複数の反応成分を反応容器へ添加して反応混合物を形成させる段階と、
反応混合物を撹拌しその混合を確かにするために、反応混合物の一部を反応容器から引き出し及び当該引き出し分を反応容器へ戻す段階と、
反応終了時に反応生成物を反応容器から引き出す段階と、を備えている合成の方法が提供されている。

Claims (14)
- 第1の下側ポート及び第2の上側ポートを有する反応容器と、
前記反応容器の温度を制御するための温度制御器と、
反応成分を保持するための複数の貯留部と、
複数の導管であって、当該複数の導管の各々が前記反応容器の前記第2の上側ポートを前記複数の貯留部のそれぞれ1つと接続している、複数の導管と、
前記第1の下側ポート及び/又は前記反応容器の内容物の圧力を測定するための少なくとも1つの圧力センサと、
前記複数の貯留部に保持されている反応成分の制御量を前記反応容器へ所望される時々に選択的に供給するための供給手段と、
前記反応容器からその前記第1の下側ポートを通して内容物の或る量を選択的に引き出し、当該引き出された内容物を前記反応容器へその前記第1の下側ポートを通して選択的に戻すための撹拌手段と、
前記供給手段及び前記撹拌手段を制御するための制御手段と、を備えている合成装置において、
前記制御手段は、前記少なくとも1つの圧力センサによる測定圧力に基づいて反応の終了または反応の段階を検出して前記供給手段を制御し、および/または前記制御手段は、前記少なくとも1つの圧力センサによる測定圧力に基づいて核酸材料が損傷しないように前記撹拌手段を制御する、核酸の合成装置。 - 前記反応容器は、その最下点に向かって先細になってゆき入口/出口ポートを前記最下点に有するように配列されている、請求項1に記載の装置。
- 前記反応容器のアスペクト比は、5:1から8:1の範囲にあり、好適には6:1から7:1の範囲にある、請求項1又は2に記載の装置。
- 前記温度制御器は、反応成分の添加及び/又は材料の引き出しと同期して、前記反応容器の温度を制御して所望の温度プロファイルをなぞらせるように配列されている、請求項1から3の何れか一項に記載の装置。
- 前記貯留部の少なくとも1つに保持されている反応成分の温度を制御するように配列されている貯留部温度制御器を更に備えている請求項1から4の何れか一項に記載の装置。
- 前記温度制御器は、サーモサーキュラー、ペルチェ加熱/冷却素子、及び/又は抵抗加熱器を備えている、請求項1から5の何れか一項に記載の装置。
- 前記供給手段及び前記撹拌手段は、往復シリンジポンプを備えている、請求項1から6の何れか一項に記載の装置。
- 前記供給手段及び/又は前記撹拌手段は、当該供給手段及び/又は当該撹拌手段を前記反応容器の前記第1の下側ポート又は第2の上側ポート及び/又は貯留部へ選択的に接続するように配列されている多方向弁を備えている、請求項1から7の何れか一項に記載の装置。

少なくとも1つの圧力センサによって前記反応容器の外から前記反応混合物の圧力を測定し、前記測定された圧力に基づいて反応の終了または反応の段階を検出して前記複数の反応成分の前記反応容器への添加を制御する、及び/又は前記測定された圧力に基づいて核酸材料が損傷しないように前記反応容器から引き出し及び前記反応容器へ戻す速度を制御する段階と、
前記反応の終了時に反応生成物を前記反応容器から引き出す段階と、を備えている核酸の合成の方法。 - 前記反応容器の前記下側ポートは、前記反応容器の下側部分に、好適には前記反応容器の最下点に設けられている、請求項10に記載の方法。
- 前記反応混合物の前記引き出し分は、前記反応混合物全体の50%又はそれ以下、望ましくは40%又はそれ以下、好適には30%又はそれ以下に及ぶ、請求項10又は11に記載の方法。
- 前記反応混合物の前記引き出し分は、前記反応混合物全体の5%又はそれ以上、望ましくは10%又はそれ以上、好適には20%又はそれ以上に及ぶ、請求項10、11又は12に記載の方法。
- 前記反応の終了に先立って前記反応混合物の試料を引き出す段階、を更に備えている請求項10から13の何れか一項に記載の方法。
