Description

アミロースのレベルが50%(w/w)よりも低いコムギ穀粒、またはこれより得られる小麦粉、全麦、デンプン顆粒またはデンプンについて説明した。

Sunstate栽培品種は、Ellison et al.,(1994)Triticum aestivum spp.vulgare(パンコムギ)cv.Sunstate,Australian Journal of Experimental Agriculture,34(6):869−869に記載されており、その全ての内容がここに参考文献として入れられている。

炭水化物の測定と分析
Regina et al.,(2006)の方法を使用して、発生中および成熟コムギ穀粒の双方からデンプンを小規模に単離した。大規模デンプン抽出は、Regina et al.,(2004)の方法に従って実施した。Megazyme(Bray,Co Wicklow,Republic of Ireland)によって提供される全デンプン分析キットを使用して、デンプン含有量を測定し、成熟、未製粉穀粒重量の百分率として、重量を基準にして計算した。次にデンプン含有量を対照植物と比較した。全穀粒重量からデンプン重量を差し引いて穀粒の全非デンプン含有量を得て、総重量の低下がデンプン含有量の低下に起因するかどうかを判定した。
内胚乳中のSBEI、SBEIIa、およびSBEIIbタンパク質の特異的発現、特にこれらのタンパク質の発現レベルまたは蓄積レベルをウエスタンブロット法によって分析した。内胚乳を全ての母体組織から切り離し、5mMのEDTA、20%グリセロール、5mMのDTTおよび1mMのPefablocを含有する、600μlの50mMリン酸カリウム緩衝液(42mMK2HPO4および8mMKH2PO4)、pH7.5中で、およそ0.2mgのサンプルを均質化した。粉砕サンプルを13,000gで10分間遠心分離し、上清をアリコートにして使用時まで−80℃で凍結した。総タンパク質量推定のために、0.25mg/mlのBSA標準の0、20、40、60、80、および100μlのアリコートを使用して、BSA標準曲線を作成した。サンプル(3μl)を蒸留水で100μlにして、1mlのCoomassie Plus Protein試薬を各サンプルに添加した。標準曲線からの0 BSAサンプルを空試験として使用して、5分後に595nmで吸光度を読み取り、サンプル中のタンパク質レベルを判定した。0.34MトリスHCl(pH8.8)、アクリルアミド(8.0%)、過硫酸アンモニウム(0.06%)、およびTEMED(0.1%)を含有する8%非変性ポリアクリルアミドゲル上で、各内胚乳からの20μgの総タンパク質量を含有するサンプルを分離した。電気泳動に続いて、Morell et al.,1997に従って、タンパク質をニトロセルロース膜に移し、SBEIIa、SBEIIbまたはSBEI特異的抗体と免疫反応させた。成熟コムギSBEIIaのN末端のアミノ酸配列AASPGKVLVPDGESDDL(配列番号16)を有する合成ペプチドを使用して、コムギSBEIIaタンパク質(抗wBEIIa)に対する抗血清を作成した(Rahman et al.,2001)。AGGPSGEVMI(配列番号17)を使用して、N末端合成ペプチドと類似様式で、コムギSBEIIb(抗wBEIIb)に対する抗血清を作成した(Regina et al.,(2005)。このペプチドは成熟SBEIIbペプチドのN末端配列に相当し、さらにオオムギSBEIIbタンパク質のN末端と同一であると考えられる(Sun et al.,1998)。N末端合成ペプチドVSAPRDYTMATAEDGV(配列番号18)を使用して、類似様式でコムギSBEIに対するポリクローナル抗体を合成した(Morell et al.,1997)。このような抗血清は、合成ペプチドで免疫化したウサギから標準法に従って得られた。
形質転換コムギ植物または導入遺伝子の存在について試験される植物のPCR分析は、Stacey and Isaac,(1994)によって記載されるミニプレップ法を使用して、1〜2cm2の新鮮な葉材料から抽出されたゲノムDNA上で実施した。ヘアピン型RNAコンストラクトの存在を判定するPCRアッセイでは、SBEIIa遺伝子からの断片(462bp)を増幅するようにデザインされた、プライマーSBEIIa−For:5’−CCCGCTGCTTTCGCTCATTTTG−3’(配列番号19)およびSBEIIa−Rev:5’−GACTACCGGAGCTCCCACCTTC−3’(配列番号20)を使用した。反応条件は、次のとおりであった:「ホットスタート」(94℃、3分間)と、それに続く30サイクルの変性(95℃、30秒間)、アニーリング(55℃、30秒間)、延長(73℃、2分間)と、それに続く1サイクルの73℃(5分間)。反応生成物は、アガロースまたはポリアクリルアミドゲル電気泳動によって分析した。
virプラスミドpAL4404およびpSB1を保有する、武装解除(disarmed)アグロバクテリウム・ツメフアシェンス(Agrobacterium tumefaciens)LBA4404株に、コムギの形質転換のための遺伝子コンストラクトを電気穿孔によって導入し、スペクチノマイシン添加培地上での選択がそれに続いた。形質転換アグロバクテリウム(Agrobacterium)株を固化YEP培地上で27℃で2日間インキュベートした。次に細菌を収集し、コムギ接種のために、400mMのアセトシリンゴンを含有するTSIM1(100mg/lのミオイノシトール、10g/lのグルコース、50mg/lのMES緩衝液を添加したMS培地、pH5.5)中に650nmで2.4の光学濃度で再懸濁した。
4つのヘアピン型RNA(dsRNA)コンストラクトを作成して、コムギのi)SBEIIa、またはii)SBEIIa、SBEIIb、およびSBEI遺伝子の発現を低下させた。各コンストラクト中で、ヘアピン型RNAをコードするDNAを、ヘアピン型RNAの内胚乳特異的発現を提供するコムギDx5遺伝子から得られる高分子量グルテニン(HMWG)プロモーター配列、およびアグロバクテリウム(Agrobacterium)からのノパリンシンターゼ遺伝子からの転写ターミネーター配列(nos3’)と連結させた。このプロモーターは、ヘアピン型RNAをコードする合成遺伝子の内胚乳特異的発現を提供する。
hp5’−SBEIIaと称される第1のコンストラクトの構築および使用については、Regina et al.,(2006)に記載される。hp5’−SBEIIaコンストラクトは、コムギSBEIIa遺伝子からのPCRによって増幅された1536bpのヌクレオチド配列を含有する(GenBank受入番号AF338431)。これは、以下を含んだ。エクソン1および2の全てとエクソン3の一部(ヌクレオチド配列部位1058〜1336、1664〜1761、および2038〜2219(SBEIIaをコードするタルホコムギ(Aegilops tauschii)cDNAのヌクレオチド配列部位1〜578、GenBank受入番号AF338431.1を含む)を含んでなり、各側にEcoRIおよびKpnI制限酵素認識部位を有する、468bpの配列(断片1)、SBEIIa(ヌクレオチド配列部位2220〜2731)のエクソン3および4の一部およびイントロン3の全てからなり、各側にKpnIおよびSacI部位を有する、512bpの配列(断片2)、およびSBEIIa(タルホコムギ(Aegilops tauschii)SBEIIa cDNAヌクレオチド配列部位1〜638、GenBank受入番号AF338431.1を含む、AF338431中のヌクレオチド配列部位1058〜1336、1664〜1761、および2038〜2279)の完全エクソン1、2、および3からなり、各側にBamHIおよびSacI部位を有する、528bpの断片(断片3)。次に断片3の配列が、断片1に対してアンチセンス方向で断片2とライゲートするように、断片1、2、および3をライゲートした。ヘアピン型RNAコンストラクトは、最初に、HMWGプロモーター配列とnos3’ターミネーターを含有するベクターpDVO3000中で生成された。
hpc−SBEIIaと称されるSBEIIaコンストラクトは、SBEIIacDNA(GenBank受入番号AF338432.1)のヌクレオチド1255〜1547に対応する293塩基対のDNA断片を含んでなり、それはSBEIIa遺伝子のエクソン12の一部、エクソン13および14、およびエクソン15の一部に対応する。SBEIIaのこの領域は、SBEIIb cDNAの対応する領域のヌクレオチド配列と約81%のみの同一性を有し、したがってSBEIIbでなく、SBEIIaのサイレンシングの特異性の可能性を増大させることから選択された。
hp3’−SBEIIaと称されるSBEIIaコンストラクトは、SBEIIacDNAのヌクレオチド2305〜2434に対応する130塩基対のDNA断片を含んでなり、SBEIIa遺伝子のエクソン21の一部、エクソン22、および3’非翻訳領域(3’UTR)の一部に対応した。
hp−comboと称されるヘアピン型RNAコンストラクトは、SBEIIa遺伝子の一部に加えて、コムギSBEI遺伝子の領域を含んでなり、i)SBEIIacDNAからのヌクレオチド1756〜2172に対応し、エクソン16の一部、エクソン17〜19、およびエクソン20の一部に対応する417塩基対の配列、およびii)SBEIcDNA(GenBank受入番号AF076679)のヌクレオチド267〜623に対応し、SBEI遺伝子のエクソン3の一部、エクソン4、およびエクソン5の一部に対応する357塩基対の配列を含有した。SBEIIa遺伝子断片は、SBEIIb遺伝子の対応する領域と約86%の同一性を有し、SBEIIbのそれらの対応する領域と100%同一性がある23個の連続したヌクレオチドのいくつかの領域を含み、そのためコムギ中のSBEIIbをコードする遺伝子ならびにSBEIIaおよびSBEIをコードする遺伝子の発現を低下させるという期待を持って、組み合わせコンストラクトがデザインされた。
PCR分析を実施して、Stacey and Isaac(1994)によって記載されるミニプレップ法を使用して、1−2cm2の新鮮葉料から抽出されたゲノムDNAを使用して、再生植物中の導入遺伝子の1つまたは複数を検出した。例えばプライマーSBEIIa−For:5’−CCCGCTGCTTTCGCTCATTTTG−3’(配列番号19)およびSBEIIa−Rev:5’−GACTACCGGAGCTCCCACCTTC−3’(配列番号20)を使用して、hp5’−SBEIIa導入遺伝子で形質転換された植物に対して、PCR反応を実施した。これらのPCR反応は、SBEIIa遺伝子からの約462bpの断片を増幅するようにデザインされた。反応条件は、次のとおりであった:「ホットスタート」(94℃、3分間)と、それに続く30サイクルの変性(95℃、30秒間)、アニーリング(55℃、30秒間)、および延長(73℃、2分間)と、それに続く1サイクルの73℃(5分間)。
T0形質転換コムギ植物から得られる、成熟T1種子からのデンプン顆粒の形態を光学顕微鏡によって観察した。25のT0hp5’−SBEIIa植物のそれぞれからの10個の個々の穀粒を分析した。各内胚乳を穏やかに粉砕してデンプン顆粒を放出し、それを水中に分散して光学顕微鏡下で視覚化した。分析した25系統の内、12系統が歪んだ顆粒がある穀粒を有したが、目視観測からは、異なる種子中の様々なレベルの歪みが明らかになった。デンプン顆粒の形態学的変化について、hpc−SBEIIa、hp3’−SBEIIa、およびhp−combo導入遺伝子で形質転換された各植物からの9個の種子を同様に分析した。この場合、残りの各半切種子をT1植物に栽培することで各系統を保存できるように、半切種子を分析した。63のhp−combo系統の内55系統は、様々なレベルの歪みがある、顆粒形態が変化した種子を有した。10のhp5’−SBEIIa系統は全て、様々なレベルの歪みがある、デンプン顆粒形態が変化した種子を有した。SBEIIa3’系統のいずれにおいても、有意なデンプン顆粒形態の変化は観察されなかった。歪んだデンプン顆粒は、内胚乳中のデンプン中のアミロースレベル上昇の指標であり、典型的に約50%のアミロースであり、または高度に歪んだデンプン顆粒では約70%のアミロースである。これは、表現型の程度の範囲が、有効なサイレンシングコンストラクトのそれぞれについて観察されたことを示した。
抗SBEIIaおよび抗SBEIIb抗体をそれぞれ使用して、ウエスタンブロット法により、SBEIIaおよびSBEIIbタンパク質のレベルについて、T1遺伝子組換え系からの4〜7個のT2発生中内胚乳を分析した。hp−combo系統の場合、抗SBEI抗体を使用して、SBEI発現もまた分析した。選択された遺伝子組換え系からの全SBEIIタンパク質レベル(SBEIIaおよびSBEIIb)は、野生型(変種NB1)中のレベルの百分率として計算され、表11に示される。実施例1に記載されるヨウ素滴定法を使用して、穀粒中の全デンプンの百分率として計算された、遺伝子組換え系からの成熟穀粒中のアミロースレベルもまた判定された(表11)。これは図5に図示される。
タルホコムギ(Aegilops tauschii)ゲノムライブラリからのSBEII遺伝子の単離およびPCRによる、それらの特性評価については、国際公開第99/14314号パンフレットおよび国際公開第200162934−A号パンフレットに記載される。A、B、およびDゲノムのSBEIIa遺伝子のイントロン5領域からのDNA配列は、国際公開第200162934−A号パンフレットに記載される。さらなる研究からは、異なるコムギ遺伝子型からのコムギSBEIIa遺伝子のその他の領域から配列を得て、例えば次のようにして同祖遺伝子をさらに特性評価することが導かれた。プライマーAR2aE12F07(5’−CATTCGTCAAATAATACCCTTGACGG−3’(配列番号21))およびAR2aE14R07(5’−CTTCACCAATGGATACAGCATCAG−3’(配列番号22))を使用して、六倍体コムギ変種CharaからSBEIIaのエクソン12〜14領域を増幅した。これから約656bpのPCR産物が生じ、それは3つの同祖遺伝子のそれぞれからの増幅断片の混合物であると推定された。TOPOベクターへのクローニングに続いて、この産物を配列決定した。エクソン12〜14間の領域をカバーする、3つの多型配列が得られた(図7)。実施例4で詳述されるように、切断後増幅多型(CAP)マーカーを使用した、Chinese Spring染色体操作系のPCR分析に基づいて、配列F1−1はDゲノムに割り当てられ、配列F1−13はBゲノムにゲノム割り当てられ、配列F1−15はAゲノムに割り当てられた。
SBEIIaおよびSBEIIb遺伝子は、どちらもコムギ2番染色体の長腕上に位置し(Regina et al.,2005;Rahman et al.,2001)、これらの報告に基づいて連結すると考えられたが、連鎖がどの程度近いのか正確に分かっていなかった。親栽培品種Baxter、Yitpi、Chara、およびWestoniaが関与する、四元交雑から得られる分離集団を使用して、SBEIIaおよびSBEIIb遺伝子の遺伝子マッピングを実施した。遺伝子間の組換えに関する集団分析は、およそ900の子孫から1個の組換えのみを明らかにした。このデータから、SBEIIaとSBEIIb間の遺伝距離はわずか0.5cMと計算された、これは2個の遺伝子間の非常に密な連鎖であった。
実施例2で得られた配列多形性に基づいて、PCRアッセイをデザインし作成して、パンコムギ中の同祖SBEIIa遺伝子を区別した。コムギSBEIIaのエクソン12〜14領域からの207bp産物を増幅するように、ネステッドプライマー対、AR2aI13ゲノムF2(5’−GTACAATTTTACCTGATGAGATCATGG−3’(配列番号29))およびAR2aI13ゲノムR2(5’−CTTCAGGAATGGATACAGCATCAG−3’(配列番号30))をデザインした。2種の制限酵素Ssp1およびMse1で消化すると、Chinese Spring(CS)からのプライマーを使用して増幅された生成物は、サイズ207bp、147bp、99bp、および108bpの4本の明確なバンドを生じた。CS染色体操作系におけるこのPCRマーカーアッセイの使用からは、207bpの生成物がAゲノムに由来し、147bpの生成物がBゲノムに由来し、99bpおよび108bpの生成物がDゲノムに由来することが明らかになった(図11)。
重イオン衝撃によるコムギの変異誘発コムギ種子の重イオン衝撃(HIB)
一般に使用される市販品種であるコムギ品種Charaにおいて、変異誘発コムギ集団を作成した。日本国埼玉県和光市の理化学研究所で行われた変異誘発では、2種類の重イオン、すなわち炭素およびネオンの供給源が使用された。変異誘発された種子を温室内に播種して、M1植物を得た。これらを自家受粉させて、M2世代を作成した。SBEIIa(ARIIaF2/ARIIaR2)およびSBEIIbのゲノム特異的PCRプライマー(ARA19F/ARA23R)(診断PCR)を使用して、およそ15,000のM2植物のそれぞれから単離されたDNAサンプルをSBEIIaおよびSBEIIb遺伝子のそれぞれの変異について個々にスクリーニングした。野生型DNAサンプルに対する各PCR反応が、A、B、およびDゲノム上のSBEIIaまたはSBEIIb遺伝子の増幅領域に対応する、3個の異なる増幅産物を生じた一方で、変異誘発M2サンプルからのPCR中の1個の断片の不在は、1個のゲノム中の対応領域の不在を示唆し、すなわちその中で遺伝子の少なくとも一部が欠失した変異対立遺伝子の存在を示唆した。このような変異対立遺伝子は、ほぼ確実にヌル対立遺伝子である。
複数ゲノム上に変異SBEII対立遺伝子を有する子孫植物および穀粒を作成する交雑のために、マイクロサテライトマーカー分析による判定で、より小さな欠失について同型接合である変異植物を選択した。交雑種からのF1子孫植物を自家受粉させ、得られたF2種子をそれらのSBEII遺伝子型について分析した。このような12のF2集団のスクリーニングは、11種類の異なる変異対立遺伝子組み合わせ(「二重ヌル」)の同定をもたらした(表14)。その特定の交雑の1200を超えるF2子孫をスクリーニングしたにもかかわらず、12回目の交雑において、B1D1遺伝子型の二重ヌル組み合わせは得られなかった。この1つの可能な説明は、SBEII遺伝子座近傍の重要遺伝子がBおよびDゲノムには存在するがAゲノムにはないことであり、したがってB1およびD1二重ヌル変異の組み合わせが、種子を生育不能にしたかもしれない。27種類の二重ヌル変異株の組み合わせが理論的には可能であり、より多くのF2集団をスクリーニングして、その他の組み合わせが同定されるであろう。
実施例1に記載されるヨウ素滴定法を使用して、実施例5に記載される単一および二重ヌル植物の穀粒デンプンのアミロース百分率を測定した。変異系統数(X軸)に対してアミロース含有量(Y軸)をプロットする散布図を図4に示す。変異株穀粒中のアミロース含有量は、27.3〜38.7%の範囲であった。野生型(未変異)Charaサンプルのアミロース含有量は、27.4%〜29.5%の範囲であった。26系統が、34%を上回るアミロース含有量を記録した。これらの26系統の内、20系統は二重ヌルであり、そのいくつかは、タイプ1またはタイプ2の組み合わせのいずれかの同一交雑からの複製であることが観察された。換言すれば、単一ヌル穀粒のアミロース含有量と比較して、タイプ1およびタイプ2二重ヌル組み合わせにおいて、アミロース含有量が顕著に増大する傾向があった。
4個を超えるSBEIIa変異対立遺伝子がある変異株系統を前の実施例で単離した欠失変異株を用いて作成するために、単一ヌルおよび二重ヌル系統のいくつかを交雑し、診断PCRアッセイを使用して、これらの交雑種のF2子孫を分析した。アッセイは、3個のSBEIIaおよび3個のSBEIIb遺伝子の存在を試験し、したがってA、B、およびDゲノムのそれぞれの中のSBEIIaおよび/またはSBEIIb遺伝子にヌル(欠失)変異を有した植物(SBEIIaおよび/またはSBEIIbに関して三重ヌル系統)を同定する試みにおいて使用された。第1の実験で実施された交雑、および親系統と可能な三重ヌル F2子孫の遺伝子型を表15に列挙する。
08/ddおよび08/bb交雑からのデンプン顆粒形態が変化した種子を播種して、結果として生じた植物を温室栽培した。実施例3に記載されるSBEIIaおよびSBEIIbのゲノム特異的プライマーを使用して、植物から抽出されたDNAを分析した。PCRアッセイからの結果は、これらの各種子が、遺伝子型A1B2、B2D2またはA1D2のホモ接合二重ヌル変異株である一方、第3の(野生型)遺伝子は、同型接合または異型接合状態のいずれかで存在することを示唆した。植物中の3個のSBEIIa遺伝子の存在または不在、および同型接合性または異型接合性をアッセイする、ゲノム特異的個別プライマー対を使用した、定量的PCR(リアルタイムPCR、Rotorgene 6000)を使用して、これらの植物からのDNAをさらに試験した。SBEIIaのために使用されたプライマー対は、Snp6for/Arev5(配列番号51/配列番号61)(Aゲノム、205bpの増幅産物)、Bsnp4/Arev5(配列番号55/配列番号61)(Bゲノム、494bpの増幅産物)、およびDSnp7for/Drev1(配列番号58/配列番号62)(Dゲノム、278bpの増幅産物)であった。SBEIIa増幅反応を正規化するために、全ての植物で等しく発現されると予測される細胞壁生合成遺伝子でありコムギの7番染色体に位置するCslF6遺伝子から、190bpの産物を増幅するプライマー対(SJ156/SJ242)を使用して、増幅を制御した。野生型植物からのDNA、2B2と称される変異誘発集団からのDNA、および野生型栽培変種Chinese Spring(CS)からのDNAを対照テンプレートとして使用した。SBEIIaプライマーとの反応で生成された相対濃度の値は、各テンプレートDNA調製品のためのCslf6プライマーの値で正規化した。可能な三重ヌル植物およびCSの値を2B2系統と比較して計算した。
S28、S22、S14、およびS24系統の種子を温室内に播種して、結果として生じた植物を自家受粉させ、各植物から種子(F3世代)を得た。植物の稔性が影響を被り、ヘッドあたりの種子数および稔性の花穂の百分率は、同時に同一条件下で栽培された、野性型、単一ヌル、およびその他の二重ヌル変異株と比較して顕著に低下したが、消滅しなかったことが観察された(表17)。
SBEIIa特異的抗体を使用して、ウエスタンブロット法により、S28植物からの1本の花穂全体から得られる発生中の内胚乳中のBEIIaタンパク質表現を分析した。花穂中の15個の内胚乳の全ては、SBEIIaのAおよびDゲノムイソ型の双方を欠くパターン(AD二重ヌル)を示し、Bゲノムから発現される1つのSBEIIaバンドのみが存在した。15個の内胚乳の内7個は、他よりも、そして対照系統NB1よりも大幅に低い、BゲノムSBEIIa発現レベルを有した。バンド強度に基づいて、各内胚乳中のSBEIIa発現を定量化した。
4個の変異SBEIIa対立遺伝子を有するA1B2変異遺伝子型がある植物(表18に要約される)からの種子の分析は、その遺伝子型について、アミロース含有量がわずかにのみ上昇し、40%未満のアミロースレベルがもたらされたことを示した。比較すると、S14、S22、S24、およびS28種子からのデータは、第5のSBEIIa変異(欠失)対立遺伝子の追加が、アミロースレベルを約67%に上昇させることを実証した。したがって4個のSBEIIaヌル対立遺伝子から最低限の5個の変異SBEIIa対立遺伝子への増加は、アミロースレベルを50%(w/w)を超えて、実際に60%(w/w)を超えて増大させる上で重要であった。この結論は、実施例2のデータから立てられた予測と一致した。対立遺伝子組成とアミロース含有量との観察された関係性は、植物中のSBEIIa変異対立遺伝子の総数が、アミロース含有量を定める上で重要であったことを示唆した(表18)。SBEIIb変異対立遺伝子の数が役割を果たすこともまた結論付けられたが、SBEIIa変異対立遺伝子の数よりも重要性に劣った。
HIB誘発欠失変異株から完全にSBEIIaを欠く三重ヌル変異株が作成できないという、上の実施例の観察は、交雑の親として使用される特定の変異体植物に依存したかもしれない。これを試験するために、これもまたHIB変異誘発から得られた追加的な親欠失変異株を使用して交雑の第2の組を実施した。38の交雑種からのF2種子を収穫してDNA抽出した。その内の12はA1B2D2遺伝子型(三重ヌル変異体)の生成を目的する交雑種からであるが、実施例7に記載されるのとは異なる親系統を使用している、25の交雑種それぞれからの少なくとも96個のDNAサンプルをPCRによってスクリーニングし、分離傾向を判定した。これらの交雑種のいずれからも、生存能力がある三重ヌルは同定されなかった。二重ヌルの回収もまた交雑に応じて変動したが、ほとんどの場合、予測された遺伝子型が得られた。6個のA1B2D2交雑種からのF2種子もまた顕微鏡的にスクリーニングして、高アミロース表現型有する種子を同定した。このような種子は、中程度の頻度で同定された。
4個の変異SBEIIa対立遺伝子と組み合わされた、2、4または6個の変異SBEIIb対立遺伝子の同型接合を有する変異株を発見する目的でタイプ3変異を伴う交雑を実施し、結果として生じる植物およびその穀粒の表現型を測定した。全てが野性型デンプン顆粒形態を示し、SBEIIbの6個のヌル対立遺伝子とともにSBEIIaのゼロまたは4個のヌル対立遺伝子を有する、A3B3D3およびA3B2D2交雑種からの三重ヌルSBEIIbが同定された。
同定された単一欠失変異株からの三重ヌルSBEIIa変異株を生成するさらなる試みにおいて、改変ストラテジーを採用した。このストラテジーでは、単一変異株の同定後に、単一SBEIIa変異を有するM2植物から未連鎖で無関係の変異を除去するために、単一変異株のいくつかの初期戻し交雑ステップを追加した。これは、変異が組み合わさると有害効果を及ぼし得る、所望のSBEIIa変異と無関係の追加的な変異を植物中に生じ得る、使用された高レベル変異誘発処理に起因する変異背景の影響を低下させるために含めた。これらの初期戻し交雑は、M2変異株と、冬コムギ栽培品種Apacheまたは春コムギ栽培品種Charaコムギのいずれかの植物との交雑によって実施された。
上述の優勢マーカーを使用した1回目のスクリーニングは、あらゆる1個のSBEIIa遺伝子について、異型接合または同型接合野生型の種子を区別できなかった。そのためTaqManベースのPCRアッセイを開発して、第3のゲノム上のSBEIIa遺伝子について、異型接合体および同型接合体を区別して、最初のスクリーニングからの遺伝子型を同定した。TaqMan分析は、半切種子に対して実施され、コムギ内胚乳は各ゲノムについて三倍体(3n)であるので、第3のゲノム上の野生型SBEIIa対立遺伝子に関する異型接合体内胚乳について、以下の2種類のプロファイルのいずれかが可能であった。野生型対立遺伝子が母親によって提供される2n、または野生型対立遺伝子が花粉を通じて父親によって提供される1n。Q−PCR TaqManベースのアッセイは、Applied Biosystems 7900HT Fast Real Time PCR System(ABI,Foster City,CA)を使用して、推定上の二重変異体コムギ種子の第3のゲノム上のSBEIIa遺伝子のコピー数を検出した。アッセイは、磁気ビーズ法(Nucleomag、カタログ番号744 400.24)によって、半切種子から抽出されるゲノムDNAを使用した。DNAを384ウェルプレートに装入し、二本鎖Q−PCR反応を各プレートについて二連で実施した。PCR反応は、プライマーSBE2aQPCRABDF4(順方向プライマー):5’−ACGATGCACTCTTTGGTGGAT−3’(配列番号31)およびSBE2aQPCRABDR4(逆方向プライマー):5’−ACTTACGGTTGTGAAGTAGTCGACAT(配列番号32)を使用して、SBEIIa遺伝子のエクソン21からの65bp断片を増幅するようにデザインされた。Q−PCR反応中に蛍光シグナルを送達するのに使用されたプローブは、SBE2aQPCRABDS4(TaqMan(登録商標)プローブMGB、FA)M5’−CAGCAGGCTTGATCAT−3’(配列番号33)であった。プライマーGamyB1F(順方向プライマー):5’−GATCCGAATAGCTGGCTCAAGTAT−3’(配列番号34)およびGamyB2R(逆方向プライマー):5’−GGAGACTGCAGGTAGGGATCAAC−3’(配列番号35)を使用して、内在性遺伝子からの配列GamyBを内部対照として使用して各サンプルのシグナル値を正規化した。反応条件は、次のとおりであった:「ホットスタート」(95℃、10分間)とそれに続く40サイクルの変性(95℃、15秒間)、アニーリング(58℃、60秒間)。7900HT Fast Real Time PCR Systemに組み込まれたRelative Quantificationマネジャーソフトウェアを使用して、反応生成物を分析した。
迅速非破壊的ハイスループット方法を開発して、高アミロース含有量と関連付けられている表現型について、単一種子をスクリーニングした。実施例4〜6に記載されるPCRベースのスクリーニング法は、15,000個の種子集団中の変異株を検出するのに成功したが、手動で各種子を切断した後に、各半切種子からのDNA調製が必要であるため時間と手間がかかった。近赤外線分光法(NIRS)を使用して、高アミロースと標準的アミロース表現型を区別し得ると判断した。近赤外線分光法(NIRS)は、コムギ種子の特性を判定するのに使用されている非破壊技術である(McClure,2003)。ワキシーデンプン表現型(低アミロース)コムギ単一種子NIRS分析は、Delwiche et al.(2006)によってデュラムコムギ上で開発された。Dowell et al(2009)は、ワキシー、部分的ワキシー、および標準的デュラムおよび六倍体コムギを分離する自動単一種子NIR仕分け装置を開発した。我々の知る限りでは、この方法は、これまで六倍体コムギ中の高アミロース種子を区別するために、使用されていない。
個々の種子中の見かけのアミロース含有量に従ってNIRS測定を較正するために、この場合は単一種子である同一サンプル上で、NIRSスペクトルデータと見かけのアミロース含有量を測定する生化学的方法とを相関させるための、数理モデルを確立してなくてはならなかった。例えば実施例1に記載される方法などの標準ヨウ素滴定法では、一定量の種子が慣例的に使用され、それはデンプン可溶化前に合わせられて、バルク(合わせた)デンプンを提供し、それは常態では、ヨウ素結合に基づいたアミロース含有量の比色分析測定に先だって脱脂される。NIRS較正目的での使用に適切にするために、この方法を修正、簡易化、縮小して、単一種子中の見かけのアミロース含有量を測定できるようにし、それによって種子間のアミロース含有量の変動を考慮に入れた。修正法は、粉砕穀粒からのデンプンを精製せず、デンプン中のアミロースと相互作用する脂質を除去せず、結果は種子からの単離デンプンの百分率でなく新鮮種子重量の百分率として表されるので、「見かけのアミロース含有量」という用語がこの文脈で使用される。これらの理由から、「見かけのアミロース含有量」について得られる値は、実施例1に記載される標準法を使用して得られる値よりもはるかに低い。
この試験では、ArizonaおよびWashingtonで行われた野外実験から得られたWMおよびWMC(対照)系統からの種子を使用した。全部で47個の胚除去半切種子を1.5ml微量遠心管に個々に入れて、それぞれに0.6mlのDMSOを添加する前に正確に秤量した。管を95℃の水浴内で20分間インキュベートし、その後、ガラス棒を使用して、サンプルを管の中で粉砕した。各混合物の体積を17mgのサンプルあたり1mlにDMSOで正確に調節し、その後、時々ボルテックスして、管を水浴内で95℃でさらに70分間インキュベートした。前述同様に、各混合物の10μlのアリコートを取り出して、それらを10μlの0.01N NaOH溶液中の0.3%I2+3%KIで処理し、H2Oで2mlに希釈して、見かけのアミロースを測定した。上述したように、各サンプルの吸光度を605nmで測定し、標準曲線を使用して、吸光度値を「見かけのアミロース」百分率に変換した。
WMおよびWMC種子上の単一種子NIRSスキャンは、Multi Purpose Analyser(MPA)NIRS分光計(Bruker Optics,F−77420Champs Sur Marnes,France)を使用して得られた。各種子をアルミニウムホイルで覆ったガラス管の底に入れ、2回スキャンした。ファイバープローブを装着したBruker MPA Multi−Purpose−Analyser spectrometer(Bruker Optics)を使用して、スペクトルを記録した。16cm−1の分解能で4000〜12,500cm−1の範囲にわたる、32個の参照スキャンおよび16個のサンプルスキャンを使用してスペクトルを記録し、1100個のデータ点がもたらされた。使用された光ファイバープローブは、固体用IN 261プローブであった。
高アミロース穀粒と対照穀粒の識別におけるNIR法を検証するために、さらに60個のWM種子と34個のWMC種子をNIRで2回スキャンして、計算される見かけのアミロース含有量を予測した。このように測定された見かけのアミロース値がWMおよびWMC集団の分布形状を獲得するようにプロットされると、2つの群がわずかな重なりを持ってほとんどは分離されることが分かる(図16)。これらの結果によれば、NIRSによる判定で30%以上の予測された見かけのアミロース表現型を有する種子は、高アミロース種子の良好な候補と見なし得た。
NIRSスクリーニングを実施して、高アミロース含有量を有する変異体種子を検出した。スクリーニングでは、21.142(B2)/Type I−20257(A1)[08/h−111]//Type I−19.832(D1)/CHARAおよび5.706(D2)/21.668(B2)//20.257(A1)/CHARAである、M80およびM85の2つの異なる交雑種からの2,700個のF2種子を使用した。したがってスクリーニングは、それぞれA1B2D1またはA1B2D2遺伝子型がある種子を同定することを目的とした。上述したように、種子あたり2回のNIRSスペクトルを記録した。
4個の種子上でPCR分析を実施して、それぞれのSBEIIa遺伝子型を判定した。最初のアッセイは、3個のゲノム上で各SBEIIa遺伝子の存在または不在を示す、優勢PCRマーカーを使用した。種子の3個は二重ヌル変異株であることが示された一方で、4個目の種子は推定上の三重ヌル変異株であった。しかし共優勢PCRマーカー(下記参照)でさらに試験すると、4個の種子は全て、SBEIIaの二重ヌル変異株(すなわち2個のゲノム中にSBEIIaを欠く)であり、第3のゲノム上の変異SBEIIa遺伝子について異型接合体であることが示された。したがってこれらの種子は、5個の変異SBEIIa対立遺伝子、および少なくとも2個の変異SBEIIb対立遺伝子を含有した。
化学的変異原アジ化ナトリウムまたはEMSを用いた処理によって生成された、変異誘発コムギ穀粒の集団をスクリーニングして、SBEIIa遺伝子に点変異を有する変異株を同定した。このスクリーニングは、野生型コムギと比較してSBEIIa−A、−Bまたは−D活性、またはSBEIIb−A、−Bまたは−D活性が減少したが消失していない変異株(部分的変異株)と並び、対応する遺伝子中の変異によりSBEIIa−A、−Bまたは−D活性、またはSBEIIb−A、−Bまたは−D活性の1つを完全に欠く変異株を検出すると期待された。変株異のスクリーニング法は、SBEIIaまたはSBEIIbに対して特異的な抗体と共にウエスタンブロット法を使用して(実施例2を参照されたい)、または次のような親和性ベースの技術によって、SBEIIaまたはSBEIIbタンパク質量を測定することに基づいた。
5mMのEDTA、5mMのDTT、0.4%プロテアーゼ阻害剤カクテル、および20%グリセロールを含有する、50mMのリン酸緩衝液、pH7.5中で、発生中の種子(開花後約15日)からの単離内胚乳を均質化し、可溶性タンパク質を抽出した。14,000gで10分間の遠心分離後、上清をゲル電気泳動のために使用した。抽出物中のタンパク質濃度をCoomassie Plus Protein Assay Reagentを使用して推定した。
SBEIIaタンパク質イソ型分離のための二次元(2D)親和性電気泳動技術において、Hoefer SE600垂直16cmスラブゲルユニット中に流し込まれた非変性ポリアクリルアミドゲルである第1次元のゲルに、タンパク質抽出物のアリコート(40または100μg)を装入した。第2次元ゲルの分離用成分は、ゲル構造内に固定化された、適量の多糖類標的(アミロペクチン、β−限界デキストリンまたはグリコーゲン)が添加された10%グリセロールを含有する、6%非変性ゲル(14×16cmまたは16×16cm、1.5mm厚さ)であった。濃縮用ゲル(多糖類を含まない)をガラスプレートフォーミングの上部から1cmに注ぎ込んで、コームを使用してウェルを形成した。ゲルを一晩4℃、定常電圧(グリコーゲンおよびβ−限界デキストリンでは100V、アミロペクチン含有ゲルでは135V)で泳動した。
電気泳動に続くSBEIIタンパク質の免疫化学的検出のために、TE 70 PWR半乾燥移動ユニット(Amersham Biosciences)を使用して、タンパク質をゲルからニトロセルロース膜に移した。転写バッファーは、39mMグリシン、48mMトリス、0.0375%SDS、および20%メタノールを含有した。移動は、0.8mA/cm2の定電流で1〜1.5時間実施した。コムギSBEIIaに対して特異的な一次ウサギポリクローナル抗体を使用するウエスタンブロット法に先だって、膜を5%脱脂乳でブロックした。
デンプン顆粒の形態および複屈折の変化
実施例2に記載される遺伝子組換え高アミロースコムギ中で、デンプンおよびデンプン顆粒の特性を調べた。走査型電子顕微鏡を使用して、デンプン顆粒サイズおよび構造の全体的変化を同定した。SBEIIa発現が低下している内胚乳からのデンプン顆粒は、非形質転対照と比較して著しい形態学的変化を示した。それらは形が高度に不規則であり、A顆粒(>10μm径)の多くは鎌形のようであった。対照的に、SBEIIb発現が低下しSBEIIa発現が無変化の内胚乳からのAおよびB(<10μm径)デンプン顆粒は、どちらも表面が滑らかで形状が球状または楕円体であり、野生型コムギデンプン顆粒と識別不能であった。
2つの独立した方法、すなわちヨウ素滴定法およびサイズ排除クロマトグラフィー(SEC)法によって、遺伝子組換えコムギ穀粒のアミロース含有量をアッセイした。アミロース含有量のヨウ素滴定測定は、実施例1記載されるように、既知の濃度精製ジャガイモアミロースおよびアミロペクチンを使用して作成される標準曲線に関連して、ヨウ素がα−1,4グルカンの直鎖領域に結合する際に誘発される色変化の測定に基づいていた。サイズ排除クロマトグラフィー法は、脱分枝されていないアミロースおよびアミロペクチンのカラムクロマトグラフィーによる分離と、それに続くカラムから溶出した画分中のデンプン濃度の測定に基づいていた。穀粒の3つの遺伝子型を分析した。1つ目は、hp−SBEIIaコンストラクトで形質転換されて、SBEIIa発現レベルが非常に低い植物;2つ目は、hp−SBEIIbコンストラクトを含有して、検出可能なSBEIIb発現がないがSBEIIaについて野生型である植物;3つ目は、非形質転換野生型対照(NB1)。SBEIIb発現を欠く植物(008)からの穀粒は、アミロース含有量が、ヨウ素滴定法による測定で27.3%であり、SEC法では32%であった。これは非形質転換対照系統NB1穀粒のアミロース含有量(ヨウ素滴定で31.8%、SECで25.5%)と顕著に異ならない。しかしSBEIIa発現が低下している穀粒(087系統)では、アミロース含有量は顕著に上昇した(ヨウ素滴定で88.5%、SECで74.4%)。087系統に関するこれらの2つの数値の差は、アミロースと良く似てヨウ素に結合し、ヨウ素滴定アッセイ中でアミロースとして測定されるが、カラムクロマトグラフィーではより大きいアミロペクチンと共に分離される、いくつかの「中間体物質」の存在であると考えられた。
イソアミラーゼ脱分枝デンプンの鎖長分布は、フルオロフォア支援炭水化物電気泳動(FACE)によって判定した。この技術は、DP1〜50の範囲内で、鎖長分布の高分解能分析を提供する。非形質転換対照の正規化鎖長分布が遺伝子組換え系の正規化分布から差し引かれるモル差プロット(molar difference plot)から、SBEIIa発現が低下した穀粒からのデンプンにおいて、DP6−12の鎖長の比率の著しい低下と、DP12を上回る対応する鎖長増大があったことが観察された。野生型と比較して、hp−SBEIIb系統からのデンプンの鎖長分布に、統計的に有意な変化は観察されなかった。
デンプンの分子量分布をサイズ排除−HPLC(SE−HPLC)によって測定した。HPLCシステムは、Auto Sampler(GBC、LC1610)および蒸発光散乱検出器(ELSD)(ALLTech、Deerfield,USA)を装着した、GBCポンプ(LC 1150、GBC Instruments,Vic,Australia)から構成された。Ultrahydrogel1000カラム、Ultrahydrogel250カラム、およびガードカラム(7.8mm×300mm、Waters,Japan)を使用して、HPLC操作中35℃を保った。´酢酸アンモニウム緩衝液(0.05M;pH5.2)を流速0.8mLmin−1で移動相として使用した。
穀粒重量の百分率としての穀粒中の全デンプン含有量の解析は、対照の52%およびhp−SBEIIb系統の50.3%と比較して、hp−SBEIIa系統(43.4%)中の内胚乳デンプン含有量のわずかな低下を明らかにした(表24)。これは阻害性コンストラクトによってSBEIIa発現が低下すると、全デンプン合成にいくらかの低下があったことを示唆した。
デンプンの糊化中の水取り込みを測定するKonik−Rose et al.,(2001)の小規模検査に従って、糊化デンプンのデンプン膨潤力を測定した。SSP推定値は、対照(9.31)およびSBEIIb低下穀粒(10.74)からのデンプンと比較して、SBEIIa低下系統からのデンプンで数値3.51と顕著により低かった(表24)。
本質的にRegina et al.,(2004)に記載されるRapid Visco Analyzer(RVA)を使用して、デンプン糊化粘度パラメータを測定した。RVAの温度プロファイルは、以下の段階を含んでなる。60℃で2分間保持、6分間で95℃に加熱、95℃で4分間保持、4分間で50℃に冷却、および50℃で4分間保持。結果(表26)は、対照コムギデンプンと比較して、SBEIIa低下穀粒からのデンプン中で、ピークおよび最終粘度が顕著により低かったことを示した。
Regina et al.,(2004)に記載される示差走査熱量測定(DSC)を使用して、デンプンゼラチン化特性を研究した。Perkin Elmer Pyris 1示差走査熱量計上でDSCを実施した。1:2の比率でデンプンおよび水を予備混合し、およそ50mgをDSCパン内に量り入れて密封し、一晩平衡化させた。1分あたり10℃の加熱速度を使用して、試験および参照サンプルを30〜130℃に加熱した。装置付属のソフトウェアを使用して、データを分析した。結果(表27)は、対照(66.6℃)と比較して、SBEIIa低下穀粒からのデンプンのゼラチン化温度終了遅延(72.6℃)を明らかに示した。対照デンプン(61.16℃)と比較して、SBEIIa低下デンプン(63.51℃)は、ピークゼラチン化温度もまたより高かった。
加圧加工に関するCSIROFood and Nutritional Sciences,Werribeeとの共同研究
小角X線散乱(SAXS)を使用して、天然デンプンと比較した高アミロースコムギデンプンの構造特性評価を実施した。研究は、a)生小麦粉の特性評価、およびb)100℃を超える温度における小麦粉またはデンプンサンプルの加圧料理中の糊化処理のリアルタイム分析、およびc)0〜10日間にわたる冷却時の構造変化と老化を含めるようにデザインされた。研究では、約25%(野生型)〜約75%の範囲にわたる、約10%刻みで増大する様々なアミロース含有量の小麦粉サンプルを使用した。
高アミロースコムギなどの穀粒を食餌中に送達する最も効果的な様式の1つは、パンを経由する。高アミロースコムギが、パンに容易に組み込み得ることを示し、製パン品質の維持を可能にする要素を調べるために、小麦粉サンプルを製造し、分析して、ベーキングで使用した。以下の方法を用いた。
コムギ穀粒を16.5%の水分含量に一晩調湿して、BRI Ltd,AustraliaのBuhler実験室規模製粉機で、またはQuadromat Junior製粉機を使用して製粉し、それに続いて篩掛けして、最終粒度150μmを得た。AACC法39−11(1999)に従った赤外線反射率(NIR)によって、またはDumas法およびAACC法44−15A(AACC51999)に従った空気乾燥器によって、サンプルのタンパク質および水分含有量を測定した。
マイクロZ−armミキサーによって、混合あたり4gの試験小麦粉を使用して小麦粉の最適吸水値を判定した(Gras et al.,(2001);Bekes et al.,(2002)。全ての混合において、それぞれ96および64rpmの迅速および緩慢ブレードの軸速度で、一定の角速度を使用した。混合は、三連で各20分間実施した。水を小麦粉に添加する前に、固形成分の混合のみによって、ベースラインを自動的に記録した(30秒間)。自動送水ポンプを使用して、水を一度に添加した。以下のパラメータは、個々の混合実験から平均を取って判定された。WA%:吸水量は500ブラベンダー単位(BU)生地粘稠度で判定された;生地形成時間(DDT):ピーク抵抗までの期間(秒)。
改変小麦粉に関する最適生地混合パラメータを判定するために、全生地質量を一定に保ちながら、10gのCSIROプロトタイプミキソグラフ中で、マイクロZ−armミキサーで測定される吸水量に相当する変動する吸水量のサンプルを混合した。各小麦粉サンプルで、以下のパラメータを記録した。MT−混合時間(秒);PR−ミキソグラフピーク抵抗(任意の単位、AU);BWPR−ピーク抵抗での帯域幅(任意の単位、AU);RBD−抵抗崩壊(%);BWBD−帯域幅崩壊(%);TMBW−最大帯域幅までの期間(秒);およびMBW−最大帯域幅(任意の単位、A.U.)。
生地伸展性パラメータは、次のように測定した:生地を10gのプロトタイプミキソグラフ中のピーク生地形成まで混合した。修正ジオメトリKieffer生地およびグルテン伸展性装置を用いて、TA.XT2iテクスチャー分析器上で、1cm/sでの伸張試験を実施した(Mann et al.,2003)。伸張試験のための生地サンプル(約1.0g/試験)をKieffer成型機で成型して、伸張試験前に30℃および90%RHで45分間休ませた。Exceed Expertソフトウェアを用いて、データからR_MaxおよびExt_Rmaxを判定した(Smewing,Themeasurement of dough and gluten extensibility usingtheSMS/Kieffer rig and theTA.TX2 texture analyzer handbook,SMS Ltd:Surrey,UK,1995;Mann,(2002)。
対照系統(ベーキング対照、NB1および008)を除いて、全てのその他のコムギ系統は大幅に上昇した吸水値を与えた(図17(a))。系統212および072は、添加レベルの増大と共に、100%の212小麦粉の添加で最高95%の吸水値をはじめとする、吸水値の増大を与えた。これらの系統からの小麦粉の組み込みレベルの増大はまた、最適ミキソグラフ混合時間の短縮をもたらした(図17(b))。吸水データと同様に、非対照系統は、添加レベルの増大と共に、比膨化体積(ローフ体積/ローフ重量)に大幅な低下を示した。影響は212系統で特に強力であった。
標準BRI Research Noodle Manufacturing Method(AFL029)を使用して、Hobartミキサー内で、黄色アルカリ麺(YAN)(100%小麦粉、32%水、1%Na2CO3)を調製した。Otake製麺機のステンレス鋼ローラー内で麺シートを形成した。休止(30分間)後、麺シートを切り分けて細断した。麺の寸法は1.5×1.5mmであった。
BRI Research中種ベーキングは、二段法を伴った。最初の段階では、全小麦粉の一部を水、酵母、およびイーストフードと混合してスポンジを作成した。スポンジを4時間発酵させた。第2の段階では、スポンジに残りの小麦粉、水、およびその他の成分を合わせて、生地を作成した。工程のスポンジ段階は200gの小麦粉で作成し、4時間発酵させた。発酵したスポンジに残りの100gの小麦粉およびその他の成分を混合して、生地を調製した。
パスタ製造に使用した方法は、Sissons et al.,(2007)に記載される。高アミロースコムギ(SBEIIa低下)および対照コムギ(NB1)からの試験サンプル小麦粉をManildraセモリナと様々な百分率で混合し(試験サンプル:0、20、40、60、80、100%)、小規模パスタ調製用の小麦粉ミックスを得た。サンプルを30%水分に調節した。所望量の水をサンプルに添加して短時間混合し、50gファリノグラフボールに移し入れてさらに2分間混合した。コーヒー豆サイズの小片に似た得られた生地をステンレス鋼チャンバーに移し入れて、7000kPaの圧力下で9分間、50℃で休ませた。次にパスタを定速で押出し、およそ48cmの長さに切断した。各サンプルで2バッチのパスタを製造した。Thermoline Temperature and Humidity Cabinet(TEC 2604)(Thermoline Scientific Equipment,Smithfield,Australia)を使用してパスタを乾燥した。乾燥サイクルは、25℃の保持温度とそれに続く65℃への増大を45分間、次に50℃で約13時間とそれに続く25℃への冷却からなった。サイクル中、湿度を制御した。引き続く試験のために乾燥パスタを7cmの長さに切断した。
本明細書に記載されるように製造されたパンをはじめとする食物サンプルの血糖指数(GI)を生体外で次のように測定した:食物サンプルを家庭用フードプロセッサ内で均質化した。およそ50mgの炭水化物に相当するサンプル量を120mlのプラスチックサンプル容器内に量り入れ、α−アミラーゼなしで100μlの炭酸塩緩衝液を添加した。炭酸塩緩衝液添加のおよそ15〜20秒後に、5mlのペプシン溶液(65mlのHCl0.02Mに溶解した65mgのペプシン(Sigma)、pH2.0、使用日に作成)を添加し、混合物を70rpmの往復流水浴内で37℃で30分間インキュベートした。培養に続いてサンプルを5mlのNaOH(0.02M)で中和して、25mlの酢酸緩衝液0.2M、pH6を添加した。次にNa酢酸緩衝液(酢酸ナトリウム緩衝液、0.2M、pH6.0、0.20M塩化カルシウムおよび0.49mM塩化マグネシウム含有)に溶解した、2mg/mLパンクレアチン(α−アミラーゼ、Sigma)とアスペルギルス・ニガー(Aspergillus niger)からの28U/mLアミログルコシダーゼ(AMG、Sigma)を含有する5ml酵素混合物を添加して、混合物を2〜5分間インキュベートした。1mlの溶液を各フラスコから1.5ml管に移し入れ、3000rpmで10分間遠心分離した。上清を新しい管に移し、冷凍庫内で保存した。残りの各サンプルをアルミニウムホイルで覆い、容器を水浴内で37℃で5時間インキュベートした。次にさらに1mlの溶液を各フラスコから収集し、先に実施したように、遠心分離して上清を移した。これもまた、吸光度の読み取りまで、冷凍庫内に保存した。
小規模研究を行って、圧延または圧扁された高アミロースコムギからの加工穀粒の難消化性デンプン(RS)含有量を判定した。技術は、水分レベル25%への1時間の穀粒の馴化を伴い、穀粒の蒸煮がそれに続いた。蒸煮に続いて、小規模ローラーを使用して、穀粒を圧扁した。次にフレークをオーブン内で120℃で35分間ローストした。SBEIIa低下高アミロースコムギ(HAW、系統85.2c)および野生型対照コムギ(栽培変種Hartog)からのおよそ200gのサンプル上で、2種類のローラー幅と3種類の蒸煮時間を使用した。試験されたローラー幅は、0.05mmおよび0.15mmであった。試験された蒸煮時間は60’、45’、および35’であった。
標準EMS処理条件を使用して、コムギ栽培品種ArcheまたはApache種子をEMS変異誘発した後に、変異植物系統集団を開発した。変異誘発種子から、約5000のApacheおよび900のArcheの個々のM1植物を栽培して自家受粉させ、各植物からの種子と引き続く子孫をそれぞれ個々のM1植物に由来する潜在的変異系統として維持した。次世代Solexa配列決定(Illumina)により、系統を3つの同祖SBEIIa遺伝子中の変異についてスクリーニングした。これを行うためには、Arche集団からの約130のM1ファミリーおよびApache集団からの96のM1ファミリーからDNAをそれぞれプールして、7個のDNAプールを調製した。プールされたDNA上で、遺伝子のスプライス部位をはじめとするエクソン領域を標的にして、同祖遺伝子あたり3または4領域についてPCRを実施した。ゲノム特異的プライマーを表30に記載する。
三重ヌルの生産のための交雑に用いるため、ApacheおよびArche変異群において同定した8個の変異株系統と、Sunstate群からの2個の変異株系統を選択した。親の1つとして点変異について同型接合である植物(表32)および2番目の親として二重ヌルSBEIIa変異株(HIB欠失変異株)を用い、全部で66回の交雑を行った。2番目の親植物は、三重ヌルを生産するため、A1D2,A2D2,A1B2およびB2D2遺伝子型の変異株を含んだ。交雑34回分のF1種子(表33)を維持し、単一種子系統によってF2種子50000個を生産するために用いた。これらを、HAW表現型について、単一種子NIRS技術を用いてスクリーニングした。全部でHAW種子2300個を同定した。全ての交雑のうち1つがHAW種子を生産した。選択した種子をグリーンハウスで播き、得られたF3植物415個のうち、分子マーカーで91個をSBEIIa遺伝子について三重ヌルであると同定した。植物85個から種子が得られ、うち67個の三重ヌル植物を、NIRS測定により高アミロースコムギ植物であると同定した。24個の植物が、GM85.2c参照穀粒よりもアミロースレベルが高い種子を生産した。アミロース含有量をヨウ素定量法により測定したところ、SBEIIa三重ヌル穀粒のいくつかがコントロール穀粒よりもアミロースレベルが高く、158%まであった。この同じ実験では、HAW GM参照穀粒は、コントロール系統よりもアミロースレベルが58%高かった(表35)。千粒重測定により、高アミロースコムギ穀粒は穀粒重量が有意に減少していないことが分かった。

Claims (17)
- 胚およびデンプンを含むコムギ穀粒において、当該胚がSBEIIa−A遺伝子の2つの同一ヌル対立遺伝子、SBEIIa−B遺伝子の2つの同一ヌル対立遺伝子およびSBEIIa−D遺伝子の2つの同一ヌル対立遺伝子を含み、前記穀粒は、ウエスタンブロット分析で測定した検出可能なSBEIIaタンパク質が欠如しており、当該SBEIIa−A、SBEIIa−BおよびSBEIIa−Dのヌル対立遺伝子の少なくとも1つが、未成熟翻訳終止変異、スプライス部位変異、または挿入である点変異を含み、前記デンプンがアミロースを含み、穀粒の抽出可能デンプンの比率として穀粒が少なくとも50%(w/w)のアミロース含有量を含み、前記穀粒が、野生型穀粒の発芽率の70%から90%、または90%から100%の発芽率を有することを特徴とするコムギ穀粒。
- 請求項1に記載の穀粒が、前記SBEIIa−BおよびSBEIIa−D遺伝子の少なくとも一部をそれぞれ欠失したBおよびDゲノム中の欠失変異であるヌル対立遺伝子を有し、前記SBEIIa−A遺伝子が点変異を含む;または前記SBEIIa−AおよびSBEIIa−D遺伝子の少なくとも一部をそれぞれ欠失したAおよびDゲノム中の欠失変異であるヌル対立遺伝子を有し、前記SBEIIa−B遺伝子が点変異を含む;または前記SBEIIa−AおよびSBEIIa−B遺伝子の少なくとも一部をそれぞれ欠失したAおよびBゲノム中の欠失変異であるヌル対立遺伝子を有し、前記SBEIIa−D遺伝子が点変異を含むことを特徴とする穀粒。
- 請求項1又は2に記載の穀粒において、前記胚が、発生中の内胚乳中で生産された場合に1つのSBEIIb遺伝子のヌル対立遺伝子およびデンプン分岐酵素活性を有するSBEIIbタンパク質を2個のみ、または、ウエスタンブロット分析で検出できるSBEIIbタンパク質を2個のみ、更に含むことを特徴とする穀粒。
- 請求項1又は2に記載の穀粒において、前記胚が、発生中の内胚乳中で生産された場合に2つのSBEIIb遺伝子のヌル対立遺伝子およびデンプン分岐酵素活性を有するSBEIIbタンパク質を1個のみ、または、ウエスタンブロット分析で検出できるSBEIIbタンパク質を1個のみ、更に含むことを特徴とする穀粒。
- 請求項1乃至4の何れか1項に記載の穀粒において、SBEIIa−B遺伝子の少なくとも一部およびSBEIIb−B遺伝子の少なくとも一部を欠く、Bゲノム中に欠失変異であるヌル変異を含む;または、SBEIIa−D遺伝子の少なくとも一部およびSBEIIb−D遺伝子の少なくとも一部を欠く、Dゲノム中に欠失変異であるヌル変異を含む;または、SBEIIa−A遺伝子の少なくとも一部およびSBEIIb−A遺伝子の少なくとも一部を欠く、Aゲノム中に欠失変異であるヌル変異を含む;ことを特徴とする穀粒。
- 請求項5に記載の穀粒において、SBEIIa−B遺伝子および/またはSBEIIb−B遺伝子の全体を欠くヌル変異を含む;または、SBEIIa−D遺伝子および/またはSBEIIb−D遺伝子の全体を欠くヌル変異を含む;または、SBEIIa−A遺伝子および/またはSBEIIb−A遺伝子の全体を欠くヌル変異を含む;ことを特徴とする穀粒。
- 請求項1乃至6の何れか1項に記載の穀粒が、当該穀粒の抽出可能なデンプンの比率としてアミロース含有量が少なくとも60%(w/w)または少なくとも67%(w/w)であることを特徴とする穀粒。
- 請求項1乃至7の何れか1項に記載の穀粒が、非遺伝子組み換え穀粒である、またはSBEIIa遺伝子の発現を低下させるRNAをコードするいかなる外来性核酸も有さないことを特徴とする穀粒。
- 請求項1乃至8の何れか1項に記載の穀粒において、当該穀粒のデンプンが、
(i)少なくとも2%の難消化性デンプンを含む;
(ii)低い相対血糖指数(GI)を含む;
(iii)低い相対アミロペクチンレベルを含む;
(iv)デンプン顆粒の歪み;
(v)顆粒複屈折の低下;
(vi)膨潤体積の低下;
(vii)鎖長分布および/または分岐発生頻度の変化;
(viii)糊化温度およびより高いピーク温度の終了遅延;
(ix)粘度低下(ピーク粘度、糊化開始温度など);
(x)アミロペクチンの分子量増大;および
(xi)野生型コムギデンプン顆粒またはデンプンに対する、%結晶化度、%A型またはB型デンプンの変化
からなる群から選択される、1つまたは2つ以上によって特徴付けられることを特徴とする穀粒。 - 請求項1乃至9の何れか1項に記載の穀粒を産生するコムギ植物が繁殖可能な雌雄であることを特徴とするコムギ植物。
- 請求項1乃至9の何れか1項に記載の穀粒から生産することを特徴とする全麦または小麦粉。
- 請求項1乃至9の何れか1項に記載の穀粒、または請求項11に記載の全麦または小麦粉を含むことを特徴とする食品原材料。
- 食品または飲料製品が、乾燥重量で少なくとも10%レベルの食品または飲料原材料を含み、当該原材料が請求項1乃至9の何れか1項に記載のコムギ穀粒、または請求項11に記載の全麦または小麦粉であることを特徴とする食品または飲料製品。
- 食料または飲料を生産する方法において、(i)請求項1乃至9の何れか1項に記載の穀粒を得るステップと;(ii)当該穀粒を加工して食料または飲料原材料を生産するステップと;(iii)ステップiiからの食品または飲料原材料を別の食料または飲料原材料に加えて当該食料または飲料を生産するステップと;を具えることを特徴とする方法。
- 穀粒を生産する方法において、(i)請求項10に記載のコムギ植物を得るステップと;(ii)当該植物からコムギ穀粒を収穫するステップと;を具えることを特徴とする方法。
- さらに前記穀粒を加工するステップを具えることを特徴とする請求項15に記載の方法。
