上に簡潔に要約されるように、本発明は、表面に新規なキメラ融合タンパク質を安定的に発現する細胞株を含む、ユニバーサル抗体媒介バイオセンサーに関する。融合タンパク質はそれらの抗原結合特異性に関わらずに抗体を結合することができ、表面に融合タンパク質を発現する細胞は、同族の抗原によって結合される抗体の架橋により活性化され得る。融合タンパク質が任意の抗体のFc領域に結合することから、それらは細胞外シグナル伝達と細胞内活性化との間の普遍的な経路としてはたらくことができる。このバイオセンサーを使用して、試料と(i)バイオセンサー細胞、及び(ii)抗原に対する結合特異性を有する抗体とを接触させることによって、試料中の選択された抗原の存在を検出することができる。一旦添加されれば、抗体は、融合タンパク質の抗体結合ドメインによる抗体のFc領域の結合を介して、キメラ融合タンパク質によって結合される。抗体による抗原の認識及び結合は抗体架橋をもたらし、それが融合タンパク質によるシグナルとしてバイオセンサー細胞中へと広がり、該細胞にて融合タンパク質の細胞内シグナル伝達ドメインが細胞活性化を引き起こす。その後、かかる活性化をアッセイし、所望に応じて、定量することができる。細胞活性化のレベルに基づき、試料中の抗原の存在に関して結論を引き出すことができる。非常におおまかに言えば、本発明のバイオセンサー細胞を使用して、細胞活性化が起こる場合、抗原が試料中に存在すると考えられる。
本発明のキメラ融合タンパク質は、それらのアミノ末端において、細胞外抗体結合ドメインを含む。例示的な抗体結合ドメインとして、限定されないが、FcγRI又はFcγRIII等のFcγ受容体(FcγR)の抗体結合ドメインが挙げられる。異なるFcγRサブタイプは、異なる抗体アイソタイプ(定常領域)に対して親和性が変化することから、本発明のバイオセンサーは、融合タンパク質中の抗体結合ドメインのアイデンティティーに基づき変化し得る。例えば、齧歯類FcγRI抗体結合ドメインは、齧歯類IgG2aだけでなく、ヒトIgG1、IgG3及びIgG4の免疫グロブリンの定常領域に対しても高親和性を有する。齧歯類FcγRIの抗体結合ドメインは、非常に高い親和性(108M−1超)で齧歯類IgG2aアイソタイプを結合する[2]。交差種結合研究は、IgG4よりもIgG1及びIgG3のより強い結合によって、ヒトFcγRIが商業的に入手可能なヒトmAbに結合することができることを実証した[3]。齧歯類FcγRIII抗体結合ドメインは、齧歯類IgG1、IgG2a、IgG2bの定常領域に対して、並びにヒトIgG1、IgG2、及びIgG4免疫グロブリンに対してより低い親和性(3×104M−1〜6×105M−1)を有する[3]が、本発明の融合タンパク質においても使用可能である。FcγRIとFcγRIIIとの間で、全てのマウス及びヒトのIg(齧歯類IgG3を除く)は、これらの2つのFc受容体のうちの1つに結合することができる。さらに、ポリクローナル抗体も、これらのFcγRに結合することができる[4]。
本発明のキメラ融合タンパク質は、それらのカルボキシ末端において、細胞内シグナル伝達ドメインを含む。好適なシグナル伝達ドメインは、他の状況において細胞の活性化を誘導することが知られているものを含む。例えば、B細胞は、生得的に、膜貫通型タンパク質CD79との複合体の形成によって抗原のB細胞受容体(BCR)結合を変換する。CD79は、B細胞の表面でヘテロダイマーを形成する2つの異なる鎖、すなわち免疫グロブリン−アルファ(Igα)及び免疫グロブリン−ベータ(Igβ)で構成される。Igα及びIgβは、細胞外ドメイン、単一の膜貫通ドメイン及び細胞質シグナル伝達ドメインを有する。Igα又はIgβの細胞内シグナル伝達領域のいずれかに融合されたCD8タンパク質の細胞外領域及び膜貫通領域を有する融合タンパク質は、シグナル伝達能力を有することが実証されている[5]。他の研究によって、プロテインキナーゼがCD8/Igα融合タンパク質のより強力な活性化因子であることが実証されている。同じ研究から、CD8架橋後にCD8/Igα融合タンパク質によるCa2+シグナル伝達を観察することができたことが更に実証された。これらの研究に基づき、一態様では、本発明の融合タンパク質は、Igαの細胞質シグナル伝達ドメインに融合された抗体結合ドメインを含む[6]。
抗体結合ドメインとシグナル伝達ドメインとの異なる組み合わせの使用により、特定の抗体に対する融合タンパク質の親和性を調整することができ、また細胞活性化のレベルを制御することができることは明らかであろう。本発明の範囲に含まれるキメラ融合タンパク質の具体例として、表1に提供されるものが挙げられる。6個の各融合タンパク質が図1に表される。
本発明は、本明細書において提供される融合タンパク質の各々をコードするヌクレオチド配列、また同様にそれらの相補鎖を含むポリヌクレオチドを含む。また、本発明は、ポリヌクレオチドを含むクローニングベクター、及びポリヌクレオチド又は発現ベクターのいずれかを含む宿主細胞を含む。かかる宿主細胞は、哺乳動物の細胞、又は限定されないがE.コリ(E. coli)及び昆虫細胞を含む非哺乳動物の細胞であってもよい。本発明は、ポリヌクレオチド及び発現ベクターによってコードされる融合タンパク質の発現を促進する条件下で宿主細胞を培養することと、細胞又は細胞培養物から融合タンパク質を回収することとを含む、本明細書に定義される融合タンパク質を産生する方法を更に含む。
齧歯類のFcγRI(配列番号15)及びFcγRIII(配列番号16)に対する配列は、クローニングされ、確認されている。抗体結合ドメイン、膜貫通ドメイン、及びシグナル伝達ドメインをコードする配列を発現ベクターへと操作することによって融合タンパク質の発現に対してキメラ融合タンパク質をコードする核酸コンストラクトを作製してもよい。例えば、FcγR受容体の抗体結合ドメイン及び膜貫通ドメインを、例えば、PCRを使用する「SOEing」によって、シグナル伝達ドメインをコードする配列とインフレームで融合してもよい[15]。FcR−γ鎖が必要な場合に(以下で検討される)コンストラクトを完成させるため、シグナル伝達ドメインのC末端及びFcR−γ鎖のN末端は、PCRによって2Aペプチドをコードする配列に付着される。FcγR−膜Igコンストラクトの構築のため、FcγR配列のC末端の制限酵素部位を、N末端において適合する制限酵素部位を含むIg定常領域に連結させるため使用してもよい。
本発明の融合タンパク質を発現するため使用され、したがって本発明のバイオセンサー細胞として役立つ可能性のある細胞株は、膜内在性タンパク質として細胞表面に該融合タンパク質を安定的に発現することができるという点、及びシグナル伝達ドメインの活性化を検出することができるという点でのみ制限される。好適な細胞株として、限定されないが、リンパ細胞及び非リンパ細胞が挙げられる。
CD8/Igα融合タンパク質を発現するリンパ細胞は、抗CD8抗体との架橋が、B細胞及びT細胞の両方における細胞内Ca2+の放出、及びIgαのリン酸化を刺激することを実証するため使用されてきた[5、6、10]。通常、内因性Igα/Igβ経路を使用してシグナル伝達を行う、マウス及びヒトのB細胞株は、本明細書に記載される融合タンパク質の発現に特に有用である。バイオセンサー細胞の産生に使用され得る好適なB細胞株として、限定されないが、Ramos細胞株、Raji細胞株、IIAI.6細胞株、及びC604細胞株が挙げられる。他の好適なB細胞株としてA20及びLK35.2が挙げられる。
HEK293細胞、CHO細胞、P815細胞、K562細胞及びCos−1細胞等の選択されたタンパク質の細胞表面発現に関するアッセイで一般に使用される、樹立されて十分に特性評価された多数の非リンパ細胞株が存在する。これらの細胞株は、これらの細胞において安定した発現を確立するのが容易であり、明確な成長特性を有することから、外来タンパク質を発現するために慣例的に使用される。しかしながら、非リンパ細胞は、Fcγ受容体発現細胞で発現される二次タンパク質であるFcRガンマ鎖(FcRγ鎖)を発現することができない。FcRγ鎖は、Fcγ受容体シグナル伝達に必要である[7]。非リンパ細胞は、FcR−γ鎖を発現しないが、かかる細胞はなおも融合タンパク質発現に対する優れた候補としてはたらき、FcR−γ鎖を共発現するように操作される場合、本発明のバイオセンサー細胞として使用することができる。
本明細書における考察から明らかなように、標的物質の検出において本発明のバイオセンサー細胞と共に使用され得る抗体のアイデンティティーは、(i)抗体が本発明の融合タンパク質によって結合され得る点、及び(ii)抗体が標的物質に結合可能である点においてのみ制限される。一旦特定の標的物質が検出のため選択されれば、該物質に対して結合特異性を有する抗体が商業的に入手可能かどうかを容易に判断することができる。入手可能でない場合は、必要とされる結合特異性を有する抗体を日常的な方法を使用して作製することができる。
標的物質の存在についてスクリーニングされ得る試料は、抗体による試料中に存在する標的物質の結合を可能とするという点においてのみ同様に制限される。好適な試料として、限定されないが、空気試料、液体試料、乾燥試料、野菜試料、及び生体試料が挙げられる。好適な空気試料として、限定されないが、エアロゾル、大気試料、人工呼吸器排出物、及びエンジン排気が挙げられる。好適な液体試料として、限定されないが、食品、飲料、水試料、医薬製剤、及びパーソナルケア製品が挙げられる。好適な乾燥試料として、限定されないが、食物、土壌、医薬製剤、可溶化スワブ試料、及びパーソナルケア製品が挙げられる。好適な野菜試料として、限定されないが、葉、果実、ナッツ、種子、花及び植物組織が挙げられる。好適な生体試料として、限定されないが、血液、血清、汗、尿、脳脊髄液、粘液、精液、便、気管支肺胞洗浄液、及び組織が挙げられる。
本発明のバイオセンサーを使用して、多様な異なる標的物質を検出することができる。当業者に明らかなように、標的物質に対する唯一の制限は、抗体による該物質の結合が可能でなくてはならないことである。標的物質は、限定されないが、生物兵器物質、アレルゲン、寄生虫抗原、真菌抗原、ウイルス抗原、細菌抗原、細胞性抗原、及び抗体等の生物学的な起源のものが挙げられる。例示的な生物兵器物質として、限定されないが、リシン、炭疽菌芽胞、ボツリヌス毒素、クロストリジウム・パーフリンジェンス(Clostridium perfringens:ウェルシュ菌)毒素、サキシトキシン、及びトリコテシン・マイコトキシンが挙げられる。例示的なアレルゲンとして、限定されないが、ツリーナッツ(tree nuts)、ピーナッツ及び動物のフケが挙げられる。例示的な細胞性抗原として、限定されないが、ヒト、霊長動物、又は限定されないが、家畜又はイヌ若しくはネコ等のコンパニオン動物等の他の哺乳動物等の被験体における疾患又は病状に関連する抗原が挙げられる。また、標的物質は、植物及び作物の物質、水生病原体、又は、病因物質、薬物及び他の化学物質、並びに限定されないが毒素及び汚染物質等の環境に見られる分子を含む。
本発明のバイオセンサー細胞を、試料中の標的物質を検出するため、場合によっては定量するためのアッセイにおいて使用することができる。上に記載されるように、抗体による該物質の結合により、またバイオセンサー細胞によって発現される融合タンパク質による抗体結合によって、細胞表面で架橋が生じ、融合タンパク質のシグナル伝達ドメインが細胞内の活性化シグナルとして結合を伝達する。例として、抗原を含む試料がバイオセンサーの細胞外表面の抗体と相互作用する場合、細胞内シグナル伝達カスケードが活性化される。
実施例1:融合タンパク質をコードするコンストラクトの産生及び発現
商業的に入手可能な齧歯類FcγRI及びIgαのcDNAを得た。2つの遺伝子の重複配列を提供するPCRプライマーを使用して2つの配列をつなぎ合わせて、配列分析によって確認されたフレーム融合のFcγRI/Igαを生じた。代替的には、FcγRIの抗体結合ドメイン及び/又は膜貫通ドメインを、受容体をコードするcDNAに由来するプライマーを用いて増幅し、またIgαの細胞内シグナル伝達ドメインを同様に増幅する。
植物病原体は、感染、並びに結果的な食物及び飼料の収穫高の損失が経済及び食糧安全保障に影響を与えることから、ウイルス性であるか細菌性であるかにかかわらず、非常に重要である。したがって、作物の管理及び輸入作物のモニタリングを支援するために植物病原体を明らかにすることと同様に、定期的に検出することができるアッセイを整備することが重要である。農場における国内作物の試験を記載する。
サルモネラ属の一種は、最も一般的な食品媒介病原体のうちの1つであり、幼い子供、老人及びその他免疫系が弱った者において重篤で、時に致命的なサルモネラ症疾患を引き起こす場合がある。サルモネラ汚染が感染した動物又はヒトの糞便との接触から生じることから、幅広い食品が、生産される卵及び肉から、また水さえからも汚染される可能性がある。現在のサルモネラ検出法として、時間がかかり、専門知識を必要とするPCR又は細菌培養が挙げられる。したがって、単純で迅速な検出アッセイが、大発生及び製品回収を予防するための食品の品質モニタリングに望ましい。鶏卵加工施設におけるサルモネラに対する試験を記載する。
リステリア(すなわち、L.モノサイトゲネス)は、およその20%の致死率を伴う重篤な細菌性疾患であるリステリア症の病因物質であり、妊婦、乳児及び免疫系の弱った者に最も危険な食品媒介病原体である。リステリアは生食肉製品(正:product)及び乳製品、並びに加工調理済み食品を汚染する場合がある。したがって、細菌を検出し、その存在をモニターする能力は、病原体の大発生及び製品回収を予防するために望ましい。すぐに食べられる食品(すなわち、デリ・ミート(deli meat)及びホットドッグ)を生産する食肉加工工場におけるリステリアの検出に対するユニバーサルバイオセンサー細胞の使用が記載される。
炭疽菌は、細菌バシラス・アンスラシス(Bacillus anthracis)の芽胞によって引き起こされる急性で致死性の疾患である。自然の土壌細菌は、放牧で育成された動物、また加工処理されていない動物の皮及び毛に由来する感染した肉との接触を通して伝染され得る。最近では、B.アンスラシスは生物兵器、及びテロ攻撃で使用するために武器化されてきた。この点において、B.アンスラシス芽胞の確実で迅速な検出は極めて重要である。被検試料は、疑わしい区域をふき取ることにより、又は溶液中での分析のためPBSに疑わしい粉末を直接懸濁することにより得られてもよい。代替的には、エアロゾル試料を疑わしい区域で収集し、また微粒子を表面に濃縮して、ユニバーサルバイオセンサー細胞に曝露してもよい。好適なエアロゾルサンプリング装置(BioFlash E)をPathSensors, Inc.から購入する。

Claims (20)
- 試料において標的物質を検出する方法であって、
(a)試料と、標的物質に対して結合特異性を有する抗体、及びバイオセンサー細胞とを接触させることと、
(b)細胞活性化について前記バイオセンサー細胞をアッセイすることと、
を含み、
前記バイオセンサー細胞が安定的にキメラ融合タンパク質を発現し、前記キメラ融合タンパク質がFcγ受容体(FcγR)抗体結合ドメインとシグナル伝達ドメインとを含み、
前記FcγR抗体結合ドメインが、配列番号1若しくは配列番号3に示されるFcγRI抗体結合ドメインである、又は配列番号1若しくは配列番号3の全長に対して少なくとも95%の配列同一性を有するその配列変異体であるか、
前記FcγR抗体結合ドメインが、配列番号2若しくは配列番号4に示されるFcγRIII抗体結合ドメインである、又は配列番号2若しくは配列番号4の全長に対して少なくとも95%の配列同一性を有するその配列変異体であり、
前記シグナル伝達ドメインが、配列番号5に示される免疫グロブリンアルファ(Igα)シグナル伝達ドメインである、又は配列番号5の全長に対して少なくとも95%の配列同一性を有するその配列変異体であるか、
