Description
用語“ペプチド”は、本明細書中で用いる場合、2〜200個のアミノ酸がペプチド結合により連結したアミノ酸配列を含む分子を表わすが、特定の態様において、非アミノ酸構造体(たとえば、連結する有機化合物)を含むことができる。本発明によるペプチドは、一般的な20種類のアミノ酸のいずれか、またはその修飾形態を含むことができ、あるいはペプチドの化学合成により、または化学修飾もしくは酵素修飾により取り込まれた非天然アミノ酸を含むことができる。用語“ポリペプチド”は、本明細書中で用いる場合、一般に比較的長いペプチドまたはタンパク質を表わす。
a.少なくとも1つのNKT細胞エピトープであって、位置P1および/またはP7に疎水性アミノ酸残基を含むエピトープを同定する;
b.位置P1および/またはP7の少なくとも1つの疎水性アミノ酸残基を除去するか、位置P1および/またはP7の少なくとも1つの疎水性アミノ酸残基を非疎水性残基で置換するか、あるいは位置P1および/またはP7に非疎水性残基を付加することにより、そのエピトープ(単数または複数)を修飾する。
a.少なくとも1つのNKT細胞エピトープであって、位置P1および/またはP7に疎水性アミノ酸残基を含むエピトープを同定する;
b.位置P1および/またはP7の少なくとも1つの疎水性アミノ酸残基を除去するか、位置P1および/またはP7の少なくとも1つの疎水性アミノ酸残基を非疎水性残基で置換するか、あるいは位置P1および/またはP7に非疎水性残基を付加することにより、そのエピトープ(単数または複数)を修飾する。
(1)所望により、VIII因子のアミノ酸配列を、P1およびP7に疎水性残基ならびにP4に脂肪族残基を含む、少なくとも1つのCD1dモチーフの存在について評価する。一般配列、たとえば[FWTHY]−X2X3−[ILMV]−X5X6−[FWTHY]を、下記のような当技術分野で周知のアルゴリズムを用いるために使用できる:

CD1dテトラマーへのペプチドSEQ ID 1の結合
SEQ ID 1を含むペプチドを化学合成により得た。このペプチドを、一夜のインキュベーションによりCD1dテトラマー(ProImmune)に装填するために用いた。
ナイーブマウスの脾臓から磁性ビーズを用いてすべてのCD4(−)細胞を除去することによりNKT細胞を調製した。
CD4+細胞を次いで、SEQ ID 1(図1のP28 WT)のペプチド(アミノ酸配列190−209に対応する)を装填したCD1dテトラマーと共にインキュベートした。
図1は、±1%のCD4+ T細胞が検出されたことを示す。さらに、SEQ ID 1を2部分に分け、配列190−200(SEQ ID 3,図1のP28 A)および200−210(SEQ ID 4,図1のP28 B)を含む合成ペプチドを調製した。
SEQ ID 3のペプチドの配列は、下記のとおりである:

この図は主要CD1d結合モチーフがSEQ ID 3に存在し、限られてはいるが有意%のNKT細胞がSEQ ID 4のペプチドによって検出されたことを示す。
したがって、主要CD1d結合モチーフはSEQ ID 3に存在すると結論された。
SEQ ID 1のペプチドを装填したJAWS2細胞を用いる免疫化によるNKT細胞の活性化
JAWS2細胞はCD1dを発現するが、MHCクラスII決定基を発現しない。SEQ ID 1のペプチドの提示をCD1dにより行なうことができることを示すために、これらの細胞を用いた。
JAWS2細胞を37℃で2時間、SEQ ID 1のペプチド(10μg/ml)と共にインキュベートし、2回、十分に洗浄した。細胞を次いでマイトマイシンで処理して細胞分裂を遮断し、4回洗浄してマイトマイシンを除去した。生理的血清中に2×106個の細胞を含有する細胞懸濁液を、腹腔内経路により3匹のナイーブなVIII因子KOマウスのシリーズそれぞれに注射した。対照として、3匹のナイーブなVIII因子KOマウスに、SEQ ID 1のペプチドと共にインキュベートしていない同数のJAWS2細胞を投与した。
CD4+細胞を次いで、SEQ ID 1のペプチドを装填したテトラマーと共にインキュベートした。
図2は、天然レパトア内に存在するNKT細胞のうち有意割合(8%)がSEQ ID 1のペプチドに対して特異的であること、およびSEQ ID 1のペプチドがJAWS2細胞により提示されるとその割合が倍増すること(16%)を示す。
このNKT細胞検出の特異性は、非装填テトラマーおよびSEQ ID 1のペプチドとは関連のないペプチドを装填したテトラマーの場合にはNKTが検出されないことにより示される。
したがって、SEQ ID 1のペプチドの提示がCD1d提示により行なわれ、この提示はNKT細胞を結合して活性化するのに十分であると結論された。
変異型VIII因子の投与はVIII因子に対する抗体の形成を低下させる
VIII因子中のCD1d結合モチーフを除去することを目的とした変異が抗VIII因子抗体の濃度を低下せることができるかどうかを判定するために、野生型または変異型VIII因子をプラスミドpGC5AM−EN中にクローニングした。オーバーラップ伸長によるスプライシングを用いるPCR(PCR and the splicing by overlap extension)(SOE−PCR)法を用いて、位置F309およびH315における変異(それぞれの場合、アラニンに)をVIII因子の配列に導入した。
天然配列または変異配列のいずれかを収容したプラスミドを配列決定により検査し、マキシプレップ(maxiprep)により調製した。
プラスミド(100μg,2ml中)をナイーブなVIII因子KO C57BL/6マウス(グループ当たり3匹)に動水圧法(hydrodynamic pressure method)(7秒以内で注入を行なった)により直接投与した;これはプラスミドを本質的に肝臓へ指向させることが当技術分野で知られている。対照マウスに同じ方法で、ただし無装填(naked)プラスミドを注入した(n=3)。
合計3回の注入を10日間隔で行なった。
最終注入の10日後、VIII因子に対する抗体の血漿濃度、およびVIII因子の機能を阻害する抗体の濃度を、それぞれELISAおよび発色アッセイ(chromogenic assay)により測定した。
図3aは、VIII因子変異体F309A−H315Aを注入したマウスにおいて3.6倍の抗体減少がみられたことを示す。図3bは、VIII因子の機能を阻害する抗体が3.6倍減少したことを示す。
これらのデータは、VIII因子の機能を阻害するものを含めて特異的抗体の濃度を有意に低下させるために、VIII因子分子中の単一のCD1d結合モチーフの除去で十分であることを示した。
CD1d結合モチーフを含むペプチドと共にインキュベートした際のヒト抗原提示細胞におけるCD1dの発現増大
ヒト抗原提示細胞がCD1d分子に関連してペプチドエピトープをプロセシングして提示する能力をもつかどうかを判定するために、ヒトマクロファージ細胞系U937を用いた。その細胞はクラスII主要組織適合性複合体を発現しないので、CD1dによる提示を評価するために用いられる。
特異的な抗CD1d抗体で検出したCD1dを発現している休止期U937細胞の割合は低い。CD1dに結合できるようにする配列モチーフをもつペプチドと共にU937細胞をインキュベートすれば、これを細胞表面におけるCD1d発現の増大により検出できるはずであると推論した。MHCクラスIIエピトープを含むペプチドについて見られたように、ペプチドの結合によりその分子(クラスII、またはこの場合にはCD1d)のコンホメーションが安定化し、その複合体を細胞表面に繋ぎ留めることができ、それの細胞内リサイクリングが低下する。
U937細胞(7×105個)を37℃で24時間、5μgの、CD1d結合モチーフを含むペプチドまたは対照類と共にインキュベートした。細胞を次いで洗浄し、蛍光標識したCD1d特異的抗体と共にインキュベートし、FACSシステムを用いて陽性細胞の数を評価した。
図4は、CD1dを発現する細胞の割合が、対照実験における6%から、CD1d結合配列を含むペプチドを用いた場合の13%に増加することを示す。ヒストグラムとして示したデータは3回測定の平均であり、星印は対照値より有意に高い結果を示す(p<0.05)。
対照は(左から右へ)、非装填U937、アルファ−gal−セラミドと共にインキュベートした細胞、および一般的なクラスII限定エピトープ(MHCII−MOG)を含む。この実験は、VIII因子配列のアミノ酸188−204に対応するヒト由来(ID SEQ 5)またはマウス由来(ID SEQ 6)VIII因子の配列を含む。
したがって、ヒト抗原提示細胞はそのペプチドと共にインキュベートした際に、ペプチドと細胞の表面にあるCD1dとの複合体を安定化すると結論される。
ヒト末梢血単核細胞(PBMC)はCD1d結合ペプチドエピトープに対して特異的なNKT細胞を含む
ヒトPBMCがCD1d結合ペプチドエピトープと反応する細胞を含有するかどうかを確認するために、T細胞受容体のValpha24−Jalpha18鎖(いわゆるインバリアント鎖)に対して特異的な抗体でコートした磁性ビーズを用いて、インバリアントNKT(iNKT)細胞をバフィーコートから調製した。
次いでこのヒトiNKT細胞(105/条件)を、ヒトVIII因子のアミノ酸残基188−204を含むペプチド(ID SEQ 5)または対照ペプチド(MHCII−MOG)のいずれかを5μg装填したマイトマイシン−C処理U937細胞(比率1:1)と共にインキュベートした。インキュベーションを37℃で2週間、10%のウシ胎仔血清および50U/mlのヒト組換えIL−2を含有するRPMI中で実施した。
培養プレートを細胞クラスターの存在について目視検査した。ID SEQ 5のペプチドを装填したU937を用いるとアルファ−gal−セラミドを装填したものと同程度に細胞クランプが生成したが、対照ペプチド(MHCII−MOG)では生成しなかったことが認められる。図5は、この所見の3つの顕微鏡視野のカウントの平均を示す(最低10個の細胞/クラスター)。
したがって、ヒト末梢血レパトアは、CD1dにより提示されるペプチドと反応するインバリアント鎖NKT細胞として規定される細胞を含有すると結論される。
CD1d結合裂溝の外側に存在するフランキング残基が効果的な細胞表面提示のために重要である
実施例1に示した実験は、ヒトVIII因子アミノ酸190−200に対応するSEQ ID 3のペプチドがCD1dテトラマーに結合し、NKT細胞の検出を可能にすることを示す;これは、この配列がCD1dに対する最小結合モチーフを含んでいたことの指標となる。このモチーフが細胞による効果的な提示に十分であるかどうかを判定するために、実施例4に報告した実験をSEQ ID 3について繰り返した。
図6は、SEQ ID 3のペプチドが、図4に示したSEQ ID 5のペプチドと対照的に、U937細胞の表面におけるCD1dの発現を増大させなかったことを示す。
したがって、CD1d結合モチーフを含むペプチドの効果的なインビボ装填にはCD1d裂溝の外側にあるフランキング領域の存在が必要であると結論される。
CD1d結合VIII因子エピトープに対して特異的なNKT細胞はCD4+およびCD8+の両系列に属する
NKT細胞が属する系列を判定するために、実施例2に記載した実験を行なった。そこで、JAWS2細胞にSEQ ID 3、Bドメイン欠失VIII因子、アルファ−gal−セラミドを装填した後、あるいは装填しないままで、VIII因子KOマウスに腹腔内(IP)注射した(細胞2×106個/マウス)。腹腔内注射の5日後、マウスを屠殺し、脾臓細胞を調製した。次いで細胞懸濁液を、SEQ ID3またはアルファ−gal−セラミドを装填しない、または装填した、CD1dテトラマーと反応させた。
faccs分析のために、CD1dテトラマーを結合する細胞を、DX5−およびCD160−特異的抗体ならびに抗CD4または抗CD8抗体で標識した。表にその結果をまとめる。

Claims (16)
- ドメイン構造A1−a1−A2−a2−B−a3−A3−C1−C2(A1、A2、B、A3、C1およびC2はドメインを表わし、a1、a2およびa3はそれらのドメインを連結する酸性領域である)をもつVIII因子分子であって、[FWTHY]−X2X3−[ILMV]−X5X6−[FWTHY]であるNKT細胞エピトープ(単数または複数)のモチーフの変異により、NKT細胞を活性化する能力が低下した、VIII因子分子:
該変異VIII因子分子において、少なくとも配列番号1で示されるアミノ酸配列内に存在するNKT細胞エピトープが、NKT細胞エピトープモチーフにおける位置P1およびP7のアミノ酸残基を、F、W、T、H、Yとは異なるアミノ酸で置換することにより除かれている。 - 位置P−1および/またはP−2および/またはP8および/またはP9のF、W、T、H、Yのうち少なくとも1つが、非疎水性残基で置換されていることを特徴とする、請求項1に記載のVIII因子分子。
- 位置P−2、P−1、P2、P6、P8および/またはP9に存在する少なくとも1つの疎水性アミノ酸残基が非疎水性残基で置換されている、請求項1に記載のVIII因子分子。
- 位置P−2、P−1、P2、P6、P8および/またはP9に存在する前記少なくとも1つの疎水性アミノ酸残基がF、W、T、HまたはYとは異なることを特徴とする、請求項3に記載のVIII因子分子。
- NKT細胞がCD4+またはCD8+系列に属することを特徴とする、請求項1に記載のVIII因子分子。
- NKT細胞エピトープがVIII因子のA1ドメイン内に存在することを特徴とする、請求項1〜5のいずれか1項に記載のVIII因子分子。
- 少なくとも配列番号1で示されるアミノ酸配列190−209および配列番号2で示されるアミノ酸配列309−316内に存在するNKT細胞エピトープが除かれていることを特徴とする、請求項6に記載のVIII因子分子。
- 分子が、Bドメインの欠失により、および/またはアミノ酸(単数または複数)の付加もしくは置換により、さらに変異していることを特徴とする、請求項1〜7のいずれか1項に記載のVIII因子分子。
- 分子が組換え発現により製造されることを特徴とする、請求項1〜8のいずれか1項に記載のVIII因子分子。
- 請求項1〜9のいずれか1項に記載のVIII因子分子を有効成分として含有する医薬。
- 請求項1に記載の変異VIII因子分子をタンパク質の形で、または遺伝子療法により投与する、請求項10に記載の医薬。
- 先天性または後天性の血友病Aの処置に使用するための、請求項11に記載の医薬。
- 敗血症性ショック、急性フィブリン溶解、多発外傷または脳内出血における出血性障害を伴う患者の処置に使用するための、請求項11に記載の医薬。
- NKT細胞を活性化する能力が低下したVIII因子分子を得るための、下記の工程:
a.少なくとも1つのNKT細胞エピトープであって、位置P1および/またはP7に疎水性アミノ酸残基を含むエピトープを同定する;
b.位置P1および/またはP7の少なくとも1つの疎水性アミノ酸残基を除去するか、位置P1および/またはP7の少なくとも1つの疎水性アミノ酸残基を非疎水性残基で置換するか、あるいは位置P1および/またはP7に非疎水性残基を付加することにより、そのエピトープ(単数または複数)を変異させる;
を含む方法であって、
NKT細胞エピトープが[FWTHY]−X2X3−[ILMV]−X5X6−[FWTHY]であり、該NKT細胞エピトープの少なくとも一つが配列番号1で示されるアミノ酸配列内に位置する、方法。 - VIII因子分子の免疫原性を低下させるための、下記の工程:
a.少なくとも1つのNKT細胞エピトープであって、位置P1および/またはP7に疎水性アミノ酸残基を含むエピトープを同定する;
b.位置P1および/またはP7の少なくとも1つの疎水性アミノ酸残基を除去するか、位置P1および/またはP7の少なくとも1つの疎水性アミノ酸残基を非疎水性残基で置換するか、あるいは位置P1および/またはP7に非疎水性残基を付加することにより、そのエピトープ(単数または複数)を変異させる;
を含む方法であって、
NKT細胞エピトープが[FWTHY]−X2X3−[ILMV]−X5X6−[FWTHY]であり、該NKT細胞エピトープの少なくとも一つが配列番号1で示されるアミノ酸配列内に位置する、方法。 - P1および/またはP7位に隣接する位置P−2、P−1、P2、P6、P8および/またはP9に存在する少なくとも1つの疎水性アミノ酸残基が非疎水性残基で置換されることを特徴とする、請求項15に記載の方法。
