以下に本発明の好ましい実施形態を説明する。以下に提供される実施形態は、本発明のよりよい理解のために提供されるものであり、本発明の範囲は以下の記載に限定されるべきでないことが理解される。従って、当業者は、本明細書中の記載を参酌して、本発明の範囲内で適宜改変を行うことができることは明らかである。また、本発明の以下の実施形態は単独でも使用されあるいはそれらを組み合わせて使用することができることが理解される。
DNAワクチンは、コードされた抗原への体液性および細胞性の免疫応答惹起することができる有力な剤形である。しかしながら、抗原特異的CD8+T細胞応答を達成するには、免疫原性には改善が必要である。本発明は、免疫原性の改善をもたらすものである。
1つの局面において、本発明は、エキソソームマーカータンパク質をコードする核酸配列と、ワクチン抗原をコードする核酸配列とを含む核酸構成物(例えば、DNAコンストラクト)を提供する。本発明の核酸構成物の代表的な実施形態であるDNAワクチンは体液性免疫のみならず、細胞性免疫応答特に細胞傷害性T細胞(CTL)をも強く誘導することができることから、感染症の予防ワクチンやがん治療ワクチンとして期待されているが、従来はヒトにおいては安全性の問題やDNAワクチンの免疫原性が弱く臨床試験において期待された効果が得られていなかった。本発明では、実施例において実証されているように、エキソソームマーカータンパク質をコードする核酸配列と、ワクチン抗原をコードする核酸配列とを含む核酸構成物を提供することにより、エキソソーム標的DNAワクチンの免疫原性が臨床適用できる程度の低減されており、このワクチンを用いたエキソソームに抗原を搭載させたエキソソームワクチンの臨床上の効果も確認されていることから、DNAワクチンの臨床応用を可能する点で注目されるべきである。例えば、実施例に記載されているように、本発明の代表的な実施形態であるエキソソーム標的DNAワクチンを筋注エレクトロポレーション法によりマウスに投与した結果、エキソソームに抗原が発現しない群と比較して強いCTLの誘導が確認される。ここで、実施例で使用されるCD63、CD9、CD81等と融合したOVA抗原をコードするプラスミドDNA(例えば、pCD63−OVA)によるin vitroトランスフェクションは、エキソソームによるOVAの運搬をもたらした。また、この強いCTLの誘導は単に膜への局在だけではなくエキソソームにターゲットした結果得られたものである事が、小胞体の膜タンパクであるカルネキシン(CALNEXIN)とオボアルブミン(OVA)抗原を融合したプラスミドDNAを作製して比較したことから判明しており、本発明の優位な効果が実施例等で実証されている。また、本発明者らは、本発明の代表的な実施形態である実施例に示すようなDNAワクチンを用いてOVAを発現しているマウスの胸腺種であるE.G−7をマウス皮下に移植し、その後DNAワクチンの接種を行ったところ、コントロール群に比べて、エキソソーム標的DNAワクチンを接種した群では腫瘍の増殖を強く抑制することが確認されており、抗がん剤としても顕著な効果が奏されることが実証されている。
別の局面において、本発明はまた、本発明の核酸構成物、本発明のDNAワクチン、本発明のタンパク質または本発明のエキソソームを含む医薬を提供する。本発明の医薬等で使用される核酸構成物、DNAワクチン、タンパク質、エキソソームは、(エキソソーム−抗原の核酸構成物)の項等おいて詳述される本発明の任意の更なる特徴を1つまたは複数含んでいてもよいことが理解される。
別の局面では、本発明は、エキソソームマーカータンパク質およびこれをコードする核酸配列の新規用途を提供する。

本実施例では、本発明者らは、エキソソームマーカータンパク質の例として、エキソソームを含む様々な原形質膜上に発現されるテトラスパニンタンパク質であるCD63に焦点を当て、CD63と融合した抗原をコードするDNAワクチンはエキソソームを標的とすることができるのか、そしてワクチンの免疫原性を改善することができるのかについて試験した。本発明者らは、CD63と融合したOVA抗原を発現するプラスミドを構築し、抗原にエキソソームを標的とさせた。本発明者らはまた、CD63融合抗原を介したDNAワクチンのエキソソームへの標的化によって、誘導または促進される免疫応答のタイプを同定しようと試み、そして、抗がんワクチンとしての有用性を評価した。
DNAコンストラクト
全長のCD63(aa 1〜238;配列番号3〜4)、CD9(aa 1〜226;配列番号7〜8)、CD81(aa 1〜236;配列番号11〜12)、およびカルネキシン(aa 1〜591;マウスNM_001110499およびNP_001103969。配列番号13〜14(いずれもマウス)のcDNAを、C57BL/6J由来のマウス肺cDNAライブラリーをテンプレートとして用いるPCRによって増幅した。cDNAフラグメントをシーケンシングによって検証し、次いで、pCI哺乳動物発現ベクター(Promega)、pCIneo−FLAG(Promega)またはpEGFP−N1(Clontech)に、以前に記載されたように導入した(Jounai, N., K. Kobiyama, M. Shiina, K. Ogata, K. J. Ishii, and F. Takeshita. 2011. J Immunol 186: 1646-1655.)。全長OVA(aa 1〜386)をコードするcDNA(V00383.1 ;CAA23682.1配列番号15〜16)を、pCI OVA(pOVA)から、PCRによって増幅した(Nagata, T., T. Higashi, T. Aoshi, M. Suzuki, M. Uchijima, and Y. Koide. 2001. Vaccine 20: 105-114.)。OVA cDNAフラグメントをシーケンシングによって検証し、次いで、pCIneo−FLAGに導入し、pOVA−FLAGと名付けた。全長OVAタンパク質を、CD63、CD9、CD81、またはカルネキシンと、N末端の位置で融合した。リンカーとして、グリシンヘキサマー(6×グリシン)を、OVAと融合遺伝子との間に挿入した。融合遺伝子をシークエンシングによって検証し、次いで、pCIベクター、pCIneo−FLAGベクターまたはpEGFP−N1ベクターに導入し、pCD63−FLAG(pCD63)、pCD63−6×Gly−OVA−FLAG(pCD63−OVA)、pCD9−6×Gly−OVA−FLAG(pCD9−OVA)、pCD81−6×Gly−OVA−FLAG(pCD81−OVA)、pCalnexin−6×Gly−OVA−FLAG(pCal−OVA)、またはpCD63−6×Gly−OVA−EGFP(pCD63−OVA−EGFP)と名付けた。全てのプラスミドを、Escherichia coli DH5αに形質転換し、製造者のプロトコルに従って、Qiagen Plasmid Endo−free Maxiprep kits(Qiagen)を用いて精製した。
293T細胞およびE.G7−OVA細胞(E.G7)細胞を、American Type Culture Collectionから購入した。FreeStyleTM 293−F細胞(293F細胞)を、Life Technologiesから購入した。293T細胞を、10%のFCSおよび50μg/mlのペニシリン−ストレプトマイシンを補充したDMEM中で、5% CO2下37℃で培養した。293F細胞を、FreeStyleTM 293Expression medium(Life Technologies)(動物血清由来のエキソソーム非含有である)中、8% CO2下37℃で培養した。E.G7細胞を、10%のFCS、50μg/mlのペニシリン−ストレプトマイシン、0.05mMの2−メルカプトエタノール、1mMのピルビン酸ナトリウム、10mMのHEPES、および1×非必須アミノ酸を補充したRPMI−1640(Life Technologies)中、5% CO2下37℃で培養した。
血清(エキソソーム)非含有293F細胞を、様々なDNAコンストラクトで、48〜72時間トランスフェクトした。次いで、培地上清を回収し、2000×gで30分間遠心分離した。製造者のプロトコルに従ってTotal Exosome Isolation kit(Life Technologies)を用いて、培養培地からエキソソームを単離した。簡潔に述べると、1.5mlのTotal Exosome Isolation reagentを3mlの細胞培養培地に添加し、そして、4℃で一晩インキュベートし、その後、10,000×g、4℃で1時間遠心分離した。エキソソームのペレットを、100μlのPBSで再懸濁した。回収したエキソソームを、電子顕微鏡、ウェスタンブロッティング、およびフローサイトメトリーアッセイによって、以下で記載されるように定性的および定量的に特性評価した。
細胞トランスフェクションを以前に記載されたように行った(Jounai, N., K. Kobiyama, M. Shiina, K. Ogata, K. J. Ishii, and F. Takeshita. 2011. J Immunol 186: 1646-1655.)。一過性のトランスフェクションを、製造者のプロトコルに従ってLipofectamine2000(Life Technologies)で行った。293F細胞(1×106/ml)を、それぞれの発現プラスミドでトランスフェクトした。トランスフェクトした細胞を、トランスフェクションの48時間後に採取した。トランスフェクトした細胞を含む培養培地を、2000×gで30分間遠心分離した。細胞をRIPAバッファーで溶解し、氷上で15分間インキュベートした。上清および単離したエキソソーム試料を、3×SDSバッファー(125mMのTris−HCl:pH6.8、4%のSDS、20%のグリセロール、および0.01%のブロモフェノールブルー)で希釈し、95℃で5分間ボイルした。イムノブロッティング分析を、抗CD63Ab(R5G2、MEDICAL&BIOLOGICAL LABORATORIES CO.,LTD)、抗カルネキシンAb(ab22595、Abcam)、または抗FLAG M2−ペルオキシダーゼ(HRP)Ab(A8592、Sigma)を用いて、以前に記載されたように行った(Jounai, N., K. Kobiyama, M. Shiina, K. Ogata, K. J. Ishii, and F. Takeshita. 2011. J Immunol 186: 1646-1655.)。
免疫金電子顕微鏡アッセイを、以前に記載されたように行った(Thery, C., S. Amigorena, G. Raposo, and A. Clayton. 2006. Current protocols in cell biology / editorial board, Juan S. Bonifacino ... [et al.] Chapter 3: Unit 3.22.)。単離したエキソソームを、1次Abとしてマウス抗FLAG Ab(Sigma)と、2次Abとして金標識した抗マウスIgG(10nm Gold)(Abcam)とを用いて免疫標識した。
293F細胞(1×106/ml)を、pCD63−OVA−EGFPでトランスフェクトした。トランスフェクトした細胞を含む培養培地を、トランスフェクション後48時間で回収し、遠心分離した。得られた上清を、INFLUX (BD Bioscience)によって特性評価した(18)。100nmベーズおよび200nmビーズ(Polysciences, Inc.)を用いて、エキソソーム画分のための適切なゲーティングを決定した(図5)。
6週齢雌性C57BL/6Jマウスを、CLEA Japan, Incから購入した。マウスを、ヌクレアーゼ非含有食塩水中、対照または融合タンパク質(OVA、CD63、CD63−OVA、CD9−OVA、CD81−OVA、またはCal−OVA)をコードするプラスミドDNA(50μg)を用いた筋肉内エレクトロポレーション(imEPT)によって免疫化した(50μl/筋肉)。
細胞性免疫応答を評価するため、脾細胞(2×106/ウェル)を調製し、20μg/mlのOVAペプチド(OVA257-264、H−2Kb制限OVAクラスIエピトープ、またはOVA323−339、I−A(d)−制限OVAクラスIIエピトープ)またはOVA抗原(Calbiochem)を含有する完全RPMI−1640培地でインキュベートした。細胞性免疫応答は、以前に記載されたように測定した(Onishi, M., et al., 2015. J Immunol 194: 2673-2682.)。
293T細胞(1×106/ml)を、各発現プラスミドでトランスフェクトした。細胞を、トランスフェクション後48時間インキュベートし、細胞および細胞培養上清におけるOVA発現の濃度を、製造者のプロトコルに従ってELISA(Institute of Tokyo Environmental Allergy)により測定した。OVA特異的血清Ab力価を、以前に記載されたように測定した(Onishi, M., et al., 2015. J Immunol 194: 2673-2682.)。
テトラマーアッセイを、以前に記載されたように行った(Kobiyama, K., et al., 2014. Proc Natl Acad Sci U S A 111: 3086-3091.)。簡潔に述べると、脾細胞を、PE標識H−2Kb OVAテトラマー(SIINFEKL)(MEDICAL&BIOLOGICAL LABORATORIES CO.,LTD.)を用いて、室温で20分間染色した。次に、細胞を、PBS中の、FITC標識抗CD8α(KT15、BioLegend)、PC標識抗TCRβ鎖(H57−597、BioLegend)、BrilliantViolet421TM標識抗CD62L(MEL−14、BioLegend)、およびAPC/Cy7標識抗CD44(IM7、BioLegend)Abによって染色した。フローサイトメトリーにより、OVAテトラマー+CD44+CD8a+TCRb+細胞数を決定した。
in vivoCTLアッセイを、以前に記載されたように行った(Kobiyama, K., et al., 2014. Proc Natl Acad Sci U S A 111: 3086-3091.)。簡潔に述べると、6週齢C57BL/6Jマウスに、DNAワクチンでワクチン接種した。21日目に、ナイーブC57BL/6Jマウス由来の脾細胞を、2または0.2μMのいずれかのCFSEにより、37℃で10分標識した。CFSEで標識した細胞を、それらを、OVA257-264(10μg/ml)と37℃で90分間インキュベートすることによりペプチドパルシングに供した。次いで、細胞を洗浄し、各処置群からの同数を、血管内経路を介して免疫化マウスに送達した。送達の24時間後に脾細胞を単離し、フローサイトメトリーによりCFSE標識細胞を分析した。
マウスの背中の毛を剃り、そして100μlのPBS中1×106のE.G7細胞をマウスの皮下に注入した。平均腫瘍体積が1cm3に達したとき、pOVA、pCD63−OVA、pCal−OVA、またはpCIneo−FLAG(陰性対照として)によって1次免疫化をマウスに施した。腫瘍予防実験では、マウスをこれらのプラスミドDNAワクチンで免疫化し、E.G7細胞を、10日後に100μlのPBS中1×106細胞で注入した。腫瘍体積は、デジタルキャリパーを用いて定期的な間隔で測定し、長さ×幅×高さで計算した。
群間の差異の統計的有意性は、Mann−Whitney U検定を用いて決定した。
エキソソームに捕捉されたCD63融合抗原はワクチンとして作用する
以前の報告は、抗原含有エキソソームワクチン接種が、Th1応答およびCTL活性化を誘導することに触れている(Wolfers, J., et al., 2001. Nat Med 7: 297-303.、Qazi, K. R., et al., 2009. Blood 113: 2673-2683)。細胞における抗原と融合したCD63の発現が、抗原のみの単純な発現より、高効率な抗原分泌をもたらすのかを調べるため、本発明者らは、CD63およびOVAの融合タンパク質をコードするプラスミドDNA(pCD63−OVA)を構築した。本発明者らは、CD63がエキソソーム膜上に発現され、既に一般的なエキソソームマーカーとして使用されていることから(Pols, M. S., and J. Klumperman. 2009. Experimental cell research 315: 1584-1592.)、CD63を介してOVA抗原をエキソソームに標的化することを策定した。加えて、対照として、本発明者らは、カルネキシン−OVA融合タンパク質をコードする1つの対照プラスミドDNA(pCal−OVA)を調製した。pCal−OVAは、コードされた抗原を小胞体膜に標的化する(Baietti, M. F., Z. et al., 2012. Nat Cell Biol 14: 677-685.;Gross, J. C., V. Chaudhary, K. Bartscherer, and M. Boutros. 2012. Nat Cell Biol 14: 1036-1045.;Lasser, C., V. S. Alikhani, K. Ekstrom, M. Eldh, P. T. 上記結果に基づいて、本発明者らは、エキソソームを標的化する抗原を組み込んだDNAワクチンは、増大した免疫原性を有しているのではないかと仮説を立てた。pCD63−OVAを有するDNAワクチンが、他の対照DNAワクチンと比較して増大した免疫原性を有するのかについて調べるため、本発明者らは、imPETによりプラスミドDNAで2回マウスを免疫化し、in vivoで効率的なトランスフェクションを誘導した。ブースト免疫化の1週間後、pCD63−OVA免疫化マウスは、pOVA免疫化マウスより、血清中抗OVA IgG1の力価が顕著に低かったが、これらの2群間で抗OVA総IgGおよびIgG2cには差が無かった(図2A)。pCD63−OVAおよびpOVAの間で抗OVA IgG2c/G1比を比較すると、pOVAと比較して、pCD63−OVA免疫化マウスにおいて、高い値の抗OVA IgG2c/G1が観察された(図2A)。次に、本発明者らは、これらの免疫化マウスにおけるOVA257-264特異的テトラマー+CD44+CD8+T細胞の頻度を確認した。pCD63−OVA免疫化マウスでは、pOVA免疫化マウスと比較して、OVA257-264特異的テトラマー+CD44+CD8+T細胞の高い頻度が観察された(図2B)。本発明者らはまた、クラスI(OVA257-264)またはクラスII(OVA323-333)OVAペプチドのいずれかで脾細胞をex vivoで刺激し、次いで、これらの脾細胞から得られたサイトカイン産生をELISAによって測定した。pCD63−OVAワクチン接種では、pOVAと比較して、各ペプチド刺激について、顕著に高いレベルのIFN−γ産生が検出された(図2C)。
次に、本発明者らは、エキソソームへのコードされた抗原の標的化の戦略が、DNAワクチン接種によるT細胞活性化を促進するための代替の戦略(例えば、小胞体のような他の原形質膜への抗原の標的化)に対して利点を有するのかについて調査した。この目的のために、本発明者らは、異なる融合タンパク質をコードする2つのプラスミドDNA:エキソソームを標的とするpCD63−OVA、およびERを標的とするpCal−OVA、の免疫原性の規模およびタイプを比較した。pCD63−OVAまたはpCal−OVAのいずれかによるimEPTを介した2つのDNAワクチン接種を受けたマウスは、類似する抗OVA Ab力価および抗OVA IgG2c/G1比を生じさせた(図2D)。しかしながら、pCal−OVAによってワクチン接種されたマウスは、pCD63−OVAによってワクチン接種されたマウスより、OVA257-264特異的テトラマー+CD44+CD8+T細胞の高い頻度を示した(図2E)。加えて、pCD63−OVAでワクチン接種したマウス由来の脾細胞は、pCal−OVAによって免疫化したマウスに由来する脾細胞より、刺激に際して顕著に高い量のIFN−γを産生した(図2F)。これらの結果は、抗原の、他の膜ではなく、エキソソーム膜への標的化が、DNAワクチンの免疫原性を改善するのに十分であることを示している。
pCD63−OVAワクチンは強力なCD8+T細胞応答を誘導する(図2)ことから、本発明者らは、CD63を発現するプラスミドDNAが、OVAを発現するDNAワクチンについてジェネティックアジュバントとして作用するのかについて調べた。CD63の潜在的なアジュバント効果を評価するために、マウスを、pOVA、pCD63、pOVAおよびpCD63の混合物、またはpCD63−OVAによって免疫化した。2次免役化の1週間後、pCD63およびpOVAを一緒に受けたマウスにおいて、pOVA単独を受けたマウスと比較して、OVA特異的CD4+およびCD8+T細胞応答の促進はなかった;しかしながら、pCD63−OVAでワクチン接種したマウスは、OVA特異的CD4+およびCD8+T細胞応答において、顕著な増加を示した(図3A、図3B)。これらの結果は、CD63が、融合タンパク質としてOVA抗原とコンジュゲートしない限り、それ自体はジェネティックアジュバントとして機能しないことを示している。したがって、抗原のエキソソーム中またはエキソソーム上への送達は、DNAワクチンの免疫原性を改善し得る。
最後に、本発明者らは、pCD63−OVAワクチンが癌ワクチンおよび/または免疫療法に適用可能であるのかについて調べた。CTL応答は癌ワクチンの有効性に重要であるため、in vivo CTL細胞毒性アッセイを行い、機能的CD8+T細胞活性のレベルを評価した。pCD63−OVAによるマウスの免疫化は、pOVA単独による免疫化によって誘導されるものと比較して、OVA特異的CD8+T細胞媒介機能的細胞毒性を顕著に増大させた(図4A)。pCD63−OVAが腫瘍成長を抑制することができるかを調べるため、OVA過剰発現マウスリンパ腫細胞であるE.G−7細胞を有する外殖同系腫瘍モデルを用いた。E.G−7腫瘍細胞の播種の前に、マウスを、pCD63−OVAまたは他の対照プラスミドDNAワクチンによって免疫化した。腫瘍播種の後、pCD63−OVA免疫化マウスにおいては、対照プラスミドDNA免疫化マウスと比較して、腫瘍成長が顕著に抑制された(図4B)。
体液性および細胞性の両方の免疫応答が、動物モデルにおいてDNAワクチン接種によって誘導された。しかしながら、ヒトでは、DNAワクチンに対する免疫学的応答が、予期されたよりも弱い場合が多数存在する。本発明者らの研究において、pCD63−OVA DNAワクチン接種の有効性が実証され、DNAワクチンの免疫原性が、エキソソームへの抗原の標的化によって改善され得ることが示唆された。本実施例の結果は、1)pCD63−OVAワクチン接種が、コードされるOVA抗原を分泌されたエキソソームに成功裡に標的化すること;2)pCD63−OVAワクチン接種が、例えば、抗原特異的および機能的細胞毒性CD8+T細胞応答のような、強力なI型免疫応答をin vivoで誘導すること;および3)pCD63−OVAワクチン接種が、同系外殖E.G−7細胞由来腫瘍の成長を、予防的および治療的の両方の様式で抑制することを示唆している。
38〜41)。これらのワクチンおよびワクチン候補は、DNAワクチンの低い免疫原性を改善するために様々な戦略を利用している。本発明者らの結果は、抗原とエキソソームマーカー(例えば、CD63)との間の融合を通じた、コードされる抗原のエキソソームへの標的化の戦略が、DNAワクチンの免疫原性を改善するための、もう1つの実行可能な方法となり得ることを示唆している。この戦略は、特に、コードされる抗原に対するCD8+T細胞応答を増大させるのに適切である。本発明者らの発見は、どのようにDNAワクチンの免疫原性および有効性を改善するかについての洞察を提供する。さらに本発明者らの発見は、DNAワクチンにおけるDNAにコードされた抗原が免疫系によってどのように送達・プロセスされ抗原特異的免疫応答を提供するのかについての生物学的および免疫学的な理解を深めるものである。
以上のように、本発明の核酸構成物は、ワクチンDNAの免疫原性の改善、免疫応答の増強、T細胞応答の増強、がんを治療および予防などの効果を達成することができることが示された。

Claims (16)
- CD63、CD81およびCD9から選択される少なくとも1つのエキソソームマーカータンパク質をコードする核酸配列と、ワクチン抗原をコードする核酸配列とを含む核酸構成物。
- 前記抗原は、がん抗原およびウイルス抗原から選択される請求項1に記載の核酸構成物。
- プラスミドDNAである請求項1に記載の核酸構成物。
- 請求項1〜3のいずれかに記載の核酸構成物を含むDNAワクチン。
- Th1型の免疫誘導を改善するための請求項4に記載のDNAワクチン。
- がんまたはウイルス疾患を対象とすることを特徴とする、請求項5に記載のDNAワクチン。
- ワクチン抗原タンパク質とエキソソームマーカータンパク質とを融合した形態であるタンパク質であって、該エキソソームマーカータンパク質はCD63、CD81およびCD9から選択される少なくとも1つである、タンパク質。
- ワクチン抗原タンパク質とエキソソームマーカータンパク質とを融合した形態で含む、エキソソームであって、該エキソソームマーカータンパク質はCD63、CD81およびCD9から選択される少なくとも1つである、エキソソーム。

- エキソソームマーカータンパク質をコードする核酸配列を有する核酸を含む、ワクチンDNAの免疫原性を改善するための組成物であって、該エキソソームマーカータンパク質はCD63、CD81およびCD9から選択される少なくとも1つである、組成物。
- 前記ワクチンDNAに含まれる抗原は、がん抗原およびウイルス抗原から選択される請求項14に記載の組成物。
