本発明は、リソソーム蓄積症に対する効果的な酵素補充療法のための、リソソーム酵素を含有し、及びN結合型グリコシル化部位を含有するリソソーム標的化部分を含有する標的治療薬、ならびにその製造方法及びその使用方法を提供する。
本発明者らは、意外なことにリソソーム酵素にN結合型グリコシル化部位を含有する標的化部分を結合することによって、リソソーム酵素をリソソームに標的化できることを発見した。本発明は、リソソーム酵素を含有し、及びN結合型グリコシル化部位を含有するリソソーム標的化部分を含有する標的治療薬を提供する。いくつかの実施形態において、本発明との関係で好適な治療用リソソーム酵素は組換えヒトNagluタンパク質である。α−N−アセチルグルコサミニダーゼ(Naglu)は、リソソーム内のヘパラン硫酸の段階的な分解を、このグリコサミノグリカンの非還元性末端からGlcNAc(N−アセチルグルコサミン)を除去することによって助ける。Nagluの不足はヘパラン硫酸の不完全な分解につながり、それに続いて不完全な分解産物がリソソーム内に蓄積する。Nagluの欠如は致死性の神経変性疾患であるサンフィリッポIIIB型の原因となる。
本発明者らは、意外なことにリソソーム酵素にN結合型グリコシル化部位を含有する標的化部分を結合することによって、リソソーム酵素をリソソームに標的化できることを発見した。したがって、本発明は、リソソーム酵素を含有し、及びN結合型グリコシル化部位を含有するリソソーム標的化部分を含有する標的治療薬を提供する。本発明との関係において好適な治療用リソソーム酵素は、リソソーム蓄積症、特にCNS病因及び/または症状を有するリソソーム蓄積症に対する発展または治療の一翼を担うことが明らかにされている任意のタンパク質であり、リソソーム蓄積症としては、アスパルチルグルコサミン尿症、コレステロールエステル蓄積症、ウォルマン病、シスチン症、ダノン病、ファブリー病、ファーバー脂肪性肉芽腫症、ファーバー病、フコシドーシス、ガラクトシアリドーシスI/II型、ゴーシェ病I/II/III型、グロボイド細胞白質ジストロフィー、クラッベ病、糖原病II、ポンペ病、GM1−ガングリオシドーシスI/II/III型、GM2−ガングリオシドーシスI型、テイ・サックス病、GM2−ガングリオシドーシスII型、サンドホフ病、GM2−ガングリオシドーシス、α−マンノシドーシスI/II型、β−マンノシドーシス、異染性白質ジストロフィー、ムコリピドーシスI型、シアリドーシスI/II型、ムコリピドーシスII/III型、I細胞病、ムコリピドーシスIIIC型偽性ハーラー・ポリジストロフィー、ムコ多糖症I型、ムコ多糖症II型、ムコ多糖症IIIA型、サンフィリッポ症候群、ムコ多糖症IIIB型、ムコ多糖症IIIC型、ムコ多糖症IIID型、ムコ多糖症IVA型、モルキオ症候群、ムコ多糖症IVB型、ムコ多糖症VI型、ムコ多糖症VII型、スライ症候群、ムコ多糖症IX型、多種スルファターゼ欠損症、神経セロイドリポフスチン症、CLN1バッテン病、CLN2バッテン病、ニーマン・ピック病A/B型、ニーマン・ピック病C1型、ニーマン・ピック病C2型、濃化異骨症、シンドラー病I/II型、ゴーシェ病及びシアル酸蓄積症が挙げられるがこれらに限定されない。
本発明者らは、意外なことにリソソーム酵素にN結合型グリコシル化部位を含有する標的化部分を結合することによって、リソソーム酵素をリソソームに標的化できることを発見した。したがって、本発明は、リソソーム酵素を含有し、及びN結合型グリコシル化部位を含有するリソソーム標的化部分を含有する標的治療薬を提供する。
リソソーム標的化部分は、当該技術分野において一般に公知の任意の方法でリソソーム酵素に結合され得る。非限定例としては、遺伝学的手段または化学的結合によって融合タンパク質を作製することが挙げられる。いくつかの実施形態では、リソソーム酵素のN末端にリソソーム標的化部分を融合させ、他の実施形態では、リソソーム酵素のC末端にリソソーム標的化部分を融合させる。いくつかの実施形態において、リソソーム標的化部分は内部に位置している、すなわちリソソーム酵素ポリペプチドの内部にある。融合タンパク質の作製は当該技術分野では標準的であり、Sambrook et al., Molecular Cloning: A Laboratory Manual, Cold Spring Harbor Laboratory Press (2001)で詳細に記載されている。本発明の範囲内では、1つより多いリソソーム標的化部分が好適なリソソーム酵素と結合され得ることを企図している。
リソソーム標的化部分は、好適なリソソーム酵素ポリペプチドのN末端またはC末端に融合され得るか、内部に挿入され得る。リソソーム標的化部分は、リソソーム酵素ポリペプチドに直接融合してもよく、リンカーまたはスペーサーによってリソソーム酵素ポリペプチドから分離してもよい。アミノ酸リンカーまたはスペーサーは、概して柔軟であるように、または2つのタンパク質部分の間にa−へリックスなどの構造を挿入するように設計される。リンカーまたはスペーサーは、GGGGGAAAAGGGG(配列番号10)のポリ「GAG」配列、GAP(配列番号11)の「GAP」配列、GGGGGP(配列番号12)の「ポリGP」配列などのように比較的短くてもよく、例えば10〜50(例えば10〜20、10〜25、10〜30、10〜35、10〜40、10〜45、10〜50)アミノ酸の長さのようにより長くすることもできる。いくつかの実施形態において、様々な短いリンカー配列が縦列反復で存在してもよい。例えば、好適なリンカーは、縦列反復で存在するGGGGGAAAAGGGG(配列番号10)の「GAG」アミノ酸配列を含有し得る。いくつかの実施形態において、そのようなリンカーは、1つ以上の「GAP」配列をさらに含有してもよく、GGGGGAAAAGGGG(配列番号10)の「GAG」配列はその枠内に入る。例えば、いくつかの実施形態において、GAPGGGGGAAAAGGGGGAPGGGGGAAAAGGGGGAP(配列番号13)のように、それぞれが「GAP」配列の枠に入っている2つの縦列「GAG」反復を含有するGAG2リンカーを用いてもよい。いくつかの実施形態において、GAPGGGGGAAAAAGGGGGGAPGGGGGAAAAAGGGGGGAPGGGGGAAAAAGGGGGGAP(配列番号14)のように、それぞれが2つの「GAP」配列の枠に入っている3つの縦列「GAG」反復を含有するGAG3リンカーを用いてもよい。いくつかの実施形態において、好適なリンカーまたはスペーサーは、配列番号14の配列と少なくとも50%、55%、60%、65%、70%、75%、80%、85%、90%、95%、98%または99%同一である配列を含有し得る。
一態様において、本発明は、本明細書に開示の融合タンパク質のいずれかをコードする核酸を提供する。一態様において、本発明は、当該核酸を含むベクターを提供する。核酸という用語は本明細書の定義の節で詳細に記載している。本明細書に記載のタンパク質及び/または本発明の他の構成要素の発現用ベクターには、プラスミドベクター及び二本鎖または一本鎖のRNAまたはDNAウイルスベクターが含まれる。ベクターは、DNAとして細胞に直接導入してもよく、ファージ及びウイルスの間接的使用により導入してもよい。ベクターは当該技術分野において一般に公知であり、Sambrook et al., Molecular Cloning: A Laboratory Manual, Cold Spring Harbor Laboratory Press (2001)で詳細に記載されている。

本発明のいくつかの実施形態において、宿主細胞はGNPTを共発現する。一般に、治療用糖タンパク質、例えばリソソーム酵素などが適切な機能を確保するためには、翻訳後修飾に大きく影響される。明細書全体を通して記載しているように、リソソームタンパク質は酸性加水分解酵素として知られる酵素の分類に属し、粗面小胞体で可溶性または膜挿入型糖タンパク質として合成される。可溶性リソソーム酵素のN結合型オリゴ糖は、ゴルジ複合体に至り、通過する過程で修飾され、マンノース−6−リン酸(M6P)認識マーカーを露出する。こうしたM6P認識マーカーは、トランスゴルジネットワーク(TGN)でカチオンマンノース−6−リン酸受容体(CI−M6PR)によって認識され、その結果、集合してAP1−クラスリン被覆小胞中に包み込まれる。エンドソームと融合した後、受容体はリガンドを放出して、新たに形成されたリソソームにリソソーム酵素を送達する。ホスホトランスフェラーゼが欠損しているI細胞病の場合のように、リソソームタンパク質がトランスゴルジネットワークでM6P受容体に結合しなくなった場合、タンパク質は細胞から分泌され、その結果、血漿、脳脊髄液、涙及び尿などの体液中のリソソーム酵素の量が過剰となる。カチオンマンノース−6−リン酸受容体は、リソソーム酵素の細胞表面への及び細胞表面からの再循環及び逆行性移動に重要なので、CI−M6PRは、酵素補充療法における治療用糖タンパク質のリソソーム媒介細胞内輸送に対する重要な標的として機能することができる。
本発明は、GNPT細胞培養系を用いた組換え標的治療用糖タンパク質の製造方法を提供する。いくつかの実施形態において、治療用糖タンパク質のマイクロスケール製造にこの細胞培養系を用いる。いくつかの実施形態において、大規模製造にこの細胞培養系を用いる。いくつかの実施形態において、製造する治療用糖タンパク質は組換えリソソーム酵素である。目的の組換えポリペプチドを製造する典型的な大規模製法としては、回分培養及び流加培養が挙げられる。回分培養プロセスは伝統的に、特定の細胞密度の種培養を大規模製造培養に接種すること、細胞増殖、生存力、及び/または生産力を促す条件(例えば好適な培地、pH、及び温度)の下で細胞を増殖させること、細胞が特定の細胞密度に達したら培養物を採取すること、ならびに発現したポリペプチドを精製することを含む。流加培養製法は、細胞の増殖中に消費される栄養及び他の成分を回分培養に補充する追加の1つまたは複数のステップを含む。いくつかの実施形態において、本発明に従う大規模製造方法は流加培養系を用いる。
本発明はまた、組換え治療用糖タンパク質の製造に有用なバイオリアクターも提供する。バイオリアクターは、例えば灌流式、回分式、流加式、反復回分式、または連続式(例えば連続撹拌槽リアクターモデル)であり得る。一般に、バイオリアクターは、培地(例えば化学的組成が明らかな栄養培地)を入れるように設計及び構成された少なくとも1つの槽を備える。槽はまた一般に、新しい栄養培地を槽内に流すように設計及び構成された少なくとも1つの流入口を備える。槽はまた一般に、廃培地を槽外に流すように設計及び構成された少なくとも1つの流出口を備える。いくつかの実施形態において、槽は、槽内の孤立した細胞が廃培地とともに少なくとも1つの流出口を通って出てしまう程度を最小限にするように設計及び構成された少なくとも1つのフィルターをさらに備え得る。バイオリアクターには、細胞増殖に適した条件を維持するように設計された1つ以上の他の構成要素が取り付けられていてもよい。例えば、バイオリアクターには、栄養培地を槽内で循環またはかき混ぜるように設計及び構成された1つ以上の循環またはミキシング装置が取り付けられていてもよい。一般に、組換え治療用糖タンパク質を発現するように改変された孤立細胞は、栄養培地中に懸濁されている。したがって、場合によっては、循環装置で孤立細胞が確実に栄養培地中に懸濁され続けるようにする。場合によっては、細胞は基質に付着している。場合によっては、細胞は、栄養培地中に浮遊した1つ以上の基質(例えばマイクロビーズ)に付着している。バイオリアクターは、槽から細胞浮遊液の試料を得るための1つ以上のポートを備え得る。バイオリアクターは、気体含有量(例えば空気、酸素、窒素、二酸化炭素)、流量、温度、pH及び溶存酸素濃度、ならびに攪拌速度/循環速度などの条件を含めた、培養の条件を監視及び/または制御するための1つ以上の構成要素によっても構成され得る。
本発明のある特定の実施形態において、実施者は、増殖細胞培養の特定の条件を定期的に監視することが有益または必要であると認める場合がある。細胞培養条件を監視することにより、実施者は、細胞培養で組換えポリペプチドまたはタンパク質が準最適レベルで産生されているかどうか、または培養が準最適産生段階に入ろうとしているかどうかを確認することができる。ある細胞培養条件を監視するためには、分析用に培養物の少量アリコートを取り出す必要がある。
本明細書に記載の様々な方法に従って製造した治療用糖タンパク質を精製または単離するために、様々な方法が用いられ得る。いくつかの実施形態において、発現した治療用糖グリコプロテインタンパク質(例えばリソソーム酵素)は培地中に分泌され得、そのため、精製プロセスの第1ステップとして例えば遠心分離または濾過によるように、細胞及び他の固形分が除去され得る。代替的にまたは追加的に、発現した治療用糖タンパク質は宿主細胞の表面に結合し得る。この実施形態では、ポリペプチドまたはタンパク質を発現した宿主細胞(例えば酵母菌)を精製のために溶解させる。宿主細胞(例えば酵母菌)の溶解は、ガラスビーズによる物理的破砕及び高pH条件への曝露を含めた、当業者にとって公知の任意の数の手段により行うことができる。
本発明は、本発明に従う標的治療薬を含有する医薬組成物をさらに提供する。典型的に、好適な医薬組成物は、少なくとも1種の医薬品として許容可能な添加物を含有し、ヒトへの投与に向けて配合される。
いくつかの実施形態において、本発明に従う医薬組成物を髄腔内投与に用いることができる。本明細書で用いる場合、髄腔内投与(髄腔内注入または髄腔内送達とも呼ぶ)は、脊柱管(脊髄を取り囲むくも膜下腔)への注入のことをいう。髄腔内投与のための様々な製剤がWO/2011/163652に記載されており、その内容を参照により本明細書に援用する。
本発明を用いてサンフィリッポ症候群B型及び他のリソソーム蓄積症を効果的に治療することができる。サンフィリッポ症候群B型、またはムコ多糖症IIIB(MPS III B)は酵素Nagluの欠損に起因する遺伝性代謝障害である。Nagluはリソソームに局在化され、グリコサミノグリカン(GAG)であるヘパラン硫酸及びデルマタン硫酸の異化において重要な役割を果たす。酵素が欠如すると、これらの基質は細胞内に蓄積し、最終的に鬱血を引き起こし、続いて細胞死及び組織破壊が生じる。酵素は広範囲に及んで発現しているため、MPS III B患者では複数の細胞型及び器官系が侵される。
複数のタンパク質がCI−M6P受容体中の異なる細胞外領域/ドメインへの結合に関係があるとされており、例えば白血病抑制因子、E1A刺激遺伝子の細胞リプレッサー及びサポシンなどはM6Pに依存する形で、また例えばインスリン様成長因子IIなどはM6Pに依存しない形で結合する。したがって、以下に記載するように、CI−M6PRに結合し、したがってリソソームへのrhNagluのCI−M6PR媒介送達を促進し得る一連のNaglu融合タンパク質を設計した。
Naglu−SapDCタンパク質は、以下に記載するようにNaglu及びサポシンタンパク質の一部の融合体である。Naglu−SapDC融合タンパク質を生成するために、サポシンの保存C末端領域及びD機能ドメイン(SapDC)をコードする配列にヒトNaglu遺伝子を結合させることによって、核酸構築物を生成した。GAG3リンカーを2つの配列間に挿入して翻訳後の2つのペプチド間の回転を可能とした。得られた全長Naglu−SapDC融合タンパク質(配列番号18)において、全長NagluはN末端を構成し、SapDCペプチドはC末端の配列を決定する。

標準的な技術を用いて、異なるNaglu融合タンパク質のそれぞれをコードする核酸を哺乳類発現ベクターにサブクローニングし、HT−1080細胞に形質移入した。その後、当該技術分野において一般に公知の技術を用いて、トランスフェクタントをスクリーニングしてHT−1080過剰発現細胞株を生成した。インビトロタンパク質をベースとしたすべてのアッセイ及び受容体結合実験用には、哺乳類細胞培養発現系を用いて、ウェーブバイオリアクター内で組換えタンパク質を製造した。発現後、各融合タンパク質に対して3段階精製プロセスを施した。まず、限外濾過(UF)装置(Pall Corporation, Port Washington, NY 11050)を用いて馴化培地を濃縮した。次に、濃縮した培地をブチルセファロースクロマトグラフィーカラム(ブチル)にかけ、その後、Qセファロースクロマトグラフィーカラム(Q)を用いて、この精製段階の溶出液をさらに精製した。保存のために、精製したタンパク質をPBS(11.9mMのリン酸ナトリウム、2.7mMのリン酸カリウム、137mMの塩化ナトリウムでpH7.4)中にバッファー交換した。得られた融合タンパク質は90%を超える純度に精製された。
N結合型グリカンの翻訳後修飾においてGNPTが果たす重要な役割、具体的には、可溶性リソソーム酵素のM6Pリン酸化を考慮して、GNPT酵素の過剰発現がリソソーム酵素上のM6Pリン酸化度を増加させ得るかどうかを評価するために実験を行った。

GNPTのガンマサブユニットの発現を促進させるために、配列番号29の核酸配列を用いて核酸構築物を生成した。
N結合型グリカンの翻訳後修飾においてGNPTが果たす重要な役割、具体的には、可溶性リソソーム酵素のM6Pリン酸化を考慮して、GNPT酵素の過剰発現がリソソーム酵素上のM6Pリン酸化度を増加させ得るかどうかを評価するために実験を行った。簡潔に言うと、標準的な方法を用いて、1種のみのリソソーム酵素(GAA、LAL、ARSA、I2S、HNSもしくはGCB)を過剰発現するか、またはリソソーム酵素及びGNPT(GAA−GNPT、ARSA−GNPT、I2S−GNPT、HNS−GNPTもしくはGCB−GNPT)を共過剰発現する組換えHT−1080細胞株を生成した。Naglu−LIFのみ(Naglu−LIF)またはNaglu−LIF及びGNPT(Naglu−LIF−GNPT)を過剰発現する類似の細胞株を生成した。細胞株のそれぞれをコンフルエントになるまで増殖させ、馴化細胞培地を収集して記載したように濃縮した。その後、対照の目的で、濃縮した試料の小部分をエンドグリコシダーゼHで処理し、処理した試料及び未処理の試料をSDS−PAGEで分離して(タンパク質はELISAまたは酵素活性アッセイにより定量し、各リソソーム酵素250ngを添加した)、抗M6P抗体を用いたウエスタンブロッティングにより解析した。図4に示すように、各リソソーム酵素とGNPTとの共発現の結果、リソソーム酵素単独での発現と比較してM6Pリン酸化シグナルが劇的に増加した。驚いたことに、単独で過剰発現したNaglu−LIFタンパク質ではいかなる形でもM6Pリン酸化が検出されないが、Naglu−LIFをGNPTと共発現させた結果、M6Pリン酸化が増加した(図4)。
GNPT過剰発現細胞株での組換えリソソームタンパク質の発現の結果、M6Pリン酸化が増加するかどうかをさらに評価するために、リソソーム酵素I2Sに関してさらなる試験を行った。標準的な方法を用いて、I2Sのみを過剰発現するか、またはI2S及びGNPTを共過剰発現する組換えHT−1080細胞株を生成した。細胞株のそれぞれをコンフルエントになるまで増殖させ、馴化細胞培地を収集して記載したように濃縮した。実施例4に記載のCI−M6PR結合アッセイを用いて、受容体親和性の何らかの変化について各タンパク質を試験した。この分析により、I2Sは、このたんぱく質のみを過剰発現した細胞から得られたものの方が、GNPTも過剰発現した細胞から得られたI2Sよりも弱くCI−M6PRに結合することが明らかとなった。(図5)。
上記のように、GNPT過剰発現細胞株を用いることによって、組換え製造した治療用糖タンパク質のM6Pリン酸化度を増加させることができた(図4)。特に、治療用融合糖タンパク質Naglu−LIFに対して、M6Pリン酸化の増加が観察された(図4)。驚いたことに、LIFに融合した場合におけるNagluタンパク質のM6Pリン酸化のこの増加は、天然Nagluタンパク質、またはGNPTを共過剰発現していない細胞で生成した非融合rhNagluに対しては観察されなかったレベルのM6Pリン酸化をもたらした。この現象をさらに評価するために、rhNaglu及びGNPT過剰発現細胞株を用いて組換え製造した様々な融合タンパク質に対してさらなる試験を行い、M6Pリン酸化度を評価した。簡潔に言うと、標準的な製法を用いて、1種のみの組換えタンパク質(Naglu−IGFII、rhNaglu、Naglu−CREGもしくはNaglu−SapDC)を発現するか、またはリソソーム酵素及びGNPT(GNPT/Naglu−SapDC、GNPT/Naglu−CREG、GNPT/rhNaglu)を共発現するクローンHT−1080細胞株を生成した。各細胞株をコンフルエントになるまで増殖させて馴化培地を収集し、90%を超える純度にそれぞれのリソソームタンパク質及び/または融合タンパク質を精製した。
GNPTとの共発現後のNaglu−LIF及びNaglu−SapDC上のM6Pの増加をさらに確認するために、単糖M6P定量アッセイを行った。この方法では、酸性加水分解を用いて分析する糖タンパク質上のすべてのグリカンを分解し、その後、遊離した単糖、例えばM6PなどをPAD−HPLC(パルスドアンペロメトリック検出高速液体クロマトグラフィー)により検出及び定量した。簡潔に言うと、M6P二ナトリウム塩を用いて分析する試料から遊離したM6Pのピークを特定した。M6P標準試料の曲線下面積を用いて標準曲線を作成し、試料から遊離したM6Pの定量を可能とした。Naglu−SapDCのM6P含有量はGNPTとの共発現後で0.75から1.84mol/mol(M6P/タンパク質)に増加し、またNaglu−LIFのM6P含有量はGNPTとの共発現後で1.07に増加した(図9)。GNPT共発現Naglu−SapDC及びGNPT共発現Naglu−LIFは両方とも、対照タンパク質のM6P含有量と非常に類似したM6P含有量を有していた(図9)。この結果は、GNPT共発現がNaglu融合タンパク質のM6P含有量を治療上有意なレベルまで増加させたことを示している。Naglu−LIF及びNaglu−SapDC上のM6Pを定量するために用いた方法を以下でより詳細に記載する。
GNPT過剰発現細胞株で産生したNaglu−SapDC及びNaglu−LIFのインビトロCI−M6PR媒介標的化及び細胞内取込も評価した(図10〜11)。比較のためにNaglu−IGFIIの細胞内取込を測定した。得られるデータを比較可能とするために、すべての試料を並行して同一条件下で実験した。

処置したマウスから組織試料も収集し、10%のNBF中で固定してパラフィン包埋のために処理した。アッセイした各組織について、ヒトNagluに特異的な抗体を用いて5μmパラフィン切片に免疫染色を施した。このデータは、Naglu−SapDCの大脳皮質ニューロンへのリソソーム送達をはっきりと示している(図13A、B)。最も際立ったことは、このデータが肝臓の肝細胞におけるNaglu−SapDCのリソソーム送達も示していることである(図13E)。これは、髄腔内送達を介してNaglu−SapDCを投与したという点で驚くべきことであり、Naglu−SapDCがその投与部位から遠くの標的に到達し、細胞に進入できることを示している。
様々な糖タンパク質の細胞内蓄積及びそれらの対応する分解産物を調べることによって、Naglu−SapDCの髄腔内投与により処置したNagluノックアウトマウスにおけるNaglu酵素のインビボ活性を評価した。そのようなアッセイの1つでは、肝臓(図14)及び脳組織(図15)中のグルコソアミノグリカン(glucosoaminoglycan)(GAG)の量を評価するように計画した。
Naglu−LIFの細胞内蓄積
ここに記載するインビボ実験では、GNPT過剰発現細胞で調製したNaglu−LIFを用いた。
治療したマウスから組織試料も収集し、10%のNBF中で固定してパラフィン包埋のために処理した。アッセイした各組織について、ヒトNagluに特異的な抗体を用いて5μmパラフィン切片に免疫染色を施した。このデータは、脊髄、小脳の髄膜、大脳皮質の髄膜のニューロン及びグリア細胞、ならびに大脳皮質の限られた領域のニューロン及びグリア細胞(図13C及びD)へのNaglu−LIFのリソソーム送達をはっきりと示している。最も際立っていることは、このデータは肝臓の肝細胞におけるNaglu−LIFのリソソーム送達も示していることである(図13F)。これは、髄腔内送達を介してNaglu−LIFを投与したという点で驚くべきことであり、Naglu−LIFがその投与部位から遠くの標的に到達し、細胞に進入できることを示している。
Naglu−SapDC治療ノックアウトマウスの分析に関連して上に記載したように、GAG及びヘパリン硫酸細胞内蓄積を調べることによってNagluのインビボ活性を評価した。図14及び図15に示すように、Naglu−LIFの髄腔内送達の結果、溶媒対照と比較して、Nagluノックアウトマウスの肝臓及び脳内それぞれのGAG総濃度が著しく減少した。脳内のGAG総濃度の減少は3週間の治療期間全体にわたって維持された(図15)。
Claims (38)
- リソソーム酵素;及び
リソソーム標的化部分;
を含み、前記リソソーム標的化部分が、M6P基を含有する少なくとも1つのN結合型グリコシル化部位を含有する、標的治療薬。 - 前記リソソーム酵素が表2から選択される、請求項1に記載の標的治療薬。
- 前記リソソーム酵素がN−アセチルグルコサミニダーゼ(Naglu)タンパク質である、請求項1に記載の標的治療薬。
- 前記Nagluタンパク質が配列番号1と少なくとも80%同一のアミノ酸配列を含む、請求項1〜3のいずれか1項に記載の標的治療薬。
- 前記Nagluタンパク質が配列番号1と少なくとも90%同一のアミノ酸配列を含む、請求項1〜4のいずれか1項に記載の標的治療薬。
- 前記Nagluタンパク質が配列番号1と少なくとも95%同一のアミノ酸配列を含む、請求項1〜5のいずれか1項に記載の標的治療薬。
- 前記Nagluタンパク質が配列番号1と同一のアミノ酸配列を含む、請求項1〜6のいずれか1項に記載の標的治療薬。
- 前記リソソーム標的化部分が、プロサポシン、E1A刺激遺伝子の細胞リプレッサー、白血病抑制因子、及びこれらの断片からなる群から選択されるタンパク質である、請求項1〜7のいずれか1項に記載の標的治療薬。
- 前記リソソーム標的化部分が配列番号4、6、または9と少なくとも80%同一の配列を含む、請求項8に記載の標的治療薬。
- 前記リソソーム標的化部分が配列番号4、6、または9と少なくとも90%同一の配列を含む、請求項8に記載の標的治療薬。

- 前記リンカーがGAPGGGGGAAAAAGGGGGGAPGGGGGAAAAAGGGGGGAPGGGGGAAAAAGGGGGGAP(配列番号15)の配列を含む、請求項15に記載の標的治療薬。
- 前記融合タンパク質が配列番号18、19または20のアミノ酸配列と少なくとも80%同一である、請求項13〜17のいずれか1項に記載の標的治療薬。
