Claims (16)
- ヒト又は動物の組織をインビトロで培養又は増殖する方法であって、
ヒト又は動物の組織から得られた試料の組織を小型化すること;
小型化した組織を2つの足場の間に置くことによってアセンブリがサンドイッチアセンブリになるように、アセンブリを生成すること;
3D灌流システムの培養チャンバの内部にアセンブリを配置すること;及び
アセンブリを3D灌流システム内で所定の時間、直接灌流することであって、小型化した組織はアセンブリを通じて培地で灌流される、直接灌流すること
を含む方法。 - 組織を小型化することが、組織を機械的に前処理することを含む、請求項1に記載の方法。
- 組織を小型化することが、組織を酵素的に前処理することを含む、請求項1又は2に記載の方法。
- アセンブリを生成する前に組織を清浄化することをさらに含む、請求項1から3のいずれか一項に記載の方法。
- 組織を清浄化することが、組織を洗浄することを含む、請求項4に記載の方法。
- 組織を清浄化することが、防腐処理を含む、請求項4又は5に記載の方法。
- アセンブリを生成することが、足場又はヒドロゲルの一方の側に格子を配置し、足場又はヒドロゲルの他方の側にさらなる格子を配置することを含む、請求項1から6のいずれか一項に記載の方法。
- 3D灌流システムの内部でアセンブリを灌流することが、交互の方向で適用される、請求項1から7のいずれか一項に記載の方法。
- 足場が細胞外マトリックスタンパク質でコーティングされる、請求項1から8のいずれか一項に記載の方法。
- 患者の腫瘍又はがんから得られた組織において、腫瘍又はがんの治療有効性をインビトロで試験する方法であって、
請求項1から9のいずれか一項に記載の方法で組織を培養すること(1);
組織に処理を施すこと(2);
組織をモニタリングすること(3);及び
モニタリングされた組織を評価すること(4)
を含む方法。 - 請求項1から10のいずれか一項に記載の方法で患者の腫瘍又はがんから得られたさらなる組織を培養すること(1’);
さらなる組織に第2の処理を施すこと(2’);
さらなる組織をモニタリングすること(3);及び
モニタリングされた組織及びさらなる組織を評価することによって第1の処理又は第2の処理を選択すること(4)
をさらに含む、請求項10に記載の方法。 - 第1の処理が組織への第1の物質の提供を含み、第2の処理がさらなる組織への第2の物質の提供を含む、請求項11に記載の方法。
- 第1の処理が第1の投与レジメンを含み、第2の処理が第2の投与レジメンを含む、請求項11又は12に記載の方法。
- 請求項1から9のいずれか一項に記載の方法で腫瘍組織を所定の時間、培養することを含む、ヒト又は動物の腫瘍組織に由来する細胞型の細胞を富化する方法。
- 腫瘍組織から前記細胞型の細胞を単離することをさらに含む、請求項14に記載の方法。
- 複数の灌流媒体を前記細胞型の細胞の増殖能に関して比較すること;
それらの能力を考慮して複数の灌流媒体から一の灌流媒体を特定すること;及び
3D灌流システムにおいてサンドイッチアセンブリを灌流するための好ましい灌流媒体を選択すること
を含む、請求項14又は15に記載の方法。
