使って、両側マンホイットニー検定またはクラスカル・ワリス検定により解析した。P値は、示した通りである。経時変化データは、2元配置分散分析とシダックの多重比較検定により、または対応のあるデータに対する反復測定2元配置分散分析とシダックの多重比較検定により解析した。別に定める場合を除き、データは平均±SEMをして示す。
我々は、神経活動のインビボでの非侵襲的、時間的制御のための系およびグルコース恒常性の中枢神経系(CNS)制御を調査するためのその使用について記載する。ニューロン活性化は、ラクダ抗GFP抗体により陽イオン伝導性一過性受容器電位バニロイド1(αGFP−TRPV1)に係留されたGFP−フェリチン融合タンパク質(GFP−フェリチン)の標的発現を介して、電波(RF)または磁界を使った遠隔操作により達成された。同じ刺激を介した神経性抑制は、TRPV1ポアを変異させて、チャネルをクロライド透過性にすることにより達成された。この系を使った視床下部腹内側核(VMH)中のグルコース感知ニューロンの活性化は、血漿中ブドウ糖およびグルカゴンを増やし、インスリンおよび刺激を受けた摂食を減らし、一方、これらのニューロンを抑制すると、血糖が低下し、血漿中インスリンが増え、摂食が抑えられた。これらの結果は、VMH中のグルコース感知ニューロンが、インスリンおよびグルカゴン分泌に対する効果を介して、グルコース恒常性を制御することができ、これは膵臓ホルモンが、血糖および行動を制御するCNS回路のエフェクター機構として機能することを示唆していることを示す。我々が採用した方法はまた、永久的移植物を不要にし、その他の神経プロセスを研究するのに適用される可能性があり、または他の細胞、分散細胞型を調節するためでさえ使用されるであろう。

(a)組み換え細胞集団を用意するステップであって、前記細胞が、第1のポリペプチドに融合した金属結合タンパク質をコードするヌクレオチド配列と第2のポリペプチドに融合した機械感受性イオンチャネルをコードするヌクレオチド配列とを含む遺伝的コンストラクトを含み、前記第1のポリペプチドが前記第2のポリペプチドの結合相手であり、前記イオンチャネルの活性化は前記タンパク質、ペプチドまたは核酸の産生を誘導する、ステップ、
(b)前記細胞を静磁界に曝し、それにより、前記イオンチャネルを活性化して前記標的遺伝子の発現を誘導し、それにより、前記タンパク質、ペプチドまたは核酸を産生するステップ、および
(c)前記タンパク質、ペプチドまたは核酸を単離するステップ、

前記第1のポリペプチドは、前記第2のポリペプチドの結合相手であり、
前記イオンチャネルの活性化は、前記タンパク質、ペプチドまたは核酸の産生を誘導し、
前記方法は、
(a)前記組み換え細胞を前記対象に投与するステップと、

前記タンパク質、ペプチドまたは核酸は、前記チャネルの活性化時に活性化される遺伝子によってコードされ、
前記方法は、
(a)前記ベクターを前記対象に投与するステップと、
