Description
(i)組換えヒトIL−32ガンマ(IL−32γ)と結合することができる;

(a)免疫媒介もしくは自己免疫性の疾患もしくは状態を処置するか、または、免疫媒介もしくは自己免疫性の疾患もしくは状態の進行を妨げる方法;
(b)免疫媒介もしくは自己免疫性の疾患もしくは状態に伴う症状を改善する方法;ならびに/あるいは
(c)免疫媒介もしくは自己免疫性の疾患もしくは状態の存在について、または、免疫媒介もしくは自己免疫性の疾患もしくは状態を発症することについての被験体の危険性を測定するために被験体を診断するか、またはスクリーニングする方法;
ただし、この場合、障害は、患者におけるIL−32の発現、上昇したIL−32活性および/または有害なIL−32活性に伴う。
(i)本発明の抗IL−32抗体またはそのIL−32結合断片の少なくとも1つのCDR;あるいは
(ii)本明細書中上記で定義されるような少なくとも1つの抗イディオタイプ抗体および/またはペプチドもしくはペプチド型化合物
を含む治療有効量のリガンド結合分子を投与することを含む方法に関連する。
−上記定義されたIL−32イソタイプに特異的に結合する本発明の抗体またはその抗原結合断片;および/または
−本発明の抗イディオタイプ抗体、および/または本発明の抗体もしくはその抗原結合断片によって特異的に認識されるエピトープを含む本発明のペプチドもしくはペプチド系化合物
からなる群より選択される少なくとも2つ、3つ、4つ、5つまたはそれ以上の成分から本質的になる治療有効量のカクテルを被験体に投与することを含む方法が提供される。
別途記載される場合を除き、本明細書中で使用されるような用語および実施形態には、国際出願公開WO2013/098419A1および同WO2013/098420A1において提供されるような、また、使用されるような定義が与えられる。補充として、本明細書中で使用されるような一般的な用語には、Oxford Dictionary of Biochemistry and Molecular Biology(Oxford University Press、1997年、2000年改訂および2003年再版、ISBN 0198506732)において提供されるような定義が与えられる。
自己寛容は、免疫系が、その生物の構成成分である抗原に反応しない過程である。自己寛容は、自己反応性T細胞およびB細胞の死滅または不活性化によって達成され、中心的(生成的)な免疫器官(胸腺または骨髄)における中枢性寛容の一部として、または最も一般的に二次免疫組織とみなされるもの(例えば、脾臓、リンパ節、腸)における末梢性寛容として起こり得る。自己寛容は、正常な免疫系の中心的な特徴である。自己寛容を確立および/または維持することができないと自己免疫になり、宿主生物の健康に深刻な影響を及ぼす自己免疫性疾患をもたらし得る。

用語「抗イディオタイプ抗体」は、抗体または他の結合分子に言及する場合、抗原結合部位付近におけるまたは抗原結合部位における抗体の可変領域上に位置する固有の抗原性ペプチド配列に結合して、これによって、所定の自己抗体により別の方法で引き起こされる特異的免疫反応を阻害する分子を含む。同様に、本発明の抗体によって特異的に認識されるエピトープを含む合成ペプチドまたはペプチド系化合物を使用することができる。

本発明に関して、宿主細胞は、細菌、昆虫、真菌、植物、動物またはヒト細胞などの原核細胞または真核細胞であり得る。好ましい真菌細胞は、例えば、サッカロマイセス属のもの、特に、S.セレビシエ種のものである。用語「原核生物の」は、本発明の抗体または対応する免疫グロブリン鎖の発現のためのDNAまたはRNA分子で形質転換またはトランスフェクトされ得るすべての細菌を含むことを意味する。原核生物宿主としては、例えば、大腸菌(E.coli)、ネズミチフス菌、セラチア・マルセッセンスおよび枯草菌などのグラム陰性ならびにグラム陽性細菌が挙げられ得る。用語「真核生物の」は、酵母、高等植物、昆虫および好ましくは哺乳動物細胞、最も好ましくはHEK293、NSO、CSOおよびCHO細胞を含むことを意味する。組換え生産手順で用いられる宿主に応じて、本発明のポリヌクレオチドによってコードされる抗体または免疫グロブリン鎖は、グリコシル化され得るか、または非グリコシル化され得る。本発明の抗体、または対応する免疫グロブリン鎖も、最初のメチオニンアミノ酸残基を含み得る。本発明のポリヌクレオチドは、当業者に一般に公知の技術のいずれかを使用して、宿主を形質転換またはトランスフェクトするのに使用され得る。さらに、機能的に連結された融合遺伝子を調製し、それらを、例えば、哺乳動物細胞および細菌で発現させるための方法は、当技術分野において周知である(Sambrook,Molecular Cloning:A Laboratory Manual,Cold Spring Harbor Laboratory,Cold Spring Harbor,NY,1989)。そこに記載されている遺伝子構築物および方法は、真核生物または原核生物の宿主における本発明の抗体または対応する免疫グロブリン鎖の発現に用いられ得る。一般に、挿入されたポリヌクレオチドの効率的な転写を促進するプロモータ配列を含有する発現ベクターが、宿主と併せて使用される。発現ベクターは、典型的には、複製起点、プロモータ、およびターミネータ、ならびに形質転換細胞の表現型選択を提供することができる特定の遺伝子を含む。DNA配列に適切な細胞源ならびに免疫グロブリンの発現および分泌用の宿主細胞は、American Type Culture Collection(「Catalogue of Cell Lines and Hybridomas」,Fifth edition(1985)Rockville,Maryland,U.S.A.(これは、参照により本明細書に組み込まれる))などの多数の供給源から得ることができる。さらに、本発明の細胞を含むトランスジェニック動物、好ましくは哺乳動物は、本発明の抗体の大規模生産に使用され得る。
様々な抗原のための酵素結合免疫吸着検定法(ELISA)としては、比色分析、化学発光および蛍光測定ベースのものが挙げられる。ELISAは、薬物、ならびに血漿および尿試料中の他の抗原性成分が低量の検出に上手く適用されており、抽出工程を伴わず、実施が簡単である。タンパク質抗原に対する抗体の検出のためのELISAでは、短い合成ペプチドをマイクロタイタープレートのプラスチック表面に直接結合する使用が多い。ペプチドは、一般に、それらの合成的性質および高速液体クロマトグラフィを使用した効率的な精製方法により非常に純粋である。短いペプチドの欠点は、それらが通常は、立体構造または不連続エピトープではなく直鎖状エピトープを表すことである。立体構造エピトープを提示するためには、長いペプチドまたは完全な天然タンパク質のいずれかを使用する。タンパク質抗原をプレートの疎水性ポリスチレン支持体に直接結合すると、結合したタンパク質が部分的または完全に変性し、立体構造エピトープが失われ得る。抗原の固定化(捕捉ELISA)を媒介する抗体でプレートをコーティングすることにより、この影響を回避することができる。
本発明との関連で使用される場合、「結合分子」は主に、抗体およびその断片に関するが、本発明の「目的の分子」に結合する他の非抗体分子を指す場合もある。ここでの「目的の分子」は、サイトカインとして知られる糖タンパク質のクラスに属するタンパク質、特に、異なるIL−32イソタイプのグループから選択されるインターロイキンである。特に好ましい実施形態では、目的の分子はIL−32γである。本発明の目的の分子は、以下の本発明の特定の実施形態の説明内でさらに詳細に定義される。本発明の結合分子としては、限定されないが、ホルモン、受容体、リガンド、主要組織適合複合体(MHC)分子、シャペロン、例えば、熱ショックタンパク質(HSP)および細胞間接着分子、例えば、カドヘリン、インテグリン、C型レクチンおよび免疫グロブリン(Ig)スーパーファミリのメンバーが挙げられる。したがって、単に明確にするために、本発明の範囲を限定するものではないが、以下の実施形態の大部分では、治療剤および診断剤の開発に好ましい結合分子の代表的なものである抗体および抗体様分子に関して議論する。
用語「抗体」および「免疫グロブリン」は、本明細書では互換的に使用される。抗体または免疫グロブリンは、上記および下記に定義される本発明の目的の分子に結合する分子であって、少なくとも重鎖の可変ドメインを含み、通常、少なくとも重鎖および軽鎖の可変ドメインを含む分子である。脊椎動物系における基本的な免疫グロブリンの構造は、比較的よく理解されている;例えば、Harlow and Lane,Antibodies:A Laboratory Manual,(Cold Spring Harbor Laboratory Press,2nd ed.1988)を参照のこと。用語「結合する」および「認識する」は、本発明の結合分子(例えば、抗体)の結合親和性に関して互換的に使用される。
軽鎖は、カッパまたはラムダ(κ、λ)のいずれかに分類される。各重鎖クラスは、カッパまたはラムダ軽鎖のいずれかと結合することができる。一般に、ハイブリドーマ、B細胞または遺伝子操作した宿主細胞のいずれかによって免疫グロブリンが産生される場合、軽鎖および重鎖は互いに共有結合し、2本の重鎖の「テール」部分は共有結合性ジスルフィド結合または非共有結合によって互いに結合する。重鎖では、アミノ酸配列は、Y構造のフォークヘッド末端にあるN末端から各鎖の下部にあるC末端へと延びている。
単鎖抗体を含む抗体断片は、可変領域を単独で、または以下のものの全部もしくは一部と組み合わせて含むことができる:ヒンジ領域、CH1、CH2およびCH3ドメイン。本発明の目的の分子に結合する断片であって、可変領域と、ヒンジ領域、CH1、CH2およびCH3ドメインとの任意の組み合わせをまた含む断片も本発明に含まれる。本発明の方法にしたがって単離されたモノクローナル抗体、特にヒトモノクローナル抗体と同等の本発明の抗体またはその免疫特異的断片は、鳥類および哺乳動物を含む任意の動物起源であり得る。好ましくは、抗体は、ヒト、マウス、ロバ、ウサギ、ヤギ、モルモット、ラクダ、リャマ、ウマまたはニワトリ抗体である。別の実施形態では、可変領域は、起源がコンドリクトイド(例えば、サメ由来)であり得る。
本発明はまた、それぞれIL−32抗体などの本発明の抗体由来のCDRを含む移植抗体(互換的に、等価物と称される)を提供する。このような移植CDRとしては、本発明の抗体のCDRが移植されたか、または1つ以上のアミノ酸置換を含むCDRが移植される動物化抗体が挙げられる。CDRは、上記のように、ヒトフレームワークまたは動物起源由来の抗体フレームワークに直接移植され得る。所望により、フレームワークライブラリを作製することによって、フレームワーク変化も組み込まれ得る。以下により詳細に記載されるように、CDRおよび/またはフレームワーク配列の最適化を独立しておよび連続的に組み合わせて実施することもできるし、または同時に実施することもできる。
上記のように、移植によって作製される抗体の例は、マウス化抗体である。本明細書で使用される場合、用語「マウス化抗体」または「マウス化免疫グロブリン」は、本発明のヒト抗体由来の1つ以上のCDR、およびマウス抗体配列に基づくアミノ酸置換および/または欠失および/または挿入を含有するヒトフレームワーク領域を含む抗体を指す。CDRを提供するヒト免疫グロブリンは「親」または「アクセプタ」と称され、フレームワーク変化をもたらすマウス抗体は「ドナー」と称される。定常領域は存在する必要がないが、存在する場合、それらは、通常、マウス抗体の定常領域と実質的に同一、すなわち、少なくとも約85〜90%、好ましくは約95%またはそれ以上同一である。したがって、いくつかの実施形態では、全長マウス化ヒト重鎖免疫グロブリンまたは軽鎖免疫グロブリンは、マウス定常領域、ヒトCDR、および多数の「マウス化」アミノ酸置換を有する実質的にヒトのフレームワークを含有する。典型的には、「マウス化抗体」は、マウス化可変軽鎖および/またはマウス化可変重鎖を含む抗体である。例えば、キメラ抗体の可変領域全体は非マウスであるため、マウス化抗体は典型的なキメラ抗体を包含しないであろう。「マウス化」の工程によって「マウス化」された改変抗体は、CDRを提供する親抗体と同じ抗原に結合し、マウスでは親抗体と比較して免疫原性が通常低い。
本明細書で使用される場合、用語「重鎖部分」は、免疫グロブリン重鎖に由来するアミノ酸配列を含む。重鎖部分を含むポリペプチドは、CH1ドメイン、ヒンジ(例えば、上部、中部および/または下部ヒンジ領域)ドメイン、CH2ドメイン、CH3ドメインまたはその変異体もしくは断片の少なくとも1つを含む。例えば、本発明に使用される結合ポリペプチドは、CH1ドメインを含むポリペプチド鎖;CH1ドメイン、ヒンジドメインの少なくとも一部およびCH2ドメインを含むポリペプチド鎖;CH1ドメインおよびCH3ドメインを含むポリペプチド鎖;CH1ドメイン、ヒンジドメインの少なくとも一部およびCH3ドメインを含むポリペプチド鎖、またはCH1ドメイン、ヒンジドメインの少なくとも一部、CH2ドメインおよびCH3ドメインを含むポリペプチド鎖を含むことができる。別の実施形態では、本発明のポリペプチドは、CH3ドメインを含むポリペプチド鎖を含む。さらに、本発明に使用される結合ポリペプチドは、CH2ドメインの少なくとも一部(例えば、CH2ドメインの全部または一部)を欠くことができる。上記のように、当業者であれば、これらのドメイン(例えば、重鎖部分)のアミノ酸配列が天然に存在する免疫グロブリン分子と異なるように、これらのドメインを改変することができると理解するであろう。
抗体のためのペプチドまたはポリペプチドのエピトープの最小サイズは約4〜5個のアミノ酸であると考えられる。ペプチドまたはポリペプチドのエピトープは好ましくは、少なくとも7個のアミノ酸を含有し、より好ましくは少なくとも9個のアミノ酸を含有し、最も好ましくは少なくとも約15〜約30個の間のアミノ酸を含有する。CDRにより、抗原性のペプチドまたはポリペプチドがその三次形態で認識され得るので、エピトープを構成するアミノ酸は連続している必要はなく、いくつかの場合には、同じペプチド鎖に存在していないことさえある。本発明において、本発明の抗体によって認識されるペプチドまたはポリペプチドのエピトープは、抗体が2つ以上のイソタイプを認識する場合には、本発明の目的とする分子、すなわち、少なくとも1つのIL−32イソタイプ、またはそれ以外のIL−32イソタイプの相同的配列の少なくとも4個、少なくとも5個、少なくとも6個、少なくとも7個、より好ましくは少なくとも8個、少なくとも9個、少なくとも10個、少なくとも15個、少なくとも20個、少なくとも25個、約15〜約30個の間または約30〜約50個の間の連続したアミノ酸または不連続なアミノ酸からなる配列を含有する。
本明細書で互換的に使用される「結合する」または「認識する」は、一般に、結合分子、例えば、抗体がその抗原結合ドメインを介して所定のエピトープに結合すること、およびその結合が抗原結合ドメインとエピトープとの間のある程度の相補性を必要とすることを意味する。この定義によれば、抗体がランダムな無関係のエピトープに結合するよりも容易に、その抗原結合ドメインを介してエピトープに結合する場合、その抗体はそのエピトープに「特異的に結合する」と言われる。用語「特異性」は、本明細書では、ある特定の抗体がある特定のエピトープに結合する相対的親和性を特定するのに使用される。例えば、抗体「A」が抗体「B」よりも所定のエピトープに対して高い特異性を有するとみなしてもよいし、または抗体「A」が、関連エピトープ「D」に対して有するよりも高い特異性でエピトープ「C」に結合すると言うこともできる。無関係のエピトープは、通常、所定の結合分子の結合特異性の評価に使用され得る非特異的抗原(例えば、BSA、カゼインまたは任意の他の特定のポリペプチド)の一部である。これに関して、用語「特異的結合」は、抗体が、非特異的抗原に対する結合のそのKDよりも少なくとも2倍低いKDで、所定の抗原に結合することを指す。本明細書で使用される場合、用語「高特異的」結合は、特定の標的エピトープに対する抗体の相対KDが、他のリガンドに対するその抗体の結合のKDよりも少なくとも10倍低いことを意味する。
免疫グロブリンまたはそれをコードするcDNAをさらに改変することができる。したがって、さらなる実施形態では、本発明の方法は、キメラ抗体、ヒト化抗体、単鎖抗体、Fab断片、二重特異性抗体、融合抗体、標識抗体またはこれらのいずれか1つの類似体を生産する工程のいずれか1つを含む。対応する方法が当業者に公知であり、例えば、Harlow and Lane「Antibodies,A Laboratory Manual」,CSH Press,Cold Spring Harbor,1988に記載されている。ファージディスプレイ技術によって前記抗体の誘導体を得る場合、本発明に提供される抗体のいずれか1つのものと同じエピトープに結合するファージ抗体の効率性を上昇させるために、BIAcoreシステムにおいて用いられるように、表面プラズモン共鳴を使用することができる(Schier,Human Antibodies Hybridomas 7(1996),97−105;Malmborg,J.Immunol.Methods 183(1995),7−13)。例えば、国際公開第89/09622号には、キメラ抗体の生産が記載されている。欧州特許出願公開第0239400号明細書および国際公開第90/07861号には、ヒト化抗体の生産方法が記載されている。本発明にしたがって用いられる抗体のさらなる供給源は、いわゆるゼノジニック抗体である。例えば、国際公開第91/10741号、国際公開第94/02602号、国際公開第96/34096号および国際公開第96/33735号には、マウスにおけるヒト抗体などのゼノジニック抗体の生産についての一般的原理が記載されている。上記のように、本発明の抗体は、完全抗体の他に、例えば、Fv、FabおよびF(ab)2を含む様々な形態で、ならびに単鎖形態で存在し得る。例えば、国際公開第88/09344号を参照のこと。
本明細書で使用される場合、用語「試料」又は「生物学的試料」は、被験体または患者から得られる任意の生物学的物質を指す。一態様では、試料は、血液、脳脊髄液(「CSF」)または尿を含み得る。他の態様では、試料は、全血、血漿、末梢血から濃縮された単核細胞(PBMC)、例えば、リンパ球(すなわち、T細胞、NK細胞またはB細胞)、単球、マクロファージ、樹状細胞および好塩基球;および培養細胞(例えば、被験体由来のB細胞)を含み得る。試料はまた、腫瘍組織を含む生検または組織試料を含み得る。さらに他の態様では、試料は、全細胞および/または細胞溶解物を含み得る。一実施形態では、試料は、末梢血単核細胞(PBMC)を含む。当技術分野において公知の方法によって試料を採取することができる。
表1に列記されるような、また、IL−32γイソタイプに関して例示的に示されるような本発明の抗IL−32抗体について特異的であるB細胞の特定、および、目的とする特異性を示す抗体の分子クローニング、同様にまた、それらの組換え発現および機能的特徴づけを一般には、国際出願公開WO2013/098419A1および同WO2013/098420A1に記載されるように行うことができる;それらにおける実施例のセクションを参照のこと。具体的には、国際公開WO2013/098419A1の118頁〜120頁における実施例1および実施例2、ならびに、国際公開WO2013/098420A1の27頁〜31頁における実施例1〜実施例4を参照のこと(それらの開示内容は参照によって本明細書中に組み込まれる)。
特に指定がない限り、用語「障害」および「疾患」は、本明細書では互換的に使用される。本明細書で使用される場合、用語「自己免疫性障害」は、個体自身の組織または器官またはその同時分離体または徴候またはそれらから生じる症状から生じるかまたそれらに対する疾患または障害である。自己免疫性疾患は適応免疫反応の脱調節によって主に引き起こされ、自己構造に対する自己抗体または自己反応性T細胞が形成される。ほぼすべての自己免疫性疾患は、炎症性成分を有する。自己炎症性疾患は主に炎症性であり、いくつかの古典的な自己炎症性疾患は、先天性炎症経路の遺伝的欠陥によって引き起こされる。自己炎症性疾患では、自己反応性T細胞または自己抗体は見られない。これらの自己免疫性および自己炎症性障害の多くにおいて、限定されないが、高ガンマグロブリン血症、高レベルの自己抗体、組織中の抗原抗体複合体沈着、副腎皮質ステロイドまたは免疫抑制性処置から恩恵を受けるもの、および罹患組織中のリンパ系細胞凝集塊を含む多くの臨床用および研究用のマーカが存在し得る。B細胞媒介性自己免疫性障害に関する理論に限定されないが、B細胞は、自己抗体産生、免疫複合体形成、樹状およびT細胞活性化、サイトカイン合成、ケモカインの直接放出、および異所性新リンパ形成に対しての病巣の提供を含む、多くの機構的な経路によりヒト自己免疫性疾患において病原性効果を示すことが考えられる。これらの経路のそれぞれが、自己免疫性疾患の病状に異なった度合いで寄与し得る。
標識剤は、本発明の抗体または抗原に直接的または間接的のいずれかでカップリングされ得る。間接的カップリングの一例は、スペーサ部分の使用によるものである。さらに、本発明の抗体は、共有結合または非共有結合によって連結されているさらなるドメインを含み得る。この連結は、当技術分野において公知の上記方法による遺伝的融合に基づくものでもよいし、または例えば国際公開第94/04686号に記載されている化学的架橋によって実施することもできる。本発明の抗体を含む融合タンパク質中に存在するさらなるドメインは、好ましくは、可撓性リンカー、有利にはポリペプチドリンカーによって連結され得、前記ポリペプチドリンカーは、前記さらなるドメインのC末端と本発明の抗体のN末端との間(またはその逆も同様である)の距離に及ぶのに十分な長さの、複数の親水性ペプチド結合アミノ酸を含む。治療的または診断的に活性な薬剤が、様々な方法によって、本発明の抗体またはその抗原結合断片にカップリングされ得る。これとしては、例えば、共有的な方法、例えばペプチド結合によって、治療的または診断的に活性な薬剤にカップリングされた、本発明の抗体の可変領域を含む単鎖融合タンパク質が挙げられる。さらなる例としては、さらなる分子に共有結合または非共有結合によってカップリングされた抗原結合断片を少なくとも含む分子が挙げられ、以下の非限定的な例示的リスト中のものが挙げられる。Traunecker,Int.J.Cancer Surp.SuDP 7(1992),51−52には、CD3に対するFv領域が、可溶性CD4または他のリガンド、例えばOVCAおよびIL−7にカップリングされている、二重特異性試薬ヤヌシンが記載されている。同様に、本発明の抗体の可変領域をFv分子に構築して、引用文献に示されている代替的リガンドにカップリングすることができる。Higgins,J.Infect.Disease 166(1992),198−202には、GP120のV3領域中の特定の配列に対する抗体に架橋されたOKT3からなるヘテロコンジュゲート抗体が記載されている。このようなヘテロコンジュゲート抗体はまた、本発明の方法の抗体中に含まれる少なくとも可変領域を使用して構築され得る。特異的抗体のさらなる例としては、Fanger,Cancer Treat.Res.68(1993),181−194およびFanger,Crit.Rev.Immunol.12(1992),101−124に記載されている抗体が挙げられる。従来の抗体を含む免疫毒素であるコンジュゲートは、当技術分野において広く記載されている。従来のカップリング技術によって毒素を抗体にカップリングしてもよいし、またはタンパク質毒素部分を含む免疫毒素を融合タンパク質として生産してもよい。本発明の抗体は、このような免疫毒素を得るための対応する方法で使用され得る。このような免疫毒素の例示は、Byers,Seminars Cell.Biol.2(1991),59−70およびFanger,Immunol.Today 12(1991),51−54によって記載されているものである。
本明細書で使用される場合、用語「処置する」または「処置」は、自己免疫性疾患および/または自己炎症性疾患の発症などの望ましくない生理的変化または障害を防止するか、または遅延(減少)させることを目的とする治療的処置および予防的または防止的手段の両方を指す。有益なまたは所望の臨床結果としては、限定されないが、検出可能であるかまたは検出不可能であるかにかかわらず、症候の軽減、疾患の程度の縮小、安定化した(すなわち、悪化していない)疾患状態、疾患進行の遅延または緩慢化、疾患状態の寛解または緩和、および(部分的であるかまたは完全であるかにかかわらず)寛解が挙げられる。「処置」はまた、処置を受けない場合に予想される生存期間と比較して生存期間を延長させることを意味し得る。処置を必要とする者としては、症状または障害を既に有する者、ならびに症状または障害を有する傾向がある者、または症状または障害の徴候を防止するべき者が挙げられる。
薬学的に許容し得る担体および投与経路は、当業者に公知の対応文献から採用することができる。当技術分野において周知の方法にしたがって本発明の医薬組成物を製剤化することができる。例えば、Remington:The Science and Practice of Pharmacy(2000)by the University of Sciences in Philadelphia,ISBN 0−683−306472,Vaccine Protocols.2nd Edition by Robinson et al.,Humana Press,Totowa,New Jersey,USA,2003;Banga,Therapeutic Peptides and Proteins:Formulation,Processing,and Delivery Systems.2nd Edition by Taylor and Francis.(2006),ISBN:0−8493−1630−8を参照のこと。適切な医薬担体の例は当技術分野において周知であり、リン酸緩衝生理食塩水溶液、水、油/水エマルジョンなどのエマルジョン、様々な種類の湿潤剤、滅菌溶液などが挙げられる。周知の従来の方法によってこのような担体を含む組成物を製剤化することができる。適切な用量で被験体にこれらの医薬組成物を投与することができる。様々な方法で、適切な組成物の投与をもたらすことができる。例としては、薬学的に許容し得る担体を含有する組成物を、経口、鼻腔内、直腸、局所、腹腔内、静脈内、筋肉内、皮下、皮下、経皮、くも膜下および頭蓋内の方法により投与することが挙げられる。点鼻薬製剤などのエアロゾル製剤は、保存剤および等張剤を有する、活性薬剤の精製した水溶液または他の溶液を含む。このような製剤は、好ましくは、鼻粘膜に適合するpHおよび等張状態に調節される。経口投与用医薬組成物、例えば、単一ドメイン抗体分子(例えば、「nanobodies(登録商標)」なども本発明で想定される。このような経口製剤は、錠剤、カプセル剤、粉末、液体または半固体形態であり得る。錠剤は、ゼラチンまたはアジュバントなどの固体担体を含むことができる。直腸投与用または経膣投与用の製剤は、適切な担体を有する坐剤として提供され得る;O’Hagan et al.,Nature Reviews,Drug Discovery 2(9)(2003),727−735も参照のこと。様々な種類の投与に適切な製剤に関するさらなる指針は、Remington’s Pharmaceutical Sciences,Mace Publishing Company,Philadelphia,PA,17th ed.(1985)および対応する最新版に見ることができる。薬物送達方法の簡単な総説については、Langer,Science 249(1990),1527−1533を参照のこと。
投与計画は、担当の医師および臨床的要因によって決定される。医学分野で周知であるように、任意のある患者への投与量は、その患者のサイズ、体表面、年齢、投与される特定の化合物、性別、投与時間および経路、一般的健康状態、同時に投与されている他の薬物を含む多数の要因に依存する。典型的な用量は、例えば、0.001〜1000μgの範囲内であり得る(すなわち、この範囲内の、発現のためのまたは発現阻害のための核酸);しかしながら、この例示的な範囲よりも少ないかまたは多い用量が、特に前述の要因を考慮して想定される。一般に、医薬組成物の定期投与としての計画は、1μg〜10mg単位/日の範囲内とするべきである。計画が持続注入である場合も、それぞれ1μg〜10mg単位/キログラム体重/分の範囲内とするべきである。定時的評価によって経過をモニタリングすることができる。非経口投与用の製剤は、滅菌水性または非水性溶液、懸濁液およびエマルジョンを含む。非水性溶媒の例は、プロピレングリコール、ポリエチレングリコール、オリーブ油などの植物油、およびエチルオレエートなどの注射可能な有機エステルである。水性担体としては、生理食塩水および緩衝媒体を含む、水、アルコール溶液/水溶液、エマルジョンまたは懸濁液が挙げられる。非経口投与用のビヒクルとしては、塩化ナトリウム溶液、リンゲルブドウ糖液、ブドウ糖および塩化ナトリウム、乳酸化リンゲル液、または固定化油が挙げられる。静脈内投与用のビヒクルとしては、体液および栄養補充液、(リンゲルブドウ糖液に基づくものなどの)電解質補充液などが挙げられる。例えば、抗微生物薬、抗酸化剤、キレート剤、および不活性ガスなどの保存剤および他の添加物も存在し得る。さらに、本発明の医薬組成物は、医薬組成物の目的用途に応じて、抗腫瘍剤および細胞毒性薬物などの薬剤をさらに含むことができる。
患者選択、APECED/APS1患者メモリーからの末梢血単核細胞(PBMC)単離、B細胞培養および抗体単離を、国際出願公開WO2013/098419A1および同WO2013/098420A1に記載されるように、ただし単離および分析された抗体の特異性が、述べられたPCT出願において具体的に使用されたIL−17およびIL−22の代わりに本明細書中上記および下記で定義されるようなIL−32イソタイプに対してのものであったという違いを伴って行った;それらにおける実施例のセクションを参照のこと、具体的には、国際公開WO2013/098419A1の117頁〜120頁における実施例1および実施例2ならびに168頁〜171頁における実施例17、また、国際公開WO2013/098420A1の27頁〜31頁における実施例1〜実施例4を参照のこと(それらの開示内容は参照によって本明細書中に組み込まれる)。
APECED(自己免疫性多腺性内分泌不全症・カンジダ症・表皮異形成、これは自己免疫性多腺性内分泌不全症1型(APS1)とも呼ばれる)の遺伝子状態に罹患する患者の血清における様々なサイトカイン特異的抗体および疾患特異的抗体の全体的な存在が、本出願人の国際出願公開WO2013/098419A1の128頁における実施例7に記載されような、また、128頁〜130頁における表1および表2に示されるようなProtoarray分析によって得られている(それらの開示内容は参照によって本明細書中に組み込まれる)。そのうえ、ELISA(酵素結合免疫吸着アッセイ)を、APECED(自己免疫性多腺性内分泌不全症・カンジダ症・表皮異形成、これは自己免疫性多腺性内分泌不全症1型(APS1)とも呼ばれる)の遺伝子状態に罹患する患者の血清におけるIL−32ガンマ対IL−32アルファ分析の示差的分析のために使用した。合計で30名の患者から得られる血清(これらはAPS1−1からAPS1−30までの符号によって示される)をアッセイにおいて使用した(図2を参照のこと)。
96ウエルのマイクロプレート(Coster、米国)をヒトIL−32γ(R&D)またはIL−32α(ImmunoTools)により被覆した。プレートをPBS−Tにより洗浄し、2%のBSA(Sigma、Buchs、スイス)を含有するPBSにより室温で1時間、ブロッキング処理した。患者血清、B細胞馴化培地または組換え抗体調製物を室温で2時間インキュベーションした。目的とする抗原に対するヒトIgGの結合を、西洋ワサビペルオキシダーゼにコンジュゲート化されたヤギ抗ヒトFcガンマ特異的抗体(Jackson ImmunoResearch,Europe Ltd.、Cambridgeshire、英国)を使用して測定し、その後、HRP活性の測定を、TMB基質溶液(TMB、Sigma、Buchs、スイス)を使用して行った。
IL−32γ(R&D)またはIL−32α(ImmunoTools)に対する本発明の例示的な抗IL−32抗体のEC50結合をELISAによって求めた。MABの連続希釈物(100,000ng/mlから1.69ng/mlまで)を抗原被覆プレート(PBSにおける1μg/mlでの一晩の被覆、続いて、洗浄、および、PBSにおける2%BSAによるブロッキング処理)と2時間インキュベーションした。プレートを続いて洗浄し、MABの結合を、抗ヒトHRPコンジュゲート化Fcガンマ特異的二次抗体(Jackson ImmunoResearch,Europe Ltd.、Cambridgeshire、英国)を用いて検出した。それぞれの抗原に対する最大結合の半分をもたらすMABの濃度(EC50、ng/ml)を、濃度の対数をODの450nmでの測定値に対してプロットすることによって得られるS字形の用量応答曲線(可変スロープ、4パラメータ)において、Prism 4 GraphPadソフトウエアを使用して計算した;結果については、図3および下記の表4を参照のこと。
14B3、19A1、26A6の各MABは、試験された最高濃度(100ug/ml)においてIL−32アルファに結合しなかった。2C2のMABは、非常に高い濃度でのみIL−32アルファに結合した。このことは、EC50結合が30μg/mlよりも大きいことを示している。
中和アッセイが、研究されたサイトカインに応答する細胞株に対して行われる。受容体に対するリガンド結合は一般に、対応するシグナル伝達経路、核への転写因子の移行を活性化し、応答因子遺伝子の転写、翻訳、および、適用可能であるならば、生成物分泌をアップレギュレーションする。使用されるサイトカイン濃度が、アッセイの感度を最大にするために用量応答曲線の直線部分の開始部から選択される。抗体の中和能を試験するために、標的サイトカインの最適な濃度を、血清、上清または精製抗体のサンプルの連続希釈物とプレインキュベーションする。結果が、陽性コントロールと陰性コントロールとの間において中間の値を示す抗体の力価または濃度として表される。インターロイキン−32受容体は未だ報告されていないが、インターロイキン−32はインビトロおよびインビボにおいて他の炎症性サイトカイン(例えば、IL−6など)を単球/マクロファージから誘導し得ることが知られている(Shoda他、Arthritis Res Ther.8(2006);R166)。したがって、分泌されたIL−6はIL−32活性の読み取りとして使用される場合があり、また、使用され、本発明の中和アッセイにおいて市販のELISAキット(Biolegend)により定量化された。
耳炎症アッセイ
耳炎症の表現型を、それぞれの耳への20μlのPBSにおけるヒトサイトカインIL−32γまたはIgGコントロール(またはPBSコントロール)の皮内注入を、30ゲージのニードルを使用して1日おきに、1日目、4日目、6日目および8日目に与えること(20μl/耳、125ng/耳、250ng/マウス/日)によって8週齢のC57BL/6J(WT)マウス(Charles Riverから得られる)において誘導した。本発明の例示的な抗IL−32の2C2抗体による処置を、誘導された耳炎症表現型を軽減させるその中和能に関してこれらの動物に関して試験した。2C2またはコントロールのヒトIgGの1回だけのIP注入[200μg/IP、100μg/IPまたは50μg/IP]を耳炎症の誘導に先立って0日目に動物に施した。マウスを11日目に屠殺した。
IL−32のマウスホモログが未だ1つも特定されていないという事実にもかかわらず、IL−32経路の少なくともいくつかの構成要素が同様にマウスに存在しているという手がかりが、例えば、上記で記載されるような誘導された耳腫大表現型として存在する。さらに、Joosten他(2006)はヒトIL−32をマウスの膝関節に注入し(これにより、関節腫大の誘導が引き起こされた)、そのような実験をRAのためのモデルとして使用している。そのような実験がまた、本発明に関連して行われている。しかしながら、動物は、測定可能な腫大をIL−32注入後に何ら明らかにしていない。これは、介在する筋肉組織を介して膝の厚さを測定することの困難さのためであるかもしれない。この事実のために、本発明は、マウスにおけるIL−32の影響を試験するための、具体的には、本発明の例示的な抗IL−32抗体の影響を試験するための、下記に記載されるような新規なアッセイを確立した。
マッピングの第1の工程として、異なった抗原結合部位に対する様々なa−IL−32MABの示差的結合が、異なる結合部位の数を求めるために調べられる。
本発明の抗体の親和性測定のために、表面プラズモン共鳴SPR測定を、国際出願公開WO2013/098419A1の163頁〜165頁での実施例14(その開示内容は参照によって本明細書中に組み込まれる)に記載されるような類似した実験構成において本発明の目的とする分子を使用して、ProteOn(商標)XPR36装置を製造者(BIO−RAD;Hercules、CA、米国)の説明書に従って使用して行った。この方法によって、本発明の例示的なIL−32抗体(2C2)の親和性が約4nMでのナノモル範囲にあることがSPRにより求められている;図8および図8C中の表を参照のこと。
Claims (22)
- ヒトモノクローナル抗インターロイキン−32(IL−32)抗体またはそのIL−32結合断片であって、
(i)組換えヒトIL−32ガンマ(IL−32γ)と結合することができる;
(ii)ヒトIL−32γにIL32アルファ(IL−32α)よりも優先的に結合することができ、かつ/または、IL−32αとは結合しない:および
(iii)IL−32γの生物学的活性を中和することができるものであり、かつ
その可変領域において、以下の6つの相補性決定領域(CDR)、即ち
(I)VHCDR1:配列番号2の位置31−37、
VHCDR2:配列番号2の位置52−67、
VHCDR3:配列番号2の位置100−107、
VLCDR1:配列番号4の位置23−35、
VLCDR2:配列番号4の位置51−57、および
VLCDR3:配列番号4の位置90−101、または

(b)(II)についての配列番号18及び配列番号20、から選択される、VH領域及びVL領域のアミノ酸配列、
を含む、請求項1に記載の抗体またはIL−32結合断片。 - IgG1またはIgG3である、請求項1又は2に記載の抗体。
- 配列番号6、配列番号8、配列番号22および配列番号24に示される、CHアミノ酸配列およびCLアミノ酸配列から選択されるアミノ酸配列、または、配列番号6、配列番号8、配列番号22または配列番号24のいずれかのアミノ酸配列に対して少なくとも90%の同一性を有するアミノ酸配列を含むCH定常領域および/またはCL定常領域を含む、請求項1〜3のいずれか一項に記載の抗体またはIL−32結合断片。
- 単鎖Fv断片(scFv)、F(ab’)断片、F(ab)断片、F(ab’)2断片および単一ドメイン抗体断片(sdAB)からなる群から選択される、請求項1〜4のいずれか一項に記載の抗体またはそのIL−32結合断片。
- 請求項1〜5のいずれか一項に記載される抗体またはそのIL−32結合断片をコードするポリヌクレオチド。
- 請求項6に記載されるポリヌクレオチドを含むベクター。
- 請求項6に記載されるポリヌクレオチドまたは請求項7に記載されるベクターを含む宿主細胞。
- 前記ポリヌクレオチドが、前記可変領域と、前記定常ドメインの少なくとも一部とをコードするcDNAである、請求項8に記載の宿主細胞。
- 抗IL−32抗体またはその免疫グロブリン鎖を調製するための方法であって、
(a)請求項8または9に記載される細胞を培養すること;および
(b)前記抗体またはその免疫グロブリン鎖を培養物から単離すること、
を含む方法。 - 請求項6に記載されるポリヌクレオチドによってコードされるか、または、請求項10に記載される方法によって得ることができる抗体またはその免疫グロブリン鎖。
- (i)検出可能に標識される;または
(ii)薬物に結合させられる、
請求項1〜5または11のいずれか一項に記載の抗IL−32抗体またはIL−32結合断片。 - 検出可能な標識が、酵素、放射性同位体、蛍光団、ペプチドおよび重金属からなる群から選択される、
請求項12に記載の抗IL−32抗体またはIL−32結合断片。 - 請求項1〜5、11〜13のいずれか一項に記載される抗IL−32抗体またはIL−32結合断片、請求項6に記載されるポリヌクレオチド、請求項7に記載されるベクター、あるいは、請求項8または9に記載される細胞を含む組成物。
- 医薬組成物であり、かつ、薬学的に許容され得る担体をさらに含む、
請求項14に記載の組成物。 - さらに、炎症性疾患を処置するために有用であるさらなる薬剤を含む、
請求項15に記載の組成物。 - 診断用の組成物またはキットであり、請求項1〜5、11〜13のいずれか一項に記載される抗IL−32抗体またはIL−32結合断片を含み、かつ、免疫または核酸に基づく診断方法において従来から使用される試薬をさらに含む、
請求項14に記載の組成物。 - 下記の方法:
(a)免疫媒介もしくは自己免疫性の疾患もしくは状態を処置するか、または、免疫媒介もしくは自己免疫性の疾患もしくは状態の進行を妨げる方法;
(b)免疫媒介もしくは自己免疫性の疾患もしくは状態に伴う症状を改善する方法;ならびに/あるいは
(c)免疫媒介もしくは自己免疫性の疾患もしくは状態の存在について、または、免疫媒介もしくは自己免疫性の疾患もしくは状態を発症することについての被験体の危険性を測定するために被験体を診断するか、またはスクリーニングする方法、
において使用されるためのものであり、
障害が患者におけるIL−32の発現に伴う、
請求項1〜5、11〜13のいずれか一項に記載の抗IL−32抗体またはIL−32結合断片、あるいは、請求項14〜17のいずれか一項に記載の組成物。 - 前記障害が、炎症性腸疾患(IBD;クローン病、潰瘍性大腸炎およびセリアック病を含む)、乾癬、関節リウマチ(RA)、強直性脊椎炎および他の形態の脊椎関節炎、乾癬性関節炎、重症筋無力症、慢性閉塞性肺疾患(COPD)、喘息、結核、白血病を含むガン、血管炎症およびアテローム性動脈硬化からなる群から選択される、
請求項18に記載の方法において使用されるための、請求項1〜5、11〜13のいずれか一項に記載の抗IL−32抗体またはIL−32結合断片、あるいは、請求項14〜17のいずれか一項に記載の組成物。 - 単離された生物学的サンプルにおいてIL−32を検出するか、または測定方法であって、前記サンプルを、請求項1〜5、11〜13のいずれか一項に記載される抗体と混合すること、前記抗体により、混合物に存在するいずれかのIL−32との複合体を形成させること、および、前記混合物に存在する前記複合体を検出することを含む、方法。
- IL−32がIL−32γである、請求項20に記載の方法。
- IL−32の発現および/またはIL−32の活性のレベルが関連する障害を処置することにおいて使用されるための医薬組成物を製造するための方法であって、
(a)抗IL−32抗体またはそのIL−32結合断片を請求項10に記載される方法に従って調製すること;
(b)前記抗体またはそのIL−32結合断片を培養物から精製すること:
(c)前記抗体またはそのIL−32結合断片を薬学的に許容され得る担体と混合して、前記医薬組成物を得ること、
を含む方法。
