Description
このため、HBVとcccDNAを完全に除去するために、より効果的な新規HBV治療用のB型肝炎ワクチンの開発が期待される。肝移植後の免疫拒絶反応が低い点からも明なように、ヒトの肝臓は免疫寛容器官である。B型肝炎ウイルス(HBV)はヒトの肝臓を感染するため、HBVに対する免疫寛容が肝臓の主な特徴である。HBV免疫寛容逆転は慢性B型肝炎患者(CHB)の免疫治療ワクチンを開発するための立脚点となる。HBV免疫寛容は、肝臓の部分的なanti−HBV免疫応答によりウイルスを効果的に排除できず、持続感染を引き起こすことに反映されているだけでなく、HBVの持続存在により全身免疫系がHBVに対して応答しないことを誘導し、たとえば、HBV寛容期にある患者がHBsAgワクチンに対して応答しないことにも反映されている。これは、現在の治療性ワクチンがCHB患者に対して効かない要因でもある。肝臓により誘導された免疫寛容及びその逆転メカニズムについての研究は、B型肝炎を治療するワクチンを開発するために、理論的根拠を提供している。
2014年に、米国Thomas H.Kingによって、「A Whole Recombinant Yeast−Based Therapeutic Vaccine that is comprised of heat−inactivated, whole recombinant Saccharomyces cerevisiae yeast cells expressing disease− related antigens」が報道されており、不活性化全組換え出芽酵母細胞で疾患関連抗原を発現させることを基礎とする治療性ワクチンである。該研究は、細胞内組換え発現タンパク質を抗原とし、不活性化全出芽酵母細胞をアジュバントとする治療性ワクチンのプラットフォームを創成している。このプラットフォームでは、番号GS4774のB型肝炎治療性ワクチンがあり、それが発現するHBV抗原は融合タンパク質としてのx−s−core抗原である。この酵母ベクターは複数種の抗原が入るMHCクラスIとクラスII抗原提示経路を提供し、強力なCD4+とCD8+細胞反応を刺激して、且つ免疫学的マウスモデルにおける抗原寛容性を破壊できる。また、該酵母ベクターはインビボで中和されにくいため、繰り返して投与して、長期間の免疫圧力を付与し、慢性細胞内感染、たとえばHCVやHBVを理想的に排除することに好適である。
2003〜2005年に、米国の科学家によって、B型肝炎ウイルスによるチンパンジー感染についての一連の研究結果が報道されている。疾患制御メカニズムは肝細胞核HBVプールの共有結合閉環状DNA(cccDNA)である。IFN−γを発生させるHBV特異的CD8+T、すなわちCTL細胞は大量で肝内に入り、HBVを感染した肝細胞にターゲットするため、cccDNAの排除、肝細胞感染HBV逆転はともに肝臓CD8+T細胞により生産するIFN−γの取り込みなどに関連する。これら結果から分かるように、cccDNAの排除は、細胞免疫反応によって媒介される二段階の過程であり、ステップ1として、細胞を損傷せずに90%以上のcccDNA分子プールを減少させることで、HBV弛緩環状デオキシリボ核酸の前駆体を排除する。ステップ2として、この過程において破損されて感染された肝細胞を増加させて、免疫逆転をトリガーさせる。
2014年に、中国科技大学の曽筑天博士によって、水動力法でHBVを持続的にキャリアするマウスを構築してHBV慢性感染者の免疫寛容状態をシミュレートしたところ、IL−12前処理及びIL−12とHBsAg VLPワクチンとの同時注射との併用療法を見出し、IL−12によるワクチン療法と呼び、HBV全身的免疫寛容を効果的に逆転して、さらにHBVを排除できることが報道されている。HBVマウスにIL−12によるワクチン療法を施した後、リンパ節内の濾胞様ヘルパーT細胞(Tfh)及び胚中心B細胞(GC B)はいずれも著しく増幅し、これに応じて、脾臓細胞におけるHBsAg特異的IgG生成細胞も著しく増加し、大部分のマウスでは、治療後期に、血清に防御抗体anti−HBsが発生し、また、T細胞のインビトロでのHBsAg刺激に対する増殖能力も、IL−12とワクチンとの併用治療を受けた後に、著しく回復することが見られた。以上の結果から明らかなように、HBV寛容マウスは、IL−12併用治療過程において、外部からのHBsAgワクチンに対する応答が回復され、それによりHBVが誘導した全身性免疫寛容が逆転されることを提示する。
前記B型肝炎治療ワクチンにおいて、前記組換えハンセヌラ・ポリモルファが発現するHBsAgは、
VLQAGFFLL、PFVQWFVGL、FLLTRILTI、WYWGPSLYSI、SLNFLGGSPV、FLGGSPVCL、LYSIVSPF、LYSIVSPFI、PFIPLLPIF、LLLCLIFLL、LLCLIFLLV、LLDYQGMLPV、LVLLDYQGML、VLLDYQGML、WLSLLVPFV、LLVPFVQWFV、GLSPTVWLSA、SIVSPFIPLL、LLPIFFCLWVの合計19個のCTLエピトープを含有することを特徴とする。
前記B型肝炎治療ワクチンにおいて、組換えハンセヌラ・ポリモルファが発現するHBsAgはadwサブタイプであり、前記組換えハンセヌラ・ポリモルファが発現するHBsAgのDNA配列はSEQ ID NO:1に示され、組換えハンセヌラ・ポリモルファが発現するHBsAgのアミノ酸配列はSEQ ID NO:2に示されることを特徴とする。
前記B型肝炎治療ワクチンにおいて、前記組換えハンセヌラ・ポリモルファが発現するHBsAgは、ウイルス様粒子構造であり、HBsAgをハンセヌラ・ポリモルファ脂質に挿入してなり、前記HBsAgの14個のシステインのうち9〜12個はジスルフィド結合、好ましくは、11個はジスルフィド結合を形成することを特徴とする。
前記B型肝炎治療ワクチンにおいて、前記不活性化組換えハンセヌラ・ポリモルファ細胞の不活性化条件は、不活性化温度が52℃〜54℃であり、不活性化時間が1時間〜3時間であることを特徴とする。
前記B型肝炎治療ワクチンにおいて、前記組換えハンセヌラ・ポリモルファの宿主であるハンセヌラ・ポリモルファ細胞株はHU−11であり、保蔵番号がCGMCC No.1218であり、前記宿主であるハンセヌラ・ポリモルファのオロチジン−5−リン酸デカルボキシラーゼ遺伝子が破壊されたDNA配列がSEQ ID NO:3に示されることを特徴とする。
前記B型肝炎治療ワクチンにおいて、HBsAg原液又はアルミアジュバントHBsAgのうちの1種をさらに含むことを特徴とする。
前記B型肝炎治療ワクチンにおいて、前記B型肝炎治療ワクチンの剤型は、プレフィルド注射液剤、注射液剤又は凍結乾燥粉末注射剤から選ばれることを特徴とする。
本発明は、SEQ ID NO:1に示されるヌクレオチド配列を含み、好ましくは、ゲノムにSEQ ID NO:1に示されるヌクレオチド配列が組み込まれている組換えハンセヌラ・ポリモルファをさらに提供する。
前記組換えハンセヌラ・ポリモルファにおいて、前記組換えハンセヌラ・ポリモルファの宿主であるハンセヌラ・ポリモルファ細胞株はHU−11(CGMCC No.1218)であり、前記宿主ハンセヌラ・ポリモルファのオロチジン−5−リン酸デカルボキシラーゼ遺伝子(HURA3)が破壊されたDNA配列がSEQ ID NO:3に示されることを特徴とする。
遺伝子合成技術を用いて、ハンセヌラ・ポリモルファMOX(メタノールオキシダーゼ)プロモーター1.5kb、ハンセヌラ・ポリモルファMOX(メタノールオキシダーゼ)ターミネーター350bp、ハンセヌラ・ポリモルファの自律複製配列HARS 1.0kb、及び出芽酵母のウリジン遺伝子ScURA3 1.1kbの5つの遺伝子素子を緊密に連結した後、pBluescripIIプラスミドに挿入して、シャトルプラスミドpMPT−02として構築する。
相同配列によって媒介される相同組み込みによってオロチジン−5−リン酸デカルボキシラーゼ遺伝子(HURA3)が破壊された組換えハンセヌラ・ポリモルファ株HU−11(CGMCC No.1218)を構築する。該菌株は、中国国内及び国外で使用されている突然変異誘発により生じた従来の栄養要求性宿主菌株に比べて、遺伝的安定性が高く、復帰突然変異率が低いという特徴を有し、遺伝的形質転換及び組換え菌株のスクリーニングを容易に実施できるとともに、野生型菌株の生理学的および生化学的特徴を保持し、組換え菌株の培養や外因性タンパク質の効率的な発現に有利し、高い工業応用価値がある。ハンセヌラ・ポリモルファ遺伝子が破壊された宿主菌HU11株のURA3遺伝子のDNAシーケンシング結果から明らかなように、遺伝子が破壊された結果、31位の塩基後に、GAACTの5個の塩基が挿入されている。GAACTの5個の塩基の挿入によりフレームシフト突然変異が発生する。フレームシフト突然変異により11位後の254個のアミノ酸コードはすべて突然変異し、突然変異により合計15個の終了コードが発生し、このことから、URA3構造遺伝子がさらに発現できないことを示している。GAACTの5個の塩基は同時に復帰突然変異を発生する確率が極めて小さく、実験によっても宿主菌HU11株の復帰突然変異率がゼロであることを確認した。このような「0」復帰突然変異率を有する宿主菌は、形質転換やスクリーニングに特に有利である。上記遺伝子ノックアウト技術によって構築されたURA3−栄養要求性宿主細胞株HU−11(CGMCC No.1218)については本出願者が出願した発明特許CN1651570Aに開示された。宿主ハンセヌラ・ポリモルファ菌HU−11のオロチジン−5−リン酸デカルボキシラーゼ遺伝子(HURA3)が破壊されたDNA配列はSEQ ID NO :5に示される。
SEQ ID NO:1に示される配列に基づいてヌクレオチド配列(以下、略称HBsAg adw2遺伝子)を合成し、HBsAg adw2遺伝子プラスミドを含むグリセロール菌として構築し、シーケンシングして正確なプラスミドをEcoRI/BamHIで二重酵素消化させ、酵素消化産物からゲル抽出して、701bpの目的断片DNAを回収する。
酵素消化し、同定して正確なプラスミドpHMPT−02をEcoRI/BamHIで二重酵素消化させ、ゲル抽出して回収したベクターDNAを連結して、ハンセヌラ・ポリモルファの細胞内プラスミドpMPT−HBS−adw2を得て、プラスミドpMPT−HBS−adw2をE.coli Competent Cell JM109(Code No.D9052)に熱形質転換して、平板にコーティングして菌体を一晩培養する。形質転換平板から単一コロニーを選択して、プラスミドDNAを抽出した後、EcoRI/BamHIで二重酵素消化させ、酵素消化結果から、すべて陽性クローンであることが分かる。シーケンシングした結果、プラスミドpMPT−HBS−adw2の結果は正確である。
組換えハンセヌラ・ポリモルファB型肝炎ウイルス表面抗原(HBsAg) adw2サブタイプエンジニアリング菌株を構築するには、出願者が開発した細胞エレクトロポレーション技術を用いて、振幅1500V、容量22μf、時間定数3−5msのRCパルスで1回電気穿孔し、pMPT−HBs adw2プラスミドを用いて、URA3−遺伝子をノックアウトしたHU−11株(CGMCC No.1218)のハンセヌラ・ポリモルファ細胞を形質転換する。MD選択培養用平皿から成長した単一コロニー形質転換体を選択して、MD液体培地に移した後、連続継代培養し、adw2サブタイプHBsAg遺伝子及び対応する調節要素をマルチコピーにして、宿主であるハンセヌラ・ポリモルファ細胞染色体に異種組み込む。千株余りの形質転換体の単一コロニーについて下記3つのステップのスクリーニングを行う。
(1)形質転換体の単一コロニーが大きく、細胞の成長速度が高いクローン株はマルチコピーの確率が高い。
(2)PCR技術によってHBsAg遺伝子と単一コピー数MOX(メタノールオキシダーゼ)遺伝子の電気泳動バンドの明るさを比較することによって、HBsAg遺伝子のコピー数を半定量的に決定する。
(3)メタノールで誘導して振盪フラスコ培養を72時間行った形質転換体の細胞が破砕した後に放出したHBsAgの発現レベルを検出する。

エンジニアリング菌のHBsAg遺伝子の増幅産物の大きさは約800bp、ハンセヌラ・ポリモルファの単一コピー遺伝子のMOX遺伝子の増幅産物の大きさは約2000bpである。スクリーニングして得られたエンジニアリング菌株のPCR増幅産物の電気泳動写真は図3に示され、図3中、1はMarkerである。最終スクリーニングして得られた組換えハンセヌラ・ポリモルファのB型肝炎ウイルス表面抗原(HBsAg)エンジニアリング菌株の番号はHS604−5である。
天壇生物社より提供したadr2サブタイプHBsAgB型肝炎表面抗原の凍結乾燥標準品10μgを希釈液で溶解して、1024ng/mL、512ng/mL、256ng/mL、128ng/mL、64ng/mL、32ng/mL、16ng/mL、8ng/mL、4ng/mL、2ng/mL、0ng/mL(希釈液)の合計11個の標準点に倍数希釈して、ラジオイムノアッセイキットでHBsAg反応を検出する。
得られたエンジニアリング菌株(番号HS604−5)について30リットルのパイロット規模発酵(ロット番号20150422)を行い、87時間発酵後、10 OD600nm1mLをサンプリングし、ガラスビーズで細胞(細胞破砕率65%)を破砕した後、サンプルを200倍希釈して、標準品とともにラジオイムノアッセイキット反応に用い、γ−カウンターで自動的に作成したフィッティング曲線から得られるHBsAg抗原の発現量は126.9(ng/mL)である。それにより組換えハンセヌラ・ポリモルファの細胞内HBsAg抗原の発現量を計算する。

組換えハンセヌラ・ポリモルファの組換えHBsAg純品(原液)の電子顕微鏡写真は図4に示される。結果から分かるように、組換えHBsAgは、高純度、高濃度であり、ウイルス様粒子(VLP)の構造が安定的である。
不活性化組換えハンセヌラ・ポリモルファの最適な条件を決定するために、下記要件を満たす必要がある。(1)不活性化組換えハンセヌラ・ポリモルファの生存率をできるだけ低下させ、5%未満にする。(2)完全な細胞構造を保持して、不活性化組換えハンセヌラ・ポリモルファの細胞内の抗原性物質が漏れないようにする。(3)抗原反応活性を低下させないように組換えハンセヌラ・ポリモルファの細胞内で発現するHBsAgウイルス様粒子(VLP)の熱安定性を維持する。以上の3つの点は不活性化組換えハンセヌラ・ポリモルファが発現するHBsAgの生産プロセスの主な条件であり、ワクチンの製造及び検証規則を制定するために根拠を提供している。不活性化組換えハンセヌラ・ポリモルファ細胞におけるHBsAgウイルス様粒子(VLP)の熱安定性は最初に解決すべき問題である。このために、異なる温度(50℃、52℃、54℃、56℃及び58℃)、及び異なる時間(1h、2h及び3h)の組換えHBsAgハンセヌラ・ポリモルファの不活性化試験を16群設計して、20℃を対照群とする。検出したところ、不活性化温度、時間の上昇に伴い、細胞壁外層の溶解度が高まり、細胞の破砕率が増大し、細胞内HBsAgVLP抗原反応性については52℃、1hで倍増するピークが現れ、細胞外HBsAg抗原反応性は、不活性化試験のすべての温度と時間の範囲で極めて低く、このことから、細胞内HBsAgVLPが外部へ漏れず、完全不活性化組換えハンセヌラ・ポリモルファ細胞の構造が保持されていることが分かる。不活性化細胞の生存率は、56℃、3hの条件で1/50,000に低下した。それにより、不活性化組換えハンセヌラ・ポリモルファ細胞の条件を最適化させるために根拠を提供している。各種の要因を配慮に入れて、不活性化プロセスの条件として、最終的に、不活性化温度52℃〜54℃、不活性化時間1時間〜3時間とする。
B型肝炎ウイルスによるチンパンジーの感染についての一連の研究結果から明らかなように、HBsAg特異的CTL T細胞は、HBVを感染した肝細胞にターゲットしてIFN−γを放出し、ステップ1として、肝細胞を損傷せずに90%以上のcccDNA分子プールを減少させ、ステップ2として、この過程において損傷されて感染された肝細胞を増加させ、免疫逆転をトリガーさせる。CTLエピトープの実験概要、予測及び特許発明などの3つの文献報道によれば、HBVカプシド抗原(Pre−S1−Pre−S2−HBsAg)の重複しないCTLエピトープは合計で23個である。本発明におけるHBsAgアミノ酸配列には、VLQAGFFLL、PFVQWFVGL、FLLTRILTI、WYWGPSLYSI、SLNFLGGSPV、FLGGSPVCL、LYSIVSPF、LYSIVSPFI、PFIPLLPIF、LLLCLIFLL、LLCLIFLLV、 LLDYQGMLPV 、LVLLDYQGML、VLLDYQGML、WLSLLVPFV、LLVPFVQWFV、GLSPTVWLSA、SIVSPFIPLL、LLPIFFCLWVの合計19個のCTLエピトープが含まれている。
プレフィルドシリンジに分注したB型肝炎治療ワクチンの完全な接種は、良好な総合的費用便益比を有する。
以下、実施例にて本発明についてさらに説明するが、下記実施例は説明するためのものであり、限定するものではなく、本発明の保護範囲は下記実施例により制限されない。
前記SEQ ID NO:1配列に基づいて、pMPT−HBsAg adw2プラスミド(すなわち前記SEQ ID NO:1配列を含む発現ベクター)を構築した。プラスミドpMPT−HBs adw2の構築プロセスは以下の工程を含む。
設計したSEQ ID NO:1に示されるDNA配列に基づいて、HBsAg adw2遺伝子を合成し、HBsAg adw2遺伝子プラスミドを含むグリセロール菌を構築し、MC407B−16と命名した。
シーケンシングして正確なMC407B−16プラスミドをEcoRI/BamHIで二重酵素消化させ、酵素消化産物についてTaKaRa PCR Fragment Recovery Kit(Code No.D301)でゲル抽出して701bpの目的断片DNAを回収し、Inset DNA6とした。
酵素消化し、同定して正確なプラスミドpMPT−02をEcoRI/BamHIで二重酵素消化させ、ゲル抽出して回収し、ベクターDNAを得て、Vector DNA6とした。
MC407A+B+C+D−77プラスミドをそれぞれプライマーRV−M、M13−47、MC407P1、MC407P2、MC407P3、MC407P4、MC407P5、MC407P6、MC407P7、MC407P8、MC407P9、MC407BF11、MC407BR11でシーケンシングした結果、プラスミドpMPT−HBs adw2の結果は正確であることが分かった。
組換えハンセヌラ・ポリモルファHBsAgエンジニアリング菌株(すなわち、前記SEQ ID NO:1 に示される配列を含むハンセヌラ・ポリモルファ宿主細胞の形質転換スクリーニング株)の構築
組換えハンセヌラ・ポリモルファの形質転換とスクリーニングの過程は図5に示される。
具体的に以下を含む。
1)細胞エレクトロポレーション法を用いて、pMPT−HBsAgプラスミドで宿主細胞であるURA3−栄養要求性ハンセヌラ・ポリモルファ細胞株HU−11(CGMCC No.1218)を形質転換した。選択培地(MD液体培地)で平皿により培養した。MD選択培養用平皿から成長した単一コロニー形質転換体を選択して、MD液体培地に移した後、連続継代培養し、adw2サブタイプHBsAg遺伝子及び対応する調節要素をマルチコピーにして、宿主であるハンセヌラ・ポリモルファ細胞の染色体に異種組み込んだ。
2)菌株スクリーニングは以下のステップを含む。
(1)ウラシル原栄養形質転換体の単一コロニーを選択する。
菌体の成長速度が高いコロニーを選定し、PCR技術によってHBsAg遺伝子のバンドの明るさを検出した後、コピー数の多いコロニーを選択し、選択培地を用いて単一コロニーを振盪フラスコ培養し、20〜400世代連続継代培養した。
(2)マルチコピーの異種組み込み形質転換クローン株をスクリーニングする。
ステップ(1)で継代培養した後、メタノールで72時間誘導培養して、ラジオイムノアッセイ(Radio immunoassay or radioimmunoassay、RIA)で形質転換体の細胞が破砕された後に放出するHBsAgの発現レベルを測定した。
(3)遊離プラスミドを除去した高コピー・高発現クローン株をスクリーニングする。
ステップ(2)でスクリーニングしたクローン株を、YPD完全培地で48時間培養した後、選択培地平皿に移してクローン化培養を行い、定量的PCRによってHBsAg遺伝子のコピー数を検出し、RIAによってHBsAg発現レベルを検出した。
(4)ステップ(3)の検出結果に基づいて、遺伝的安定性に優れた組換えハンセヌラ・ポリモルファHBsAgエンジニアリング菌の初代菌株を選択した。
主なプロセス過程及び操作ポイントは以下のとおりである。
30リットルのパイロット規模発酵の主なプロセス
1)シード培地200mlで、液体窒素を用いて貯蔵した菌種を解凍して、培地に接種し、2つの0.5L振盪フラスコに等分して、31℃で22時間培養して一次シードとした。
2)シード培地1600mlを用いて一次シードを二次シード培地に移して、6個の1L振盪フラスコに等分して、31℃で20時間培養して二次シードとした。
3)12L発酵培地をpH5.5に調整して30L発酵缶に投入し、二次シードを接種した後、30〜31℃で、グリセリン、メタノールの2種の炭素源で、成長、抑制解除及び誘導の3つの時期を経て、合計85−96時間培養して、誘導を停止して2〜3時間後、細胞を収穫した。凍結細胞をホモジネートした。
(1)成長時期における流加操作は、溶存酸素が有意に低下して、基礎培地が消費されたときに実施し始め、且つ、基礎培地の消費量の増加に伴い徐々に流加速度を向上させ、溶存酸素が戻る前の2−3時間に流加を終了する。
(2)成長時期の後期に、溶存酸素の上昇をモニタリングして、溶存酸素の最低値を記録し、溶存酸素が70〜80%まで戻ったときに、流加を開始させて、抑制解除時期に入る。
(3)抑制解除時期の後期に、流加終了後、溶存酸素が戻り始める。溶存酸素が70−80%まで戻ったときに、メタノールを流加して溶液を誘導し、メタノール濃度を3〜5‰に制御し、メタノール検出流加コントローラで流加速度を制御する。
細胞収獲時のメタノール残留を減少させるように、発酵終了前の2〜3時間に、メタノール流加を停止する。
1.塩化カルシウム溶液の調製
CaCl2 11.33gを正確に秤量して、洗浄した三角フラスコに投入し、適量の脱イオン水を加えて溶解した後、200mlに定容した。
下記試薬を正確に秤量した。
下記試薬を正確に秤量した。
以上の4種類の溶液をそれぞれ除菌して濾過して、使用時まで保存した。
110℃で30分間高圧蒸気滅菌を行うことで、500ml三角フラスコを2個、1000ml三角フラスコを6個、100mlと500mlのメスシリンダーをそれぞれ1つ充填前に滅菌した。
クリーンベンチにおいて、無菌操作により滅菌したグリセリン水溶液をそれぞれ100ml秤取して、それぞれ滅菌後の2個の500ml三角フラスコに投入して、塩化カルシウム溶液1ml、微量元素溶液1ml、ビタミン溶液0.5ml、痕跡元素溶液0.25mlをそれぞれ加えて、加えた溶液を均一に振とうさせた。
クリーンベンチにおいて、無菌操作技術により滅菌後のグリセリン水溶液1600mlを秤取して、滅菌後の2000ml三角フラスコに投入して、塩化カルシウム溶液16ml、微量元素溶液16ml、ビタミン溶液8ml、痕跡元素溶液4mlをそれぞれ加えた。
試薬として、NH4H2PO4175g、MgSO4・7H2O 40g、KCl 44g、NaCl 4.4gを正確に秤量して、2000mlの脱イオン水に溶解した。
500ml小ビーカーにグリセリン520gを秤量して、消泡剤10mlを加えて、オンラインで滅菌し、次に、塩化カルシウム溶液175ml、微量元素溶液175ml、ビタミン溶液88ml、痕跡元素溶液44mlを加えた。
1000ml三角フラスコに、NH4H2PO487g、グリセリン260gを秤取して、脱イオン水を500ml加え、包まれた流加管路に加えて、110℃で30分間滅菌した。
5000ml三角フラスコに、グリセリン1800gを秤取して、脱イオン水660mlを加え、包まれた流加管路に加えて、110℃で30分間滅菌し、冷却後、無菌操作により濾過除菌した塩溶液540mlを加えた。
1000ml三角フラスコに、グリセリン400mlを秤取して、包まれた流加管路に加え、110℃で30分間滅菌し、冷却後、無菌操作によりメタノール1600mlを加えた。
実施例3で得られた発酵液について、細胞を収穫して細胞を洗浄し、詳細な精製ステップについては、以下の文献を参照すればよい。李津、孔艷.組換えB型肝炎治療ワクチンの生産プロセス。李津、兪詠霆、董徳詳編集:生物製薬装置及び分離精製技術.第1版.北京:化学工業出版社、2003:348−349。さらに、上記収獲した細胞をホモジナイザーで破砕して、HBsAgを放出させ、0.22μmの微多孔ろ過器で濾過して、細胞破片を除去し、次に、300Kの限外ろ過器で限外ろ過して小分子不純物を除去し、シリカゲル吸着処理法によりHBsAgを抽出し、最後に、ブチルアガロース疎水性クロマトグラフィー方法で精製した。
不活性化組換えハンセヌラ・ポリモルファ細胞の最適な条件試験
不活性化組換えハンセヌラ・ポリモルファの最適な条件を決定するのに、以下の要求を満たす必要がある。
(1)不活性化組換えハンセヌラ・ポリモルファの生存率をできるだけ低下させ、5%未満にする。(2)完全な細胞構造を保持し、多価の不活性化組換えハンセヌラ・ポリモルファのアジュバント活性作用を発揮させる。(3)組換えハンセヌラ・ポリモルファ細胞内で発現するHBsAgウイルス様粒子(VLP)の完全性を保持して、抗原性を低下させないようにする。以上の3つの点は不活性化組換えハンセヌラ酵にHBsAgを発現する生産プロセスの主な条件であり、ワクチンの製造及び検証規則を制定するために根拠を提供している。不活性化組換えハンセヌラ・ポリモルファ細胞におけるHBsAgウイルス様粒子(VLP)の熱安定性は最初に解決すべき問題である。
ハンセヌラ・ポリモルファエンジニアリング菌(菌株番号HS604−5)細胞の培養は、発酵又は振盪フラスコ誘導培養を経た後、細胞を遠心法でリン酸塩緩衝液(PBS)において3回洗浄して、沈殿したハンセヌラ・ポリモルファを、体積を計算したPBSに懸濁させた。細胞をOD600nmでカウントして、PBSで10 OD600nm/ミリリットルに希釈し、試験管ごとに2ミリxリットルとし、試験群ごとに、破砕試験管と非破砕試験管の2本を設置し、16個の試験群では、合計32本の細胞サンプル試験管を必要とした。
設定温度の水浴に入れて、所定時間にかけて組換えハンセヌラ・ポリモルファを不活性化させた。
不活性化組換えハンセヌラ・ポリモルファを、クロラムフェニコール含有完全培地を入れた寒天皿において37℃で3日間培養し、生存率をカウントした。
不活性化ハンセヌラ・ポリモルファを4℃で貯蔵して、さらなる使用に供する。

- HBsAgを発現する不活性化全組換えハンセヌラ・ポリモルファ細胞によるB型肝炎治療ワクチンであって、
前記B型肝炎治療ワクチンは前記不活性化全組換えハンセヌラ・ポリモルファ細胞をアジュバントとし、
前記抗原としてのHBsAgの組換えハンセヌラ・ポリモルファ細胞内での発現量が108細胞あたり6〜10μgのHBsAgを含有し、
前記HBsAgは、合計16〜21個のHBsAg特異的CTLエピトープを含有し、
前記組換えハンセヌラ・ポリモルファが発現するHBsAgのDNA配列はSEQID NO:1に示されることを特徴とするB型肝炎治療ワクチン。 - 前記組換えハンセヌラ・ポリモルファが発現するHBsAgは、

- 前記組換えハンセヌラ・ポリモルファが発現するHBsAgはウイルス様粒子構造であり、HBsAgをハンセヌラ・ポリモルファ脂質に挿入してなり、前記HBsAgの14個のシステインのうち9〜12個はジスルフィド結合を形成することを特徴とする請求項1に記載のB型肝炎治療ワクチン。
- 前記組換えハンセヌラ・ポリモルファの宿主であるハンセヌラ・ポリモルファ細胞株はHU−11であり、保蔵番号がCGMCC No.1218であり、前記宿主であるハンセヌラ・ポリモルファのオロチジン−5−リン酸デカルボキシラーゼ遺伝子が破壊されたDNA配列がSEQ ID NO:3に示されることを特徴とする請求項1に記載のB型肝炎治療ワクチン。
- 剤型が、プレフィルド注射液剤、注射液剤又は凍結乾燥粉末注射剤から選ばれることを特徴とする請求項1〜5のいずれか1項に記載のB型肝炎治療ワクチン。
- HBsAg原液又はアルミアジュバントHBsAgのうちの1種をさらに含むことを特徴とする請求項1に記載のB型肝炎治療ワクチン。
