Description
(1)細胞及び培地を収容可能であり、前記培地に接するように細胞測定電極及び参照電極を備え、内面に前記細胞測定電極及び前記参照電極の少なくとも一方が設けられた細胞培養容器と、
前記細胞測定電極及び前記参照電極の少なくとも一方に接続され、前記細胞培養容器内の細胞量に応じた量を測定可能な測定装置と、
を備える細胞培養システム。
(2)前記細胞培養容器内の細胞を観察するための顕微鏡装置をさらに備える上記(1)に記載の細胞培養システム。
(3)前記細胞培養容器が複数段積層されている上記(1)及び(2)に記載の細胞培養システム。
(4)細胞及び培地を収容可能であり、前記培地に接するように細胞測定電極及び参照電極を備え、内面に前記細胞測定電極及び前記参照電極の少なくとも一方が設けられた細胞培養容器を複数段積層させ、それぞれの前記細胞培養容器内に細胞及び培地を収容し、細胞培養を行う工程と、
前記細胞測定電極及び前記参照電極の少なくとも一方に接続された測定装置によって、それぞれの細胞培養容器における細胞量に応じた量を測定する工程と、
複数段積層した細胞培養容器のうち、最下層の細胞培養容器における細胞を顕微鏡装置により観察する工程と、
顕微鏡装置による観察の結果、最下層の細胞培養容器において所定量を超える細胞が確認されたときに、最下層の細胞培養容器における細胞量に応じた量と、最下層の細胞培養容器以外の細胞培養容器における細胞量に応じた量とを比較する工程と、
を有し、
最下層の細胞培養容器以外の細胞培養容器における細胞量に応じた量が、最下層の細胞培養容器における細胞量に応じた量よりも大きい場合に、細胞培養を完了してそれぞれの細胞培養容器から細胞を回収する細胞培養方法。
(5)前記細胞量に応じた量が、細胞呼吸により生ずる水素イオン量である上記(4)に記載の細胞培養方法。
(6)前記細胞量に応じた量が、細胞量に応じて変化するインピーダンスである上記(4)に記載の細胞培養方法。
本発明に係る細胞培養システムの一実施形態について、図1及び図2に基づき説明する。本発明の細胞培養システムは、図1に示すように、細胞及び培地を収容可能であり、内面に電極10が設けられた細胞培養容器1と、電極10に接続され、細胞培養容器1内の細胞量に応じた量を測定可能な測定装置2とを備えている。
(実施例1)細胞電位(水素イオン量測定)
シャーレ状の細胞培養容器の底面に、細胞測定電極としてITO電極を設置し、細胞培養容器内を培地で満たした上で、細胞測定電極上で細胞を培養した。参照電極として白金電極を用い、細胞測定電極の上方に、培地に浸るように設置した。そして、参照電極をGNDとし、細胞測定電極に電圧測定装置(入力インピーダンス無限大)を接続し、60秒間、細胞測定電極における電位を測定した。
シャーレ状の細胞培養容器の底面に、細胞測定電極として35mm×45mmのITO電極(厚さ150nm)、及び参照電極として15mm×45mmのITO電極(厚さ150nm)を設け、細胞培養容器内を培地で満たした上で、細胞測定電極上で細胞を培養した。そして、細胞測定電極及び参照電極の間に、2Vの直流電圧を60秒間印加し、電圧印加時の電流値を測定し、R=V/Iにより直流インピーダンスの値を算出した。培養した細胞が絶縁層となって電極を覆うため、細胞が多いほどインピーダンスの値は高くなる。

- 細胞及び培地を収容可能であり、前記培地に接するように細胞測定電極及び参照電極を備え、内面に前記細胞測定電極及び前記参照電極の少なくとも一方が設けられた細胞培養容器が複数段積層され、
前記細胞測定電極及び前記参照電極の少なくとも一方に接続され、前記細胞培養容器内の細胞量に応じた量を測定可能な測定装置と、
複数段積層された前記細胞培養容器のうち、最下層の細胞培養容器内の細胞を観察するための顕微鏡装置と、
を備える細胞培養システム。 - 前記細胞量に応じた量が、細胞呼吸により生ずる水素イオン量である請求項1に記載の細胞培養システム。
- 前記細胞量に応じた量が、細胞量に応じて変化するインピーダンスである請求項1に記載の細胞培養システム。
- 細胞及び培地を収容可能であり、前記培地に接するように細胞測定電極及び参照電極を備え、内面に前記細胞測定電極及び前記参照電極の少なくとも一方が設けられた細胞培養容器を複数段積層させ、それぞれの前記細胞培養容器内に細胞及び培地を収容し、細胞培養を行う工程と、

顕微鏡装置による観察の結果、最下層の細胞培養容器において所定量を超える細胞が確認されたときに、最下層の細胞培養容器における細胞量に応じた量と、最下層の細胞培養容器以外の細胞培養容器における細胞量に応じた量とを比較する工程と、
を有し、
