Description
(1)以下の(a)又は(b)で示す遺伝子で構成されるオキシトシン感受性予測マーカー。
(a)配列番号2、4、6、8、及び10で示すアミノ酸配列からなる群から選択されるタンパク質をコードする遺伝子
(b)配列番号12で示すアミノ酸配列からなるタンパク質をコードする遺伝子
(2)前記(a)に記載の配列番号2、4、6、8、及び10で示すアミノ酸配列からなる群から選択されるタンパク質をコードする遺伝子において、それぞれcg12513795、cg27278382、cg19104656、cg16553574、及びcg04861640で示されるメチル化標的部位のシトシンがメチル化された遺伝子であって、オキシトシンに対する高感受性を予測することのできる、(1)に記載のオキシトシン感受性予測マーカー。
(3)前記(a)に記載の配列番号2、4、6、8、及び10で示すアミノ酸配列からなるタンパク質をコードする遺伝子が、それぞれ配列番号1、3、5、7、及び9で示す塩基配列からなる、(1)又は(2)に記載のオキシトシン感受性予測マーカー。
(4)前記(b)に記載の遺伝子の塩基配列において、rs53576、rs11706648、rs17049528、rs4686300、rs237900、rs2254298、rs11131149、rs2268495、rs237887、及びrs237878からなる群から選択される1又は2以上のSNPがマイナーアレルである、(1)に記載のオキシトシン感受性予測マーカー。
(5)前記rs53576、rs11706648、rs17049528、rs4686300、及びrs237900からなる群から選択されるSNPがそれぞれGG、AA、GG、CC、及びGGではない遺伝子であって、オキシトシンに対する低感受性を予測することのできる、(4)に記載のオキシトシン感受性予測マーカー。
(6)前記rs2254298、rs11131149、rs2268495、rs237887、及びrs237878からなる群から選択されるSNPがGG、GG、GG、GG、及びTTではない遺伝子であって、オキシトシンに対する高感受性を予測することのできる、(4)に記載のオキシトシン感受性予測マーカー。
(7)前記(b)に記載の配列番号12で示すアミノ酸配列からなるタンパク質をコードする遺伝子が配列番号11で示す塩基配列からなる、(1)、及び(4)〜(6)のいずれかに記載のオキシトシン感受性予測マーカー。
(8)被験者のオキシトシンに対する感受性を予測する方法であって、前記被験者由来の試料からゲノムDNAを抽出する工程、抽出したゲノムDNAにおいて配列番号2、4、6、8、及び10で示すアミノ酸配列からなる群から選択される一以上のタンパク質をコードする遺伝子の、それぞれcg12513795、cg27278382、cg19104656、cg16553574、及びcg04861640で示すメチル化標的部位のメチル化の有無を検出する工程、前記遺伝子において前記メチル化標的部位がメチル化されているときに前記被験者のオキシトシンに対する感受性が有意に高いと予測する工程を含む前記方法。
(9)配列番号2、4、6、8、及び10で示すアミノ酸配列からなるタンパク質をコードする遺伝子が、それぞれ配列番号1、3、5、7、及び9で示す塩基配列からなる(8)に記載の方法。
(10)被験者のオキシトシンに対する感受性を予測する方法であって、前記被験者由来の試料からゲノムDNAを抽出する工程、抽出したゲノムDNAにおいて配列番号12で示すアミノ酸配列からなるタンパク質をコードする遺伝子の、rs53576、rs2254298、rs11706648、rs2268495、rs11131149、rs17049528、rs4686300、rs237878、rs237887、及びrs237900からなる群から選択される1又は2以上のSNPを同定する工程、前記SNPがマイナーアレルであったときに前記被験者のオキシトシンに対する感受性が有意に高い又は低いと予測する工程を含む前記方法。
(11)前記遺伝子が配列番号11で示す塩基配列からなる、(10)に記載の方法。
(12)前記感受性を予測する工程において、rs53576、rs11706648、rs17049528、rs4686300、及びrs237900からなる群から選択されるSNPがそれぞれGG、AA、GG、CC、及びGGではないときに前記被験者のオキシトシンに対する感受性が有意に低いと予測する、(10)又は(11)に記載の方法。
(13)前記感受性を予測する工程において、rs2254298、rs11131149、rs2268495、rs237887、及びrs237878からなる群から選択されるSNPがそれぞれGG、GG、GG、GG、及びTTではないときに前記被験者のオキシトシンに対する感受性が有意に高いと予測する、(10)又は(11)に記載の方法。
(14)自閉スペクトラム症患者のオキシトシン投与による治療効果を推定する方法であって、前記(8)、(9)又は(13)に記載のオキシトシン感受性予測方法における感受性を予測する工程で、被験者のオキシトシンに対する感受性が有意に高いと予測されたときに、被験者である自閉スペクトラム症患者のオキシトシン投与による治療効果が期待できると推定する工程を含む前記方法。
(15)自閉スペクトラム症患者のオキシトシン投与による治療効果を推定する方法であって、前記(12)に記載のオキシトシン感受性予測方法における感受性を予測する工程で、被験者のオキシトシンに対する感受性が有意に低いと予測されたときに、被験者である自閉スペクトラム症患者のオキシトシン投与による治療効果が期待できないと推定する工程を含む前記方法。
1−1.概要
本発明の第1の態様は、オキシトシン感受性予測マーカーである。本発明のオキシトシン感受性予測マーカーは、オキシトシンに対する感受性と有意に相関する部位を有する遺伝子で構成されるバイオマーカーである。被験者から当該マーカーを検出することで、その被験者のオキシトシンに対する感受性を予測でき、またその感受性の予測結果から被験者におけるオキシトシンのASDに対する治療効果を予測することが可能となる。
本明細書において「オキシトシン感受性予測マーカー」とは、被験者のオキシトシンに対する感受性を予測できるバイオマーカーをいう。その実体は、後述する特定の構造を有する遺伝子で構成される。
本発明のオキシトシン感受性予測マーカーは、特定のメチル化標的部位のメチル化を指標とする5種のメチル化指標遺伝子、及び特定位置のSNPが特定アレルの塩基配列であることを指標とするSNP指標遺伝子の2群で構成される。以下、各群の遺伝子について、具体的に説明をする。
オキシトシン感受性予測マーカーの第1群は、メチル化指標遺伝子で構成される。
「メチル化指標遺伝子」は、5種の遺伝子、すなわちC3orf59遺伝子、UQCRC1遺伝子、VARS2遺伝子、ZNF92遺伝子、及びZSCAN26遺伝子からなる。
本明細書においてメチル化指標遺伝子は、エクソン及びイントロンからなる実質的な遺伝子領域のみならず、非翻訳領域(5’UTR及び3’UTRを含む)を含む転写領域、及びその発現制御領域(エンハンサー、プロモーター、CpGアイランド、及びshore領域を含む)を包含する1つの遺伝子発現単位を意味する。ここで、「shore領域」とは、CpGアイランドの両側に位置するそれぞれ2000bpまでの領域をいう。各メチル化指標遺伝子は、その塩基配列中にオキシトシンの感受性及びASD改善度と強い相関性を示すメチル化標的部位を包含している。したがって、被験者のゲノムDNA上の各遺伝子における当該メチル化標的部位のシトシンのメチル化を指標として、その被験者のオキシトシン感受性を予測することができる。具体的には、メチル化標的部位のシトシンがメチル化されていた場合、その被験者は、オキシトシン感受性が高いと予測される。それ故、被験者がASD患者であれば、オキシトシン投与による治療効果が期待できると推定できる。
(1)C3orf59遺伝子
C3orf59(chromosome 3 open reading frame 59)遺伝子は、MB21D2(Mab-21 domain containing 2)遺伝子とも呼ばれ、ヒトでは第3染色体上に存在し、配列番号2で示すアミノ酸配列からなる機能未知のC3orf59タンパク質をコードする。
UQCRC1(ubiquinol-cytochrome c reductase core protein 1)遺伝子は、ヒトでは第3染色体上に存在し、配列番号4で示すアミノ酸配列からなるUQCRC1タンパク質、すなわちユビキノール-シトクロムcレダクターゼコアタンパク質1をコードする。ユビキノール-シトクロムcレダクターゼは、ミトコンドリアの電子伝達系において第3番目の複合体(complex III)として機能し、UQCRC1タンパク質は、そのサブユニットの一つを構成している。
VARS2(valyl-tRNA Synthetase 2)遺伝子は、ヒトでは第6染色体上に存在し、配列番号6で示すアミノ酸配列からなるVARS2タンパク質、すなわちミトコンドリアアミノアシルtRNA合成酵素2(mitochondrial aminoacyl-tRNA synthetase2)をコードする。ミトコンドリアアミノアシルtRNA合成酵素2は、バリンをtRNA(Val)にエステル結合させてアミノアシルtRNA(Val)を合成する反応を触媒する。
ZNF92(zinc finger protein 92)遺伝子は、ヒトでは第7染色体上に存在し、配列番号8で示すアミノ酸配列からなるZNF92タンパク質、すなわちジンクフィンガータンパク質92をコードしている。ZNF92タンパク質は、亜鉛(Zn)と錯体を形成するタンパク質の1種で、亜鉛は、オキシトシンの安定性に関与することが報告されていることから、この遺伝子のメチル化はASDの病態形成と密接に関係することが予測されている。
ZSCAN26(zinc finger and SCAN domain containing 26)遺伝子は、ZNF187(zinc finger protein 187)遺伝子とも呼ばれる。ヒトでは第6染色体上に存在し、配列番号10で示すアミノ酸配列からなるZSCAN26タンパク質をコードしている。ZSCAN26タンパク質は、ZNF92タンパク質と同様に、亜鉛(Zn)と錯体を形成するタンパク質の1種であり、この遺伝子のメチル化はASDの病態形成と密接に関係することが予測されている。
オキシトシン感受性予測マーカーの第2群は、SNP指標遺伝子で構成される。
本明細書における「SNP指標遺伝子」は、オキシトシン受容体遺伝子(oxytocin receptor:OXTR)(本明細書では、しばしば「OXTR遺伝子」と表記する)である。OXTR遺伝子は、7回膜貫通型Gタンパク質共役受容体で、オキシトシンの結合により、細胞内でGpタンパク質と結合して細胞内シグナル伝達を活性化する機能を有する。ヒトでは第3染色体上に存在し、配列番号12で示すアミノ酸配列からなる。OXTR遺伝子の塩基配列には、例えば、配列番号11で示す配列が挙げられる。
前記各SNPが集団内に多数の割合で存在するメジャーアレル、又は少数の割合で存在するマイナーアレルのいずれであるかを指標として、OXTR遺伝子内にそのSNPを有する被験者のオキシトシンに対する感受性が高いか、又は低いかを予測することができる。各SNPのメジャーアレルは、表1に示す塩基となる。例えば、rs53576のメジャーアレルはGGである(本明細書では「rs53576GG」と表記する。他のアレルについても同様とする。)。
2−1.概要
本発明の第2の態様は、被験者におけるオキシトシン感受性予測方法である。本発明のオキシトシン感受性予測方法は、医師の介在を必要とすることなく、第1態様に記載のオキシトシン感受性予測マーカーを指標として、被験体のオキシトシンに対する感受性を予測することができる方法であり、医療行為を補助する方法である。
本発明のオキシトシン感受性予測方法は、使用するオキシトシン感受性予測マーカー群、すなわちメチル化指標遺伝子及びSNP指標遺伝子によって以下の3つの方法に分けることができる。
本方法は、ゲノムDNA上のメチル化指標遺伝子のメチル化頻度を測定することで、被験者のオキシトシン感受性を予測する方法である。
「ゲノムDNA抽出工程」とは、被験者由来の試料からゲノムDNAを抽出する工程をいう。
本明細書において「試料」とは、ゲノムDNAを含み得る生物学的試料をいう。例えば、細胞(組織、器官を含む)及び細胞を含み得る体液が該当する。本明細書において「体液」とは、個体から直接採取される流体試料をいう。例えば、血液(全血、血清、血漿及び間質液を含む)、リンパ液、脳脊髄液、神経根周囲液、滑液、涙液、鼻汁、唾液、尿、汗、乳、痰、膣液、精液、胸水、腹水等が該当する。本発明の試料は、いずれの細胞又は体液であってもよいが、好ましくは個体から採取する際に侵襲性の低い細胞又は体液である。例えば、細胞であれば、口腔粘膜、鼻粘膜、膣粘膜及び腸粘膜を構成する上皮細胞、毛母細胞、及び角質細胞が挙げられる。また、体液であれば、血液、唾液、鼻汁、痰、膣液、及び精液が挙げられる。本発明の方法に必要なゲノムDNAを容易かつ十分量抽出できる点で特に好ましい試料は、血液である。血液は末梢血でよい。
「メチル化検出工程」とは、前記ゲノムDNA抽出工程で得られたゲノムDNAにおいて、第1態様に記載のオキシトシン感受性予測マーカーであるメチル化指標遺伝子、すなわちC3orf59遺伝子、UQCRC1遺伝子、VARS2遺伝子、ZNF92遺伝子、及びZSCAN26遺伝子のいずれか1又は2以上の遺伝子について、第1態様に記載の特定のメチル化標的部位におけるシトシンのメチル化の有無を検出する工程である。2以上、3以上、4以上、又は全てのメチル化指標遺伝子について、特定のメチル化標的部位のメチル化を調べることは好適である。特定のメチル化標的部位がメチル化されたメチル化指標遺伝子が多いほど、換言すればメチル化指標遺伝子のメチル化頻度が高いほど、被験者に対するオキシトシン感受性の予測精度を向上できるからである。
メチル化指標遺伝子を用いた方法において「感受性予測工程」とは、前記メチル化検出工程の検出結果から、前記被験者のオキシトシンに対する感受性を予測する工程である。
本方法は、OXTR遺伝子のSNP情報を解析することで、被験者のオキシトシンに対する感受性を予測する方法である。
「ゲノムDNA抽出工程」とは、被験者由来の試料から核酸を抽出する工程をいう。本工程の核酸は、ゲノムDNAに限らずRNAを包含する。抽出するRNAは、OXTR mRNAを含むRNAであれば、総RNA及びmRNA(pre-mRNA及びmature mRNAを含む)を問わない。
「SNP同定工程」とは、前記核酸抽出工程で得られた核酸において、第1態様に記載のオキシトシン感受性予測マーカーであるOXTR遺伝子上の特定のSNPを同定する工程である。同定すべきSNPは、rs53576、rs2254298、rs11706648、rs2268495、rs11131149、rs17049528、rs4686300、rs237878、rs237887、及びrs237900からなる群から選択される1又は2以上のSNPセットである。複数のSNPセットを組み合わせて同定することで、オキシトシン感受性の予測精度が向上し、またオキシトシン投与によるASDの治療効果との関連性が有意に高くなるからである。SNPの組み合わせは、例えば、rs53576とrs2254298の組み合わせ、rs53576とrs11706648の組み合わせ、rs53576とrs2268495の組み合わせ、rs11706648とrs2254298の組み合わせ、rs11706648とrs2268495の組み合わせ、rs2254298とrs2268495の組み合わせ、rs53576、rs11706648及びrs2254298の組み合わせ、rs53576、rs11706648及びrs2268495の組み合わせ、rs11706648、rs2268495及びrs2254298の組み合わせ、そしてrs53576、rs11706648、rs2268495及びrs2254298の組み合わせが挙げられるが、これらに限定はされない。上記全てのSNPセットの組み合わせであってもよい。
SNP指標遺伝子を用いた方法において「感受性予測工程」とは、メチル化指標遺伝子を用いた方法における「感受性予測工程」とは異なり、前記SNP同定工程の結果から、前記被験者のオキシトシンに対する感受性を予測する工程である。
本方法は、前述のメチル化指標遺伝子を用いた方法とSNPを用いた方法を組み合わせた方法である。2つのオキシトシン感受性予測方法を組み合わせることで、被験者のオキシトシン感受性に関して、より精度の高い予測を行うことができる。
本発明の第3の態様は、第2態様のオキシトシン感受性予測方法に従属する方法であって、第2態様の予測結果から、ASD患者にオキシトシンを投与したときの治療効果を事前に推定する。
「治療効果推定工程」とは、第2態様のオキシトシン感受性予測方法における感受性予測工程の予測結果から、ASD患者のオキシトシン投与による治療効果が期待できるか、又はできないかを推測する工程である。
本態様において、対象となる被験者は、ASD患者である。
(目的)
オキシトシン投与によるASD患者の治療効果と相関するメチル化指標遺伝子を探索する。
ゲノムDNA上の全メチル化部位(メチル化標的部位)に関して、オキシトシン投与によるASD患者の治療効果と相関するメチル化標的部位を網羅的に探索し、有意性が認められるメチル部位が位置する遺伝子をオキシトシン感受性予測マーカーとして単離した。
オキシトシンの反復経鼻投与によるASD患者における社会相互性の改善を図1及び表3に、また社会相互性の改善度と有意に相関するメチル化標的部位を図2に、示す。
(目的)
オキシトシン投与によるASD患者の治療効果と相関するOXTR遺伝子におけるSNPを探索する。
SNPは、全ゲノム中に存在する60万箇所のSNPデータバンクに含まれ、OXTR遺伝子上に存在する全SNPと、前記SNPデータバンクには含まれていないがASDや社会行動との関係がしばしば報告されているOXTR遺伝子上のSNPの、計22SNPを対象とした。表4に本実施例の対象とした22箇所のSNPを示す。
Differenceij=min( |SNPi−SNPj|, Nparticipant−|SNPi−SNPj|)
を用いて多重共線性を調べた。ここで、Differenceijは、SNPパターンi及びj間の違いを示す。また、Nparticipantは、実験対象者数(ここでは38)を示す。Differenceijが2以下のSNPを1つのパターンにまとめたところ、残った16 SNPのうち2 SNP(表4のNo.1、2)を1つのパターンにまとめることができた。回帰分析によるこの方法は、目的とするSNP間の連鎖不平衡(LD)の推定と実質的に同じである。
図3及び表1に結果を示す。
図3に示すオキシトシン投与に対する6つの応答指標(a〜f)のいずれかにおいて、破線で示すチャンスレベルよりも有意に高い10 SNPをオキシトシン感受性予測における有用なSNPとして選択した(表1)。これらの10 SNPは、比較的偏りなく分布し、互いに実質的に異なっていた(Differenceij≧7, variance infration factor<1.9)。OXTR遺伝子におけるこれらのSNPは、被験者のオキシトシン感受性と有意に相関する指標となり得ることが明らかとなった。
(目的)
実施例2で選択されたOXTR遺伝子上の10 SNPを組み合わせたときに被験者におけるオキシトシン効果を正確に予測し得るかを検証する。
実施例2で選択された10 SNPを組み合わせて前記6つのオキシトシン応答指標についてオキシトシンの予測効果をSVR(Support Vector Regression)(Bishop CM. Pattern Recognition and Machine Learning. Springer Verlag, 2006.)によって算出し、オキシトシン感受性の高い当事者をそれが低い当事者から識別する事を試みた。
図4及び図5A及び図5Bに結果を示す。
図4のAで示すように、6つのオキシトシン応答指標について回帰分析は有意な精度で実験的に観察された効果を予測することができ(PBonferroni-corrected<0.05 in F tests, adjusted R2>0.27)、予測効果と実測効果の間で有意に高い相関性が認められた。また、Aで示した回帰分析に基づいて、当事者をオキシトシン応答群と応答の比較的弱い群に分類するための機械学習法に基づく予測精度を計算した。その結果、Cで示すように、全ての応答指標において、分類精度は有意に高かった(PBonferroni-corrected P<0.05)。
(目的)
15 SNPの機能的な影響を調べるため、回帰分析中の10 SNPがメジャーアレルであるかマイナーアレルであるかによって、オキシトシン効果を増強するか、低減するかを検証した。
例えば、仮にrs53576がACC活性に関して直線回帰においてポジティブな重みを割り当てられた場合、GGはrs53576のメジャーアレルであり、GA/AAアレルよりも大きなダミー値で表されることからrs53576GGを有する個体は、オキシトシン投与後にACC活性が増加することを示すはずである。
図6に結果を示す。(a)〜(f)は、オキシトシンに対する6つの応答指標を示している。全てのSNPで、0値に対して有意差が認められた(t(3)>7.6、PBonferroni-corrected<0.05)。注目すべき点として、6つの応答指標はそれぞれ独立した異なる指標であるにもかかわらず、各SNPの性質は全ての応答指標で同じであった。例えば、rs53576は、6つの応答指標の全てで常にポジティブな値を示した。これは、rs53576のメジャーアレルであるrs53576GGを有する個体がマイナーアレルであるrs53576GA/AAを有する個体よりも行動レベル及び神経レベルの両方でオキシトシンに対して応答性が高いことを示唆している。一方、rs2254298は、応答指標で常にネガティブな値を示した。これはマイナーアレルであるrs2254298GA/AAを有する個体がメジャーアレルであるrs2254298GGを有する個体よりもオキシトシンに対して応答性が高いことを示唆している。

- 以下の(a)〜(f)で示す遺伝子のメチル化標的部位におけるシトシンがメチル化されているとき、オキシトシンに対する感受性が高いと予測する、オキシトシン感受性を予測するためのバイオマーカーとしての前記遺伝子の使用。
(a)配列番号2で示すアミノ酸配列からなるタンパク質をコードする遺伝子のcg12513795で示されるメチル化標的部位、
(b)配列番号4で示すアミノ酸配列からなるタンパク質をコードする遺伝子のcg2727838で示されるメチル化標的部位、
(c)配列番号6で示すアミノ酸配列からなるタンパク質をコードする遺伝子のcg19104656で示されるメチル化標的部位、

抽出したゲノムDNAにおいて配列番号2、4、6、8、及び10で示すアミノ酸配列からなる群から選択される一以上のタンパク質をコードする遺伝子の、それぞれcg12513795、cg27278382、cg19104656、cg16553574、及びcg04861640で示すメチル化標的部位のメチル化の有無を検出する工程、
前記遺伝子において前記メチル化標的部位がメチル化されているときに前記被験者のオキシトシンに対する感受性が有意に高いと予測する工程
を含む前記方法。 - 配列番号2、4、6、8、及び10で示すアミノ酸配列からなるタンパク質をコードする遺伝子が、それぞれ配列番号1、3、5、7、及び9で示す塩基配列からなる、請求項6に記載の方法。
- 被験者のオキシトシンに対する感受性を予測する方法であって、
前記被験者由来の試料から核酸を抽出する工程、
抽出したゲノムDNAにおいて配列番号12で示すアミノ酸配列からなるタンパク質をコードする遺伝子の、rs53576、rs2254298、及びrs11706648からなる群から選択される1又は2以上のSNPを同定する工程、及び
前記rs53576で示すSNPがGGのとき、
前記rs11706648で示すSNPがAAのとき、又は
前記rs2254298で示すSNPがGG以外のとき、
オキシトシンに対する感受性が有意に高いと予測する工程
を含む前記方法。 - 前記遺伝子が配列番号11で示す塩基配列からなる、請求項8に記載の方法。
- 自閉スペクトラム症患者のオキシトシン投与による治療効果を推定する方法であって、請求項8又は9に記載のオキシトシン感受性予測方法における感受性を予測する工程で、被験者のオキシトシンに対する感受性が有意に高いと予測されたときに、被験者である自閉スペクトラム症患者のオキシトシン投与による治療効果が期待できると推定する工程を含む前記方法。
