GLP−1(Glucagon−like peptide 1)はブドウ糖依存したインスリン促進分泌ペプチドであり、GLP−1(7−36)アミドとGLP−1(7−37)の2種類の活性形式がある。GLP−1は体内でGタンパク質共役受容体(GPCRs)に属するGLP−1受容体と結合でき、膵島β細胞のインスリン分泌を促進でき、グルカゴンの分泌を抑制でき、血糖をコントロールでき、2型糖尿病を効果的に治療するポリペプチドホルモンであるが、体内でジペプチジルペプチダーゼIV(Dipeptidyl Peptidase IV、DPP−IV)の分解を受け、その半減期が2min程度だけで、臨床使用に高剤量の頻繁注射が必要となり、薬物として臨床使用は厳しく制限されている。
1.担体構築
コードZ1Z2を有する遺伝子配列(例えばSEQ ID NO:5中の73〜663位、以下Z配列と略称)と、コードTelethonin のN末端(TeN)を有する遺伝子配列(例えばSEQ ID NO:1中の73〜348位、以下T配列と略称)を、NcoI/KpnI酵素切断部位を介して改良後のpET24dの担体に(pET24dに6xHisタグとTEVプロテアーゼ酵素切断部位を導入)クローンして、T配列でのCysをSer(Zou PJ et al,J Biol Chem.24 Jan 2003,278(4):2636−2644)に突然変異し、pET−Z1Z2(原pET24d担体以外の配列部分はSEQ ID NO:5の通り)とpET−TeN(原pET24d担体以外の配列部分はSEQ ID NO:3の通り)プラスミッドを得た。
TeN−F:SEQ ID NO:9、ここで、第1〜48位はlinkerコドンであり、
TeN−R:SEQ ID NO:10。
GLP−1−F:SEQ ID NO:11、ここで、5’末端は燐酸化されており、
GLP−1−R:SEQ ID NO:12、ここで、5’末端は燐酸化されている。
pET−Z1Z2、pET−TeNとpET−24d−GLTeをそれぞれE.coli BL21(DE3)コンピテントセルに転化し、シングルコロニーをピックアップして60mg/mlカナマイシン含有のLB培地に入れて、37℃振とうの一晩培養後、1%の接種量で次の1級に切り替え、37℃で菌液OD600が約0.4〜0.6になるまで培養し続けて、最終濃度が1.0mmol/LのIPTGに添加して、30℃で12h誘導発現した。発現時間が終了後、4℃、5000g条件で10min遠心して、菌体を収集した。
それぞれ等体積のTeNとGLTeをZ1Z2に1滴ずつ滴下し、滴下しながら揺れ、最終尿素の濃度が4Mになり、そして、混合溶液を透析液I(25mM Tris/HCl、300mM NaCl、pH 8.0)に4h透析して、透析液II(25mM Tris/HCl,pH8.0)に換えて一晩透析した。最終的に、それぞれZ1Z2/TeN複合体とZ1Z2/GLTe複合体を形成した。
大腸菌で発現したタンパク質に大量の内毒素が含まれ、内毒素は後期の細胞や動物試験に深刻な影響を与えるため、それを除去しなければならない。Qイオン交換カラムで精製したZ1Z2/TeN複合体とZ1Z2/GLTe複合体を内毒素除去カラム(ToxinEraserTM Endotoxin Removal kit、Genscript、Nanjing、China)に添加して内毒素を除去し、内毒素検出キット(ToxinSensorTM Chromogenic LAL Endotoxin Assay Kit、Genscript)で内毒素含有量を検出し、最終的に、Z1Z2/TeN複合体とZ1Z2/GLTe複合体中の内毒素含有量が2EU/mlより小さくなった。
ラットインスリン腫細胞RINm5Fを10%FBS(Life Technology)含有のRPMI 1640培地(Life Technology)に、37℃で、5%CO2培養器に培養した。5000個RINm5F細胞を96ウェル細胞培養プレートに接種し、一晩培養し、血清を含有しないRPMI 1640培地で2回洗浄した後、異なる濃度のZ1Z2/GLTe複合体とGLP−1(血清を含有しない培地で希釈し、100μMのIBMXを添加)をそれぞれRINm5F細胞と37℃で20min孵化し、そして、細胞を分解し、細胞内cAMPの含有量をcAMP−GloTM Assayキット(Promega)説明書により測定した。
Z1Z2/GLTeによるブドウ糖のコントロール作用を確定するために、経口ブドウ糖負荷試験(Oral glucose tolerance test、OGTT)を行った。250gぐらいのラットをランダムにZ1Z2/TeN群、Z1Z2/GLTe群、GLP−1群とPBS群の4群(n=5)に分け、前の3群にそれぞれ体重別に25nmol/kg腹腔注射し、PBS群に等体積のPBSを投与し、血糖計で血糖濃度を測定した。30min後、1kg体重ごとに2gブドウ糖を胃内投与し、このとき0minと計時し、その後10、30、60、90、120minで血糖値を測定した。曲線下面積(Area of under curve、AUC)をGraphpad Prism6.0ソフトウェアで計算した。この過程で、それぞれ0、10、30minで尾静脈から採血し、EDTA−Na2含有の遠心管に添加し、4000rpmで10min遠心して、上清液を取って、Rat/Mouse Insulin Elisa kit(Millipore)でそのインスリン含有量を測定した。
Z1Z2/GLTeとGLP−1はそれぞれ腹腔(4nmol)と静脈(1nmol)でSDラットの体内に注入し、そしてそれぞれ0、0.5、1、1.5、2、4、6、10hで尾から採血して、EDTA−Na2で処理したEp管に滴下し、採血後にすぐに(30s以内)DPP−4抑制剤を添加した。サンプルでの活性GLP−1濃度をActive GLP−1 Elisa kit(Millipore)で測定した。
ストレプトゾトシン(STZ)を45mg/kg体重別にKMマウスに腹腔注射し、5d連続注射してから、正常に10d給餌した。そして、一晩欠食(12〜16h)した後、血糖値を測定し、空腹時血糖値>11.1mMを糖尿病のモデルと判断できる。試験は正常群(即ち、非糖尿病モデル群)、糖尿病マウス対照群(PBS注射)と糖尿病マウス試験群(Z1Z2/GLTe注射)の3群に分け、1群が6匹で、3かご分けて飼育した。試験群では、毎日午前9時、体重で25nmol/kgのZ1Z2/GLTeを腹腔注射するが、対照群と正常群では、それぞれPBSを腹腔注射した。4dごとに一晩欠食(12〜16h)し、第4dで空腹時血糖値を測定した。
1.Z1Z2/GLTe発現精製
図2から分かるように、一連の精製後、Z1Z2/GLTeはSDS−PAGE図でZ1Z2(上)とGLTe(下)の2本の非常に純粋なストライプを示し、Native−PAGE図でZ1Z2/GLTe複合体である1本の非常に純粋なストライプを示した。これは、純度の高い、後続試験に用いるZ1Z2/GLTe複合体を得ることを表明した。
GLP−1Rは細胞表面発現のGタンパク質共役受容体であり、GLP−1は細胞表面発現のGLP−1Rと結合することで、下流の信号経路を活性化にして、cAMPを生成することができる。生成したcAMPの量により、受容体との結合状況を反映することができる。その結果、Z1Z2/GLTeとGLP−1はいずれもGLP−1Rと結合することができ、しかも濃度依存の方式でcAMPの生成を刺激することができる。Z1Z2/GLTeとGLP−1のEC50値はそれぞれ0.35±0.05nMと0.24±0.03nM(図3)である。これは、Z1Z2/GLTeがGLP−1Rと結合でき、かつGLP−1Rを強力に活性化でき、GLP−1Rの興奮剤の一種であることを表明した。
GLP−1は体内で膵島β細胞のインスリン分泌を促進でき、グルカゴンの分泌を抑制でき、血糖をコントロールすることができる。その結果、Z1Z2/GLTeはラット血糖を著しく下げることができる。ブドウ糖を経口摂取したとき、血糖含有量が急激に上がり、GLP−1とZ1Z2/GLTeが血糖値を著しく下げることができるが、GLP−1は30minだけで効果を発揮して、Z1Z2/GLTeは全体の120 min以内で血糖を効果的にコントロールすることができる(図4A、B)。インスリンの生成から見れば、Z1Z2/GLTeとGLP−1はいずれも血液中のインスリン含有量を著しく増えるため、血糖値を下げる作用を効果的に発揮することができる(図4C)。
試験データによると、Z1Z2/GLTeは腹腔注射(図5A)や静脈注射(図5B)を問わず、いずれもGLP−1半減期を著しく延長することができる。Z1Z2/GLTeを腹腔注射した2h後、血漿中の濃度が最大に達するが、GLP−1は0.5hに最大に達し、その後、GLP−1が急速に低下し、1hに測定下限に達し、Z1Z2/GLTeを腹腔注射した24h後も200pM以上の濃度を測定することができる。これは、Z1Z2/GLTeがGLP−1の安定性を著しく増加し、その半減期を延長させ、その作用を発揮する時間を増加することを表明した。静脈注射後、Z1Z2/GLTe濃度は徐々に低下するが、GLP−1は急速に低下し、0.5hに検出下限に達する。GLP−1の静脈注射の半減期は1〜2min(Kim BJ et al,J Pharmacol Exp Ther,2012,334(3):682−692)であるが、本試験にZ1Z2/GLTeは67±3.3minであり、GLP−1半減期を著しく延長した。
Z1Z2/GLTeは原始GLP−1と比べてより長い半減期があるが、その活性は弱まっていないので、GLP−1よりは長い時間の血糖コントロール能力を持つ。糖尿病マウスの血糖のコントロール試験(図6)から分かるように、毎日、25nmol/kgのZ1Z2/GLTeを腹腔注射すると、糖尿病マウスの空腹時血糖レベルを効果的に下げることができる。毎日1回の注射で、空腹時血糖レベルを長期間に低い範囲内にコントロールすることができる。注射が停止した(28d)後、そのZ1Z2/GLTe群の血糖値がまた上昇する。
GLP−1(7−37)はそのアミノ酸配列の前の2つのアミノ酸がHAであるため、血液中にDPP−4の切断作用を受けやすくて活性を失うため、GLP−1(7−37)のアミノ酸配列中の第8位アミノ酸をGlyに突然変異することで、DPP−4による酵素切断を効果的に抑えることができ、半減期をより長く延長することができる。
1.担体構築
下記のプライマーを用いて実施例1での担体pET−24d−GLTeを部位特異的突然変異し、GLP−1(7−37)アミノ酸配列中の第8位アミノ酸AlaをGlyに突然変異した。

シークエンシング成功後、pET−24d−GLTe−Gと名付けて、原pET24d担体以外の配列の部分は以下SEQ ID NO:13の通りであり、そのコードのアミノ酸配列が例えばSEQ ID NO:14である。SEQ ID NO:14で、第25〜26位の間はTEV酵素の酵素切断部位であり、第3〜8位は6xHisタグであり、第25〜55位はGLP−1アミノ酸配列であり、第56〜70位はlinkerであり、第26位は突然変異したアミノ酸である。
実施例1と同じ方法ステップを用いて行い、純度の高いかつ内毒素含有量が2EU/mlより小さいZ1Z2/GLTe−G複合体を得ることができ、他の複合体、例えばZ1Z2/TeN複合体は直接に実施例1での担体やその発現産物や内毒素除去後の産物から得ることができる。
Z1Z2/GLTe−Gが血糖を効果的にコントロールできるかどうか、異なる濃度と血糖コントロールの関係を確定するために、異なる濃度のZ1Z2/GLTe−GのOGTT試験を行った。250gぐらいのSDラットをランダムに1nmol/kg群、5nmol/kg群、25nmol/kg群とPBS群の4群(n=5)に分け、上述の濃度剤量のZ1Z2/GLTe−GとPBSを腹腔注射でそれぞれラットの体内に注入して、30min後に1kg体重ごとに2gのブドウ糖を胃内に投与し、このとき0 minと計時し、その後10、30、60、90、120minにて血糖計で血糖値を測定した。
1nmolのZ1Z2/GLTe−Gをラット尾からラット体内に静脈注射し、このとき0hと計時し、その後、1、2、4、8、24hで採血し、予めEDTA−Na2で処理したEp管に滴下した。サンプルでの活性GLP−1濃度をGLP−1(Total)Elisa kit (Millipore)で測定した。
1.活性測定
異なる濃度のZ1Z2/GLTe−G複合体に対してOGTT試験を行い、その用量依存性の血糖値を下げることができる(図7A)。たとえ1nmol/kgの時でも血糖値を下げる作用を発揮することができ、25nmol/kgの時で、血糖値を下げる作用が最も顕著になる。これは、GLP−1配列中の第8位アミノ酸AをGに突然変異するとき、その活性は影響を受けず、依然として血糖値を下げる作用を発揮することができることを十分に表明した。
Z1Z2/GLTe−G複合体をSDラット体内に静脈注射した後、一定時間ごとに尾から採血して、GLP−1(Total)Elisa Kit(Millipore)で血漿中のZ1Z2/GLTe−Gを測定した(図8)。その低下速度が遅いことを発見し、計算により、その半減期は約4hで、突然変異前(67min)の4倍にもなり、原GLP−1(1〜2min)の約200倍であり、その半減期が大幅に延長され、よりよい臨床使用に基礎を打ち立った。
試験:
1.担体構築
遺伝子全合成の方式を採用して、GLP−1(8G)のコード配列をコードZ1Z2を持つ遺伝子配列のN末端に接続し、改良後のpET24d担体に構築し、pET−24d−GLZと名付ける(原pET24d担体以外の配列部分は以下のSEQ ID NO:17の通りであり、そのコードのアミノ酸配列が例えばSEQ ID NO:18である。SEQ ID NO:18で、第24〜25位の間はTEV酵素の酵素切断部位であり、第3〜8位は6xHisタグであり、第25〜55位はGLP−1(8G)アミノ酸配列であり、第56〜66位はlinkerであり、第26位は突然変異したアミノ酸である)。GLP−1(8G)とZ1Z2との間のLinkerはSEAAAKEAAAKである。
実施例1と同じ方法ステップを用いて行い、純度の高いかつ内毒素含有量が2EU/mlより小さいGLZ/TeN複合体を得ることができ、他の複合体、例えばZ1Z2/GLTe−G複合体は前の実施例から得ることができる。
GLZ/TeN複合体が血糖値を効果的に下げるかどうか、およびZ1Z2/GLTe−G複合体と比べて、その活性はどうでしょうか、OGTT試験を行った。
1nmolのGLZ/TeNをSDラット(250gぐらい)の尾静脈(Intravenous,i.v)と皮下(Subcutaneous,s.c)でラット体内に注入し、このとき0hと計時し、それぞれ0.05、0.5、2、4、8、11、24、48hにラット尾静脈から採血し、予めEDTA−Na2で処理したEp管に滴下し、採血後にすぐに(30s以内)プロティナーゼ抑制剤Aprotininを添加し、サンプルでのGLP−1(8G)濃度をGLP−1(Total)Elisa kit(Millipore)で測定した。
25gぐらいのC57BL/6マウスをランダムにPBS群、GLZ/TeN群とZ1Z2/GLTe−G群の3群(n=5)に分け、各マウスごとに1かごであり、各群のマウスを初日夜の7時から翌日の9時まで欠食し、各群のマウスの空腹体重を秤量し、そしてマウスの鼠餌を早めに計量してパケットし、GLZ/TeN群とZ1Z2/GLTe−G群について、体重別に20nmol/kgのGLZ/TeNとZ1Z2/ GLTe−G複合体を腹腔注射し、PBS群について、等体積のPBSを投与し、このとき0hと計時し、薬物注射後、すぐにあらかじめ計量したマウスの鼠餌を与えて、0、2、5、8、24、36、48hに鼠餌および血糖値を測定した。
C57BL/6マウスを6匹の対照群と15匹の試験群の2群に分け、対照群に正常飼料(Standard)を給餌し、試験群に高糖分高脂飼料(HFD)(1%コレステロール、20%蔗糖、12%ラード、10%卵黄粉と2%コール酸ナトリウム)を給餌し、飼料は北京軍事医学科学院から購入し、対照群と試験群に対して異なる飼料を2ヶ月給餌し、この間で15dごとに体重を秤量し、空腹12h後、対照群(6匹)と試験群をそれぞれ2g/kgのブドウ糖を胃内投与し、OGTTを行い、異なる時点で血糖値を測定し、インスリン抵抗効果を測定し、インスリン抵抗効果があったと、また小剤量で連続5dにSTZ誘導型糖尿病モデルを注射し、引き続き給餌7d後、血糖値を測定して、空腹時血糖値>11.1mMになるとモデル成功と考えられる。試験群からモデル成功の12匹マウスをランダムにGLZ/TeN群とPBS群の2群に分け、20nmol/kgのGLZ/TeN複合体或いは同量のPBSを毎日1回腹腔注射し、血糖値を測定した。
1.発現、精製、内毒素の除去
pET−24d−GLZとpET−TeNプラスミッドをそれぞれE.coli BL21(DE3)に転化して発現させ、GLZ/TeN複合体を形成し、かつ系列操作を経て、純度の高いGLZ/TeN複合体を得た。
GLZ/TeN複合体がZ1Z2/GLTe−Gより強い活性を持つかどうかか、OGTTで直接に対比することができる。OGTT試験結果によると、腹腔注射濃度が10nmol/kgであるとき、GLZ/TeNの活性がZ1Z2/GLTe−Gより著しく高くなる。SDラットにブドウ糖を胃内投与するとき、GLZ/TeNとZ1Z2/GLTe−G群が明らかに血糖値を下げ、GLZ/TeNがZ1Z2/GLTe−Gより高い活性を持ち、血糖を下げる能力が高く、30min時にその最高血糖値がそれぞれ8.3と5.7mMである(図10A)。AUCデータからわかるように、GLZ/TeNとZ1Z2/GLTe−G複合体はいずれもAUCを非常に顕著に下げることができ(図10B)、GLZ/TeNがZ1Z2/GLTe−Gより強い抑制力を持つ。
GLZ/TeN複合体が皮下(s.c)や静脈(i.v)でSDラットに注射された後、GLP−1 ELISAキットで血漿中のGLP−1含有量を測定した。その含有量の低下が遅いことを発見し、計算により、皮下注射の半減期は19.8±3.2hであり、静脈注射の半減期は18.7±2.3hである(図11)。その静脈注射の半減期はZ1Z2/GLTe−G複合体(4h)の4倍でもあり、さらに向上した。これは、GLZ/TeN複合体に2つのGLP−1分子があり、その分子量をさらに増えて、腎臓の濾過作用を緩め、しかもそれとGLP−1Rの結合の親和性がより大きくなり、その半減期が長くなるためである。
1回で体重別に薬物を腹腔注射した後、結果によると、Z1Z2/GLTe−GおよびGLZ/TeN複合体はいずれも8h内でマウスの摂食量を著しく抑えることができ、24h後、抑制作用が消失するが、Z1Z2/GLTe−G群がGLZ/TeN群と比べ、明らかな差異がない(図12A)。
HFD飼料で2ヶ月連続給餌すると、体重から分かるように(図13A)、マウスの体重は2ヶ月内で正常飼料給餌の対照群より明らかに増えた。
試験:
1.担体構築
まず、遺伝子全合成の方式でGLP−1(8G)のコード配列を(G4S)3linkerのコード配列によりZ1Z2を持つコード配列のN末端に接続し、改良後のpET24d担体に構築して、pET−24d−GLZ−synと名付ける。そして、プライマーを設計することにより、それぞれpET−24d−GLZ−syn、pET−TeN、pET−Z1Z2をテンプレートとして新しい発現担体を構築し、そのN末端がGLP−1(8G)付きのZ1Z2、Telethonin分子のN末端のβ−プリーツシート領域およびZ1Z2分子の間をlinkerで接続し、1つの大きい融合タンパク質をワンステップ発現して形成することができる。
pET−24d−GLRecomプラスミッドをE.coli BL21(DE3)菌株に転化し、1mM ITGの誘導下で、16℃振とうに24h発現し、GLRecomタンパク質が上清液に発現させ、後でTEV酵素切断、二次Niカラム、Qカラム精製などの過程を経て、純度の高いGLRecomタンパク質を得、そして、実施例1と同じ方法ステップを用いて内毒素の除去を行い、純度の高いかつ内毒素含有量が2EU/mlより小さいGLRecomタンパク質を得た。
GLRecomが血糖値を効果的に下げるかどうか、およびZ1Z2/GLTe−Gと比べて、その活性はどうでしょうか、OGTT試験を行った。
2.8nmol GLRecomをSDラット(250gぐらい)の尾静脈(Intravenous,i.v)でラット体内に注入し、このとき0hと計時し、それぞれ0.05、0.5、2、5、10、24、48hにラット尾静脈から採血し、予めEDTA−Na2で処理した遠心管に滴下し、採血後、すぐに(30s以内)プロティナーゼ抑制剤Aprotininを添加し、サンプルでのGLP−1(8G)濃度をGLP−1(Total)Elisa kit(Millipore)で測定した。
1.発現、精製、内毒素の除去
GLRecomタンパク質で、GLP−1(8G)付きの一番目のZ1Z2分子とTelethoninβ−プリーツシート領域の間にSGSGSAPGTPGGGGSGGGGSGGGGSをlinkerとして設計し、Telethoninβ−プリーツシート領域と二番目のZ1Z2分子の間にGASGPAGSPTGSGPGSAGSGPGSAGをlinkerとして設計し、組換えタンパク質を形成し、発現、精製ステップの減少に有利するため、精製に必要な時間やコストを減らすことができる。GLRecomタンパク質は一連の精製過程を経た後、分子量が60.0 kDaのタンパク質を得(図14)、上清液に発現させ、操作は簡単で、しかも純度の高いタンパク質を得やすい。
GLRecomとZ1Z2/GLTe−G複合体にいずれも1つのGLP−1分子が含まれ、そのGLP−1分子中のN末端のAlaがGlyに置換される。OGTT結果によると、GLRecomとZ1Z2/GLTe−G複合体はいずれも血糖値を著しく下げる作用がある。マウスに2g/kgブドウ糖を投与した後、PBS群のマウスの血糖値が急速に上昇するが、GLRecomとZ1Z2/GLTe−G複合体群はPBS群より顕著に低下する。120minになると、GLRecomとZ1Z2/GLTe−G複合体群も血糖値レベルを著しく下げることができる(図15A)。AUCデータによると、GLRecomとZ1Z2/GLTe−G複合体群がマウスの血糖レベルを顕著に下げ、マウスの経口ブドウ糖耐性を増加させることを直感的に表明し(図15B)、GLRecomとZ1Z2/GLTe−G複合体が血糖値レベルを下げる点で差異がない原因は、その構造中に1つだけのGLP−1分子がある可能である。
GLRecomタンパク質が血液中に長い半減期を有し、濃度の低下が遅くて(図16)、その半減期を計算すると、13.4±0.8hであり、Z1Z2/GLTe−G複合体(4h)の3倍以上でもある。これは、GLRecomが大きな分子量(60.0kDa)を有するが、Z1Z2/GLTe−Gの分子量が56.7kDaであり、または、GLRecomタンパク質中の分子間のlinkerがフレキシブルの非常に高いLoopを形成するためであり、コンパクトなZ1Z2/GLTe−Gに対して、これらのlinkerはGLRecomの体積を増え、プロティナーゼの分解をより阻止し、腎臓による濾過作用をより緩め、その半減期を延長することができる。
ソマトスタチン(Somatostatin,SST)は環状ポリペプチドであり、中枢神経系、末梢神経系や胃腸の中で広く分布され、成長ホルモンの分泌と放出を著しく抑制することができ、膵臓ホルモン、胃泌素などの分泌を抑制することもできる。SSTがインスリンやグルカゴンの分泌をコントロールすることができる。SSTはSST14とSST28の2つの活性形式がある。SSTはGPCRsに属するSST受容体と結合して、下流の通路を活性化する。主に末端肥大症、肝硬変門脈高圧による食道静脈出血、急性膵炎およびその合併症、並びに膵液瘻、胆汁瘻および腸瘻などを治療する。しかし、その体内の半減期は2−3min(Afargan M et al,Endocrinology, 2001,142(1):477−486)だけで、その臨床使用は厳しく制限されている。

遺伝子全合成の方式で、それぞれSST遺伝子をTeN突然変異体Loop(LT1)中の遺伝子配列に挿入する担体pET−24d−SSTe(原pET24d担体以外の配列部分はSEQ ID NO:27の通りであり、そのコードのアミノ酸配列が例えばSEQ ID NO:28である。SEQ ID NO:28で、第25〜26位の間はTEV酵素の酵素切断部位であり、第3〜8位は6xHisタグであり、第43〜54位はGLP−1アミノ酸配列であり、第57〜58位はlinkerである。)を得た。
実施例1と同じ方法ステップを用いて行い、純度の高いかつ内毒素含有量が2EU/mlより小さいZ1Z2/SSTe複合体を得ることができる。
250gぐらいのSDラットを12〜16h欠食した後、ランダムに3群分け、2群は試験群であり、体重別に200nmol/kgのSSTとZ1Z2/SSTe複合体を腹腔注射し、もう1群は等体積のPBSを注射した。0、30、60、120min時にそれぞれ尾から採血し、EDTA−Na2含有の遠心管に添加し、4000rpmで10min遠心して、上清液を取った。上清液での成長ホルモンの濃度をGrowth hormone ELISA kit(Millipore)キットで測定した。
Z1Z2/SSTe 0.28mgを尾静脈からラットの体内に注入し、それぞれ0、0.5、1、2、4、8、24hで尾静脈から採血し、EDTA−Na2含有の遠心管に添加し、4000rpmで10min遠心して、上清液を取った。上清液でのZ1Z2/SSTe中のSSTをSomatostatin (SST) EIA kit (Phoenix pharmaceuticals)で測定した。
1.発現、精製、内毒素の除去
図17から分かるように、一連の精製後、Z1Z2/SSTeはSDS−PAGE図で2本の非常に純粋なストライプを示すが、Native−PAGE図でZ1Z2/SSTe複合体である1本の非常に純粋なストライプを表しました。これは、純度の高い、後続試験に用いるZ1Z2/SSTe複合体を得ることを表明した。
SSTが体内で成長ホルモンの分泌を抑えることができるので、ラット体内の成長ホルモンの含有量を測定することにより、Z1Z2/SSTe複合体の活性を検証することができる。Z1Z2/SSTe複合体がラット体内に注入された後、ラット成長ホルモンのレベルを著しく抑制した。SSTおよびZ1Z2/SSTeがラット体内に注入された後、初期ではいずれも体内成長ホルモンの分泌を明らかに抑制するが、SSTの半減期(2〜3min)が短いため、SSTの分解につれて、その抑制効果を次第に弱め、最後に2hで対照レベルまで回復し、Z1Z2/SSTeが120min内で成長ホルモンのレベルを明らかに抑制し、より長い半減期を有することを説明した(図18)。それは原SSTと類似する生物学的活性を持ち、かつ成長ホルモンへの抑制作用をより長い期間で発揮することができることを表明した。
Z1Z2/SSTe複合体の静脈注射後、血漿中のSST含有量が図19に示すように、その計算の半減期は6.62±0.43hであり、原SST(2〜3 min)より約200倍に延長した。これは、Z1Z2/SSTe複合体はSSTとその受容体の結合に影響しない場合で、その半減期を大幅に延長して、より長く薬効を発揮させることができることを表明した。
試験:

遺伝子全合成の方式で、それぞれSST遺伝子をZ1Z2分子のLoop中の遺伝子配列に挿入する担体pET−24d−ZSST(原pET24d担体以外の配列部分は以下のSEQ ID NO:29の通りであり、そのコードのアミノ酸配列が例えばSEQ ID NO:30である。SEQ ID NO:30で、第24〜25位の間はTEV酵素の酵素切断部位であり、第3〜8位は6xHisタグであり、第171〜184位はSSTアミノ酸配列であり、第185〜186位はlinkerである。)を得た。
実施例1と同じ方法ステップを用いて行い、純度の高いかつ内毒素含有量が2EU/mlより小さいZSST/TeN複合体を得ることができる。

1.発現、精製、内毒素の除去
SDS−PAGE図で2本の非常に純粋なストライプを示すが、Native−PAGE図で1本の非常に純粋なストライプを示し、純度の高いZSST/TeN複合体を得ることを表明した。ZSST/TeN複合体の内毒素は内毒素除去樹脂で処理された後、含有量を2EU/mlの範囲内で制御し、後続の試験に使うことができる。
ZSST/TeN複合体がSDラット体内に腹腔注入された後、ラット成長ホルモン(GH)のレベルを著しく抑制した。SSTおよびZ1Z2/SSTeはラット体内に注入された後、短い時間内で成長ホルモンの分泌を抑制するが、SSTの半減期(2〜3min)が短くて、抑制作用が弱いため、2h後に対照レベルまで回復し、Z1Z2/SSTeは4h内で成長ホルモンのレベルを明らに抑制することができ、8h後に対照レベルまで回復し、より長い半減期を有する。そして、ZSST/TeN複合体が12hに成長ホルモンの分泌を著しく抑制することができ、より長い半減期を有することを表明した(図20)。ZSST/TeN複合体が2つのSST分子を有するので、より長い半減期を有するとともに、より強い成長ホルモンの分泌の抑制作用を有する。
ZSST/TeN複合体がSDラットに静脈注射された後、血漿中のSST含有量がELISAキットで測定し(図21)、その計算の半減期は14.3±1.7hであり、原SST(2〜3min)より数百倍も延長し、Z1Z2/SSTe複合体よりも大幅に延長した。これは、ZSST/TeN複合体はSSTとその受容体の結合に影響しない場合で、その半減期を顕著に延長することを表明した。
PYY(peptide Tyrosin−tyrosin)は腸のL−細胞の摂食後で分泌した36個のアミノ酸のポリペプチドである。研究によると、その体内でPYY1−36とDPP−4酵素切断後のPYY3−36の2つの形式がある。PYY3−36は視床下部と外週神経系のY2受容体に作用することにより、食欲の抑制、胃排出の緩め、体重の軽減の作用を発揮させる。動物と人体の試験をしたところ、PYY3−36の外週注射が摂食量を明らかに減少することができる。よって、PYY3−36が肥満治療の有効な薬物と認められる。しかし、PYYは体内で、プロティナーゼの分解と腎臓の濾過作用で、半減期が短くて、8minだけあるため、その臨床使用に制限した。

遺伝子全合成の方式で、PYY3−36コード配列をTelethoninβ−プリーツシート領域のコード配列のC末端の遺伝子配列に接続する担体pET−24d−TePY(原pET24d担体以外の部分はSEQ ID NO:31の通りであり、そのコードのアミノ酸配列が例えばSEQ ID NO:32である。SEQ ID NO:32で、第24〜25位の間はTEV酵素の酵素切断部位であり、第3〜8位は6xHisタグであり、第120〜153位はPYYアミノ酸配列であり、第117〜119位はlinkerである。)を得た。
実施例1と同じ方法ステップを用いて行い、純度の高いかつ内毒素含有量が2EU/mlより小さいZ1Z2/TePY複合体を得ることができる。
Z1Z2/TePY複合体が体内で摂食を抑制する作用を発揮できるかどうかを確定するために、オスKMマウスを用いて摂食試験を行った。30gぐらいのKMマウスをランダムにPBS群とZ1Z2/TePY群の2群に分け、そして夜6時から翌朝9時まで15h欠食させ、各マウスの空腹体重;各々マウス空腹体重別にZ1Z2/TePY群に200nmol/kg薬物を腹腔注射し、PBS群に等体積のPBSを投与し、このとき0hにして、すぐ事前に重量が秤量した食べ物を投与し、しばらくの時間ごとに食べ物を秤量し、各時間帯のマウスの摂食量/体重をもってマウスの摂食量と表す。
9.0nmol Z1Z2/TePY複合体をSDラット(250gぐらい)の尾静脈(Intravenous,i.v)からラット体内に注入し、このとき0hと計時し、それぞれ0.05、1、2、4、7、10、24hにラット尾静脈から採血し、予めEDTA−Na2で処理した遠心管に滴下し、採血後、すぐに(30s以内)プロティナーゼ抑制剤Aprotininを添加し、サンプルでのPYY濃度をPYY3−36 EIA kit(Phoenix)で測定した。
1.発現、精製、内毒素の除去
pET−24d−TePYがそれぞれE.coli BL21(DE3)に転化され、IPTGで誘導発現を行った。そして、それぞれNi2+−NTA親和クロマトグラフィーカラム精製、TEVプロティナーゼ切断、Ni2+−NTA親和クロマトグラフィーカラム二次精製、Qイオン交換カラム精製や内毒素除去などのステップを経て、最終的に純度の高いタンパク質複合体Z1Z2/TePYを得、後続の動物試験に使うことができる。
Z1Z2/TePY 200nmol/kgをマウスの体内に腹腔注射した後、8h内で摂食量(p<0.05)を顕著に低下させることができるが、8h後にその抑制作用が消えることを発見した(図22)。
測定によると、Z1Z2/TePY複合体が体内で濃度の低下が遅くて、その半減期を計算すると、静脈注射では6.4±0.8hであり、天然PYY(8min)より大きく延長した(図23)。
Claims (16)
- ポリペプチドまたはタンパク質薬物とポリペプチドAおよび/またはポリペプチドBで構成する融合タンパク質を含み、この融合タンパク質とポリペプチドA、ポリペプチドBまたは別の前記融合タンパク質がβ―プリーツシート構造を形成し、前記ポリペプチドAはtitin分子のN末端の2つのIgドメイン(Z1Z2)を含むポリペプチドであり、前記ポリペプチドBはTelethonin分子のN末端のβ―プリーツシート領域を含むポリペプチドであることを特徴とする薬用作用を有する融合タンパク質複合体。
- 前記融合タンパク質で、ポリペプチドまたはタンパク質薬物がポリペプチドAおよび/またはポリペプチドBの1つまたは複数の下記部位、即ちポリペプチドBのN末端(NT)、C末端(CT)および内部の2つのloop領域、2つのポリペプチドAのそれぞれのN末端、C末端および4つのloop領域に位置することを特徴とする請求項1に記載の薬用作用を有する融合タンパク質複合体。
- 前記ポリペプチドAはSEQ ID NO:6中の31〜221位に示すアミノ酸配列を含む断片、その保守的変異体、生物活性断片または誘導体であり、前記ポリペプチドBはSEQ ID NO:2中の30〜116位に示すアミノ酸配列を含む断片、その保守的変異体、生物活性断片または誘導体であることを特徴とする請求項1に記載の薬用作用を有する融合タンパク質複合体。
- 前記ポリペプチドAはSEQ ID NO:6中の31〜221位に示すアミノ酸配列に対して少なくとも60%相同性を有する配列を含むポリペプチド断片であり、前記ポリペプチドBはSEQ ID NO:2中の30〜116位に示すアミノ酸配列に対して少なくとも60%相同性を有する配列を含むポリペプチド断片であることを特徴とする請求項1に記載の薬用作用を有する融合タンパク質複合体。
- GLP−1、GLP−1(8G)、SSTまたはPYYとポリペプチドAおよび/またはポリペプチドBで構成する融合タンパク質を含み、この融合タンパク質とポリペプチドA、ポリペプチドBまたは別のGLP−1、GLP−1(8G)、SSTまたはPYYとポリペプチドAおよび/またはポリペプチドBで構成する融合タンパク質がβ―プリーツシート構造を形成し、前記ポリペプチドAはtitin分子のN末端の2つのIgドメイン(Z1Z2)を含むポリペプチドであり、前記ポリペプチドBは、Telethonin分子のN末端のβ―プリーツシート領域を含むポリペプチドであることを特徴とする請求項1に記載の薬用作用を有する融合タンパク質複合体。
- 前記融合タンパク質複合体は、SEQ ID NO:8またはSEQ ID NO:4中の第25〜161位に示すアミノ酸配列を含む断片、その保守的変異体、生物活性断片または誘導体および2つのSEQ ID NO:6中の31〜221位に示すアミノ酸配列を含む断片、その保守的変異体、生物活性断片または誘導体が結合して構成された、β―プリーツシート構造を有する複合体であることを特徴とする請求項5に記載の薬用作用を有する融合タンパク質複合体。
- 前記融合タンパク質複合体は、2つのSEQ ID NO:18中の第25〜260位に示すアミノ酸配列を含む断片、その保守的変異体、生物活性断片または誘導体および1つのSEQ ID NO:2またはSEQ ID NO:4中の30〜116位に示すアミノ酸配列を含む断片、その保守的変異体、生物活性断片または誘導体が結合して構成された、β―プリーツシート構造を有する複合体であることを特徴とする請求項5に記載の薬用作用を有する融合タンパク質複合体。
- 前記融合タンパク質複合体は、SEQ ID NO:28中の第25〜132位に示すアミノ酸配列を含む断片、その保守的変異体、生物活性断片または誘導体および2つのSEQ ID NO:6中の31〜221位に示すアミノ酸配列を含む断片、その保守的変異体、生物活性断片または誘導体が結合して構成された、β―プリーツシート構造を有する複合体であることを特徴とする請求項5に記載の薬用作用を有する融合タンパク質複合体。
- 前記融合タンパク質複合体は、2つのSEQ ID NO:30中の第25〜237位に示すアミノ酸配列を含む断片、その保守的変異体、生物活性断片または誘導体および1つのSEQ ID NO:2またはSEQ ID NO:4中の30〜116位に示すアミノ酸配列を含む断片、その保守的変異体、生物活性断片または誘導体が結合して構成された、β―プリーツシート構造を有する複合体であることを特徴とする請求項5に記載の薬用作用を有する融合タンパク質複合体。
- 前記融合タンパク質複合体は、SEQ ID NO:32中の第25〜153位に示すアミノ酸配列を含む断片、その保守的変異体、生物活性断片または誘導体および2つのSEQ ID NO:6中の31〜221位に示すアミノ酸配列を含む断片、その保守的変異体、生物活性断片または誘導体が結合して構成された、β―プリーツシート構造を有する複合体であることを特徴とする請求項5に記載の薬用作用を有する融合タンパク質複合体。
- 薬用作用を有する融合タンパク質であって、前記融合タンパク質は、直接的または間接的に順次に接続した第一ポリペプチドAと、ポリペプチドBと、第二ポリペプチドAとを含み、任意の前記ポリペプチドAおよび/またはポリペプチドBの1つまたは複数の部位に挿入、または接続した1つまたは複数のポリペプチドまたはタンパク質薬物を含み、前記2つのポリペプチドAとポリペプチドBがフレキシブルlinkerを介してβ―プリーツシート構造を形成し、前記第一ポリペプチドAおよび第二ポリペプチドAは、titin分子のN末端の2つのIgドメイン(Z1Z2)を含むポリペプチドであり、前記ポリペプチドBは、Telethonin分子のN末端のβ―プリーツシート領域を含むポリペプチドであることを特徴とする薬用作用を有する融合タンパク質。
- ポリペプチドまたはタンパク質薬物が任意の前記ポリペプチドAおよび/またはポリペプチドBの1つまたは複数の下記部位、即ちポリペプチドBのN末端(NT)、C末端(CT)および内部の2つのloop領域、2つのポリペプチドAのそれぞれのN末端、C末端および4つのloop領域に位置することを特徴とする請求項11に記載の薬用作用を有する融合タンパク質。
- 前記第一ポリペプチドAおよび/または第二ポリペプチドAはSEQ ID NO:6中の31〜221位に示すアミノ酸配列を含む断片、その保守的変異体、生物活性断片または誘導体であり、前記ポリペプチドBはSEQ ID NO:2中の30〜116位に示すアミノ酸配列を含む断片、その保守的変異体、生物活性断片または誘導体であることを特徴とする請求項11に記載の薬用作用を有する融合タンパク質。
- 前記第一ポリペプチドAおよび/または第二ポリペプチドAはSEQ ID NO:6中の31〜221位に示すアミノ酸配列に対して少なくとも60%相同性を有する配列を含むポリペプチド断片であり、前記ポリペプチドBはSEQ ID NO:2中の30〜116位に示すアミノ酸配列に対して少なくとも60%相同性を有する配列を含むポリペプチド断片であることを特徴とする請求項11に記載の薬用作用を有する融合タンパク質。
- 前記融合タンパク質は、直接的または間接的に順次に接続した第一ポリペプチドAと、ポリペプチドBと、第二ポリペプチドAとを含み、任意の前記ポリペプチドAおよび/またはポリペプチドBの1つまたは複数の部位に挿入、または接続した1つまたは複数のGLP−1、GLP−1(8G)、SSTまたはPYYのポリペプチドまたはタンパク質薬物を含み、前記2つのポリペプチドAとポリペプチドBがフレキシブルlinkerを介してβ―プリーツシート構造を形成し、前記第一ポリペプチドAおよび第二ポリペプチドAは、titin分子のN末端の2つのIgドメイン(Z1Z2)を含むポリペプチドであり、前記ポリペプチドBは、Telethonin分子のN末端のβ―プリーツシート領域を含むポリペプチドであることを特徴とする薬用作用を有する請求項11に記載の薬用作用を有する融合タンパク質。
