Description
金ナノ粒子を含み、
前記金ナノ粒子は、マトリックスメタロプロテアーゼの酵素活性部位の亜鉛イオンと配位結合可能な官能基を含む保護剤を表面に有することを特徴としている。
(A) (CH2)nCOOHまたはその塩、あるいは対応するカルボキシレートイオン(nは0〜3の整数を示す。)
(B) (CH2)nOHまたはその塩、あるいは対応するアルコキシドイオン(nは0〜3の整数を示す。)
(C)炭素数1〜20の飽和または不飽和の炭化水素基
上記のような低分子化合物の保護剤の他、次のような高分子保護剤も使用することができる。
<合成例1>
(1)化合物1の合成
下記式で表される化合物1を合成した。
赤外線吸収スペクトル2291cm-1:N≡N+伸縮振動、1728cm-1:C=O伸縮振動、808 cm-1:C−H面外変角振動
テトラクロロ金(III)酸四水和物(1.00g、2.43mmol)をイオン交換水(99.5mL)に溶解させ、N2を15分間フローし、脱気した。N2雰囲気下、化合物1(0.573g、2.43mmol)を加え、5分間攪拌させた後、イオン交換水80.5mLで溶解させた水素化ホウ素ナトリウム(0.0459g、1.21mmol)を5〜10℃、2時間で滴下した。滴下後、1時間攪拌し、紫色分散液が得られた。得られた分散液をろ過、遠心分離、水洗、溶剤洗浄等で精製し、イオン交換水で濃度調整することにより、金ナノ粒子中の金純度97.9質量%、金含有量50g/Lの黒紫色水分散液が得られた。金ナノ粒子の平均粒径は透過型電子顕微鏡(日立製作所製、H-7500)を用いて求めた。
紫外−可視吸収スペクトルのピーク波長550nm、平均粒径19nm、ゼータ電位-34.77mV
テトラクロロ金(III)酸四水和物(0.264g、0.641mmol)をイオン交換水(1490mL)に溶解させ、N2を30分間フローし、脱気した。N2雰囲気下、0.2Mクエン酸ナトリウム水溶液59.6mLを室温下、30分で滴下した。滴下後、80℃で25分攪拌することにより、紫色分散液が得られた。得られた分散液をろ過、遠心分離、水洗、溶剤洗浄等で精製し、イオン交換水で濃度調整することにより、金ナノ粒子中の金純度99.7質量%、金含有量10g/Lの紫色水分散液が得られた。金ナノ粒子の平均粒径は透過型電子顕微鏡(日立製作所製、H-7500)を用いて求めた。
紫外−可視吸収スペクトルのピーク波長529nm、平均粒径15nm、ゼータ電位-42.37mV
テトラクロロ金(III)酸四水和物(0.333g、0.809mmol)をイオン交換水(770mL)に溶解させ、N2を15分間フローし、脱気した。N2雰囲気下、ポリアクリル酸ナトリウム(アルドリッチ社製、分子量〜5100、0.228g)を加え、30分間攪拌させた後、エタノール100mLで溶解させた水素化ホウ素ナトリウム(0.0038g、0.100mmol)を室温下、80分で滴下した。滴下後、1時間攪拌し、紫色分散液が得られた。得られた分散液をろ過、遠心分離、水洗、溶剤洗浄等で精製し、イオン交換水で濃度調整することにより、金ナノ粒子中の金純度93.8質量%、金含有量10g/Lの紫色水分散液が得られた。金ナノ粒子の平均粒径は紫外−可視分光光度計(日本分光製、V-550)を用いて測定した表面プラズモン共鳴のピーク波長から、J. Phys. Chem. C 2007, 111, 14664-14669記載の計算式をもとに算出した。
紫外−可視吸収スペクトルのピーク波長536nm、平均粒径57nm、ゼータ電位-41.80mV
テトラクロロ金(III)酸四水和物(1.00g、2.43mmol)をイオン交換水(2427mL)に溶解させ、N2を30分間フローし、脱気した。N2雰囲気下、ポリビニルピロリドン(和光純薬株式会社製、分子量約40000、0.808g)を加え、30分間攪拌させた後、エタノール303mLで溶解させた水素化ホウ素ナトリウム(0.0114g、0.301mmol)を室温下、80分で滴下した。滴下後、1時間攪拌し、紫色分散液が得られた。得られた分散液をろ過、遠心分離、水洗、溶剤洗浄等で精製し、イオン交換水で濃度調整することにより、金ナノ粒子中の金純度98.4質量%、金含有量10g/Lの紫色水分散液が得られた。金ナノ粒子の平均粒径は透過型電子顕微鏡(日立製作所製、H-7500)を用いて求めた。

テトラクロロ金(III)酸四水和物(0.209g、0.507mmol)をイオン交換水(1015mL)に溶解させ、N2を30分間フローし、脱気した。N2雰囲気下、ポリビニルピロリドン(和光純薬株式会社製、分子量約40000、28.24g)を加え、30分間攪拌させた後、イオン交換水63mLで溶解させた水素化ホウ素ナトリウム(2.38mg、0.0629mmol)を室温下、5分で滴下した。滴下後、1時間攪拌し、赤紫色分散液が得られた。得られた分散液をろ過、遠心分離、水洗、溶剤洗浄等で精製し、イオン交換水で濃度調整することにより、金ナノ粒子中の金純度99.0質量%、金含有量10g/Lの赤紫色水分散液が得られた。金ナノ粒子の平均粒径は透過型電子顕微鏡(日立製作所製、H-7500)を用いて求めた。

テトラクロロ金(III)酸四水和物(0.209g、0.507mmol)をイオン交換水(509mL)に溶解させ、N2を30分間フローし、脱気した。N2雰囲気下、ポリビニルピロリドン(和光純薬株式会社製、分子量約40000、0.564g)を加え、30分間攪拌させた後、イオン交換水63mLで溶解させた水素化ホウ素ナトリウム(2.38mg、0.0629mmol)を室温下、80分で滴下した。滴下後、1時間攪拌し、紫色分散液が得られた。得られた分散液をろ過、遠心分離、水洗、溶剤洗浄等で精製し、イオン交換水で濃度調整することにより、金ナノ粒子中の金純度99.5質量%、金含有量10g/Lの紫色水分散液が得られた。金ナノ粒子の平均粒径は透過型電子顕微鏡(日立製作所製、H-7500)を用いて求めた。

テトラクロロ金(III)酸四水和物(0.167g、0.405mmol)をイオン交換水(813mL)に溶解させ、N2を20分間フローし、脱気した。N2雰囲気下、ポリビニルアルコール(岩井化学薬品製、分子量約80000、0.0352g)を加え、30分間攪拌させた後、エタノール50mLで溶解させた水素化ホウ素ナトリウム(0.0019g、0.0502mmol)を室温下、80分で滴下した。滴下後、1時間攪拌し、紫色分散液が得られた。得られた分散液をろ過、遠心分離、水洗、溶剤洗浄等で精製し、イオン交換水で濃度調整することにより、金ナノ粒子中の金純度72.0質量%、金含有量10g/Lの紫色水分散液が得られた。金ナノ粒子の平均粒径は紫外−可視分光光度計(日本分光製、V-550)を用いて測定した表面プラズモン共鳴のピーク波長から、J. テトラクロロ金(III)酸四水和物(0.314g、0.762mmol)をイオン交換水(765mL)に溶解させ、N2を20分間フローし、脱気した。N2雰囲気下、カルボキシメチルセルロースナトリウム(和光純薬株式会社製、分子量100000〜110000、0.3068g)を加え、30分間攪拌させた後、エタノール94mLで溶解させた水素化ホウ素ナトリウム(0.0036g、0.0952mmol)を室温下、120分で滴下した。滴下後、1時間攪拌し、紫色分散液が得られた。得られた分散液をろ過、遠心分離、水洗、溶剤洗浄等で精製し、イオン交換水で濃度調整することにより、金ナノ粒子中の金純度89.7質量%、金含有量10g/Lの紫色水分散液が得られた。金ナノ粒子の平均粒径は紫外−可視分光光度計(日本分光製、V-550)を用いて測定した表面プラズモン共鳴のピーク波長から、J. Phys. Chem. C 2007, 111, 14664-14669記載の計算式をもとに算出した。
紫外−可視吸収スペクトルのピーク波長537nm、平均粒径59nm、ゼータ電位-22.61mV
テトラクロロ金(III)酸四水和物(0.209g、0.507mmol)をイオン交換水(509mL)に溶解させ、N2を30分間フローし、脱気した。N2雰囲気下、ポリビニルピロリドン(和光純薬株式会社製、分子量約40000、0.564g)を加え、30分間攪拌させた後、エタノール63mLで溶解させた水素化ホウ素ナトリウム(1.192mg、0.0315mmol)を室温下、80分で滴下した。滴下後、20時間攪拌し、紫色分散液が得られた。得られた分散液をろ過、遠心分離、水洗、溶剤洗浄等で精製し、イオン交換水で濃度調整することにより、金ナノ粒子中の金純度99.9質量%、金含有量10g/Lの紫色水分散液が得られた。金ナノ粒子の平均粒径は透過型電子顕微鏡(日立製作所製、H-7500)を用いて求めた。
紫外−可視吸収スペクトルのピーク波長561nm、平均粒径65nm、ゼータ電位-38.63mV
テトラクロロ金(III)酸四水和物(0.042g、0.102mmol)をイオン交換水(25.0mL)に溶解させ、N2を15分間フローし、脱気した。N2雰囲気下、相間移動触媒としてテトラ-n-オクチルアンモニウムブロミド(0.131g、0.240mmol)を溶解したトルエン溶液(25.0mL)を加え、1時間攪拌した。さらに、n−オクチルアミン(0.143g、1.106mmol)を溶解したトルエン溶液(25.0mL)を加え、5分間攪拌した。イオン交換水25.0mLで溶解させた水素化ホウ素ナトリウム(0.059g、1.560mmol)を室温下、30分で滴下した。滴下後、24時間攪拌し、トルエン層を分取することにより、金ナノ粒子が分散した紫色分散液が得られた。得られたトルエン分散液をろ過、遠心分離、水洗、溶剤洗浄等で精製し、トルエンで濃度調整することにより、金含有量10g/Lの紫色分散液が得られた。金ナノ粒子の平均粒径は紫外−可視分光光度計(日本分光製、V-550)を用いて測定した表面プラズモン共鳴のピーク波長から、J. Phys. Chem. C 2007, 111, 14664-14669記載の計算式をもとに算出した。

(1)化合物2の合成
下記式で表される化合物2を合成した。
赤外線吸収スペクトル2917cm-1:C−H伸縮振動、2847cm-1:C−H伸縮振動、2297cm-1:N≡N+伸縮振動、820cm-1:C−H面外変角振動
テトラクロロ金(III)酸四水和物(0.209g、0.507mmol)をイオン交換水(16.0mL)に溶解させ、N2を15分間フローし、脱気した。N2雰囲気下、化合物2(0.337g、1.014mmol)を溶解したトルエン溶液(30.0mL)を加え、2時間攪拌させた後、トルエン層を分取し、水洗を2回行った。イオン交換水8.0mLで溶解させた水素化ホウ素ナトリウム(0.0461g、1.219mmol)を室温下、40分で滴下した。滴下後、1時間攪拌し、トルエン層を分取することにより、金ナノ粒子が分散した紫色分散液が得られた。得られたトルエン分散液をろ過、遠心分離、水洗、溶剤洗浄等で精製し、トルエンで濃度調整することにより、金含有量10g/Lの紫色分散液が得られた。金ナノ粒子の平均粒径は紫外−可視分光光度計(日本分光製、V-550)を用いて測定した表面プラズモン共鳴のピーク波長から、J. Phys. Chem. C 2007, 111, 14664-14669記載の計算式をもとに算出した。

特許文献4(特開2008−069097号公報)実施例2の方法を参考にして白金ナノ粒子を合成した。ヘキサクロロ白金(IV)酸六水和物(0.345g、0.666mmol)をイオン交換水(500mL)に溶解させ、N2を10分間フローし、脱気した。N2雰囲気下、ポリアクリル酸ナトリウム(アルドリッチ社製、分子量〜5100、7.83g)を加え、30分間攪拌させた後、エタノール500mLを加え、N2雰囲気下を保ちながら、100℃、2時間攪拌し、黒色分散液が得られた。得られた分散液をろ過、遠心分離、水洗、溶剤洗浄等で精製し、イオン交換水で濃度調整することにより、白金ナノ粒子中の白金純度64.8質量%、白金含有量10g/Lの黒色水分散液が得られた。白金ナノ粒子の平均粒径は透過型電子顕微鏡(日立製作所製、H-7500)を用いて求めた。
紫外−可視吸収スペクトルのピーク波長239nm、平均粒径4nm、ゼータ電位-41.37mV
特許文献4(特開2008−069097号公報)実施例1の方法を参考にして白金ナノ粒子を合成した。ヘキサクロロ白金(IV)酸六水和物(0.344g、0.664mmol)をイオン交換水(500mL)に溶解させ、N2を10分間フローし、脱気した。N2雰囲気下、ポリビニルピロリドン(和光純薬株式会社製、分子量約40000、2.934g)を加え、30分間攪拌させた後、エタノール500mLを加え、N2雰囲気下を保ちながら、100℃、2時間攪拌し、黒色分散液が得られた。得られた分散液をろ過、遠心分離、水洗、溶剤洗浄等で精製し、イオン交換水で濃度調整することにより、白金ナノ粒子中の白金純度95.8質量%、白金含有量10g/Lの黒色水分散液が得られた。白金ナノ粒子の平均粒径は透過型電子顕微鏡(日立製作所製、H-7500)を用いて求めた。
紫外−可視吸収スペクトルのピーク波長249nm、平均粒径7nm、ゼータ電位-36.19mV
硝酸銀(0.2592g、1.53mmol)をイオン交換水(150mL)に溶解させ、N2を20分間フローし、脱気した。N2雰囲気下、化合物1(0.108g、0.458mmol)を加え、5分間攪拌させた後、イオン交換水45mLで溶解させた水素化ホウ素ナトリウム(0.0058g、0.153mmol)を5〜10℃、2時間で滴下した。滴下後、1時間攪拌し、黒黄色分散液が得られた。得られた分散液をろ過、遠心分離、水洗、溶剤洗浄等で精製し、イオン交換水で濃度調整することにより、銀ナノ粒子中の銀純度94.0質量%、銀含有量10g/Lの黒黄色水分散液が得られた。銀ナノ粒子の平均粒径は透過型電子顕微鏡(日立製作所製、H-7500)を用いて求めた。
紫外−可視吸収スペクトルのピーク波長420nm 平均粒径16nm
[細胞毒性試験]
線維芽細胞L929およびマクロファージ細胞RAW264.7を理研バイオリソースセンターから入手した。細胞の培養はα-MEMに10% FBSを添加した培地を用いて行なった。細胞を1×104個/wellになるように96穴プレートに200μL播種し、37℃、5% CO2条件下で72時間培養し、合成例1〜8の各金ナノ粒子分散液を表1に示す濃度で添加し、24時間培養した。細胞をPBS(リン酸緩衝生理食塩水)で2回洗浄した後、培地を加えたところに細胞毒性測定用試薬(Cell Counting Kit-8、同仁化学研究所)を添加し、37℃で1時間反応を行なった。反応後、分取した上澄みの吸光度(450nm)を測定し、細胞生存率を算出した。結果を表1に示す。なお、細胞生存率90%以上を細胞毒性なし、90%未満を細胞毒性ありとして、細胞毒性を評価した。
実施例1〜8と同様にして、合成例15〜17の白金および銀ナノ粒子分散液を表1に示す濃度で添加し細胞毒性試験を行ない、細胞生存率を算出した。結果を表1に示す。
[遺伝毒性試験]
実施例1〜8と同様に合成例1、2、5、6、9の各金ナノ粒子分散液を表2に示す濃度で添加し培養した線維芽細胞L929およびマクロファージ細胞RAW264.7について、コメットアッセイキット(Trevigen製)を用いて試験を行った。具体的には、細胞をPBS(リン酸緩衝生理食塩水)で洗浄した後、コメットスライド上で低融点アガロースゲルと混和し、細胞溶解液(0.3M NaCl, pH 7.5)中で20分間溶解した。21V、300mAで30分間、電気泳動を行った後、スライドをバッファーおよび無水エタノールで5分間洗浄した。サイバーグリーン色素(Invitrogen製)でDNA染色した後、蛍光顕微鏡(ニコン製、TS1200-E)を用いて細胞を観察し、画像解析によりDNA損傷を受けている細胞数の解析を行った。結果を表2に示す。なお、金ナノ粒子を添加していない細胞で行った試験(コントロール試験)のDNA損傷率を100%とし、DNA損傷率110%以下を遺伝毒性なし、111%以上を遺伝毒性ありとして、遺伝毒性を評価した。
実施例9〜13と同様にして、合成例15の白金ナノ粒子分散液を表2に示す濃度で添加し培養した線維芽細胞およびマクロファージ細胞について、遺伝毒性試験を行ない、DNA損傷率を算出した。結果を表2に示す。
[MMP活性測定試験]
MMP−1、MMP−2、MMP−8、MMP−9の活性測定キット(ANASPEC製)を用いて、96穴プレートにアッセイバッファー130μL、基質溶液50μL、合成例1〜9の各金ナノ粒子分散液を表3に示す濃度で加えたところに、MMP溶液20μLを添加した。30分間反応した後、蛍光強度をPerkinElmer社製 1420 Multilabel Counter ARVO MX(測定波長 485/535nm)を用いて測定を行ない、MMP−1、MMP−2、MMP−8、MMP−9それぞれの阻害率を算出した。結果を表3に示す。
実施例14〜22と同様にして、合成例15〜17の白金および銀ナノ粒子分散液を表3に示す濃度で添加しMMP−1活性測定試験を行ない、MMP−1の阻害率を算出した。結果を表3に示す。
[樹脂組成物の弾性率測定]
合成例1〜4、8、10〜14の各金ナノ粒子分散液をろ過、遠心分離、減圧留去、減圧乾燥等の操作を行なうことにより、金ナノ粒子の乾燥微粒子を得た。組成物Iとして、Bis-GMA(ビスフェノールA・グリシジルメタクリル酸)30質量%、TEDMA(トリエチレングリコールジメタクリレート)70質量%、BPO(過酸化ベンゾイル)1質量%を混合したところに、得られた乾粉微粒子5質量%を添加した。また、組成物IIとして、Bis-GMA 30質量%、TEDMA 70質量%、DMPT(N,N-ジメチル-p-トルイジン)0.5質量%を混合した。熱分析用サンプルパン内で組成物IおよびIIを1:1(質量%)の比率で混合した後、窒素雰囲気下、23℃、30分で硬化した。硬化したサンプルについて、熱機械分析装置(セイコーインスツルメンツ製、TMA/SS6200)を用いて、25℃、0〜1400mN、100mN/min条件で測定を行ない、得られた応力・歪曲線から弾性率を求めた。結果を表4に示す。
実施例23〜32と同様にして、比較例8では、合成例15の白金ナノ粒子を用いて、比較例9では、合成例17の銀ナノ粒子を用いて、比較例10では、金属ナノ粒子を添加せずに、樹脂組成物の弾性率測定を行なった。結果を表4に示す。
[DPPHラジカル消去法による抗酸化作用の評価]
DPPH(1,1-ジフェニル-2-ピクリルヒドラジル)ラジカル試薬を溶解したエタノール溶液に、合成例1〜4、7、8の各金ナノ粒子分散液を表5に示す濃度で添加した。48時間後、遠心分離により金ナノ粒子を取り除いた後、紫外−可視分光光度計(日本分光製、V-550)による測定および目視による色差により抗酸化作用を判定した。結果を表5に示す。
Claims (2)
- マトリックスメタロプロテアーゼ(MMP)阻害剤であって、
金ナノ粒子を含み、
前記金ナノ粒子は、平均粒径が10〜30nmであり、かつ、保護剤を表面に有し、
前記保護剤は、マトリックスメタロプロテアーゼの酵素活性部位の亜鉛イオンと配位結合可能であり、かつ、N、P、O、およびSから選択される電子供与原子を有する官能基を含み、分子量が258以下であり、
前記マトリックスメタロプロテアーゼが、MMP-1、MMP-2、MMP-8、MMP-9である
マトリックスメタロプロテアーゼ阻害剤。 - 前記保護剤は、前記金ナノ粒子に共有結合する官能基と、前記マトリックスメタロプロテアーゼの亜鉛イオンと配位結合可能な官能基とを含む、請求項1に記載のマトリックスメタロプロテアーゼ阻害剤。
