Description
例示的な装置の作製
SPME−TM装置を次のように作製した。長さ2.5cm、幅0.4cmのステンレス鋼メッシュ(74×74本ワイヤ/インチ、ワイヤ直径=0.004インチ)を濃塩酸(37v/v%)中で5分間エッチングした、水で洗浄し、メタノール中で超音波処理により洗浄した。エッチングされたメッシュの写真は、図2に示される。被覆する前に表面の酸化または著しい変化を防ぐために、エッチングされたメッシュを乾燥器内で不活性雰囲気に保存した。
例示的な装置の分析工程
SPME−TMを使用する例示的な分析工程は、図8に示される。ステップ(40)で示すように、試料基質(300〜1500μL)を含有するバイアルに、実験例1に従って作製された被覆メッシュ基材を挿入し、試料を高速(3200rpmの渦動撹拌、t≦1分)で撹拌することによって、抽出および濃縮を行った。ステップ(42)で、被膜表面に付着した少なくとも一部の人工物質を除去するために、水を含むバイアル(1500μL、t≦10秒)中で被覆メッシュ基材を洗浄した。被覆メッシュ基材を容易且つ迅速に交換することができるホルダに、被覆メッシュ基材を設置した。次いで、メッシュを移動する自動リニアレールに、ホルダをDARTノズルとMS注入口との間(3者を互いに同軸で0°の角度で)に配置する。ステップ(44)に示すように、準安定気体流をメッシュに流すことによって、被覆粒子の表面に吸着された化合物を同時に脱離およびイオン化した。抽出または予備濃縮した分析物のイオンを大気圧界面(API)に輸送し、タンデム質量分析法によって分析した。
例示的な装置の検出能力
SPMEに精通していない科学者が、短時間で抽出および濃縮を行うことができないと誤って想定している。本開示の文脈において、短い抽出時間とは、60秒以下の抽出時間を意味すると理解すべきである。抽出相と基質との間の大きな表面接触は、高い速度の物質移動を容易にする。薄い被覆は、迅速な平衡時間およびMS装置への効率的な脱離を保証する。さらに、SPMEに精通していない科学者が、より低いLOD/LOQを達成するために、平衡状態で抽出を行うべきであると想定している。
例示的な装置の装置内再現性および装置間再現性
他のアンビエント質量分析装置と比較して、実験例1に開示された装置の特別な特徴は、その再利用性である。9つの独立した装置(n=36)を用いて、コカインおよびジアゼパムを添加した1.5mLのPBS溶液に行われた抽出は、それぞれ4.7%および3.2%未満の装置内再現性/装置間再現性を示した(表1〜5)。勿論、実験で確認したことは、薄い被膜を用いた、分析物の効率的な物質移動(迅速な抽出)だけでなく、効果的な脱離も達成されることである。さらに、第2回の脱離およびイオン化サイクルで得られたシグナル(持ち越し汚染)がDZPの定量(図14A)に使用されたシグナルの約5%であることが判明したにも関わらず、DZPおよびコカインの検出が同時に実施できることは、重要である。したがって、DARTの実験条件は、DZPのために排他的に最適化されていなかった。これによって、一回目の脱離およびイオン化サイクルの後に少量の分析物が依然残っている理由を説明することができる。それにも関わらず、脱離およびイオン化サイクルの直後に、(例えば、50:25:25のメタノール、イソプロパノールおよびアセトニトリルの混合物で)洗浄工程を行うことによって、持ち越し汚染が無視できるようになった(≦0.4%、図14B)。被膜の化学的性質および目標分析物に対する親和性の両方に応じて、洗浄工程を最適化することができる。試料間の分析物濃度に非常に大きなばらつきがあった場合(例えば、低い場合に、pptレベル、高い場合に、ppmレベルまで)、好ましくは、装置の使用を1回のみに制限することができる。さもなければ、洗浄サイクルの後でも、少量の分析物が被膜上に残り、偽陽性をもたらす可能性がある。50ppb以上の濃度および被膜に対して高親和性を有する化合物を抽出する場合、より短い時間(≦30秒)で抽出を行うことができる。より短い抽出時間で抽出を行うことは、濃縮された分析物の量を減少することができ、洗浄工程を含めば、DARTまたは他の溶媒型または熱型イオン化手法によって脱離されていない分析物を完全に除去することができる。
複合基質中の薬物の定量における例示的装置の応用
MS分析は、複雑な試料に関する有意な量の情報を提供する。しかしながら、従来のMS分析を行う前に必要な試料の前処理は、大量の労働および時間が必要とされると共に、複雑である。本明細書中に開示された装置および方法による分析の速さおよび容易さによって、生物試料中の対照物モニタリングおよび治療薬物モニタリング(TDM)は、より少ない労力および/または少ない時間で実施することができる。実験例1に開示された装置を用いて、尿中および血漿中のコカインおよびDZPの定量を行った。図15〜18は、両方の基質から得られた直線性をまとめたものである。PBSと同様に、尿中のコカインおよび血漿中のコカインについて、それぞれ2μg/mLおよび5μg/mLのLOQが測定された。したがって、基質の影響は、開示された方法によって提供された試料の洗浄によって著しく低減され、低結合を有する分析物は、基質に依存せず、同等の検出限界を与える。洗浄工程は、抽出中において、被覆された撚糸に機械的に付着した塩および生体分子を除去するため、信頼できる高感度を与えることによって質量分析計の動作時間を延長すると共に、装置のメンテナンスを最小限に抑えることができる。コカインとは異なり、血漿中DZPのLOQ(497pg/mL)は、尿およびPBS中のLOQ(それぞれ19μg/mLおよび28μg/mL)に比べて、著しく高かった。しかしながら、留意すべきことは、DZPの98%が血漿タンパク質と結合しており、本明細書に記載の装置は、基質中に存在する分析物の遊離部分のみを抽出することである。最後に重要なことは、TM構成が抽出種とイオン化種との間に均一な相互作用をもたらすため、SPME−TMを用いて標準校正を不要として定量をすることもできることである(図19〜図21)。しかしながら、平衡状態(t≦1分)で抽出を行わない場合、再現性のある結果を得るために、変数を精確且つ適切に制御すべきである。そのような変数には、試料の採取時間、対流(撹拌速度および撹拌の均一性)、および被膜厚さの均一性が含まれる。最初の2つのパラメータは、自動抽出システムまたは洗浄システムを使用して制御することができる。自動化で被覆工程のパラメータを制御する場合、メッシュ間の再現性を高めることができる。
複数の対照物の分析
現在、複雑な基質中の対照物の分析を可能にする有力なLC−MS/MSまたはGC−MS/MS法の開発に、多くの研究者が努力している。試料中の成分の複雑さを考慮すると、このような手順は、煩雑で広範な試料の調製工程を必要とする。したがって、迅速、定量的および直接的な分析を可能にする手法が強く求められている。概念実証として、実験例1に記載された装置を用いて、20ng/mLでPBSに添加された21種の禁止物質を同時にモニタリングした。選択的反応モニタリング(SRM)を用いて、各化合物を専一に同定した。LODは、PBS中のコカインおよびジアゼパムについて得られた結果に基づいて、暫定的に予測された(表6)。DARTソースパラメータが各分析物用に最適化されていないにも関わらず、すべての物質が検出され、16個の化合物で50pg/mL未満の仮り検出限界が得られた(例えば、ヘロインのLogP=1.52、プロプラノロールのLogP=3.48、スタノゾロールのLogP=5.53、図22A〜C)。これらの実験例に記載された方法および装置が例示的な抽出相の物理化学的特性から感度および選択性を得られる限り、特定の標的化合物に対してより大きな親和性を有する他の被膜を使用することができる。勿論、感度または速さを失うことなく、本明細書に記載された方法および装置を用いて、単一の分析で多数の物質をスクリーニングする能力は、この技術の特筆すべき特性である。よって、この技術は、廃水中のパーソナルケア製品または食料品中の殺虫剤のモニタリングなどの他の用途に使用することができる。

- 質量分析計で分析されるイオン化分子を生成する装置であって、
約0.2μm〜約100μmの実質的に均一な厚さの抽出相で被覆した被覆メッシュ基材を備え、
前記抽出相は、基質から目標分子を吸着するポリマ、または基質から目標分子を吸着するような寸法に設計された細孔を有するポリマおよび固相マイクロ抽出(SPME)粒子を含み、
前記被覆メッシュ基材は、前記被覆メッシュ基材を通って流体が流れることを可能にするのに十分な開口構造を有し、
