Description
−架橋溶液の組成(例えば、バッファ、酵素、酵素の基質、使用されるポリマー及び/又はそれらの置換基)、
−電極の位置、
−加えられる電流又は電圧、及び/又は
−(結合)過程の継続時間、結合過程は例えばパラメーターを変化させるために、同様に中断されうる。これは特に、事前に用意された(本発明による方法、又はその他のゲル化過程のいずれかを用いて生産された)(ハイドロ)ゲルが局所的に機能化される、本発明の1実施形態に及ぶ。本発明の好ましい実施形態では、空間的に構成されたハイドロゲルは前記結合反応によって形成され、前記ハイドロゲルは具体的には結合反応の局所的阻害によって形成され、前記阻害は具体的には前記pHを局所的に変化させることによって制御され、具体的にはその結果前記変化したpHが溶液中の酵素の酵素活性に影響を与え、かつ、具体的には前記酵素が、溶液中の第1及び第2化合物に含まれる第1及び第2部分を、前記第1及び第2部分の間の共有結合を形成することによって、前記ハイドロゲルに変換する。
図6に示されるような溶液のpHの局所的な変化は、溶液の電解を誘起する、(上記のような)電極11、12、13を経由する溶液への電流の通電によって達成される。
本発明の他の実施形態は、上記の、ただしそれぞれpH5及びpH11の、前駆体、活性化第XIII因子及び蛍光標識(Lys−FITC)を含む溶液を調製することによって実現される。いずれのpH条件下でも、活性化第XIII因子の活性が阻害されるので、電流が溶液に流れない場合、前駆体は架橋されないことになる。
本発明による実施形態の他の実施例では、前駆体としてLys−PEG20L及びGln−PEG22Gを用いるハイドロゲルの形成の空間的制御能力は、空間的に定義された生物学的微小環境を作り出すために用いられうる。具体的にはLys−PEG20L、Gln−PEG22G及び具体的にはLys−又はGln−で標識された生体分子24L、25G又は標識を用いて生産されるハイドロゲル200はTG−PEG(トランスグルタミネーション媒介PEG)を指すことになる。具体的には3次元ハイドロゲル200中のマイクロチャネルの形成はインビトロ骨格の開発における課題である。クロバックらは該課題に、重合前にステンレス鋼微小針を配置し、かつ、その後マイクロチャネルを形成するために前記針を除去することによって対処した[20、21]。前駆体及び酵素の溶液への電圧の印加なしにハイドロゲルの形成が生じ、かつ具体的には金属ワイヤーをそのような(構成されていない)ハイドロゲル中にマイクロチャネルを作り出すための鋳型として用いるような従来の方法は、ワイヤー表面へのハイドロゲルの強すぎる接着のために、頻繁にマイクロチャネル及びハイドロゲルの崩壊をもたらす。
マイクロチャネルの生産は、細胞の存在下でさえも、それらの生存に影響を与えることなく実行しうる。図13は0(図13a)及び5μA/mm2(図13b)における、チャネルの生産中のハイドロゲル200に懸濁された間葉系幹細胞(MSCs)の生細胞/死細胞アッセイを示す。ほとんど死んだ細胞(白い矢印)は見えないように、細胞の生存に違いは認められなかった。生細胞/死細胞アッセイにおいて、106mLのヒト骨髄由来のMSCsをTG−PEG前駆体溶液に再懸濁する。溶液は、電極(タングステンワイヤー)除去前に陽極分極し(図13b)、かつ陽極分極せず(図13a)、架橋する。生細胞/死細胞アッセイ(シグマ・アルドリッチ社、スイス国)はメーカーのプロトコルに従って実行する。前記アッセイは死細胞に対しては赤色スペクトル領域で発光する蛍光標識、及び生細胞に対しては緑色スペクトル領域で発光する蛍光標識に基づく。
該実施例では、TG−PEGから作られる微小環境は、例えば細胞のような生物学的存在の生存を支持する幾つかのタンパク質、ペプチド及びその他の生体分子を含む、ハイドロゲル200になりうる。
PDMS反応室100の調製。ポリジメチルシロキサン(PDMS)反応室200は以下のように作成する:シリコンエラストマー及び硬化剤(シルガード184、ダウ・コーニング・コーポレーション、アメリカ合衆国)を(10:1の質量で)ARE−250ミキサー(シンキー・コーポレーション、日本)で、2000rpmで3分間混合する。混合物をその後、電極11,12、13のための反応室100の壁101の挿入穴105、106を作るために、直径500μmのステンレス鋼ワイヤーが配置されたポリ(メチルメタクリレート)(PMMA)の型に流し込む。混合物はその後真空室で30分間脱気し、60℃で4時間固める。ステンレス鋼ワイヤー及びPDMS反応室100をPMMA型から除去し、イソプロパノール(IPA)で洗い、酸素プラズマで洗浄し(300Wで1分、プラズマ−システム100、テクニクス・プラズマ・ゲーエムベーハー、ドイツ国)、そして最後に顕微鏡ガラスカバースリップ103の上に押し付ける。真っ直ぐなタングステンワイヤー11、12、13(W、直径500μm、アドベント・リサーチ・マテリアルズ社、英国)をPDMS反応室100に挿入し、そして2つの電極機構として定電位定電流電解装置(PGU−10V−1A−IMP−S及びECMwinコンピューターインターフェース、エレクトロニクラボ・ピーター・シュレムス、ドイツ国)に接続する。
保留の(pending)活性化第XIII因子(FXIIIa)ペプチド基質、グルタミン受容体基質(n−PEG−Gln)又はMMP感応リンカー(n−PEG−MMP感応−Lys)を含むリジン供与体基質を含む8腕のPEG20、22前駆体は他の場所に記載されるように生産し、かつ特徴付ける[4]。簡潔には、分子量40000の8腕のPEGはネクター(ハンツビル、アラバマ州、アメリカ合衆国)から購入する。ジビニルスルホンはアルドリッチ(ブフス、スイス国)から購入する。PEGビニルスルホン(PEG−VS)は他の場所に記載されるように生産し、かつ特徴付ける[22]。活性化第XIII因子ペプチド基質、H−NQEQVSPL−ERCG−NH2(TG−Gln)、Ac−FKGG−GPQGIWGQ−ERCG−NH2(MMP感応−Lys)及び接着リガンドAc−GCYGRDGSPG−NH2(TG−Gln−RGD)はネオエムピーエス(ストラスブール、フランス国)から入手する(免疫等級、C18精製、HPLC分析:>90%)。NQEQVSPLカセットはα2−プラスミンインヒビターの活性化第XIII因子基質部位に対応し[23]、FKGGカセットは最適化された活性化第XIII因子基質部位に対応し[24]、かつERGGカセットはビニルスルホン反応性システインに対応する[25]。別々の小瓶のTG−Gln、TG−MMP感応−LysをVS基に対して1.2倍モル量過剰に添加し、そして0.3Mのトリエタノールアミン中(pH8.0)で37℃、2時間反応させる。生産物を超純水で3日間、4℃で透析(スネーク・スキン、MWCO・10K、ピアース、ロックフォード、イリノイ州、アメリカ合衆国)する。透析後、無塩の生産物(8−PEG−MMP感応−Lys及び8−PEG−Glnのそれぞれ)を凍結乾燥する。
活性化第XIII因子(200U/ mL、フィブロガミン、CSLベーリング、スイス国)1mLを、100μLのトロンビン(20U/mL、シグマ−アルドリッチ、スイス国)で30分間、37℃で活性化させる。少量の活性化された活性化第XIII因子はさらなる利用のために−80℃で保存しうる。最終乾燥質量含有率が1.5%のハイドロゲルは、50mMの塩化カルシウムを含み、様々なモル濃度及びpH(上記を参照)である、Trisバッファ(Tris)中のn−PEG−Gln及びn−PEG−MMP感応−Lysの化学量論的に均等な([Lys]/[Gln]=1)前駆体溶液によって調製する。
TG−PEG重合への電気化学の影響を調査するために、TG−PEG、Tris(50mM又は10mM、pH5、7.6又は11)、塩化カルシウム及び例えばLys−FITC、TG−Alexa561又は蛍光ポリスチレンビーズ(フルオレスブライト・プレーンYG20・微小ミクロスフィア、ポリサイエンス社)のような蛍光剤を含む60μLの溶液を活性化第XIII因子と混合した。混合物は直ちにPDMS反応室に注ぐ必要がある。TG−PEGの架橋は、定電流状態で流される直流電流の存在下で6分間進行させる。電流密度は0から8μA/mm2の間の範囲で変化しうる。
TG−PEGハイドロゲル−電極界面はLSM510共焦点レーザー走査顕微鏡(カール・ツァイス・アーゲー、ドイツ国)を用いて撮像する。タングステンワイヤーの最大断面(500μm)を得るために、焦点面を調整する可能性がある。FITCは0.7%のレーザー出力での490nmの励起、及び505−550nmの発光バンドパスフィルターによって検出する。Alexa561は515nmの励起、及び575−615nmの発光バンドパスフィルターによって検出しうる。強度プロファイルは電極の平均強度として最小強度を設定し、そして末端200μmの平均強度の値を標準化する(最大強度)ことによって、500×500μmの視野で取得しうる。少なくとも1条件につき3サンプルを分析する必要があり、そして1電極につき2つの画像を得る。
除去時のタングステンワイヤー11、12、13のTG−PEGハイドロゲルへの接着及びその後のマイクロチャネルの形成への電気化学の影響を調査するために、ハイドロゲル200内に分散する20μmのポリスチレン粒子の移動をライカ蛍光顕微鏡(BM550B、ライカ・マイクロシステムズ、ドイツ国)を用いて追跡する。TG−PEGハイドロゲル200を上記のように調製し、そしてタングステンワイヤー11、12、13を手動でハイドロゲル11、12、13から引き抜く。画像は100ミリ秒ごとに記録し、そして粒子は488nmの励起光で検出する。粒子の軌道は前述の画像Jスクリプトを用いて計算する[26]。
チャネル形成後、1%(v/v)のペニシリン/ストレプトマイシン溶液(P/S、ギブコ・ライフ・テクノロジーズ、カタログ番号15140−122)を添加した、血清を含まないDMEM/F−12+GlutaMAXTM(ギブコ・ライフ・テクノロジーズ、カタログ番号31331−028)中の、106/mLのヒト骨髄由来の間葉系幹細胞(MSCs)の溶液をチャネル内に分散する。ハイドロゲルをその後、ヒト血小板由来の成長因子BB(PDFG−BB、10ng/ml、ペプロテック、カタログ番号100−14B)を添加した培地に入れ、そして7日間培養し続けた。明視野画像はツァイス・アクシオウェルト200M倒立顕微鏡で得る。
HEK−IL4レポーター細胞は前述のように生産する[6]。簡潔には、HEK293T細胞をpHW40(PSTAT6−eYFP−pA)及び構成的発現ベクターSTAT6(オープン・バイオシステムズ、ハンツビル、アラバマ州、クローンID5530399から得られる)で形質転換する。構成的mCherry発現プラスミド(pMK47)はインターナルコントロールとして用いる。3次元IL4応答アッセイにおいて、106/mLのレポーター細胞はTG−PEG前駆体溶液で再懸濁し、そして10%(v/v)ウシ胎仔血清(FCS、ギブコ・ライフ・テクノロジーズ、カタログ番号.10500)及び1%(v/v)P/Sを添加したDMEM/F−12+GlutaMAXTMで24時間培養する。蛍光及び明視野画像はライカDMI6000B倒立顕微鏡によって得る。

Claims (15)
- 結合反応は、共有結合の形成であること;及び
前記結合反応を制御するために、第1電極(11)経由で溶液に電解誘導の電流を流すことによって、溶液のpHが前記電極(11)の近傍で局所的に変化すること;及び
空間的に構成されたハイドロゲル(200)が前記結合反応によって形成され、前記ハイドロゲル(200)が前記結合反応の局所的阻害によって形成されること
を特徴とする、第1電極(11)の近傍の溶液中の結合反応の制御方法。 - 前記阻害が前記pHを局所的に変化させることによって制御され、その結果前記変化したpHが溶液中の酵素の酵素活性に影響を与え、かつ、前記酵素が溶液中の第1及び第2化合物(20、22)に含まれる第1及び第2部分(21、23)を、前記第1及び第2部分(21、23)の間に共有結合を形成することによって、前記ハイドロゲル(200)に変換することを特徴とする、請求項1に記載の方法。
- 前記溶液を流れる前記電流を変化させることによって、前記結合反応が空間的に及び/又は時間的に限定されることを特徴とする、請求項1又は2に記載の方法。
- 前記酵素がアミノアシルトランスフェラーゼであることを特徴とする、請求項1〜3のいずれか1項に記載の方法。
- 前記第1及び/又は前記第2化合物がポリマーを含み、又はポリマーから成り、前記ポリマーは天然ポリマーであり、又は合成ポリマーであることを特徴とする、請求項2〜4のいずれか1項に記載の方法。
- 前記第1化合物(20)及び/又は前記第2化合物(22)はポリエチレングリコール(PEG)を含むことを特徴とする、請求項2〜5のいずれか1項に記載の方法。
- 前記共有結合の前記形成が、縮合反応、結合反応、連結反応、架橋反応又は重合反応であることを特徴とする、請求項1〜6のいずれか1項に記載の方法。
- 前記溶液中で電極(11、12)によって前記電圧が誘起され、かつ、前記溶液の前記pHが1〜14の間の範囲にあるように、前記電圧が調整されることを特徴とする、請求項1〜7のいずれか1項に記載の方法。
- 前記溶液に加えられる前記電圧に応じて、前記酵素の前記酵素活性が、局所的に阻害され、低下し又は促進されることを特徴とする、請求項2〜8のいずれか1項に記載の方法。
- 前記溶液が第3部分を含む第3化合物(24、25)を含み、前記酵素によって前記第3部分は前記各第1又は第2部分と変換可能であることを特徴とする、請求項2〜9のいずれか1項に記載の方法。
- 以下のパラメ−ターの少なくとも1つが、前記結合反応中に変化することを特徴とする、請求項1〜10のいずれか1項に記載の方法:
−以下の少なくとも1つの前記溶液の組成:バッファ、酵素、前記酵素の基質、使用されるポリマー及び/又はそれらの置換基、
−電極の位置、
−加えられる電流又は電圧、及び/又は
−結合反応の継続時間。 - 前記結合反応が前記結合反応のパラメーターを変化させるために中断されることを特徴とする、請求項1〜15のいずれか1項に記載の方法。
- 請求項1〜12のいずれか1項に記載の方法を用いる、ハイドロゲルを生産するためのデバイスであって、
−溶液を保持するように設計される反応室(100)、及び
−前記反応室(100)に配置される第1及び第2電極(11、12)、
を備え、
及び、前記第1及び/又は第2電極は、細長い素子として形成され、第1及び第2電極の間に電圧が加えられるとき、電極の近傍でハイドロゲルの形成が阻害されるように設計され、前記ハイドロゲルの形成が阻害されるとき、空洞のチャネルを備えるハイドロゲルが形成され、及び前記ハイドロゲルから電極が除去される場合に、前記電極は前記ハイドロゲルを破壊し、変形し、又は損傷することなく、反応室内の前記ハイドロゲルから除去できる、
前記デバイス。 - 前記第1及び/又は第2電極(11、12)が、前記ハイドロゲルから前記第1及び/又は第2電極(11、12)を除去するために、前記反応室(100)に対して動かされるように設計され、及び/又は前記反応室(100)から解放されるように設計されることを特徴とする、請求項13に記載のデバイス。
- 前記第1及び/又は第2電極(11、12)が、金属、タングステン、半導体、導電性ポリマー材料、又はこれらの材料の組み合わせを含むことを特徴とする、請求項13又は14に記載のデバイス。
