本発明の他の側面によれば、上述の接着細胞へのナノ粒子取り込み方法を使用して接着細胞の細胞質ゾル中へ金属ナノ粒子を取り込ませ、前記細胞質ゾル内の蛍光タンパク質を前記取り込まれた金ナノ粒子の表面に結合させる、細胞質内にプラズモンアンテナを作製する方法が与えられる。
ここで、前記金属ナノ粒子は金ナノ粒子であってよい。
本発明の更に他の側面によれば、金属ナノ粒子が取り込まれている、光音響イメージング、X線透視撮影またはX線CTによって識別可能な接着細胞が与えられる。
本発明の更に他の側面によれば、上述の光音響イメージング、X線透視撮影またはX線CTによって識別可能な接着細胞を生体内に導入することによる、光音響、X線透視またはX線CTによるイメージング方法が与えられる。
金ナノ粒子表面に、プラスミドDNAやsiRNA等の核酸、システイン残基(アミノ酸)を含むタンパク質を結合した複合粒子を合成する。または、脂質とヘキサデカンチオールとの混合溶液により金ナノ粒子表面上に脂質二重膜を形成することで被覆を行い、その被覆界面へ脂質結合性分子等を利用してタンパク等の生体分子を導入する。このようにして生体分子を導入した複合粒子を、低張浸透圧下で細胞へ作用させることで細胞質内へ直接・迅速・効率的に分子を導入できる。またリソソーム(低pH、多様な分解酵素を含む)を経由することを回避することで細胞質ゾル中へ導入されるタンパク質の変性や分解を防止することができる。
例えば、蛍光タンパク質を細胞内へ導入することによって、細胞内の機能のための測定を行うことがある。この際に、細胞質内へ金ナノ粒子を導入し、その表面へ蛍光タンパク質を結合させることにより、細胞質内にプラズモン・アンテナを構築することで、細胞からの蛍光シグナルを増強させて検出感度を上げることができる。
金属粒子、特に多量の電子を持つ金粒子を濃縮した細胞は、たとえば、光音響イメージング、X線透視撮影またはX線CTによって周囲と区別できる可能性がある。小動物(マウス等)から取り出した後に分離・分化させた細胞、またはヒトなど他動物由来の細胞(癌細胞株など)に対して、本特許の方法で特定の細胞に金ナノ粒子を大量に導入し生きている動物内に移植する。これにより、X線CTによるイメージングを行うことによって、個体内における細胞レベルでの動態観察が可能となる。本発明の方法を使用すれば大量の金ナノ粒子を接着細胞内へ取り込ませることができるため、X線CTによって識別可能な金ナノ粒子入り細胞が得られる。
先ず、本発明ではない、すなわち低浸透圧環境を使用しない従来の方法によってGNPがどのように細胞内へ取り込まれるかを、蛍光顕微鏡を使用して調べた。GNPの蛍光標識付けを行うため、大腸菌内に組み換え型mCherry蛍光タンパク質を発現させて、それを精製した。野生型mCherryタンパク質は内部システイン残基を有していないため、ペプチドのN−終端にシステインを遺伝子工学的に導入することで、タンパク質を硫黄−金結合を介してGNPに結合させた。細胞内に取り込まれたナノ粒子を撮影するため、HeLa細胞を5μg/mLのmCherry結合GNPとともに培養基中で2時間培養した。洗浄(washout)後、細胞中に存在する直径が5nm及び100nmの両方の粒子を含む個々の蛍光GNPを、図1A及び図1Bに示すように撮影することができた。この結果、上記標識付けは有効であり、また金表面によるmCherry蛍光の消光は検出のための重大な問題でないことが確認された。二色撮影により、ほとんどのGNPがリソソームマーカーであるLamp−1 GFPと同じ位置にあることがわかった。なお、添付図面は白黒化されているが、これらの白黒写真上では、図1A及び図1Bの下段右端にあるマージされた写真上で、GNPの蛍光とLamp−1 GFP(図中ではLamp1と表記)の蛍光とが重なっている場所では蛍光の輝度が加算されて輝度が高められていることから、上記事項が確認できる。一方、写真は示さないが、ほとんどのGNPは初期エンドソームマーカーであるRab5−GFPとは同じ位置にないことも分かった。従って、広いサイズ範囲のGNPが細胞中に取り込まれたが、予想通り、これらはエンドサイトーシスを経てリソソーム内に輸送された。
生細胞の細胞質中にナノ粒子を導入するため、2つの異なる方法を使用した。第1の方法として、培養の際のナノ粒子濃度を高くした。第2の方法では、培養中に浸透圧を細胞に印加した。これらの処理により、リソソーム経路を飽和させて、過剰の粒子が細胞質ゾル中に漏出することを期待した。更に、細胞膜に印加される浸透圧力により一部の粒子が細胞膜を透過するかもしれない。これらの実験では、修飾されていない100nmのGNPを使用することで、表面修飾により起こるかもしれないアーティファクトを回避した。これらの修飾されていないGNPは反射フィルターを使用することで撮影できた。それは、これらの粒子は背景反射よりもはるかに強く光を拡散するからである(詳細は後述)。
フローサイトメトリーに基づく測定を行うことによって、個々の細胞の細胞質ゾル中へのGNPの取込みを統計的に解析した。従来の研究で、細胞からの後方散乱を細胞中の金属ナノ粒子の取込みの測定に使用できることが示されている。細胞中のGNPがフローサイトメトリーによって検出できるかどうかを調べるため、先ず、mCherryで蛍光標識付けされたGNPが取り込まれている細胞と修飾されていないGNPが取り込まれている細胞の夫々からの蛍光信号及び後方散乱(BSC)信号とを比較した。5μg/mLの濃度のGNPとともに2時間培養したHeLa細胞をフローサイトメーターを使って測定した。期待されたように、修飾されたGNPとともに培養された場合とmCherryで標識付けられたGNPとともに培養された場合の細胞集団は何れもGNPなしの対照細胞と比較して平均的な後方散乱信号が増大するが、mCherry−GNPとともに培養された細胞集団だけが大きな赤色蛍光を示すことを見出した。これらの測定結果を図4Aに示す。これに加えて、図4Aのドットプロット上で個々の細胞からのシグナルを個別に検討した所、増大した赤色蛍光信号を示す個々の細胞から、より大きな後方散乱信号が観察されたが、このことはこれらの細胞がmCherry−GNPを含むことも示している。これについても図4Aを参照されたい。これらの結果から、平均のBSC信号を測定することによってGNPの取込みを統計的に測定できることが示される。図4B及び図4Cに示すように、HeLa細胞とHEK293細胞の両者へのGNPの取込みは、同様に検出可能である。しかし、mCherry−GNPを含む細胞のBSC信号は、何れのタイプの細胞についてもその中に修飾されていないGNPを有する細胞のBSC信号よりもわずかに小さかった。mCherryによる修飾は一部のGNPの凝集を引き起こし、大きすぎて細胞内に入ることができない過大な集塊を生成したのかもしれない。
これまでに説明した圧力誘起取込み機構の理解に基づいて考えれば、このプロセスは材料固有のものではなく、他の材料でも浸透圧によって細胞質ゾル中へ取り込まれる。実際図5Aに示すように、銀ナノ粒子(直径30nm)、酸化チタンナノ粒子(直径40nm)及びメソポーラスシリカナノ粒子(直径200nm、細孔サイズ4nm)について、低浸透圧下での取込みの増大が観察された。これらのナノ粒子は、一定濃度33pMで細胞とともに培養した。なお、この濃度は直径100nmのGNPについての20μg/mLの濃度と等価である。従って、浸透圧誘起取込みは、材料の性質及び粒子サイズとは独立に起こったものである。これらの結果から、ナノ粒子と細胞膜との表面相互作用は低浸透圧条件下での取込みについては非常に重要というわけではないかもしれない。
細胞質ゾル中のGNPを使用して細胞の蛍光信号を変調することを検討した。これは診断及びイメージング用途に有用となる可能性がある。図2に示されるGNPと蛍光タンパク質との間の相互作用を調べることで、これらの用途の開発を試みた。mCherryタンパク質(野生型及びC−mCherry−C)に加えて、緑及び近赤外線(NIR)蛍光タンパク質であるAcGFP及びiRFPも発現させて、GNPがどのようにこれらのたんぱく質の蛍光を細胞内で変調することができるかを調べた。先にC−mCherry−Cについて行ったように、複数のシステイン残基をAcGFP及びiRFPに付加した(以下、C−GFP−C及びC−iRFP−Cと称する)。ただし、ここで野生型AcGFP及びiRFPは内部にシステイン残基を含む。これにより、図6A中のC−GFP−C、C−mCherry−C及びC−iRFP−Cのグラフに示すように、改変されたタンパク質(C−GFP−C、C−mCherry−C及びC−iRFP−C)を発現した細胞の蛍光信号は取り込まれた細胞質ゾル内GNPによって増強されることがわかった。これと対照的に、図6A中のmCherry(WT)のグラフからは、野生型mCherryを発現した細胞についてはこのような増強は観察されなかった。先に、C−mCherry−Cとは対照的に、図2Aにあるように、野生型のmCherryはGNPに結合しないことを示した。従って、これらの結果から、GNPと蛍光タンパク質との相互作用は蛍光を増強するように機能したことが証明される。
実施例中で使用した100nmのGNPの水溶液中での吸収スペクトルは550〜570nmに最大値を有していた。GNPに見られる強い光吸収はLSPRによるものであり、単一のGNPのLSPRによって生成された電磁(EM)場の強度はこれらの波長の周囲で最も大きくなる。プラズモン増強蛍光はまた、励起光の波長がこれらの波長に近いとき最も大きくなることが知られる。しかしながら、上の表に示すように、iRFPについてのプラズモン増強(690nmで励起)の方がmCherryについてのプラズモン増強(587nmで励起)よりも大きいことが観察された。この食い違いは細胞質ゾル中でのGNPのLSPRの赤方偏移で説明できるだろう。つまり、赤方偏移したLSPRがiRFP励起スペクトルともっと良くオーバーラップした可能性があるということである。LSPRのこのような赤方偏移は、タンパク質の結合あるいはGNPのクラスタ形成等のGNPの表面状態の変化によって引き起こすことができる。GNP表面へのタンパク質の結合は屈折率を変化させ、その結果、LSPR波長を約10nm以下だけ赤方偏移させる。これとは対照的に、GNPクラスタは単一粒子の場合に比べて強い電磁場をサポートし、クラスタのLSPRはもっと大きな規模で赤方偏移及び帯域の拡大をもたらす傾向がある。iRFPの場合の励起波長は単一のGNPのLSPR波長よりも100nm程度長いので、GNPのクラスタ形成の方が、単粒子へのタンパク結合と比較すると、上記観察結果をよりよく説明できる可能性がある。
低浸透圧によって細胞質ゾル中へのナノ粒子の取込みが引き起こされることを示した。また、本願発明者の解析により、これらの取り込まれたナノ粒子の大部分は圧力のかかった細胞膜を通して直接浸透したものであることが示唆される。浸透圧ストレスの下でのこのような浸透がどのようにして起こり得るかは、物理化学の基本的なテーマである。しかしながら、この機構を完全に解明するためにはさらに研究を進めることが依然として必要とされる。というのも、これは脂質膜の生物物理学の単純な問題ではないからである。脂質だけの小胞では膜構造は浸透圧によって劇的に変化し、あるいは破裂する。しかし、細胞膜の構造は低浸透圧であっても維持されることが観察された。このような安定性・堅牢性は、物理的にはアクチンフィラメントのような細胞内の物理的構造によって、また、化学的には細胞膜の損傷を治癒するためのイオンチャネル及び輸送体による制御された分子輸送や脂質の輸送によって成り立っている。更には、より生理学的な視点からの浸透圧の研究も重要である。例えば、最近の研究によれば、膜の浸透圧膨張は動物での細胞内シグナル伝達で積極的な役割さえも演じている可能性があることが示唆されている。
<化学薬品及び遺伝子素材>
金属ナノ粒子及び無機ナノ粒子は、金ナノ粒子については田中貴金属(東京)から、銀ナノ粒子はグリーンケム株式会社(堺)から、酸化チタンは石原産業(大阪)から、またメソポーラスシリカはSigma Aldrich(米国ミズーリ州)から入手した。これらナノ粒子の特性も生業業者から提供されたものである。ブレフェルジンA及びコルヒチンはSigma Aldrichから入手した。カルセインAM、DiBAC4(3)は同人化学研究所(熊本)から入手した。Alexa555で標識が付けられたファリオジンはThermo Fischer(米国マサチューセッツ州)から入手した。BSA及びTriton X−100は和光純薬工業(大阪)から入手した。野生種のLamp1及びRab5の遺伝子はDr. Walther Mothes及びDr. Ari Heleniusから入手したベクターから、pAcGFP−N1/C1ベクター(タカラバイオ(滋賀))へサブクローニングした。C−AcGFP−C及びC−mCherry−C遺伝子は3つの追加のシステイン残基を含む配列を野生型AcGFP、mCherry遺伝子のN−末端及びC−末端の両方に導入することで構築した。既にシステイン残基をN−、C−両末端の近傍に有するiRFPへは2つのシステイン残基を含む配列を両末端に付加することでC−iRFP−C遺伝子を構築した(iRFP遺伝子はDr. Vladislav Verhushaから入手)。これらの遺伝子及び野生型mCherryはpAcGFP−N1ベクター内のAcGFP遺伝子と組み替えてクローニングした。
可溶性のcys−mCherryタンパク質を発現し精製するため、N−末端での一つのシステインとmCherryとの融合遺伝子をpGex6p1ベクター(GE Healthcare)中でクローニングし、IPTG誘導を使ってBL21−DE3大腸菌中にcys−mCherryタンパク質を発現させた。可溶性のGFPタンパク質を発現し生成するため、AcGFPをpET30aベクター(EMD millipore)中でクローニングし、IPTG誘導を使ってBL21−DE3大腸菌中にAcGFPタンパク質を発現させた。タンパク質(mCherry及びGFP)の精製は、mCherryについては、グルタチオン−セファロース4B(GE)を使用し、またそれに続いてPreScissionプロテアーゼ(GE)による消化(digestion)を行った。GFPの方はNi−NTA His−Band Resin(EMD millipore)を使用した。両者ともその後にゲル濾過精製を行った。タンパク質濃度は分光光度計を使用して測定した。GNPへの標識付けは、精製したcys−mCherryと0.1mMのTHCPの存在の下でpH=7で混合して2時間以上振盪することで行った。
血清入りDMEM培地中で継代維持されているHeLa細胞、HEK293細胞及びL929細胞を、イメージング実験の24時間前にLipofectamine 2000(Thermo Fischer)を使用して遺伝子導入を行った。蛍光顕微鏡、反射顕微鏡及び暗視野顕微鏡観察については細胞をガラスボトムディッシュ(松浪硝子工業(大阪))上で培養したが、他の試験では通常の細胞培養皿及びマイクロプレート上で培養した。細胞増殖試験では、対照細胞及び各種の浸透圧下でGNPとともに培養する細胞はGNPとともに2時間培養した後のいくつかの時点で試験に付された。Cell Counting Kit−8(同仁化学研究所)を使用し、460nmにおけるWST−ホルマザンの吸収を、マイクロプレートリーダー(日立ハイテクノロジーズ(東京))を使用して測定した。
細胞への2時間の低浸透圧印加のため、培地の2/3を水及び/またはGNPあるいは他のナノ粒子を含有する緩衝液の混合液で置換した。タンパク質濃度一定での各種のオスモル濃度を比較するため、培地(血漿入りのDMEM)の1/3の体積を残して、最終的に300mOsm/L溶液とするものについては残りを2/3体積のHEPES緩衝生理食塩水(300mOsm/Lに調整したHBS)で、最終的に200mOsm/L溶液とするものについては1/3体積のHBSと1/3体積の水との混合物で、また最終的に100mOsm/L溶液とするものについては2/3体積の水で置換した。溶液のpHは実験の間中ほぼ7に保持した。たんぱく質(GFP)及び蛍光低分子(セルセイン−AM及びDiBAC4(3))の取り込み実験においては、細胞を低浸透圧下に2時間置いた後に、低浸透圧状態を維持したままで各分子を添加することで行った。エンドサイトーシス阻害剤添加の実験においては、ナノ粒子を添加する1時間前に各阻害剤(5μMのBFA及び10μMのコルヒチン)を細胞に添加して、GNP取り込みの間、阻害剤が含まれたままとした。過剰のナノ粒子及び分子を洗い流してから、細胞の像を撮影し、あるいはサイトメトリー分析のためにEDTA及び/またはトリプシンで細胞を剥離してPBS中に分散させた。分子送達のために、PBS中で1mg/mLのメソポーラスシリカをAlexa555で標識された6μMのファロイジンによりローディングした。遠心分離による洗浄後、収集されたナノ粒子を33pMの濃度で細胞に添加した。
Cascade II EMCCDカメラ(米国アリゾナ州TusconのRoper)を備えたLeica AF6000LX顕微鏡及び100×の油浸レンズを使用して蛍光イメージングを行った。非蛍光性のGNPの散乱光は、535nmの入射光及び反射光を通すフィルターキューブを使用して検出した。DS−Ri1ディジタルカメラ及び40×水浸レンズを備えたNikon ECLISPE 80i顕微鏡を使用して暗視野顕微鏡撮影を行った。散乱スペクトルは、光ファイバーを介して顕微鏡に結合された小型グレーティング分光器(Ocean Optics UBS4000、スペクトル分解能1.55nm)を使用して測定した。収集容積(collection volume)は約1μm2であり、取得時間は1秒であった。像中のGNPスポットはImageJ(NIH)を使用し、バックグラウンドシグナル減算及び閾値処理を全ての像に同じ態様で適用することで検出を行った。RGBの数値化のために、同じ条件(露出時間等)で撮影された元画像を比較した。細胞ソーターSH800(ソニー)を使用してフローサイトメトリー解析を行い、またレーザー線(488nm、561nm及び638nm)及び放射フィルターセット(緑:500nm〜550nm、赤:570nm〜630nm、遠赤色:650nm〜680nm、近赤外:755nm〜815nm)を使用して蛍光及び散乱検出を行った。
Claims (11)
- 基板に固定された接着性細胞、又は、基板上に沈殿し付着した浮遊系細胞である接着細胞を、ナノ粒子が分散しており、浸透圧が生理学的な環境における浸透圧よりも低い水溶液に生体内での温度領域で浸漬する、接着細胞へのナノ粒子取り込み方法。
- 前記ナノ粒子の直径は30nm〜1000nmである、請求項1に記載の接着細胞へのナノ粒子取り込み方法。
- 前記水溶液の浸透圧モル濃度が250mOsm/L以下である、請求項1又は2に記載の接着細胞へのナノ粒子取り込み方法。
- 前記ナノ粒子は第1の物質からなる担持体粒子と、前記第1の物質とは異なる第2の物質との複合粒子である、請求項1から3の何れかに記載の接着細胞へのナノ粒子取り込み方法。
