Description
前記化学式(I)でn及びmはそれぞれ独立的に5−500である。また、前記化学式(I)のイミダゾール基はトリアゾール基(triazole)またはピペラジン基(piperazine)に置換されることができる。
前記化学式(II)でn及びmはそれぞれ独立的に5−500である。また、前記化学式(II)のイミダゾール基はトリアゾール基(triazole)またはピペラジン基(piperazine)に置換されることができる。さらに、前記化学式(II)のフェニル基はビフェニル基(bipHenyl)、ナフチル基(napHthyl)またはインドール基(indole)に置換されることができる。
本発明の光力学的治療用自己組立型薬学の組成物において、MRI造影用ナノ粒子を追加で含めることができる。前記MRI造影用ナノ粒子は酸化鉄ナノ粒子であることができる。また、前記酸化鉄ナノ粒子の大きさは1nmないし100nmであってもよい。
(実施例1.リガンドの合成)
ポリ(エチレングリコール)−ポリ(β−ベンジル−L−アスパルテート)(PEG−PBLA)を鋳型として使用して、リガンドA及びリガンドBを合成した(化3)。
PEG−PBLAを製造するために、β−ベンジル−L−アスパルテートN−カルボキシ無水物(BLA−NCA)(3g、12mmol)を、DMF(20mL)及びCH2Cl2(50mL)混合物内、40℃で末端の1級アミノ基のα−メトキシ−ω−アミノ−ポリ(エチレングリコール)(MW=2,000Da、240mg、120mol)から開始して重合した。PEG−PBLAをエーテル(3L)で3回沈殿させて精製した。PEG−PBLA−Ce6(プラットフォームリガンド)を合成するために、Ce6を従来のカルボジイミド反応を通じて、PEG−PBLAのアミン基に付着した。前記PEG−PBLA(0.5g、32.5mol)と、Ce6(23.4mg、39.0μmol)、ジシクロヘキシルカルボジイミド(9.6mg、46.8μmol)及びN−ヒドロキシスクシンイミド(5.4mg、46.8μmol)の混合物をDMSO(5mL)にそれぞれ独立的に溶解し、前記溶液を縮合反応の前に3時間十分に撹拌した。次に前記二つの反応溶液を混合して室温で撹拌した。24時間後、前記反応混合物を濾過して不溶性副産物(例えば、ジシクロヘキシルウレア)を除去し、2日間にわたって脱イオン水に透析した(Spectra/Por:分子量遮断の大きさ、MWCO:1,000Da)。前記最終的な溶液を凍結乾燥し、プラットフォームのリガンドを得た。
(実施例2.ESIONの合成)
ESIONを、以前に報告された方法(Kim,B.H.;Lee,N.;Kim,H.;An,K.;Park,Y.I.;Choi,Y.;Shin,K.;Lee,Y.;Kwon,S.G.;Na,H.B.;Park,J.G.;Ahn,T.Y.;Kim,Y.W.;Moon,W.K.;Choi,S.H.;Hyeon,T.J.Am.Chem.Soc.2011、133、12624)を使用してオレイルアルコール存在下でアイロン−オレアート錯化合物の熱分解を通じて合成した。簡単に、1.8gのアイロン−オレアート錯化合物(2mmol)、0.57gのオレイン酸(2mmol)及び1.61gのオレイルアルコール(6mmol)を室温で10gのジフェニルエーテルに溶解させた。前記混合物を10℃/minの一定の昇温速度で250℃に加熱した上で、不活性大気の下で30分間、前記温度を維持した。前記反応が進むことによって、初期の褐色透明な溶液が黒い色に変わった。前記反応後、ナノ粒子を含有する前記混合物を加熱器から外して室温で冷却した後、50mLのアセトンを添加し、前記ナノ粒子を沈殿させた。前記ナノ粒子を4時間40,000rpmで遠心分離してペレット化し、前記上澄液をデカント(decant)しており、前記ナノ粒子をn−ヘキサンまたはクロロホルムに再分散させた。
(実施例3.PMNの製造)
自己組立のために、15mgのリガンドA及び15mgのリガンドBをDMSO3mL内で混合して製造した溶液を、5mLのクロロホルム内のコロイド状のESION(0.4mgのFe/mL)にゆっくりと添加した。前記混合物を30分間、室温で振盪培養(incubated on a shaker)した。次に、クロロホルムを真空下に蒸発させて完全に除去し、全体量が5mLになるまでDMSO内の前記コロイド状の懸濁液に脱イオン水を添加した。透析膜(Spectra/Por;MWCO:12,000Da)を使用し、前記DMSOを脱イオン水に完全に置換させた。余剰のリガンドを遠心分離して除去し、スピンフィルター(Millipore、MWCO:100,000Da、10,000、10分)で3回ないし5回洗浄した。こうして生成されたナノ粒子を水に再分散させた。
(実施例4.自己組立リガンドの製造)
3mLのDMSOに15mgのリガンドA及び15mgのリガンドBの混合溶液を5mLのクロロホルムにゆっくりと添加した。前記混合物を30分間、室温で振盪培養した。以後、クロロホルムを真空下で蒸発させて完全に除去した。その後、全体量が5mLになるまでDMSO内のコロイド状の溶液に脱イオン水を添加した。DMSOを透析膜(Spectra/Por;MWCO:12,000Da)を使用して脱イオン水に完全に置換させた。余剰のリガンドを遠心分離して除去し、スピンフィルター(Millipore、MWCO:100,000Da、10,000、10min)で3回ないし5回洗浄した。こうして生成されたナノ粒子を水に再分散させた。
(実施例5.pH−非感応性ナノ粒子組立体(InS−NP)の製造)
3mLのDMSOに30mgのプラットフォームリガンドを混合して製造された溶液を、5mLのクロロホルム内のコロイド状のESION(0.4mgのFe/mL)懸濁液にゆっくりと添加した。前記混合物を30分間、室温で振盪培養した。次に、クロロホルムを真空下に蒸発させて完全に除去した。その後、全体量が5mLになるまでDMSO内のコロイド状の溶液に脱イオン水を添加した。DMSOを透析膜(Spectra/Por;MWCO:12,000Da)を使用して脱イオン水に完全に置換させた。余剰のリガンドを遠心分離して除去し、スピンフィルター(Millipore、MWCO:100,000Da、10,000、10分)で3回ないし5回洗浄した。こうして生成されたナノ粒子を水に再分散させた。
(実施例6.材料の特性分析)
TEM測定は200kVで作動するJEOL EM−2010顕微鏡で遂行した。粉末X線回折パタ−ンを回転陽極及びCu Kα放射線源(λ=0.15418nm)を備えたRigaku D/Max−3C回折分析器で得た。本発明者たちはICPS−7500分光器(Shimadzu)を使用する誘導結合プラズマ発光分光分析(inductively coupled plasma atomic emission spectroscopy、ICP−AES)及び誘導結合プラズマ光学的放出分光計(inductively coupled plasma−optical emission spectrometer、ICP−OES)(PerkinElmer Optima 4300 DV)で元素分析を遂行した。UV/visible吸収スペクトルはUV−2450分光光度計(Shimadzu、Japan)で収集した。粒径及びゼ−タ電位はZetasizer Nano ZS(Malvern Instruments、Malvern、UK)で測定した。大きさデ−タ分析のために、素材屈折率(2.42)、素材の吸光度(0.2)、分散剤屈折率(1.33)、及び分散剤粘度(0.8872)を使用した。磁気的特性は超伝導光子干渉法(superconducting quantum interface device、SQUID)磁力計(Quantum Design MPMS XL)を使用して調査した。水力学的大きさは25℃で動的光散乱(dynamic light scattering、DLS)(Zetasizer Nano ZS(Malvern Instruments、Malvern、UK)機器を使用して、獲得した。
(実施例7.臨界凝集濃度(CAC)の測定)
2回蒸留された蒸留水内のHoechst33342保存液(1.4×10−3M)を製造し、4℃で保存した。1.0×10−3mg/mLないし1.0mg/mLの濃度で前記Hoechst33342溶液を試料を含有する溶液と混合した。各試料溶液内のHoechst33342の最終濃度は7.0×10−4Mだった。それぞれλex=355nm及びλem=457nmのRF−5310PC(Shimadzu、Japan)を使用して蛍光を測定し、スリット幅はそれぞれex=3nm及びem=3nmだった。
(実施例8.他のpHでPMNの透過率の測定)
PMN溶液(2mg/mL、Ce6付着なし)の光透過率の測定値を500nmでUV−Visible分光光度計を使用して獲得し、そのとき0.1N塩酸を添加して溶液のpH値が7.5から4.0に徐々に減少し、0.1N NaOHの溶液を添加し、前記溶液のpHを4.0から7.5に増加させた。
(実施例9.pH−依存的蛍光強度の測定)
PMNまたは自己組立リガンド(2mg/mL、650nmの励起及び675nmの放出)の蛍光強度を、0.1N塩酸溶液を添加して溶液のpHを7.5から4.0に徐々に減少させながら、蛍光プレートリーダー(fluorescence plate reader(Tecan Genios、Durham、NC))を使用して測定した。
(実施例10.pH−依存的一重項酸素の生成(SOG)の測定)
SOG(singlet oxygen generation)を評価するため、他のpHの試料(2mg/mL、100μL)を一重項酸素センサーグリーン(singlet oxygen sensor green(SOSG)、2.0μM、100μL)と混合した。4分間5mW/cm2強度で、670nmのレーザー源(Institute of Electronics)を照射(irradiation)してSOGを誘導した。試料のSOGを決定するための照射後SOSG蛍光を検出した(λex=494nm、λem=534nm)。SOSG蛍光の増加とバックグラウンドの比較により、SOGを評価した。
(実施例11.細胞培養)
HCT116(人間の結腸癌、KCLB No.10247)細胞、CT26(マウスの結腸直腸癌種の細胞、KCLB No.80009)及びM2−10B4(マウスの骨髄間質細胞、KCLB No.21972)を韓国細胞銀行(Korean Cell Line Bank)から得た。HCT116細胞を、10%ウシ胎児血清(FBS)及び1%ペニシリン/ストレプトマイシンが添加された10mLのRoswell Park Memorial Institute(RPMI−1640)培地で培養した。CT26細胞を、10%FBSと1%ペニシリン/ストレプトマイシンが添加されたDulbecco’s Modified Eagle Medium(DMEM)で培養した。M2−10B4を、10%FBS及び1%ペニシリン/ストレプトマイシンが添加されたRPMI−1640の培地で培養した。すべての細胞の培養条件は37℃、湿度100%及びCO25%だった。24時間、CT26細胞に露出されるDOXの投与量を増加(1、5、10、25、50、100、500、1000、及び5000ng/mL)した後、次の投与量に露出される前に3日ないし4日の回復期間を与え、薬物耐性CT26細胞(CT26/MDR)を成長させた。次に、CT26/MDRを冷凍させ、液体窒素に保存した。CT26/MDRの新しい分割量(fresh aliquots)を全ての実験に使用して薬物敏感性表現型(drug sensitive phenotype)に転換されないようにした。
(実施例12.pH−依存的細胞吸収(cell uptake))
HCT116細胞を他のpHでPMN、InS−NP及びCe6に露出させ、それぞれの細胞の吸収を確認した。前記細胞を2時間培養し、洗浄し、収穫した後、DPBSに再懸濁させた。試験管内(in vitro)細胞の吸収を流動細胞計数器(Beckman、San Jose、CA、USA)を使用して定量した。各試料10,000細胞(ゲ−トイベント(gated events))を計数し、遊離したCe6の蛍光をログ設定(logarithmic setting)(FL4;λem=670nm)で検出した。これらの蛍光(FL4)が非処理細胞の懸濁液の細胞蛍光に比べて高いと、細胞を陽性と計数した。各実験をCXP分析プログラムを使用して統計的に分析した。
(実施例13.共焦点レーザー走査顕微鏡)
共焦点レーザー走査顕微鏡(LSM 510 Meta;Zeiss、Germany)を使用して蛍光映像化を遂行した。120μmの最適のピンホ−ル(pinhole)サイズを選定して、焦点平面の上と下の焦点はずれ(out−of−focus)平面で放出された蛍光を排除した。励起/放出、波紋はDAPIについて340/488nm、FITCについて490/520NM、RITCについて555/578nmそしてCe6について650/670nmであった。蛍光映像をLSM Image Browser software(Zeiss)を使用して分析した。
(実施例14.細胞MR映像化)
PMNによって細胞を標識するために、HCT116細胞を10mLの培地の培養皿に接種し、一夜にかけて成長させた。次いで、0、12.5、25及び50mg/mLのPMNを添加した。4時間後、前記細胞をPBSで2回洗浄し、1mLのトリプシン/EDTAを添加して分離させた。遠心分離後、細胞を培養培地に分散させ、1.5mLの試験管に移した。5分間、1,000rpmで遠心分離して細胞ペレットを製造した。T1−加重MR映像を1.5 T MRスキャナー(Signa Excite、GE Healthcare)のヘッドコイルを使用して、獲得した。
(実施例15. 薬物動態分析)
血漿濃度−時間の実験のため、マウスの尾静脈を通じてPMN及びInS−NPをそれぞれ(2mgのFe/kg)注射した。前記注射後1分、30分、1時間、3時間、8時間及び20時間で血液を収集した。10分間、1,000rpmで遠心分離して赤血球から血漿を分離した。その後、各血液試料用血漿(100μL)を12時間室温で70%硝酸(1mL)と混合した後、遠心分離(5分、10,000rpm)し、上澄液を2%硝酸で5倍希釈した後に誘導結合プラズマ発光分光分析(inductively coupled plasma optical emission spectrometer analysis)(ICP−OES、Optima 4300 DV、Perkin−Elmer)に使用した。PMNまたはInS−NP(2mgFe/kg)を注射してから12時間後に前記腫瘍の鉄の吸収を測定した。切開した腫瘍組織の重さを測定して、均質化しており、シンチレーション混合物(scintillation mixture)で処理した。多量の60%硝酸を各試料に添加し、組織を60℃で24時間培養した後、前記溶液を30分間、13,000rpmで遠心分離し、上澄液を2%硝酸で10倍希釈した。腫瘍内鉄吸収量の測定をICP−OESで遂行し、腫瘍内の前記ナノ粒子の吸収をgFe/g組織で計算した。
(実施例16.免疫組織化学)
腫瘍組織を10%中性緩衝ホルマリンに固定し、5ミクロンの厚さでスライスして冷凍切開した。SDF−1及びP−gpを抗−SDF−1抗体(abcam)及び抗−P−糖蛋白質抗体(abcam)で免疫標識(immunolabel)し、以後、追加で共焦点レーザー走査顕微鏡の分析をするため、前記試料をRITC(λex/λem、555/578nm)またはFITC(λex/λem、490/520nm)が接合された(conjugated)二次抗体を使用して室温で60分間、培養した。
(実施例17.腫瘍組織学)
組織学的分析のために、対照群及び処理されたマウスの腫瘍組織を10%中性緩衝ホルマリンに固定し、5ミクロンの厚さでスライスして冷凍切開し、ヘマトキシリン・エオシン(matoxylin&eosin、H&E)で染色し、TUNEL法で標識化し、デジタル顕微鏡(Leica QWin)及び共焦点レーザー走査顕微鏡(LSM 510 Meta;Zeiss、Germany)で検査した。
(実施例18.PMNの生体内での毒性評価)
健康なヌ−ドマウスにPMNまたは生理食塩水(対照群)の懸濁液を静脈注射した。2週間後、前記マウスを犠牲にし、主要臓器を収集した。心臓、肝臓、脾臓、腎臓 及び肺をH&Eで染色した。ALT、AST、ALP及びD−LDHの試薬キットを、眼球摘出によって採取した血液から分離した血清を分析するために使用した。
(実施例19.試験管内での細胞吸収)
腫瘍細胞のPMNの吸収を、他のpH条件(pH7.4及び6.8)で、KODAK In Vivo Image Station(Image Station 4000 MM;Kodak、New Haven、CT、USA)及び流動細胞分析機(Beckman、San Jose、CA、USA)を使用してモニタリングした。HCT116細胞(1×105cells/mL)を37℃、RPMI−1640の培地(pH7.4または6.8、1%のペニシリンとストレプトマイシン添加)で2時間の間、各試料と一緒に培養した上で分析した。顕微鏡観察中の光脱色(photobleaching)を改善するために、アンチ−フェードマウンティングソリューション(anti−fade mounting solution)(5%N−プロピルガレート、47.5%グリセリン、及び47.5%Tris−HCl、pH8.4)の一滴を前記細胞に添加した。
(実施例20.生体内での研究)
すべての生体研究はアメリカ合衆国国立保健院(National Institutes of Health)が出版したthe Guide for the Care and Use of Laboratory Animals(NIH publication no.85−23、1985、revised 1996)を受けて、韓国カトリック大学(Republic of Korea)のIACUC(Institutional Animal Care and Use Committee)から承認されたプロトコルのガイドラインのもとにマウスを保存した。
(実施例21.pH−感応性磁気ナノ組成物(PMN)の特性)
本発明はCe6がグラフティングされたポリ(エチレングリコール)−ポリ(−ベンジル−L−アスパルテート)(PEG−PBLA−Ce6)を合成して、側面のベンジルエステル基が求核攻撃を通じて第一級アミンと直ちに反応するプラットフォームリガンドを提供した。イミダゾール(pKa=約6.8)はイオン化される官能基として容易に統合され、腫瘍の微細環境にpH感応性を付与した。このようなプラットフォームに基づいて、二つのリガンド誘導体:リガンドA及びBを追加で設計した。リガンドAについて、カテコール基 (catechol group)を追加することによって自己組立(self−assembly)を容易にしたが、これは前記カテコール基が酸化鉄ナノ粒子に対する高親和力アンカー(high−affinity anchor)として作用することができるからである。これと対照的に、3−フェニル−1−プロピルアミンを使用してリガンドBの疎水性(hydrophobicity)を調節して、約5.5の腫瘍エンドソーム/リソソームのpHによって活性化されるPMNの臨界相転移(critical phase transition)を生成した。ESION、リガンドA及びリガンドBの共組立(coassembly)によって前記PMNを製造した(化4)。
ESION(約3nm)が前記リガンドのペプチドブロックの水力学的大きさ(約60個の残基;約20nm長さ)より非常に小さいために、カテコールが固定された(catechol−anchored)リガンドAがESIONの側面を取り囲むことが可能だった。したがって、前記機能化がESIONをコロイド状のマグネト−コアシェル(colloidal magneto−core shell)構造体の内部に誘導し自己組立されることを可能にするために、このような機能化されたESIONは従来の両親媒性二重ブロック共重合体(diblock copolymer)の高分子−金属類似体(polymer−metal analogue)として見なしうる(化5)。そして、前記疎水性コアはESIONと、腫瘍−感知高分子リガンドの疎水性ブロックで構成される。
透過電子顕微鏡(TEM)の写真(図5a)によると前記PMNの粒子の大きさは約60nmだった。前記PMNは水によく分散されており、動的光散乱法(DLS)によって測定された水力学的直径は70±5nmであることが明らかになっており(図5a)、これは受動性腫瘍標的化(passive tumor targeting)のための透過度の増加及び保有の効果(enhanced permeability and retention(EPR)effect)に適合する。対照群として、ESION及びプラットフォームリガンドを組み立ててpH−非感応性ナノ粒子組立体(pH−insensitive nanoparticle assembly(InS−NP)を製造し、(図6)、これはPMNと大きさが類似した。
(実施例22.試験管内及び生体内でのMRI)
Litz Coil(直径、100mm;長さ、85mm;DOTY Scientific Inc.、NC、USA)を使用する1.5 T MRスキャナー(Signa Excite;GE Healthcare)を使用してMRI検査を遂行した。室温の他のpH値の培地内で、他の濃度のPMNに対して、高速スピンエコー(fast spin echo、FSE)シークエンスを使用してスピン−格子及びスピン−スピンの弛緩時間(T1及びT2)を測定した。T1を測定するために、、視界(Field of View、FOV)は75×75mm、スライス厚(SL)=3mm、エコー時間(TE)=9.3msそして反復時間(TR)=505.2、525.0、545.0、565.0、585.0、605.0、625.0、645.0、665.0、685.0、705.0、730.0、755.0、805.0、855.0、905.0、955.0、1005.0、1055.0、1105.0msに設定した。T2測定のため、次のようなパラメータが使用された:FOV=100mm*100mm、SL=3mm、TR=4000ms、TE=10.9、21.7、43.5、54.4、65.2、87.0、119.6、141.3、163.1、174.0ms。垂直弛緩度(relaxivitiy)(R1)及び水平弛緩度(R2)をri=(1/Ti1/Ti0)/cから計算し、前記cはmM単位のPMNの鉄の濃度であり、Tiは、濃度cでの弛緩時間であり、Ti0は水の弛緩の時間で、T1及びT2についてiは1及び2である。FSEシークエンス(FOV=100mm*100mm、SL=3mm)を使用して細胞MR映像を得た。T1を測定するために、TR=400ms及びTE=10msが使用されたが、T2測定のためにはTR=4000ms及びTE=86.72msが使用された。生体内での腫瘍のMR診断のため、2mgFe/kg体重の投与量でPMNを尾静脈に注射した。次に、マウスを1.5 T MRスキャナー(Signa Excite、GE Healthcare)に位置させており、FSEシークエンスは、次のパラメータを使用した:FOV=90mm×90mm、SL=2mm、フリップ角(FA)=90°、T1 MR映像化についてTR=400ms、TE=10ms、及びT2 MR映像化についてTR=3000ms、TE=101ms。横断面の映像を得、Onis Dicom Viewer(DigitalCore、Tokyo、Japan)を使用して分析した。
(実施例23.試験管内及び生体内での光力学治療(PDT))
試験管内でのPDTのため、HCT116細胞(5×104/well)を前述した材料とともに培養した。以後、2分間670nmのレーザーの供給源(Institute of Electronics)で前記細胞を調査することによって光感作の実験を実施した。細胞レベルでの出力を5mW/cm2で固定した。細胞生存率をMTT分析を用いて評価した。吸光度強度をマイクロプレートリーダー(Bio−Tek、VT、USA)を使用して595nmで測定した。

- 外部から照射される光と酸素の作用によって化学作用を引き起こす光感作剤を使用して、治療が要求される特定の組織の細胞を選択して殺すことにより病気を治療するための光力学的自己組立型薬学の組成物であって、
光感作剤と、特定の範囲の水素イオン濃度(pH)条件でpH依存的に分離されるリガンドAと、特定範囲の水素イオン濃度条件で表面電荷が変わるリガンドBと、を含み、
前記光感作剤が、クロリンE6(Ce6)であり、
前記光感作剤は、前記リガンドA及び前記リガンドBに結合されており、
前記リガンドAは、下記の式(I)で表され、

前記pH依存的に分離されるとは、前記特定の範囲の水素イオン濃度(pH)条件において、前記リガンドA及び前記リガンドBの疎水性相互作用が弱まることにより、自己組み立てされた状態から分離されることである、光力学的自己組立型薬学の組成物の製造方法。 - 前記(i)段階の混合溶液の溶媒がDMSO、DMF及びTHFよりなる群から選ばれることを特徴とする、請求項5に記載の光力学的治療用自己組立型薬学の組成物の製造方法。
- 前記(i)段階の有機溶媒がクロロホルム、ジクロロメタン及び1、2―ジクロロエタンよりなる群から選ばれるのを特徴とする、請求項5に記載の光力学的治療用自己組立型薬学の組成物の製造方法。
- 前記リガンドAが
(i)DMFとCH2Cl2の混合物内でβ-ベンジル-L-アスパルテートN-カルボキシ無水物(BLA―NCA)を重合してポリ(エチレングリコール)-ポリ(β-ベンジル-L-アスパルテート)(PEG―PBLA)を製造する段階と、
(ii)前記(i)段階で製造されたPEG―PBLAにクロリンe6(Ce6)を結合させ、プラットフォームのリガンドを製造する段階と、
(iii)前記プラットフォームリガンドを1―(3―アミノプロピル)イミダゾール及びドーパミンを使用してアミノ分解させてリガンドAを製造する段階を含める方法によって製造されたものであることを特徴とする、請求項5に記載の光力学的治療用自己組立型薬学の組成物の製造方法 。 - 前記リガンドBが
(i)DMFとCH2Cl2の混合物内でβ-ベンジル-L-アスパルテートN―カルボキシ無水物(BLA―NCA)を重合してポリ(エチレングリコール)-ポリ(β-ベンジル-L-アスパルテート)(PEG―PBLA)を製造する段階と、
(ii)前記(i)段階で製造されたPEG―PBLAにクロリンe6(Ce6)を結合させ、プラットフォームのリガンドを製造する段階と、
(iii)前記プラットフォームリガンドを1―(3―アミノプロピル)イミダゾール及び3―フェニル―1―プロピルアミンを使用してアミノ分解させてリガンドBを製造する段階を含める方法によって製造されたものであることを特徴とする、請求項5に記載の光力学的治療用自己組立型薬学の組成物の製造方法 。 - 前記(i)段階のリガンド組成物とMRI造影剤用ナノ粒子を混合する段階を追加で含めることを特徴とする、請求項5に記載の光力学的治療用自己組立型薬学の組成物の製造方法 。
- 前記MRI造影剤用ナノ粒子が酸化鉄ナノ粒子であることを特徴とする、請求項10に記載の光力学的治療用自己組立型薬学の組成物の製造方法。
- 前記酸化鉄ナノ粒子の大きさが1nmないし100nmであることを特徴とする、請求項11に記載の光力学的治療用自己組立型薬学の組成物の製造方法 。
