1.3〜5MUの通常型IFN−α、このIFN−αは、患者の忍容性に応じて調節することができ、1週間に3回または2日1回、6カ月間皮下注射する。
2.180μgのポリエチレングリコール−IFN−α 2aを1週間に1回1年間皮下注射する。
3.1.0〜1.5μg/kgのポリエチレングリコール−IFN−α 2bを1週間に1回1年間皮下注射する。
4.ラミブジン(LAM):100mgのラミブジンを1日1回経口投与する。
5.アデホビルジピボキシル(ADV):10mgのアデホビルジピボキシルを1日1回経口投与する。
6.エンテカビル(ETV):0.5mgのエンテカビルを1日1回経口投与する。
7.テルビブジン(LdT):600mgのテルビブジンを1日1回経口投与する。
8.フマル酸テノホビルジソプロキシル(TDF):TDF(中国において未承認)は、アデホビルジピボキシルと類似する構造を有するが、腎臓に対する毒性が少ない。治療用量は、日量300mgである。
組換えB型肝炎ワクチンと組み合わせたGM−CSFの免疫プロトコルは、組換えB型肝炎サブユニットワクチンに対して、C57BL/6の免疫応答を向上させる。
材料:遺伝子操作された(CHO)B型肝炎ワクチン(組換えB型肝炎ワクチン(CHO)) 10μg/ml、注射用の組換えヒト顆粒球/マクロファージコロニー刺激因子 300μg/バイアル、CHOにより発現された遺伝子操作された(CHO)B型肝炎ワクチン保存液(HBsAg保存液)、これらは全て、North China Pharmaceutical Group、Jintan BioTechnology Co.,Ltdにより提供される。
(1)抗原コーティング:96ウェルマイクロプレート上に、1ug/mlの抗原を100ul/ウェルでコートし、4℃で一晩置いた。
(2)ブロッキング:プレートを、PBST(PBSに溶解した0.05%Tween20)で、各回5分間継続させて3回洗浄した。ついで、5%スキムミルクを、100μl/ウェルで37℃において1時間使用して、プレートをブロックした。
(3)血清添加:プレートを、PBSTで各回5分間継続させて3回洗浄した。未処置のマウス血清を対照として使用して、2倍系列希釈したマウス血清を100μl/ウェルで添加した。ついで、プレートを37℃で1時間インキュベートした。
(4)二次抗体添加:プレートを、PBSTで各回5分間継続させて3回洗浄した。各ウェルに、HRP標識ヤギ抗マウスIgG(1:1000)を100μl/ウェルで添加した。ついで、プレートを37℃で1時間インキュベートした。
(5)発色:プレートを、PBSTで各回5分間継続させて3回洗浄した。基質であるTMBを100μl/ウェルで添加して、発色させた。ついで、プレートを、37℃において暗所に15分間置き、発色させた。
(6)反応停止:0.2M H2SO4を、50μl/ウェルで添加して、発色を停止させた。
(7)読取り:光学密度をOD450nm/620nmで測定した。サンプルウェルのOD値が対照ウェルの2倍である場合、陽性と考えられる。
(1)抗原コーティング:96ウェルマイクロプレート上にウサギIgG(2μg/ml)およびVP1抗原(1μg/ml)を100μl/ウェルでコートした。ついで、プレートを、4℃で一晩置いた。
(2)ブロッキング:プレートを、PBST(PBSに溶解させた0.05% Tween20)で、各回5分間継続させて3回洗浄した。3% BSA溶液を100μl/ウェルで添加して、プレートを37℃で1時間ブロックした。
(3)血清添加:プレートを、PBSTで各回5分間継続させて3回洗浄した。マウス抗ウサギIgGを20ng/mlから10回連続2倍系列希釈で希釈した。マウス血清を1:100で希釈した。ついで、両方を、100μl/ウェルで、ウェルにトリプリケートで添加した。ついで、プレートを、37℃で1時間インキュベートした。
(4)二次抗体添加:プレートを、PBSTで各回5分間継続させて3回洗浄した。HRP標識ヤギ抗マウスIgG1およびIgG2a(1:1000)を、100μl/ウェルでウェルに添加した。ついで、プレートを、37℃で1時間インキュベートした。
(5)発色:プレートを、PBSTで各回5分間継続させて3回洗浄した。基質であるTMBを、100μl/ウェルで添加して、発色させた。ついで、プレートを、37℃において暗所に15分間置き、発色させた。
(6)反応停止:0.2M H2SO4を、50μl/ウェルで添加して、発色を停止させた。
(7)読取り:光学密度を、OD450nm/620nmで測定した。サンプルウェルのOD値が対照ウェルの2倍である場合、陽性と考えられる。
(8)検量線をプロットし、抗体量を算出した。
T細胞のin vitro増殖活性を、MTTにより測定した。
(1)本実験に使用した全ての材料を、予め滅菌しておく必要がある。
(2)マウスを脱臼により屠殺にし、70%エタノールに15分間浸漬させた。
(3)マウスの脾臓を、無菌条件下において超清浄作業台上で取り出し、UV光により20分間予め滅菌しておき、2ml RPMI1640培地を含む細胞培養皿に入れた。
(4)銅スクリーンを焼き、冷却し、ついで、プレート内に入れた。脾臓を、滅菌シリンジで砕き、細胞懸濁液として調製した。ついで、細胞懸濁液を、13mlの細胞遠心管内にろ過した。
(5)遠心管をメンブランでシールし、2000rpmで10分間遠心分離した。
(6)上清を廃棄し、ついで、2〜3mlの赤血球溶解バッファーを、管内に添加して、細胞を懸濁させた。溶解の2分後、同量のRPMI1640培地(またはウシ胎児血清)を添加して、反応を停止させた。管を、メンブランでシールし、2000rpmで10分間遠心分離した。
(7)上清を廃棄し、ついで、3〜4mlのRPMI1640培地(2%ウシ胎児血清含有)を、管内に添加して、細胞を懸濁させた。
(8)細胞懸濁液を、37℃においてガラス繊維でゆっくりろ過して、細胞とガラス繊維とを完全に組み合わせて、B細胞を除去した。
(9)血球計数チャンバを使用して、細胞の密度をカウントした。
(10)細胞密度を、RPMI1640培地(2%ウシ胎児血清含有)で、3〜4×106個/mlに調節した。
(11)濃度調節した細胞懸濁液を、96ウェルプレートに100μl/ウェルで添加した。
(12)抗原を滅菌し、特定の濃度に希釈した。ついで、20μlの刺激物質を、各ウェルに添加した(刺激物質の最終濃度を、ConA 10μg/ml、5μg/ml、BSA/OVA 2μg/mlとした。刺激物質を希釈して、種々の濃度で添加することができる)。刺激物質を含まない対照細胞のみのウェルおよび培地のみのウェルを設定した。
(13)細胞をインキュベータ内に入れ、37℃での5% CO2下において48〜72時間インキュベートした。ついで、MTT法を、発色に使用した(20μlのMTTを、各ウェル内に添加し、3〜4時間後にデータを読み取った)、実験サンプルOD−培地OD。
(14)上清を捨て、150μlのジメチルスルホキシドを各ウェルに添加した。ついで、プレートを、振とうベッド上に置いて、低速で10分間振とうさせて、結晶を完全に溶解させた。490nmでのOD値を、マイクロプレートリーダ(Magellan、Tecan Austria GmbH)により測定した。
(15)算出結果:SI=(刺激したウェルのOD−培地のみのOD)/(未刺激のウェルのOD−培地のみのOD)。
(1)純粋なT細胞をマウスから10%培地に単離し、細胞1×107個/mlに希釈した。
(2)100ulの細胞および最終濃度10μg/mlでのショートペプチドを96細胞プレートに添加した。最終濃度10μg/mlでのCD28モノ抗体も、同時刺激シグナルのために添加することができる。十分混合した後、混合物を、37℃での5%CO2下においてインキュベートした。
(3)4〜6時間の刺激後、モネンシンタンパク質阻害剤を、2μl/ウェルで各ウェルに添加した。
(4)2時間のモネンシン阻害後、細胞を、2mlのPBSにより2000rpmで5分間遠心分離し、100〜200μlのPBSで再懸濁させた。非特異的結合染色を排除するために、精製FcII/III受容体抗体(CD16/32)を、1μg/106個の細胞の使用に応じて添加し、氷浴中において15〜20分間インキュベートし、2mlのPBSにより2000rpmで5分間遠心分離した。
(5)細胞を、4%パラホルムアルデヒドを含む200μlのPBS溶液に再懸濁させ、室温で10〜15分間インキュベートし、ついで、2mlのPBSにより2000rpmで5分間遠心分離した。
(6)細胞を、200μlの0.1%サポニンに再懸濁させ、4℃で10分間インキュベートし、ついで、2mlのPBSにより2000rpmで5分間遠心分離した。
(7)表面分子および細胞内サイトカインを染色した。
2種類の蛍光抗体を、説明書に基づいて細胞に同時に添加し、氷浴中において20〜30分間インキュベートし、2mlのPBSにより2000rpmで5分間遠心分離した。細胞を、300μlのPBSに再懸濁させた。細胞懸濁液を、機器による測定および分析専用のFACSチューブ内に、銅スクリーンを通してろ過した。
脾臓細胞を、未処置のマウスの赤血球を破壊および単離することにより得た。
(1)脾臓細胞を、2つの培養皿に均等に分けた。一方の皿では、50μgのT細胞エピトープペプチドフラグメントをインキュベートした。一方、他方では、ペプチドフラグメントを添加せずにインキュベートした。各皿において1〜2mlの容量で、皿を、37℃での5%CO2下において4時間インキュベートした(この工程において、目的細胞数を、実験群の数に応じて増加させ得る)。
(2)細胞を、15mlのファルコンチューブ内に移し、3000rpmで5分間遠心分離した。
(3)低分子量ペプチドとインキュベートしていない目的細胞を、低濃度のCFSE(0.5μM)で染色した。一方、低分子量ペプチドとインキュベートした目的細胞を、高濃度のCFSE(5μM)で染色した。両方とも、37℃において暗所で15分間穏やかに振とうした。
(4)同量のウシ胎児血清を添加して、染色後の反応を停止させた。溶液を3000rpmで5分間遠心分離した後、上清を廃棄した。ついで、細胞を、10mlのPBSで3回洗浄した。
(5)低濃度および高濃度で染色した等量の目的細胞を互いに混合し、マウスあたりに細胞2×107個で、尾静脈から実験マウスに再度注射して、in vivoにおける細胞の細胞傷害活性を行った。
(6)マウスを、注射の4時間後に屠殺にした。脾臓細胞を、暗所で単離した。
(7)サンプルを、銅スクリーンによりろ過し、機器による測定および分析専用のFACSチューブ内に移した。
1.GM−CSFがHBVワクチンの体液性応答に影響を及ぼし得るかどうかを試験するために、GM−CSFを、HBVワクチンの先にまたはHBVワクチンと同時に注射した。血清中におけるHBsAgに対する総IgGを、ブースト免疫化の7日後に測定した。HBVワクチンのみを注射された群と比較して、GM−CSFと共に注射された群におけるIgGレベルは、有意に向上していた(図1−A)。IgG2aレベルは、免疫化前3日にGM−CSFを注射された群において、有意に向上した(p<0.05)。対照的に、IgG1レベルは、GM−CSFとHBVとを同時に注射された群において、有意に向上した(図1−A)。GM−CSFによる前処置は、Th1応答を向上させ、一方、GM−CSFとワクチンとの同時注射は、Th2応答を向上させ得ることが、結果から示される。
GM−CSFがT細胞応答に影響を及ぼし得るかどうかをさらに観察するために、マウスの脾臓細胞を、ブースト免疫化の1週間後に、無菌で取り出した。非特異的抗原対照としてBSA、陽性対照としてConAおよび陰性対照として培地を使用して、T細胞の増殖を、rHBsAg抗原で刺激した。T細胞増殖は、GM−CSFの事前注射された群において、有意に増大した。しかしながら、T細胞の増殖レベルは、GM−CSFとHBVワクチンとを同時に注射された群では低かった(図1−B)。このことは、GM−CSFの前処置が、抗原特異的T細胞応答を促進し得たことを示している。
異なる時間において注射されたGM−CSFのCTL応答における効果を理解するために、2回目の免疫化の7日後に、フローサイトメトリにより、CTL応答を測定した。図1−Cに示されたように、目的細胞の特異的殺傷率は、GM−CSFを前処置した群において30.01%である。殺傷率は、10.26%の特異的殺傷率を有する免疫核酸ワクチン単独による群より非常に高い。一方、GM−CSFとHBVとを同時に注射された群では、特異的殺傷率は、13.6%である。GM−CSFの前処置は、in vivo CTL応答のレベルを向上させ得たが、GM−CSFとHBVとを同時に注射された群において明らかな変化が無かったことが、結果から示された。
サイトカインは、細胞の分化および増殖を調節し、細胞が機能するように誘導し得る。したがって、サイトカインは、免疫応答を調節するのに重要な役割を果たしている。この実験では、CD4+T細胞における抗原特異的IL−4およびIFN−γならびにCD8+T細胞における抗原特異的IFN−γの発現レベルを、細胞内サイトカイン染色により測定した。図2に示されるように、GM−CSFで前処置した群における(CD4+細胞での)IL−4、IFN−γおよび(CD8+細胞での)IFN−γのレベルは、HBV単独注射群と比較して、有意に改善された。一方、GM−CSFとHBVワクチンとを同時に注射された群において、対照群と比較して、IFN−γの発現レベルは向上せず、IL−4の発現レベルのみが向上した。GM−CSFで前処置した群は、Th1およびTh2両方のサイトカイン発現を向上させ得るが、GM−CSFとHBVワクチンとの同時注射は、Th2の発現のみを誘導し得たことが、結果から示された。
HBsAgトランスジェニックマウス(C57BL/6J−Tg(A1b1HBV)44Bri/Jf4J)を、Shanghai Public Health Clinical Centerから購入した。Shanghai Public Health Clinical Centerは、Fudan Universityの加盟団体である。「B型肝炎表面抗原用酵素結合免疫吸着法診断キット」およびB型肝炎表面抗原標準を、Beijing Kinghawk Pharmaceutical Co.,LTDから購入した。他の実験材料、主な試薬および機器は、実施形態1に列記されたのと同様である。
動物群および免疫化
35匹のHBsAgトランスジェニックマウス(C57BL/6J−Tg(A1b1HBV)44Bri/Jf4J)(5000−10000pg/mlの本来のHBsAg濃度を有する)を、以下の表「実験免疫群」に列記されたように、1群あたり7匹のマウスを含む5つの群にランダムに分割した。各群において、生理食塩水に溶解させたワクチンまたはGM−CSFを、動物あたりに100μlで頸部皮下注射により注射した。各群のマウスを、1回目、2回目および3回目の免疫化の間に14日の間隔をあけて、ならびに、3回目と4回目との免疫化の間に8週の間隔をあけて、4回免疫化した。マウスの眼窩の血液を、2週間に1回収集し、トランスジェニックマウスの血清中のB型肝炎表面抗原およびB型肝炎表面抗原抗体の濃度をELISAにより測定した。
1.組換えB型肝炎ワクチンとの組み合わせたGM−CSFの戦略が、HBsAgトランスジェニックマウスにおける免疫寛容を打破し、B型肝炎表面抗原の体液性免疫応答を誘導し得るかどうかを試験するために、GM−CSFを、HBVワクチンの前またはHBVワクチンと共に注射した。HBsAgおよびHBsAgに対する総IgGの濃度を、2週間に1回測定した。HBVを単独で注射された群と比較して、総IgGレベルは、3GM−CSF+HBVによる群において有意に向上した(図3)。B型肝炎表面抗原のレベルは、3日前にGM−CSFを注射された群(3GM−CSF+HBV)における4回目の免疫化の6週間後に、有意に低下した(p<0.05)。3GM−CSF+HBV群(すなわち、3日前にGM−CSFを注射された群)は、トランスジェニックマウスの免疫寛容を打破し、HBsAgトランスジェニックマウスにおけるB型肝炎表面抗原の体液性免疫応答を誘導し、さらに、B型肝炎ウイルス表面抗原を低レベルで維持し得たことが、結果から示された。一方、HBVワクチンを単独で注射された群およびGM−CSFとHBVとを同時に注射された群において、B型肝炎表面抗原に対する抗体は、有意に増大しなかった。
HBsAgトランスジェニックマウス(C57BL/6J−Tg(A1b1HBV)44Bri/Jf4J)を、Shanghai Public Health Clinical Centerから購入した。Shanghai Public Health Clinical Centerは、Fudan Universityの加盟団体である。B型肝炎ウイルス表面S抗原に対する一次および二次免疫組織化学抗体を、Shanghai Long Island Biotec.Co.,Ltdから購入する。他の実験材料、主な試薬および機器は、実施形態1に列記されたのと同様である。
35匹のHBsAgトランスジェニックマウス(C57BL/6J−Tg(A1b1HBV)44Bri/Jf4J)(5000−10000pg/mlの本来のHBsAg濃度を有する)を、以下の表に列記されたように、1群あたり7匹のマウスを含む5つの群にランダムに分割した。各群において、生理食塩水に溶解させたワクチンまたはGM−CSFを、動物あたりに100μlで頸部皮下注射により注射した。各群におけるマウスを、それぞれ14日の間隔をあけて3回免疫化した。遅延型過敏反応(DTH)を、3回目の免疫化の12日後に行った。マウスを、3回目の免疫化の15日後に屠殺にした。脾臓を単離し、肝臓の免疫組織化学を行った。
3回目の免疫化の12日後に、実験群および対照群両方における全てのマウスについて、rHBsAg抗原(2μg)を、右後肢の足蹠に注射し、生理食塩水を、左後肢の足蹠に注射した。足蹠の厚みを、注射の24、48および72時間後に、ノギスで測定し、次式:膨潤厚み(mm)=右足蹠の厚み−左足蹠の厚みにより算出した。膨潤厚みの値は、遅延型過敏反応(DTH)レベルを反映している。
HBsAgトランスジェニックマウスを、最後の免疫化の14日後に麻酔し、肝臓組織を固定化し、包埋し、切片化した。H&E染色および免疫組織化学実験を個々に行った。免疫組織化学に使用した抗体を、抗HBsAg抗体とした。
(1)サンプリング:トランスジェニックマウスを、麻酔により屠殺にし、マウスの肝臓組織を取り出した。
(2)固定化:肝臓組織を、Bouin液に直ちに入れた。
(3)脱水および透過性:低濃度から高濃度でのアルコールを使用して、組織を脱水した。
(4)パラフィン浸漬:組織を、パラフィンIに56℃〜58℃で約1時間、パラフィンIIに56℃〜58℃で約1時間、および、パラフィンIIIに56℃〜58℃で約1〜2時間浸漬した。
(5)包埋:組織のブロックを、内側にパラフィンが塗布された予め折りたたまれた小型の紙箱に入れた。
(6)パラフィンブロックのトリミング:パラフィンブロックを、パラフィンテープの形成を容易にするために、台形にトリミングした。
(7)切片:切片の厚みを、約8〜10μmとする。
(8)取付け:パラフィンテープを、38℃に調節した温度の水浴伸長機に浮かべた。パラフィンテープを伸ばした後に、ガラススライドを使用して、パラフィンテープを回収し、顕微鏡観察下で観察して、組織の形態を確認および選択した。
(9)トースティング:取付けた切片を、41℃より高く、50℃より低い温度でのオーブンに、少なくとも4時間入れて、組織をスライドにしっかり取り付けた。
(1)勾配におけるエタノールを使用して、良好な組織形態を有する切片を再水和し、ついで、PBS(0.01M、pH7.4)で、各回5分間継続させて3回浸漬させた。
(2)抗原アンマスキング:切片を、クエン酸バッファーを含む容器に入れ、マイクロ波で高温において、5分×3回加熱した。液体が減少した場合、希釈した熱水を必要に応じて添加した。
(3)切片を、ブロッキングに使用した血清またはBSAで、37℃でのインキュベータ中で少なくとも1時間ブロックした。
(4)血清を、洗浄することなく排出した。適切な比に希釈した一次抗体を、切片を覆うのに使用し、ついで、27℃で1時間または4℃で一晩ブロックした。
(5)切片を、各回5分間継続させて3回PBSで洗浄した。
(6)適切な比に希釈したビオチン標識二次抗体(1%BSA−PBSを使用して希釈)を、切片に滴下し、ついで、27℃で30〜40分間インキュベートした。
(7)切片を、各回5分間継続させて3回PBSで洗浄した。
(8)適切な比に希釈した三次抗体を、切片に滴下し、ついで、27℃で30分間インキュベートした。
(9)アルカリホスファターゼを使用して、暗所で発色させた。陽性のAP発色の結果は、特定の赤色であるべきである。
(10)切片を、水道水ですすいだ。
(11)再染色:染色手順は、Harrisヘマトキシリンを使用して1〜3分間染色し、弱酸溶液(1〜2滴の1M塩酸を希釈水に添加し得る)を使用して1分間鑑別し、続けて、水道水を使用して3分間すすぐというものである。エオシン−フロキシン染色を、1分間直接行った。
(12)切片を脱水し、組織形態を観察するために、ドラフトで乾燥させた。
切片を、保存用ボックス中で保存し、写真撮影し、適切なスケール、カラー調節および適切に陽性の結果の選択で観察した。
1.組換えB型肝炎ワクチン免疫化と組み合わせたGM−CSFは、B型肝炎ウイルス表面抗原トランスジェニックマウスの遅延型過敏反応レベルを向上させた。遅延型過敏反応は、T細胞により媒介される免疫応答の一種である。組換えB型肝炎ワクチンとの組み合わせたGM−CSFの免疫戦略がin vivoでの強力な細胞性免疫レベルを誘導し得るかどうかを評価するために、遅延型過敏反応レベルを、in vivo指標として使用した。実験群に基づいて、免疫化を行った。HBsAgトランスジェニックマウス(C57BL/6J−Tg(A1b1HBV)44Bri/Jf4J)を3回免疫化した12日後に、実験群および対照群からの全てのマウスについて、rHBsAgを、右後肢の足蹠に注射し、生理食塩水を、左後肢の足蹠に注射した。右および左の足蹠厚みを、注射の24、48および72時間後に測定した。B型肝炎ワクチンでのみ免疫化された群が、対照群より高いDTHレベルを有し、一方、組換えB型肝炎ワクチンと組み合わせたGM−CSFの免疫戦略による群(3*GM−CSF+HBVおよび30μg GM−CSF+HBVの群)におけるDTHレベルは、B型肝炎ワクチンを単独で注射された群と比較して、有意に向上したことが、図4−Bに示されている。このことは、組換えB型肝炎ワクチンと組み合わせたGM−CSFの免疫戦略による群(3*GM−CSF+HBVおよび30μg GM−CSF+HBVの群)が、in vivoでの強力な細胞性免疫応答を誘導したことを示している。
1.マウスを従来通り屠殺にし、75%エタノールに5〜10分間浸漬した。
1.B型肝炎表面抗原を、2回目の免疫化の後14日に、ELISAにより測定した。(図6−A)HBVワクチンの注射の1日前でのIFN−α1bの注射は、対照群と比較して、B型肝炎表面抗原に対する抗体レベルを有意に低下させ得ることが見出されている。特に、体液性免疫のレベルは、IFN−α1bとB型肝炎ワクチンとの同時投与による群において有意に抑制された。B型肝炎ワクチン注射の1日前、または、同注射と同時のI型インターフェロンの投与は、B型肝炎ワクチンの体液性免疫レベルに影響を及ぼし得ることが証明されている。
1.既存技術と比較して、抗ウイルス剤は、ウイルス複製を一時的に阻害するのに本発明の医薬組成物に使用される。ついで、ワクチンに加えて免疫調節剤が、免疫化に使用される。免疫化は、B型肝炎ワクチンの免疫応答を効果的に向上させ、単独の抗ウイルス剤の薬剤耐性により引き起こされる応答欠如を避け、慢性HBV感染の処置についての新たな方向性を導き得る。
2.実施形態1〜3に示されたように、既存の抗ウイルス療法と比較して、本発明における抗ウイルス剤の免疫組み合わせが、体液性および細胞性の免疫レベルを効果的に刺激し、免疫応答を有意に向上させ得る。
3.本発明における医薬組成物は、小さなコストで使用されるのに都合が良く、最少の副作用を有し、普及が容易である。
4.本発明における医薬組成物は、免疫寛容により刺激された抗体応答を打破し、Th1、Th2、Th17、Tc1およびTc17の刺激を含む生物のCD4+およびCD8+のT細胞応答を刺激し、免疫応答により効果的にウイルスを除去できるだけでなく、治癒後の感染の再発も予防することができる。
Claims (9)
- 抗ウイルス剤と、免疫調節剤と、組換えB型肝炎ワクチンとからなる、ウイルス免疫療法用の組み合わせ医薬であって、抗ウイルス剤は、ポリエチレングリコール−IFN−α 2aであり、免疫調節剤は、組換えヒト顆粒球マクロファージコロニー刺激因子(rhGM−CSF)であり、組換えB型肝炎ワクチンは、遺伝子操作されたB型肝炎ワクチンであり、遺伝子操作されたB型肝炎ワクチンと組換えヒト顆粒球マクロファージコロニー刺激因子(rhGM−CSF)とは、1:1〜30の重量比を有する、ウイルス免疫療法用の組み合わせ医薬。
- 抗ウイルス剤、免疫調節剤および組換えB型肝炎ワクチンは、混合物としての投与または分離した方法での投与に適しており、投与は、皮下もしくは筋肉内注射によるか、または、それぞれ注射と組み合わせた経口投与によるか、または、異なる順序で異なる時間においてである、請求項1に記載のウイルス免疫療法用の組み合わせ医薬。
- 抗ウイルス剤、免疫調節剤および組換えB型肝炎ワクチンの投与順序は、遺伝子操作されたB型肝炎ワクチンの投与の前に抗ウイルス剤が投与されるか、または、遺伝子操作されたB型肝炎ワクチンの投与の前に組換えヒト顆粒球マクロファージコロニー刺激因子が投与されるかである、請求項2に記載のウイルス免疫療法用の組み合わせ医薬。
- B型肝炎を処置するための医薬の製造における、請求項1に記載のウイルス免疫療法用の組み合わせ医薬の使用。
- B型肝炎は、慢性B型肝炎である、請求項4に記載の使用。
- 組み合わせ医薬は、皮下もしくは筋肉内注射による投与、または、混合物としての投与、または、それぞれ注射と組み合わせた経口投与、または、異なる順序で異なる時間における投与に適している、請求項4に記載の使用。
- 投与中、遺伝子操作されたB型肝炎ワクチンの投与の前に抗ウイルス剤が投与される、請求項6に記載の使用。
- 投与中、遺伝子操作されたB型肝炎ワクチンの投与の前に組換えヒト顆粒球マクロファージコロニー刺激因子が投与される、請求項6に記載の使用。
- 抗ウイルス剤と、免疫調節剤と、組換えB型肝炎ワクチンとからなる、ウイルス免疫療法用の医薬組成物であって、抗ウイルス剤は、ポリエチレングリコール−IFN−α 2aであり、免疫調節剤は、組換えヒト顆粒球マクロファージコロニー刺激因子(rhGM−CSF)であり、組換えB型肝炎ワクチンは、遺伝子操作されたB型肝炎ワクチンであり、遺伝子操作されたB型肝炎ワクチンと組換えヒト顆粒球マクロファージコロニー刺激因子(rhGM−CSF)とは、1:1〜30の重量比を有する、ウイルス免疫療法用の医薬組成物。
