Description
ジャガイモ配列番号7および8を標的とするRNAi構築物
3つの異なるRNAi構築物を作製し、それらは以下を保有する/標的とする:
1.配列番号7の5’末端:コーディング配列−159−200に相当する(開始から−159は、5’utr中にあることを意味する)。
2.配列番号7および8の5’末端のキメラ:コーディング配列4−199+1−204に相当する。
3.配列番号7の中央部分(配列番号8の中央に高度に相同):コーディング配列334−743に相当する。
T1(第一世代トランスジェニック)植物から切断葉を採取し、次いで湿った綿−ウールまたはオアシスにおいて葉柄付きで100%RHでトレーに置いた。フィトフトラ・インフェスタンス(P.inf)培養物(ライ−スクロース−アガープレート)からP.inf遊走子/胞子嚢を採取し、次いで10e3の胞子嚢(10e5/ml)を含有する10μl液滴の胞子懸濁液を中央脈(midvein)の両側に滴下した。トレーを18Cでインキュベートした。葉の感染率を、11日目に、1.完全に感染/過剰生育、2.部分的に感染(10−50%の領域)、および3.クリーン(<10%の領域)としてスコア化した。
トランスポゾン挿入株をコレクション/ライブラリー(Vandenbussche et al., 2008, Plant Journal 54)から同定した。2dTph1トランスポゾン挿入アレルは配列番号9で、かつ3dTph1トランスポゾン挿入アレルは配列番号10で見出された。いくつかの交雑を行い、二重変異を生成した。
植物を標準的な用土で、個別に鉢植えし、23Cで生育させた。
P.ニコチアナエ(P.nicotianae)の胞子を培養物(リマ−マメ−アガーまたはV8−アガープレート)から採取し、次いで10e4(アッセイ Sept)胞子を含有する、2mlの胞子懸濁液を各植物の土壌に滴下した。植物崩壊(plant collapse)を定期的に監視した。
配列番号11および12の両方の過剰発現を提供するため、または配列番号11および12の両方の発現抑制を提供するためのいずれかのために、2つの構築物でトマト植物を形質転換した。
トマト配列番号11を発現抑制する構築物を、配列番号11のCDSの中央の部分と同一の300bpフラグメントのゲートウェイクローニングを用いて生成した。

植物がペチュニアであって、フィトフトラ抵抗性がフィトフトラ・ニコチアナエ(Phytophthora nicotianae)に対する抵抗性であり、かつ少なくとも2つの遺伝子の組み合わせが、配列番号3および配列番号4に記載のタンパク質または配列番号3および配列番号4と少なくとも90%の配列同一性を有するタンパク質をコードする遺伝子である、または
植物がトマトであって、フィトフトラ抵抗性がフィトフトラ・インフェスタンス(Phytophthora infestans)に対する抵抗性であり、かつ少なくとも2つの遺伝子の組み合わせが、配列番号5および配列番号6に記載のタンパク質または配列番号5および配列番号6と少なくとも90%の配列同一性を有するタンパク質をコードする遺伝子である、
植物。 - ナス科に属する植物であって、前記植物はフィトフトラ抵抗性を提供する遺伝的形質を含み、かつ前記抵抗性形質は、2つの遺伝子の組み合わせの発現、もしくは翻訳を下方制御することによって、またはフィトフトラ感受性の植物において前記遺伝子によりコードされるタンパク質の活性を減少させることによって得られ、ここで、前記2つの遺伝子は、配列番号1および2、もしくは配列番号3および4、もしくは配列番号5および6、またはそれらと少なくとも90%の配列同一性を有するタンパク質の組み合わせをコードする、植物。
- ナス科に属する植物がジャガイモ、ペチュニアおよびトマトからなる群から選択される、請求項2に記載の植物。
