培養空間が400mm3以下の大きさであること、及び、
培養空間の体積をVmm3、細胞が移動可能な空間の中に存在する間葉系細胞の細胞数をNとしたとき、N/V比が35以上3000以下であること;
培養空間の培地中に存在する間葉系細胞の割合、及び
間葉系細胞の移動距離可能な領域を制限すること。

Claims (12)
- いずれも未分化の内胚葉細胞、外胚葉細胞及び中胚葉細胞からなる群から選択される一種以上の細胞である幹細胞由来細胞と、
血管内皮細胞と、
未分化の間葉系細胞と、
を含む三以上の細胞からなる集団を、所定の領域内で三次元培養する、
培養方法であって、
前記領域は細胞が移動可能な空間からなり、
前記空間の体積をV mm3とし、前記空間に播種される前記間葉系細胞の細胞数をNとしたとき、前記Vは1以下であり、かつN/Vは35以上3000以下であり、
前記集団は細胞数Xの前記幹細胞由来細胞と細胞数Yの前記間葉系細胞とからなり、
X:Yは20:1から100:1の範囲内にある、
ことを特徴とする培養方法。 - 培養に供する細胞の総数に対する前記間葉系細胞の数の割合は、0.5%以上5%未満である、
ことを特徴とする請求項1に記載の培養方法。 - いずれも未分化の内胚葉細胞、外胚葉細胞及び中胚葉細胞からなる群から選択される一種以上の細胞である幹細胞由来細胞と、
血管内皮細胞と、
未分化の間葉系細胞と、
を含む三以上の細胞からなる集団を、所定の領域内で三次元培養する、
培養方法であって、
前記領域は細胞が移動可能な空間からなり、
前記空間の体積をV mm3とし、前記空間に播種される前記間葉系細胞の細胞数をNとしたとき、前記Vは1以下であり、かつN/Vは35以上3000以下であり、
培養に供する細胞の総数に対する前記間葉系細胞の数の割合は、0.5%以上2.9%未満である、
ことを特徴とする培養方法。 - 前記集団は細胞数Xの前記幹細胞由来細胞と細胞数Yの前記間葉系細胞とからなり、
X:Yは20:1から100:1の範囲内にある、
ことを特徴とする請求項3に記載の培養方法。 - 前記領域はマイクロ容器で形成されており、
前記N/Vは100以上300以下であり、
前記X:Yは20:1から50:1の範囲内にあり、
前記マイクロ容器の相当直径をEとし、前記マイクロ容器の深さをDとしたとき、E:Dは1:0.5から1:2の範囲内である、
ことを特徴とする請求項1、2及び4のいずれか一項に記載の培養方法。 - 前記三次元培養によって得られる細胞塊は器官芽である、
ことを特徴とする請求項1乃至5のいずれか一項に記載の培養方法。 - 前記三次元培養によって得られる細胞塊はスフェロイド形状であり、
前記スフェロイド形状の細胞塊の直径は20μmから2mmである、
ことを特徴とする請求項1乃至6のいずれか一項に記載の培養方法。 - 前記幹細胞由来細胞は胎生幹細胞又は人工多能性幹細胞に由来する、
ことを特徴とする請求項1乃至7のいずれか一項に記載の培養方法。 - 前記幹細胞由来細胞は人工多能性幹細胞に由来し、
前記幹細胞由来細胞は内胚葉細胞である、
ことを特徴とする請求項1乃至8のいずれか一項に記載の培養方法。 - 前記領域はマイクロ容器で形成されており、
前記マイクロ容器の相当直径は20μm以上2.5mm以下であり、
前記マイクロ容器の深さは20μm以上1000μm以下である、
ことを特徴とする請求項1乃至9のいずれか一項に記載の培養方法。 - 前記マイクロ容器は底部、及びホーン部を備え、
前記ホーン部は壁を有し、
前記壁は1度以上20度以下のテーパ角を有し、
前記底部は半球形状又は円錐台形状の空間を有する、
ことを特徴とする請求項10に記載の培養方法。 - 前記マイクロ容器は、細胞と接触する培養面を有し、
前記培養面は、リン脂質、リン脂質・高分子複合体、ポリ(2−ヒドロキシエチルメタクリレート)(PHEMA)、ポリビニルアルコール、アガロース、キトサン、ポリエチレングリコール、及びアルブミン、のグループから選択される一つ又はこれらの組合せからなるポリマーでコートされている、
ことを特徴とする請求項10又は11に記載の培養方法。
