Description
前記骨格構造体の表面に形成された乳酸−グリコール酸共重合体を含むコート層と、を有する細胞工学用支持体の製造方法であって、
ポリ−D−乳酸、またはポリ−L−乳酸を含む顆粒状の多孔質体である第1の顆粒状多孔質物質と、乳酸−グリコール酸共重合体を含む顆粒状の多孔質体である第2の顆粒状多孔質物質とを混合して混合体を調製する混合工程と、
前記混合体を型に入れ加圧・加熱する加圧・加熱工程と、を有し、
前記加圧・加熱工程では、前記乳酸−グリコール酸共重合体の融点以上、前記第1の顆粒状多孔質物質に含まれるポリ−D−乳酸、またはポリ−L−乳酸の融点未満まで加熱する細胞工学用支持体の製造方法を提供する。
[第1の実施形態]
本実施形態では細胞工学用支持体の製造方法の一構成例について説明する。
(混合工程)
混合工程においては、ポリ−D−乳酸、またはポリ−L−乳酸を含み、顆粒状の多孔質体である第1の顆粒状多孔質物質と、乳酸−グリコール酸共重合体を含み、顆粒状の多孔質体である第2の顆粒状多孔質物質とを混合して混合体を調製できる。
(加圧・加熱工程)
次に、混合体を型に入れ加圧・加熱する加圧・加熱工程について説明する。
[第2の実施形態]
次に、本実施形態の細胞工学用支持体の製造方法の他の構成例について説明する。
(骨格構造体準備工程)
まず、骨格構造体準備工程について説明する。骨格構造体準備工程では、ポリ−D−乳酸、またはポリ−L−乳酸を含む骨格構造体を準備することができる。
60℃以上とすることで、ポリ−D−乳酸、またはポリ−L−乳酸同士の結合を十分に高めることができ、形状安定性を特に高めることができる。また、200℃以下とすることで、ポリ−D−乳酸、またはポリ−L−乳酸が変性することを確実に防ぐことができる。
(熱処理工程)
次に熱処理工程について説明する。熱処理工程では、骨格構造体の表面に乳酸−グリコ−ル酸共重合体の粉末を分散した後、熱処理を行うことができる。
[実施例1]
以下の手順により細胞工学用支持体を作製した。
(混合工程)
まず、以下の手順により、ポリ−L−乳酸からなり、顆粒状の多孔質体である第1の顆粒状多孔質物質を準備した。
(加圧・加熱工程)
得られた混合体を内径9mm×高さ10mmのガラス容器内に高さが約7mmとなるように入れた。そして、直径9mmのチタン棒で1500g/cm2で加圧した状態の体積を保って50℃で20分間加熱して、直径9mm×高さ約7mmの円柱形状をした細胞工学用支持体が得られた。
[実施例2]
第1の顆粒状多孔質物質として、ポリ−D−乳酸からなる顆粒状の多孔質体を用いた点以外は、実施例1と同様にして細胞工学用支持体を作製し、評価を行った。
[実施例3]
以下の手順により細胞工学用支持体を作製した。
(骨格構造体準備工程)
まず、以下の手順により、ポリ−L−乳酸を含有する骨格構造体を準備する骨格構造体準備工程を実施した。
(熱処理工程)
次に得られた骨格構造体を熱処理するための加熱炉内に配置し、該骨格構造体の表面に、平均粒径が200μmの乳酸−グリコ−ル酸共重合体粉末を分散した。なおこの際、骨格構造体と、分散する乳酸−グリコール酸共重合体粉末との質量の合計のうち、分散する乳酸−グリコール酸共重合体粉末の割合が20質量%となるように、乳酸−グリコール酸共重合体粉末を分散した。
[実施例4]
ポリ−D−乳酸を含有する骨格構造体を用いた点以外は実施例3と同様にして、細胞工学用支持体を作製し、評価を行った。

Claims (2)
- ポリ−D−乳酸、またはポリ−L−乳酸を含む骨格構造体と、
前記骨格構造体の表面に形成された乳酸−グリコール酸共重合体を含むコート層と、を有する細胞工学用支持体の製造方法であって、
ポリ−D−乳酸、またはポリ−L−乳酸を含む、顆粒状の多孔質体である第1の顆粒状多孔質物質と、乳酸−グリコール酸共重合体を含む、顆粒状の多孔質体である第2の顆粒状多孔質物質とを混合して混合体を調製する混合工程と、
前記混合体を型に入れ加圧・加熱する加圧・加熱工程と、を有し、
前記加圧・加熱工程では、前記乳酸−グリコール酸共重合体の融点以上、前記第1の顆粒状多孔質物質に含まれるポリ−D−乳酸、またはポリ−L−乳酸の融点未満まで加熱する細胞工学用支持体の製造方法。 - ポリ−D−乳酸、またはポリ−L−乳酸を含む骨格構造体と、
前記骨格構造体の表面に形成された乳酸−グリコール酸共重合体を含むコート層と、を有する細胞工学用支持体の製造方法であって、
ポリ−D−乳酸、またはポリ−L−乳酸を含む、顆粒状の多孔質体を型に入れ、500g/cm 2 以上3000g/cm 2 以下、60℃以上200℃以下で、加圧・加熱し、ポリ−D−乳酸、またはポリ−L−乳酸を含む前記骨格構造体を準備する骨格構造体準備工程と、
前記骨格構造体の表面に乳酸−グリコ−ル酸共重合体の粉末を分散した後、熱処理を行う熱処理工程と、を有し、
前記熱処理工程では、前記乳酸−グリコール酸共重合体の融点以上、前記骨格構造体に含まれるポリ−D−乳酸、またはポリ−L−乳酸の融点未満まで加熱する細胞工学用支持体の製造方法。
